1. Development of a Herb-Based Dietary Ingredient with Potential Nootropic Properties: From Bench to Bedside.

Abstract

Cognitive decline is a natural process that occurs during aging. Several natural solutions called nootropics exist that can help mitigate this process. However, in some cases, there is a lack of scientific evidence on their potential mechanisms and efficacy. To this end, a total of nine different herb extracts with potential nootropic activity were evaluated. These herb extracts were tested in human neural stem cell cultures exposed to oxidative stress, where reactive oxygen/nitrogen species, cell survival and trophic factor expression were analyzed. Of these extracts, Salvia officinalis (sage) was observed to possess the most significant effect. Its combination with rutin demonstrated additional benefits than with only sage. A randomized, double-blind placebo-controlled clinical study was then performed, testing the effects of the combination at two different doses for 12 weeks in healthy volunteers aged 50+ with mild cognitive impairment. As a result, the blend significantly improved several characteristics related to memory, particularly memory recall, with a more noticeable effect in the higher dose. Therefore, this study reveals the potential nootropic effect of a blend comprising Salvia officinalis and rutin. 6. Skin and Scalp Health Benefits of a Specific Botanical Extract Blend: Results from a Double-Blind Placebo-Controlled Study in Urban Outdoor Workers.

Abstract

Environmental pollution is increasingly recognized as a significant contributor to skin and scalp damage. Oral supplementation with a specific blend of four standardized botanical extracts (Rosmarinus officinalis, Lippia citriodora, Olea europaea leaf, and Sophora japonica) has been previously demonstrated to enhance skin health in individuals exposed to high environmental stress. Thus, it might represent a convenient strategy to also improve their scalp health aspect, particularly in subjects with sensitive scalps. To support these effects, a 12-week double-blind, randomized, placebo-controlled trial was performed in 66 women working outdoors in urban areas with high particulate matter (PM) levels and taking 250 mg of the botanical blend daily. Product efficacy was measured as follows: skin antioxidant status (FRAP); skin and scalp moisturization (corneometer), transepidermal water loss (tewameter), and oiliness (sebumeter); skin radiance and colour (spectrophotometer), elasticity and firmness (cutometer) and wrinkle depth (image analysis); and scalp clinical evaluation. Despite constant exposure to increased levels of PM, the tested product positively influenced all monitored parameters compared to both baseline and the placebo-treated group, in as early as 4 weeks. At the end of the study, key improvements included increased skin FRAP (21.9%), moisturization (9.5%), radiance (24.9%) and reduced wrinkle depth (−16.5%), dark spot pigmentation (−26.2%), and skin oiliness (−19.3%). For the scalp, moisturization increased (14.1%), TEWL decreased (−13.8%), and sebum content reduced by 16.2%. Additionally, 71% of subjects with sensitive scalps experienced reduced redness. 8. Dietary Supplementation with an Extract of Aloysia citrodora (Lemon verbena) Improves Sleep Quality in Healthy Subjects: A Randomized Double-Blind Controlled Study.

Abstract

Seventy-one healthy subjects with sleep disturbances participated in a randomized, double-blind controlled trial in which dietary supplementation with an extract of Aloysia citrodora (lemon verbena) (n = 33) or placebo (n = 38) was administered for 90 days. There were between-group differences in favor of the experimental group in the visual analogue scale (VAS) for sleep quality (6.5 ± 1.6 vs. 5.5 ± 2.1, p = 0.021) as well as in the overall score (5.8 ± 2.4, p = 0.008) and scores for sleep latency (1.6 ± 1.0 vs. 1.9 ± 0.7, p = 0.027) and sleep efficiency (84.5 ± 12.8 vs. 79.8 ± 13.6, p = 0.023) in the Pittsburgh Sleep Quality Index (PSQI). Sleep-related variables (latency, efficiency, wakefulness after sleep onset, awakenings) assessed by actigraphy also showed better scores in the experimental group (p = 0.001). Plasma nocturnal melatonin levels also increased significantly in the experimental group (199.7 ± 135.3 vs. 174.7 ± 115.4 pg/mL, p = 0.048). Changes in anthropometric parameters and physical activity levels were not found. In summary, a dietary supplement of lemon verbena administered for 3 months was associated with a significant improvement in sleep quality as compared with placebo in a population of healthy subjects with sleep problems. Abstract

Consumer digital technology is rapidly evolving, allowing users to manage their health in a simple, non-invasive manner. However, there are few studies revealing if using digital technology as part of an intervention really has an impact in consumer health compared with traditional strategies. The objective of the current study is to compare two groups (MTB; n = 18, 46.1 ± 10.4 years and MTBAPP; n = 19, 45.3 ± 6.40 years) of overweight, prehypertensive individuals in losing weight and lowering their blood pressure. Both were provided with nutritionist-guided recommendations, a wearable tracking device and a dietary supplement that has previously been proven to help lose body weight and lower blood pressure. In addition, one of the groups (MTBAPP) used a mobile app specifically designed for the intervention. Blood pressure, body composition, triglyceride level, peak expiratory flow, forced expiratory volume in the first second and maximum oxygen volume were measured at different time points. In addition, participants were monitored with an activity bracelet throughout the intervention. As a result, both groups significantly lost body weight, while the group using the app additionally improved blood pressure levels and lowered fat mass. Furthermore, the app users significantly increased the number of daily steps and decreased sedentary time. In conclusion, the addition of a mobile app with daily reminders to follow healthy lifestyle recommendations increased physical activity and overall improved blood pressure and fat mass levels when compared with a group performing the same intervention but in absence of the mobile application.

13. Anxiolytic Effect and Improved Sleep Quality in Individuals Taking Lippia citriodora Extract

Abstract

The current fast-moving, hectic lifestyle has increased the number of individuals worldwide with difficulties in managing stress, which in turn is also affecting their sleep quality. Therefore, the objective of the current study was to assess a natural plant-based dietary supplement comprised of lemon verbena (Lippia citriodora) extract, purified in phenylpropanoids, in alleviating stress and improving quality of sleep. A double-blind, placebo-controlled study was conducted for 8 weeks, followed by a 4-week washout period. Both validated questionnaires and functional tests were performed during the study, whereas questionnaires were used after the washout. As a result, the group taking the lemon verbena extract significantly reduced their perception of stress after 8 weeks, which was corroborated by a significant decrease in cortisol levels. After the washout period, the subjects reported to present even lower stress levels, due to the lasting effect of the ingredient. As for sleep quality, the subjects taking the supplement reported feeling better rested, with a stronger effect observed in women. Sleep tracking using a wearable device revealed that the supplement users improved their times in the deeper stages of sleep, specifically their percentage of time in deep sleep and REM. In conclusion, lemon verbena extract purified in phenylpropanoids is revealed as a natural solution to help individuals to improve their stress and sleep quality.

Abstract

This paper investigates the impact of a polyphenol-enriched supplement comprised of hibiscus (Hibiscus sabdariffa) and lemon verbena (Lippia Citriodora) (brand name Metabolaid®) on gut microbiota composition and functionality, by performing fermentations with human faecal microbiota under gut-simulated conditions. Results indicated that Metabolaid®, in a dose-dependent manner, modulated the gut bacteriome to higher relative abundances of some major genera (>3% of relative abundance) such as Bifidobacterium, Blautia and Faecalibacterium and of other minor genera such as Prevotella and Akkermansia. Metabolaid® also promoted increases in short-chain fatty acids (SCFAs) production, particularly butyric acid. On the other hand, it was confirmed that the compounds present in the Metabolaid® supplement (anthocyanins, verbascoside and hibiscus acid) were rapidly metabolized by the gut microbiota. Overall, these results reveal a certain level of gut microbiota-modulating properties of Metabolaid® that could be implicated, at least partially, in the mechanisms behind the potential beneficial effects of this supplement in obesity.

Abstract

Background: Air pollution exposure is one of the major threats to skin health and accelerates skin ageing mainly through oxidative stress mechanisms. Since it is difficult to minimize skin exposure to air pollutants, especially in urban areas, strategies to protect the skin are needed. Plant phenolic compounds have been found to be effective in attenuating cellular oxidative stress and inflammation induced by different air pollutants and a dietary approach based on these compounds could provide an efficient protection measure. Objective: Here we investigated the efficacy of a commercially available polyphenol-enriched dietary supplement (Zeropollution®) in reducing pollution-induced oxidative stress and in improving different skin parameters related to skin ageing of Caucasian and Asian subjects exposed to air pollution. Zeropollution is composed of four standardized herbal extracts: Olea europaea leaf, Lippia citriodora, Rosmarinus officinalis, and Sophora japonica. Design: A double-blind randomized, parallel group study was carried out on 100 outdoor workers living in a polluted urban European area (Milan) to assess the efficacy of the dietary supplement. The total antioxidant capacity on saliva (FRAP), the oxidative damage on skin (lipoperoxides content), skin moisturization (corneometer), transepidermal water loss (tewameter), skin radiance and colour (spectrophotometer), skin elasticity (cutometer), skin sebum content (sebumeter), and the skin roughness (image analysis) were measured. Results: Both inter-group and intra-group analysis proved that the dietary supplement improved all clinical and biochemical-monitored parameters, in both Caucasian and Asian individuals. Some of the positive effects such as decreased wrinkle depth, increased elasticity and firmness, improved skin moisturization and transepidermal water loss, and reduced dark spots pigmentation were statistically significant as early as 2 weeks of product consumption. Conclusions: The results of the study indicate reduced oxidative stress-induced skin damage in both Asian and Caucasian women living in a polluted urban area. Therefore, the oral intake of this four-plant based supplement could be considered a complementary nutrition strategy to avoid the negative effects of environmental pollution exposure. Abstract

Abstract: The rhizome of the plant Hypoxis rooperi (“African potato”) is known for its traditional and ethnomedical uses in the treatment of benign prostatic hyperplasia and other diseases. We have characterized an extract derived from H. rooperi, isolated its major bioactive compound, hypoxoside, and obtained its aglycone, rooperol, by enzymatic digestion. Absorption, fluorescence emission and bidimensional NMR complete spectral data of these compounds were obtained. The antioxidant capacity of both compounds was fully analyzed through the thiobarbituric acid reactive substances (TBARS) and Trolox equivalent antioxidant capacity (TEAC) assays, and it was compared to catechins and olive biophenolics. Both compounds showed a strong antioxidant capacity, although rooperol exhibited a higher antioxidant activity against lipid peroxidation which correlated to its strong affinity for phospholipid membranes as derived from its extremely high lipid/water partition coefficient (K p =3.4×104). The study of the lipophilic (EtOH) and hydrophilic (water) TEAC values revealed that more hydrophobic compounds, had greater lipophilic TEAC values than hydrophilic ones, probably indicating that lipophilic TEAC assay may be more reliable for these compounds. The H. rooperi extract also showed higher antioxidant efficacy compared to other strong antioxidant herbal extracts, such as olive leaf or green tea. Moreover, neither evidence of acute oral toxicity nor adverse effects were observed when the H. rooperi commercial extract containing 45% hypoxoside was used at a dosage of 2000mg/kg. The results obtained in this work may contribute to understanding the biological activity described for these dicatechols and the African potato extract for food and cosmetic applications. Abstract

Lemon verbena ( Lippia citriodora ) has been used as a food spice, cosmetic, and in traditional medicine formulations to treat asthma and diabetes in South America and Southern Europe. Hibiscus flower ( Hibiscus sabdariffa L.) is used in traditional Chinese medicine in the form of a tea to treat hypertension and inflammation. In the present study, we examined the synergistic effects of a formula of Metabolaid ® (MetA), a combination of lemon verbena and hibiscus-flower extracts, on obesity and its complications in high-fat-diet (HFD)-induced obese mice. The results showed that MetA decreased body weight, white adipose tissue (WAT), and liver weight. Additionally, serum and hepatic lipid profiles, glucose levels, glucose tolerance, and cold-induced thermogenesis were significantly improved. Appetite-regulating hormones adiponectin and leptin were significantly increased and decreased, respectively, while the inflammatory-related factors tumor necrosis factor (TNF)-α and interleukin (IL)-6 were downregulated by MetA. Adipogenesis-activating gene expression was decreased, while increased thermogenesis-inducing genes were upregulated in the WAT, correlating with increased phosphorylation of AMPK and fatty-acid oxidation in the liver. Taken together, these results suggest that MetA decreased obesity and its complications in HFD mice. Therefore, this formula may be a candidate for the prevention and treatment of obesity and its complications.
