Serotiny degree may hypothetically depend on mother plant age and fire severity, and fire severity can strongly affect the regeneration of Pinus pinaster Aiton, which is an obligate seeder species with aerial seed banks stocked in cones showing a certain degree of serotiny. The main objective of this study was to evaluate the effect of serotiny and mother plant age on seed germination of P. pinaster in relation to fire severity. For this purpose, cones were collected in two stands from mother trees of different ages. Serotinous cones were opened in oven at increasing temperatures to determine the number of seeds released. The length of cones and the number and weight of seeds were measured. Seeds from both classes of mother plant age were subjected to 34 different treatments simulating different fire severity levels, and their germination was recorded. The results showed that serotiny was significantly higher in cones from young mother trees. Seed germination in the control groups from serotinous cones was always higher than that from non-serotinous cones without thermal shock. Fire severity decreased seed germination as fire severity increases in both mother age classes. Mother plant age was related to the serotiny level, which was higher in young than in old trees, suggesting a strong selection in the population due to a recent fire.

A total of 163 clinical samples (128 pulmonary and 35 extra-pulmonary) were processed using four PCR assay kits: Abbott RealTime MTB RIF/INH, Genotype MTBDRplus, Xpert/MTB RIF, and Anyplex MTB/MDR. The results of phenotypic drug-susceptibility testing using BACTECMGIT 960 were used as reference. The sensitivity and specificity of the new Abbott RealTime MTB RIF/INH assay in comparison with phenotypic testing was 96.3% (95%CI 87.32%–100%) for RIF and 100% (95%CI 99.3%–100%) for INH; the sensitivity was 78.8% (95%CI 66.8%–90.9%) and the specificity was 100% (95%CI 98.9%–100%). The Abbott RealTime MTB RIF/INH test could be a valid method for detecting the most common mutations in strains resistant to RIF and INH. It has long been assumed that lead glazing technology preceded glassmaking in the Western world and that the technological transfer was from glazes to glass. Here, we present new evidence for the reverse, the indigenous innovation of glassmaking and its transfer to glazes in early Islamic al-Andalus (Spain). Compositional analyses show that Islamic lead glazes from Córdoba are intimately related to a distinct type of high-lead glass, suggesting a connection between the two technologies. The archaeological remains from a pottery workshop indicate that the glazing process initially involved the production of a lead glass and is not linked to earlier Roman or other contemporary glazing technologies. The data also demonstrate that the potters not only used the same materials and techniques but borrowed stylistic and decorative models from glassmaking. [ABSTRACT FROM AUTHOR]
