Abstract Plastic pollution is a significant environmental challenge of contemporary age. Polystyrene (PS), among the most commonly used plastic polymers worldwide, is highly durable and difficult to degrade. Despite various disposal strategies, PS continues to impact biodiversity, human health, and ecosystems. Recently, the scientific community has focused on the potential role of microorganisms for plastic biodegradation, particularly those from the gut of plastivorous insects. In a previous study, three bacterial strains, each representing a distinct taxonomic group (Klebsiella, Pseudomonas, and Stenotrophomonas), were isolated from Alphitobius diaperinus larvae after rearing on a PS diet and enriched in a medium with PS as the sole carbon source. The Stenotrophomonas sp. strain, here identified as S. indicatrix, showed the greatest potential for PS degradation. The present study investigates the genetic profile of the newly isolated S. indicatrix strain DAI2m/c through genome sequencing, to identify enzyme-encoding genes involved in the intracellular metabolic pathways responsible for the biodegradation of the styrene monomer. Our findings indicate that the genome of S. indicatrix strain DAI2m/c encodes all enzymes required for one of the two recognized styrene degradation pathways, suggesting its ability to convert styrene into byproducts that are then utilized for cellular energy production.

The complete mitochondrial genome of the shining leaf chafer Mimela junii was sequenced and is herein described. The mitogenome consists of a circular molecule of 16,805 bp, with an overall AT content of 75.7%. It encodes for 13 protein-coding genes (PCGs), 22 transfer RNA genes (tRNAs), two ribosomal RNA genes (rRNAs) and contains a non-coding Control Region (CR) characterized by the presence of tandem repeats. The gene order corresponds to the ancestral Pancrustacea model and mitogenome characteristics are congruous with those of hexapods. In the phylogenetic analysis, M. junii is nested within a paraphyletic Anomala with high support, and is herein associated with Anomala corpulenta with medium/low support.

Children experiencing neurodevelopmental delay have motor, linguistic, and cognitive problems that affect their skills in various domains, especially interpersonal ones. Since music therapy is based on musical exchange, these three case studies explore how child–music therapists' interactions change within and between sessions. Moreover, we evaluated whether the children's and parents' musical interaction changed in everyday life. Three children experiencing neurodevelopmental delay participated in 70 psychodynamic music therapy sessions, relying primarily on musical improvisation techniques. The initial (i.e. pre-test) and final sessions (i.e. post-test) were video recorded and codified with the Kategoriensystem Musik Therapie (KAMUTHE) in the 10th and 30th minutes. Videos were analyzed using the Behavioral Observation Research Interactive Software (BORIS). In addition, parents completed the Music in Everyday Life (MEL) Scale to evaluate how they experienced musical activities with their children. Reliable Change Indexes (RCIs) were calculated to assess significant changes in everyday musical activities. Based on video analysis, three children were more active at the 30th minute and the post-test, while Music Therapists stimulated them less. In addition, therapists adapted the activities and stimuli based on the children's responses. Based on RCIs, parents reported an increased musical engagement with two children. Music therapy might help children experiencing neurodevelopmental delays modify their interaction style and improve motor and interpersonal skills. [ABSTRACT FROM AUTHOR]

Two new mite species belonging to the genus Stereotydeus Berlese, 1901 were discovered from locations along the coast of Victoria Land, continental Antarctica. Previous records of this genus in the area under study only reported the presence of S. belli and S. mollis. Although those studies included no morphological analyses, it has since been assumed that only these species were present within the area. Specimens of S. ineffabilis sp. nov. and S. nunatakis sp. nov. were obtained, sometimes in sympatry, from four different localities in Central and South Victoria Land and are here described and illustrated using optical and scanning electron microscopy (SEM) techniques. Features useful for identification of the two new Stereotydeus species include the size of the specimens, the length of the apical segment of pedipalps, the presence/absence of division of the femora, the position of solenidia, the shape and disposition of the rhagidiform organs on the tarsi, the shape of the apical setae of the tarsi, the numbers of aggenital setae and the position of the anal opening. A key to 14 of the 15 currently described Antarctic and sub-Antarctic Stereotydeus species is provided.

Complete mitochondrial genome data are frequently applied to address phylogenetic/phylogeographic issues at different taxonomic levels in ecology and evolution. While sample preparation/sequencing is becoming more and more straightforward thanks to dropping costs for next-generation sequencing (NGS), data preparation and visualization remains a manually intensive step that may lead to errors if improperly conducted. We have elaborated, and here introduce, EZmito, a simple and intuitive, freely accessible Web Server aimed at automating some of these tasks. EZmito is divided into three main tools: EZpipe that assembles DNA matrices for phylo-mitogenomic analyses; EZskew that calculates genome, strand, and codon nucleotide compositional skews and EZcodon which computes Relative Synonymous Codon Usage statistics as well as amino acid usage frequency over multiple mitogenomes. Output is produced in tabular format as well as publication-quality graphics.

Abstract Collembola are a key component of the soil biota globally, playing an important role in community and ecosystem dynamics. Equally significant are their associated microbiomes, that can contribute to key metabolic functions. In the present study, we investigated the bacterial community composition of four Antarctic springtail species to assess if and how the extreme Antarctic environment has shaped the collembolans’ microbiomes. Springtails were collected from two biogeographical regions, the maritime and the continental Antarctic. From each region, two endemic species, belonging to the genera Cryptopygus (Isotomidae, Entomobryomorpha) and Friesea (Neanuridae, Poduromorpha), were included. This experimental design allowed us to quantify the relative importance of ecological factors (different regions of occurrence) and/or phylogenetic divergence in the host (different Orders) in shaping the Collembola microbiome. The diversity and richness of springtail microbiomes was lower in the Antarctic taxa compared to published information from species from temperate regions. The microbiome composition was predominantly species-specific, with a limited core microbiome shared across the four species examined. While both geographic origin and host species influenced the associated microbiomes, the former was the prevalent driver, with closer similarity between springtails from the same bioregion than between those belonging to the same genus.

The complete mitochondrial genome of the true bug (Homoptera) Nysius cymoides (Spinola, 1837) is herein described and used for phylogenetic comparison with other species of Lygaeoidea. The mtDNA has a gene order and other molecular features typically observed in hexapods, and a long A + T-rich region, due to the occurrence of several repeat units. The phylogenetic analyses support the monophyly of all families except Rhyparochromidae.

The samurai wasp Trissolcus japonicus (Ashmead, 1904) is a parasitoid hymenopteran that came into the limelight as the natural enemy of Halyomorpha halys. Here, we present the complete sequence of the mitochondrial genome of the CREATJ laboratory strain, naturally recovered in Italy in 2018. The molecule conforms to the typical model of animal mitochondrial genomes. Gene order is identical to that of its congeneric Trissolcus basalis. Phylogenetic analysis confirms its placement within monophyletic Scelionidae and Telenominae as the sister group of T. basalis.

Flavonols are known for causing undesirable deposits in both red and white wines. Among flavonols, quercetin is widely considered the principal factor determining this phenomenon. One of the most accredited hypotheses claims that glycosylated derivatives of quercetin undergo hydrolysis of the glycosylic bond during the fermentation and the wine ageing, releasing quercetin aglycone, which is much less soluble in water solution and causes the precipitation. Our work describes the dynamics of quercetin-derived deposition in Chianti wines and purposes a new method, based on the enzymatic quercetin glycoside hydrolysis of the glycosidic bond, to prevent the unpleasant deposit formation during the wine ageing. In our study, forty-four monovarietal wines obtained from 7 different Italian grape varieties were compared in the content of total quercetin-3-glycosides (rutin, quercetin-3-glucuronide, quercetin-3-glucoside) and quercetin aglycone. The data confirmed the literature revealing Sangiovese as the richest in quercetin. We tested then, in a Sangiovese wine, four fining agents (PVPP, PVPP/PVI, bentonite and a vegetal protein) for quercetin removal, showing that only the PVPP had a modest aglycone removal activity. Then, the kinetics of deposit formation was studied in three Chianti wines which differed in the initial content of quercetin aglycone. This investigation highlighted that the chemical equilibrium of quercetin changes over time as the turbidity slowly increases, as previously documented. The comparison of the three dynamics also permitted us to conclude that different wines show a different ability to keep in solution quercetin. Finally, a new approach for deposit prevention was studied by a precocious Chianti wine treatment with a pectolytic enzyme having secondary glycosidase activity. This enzyme significantly accelerated the hydrolysis of glycosylated quercetins into their aglycone, which could enhance the deposition before bottling, without serious wine colour depletion. Our study represents the first evidence of the promising potential of using the pectolytic enzyme with secondary glycosidase activity to prevent quercetin deposit during Chianti ageing, in a way that is compatible with organic wine production.

We describe and delimit with integrative taxonomy the new springtail species Desoria calderonis sp. nov. (Collembola: Isotomidae). This cryophilic species is strictly linked to the supraglacial stony debris of the isolated Calderone glacier (Central Apennines, Italy), one of the southernmost glaciers of Europe. Desoria calderonis sp. nov. could belong to the nivalis-complex, a group of European mountain species included in the violacea-group. Genetic analysis (COI mtDNA barcoding) confirms the morphological attribution to the genus Desoria Nicolet in Desor, 1841, but highlights that the genus, in its current definition, is polyphyletic. We specify the peculiar micro-habitat preferences and highlight the threat of extinction for this cryophilic species in the context of the ongoing climate change and subsequent risk of complete disappearance of the glacier.

The silverfish Neoasterolepisma foreli belongs to the family Lepismatidae within Zygentoma and is well known for the peculiar habit of living in strict association with ant nests (myrmecophily). In this study, we describe its mitochondrial genome, a circular molecule of 15,398 bp including the canonical 13 PCGs, 22 tRNAs, 2 rRNAs, as well as a 403 bp AT-rich region. A phylomitogenomic analysis of the new sequence, alongside basal hexapod mtDNAs, confirmed the monophyly of all orders, with some uncertainty over the position of the enigmatic Tricholepidion gertschi that would make Zygentoma paraphyletic. Neoasterolepisma foreli is recovered in a basal position within family Lepismatidae, at odd with our current understanding of the group that would, in turn, suggest a closer relationship with the genus Lepisma (Mendes, ).

Symphypleona (sensu stricto) are a group of Collembola (=springtails) that, despite displaying some variation in gene order, have been poorly investigated under the phylomitogenomic perspective. How families and subfamilies of this taxon are evolutionary related is still partially unknown. For this reason we sequenced, and herein described, the complete mitochondrial genome sequence of Allacma fusca (Sminthuridae). This sequence, alongside others from the literature, is here used to study the phylogenetic relationships among Symphypleona.

The gut microbiome can play a fundamental role in several processes associated with an organism’s ecology, and research on the microbiota of wild animals has flourished in the last decades. Microbiome composition can vary across and within species according to taxonomy and environmental variability, including the availability of food resources. Species with a large distribution may exhibit spatial patterns acting at local/regional scales. We considered one of the most widespread and ecologically important predators in the world, i.e., the grey wolf Canis lupus, for which microbiome data is unduly limited. We studied four packs in different ecological conditions in Italy—two packs from a Mediterranean coastal area and two packs from an Alpine range—using an amplicon sequencing barcoding approach. Overall, our results are consistent with food habits entailing a diet largely based on wild prey and agree with findings obtained on other species of canids. If confirmed through a larger sample, they would support the hypothesis of an influence of the shared evolutionary history across canids on the composition of the gut microbiome. Some emerging differences were observed among packs in terms of species composition (Jaccard) and diversity, providing partial support to recent indications on pack identity as a significant determinant of microbiome composition. These results should be considered preliminary results of gut microbiome composition in our study areas.

A new cave-dwelling species, Deuteraphorura pieroluccii sp. n., is described from northern Italy. The size of adult specimens, number, and distribution of dorsal and ventral pseudocelli, and number of compound vesicles of the post antennal organ were used to distinguish it among other congeneric species.

In the harsh Antarctic terrestrial ecosystems, invertebrates are currently confined to sparse and restricted ice free areas, where they have survived on multi-million-year timescales in refugia. The limited dispersal abilities of these invertebrate species, their specific habitat requirements, and the presence of geographical barriers can drastically reduce gene flow between populations, resulting in high genetic differentiation. On continental Antarctica, mites are one of the most diverse invertebrate groups. Recently, two new species of the free living prostigmatid mite genus Stereotydeus Berlese, 1901 were discovered, bringing the number of Antarctic and sub-Antarctic species of this genus up to 15, of which 7 occur along the coast of Victoria Land and in the Transantarctic Mountains. To examine the biodiversity of Stereotydeus spp., the present study combines phylogenetic, morphological and population genetic data of specimens collected from nine localities in Victoria Land. Genetically distinct intraspecific groups are spatially isolated in northern Victoria Land, while, for other species, the genetic haplogroups more often occur sympatrically in southern Victoria Land. We provide a new distribution map for the Stereotydeus species of Victoria Land, which will assist future decisions in matters of the protection and conservation of the unique Antarctic terrestrial fauna.

Abstract Gregarines (Apicomplexa) are a diverse group of protozoan parasites, which infects gut and other body cavities of invertebrate hosts. In reproductive system of insects, gregarine has been reported only in the accessory glands and spermathecae of females; therefore, this is the first report of a gregarine species in seminal vesicles of insects. Different developmental stages, including sporozoytes, oocysts and trophozoites were described from morphological descriptions using light and electron transmission microscopy. The parasites were described in seminal vesicles of the beetle Tribolium castaneum a model organism and an important insect pest. DNA sequence analysis suggests that the protozoan parasite was an Ascogregarina sp.

Springtails and mites are the dominant groups of terrestrial arthropods in Antarctic terrestrial ecosystems. Their Antarctic diversity includes a limited number of species, which are frequently endemic to specific regions within the continent. Advances in molecular techniques, combined with the re-evaluation of morphological characters and the availability of new samples, have recently led to the identification of a number of new springtail species within previously named, but ill-defined, species entities described in the last century. One such species, the neanurid Friesea grisea, originally described from sub-Antarctic South Georgia, was for many years considered to be the only known springtail with a pan-Antarctic distribution. With the recent availability of new morphological and molecular data, it has now been firmly established that the different representatives previously referred to this taxon from the Antarctic Peninsula and Victoria Land (continental Antarctica) should no longer be considered as representing one and the same species, and three clearly distinct taxa have been recognized: F. antarctica, F. gretae and F. propria. In this study, the relationships among these three species are further explored through the sequencing of the complete mtDNA for F. gretae and the use of complete mitogenomic as well as cytochrome c oxidase I data. The data obtained provide further support that distinct species were originally hidden within the same taxon and that, despite the difficulties in obtaining reliable diagnostic morphological characters, F. gretae is genetically differentiated from F. propria (known to be present in different locations in Northern Victoria Land), as well as from F. antarctica (distributed in the Antarctic Peninsula).

The complete mitochondrial genome of the machilid Trigoniophthalmus alternatus (Silvestri 1904) is herein described and applied to phylogenetic analyses, inclusive of the most early-divergent lineages of hexapods. Both gene content and order generally conform with the organization of the arthropods’ mitochondrial genome. One gene translocation involving trnA is the autapomorphic character observed in this species. Another peculiar molecular feature is the long size of the A + T-rich region, due to the occurrence of repeat units. The phylogenetic analyses support the typical placement, along the hexapods’ tree, of Ectognatha, Monocondylia and Dicondylia, with Diplura as the adelphotaxon of all true insects.

The complete mitochondrial genome of the springtail Bourletiella arvalis (Fitch, 1863) is herein described and applied to a Bayesian phylogenetic analysis, inclusive of all the Collembola mitochondrial DNAs sequenced so far. The gene content and order, as well as the nucleotide composition, conform with the well-known features of hexapods’ mitochondrial genomes. The phylogenetic analysis supports the monophyly of Collembola, Poduromorpha, Entomobryomorpha and Symphypleona. However, no mtDNA from Neelipleona is available to date, therefore limiting the application of mitochondrial genomes to further investigate springtail systematics.
