4. Futures Education: Curriculum and Educational Practices in Australia, Spain, and Chile

Abstract

The images of the future among young people have been conditioned by the stories present in the media, films, books, and also in school. Educational curriculums are made up of a selection of knowledge that privileges some ways of understanding the future over others. Young people often imagine a future that is in economic, social, and/or climate crisis. However, they also imagine a bright future for themselves, detached from the future they imagine for society. In this article, we present a qualitative analysis of the curriculums of Australia, Spain, and Chile, together with interviews with teachers from these countries. We investigate the presence and absence of futures education in these curriculums, their degree of development regarding futures education, and make a first analysis of the influence of futures education in schools. To do this we identify four dimensions: situate in time, anticipate, imagine alternative futures, and social action. The results show that, while the Australian curriculum explicitly includes education for the future, the Spanish and Chilean curriculum include it only tangentially. In addition, the socio-cultural context of schools and the will of the teaching staff are elements that determine the implementation of futures education in the school context.

7. National Curricula and Citizenship Education in Populist Times. The Cases of Brazil and Spain

Abstract

Despite increasing attention to citizenship education since the turn of the 21st century, the recent spread of authoritarian populism worldwide has raised relatively little attention in educational policy and research. As a result, the possibilities and limitations that national curricula offer to educators to deal with this phenomenon are still rather uncertain. In this article, we develop an analytical framework based on the key features of authoritarian populism and critical citizenship education to compare the elements and scope for addressing populism in the national curricula of Brazil and Spain, two countries where national populism is particularly widespread. This paper examines the extent to which national curricula in these countries include goals and content that enable teachers to address the complexities of this phenomenon ranging from political polarisation through to the exaltation of national identities.

10. Chordal graphs with bounded tree-width

Abstract

Given $t\geq 2$ and $0\leq k\leq t$, we prove that the number of labelled $k$-connected chordal graphs with $n$ vertices and tree-width at most $t$ is asymptotically $c n^{-5/2} \gamma^n n!$, as $n\to\infty$, for some constants $c,\gamma >0$ depending on $t$ and $k$. Additionally, we show that the number of $i$-cliques ($2\leq i\leq t$) in a uniform random $k$-connected chordal graph with tree-width at most $t$ is normally distributed as $n\to\infty$. The asymptotic enumeration of graphs of tree-width at most $t$ is wide open for $t\geq 3$. To the best of our knowledge, this is the first non-trivial class of graphs with bounded tree-width where the asymptotic counting problem is solved. Abstract

ObjectivesEarly- and late-onset Alzheimer's disease (EOAD and LOAD) share the same neuropathological traits but show distinct cognitive features. We aimed to explore baseline and longitudinal outcomes of global and domain-specific cognitive function in a well characterized cohort of patients with a biomarker-based diagnosis.MethodsIn this retrospective cohort study, 195 participants were included and classified according to their age, clinical status, and CSF AD biomarker profile: 89 EOAD, 37 LOAD, 46 young healthy controls (age ≤ 65 years), and 23 old healthy controls (>65 years). All subjects underwent clinical and neuropsychological assessment, neuroimaging, APOE genotyping and lumbar puncture.ResultsWe found distinct neuropsychological profiles between EOAD and LOAD at the time of diagnosis. Abstract

We determine the number of labelled chordal planar graphs with $n$ vertices, which is asymptotically $c_1\cdot n^{-5/2} \gamma^n n!$ for a constant $c_1>0$ and $\gamma \approx 11.89235$. We also determine the number of rooted simple chordal planar maps with $n$ edges, which is asymptotically $c_2 n^{-3/2} \delta^n$, where $\delta = 1/\sigma \approx 6.40375$, and $\sigma$ is an algebraic number of degree 12. The proofs are based on combinatorial decompositions and singularity analysis. Chordal planar graphs (or maps) are a natural example of a subcritical class of graphs in which the class of 3-connected graphs is relatively rich. Abstract

BackgroundThe literature on associations of circulating concentrations of minerals and vitamins with risk of colorectal cancer is limited and inconsistent. Evidence from randomized controlled trials (RCTs) to support the efficacy of dietary modification or nutrient supplementation for colorectal cancer prevention is also limited.ObjectivesTo complement observational and RCT findings, we investigated associations of genetically predicted concentrations of 11 micronutrients (β-carotene, calcium, copper, folate, iron, magnesium, phosphorus, selenium, vitamin B-6, vitamin B-12, and zinc) with colorectal cancer risk using Mendelian randomization (MR).MethodsTwo-sample MR was conducted using 58,221 individuals with colorectal cancer and 67,694 controls from the Genetics and Epidemiology of Colorectal Cancer Consortium, Colorectal Cancer Transdisciplinary Study, and Colon Cancer Family Registry. Inverse variance-weighted MR analyses were performed with sensitivity analyses to assess the impact of potential violations of MR assumptions.ResultsNominally significant associations were noted for genetically predicted iron concentration and higher risk of colon cancer [ORs per SD (ORSD): 1.08; 95% CI: 1.00, 1.17; P value = 0.05] and similarly for proximal colon cancer, and for vitamin B-12 concentration and higher risk of colorectal cancer (ORSD: 1.12; 95% CI: 1.03, 1.21; P value = 0.01) and similarly for colon cancer. A nominally significant association was also noted for genetically predicted selenium concentration and lower risk of colon cancer (ORSD: 0.98; 95% CI: 0.96, 1.00; P value = 0.05) and similarly for distal colon cancer. These associations were robust to sensitivity analyses. Nominally significant inverse associations were observed for zinc and risk of colorectal and distal colon cancers, but sensitivity analyses could not be performed. None of these findings survived correction for multiple testing. Genetically predicted concentrations of β-carotene, calcium, copper, folate, magnesium, phosphorus, and vitamin B-6 were not associated with disease risk.ConclusionsThese results suggest possible causal associations of circulating iron and vitamin B-12 (positively) and selenium (inversely) with risk of colon cancer.

22. Assessing the Relationship between School Failure and Suicidal Behavior in Adolescents and Young Adults: A Systematic Review and Meta-Analysis of Longitudinal Studies

Abstract

There is an increasing acknowledgment that suicidal behaviors are negatively affected by school failure in adolescents and young adults. However, no systematic reviews with meta-analysis have been performed to evaluate the strength of this association. We assessed the magnitude of association of school failure as risk factor for suicide behavior in adolescents and young adults. Six international databases were searched up until January 2017. Inclusion criteria were: (1) assessment of suicide attempt or suicide; (2) case-control or cohort studies; (3) subjects aged 12-26 years; (4) school failure evaluated, which includes low academic performance, school dropout, school expulsion, and grade repetition. Random effect models were used; population attributable risks (PARs) were estimated. From 26,883 potentially eligible articles, 14 (62,298 individuals) were included. Meta-analyses could only be performed for suicide attempt. A history of school dropout (OR 6.44, 95% CI 3.03-13.65), low academic performance (OR 1.48, 95% CI 1.22-1.81), and school failure (OR 1.98, 95% CI 1.49-2.64) were significant risks of suicide attempts. Highest PAR estimates were 9.9% for school dropout, 5.1% for low academic performance, and 6.7% for any school failure. Only one study has been found for suicide, but results suggest an association with school dropout. Results show that the students who had school failure are at higher risk of suicide attempt and probably for suicide. The development of effective programs to prevent suicidal behaviors in educational settings may be an effective strategy to reduce these behaviors. However, reliable conclusions from this review are limited by the small amount of included studies for analysis.

Abstract

The persistence of HIV reservoirs, including latently infected, resting CD4+ T cells, is the major obstacle to cure HIV infection. CD32a expression was recently reported to mark CD4+ T cells harboring a replication-competent HIV reservoir during antiretroviral therapy (ART) suppression. We aimed to determine whether CD32 expression marks HIV latently or transcriptionally active infected CD4+ T cells. Using peripheral blood and lymphoid tissue of ART-treated HIV+ or SIV+ subjects, we found that most of the circulating memory CD32+ CD4+ T cells expressed markers of activation, including CD69, HLA-DR, CD25, CD38, and Ki67, and bore a TH2 phenotype as defined by CXCR3, CCR4, and CCR6. CD32 expression did not selectively enrich for HIV- or SIV-infected CD4+ T cells in peripheral blood or lymphoid tissue; isolated CD32+ resting CD4+ T cells accounted for less than 3% of the total HIV DNA in CD4+ T cells. Cell-associated HIV DNA and RNA loads in CD4+ T cells positively correlated with the frequency of CD32+ CD69+ CD4+ T cells but not with CD32 expression on resting CD4+ T cells. Using RNA fluorescence in situ hybridization, CD32 coexpression with HIV RNA or p24 was detected after in vitro HIV infection (peripheral blood mononuclear cell and tissue) and in vivo within lymph node tissue from HIV-infected individuals. Together, these results indicate that CD32 is not a marker of resting CD4+ T cells or of enriched HIV DNA-positive cells after ART; rather, CD32 is predominately expressed on a subset of activated CD4+ T cells enriched for transcriptionally active HIV after long-term ART.

Abstract

Non-small-cell lung cancer patients with activating epidermal growth factor receptor (EGFR) mutations typically benefit from EGFR tyrosine kinase inhibitor treatment. However, virtually all patients succumb to acquired EGFR tyrosine kinase inhibitor resistance that occurs via diverse mechanisms. The diversity and unpredictability of EGFR tyrosine kinase inhibitor resistance mechanisms presents a challenge for developing new treatments to overcome EGFR tyrosine kinase inhibitor resistance. Here, we show that Akt activation is a convergent feature of acquired EGFR tyrosine kinase inhibitor resistance, across a spectrum of diverse, established upstream resistance mechanisms. Combined treatment with an EGFR tyrosine kinase inhibitor and Akt inhibitor causes apoptosis and synergistic growth inhibition in multiple EGFR tyrosine kinase inhibitor-resistant non-small-cell lung cancer models. Moreover, phospho-Akt levels are increased in most clinical specimens obtained from EGFR-mutant non-small-cell lung cancer patients with acquired EGFR tyrosine kinase inhibitor resistance. Our findings provide a rationale for clinical trials testing Akt and EGFR inhibitor co-treatment in patients with elevated phospho-Akt levels to therapeutically combat the heterogeneity of EGFR tyrosine kinase inhibitor resistance mechanisms.EGFR-mutant non-small cell lung cancer are often resistant to EGFR tyrosine kinase inhibitor treatment. In this study, the authors show that resistant tumors display high Akt activation and that a combined treatment with AKT inhibitors causes synergistic tumour growth inhibition in vitro and in vivo.

33. Decreased practice effects in preclinical Alzheimer's disease: a multicenter, longitudinal, cohort study.

Abstract

Background: Practice effects are a well‐known cognitive phenomenon that is reduced in patients with Alzheimer's disease (AD). We aimed to investigate whether cognitively unimpaired (CU) individuals within the Alzheimer's continuum (i.e., positive amyloid‐β biomarker) display decreased practice effects on serial neuropsychological testing. Methods: We included 310 CU from four Spanish research centers, classified into controls (n = 250) or Aβ+ (n = 60). In the main cohort (Cohort A; n = 209), participants underwent neuropsychological assessment at baseline and annually during a 2‐year follow‐up (FU1 and FU2). A "long‐term cohort" (Cohort B; n = 101) was employed to assess practice effects over longer time periods (i.e., two follow‐up sessions at years 3 and 6 from baseline). Practice effects were defined as simple discrepancy scores (SDS) by subtracting Z‐scores at FU2 from Z‐scores at baseline for each neuropsychological variable, and linear mixed effects models (LME) were run to assess the temporal evolution of practice effects according to the two groups. Results: There were no cross‐sectional differences between the control and Aβ+ groups in none of the neuropsychological scores at baseline (Fig. 1). The Aβ+ group displayed lower practice effects than the controls in terms of SDS in several neuropsychological outcomes (Fig. 2). In Cohort A, LME showed negative slopes by the Aβ+ group in verbal memory measures such as the free learning score (β = ‐0.37, SD = 0.12, p = 0.0034), delayed free recall (β = ‐0.43, SD = 0.15, p = 0.0047) and delayed total recall (β = ‐0.46, SD = 0.17, p = 0.0069) from the Free and Cued Selective Reminding Test; as well as in language tasks (Boston Naming Test; β = ‐0.26, SD = 0.087, p = 0.0025) and executive function measures (Trail Making Test; β = ‐0.33, SD = 0.12, p = 0.0094) (Fig. 3A). In Cohort B, similar findings were observed in visual memory measures, such as the Rey‐Osterrieth Complex Figure immediate (β = ‐0.80, SD = 0.35, p = 0.024) and delayed (β = ‐1.25, SD = 0.34, p = 0.00038) recall (Fig. 3B). Conclusions: Individuals with normal cognition who are in the Alzheimer's continuum show decreased practice effects over serial neuropsychological testing. Our findings suggest the reduction of practice effects, particularly in memory measures, as an indicator of subtle cognitive decline in the earliest phase of the Alzheimer's continuum and could be particularly relevant for the design and interpretation of primary prevention trials on disease‐modifying therapies. 41. Prevalence and Characteristics of Autism Spectrum Disorder among Spanish School-Age Children

Abstract

The present study aims to assess the prevalence of autism spectrum disorder (ASD) in preschool and school-age children following a two-phase procedure. The screening phase was performed on a sample of 5555 children taking into account parent and teacher information. The individual assessment included the ADI-R, ADOS-2 and Wechsler scales. The estimated prevalence was 1.55% in preschoolers and 1.00% in school-age children. Between 1.84 and 2.59% of the children exhibited subclinical diagnosis. The male-to-female ratio was around 4:1. Most of the children exhibited mild and moderate nuclear symptoms, and the girls showed less severe communication problems. Previous diagnosis was found in 62-71% of the children. Prevalence estimates are close to the 1% international ratings and much higher than previous national reports suggested.

Abstract

Strategies for the early detection of autism spectrum disorders (ASD) in people with intellectual developmental disorder (IDD) are urgently needed, but few specific tools have been developed. The present study examines the psychometric properties of the EVTEA-DI, a Spanish adaptation of the PDD-MRS, in a large randomized sample of 979 adults with IDD. Factorial solution analysis suggested a three-factor solution (stereotyped behavior, communication, and social behavior). The EVTEA-DI showed good reliability and convergent validity when compared to the Childhood Autism Rating Scale. Discriminative validity analysis resulted in an acceptable global sensitivity of 70% and a high specificity of 90%. The EVTEA-DI proved to be a valid screening tool in ASD assessment of the adult Spanish population with IDD.

Abstract

Patients with acute respiratory distress syndrome often require mechanical ventilation to maintain adequate gas exchange and to reduce the workload of the respiratory muscles. Although lifesaving, positive pressure mechanical ventilation can potentially injure the lungs and diaphragm, further worsening patient outcomes. While the effect of mechanical ventilation on the risk of developing lung injury is widely appreciated, its potentially deleterious effects on the diaphragm have only recently come to be considered by the broader intensive care unit community. Importantly, both ventilator-induced lung injury and ventilator-induced diaphragm dysfunction are associated with worse patient-centered outcomes.

Abstract

Background: Neuropsychological performance guides diagnostic and therapeutic decision‐making on Alzheimer's disease (AD) and related disorders. Despite broad recognition that amyloid‐beta (Aβ) impacts cognition during preclinical AD, the added value of Aβ‐negative norms remains uncertain. Furthermore, normative modeling is constrained by limitations inherent to traditional methods. Here, we derived next‐generation norms (NGN) for a comprehensive neuropsychological battery and compared their utility with that of traditional norms (TN). Methods: This multicenter study included 2405 non‐demented participants conventionally identified as cognitively normal (CN, n = 987) or with mild cognitive impairment (MCI, n = 1418) (Table 1). All participants had extensive neuropsychological data and CSF AD biomarkers at baseline. Based on the performance of 774 Aβ‐negative CN individuals, we derived NGN using generalized additive models for location, scale, and shape (GAMLSS). First, we compared the accuracy of NGN and TN to discriminate Aβ‐positive from Aβ‐negative MCI participants. Next, we actuarially reclassified all participants as CN or MCI according to both TN and NGN, and we examined the concordance of these classifications with AD biomarkers. For a subset of participants with clinical follow‐up (n = 1318), linear mixed‐effects modeling was used to capture the longitudinal change in Clinical Dementia Rating Scale‐Sum of Boxes. The Akaike Information Criteria (AIC) was employed to select the best model. Results: Age‐, education‐, and sex‐adjusted percentiles were obtained for 14 neuropsychological measures across main cognitive domains (memory, language, attention, executive functioning, and visuospatial skills) (Table 2). NGN outperformed TN at detecting prodromal AD for verbal memory measures, with all but one of the differences reaching statistical significance (p<0.05). Most participants were actuarially reclassified as either CN (43%) or MCI (52%) based on both TN and NGN. Among conflicting diagnoses (5%), the MCI/CN group (3%) ‐individuals diagnosed with MCI according to TN but considered CN as per NGN‐ was analogous to the CN/CN group, while the CN/MCI (2%) paralleled the MCI/MCI group in their rates of Aβ‐positivity (Figure 1). Cognitive status based on NGN better predicted clinical progression than TN (AIC = 5195.5 vs 5206.1, respectively). Conclusion: NGN incorporating Aβ status and GAMLSS yield stronger associations with biomarkers and disease progression than TN. This has direct implications for clinical practice and research.
