Abstract

Introduction Lung cancer (LC) is the most common cause of cancer-related deaths worldwide. Its early detection can be achieved with a CT scan. Two large randomised trials proved the efficacy of low-dose CT (LDCT)-based lung cancer screening (LCS) in high-risk populations. The decrease in specific mortality is 20%–25%.Nonetheless, implementing LCS on a large scale faces obstacles due to the low number of thoracic radiologists and CT scans available for the eligible population and the high frequency of false-positive screening results and the long period of indeterminacy of nodules that can reach up to 24 months, which is a source of prolonged anxiety and multiple costly examinations with possible side effects.Deep learning, an artificial intelligence solution has shown promising results in retrospective trials detecting lung nodules and characterising them. However, until now no prospective studies have demonstrated their importance in a real-life setting.Methods and analysis This open-label randomised controlled study focuses on LCS for patients aged 50–80 years, who smoked more than 20 pack-years, whether active or quit smoking less than 15 years ago. Its objective is to determine whether assisting a multidisciplinary team (MDT) with a 3D convolutional network-based analysis of screening chest CT scans accelerates the definitive classification of nodules into malignant or benign. 2722 patients will be included with the aim to demonstrate a 3-month reduction in the delay between lung nodule detection and its definitive classification into benign or malignant.Ethics and dissemination The sponsor of this study is the University Hospital of Nice. The study was approved for France by the ethical committee CPP (Comités de Protection des Personnes) Sud-Ouest et outre-mer III (No. 2022-A01543-40) and the Agence Nationale du Medicament et des produits de Santé (Ministry of Health) in December 2023. The findings of the trial will be disseminated through peer-reviewed journals and national and international conference presentations.Trial registration number NCT05704920.

Abstract

Synchrotron Laue microdiffraction and Digital Image Correlation measurements were coupled to track the elastic strain field (or stress field) and the total strain field near a general grain boundary in a bent bicrystal. A 316L stainless steel bicrystal was deformed in situ into the elasto-plastic regime with a four-point bending setup. The test was then simulated using finite elements with a crystal plasticity model comprising internal variables (dislocation densities on discrete slip systems). The predictions of the model have been compared with both the total strain field and the elastic strain field obtained experimentally. While activated slip systems and total strains are reasonably well predicted, elastic strains appear overestimated next to the grain boundary. This suggests that conventional crystal plasticity models need improvement to correctly model stresses at grain boundaries.

4. Evidence of Dislocation Mixed Climb in Quartz From the Main Central and Moine Thrusts: An Electron Tomography Study.

Abstract

In this study we apply electron tomography to characterize 3D dislocation microstructures in two quartz mylonite specimens from the Moine and Main Central Thrusts, both of which accommodated displacements by dislocation creep in the presence of water. Both specimens show dislocation activity with dislocation densities of the order of 3–4 × 1012 m−2 and evidence of recovery from the presence of subgrain boundaries. 〈a〉 slip occurs predominantly on pyramidal and prismatic planes (〈a〉 basal glide is not active). [c] Glide is not significant. On the other hand, we observe a very high level of activation of 〈c + a〉 glide on the 101‾0 $\left\{10\overline{1}0\right\}$, 101‾1 $\left\{10\overline{1}1\right\}$, 112‾n $\left\{11\overline{2}n\right\}$ (n = 1,2) and even 213‾1 $\left\{21\overline{3}1\right\}$ planes. Approximately 60% of all dislocations show evidence of climb with a predominance of mixed climb, a deformation mechanism characterized by dislocations moving in a plane intermediate between the glide and the climb planes. This atypical mode of deformation demonstrates comparable glide and climb efficiency under natural deformation conditions. It promotes dislocation glide in planes not expected for the quartz structure, probably by inhibiting lattice friction. Our quantitative characterization of the microstructure enables us to assess the strain that dislocations can generate. We show that glide systems indicated by the observed dislocations are sufficient to accommodate any arbitrary 3D strain by themselves. Although historically dislocation glide has been regarded as being primarily responsible for producing strain, activation of climb can also directly contribute to the finite strain. On the basis of this characterization, we propose a numerical modeling approach for attempting to characterize the local stress state that gave rise to the observed microstructure. Plain Language Summary: In this study, we characterize dislocation microstructures generated in quartz deformed in natural shear zones by advanced transmission electron microscopy to better understand the geological deformation conditions and the response of quartz to applied stresses. We characterize the crystal defects responsible for deformation of the mineral and their three‐dimensional geometries. We highlight an original deformation mechanism activated at very low natural strain rates that involves simultaneous dislocation glide and climb outside of their crystallographic glide planes. Abstract

Progressive deformation of upper mantle rocks via dislocation creep causes their constituent crystals to take on a non-random orientation distribution (crystallographic preferred orientation or CPO) whose observable signatures include shear-wave splitting and azimuthal dependence of surface wave speeds. Comparison of these signatures with mantle flow models thus allows mantle dynamics to be unraveled on global and regional scales. However, existing self-consistent models of CPO evolution are computationally expensive when used in 3-D and/or time-dependent convection models. Here we propose a new method, called ANPAR, which is based on an analytical parameterisation of the crystallographic spin predicted by the second-order (SO) self-consistent theory. Our parameterisation runs approximately 2-3 x 10^4 times faster than the SO model and fits its predictions for CPO and crystallographic spin with a variance reduction > 99%. We illustrate the ANPAR model predictions for three uniform deformations (uniaxial compression, pure shear, simple shear) and for a corner-flow model of a spreading ridge., Comment: Submitted to GJI December 2014

Abstract

The load transfer induced by the elastic and plastic phase anisotropies of a Ti–10V–2Fe–3Al titanium alloy is studied. The microstructure consists in α nodules embedded in elongated β grains. EBSD performed on the alloy shows no crystallographic texture neither for α nor β phase. Tensile tests along the elongation direction, at a strain rate of 2 x 10-3 s-1 give a yield stress of 830 MPa with 13% ductility. Simulations based on an advanced two-phase polycrystalline elasto-viscoplastic self-consistent (EVPSC) model predict that the β phase first plastifies with a sequential onset of plasticity starting from oriented β grains, then and finally oriented β grains. This leads to a strong load transfer from the β grains to the α nodules whose average behavior remains elastic up to high stresses (~940 MPa). However, additional simulations considering exclusively β grains of specific orientation show that the behavior of α nodules is strongly dependent on the β texture in which they are embedded. Especially, in β grains, which plastify the latest, the model predicts the onset of plasticity in favorably orientated α nodules. Moreover, the orientation spread within the β grains can modify the average plastic behavior of α phase. In future, these results will be compared to data obtained from in-situ High Energy XRD and SEM/EBSD experiments.

Abstract

INTRODUCTION: DLL3, an atypical Notch ligand, is expressed in SCLC tumors but is not detectable in normal adult tissues. Rovalpituzumab tesirine (Rova-T) is an antibody-drug conjugate containing a DLL3-targeting antibody tethered to a cytotoxic agent pyrrolobenzodiazepine by means of a protease-cleavable linker. The efficacy and safety of Rova-T compared with topotecan as second-line therapy in patients with SCLC expressing high levels of DLL3 (DLL3-high) was evaluated. METHODS: The TAHOE study was an open-label, two-to-one randomized, phase 3 study comparing Rova-T with topotecan as second-line therapy in DLL3-high advanced or metastatic SCLC. Rova-T (0.3 mg/kg) was administered intravenously on day 1 of a 42-day cycle for two cycles, with two additional cycles available to patients who met protocol-defined criteria for continued dosing. Topotecan (1.5 mg/m2) was administered intravenously on days 1 to 5 of a 21-day cycle. The primary end point was overall survival (OS). RESULTS: Patients randomized to Rova-T (n = 296) and topotecan (n = 148) were included in the efficacy analyses. The median age was 64 years, and 77% had the extensive disease at initial diagnosis. The median OS (95% confidence interval) was 6.3 months (5.6-7.3) in the Rova-T arm and 8.6 months (7.7-10.1) in the topotecan arm (hazard ratio, 1.46 [95% confidence interval: 1.17-1.82]). An independent data monitoring committee recommended that enrollment be discontinued because of the shorter OS observed with Rova-T compared with topotecan. Safety profiles for both drugs were consistent with previous reports. CONCLUSIONS: Compared with topotecan, which is the current standard second-line chemotherapy, Rova-T exhibited an inferior OS and higher rates of serosal effusions, photosensitivity reaction, and peripheral edema in patients with SCLC. Abstract

International audience; Seismic anisotropy is a critical material property which allow us to identify structures, deformation behaviour and flow in the Earth’s mantle. Interpreting seismic signals, however, requires extensive knowledge of the underlying physical properties of minerals.Large scale seismic anisotropy can arise from the anisotropy of individual crystals and their alignment due to specific mechanisms. In the upper mantle, strong seismic anistotropy is explained by lattice preferred orientation (LPO) of olivine due to mantle flow. In the mantle transition zone, seismic anisotropy is weaker and there is no consensus on the source of this anisotropy. The 410 km depth discontinuity, which separates these two regions, is attributed to the transformation of olivine into its high pressure polymorph wadsleyite. Experiments show that this transformation can happen either by a martensitic-like mechanism or by a non-martensitic-like mechanism. In the first case, the LPO of the parent olivine will be inherited by the daughter wadsleyite whereas in the second case, the daughter wadsleyite will form with a random orientation. Hence, if this transformation is martensitic-like, it can be a source for seismic anisotropy in the mantle transition zone.Here we present a project in which an experimental study of the olivine-wadsleyite transformation is used to make inferences about the prevalent transformation mechanism at mantle conditions. We rely on laboratory experiments to produce the transformation at the conditions of the 410 km depth discontinuity by compressing either polycrystals or single crystals of olivine in diamond anvils cells at pressures up to 20 GPa and temperatures of 1600-1800 K. Using multigrain crystallography we evaluate the orientation and position of individual grains in the sample. We then use the experimental LPO to model the seismic anisotropy resulting from the transformation. Finally, we simulate different observables as anisotropy or seismic reflections from the Earth’s transition zone and compare our predictions with real seismic data.

Abstract

The viscoplastic behavior of polycrystalline Mg$_2$SiO$_4$ wadsleyite aggregates, a major high pressure phase of the mantle transition zone of the Earth (depth range: 410 -- 520 km), is obtained by properly bridging several scale transition models. At the very fine nanometric scale corresponding to the disloca-tion core structure, the behavior of thermally activated plastic slip is modeled for strain-rates relevant for laboratory experimental conditions, at high pressure and for a wide range of temperatures, based on the Peierls-Nabarro-Galerkin model. Corresponding single slip reference resolved shear stresses and associated constitutive equations are deduced from Orowan's equation in order to describe the average viscoplastic behavior at the grain scale, for the easiest slip systems. These data have been implemented in two grain-polycrystal scale transition models, a mean-field one (the recent Fully-Optimized Second-Order Viscoplastic Self-Consistent scheme of [44]) allowing rapid evaluation of the effective viscosity of polycrystalline aggregates , and a full-field (FFT based [45] [33]) method allowing investigating stress and strain-rate localization in typical microstructures and heterogeneous activation of slip systems within grains.
