Abstract

[ES] Nuestro objetivo fue investigar fa incidencia de la resistencia a antimicrobianos en bacterias del ácido láctico (SAL) aisladas de la fermentación natural de aceitunas Aloreña. Para ello, los métodos fenotípicos y genotípicos permitieron determinar que las SAL poseían una resistencia intrínseca múltiple a varios antibióticos, aunque eran sensibles a los biocidas. Así mismo, hemos determinado la respuesta de las SAL a diferentes tipos de estrés obteniéndose una respuesta fenotípica uniforme, sin embargo el repertorio de genes inducidos y reprimidos diferían en función del tipo de estrés y de fa cepa SAL. Y para mejorar la tolerancia al estrés de las SAL, la adaptación al estrés antimicrobiano de una cepa seleccionada ( Lactobacillus pentosus MP-10) permitió ajustar el arsenal proteómico de dicha bacteria como una estrategia de supervivencia. De otra parte, hemos analizado a nivel genómico fa seguridad de Lb. pentosus MP-10 con potencial probiótico y los resultados obtenidos confirmaron su inocuidad. [EN] Our objective was to investigate the incidence of antimicrobial resistance in lactic acid bacteria (LAS) isolated from the natural fermentation of Aloreña olives. For this, phenotypic and genotypic methods allowed to determine that LAB possessed a muftiple intrinsic resistance to severa! antibiotics, although they were sensitive to biocides. Also, we have determined the response of LAS to different types of stress obtaining a uniform phenotypic response, however the repertoire of genes up-regulated and down-regulated differed depending on the type of stress and the LAS strain. With the aim to improve stress tolerance of LAS, antimicrobial stress adaptation of a selected strain (Lactobacillus pentosus MP- 1O) allowed to adjust the proteomic arsenal of such bacteria as a survival strategy. On the other hand, we have analyzed the safety of a potentially probiotic Lb. pentosus MP-10 at genomic level and the results confirmed its safety. Tesis Univ. Jaén. Abstract

In this study, we compared the activity of four different xylanases (GH 5_34, 8, 10 and 11) for the production of arabinoxylan-oligosaccharide or (A)XOS from cereal arabinoxylans as potential prebiotics. In terms of efficiency, the AcXyn10A and TlXyn11 produced larger quantities of small (A)XOS of mainly X1-X3 in a shorter period of time. The BXyn8 was most restricted by high arabinose substitution but could still accommodate them at the +2 subsite. The GpXyn5_34 preferred high arabinose substitution, requiring an arabinose at the -1 subsite for cleavage. Both the BXyn8 and GpXyn5_34 produced longer (A)XOS that were mostly linear and substituted, respectively. During fermentation of (A)XOS on selected gut bacteria, the long-substituted products of GpXyn5_34 stunted the growth of B. ovatus and B. adolescentis, while the mid-length, linear products of BXyn8 favoured the growth of both bacteria., QC 20220210
