11. Anatomical, functional and dynamic MRI features of the G144 GBM model developed in NOD-SCID mice

Abstract

Preclinical models are needed to study glioblastoma multiforme, the most lethal malignant brain tumor. Human G144-glioblastoma stem cells have been used in in vitro studies but there are few studies in vivo, especially using MRI approaches. We aimed to evaluate in vivo, for the first time to our knowledge, a G144 glioblastoma model by multiparametric-MRI. NOD-SCID mice were orthotopically injected with G144 cells, tumor was characterized by multiparametric-MRI and dynamic contrast enhancement studies. G144 tumors presented heterogeneous imaging pattern compared to other GBM models with higher contrast uptake areas and edema related features.

Email: paulomoreira@por.ulusiada.pt The clients’ belief systems are components of Effective Therapy Relationships. Thus, it is desirable to include clients’ beliefs about their psychological problems on systematic assessment protocols underlying the process of systematic treatment selection and of tailoring the treatment to the person. However, assessment instruments which specifically capture clients’ beliefs about their psychological problems are scarce. The objective of the studies presented was to evaluate the psychometric properties of the Beliefs About Psychological Problems Inventory (BAPPI), an assessment instrument of the clients’ beliefs about their psychological problems. Study 1 (Exploratory Factor Analysis) involved 200 participants, and Study 2 (Confirmatory Factor Analysis and other validity studies), involved 545 participants. Results revealed that the BAPPI presents a stable factorial structure of six dimensions (Psychodynamic, Humanistic, Biomedical, Cognitive-Behavioral, Systemic, and Eclectic/Integrative). Altogether, analyses of items, internal consistency, reliability, and external validity revealed that the BAPPI is a valid assessment instrument for use in mental health research and practice, especially in the process of systematic treatment selection and, therefore, of matching/tailoring the treatment to the client’s characteristics.

14. Multiparametric MRI characterization of the G144 GBM model

Trabajo presentado en el ESMRMB 38th Annual Scientific Meeting, celebrado en modalidad virtual del 07 al 09 de octubre de 2021., [Introduction]: Different orthotopic murine preclinical models have been developed to study the diagnosis, progression and therapy validation of glioblastoma multiforme (GBM)1, the most frequent and lethal malignant brain tumor2. Among them, the xenograft model established by stereotactic injection of the human G144 stem cells emerges as one of the best approaches to replicate the human GBM features in immunocompromised mice and to address GBM genetics and histopathology3,4,5. Magnetic resonance imaging (MRI) is a powerful tool to obtain structural, functional and molecular information on the physiopathology of intracranial tumors, but very few studies have been performed with this GBM model4. The aim of this work is to characterize the preclinical G144 glioma model by multiparametric (MP)-MRI approaches. [Methods]: The orthotopic GBM was developed in NOD-SCID mice (n=5) by injecting 105 G144 cells in the right caudate nucleus. MRI studies were carried out in a Bruker 7T system. Tumor growth was followed up by T2 weighted (T2W) images, and MP-MRI studies were performed when the tumor volume was ~70 mm3: T2 maps, magnetization transfer (MT) images, diffusion weighted images and dynamic contrast enhancement (DCE) using the intravenous injection of 0.25 mmol/Kg of Gadovist® (Bayer) as contrast agent (CA). Parametric maps were generated with a home-made software built in Matlab and relative contrast enhancement (RCE) maps were obtained using the software DCE@ourlab6. Regions of interest (ROIs) were manually selected to quantify the obtained parameters. [Results]: Tumors were visualized in T2W MRI 30-50 days post cell-injection (p.i.) and the volume to perform the MP-MRI studies was reached 53-104 days p.i. Hyperintensity spots were heterogeneously observed in the tumor in T2W and in contrast-enhance T1W (CE-tumor) images (Figure 1). DCE studies showed the progressive and faster accumulation of CA in the CE-tumor ROI than in the rest of the ROIs (Figure 2). CEtumor also showed higher T2 and Mean Diffusivity (MD) values, and lower MT and Fractional Anisotropy (FA) values (Figure 3). [Discussion]: To our knowledge, this is the first MP-MRI characterization of a GBM humanized mouse model established from G144 cells. The results point that necrosis, inflammatory processes and loss of structural integrity of the tissue are occurring in the CE-tumor area compared with the rest of the ROIs, similarly to MRI characteristics from human GBM patients7. In order to improve the knowledge of this GBM model utility, further experiments will be performed to evaluate different therapies effects in the MRI parameters.

Abstract

Gold nanoparticles (AuNPs) are interesting for the design of new cancer theranostic tools, mainly due to their biocompatibility, easy molecular vectorization, and good biological half-life. Herein, we report a gold nanoparticle platform as a bimodal imaging probe, capable of coordinating Gd3+ for Magnetic Resonance Imaging (MRI) and 67Ga3+ for Single Photon Emission Computed Tomography (SPECT) imaging. Our AuNPs carry a bombesin analogue with affinity towards the gastrin releasing peptide receptor (GRPr), overexpressed in a variety of human cancer cells, namely PC3 prostate cancer cells. The potential of these multimodal imaging nanoconstructs was thoroughly investigated by the assessment of their magnetic properties, in vitro cellular uptake, biodistribution, and radiosensitisation assays. The relaxometric properties predict a potential T1- and T2- MRI application. The promising in vitro cellular uptake of 67Ga/Gd-based bombesin containing particles was confirmed through biodistribution studies in tumor bearing mice, indicating their integrity and ability to target the GRPr. Radiosensitization studies revealed the therapeutic potential of the nanoparticles. Moreover, the DOTA chelating unit moiety versatility gives a high theranostic potential through the coordination of other therapeutically interesting radiometals. Altogether, our nanoparticles are interesting nanomaterial for theranostic application and as bimodal T1- and T2- MRI / SPECT imaging probes.

43. Fragmentation Reactions of Molecular Ions and Dications of Indoleamines

Abstract

The combination of techniques such as two-dimensional mass spectrometry and mass analysed ion kinetic energy spectrometry (with and without collision gas) in the study of gas-phase ion chemistry of indole, gramine, tryptamine and serotonine enabled us to establish the fragmentation pathways of their singly- and doubly-charged molecular ions generated by electron impact. The measurement of the kinetic energy released in the charge separation reactions of the molecular dications produced information on the intercharge distance on the transition state for the fragmentation, assuming that it arises entirely from the coulombic energy released. For the singly-charged molecular ions of indoleamines, loss of the neutral molecule CH2=NR (R = H or CH3), by hydrogen transfer from the amino (or methyl) group to the indole nucleus, is the only fragmentation occurring in the third field-free region of the mass spectrometer. Upon collision of the molecular ion with He gas, a competing process of bond cleavage by loss of the radical •CH2NH2(or •N(CH3)2for gramine) is observed for tryptamine and serotonine. The doubly-charged ions dissociate by elimination of neutral molecules or radicals to generate doubly-charged fragment ions or by charge separation reactions. The intercharge distance, calculated from the kinetic energy released in the fragmentation, was, whenever possible, correlated with charge location in the ion.

Abstract

The identification of testosterone, pregnelonone, and 17a-hydroxyprogesterone by tandem mass spectrometry and of progesterone by sequential product ion scanning in the shrimp gonads of Penaeus japonicusis described. The identification of these substances in biological samples is usually done by gas chromatography-mass spectrometry and involves several liquid chromotographic purification steps followed by derivatization. The utilization of tandem mass spectrometry in this analysis has simplified considerably the sample pretreatment and provided a very simple method of screening these substances in complex mixtures.

It is shown by mass-analyzed ion kinetic energy spectrometry that the metastably decomposing molecular ions of octopamine ( p-HOC 6H 4CH(OH)CH 2NH 2) and synephrine ( p-HOC 6H 4CH(OH)CH 2NHCH 3) yield only protonated methylamine and dimethylamine, respectively, as product ions. From deuterium labeling and variation of the internal energy of the molecular ions, experimental support has been obtained that these product ions are generated via the occurrence of a distonic ion-neutral complex. In the case of octopamine, this complex would consist of a nitrogen-protonated aminomethyl radical and p-hydroxylbenzaldehyde in which the former species abstracts the aldehydic or phenolic hydrogen atom from the latter to give protonated dimethylamine.

Abstract

Having 2 or more relatives involved in the informal care of people with dementia is frequent worldwide. There are, however, few comparisons of primary and secondary caregivers and even fewer of those who are caring for the same person. Our study aimed to contrast these 2 experiences of caregiving. We compared 2 related samples of 61 primary and 61 secondary family caregivers of the same persons with dementia in a nonrandomized cross-sectional study. Caregivers' main outcome assessments were the Zarit Burden Interview (for subjective burden), the General Health Questionnaire (for psychological distress), and the Positive Aspects of Caregiving scale. We controlled for caregiver variables (e.g., demographics, caregiving arrangements, social support, sense of coherence) and the neuropsychiatric symptoms of dementia. Subjective burden was higher in primary than secondary caregivers (p = .013), but positive aspects of caregiving did not differ (p = .150). Psychological distress was high at clinically relevant levels in primary and secondary caregivers, without statistically significant differences between groups (p = .456). The findings demonstrate that notwithstanding the difficulties faced by primary caregivers, secondary caregivers may also experience clinically significant distress. Therefore, their needs for assistance and support should be addressed more systematically. These findings call for systemic family-focused interventions in dementia that address the support each person provides or might provide, as well as the psychological distress each person may feel. Abstract

Among all major organs, the brain is one of the most susceptible to the inexorable effects of aging. Throughout the last decades, several studies in human cohorts and animal models have revealed a plethora of age-related changes in the brain, including reduced neurogenesis, oxidative damage, mitochondrial dysfunction and cell senescence. As the main immune effectors and first responders of the nervous tissue, microglia are at the center of these events. These cells experience irrevocable changes as a result from cumulative exposure to environmental triggers, such as stress, infection and metabolic dysregulation. The age-related immunosenescent phenotype acquired by microglia is characterized by profound modifications in their transcriptomic profile, secretome, morphology and phagocytic activity, which compromise both their housekeeping and defensive functions. As a result, aged microglia are no longer capable of establishing effective immune responses and sustaining normal synaptic activity, directly contributing to age-associated cognitive decline and neurodegeneration. This review discusses how lifestyle and environmental factors drive microglia dysfunction at the molecular and functional level, also highlighting possible interventions to reverse aging-associated damage to the nervous and immune systems., (Copyright © 2021 The Author(s). Abstract

Water soluble phthalocyanines bearing either four PEG500 or four choline substituents in the macrocyclic structure, as well as their Zn(II) and Mn(III) complexes were synthesized. The metal-free and Zn(II) complexes present relatively high fluorescence quantum yields (up to 0.30), while the Mn(III) complexes show no fluorescence as a consequence of rapid non-radiative deactivation of the Mn(III) phthalocyanine excited states through low-lying metal based or charge-transfer states. The effect of DMSO on the aggregation of the phthalocyanines was studied. It was not possible to obtain the Mn(II) complexes by reduction of the corresponding Mn(III) complexes due to the presence of electron donating substituents at the periphery of the phthalocyanines. The (1)H NMRD plots of the PEG500 and choline substituted Mn(III)-phthalocyanine complexes are typical of self-aggregated Mn(III) systems with r1 relaxivities of 4.0 and 5.7mM(-1)s(-1) at 20MHz and 25°C. The Mn(III)-phthalocyanine-PEG4 complex shows no significant cytotoxicity to HeLa cell cultures after 2h of incubation up to 2mM concentration. After 24h of cell exposure to the compound, significant toxicity was observed for all the concentrations tested with IC50 of 1.105mM., (Copyright © 2015 Elsevier Inc. All rights reserved.)
