The understanding of the halo current properties during disruptions is key to design and operate large scale tokamaks in view of the large thermal and electromagnetic loads that they entail. For the first time, we present a fully self-consistent model for halo current simulations including neutral particles and sheath boundary conditions. The model is used to simulate Vertical Displacement Events (VDEs) occurring in the COMPASS tokamak. Recent COMPASS experiments have shown that the parallel halo current density at the plasma-wall interface is limited by the ion saturation current during VDE-induced disruptions. We show that usual MHD boundary conditions can lead to the violation of this physical limit and we implement this current density limitation through a boundary condition for the electrostatic potential. Sheath boundary conditions for the density, the heat flux, the parallel velocity and a realistic parameter choice (e.g. Spitzer $\eta$ and Spitzer-H\"arm $\chi_\parallel$ values) extend present VDE simulations beyond the state of the art. Experimental measurements of the current density, temperature and heat flux profiles at the COMPASS divertor are compared with the results obtained from axisymmetric simulations. Since the ion saturation current density ($J_{sat}$) is shown to be essential to determine the halo current profile, parametric scans are performed to study its dependence on different quantities such as the plasma resistivity and the particle and heat diffusion coefficients. In this respect, the plasma resistivity in the halo region broadens significantly the $J_{sat}$ profile, increasing the halo width at a similar total halo current., Comment: Submitted to Plasma Physics and Controlled Fusion via the special issue of the Joint Varenna-Lausanne international workshop (October 12-16, 2020)

- Abstract
Within the 9th European Framework programme, since 2021 EUROfusion is operating five tokamaks under the auspices of a single Task Force called ‘Tokamak Exploitation’. The goal is to benefit from the complementary capabilities of each machine in a coordinated way and help in developing a scientific output scalable to future largre machines. The programme of this Task Force ensures that ASDEX Upgrade, MAST-U, TCV, WEST and JET (since 2022) work together to achieve the objectives of Missions 1 and 2 of the EUROfusion Roadmap: i) demonstrate plasma scenarios that increase the success margin of ITER and satisfy the requirements of DEMO and, ii) demonstrate an integrated approach that can handle the large power leaving ITER and DEMO plasmas. The Tokamak Exploitation task force has therefore organized experiments on these two missions with the goal to strengthen the physics and operational basis for the ITER baseline scenario and for exploiting the recent plasma exhaust enhancements in all four devices (PEX: Plasma EXhaust) for exploring the solution for handling heat and particle exhaust in ITER and develop the conceptual solutions for DEMO. The ITER Baseline scenario has been developed in a similar way in ASDEX Upgrade, TCV and JET. Key risks for ITER such as disruptions and run-aways have been also investigated in TCV, ASDEX Upgrade and JET. Experiments have explored successfully different divertor configurations (standard, super-X, snowflakes) in MAST-U and TCV and studied tungsten melting in WEST and ASDEX Upgrade. The input from the smaller devices to JET has also been proven successful to set-up novel control schemes on disruption avoidance and detachment.

- Abstract
The tokamak à configuration variable (TCV) continues to leverage its unique shaping capabilities, flexible heating systems and modern control system to address critical issues in preparation for ITER and a fusion power plant. For the 2019-20 campaign its configurational flexibility has been enhanced with the installation of removable divertor gas baffles, its diagnostic capabilities with an extensive set of upgrades and its heating systems with new dual frequency gyrotrons. The gas baffles reduce coupling between the divertor and the main chamber and allow for detailed investigations on the role of fuelling in general and, together with upgraded boundary diagnostics, test divertor and edge models in particular. The increased heating capabilities broaden the operational regime to include Te/Ti ∼1 and have stimulated refocussing studies from L-mode to H-mode across a range of research topics. ITER baseline parameters were reached in type-I ELMy H-modes and alternative regimes with 'small' (or no) ELMs explored. Most prominently, negative triangularity was investigated in detail and confirmed as an attractive scenario with H-mode level core confinement but an L-mode edge. Emphasis was also placed on control, where an increased number of observers, actuators and control solutions became available and are now integrated into a generic control framework as will be needed in future devices. The quantity and quality of results of the 2019-20 TCV campaign are a testament to its successful integration within the European research effort alongside a vibrant domestic programme and international collaborations.

- Abstract
Analysis of the divertor edge localized mode (ELM) electron temperature at a uniquely high temporal resolution (10(-5) s) was reported at the JET tokamak (Guillemaut et al 2018 Nucl. Fusion 58 066006). By collecting divertor probe data obtained during many dozens of ELMs, the conditional-average (CAV) technique yields surprisingly low peak electron temperatures, far below the pedestal ones (70%-99% reduction!) which we, however, question. This result was interpreted through the collisional free-streaming kinetic model of ELMs, by a transfer of most of the electron energy to ions, implying a high tungsten sputtering for unmitigated ELMs in future fusion devices like ITER. Recently, direct microsecond temperature measurements on the COMPASS tokamak, however, showed that the electron temperature peak of ELM filaments measured in the divertor is reduced by less than a third with respect to the pedestal one. This was further confirmed by a dedicated 1D particle-in-cell (PIC) simulation and tends to prove that the pedestal electrons can transfer only their parallel energy to ions (due to low collisionality), thus less than a third, as is predicted by the collisionless free-streaming model. This finding strongly contradicts the JET observations. We have therefore compared the CAV to the direct (microsecond) ball-pen and Langmuir probes measurements in COMPASS and found very good agreement between them. Revisiting the aforementioned JET CAV analysis indeed shows that the electron temperatures are much higher than previously reported, close to those predicted by the PIC simulation, and thus the ion energy seems to not significantly increase in the scrape-off layer., QC 20230425

- Abstract
Electron cyclotron wall condition (ECWC) discharges are characterised in ASDEX Upgrade with full tungsten plasma facing components and X2 polarised waves launched from the equatorial ports, relevant to ECWC conditions in ITER Pre-Fusion Power Operation phase 1. The characterisation of the deuterium plasmas is based on experimental inputs such as electron density measurements, in-vessel pressure measurements, poloidal field maps obtained from the measured coil currents, as well as advanced tomographic methods on camera images filtered at the hydrogen Balmer lines. TOMATOR-1D simulations and collective Thomson scattering radiometer spectra complement the findings. The cold, high density and partially ionized toroidal plasmas show significant levels of stray radiation. The measured radiation includes waves at half of the gyrotron frequency suggesting the occurrence of parametric decay instabilities at 2nd harmonic upper hybrid resonance that locates at the low field side of the resonance. A displacement of the plasma emission along the resonance layer is observed at higher discharge power in discharges with a vertical poloidal magnetic field only. By optimizing the poloidal field pattern, along with the location of the electron cyclotron heating (ECH) resonance, the strongest surface interaction regions for the charged particles can be controlled. Directing plasma flux to inner wall surfaces, and same for the inner divertor apron, is found less effective in ASDEX Upgrade due to magnetic mirror effects and outward convective flows. Modeling however predicts the presence of an intense and uniform flux of low energy atoms produced at the ECH absorption layer that may be effective for conditioning the high field side surfaces after use of the disruption mitigation system.

- Abstract
The tokamak a configuration variable (TCV) continues to leverage its unique shaping capabilities, flexible heating systems and modern control system to address critical issues in preparation for ITER and a fusion power plant. For the 2019-20 campaign its configurational flexibility has been enhanced with the installation of removable divertor gas baffles, its diagnostic capabilities with an extensive set of upgrades and its heating systems with new dual frequency gyrotrons. The gas baffles reduce coupling between the divertor and the main chamber and allow for detailed investigations on the role of fuelling in general and, together with upgraded boundary diagnostics, test divertor and edge models in particular. The increased heating capabilities broaden the operational regime to include T (e)/T (i) similar to 1 and have stimulated refocussing studies from L-mode to H-mode across a range of research topics. ITER baseline parameters were reached in type-I ELMy H-modes and alternative regimes with `small' (or no) ELMs explored. Most prominently, negative triangularity was investigated in detail and confirmed as an attractive scenario with H-mode level core confinement but an L-mode edge. Emphasis was also placed on control, where an increased number of observers, actuators and control solutions became available and are now integrated into a generic control framework as will be needed in future devices. The quantity and quality of results of the 2019-20 TCV campaign are a testament to its successful integration within the European research effort alongside a vibrant domestic programme and international collaborations.

- Abstract
With the aim to discover new antituberculous molecules, three novel series of 23 hydroxamic acids, 13 hydrazides, and 9O-alkyl/O-acyl protected hydroxamic acid derivatives have been synthesized, and fully characterized by spectral 1H NMR, 13C NMR, HRMS) analysis. These compounds were further biologically screened for their in vitro antibacterial activities against three pathogenic mycobacteria - M. abscessus S and R, M. marinum, and M. tuberculosis – as well as for their toxicity towards murine macrophages by the resazurin microtiter assay (REMA). Among the 45 derivatives, 17 compounds (3 hydroxamic acids, 9 hydrazides, and 5O-alkyl/O-acyl protected hydroxamic acids) were nontoxic against murine macrophages. When tested for their antibacterial activity, hydroxamic acid 9 h was found to be the most potent inhibitor against M. abscessus S and R only. Regarding hydrazide series, only 7h was active against M. abscessus R, M. marinum and M. tuberculosis; while the O-acyl protected hydroxamic acid derivatives 14d and 15d displayed promising antibacterial activity against both M. marinum and M. tuberculosis. Since such hydroxamic- and hydrazide-chelating groups have been reported to impair the activity of the peptide deformylase, in silico molecular docking studies in M. tuberculosis peptide deformylase enzyme active site were further performed with 7h in order to predict the possible interaction mode and binding energy of this molecule at the molecular level. © 2022 Elsevier Ltd

- Abstract
The tokamak a configuration variable (TCV) continues to leverage its unique shaping capabilities, flexible heating systems and modern control system to address critical issues in preparation for ITER and a fusion power plant. For the 2019-20 campaign its configurational flexibility has been enhanced with the installation of removable divertor gas baffles, its diagnostic capabilities with an extensive set of upgrades and its heating systems with new dual frequency gyrotrons. The gas baffles reduce coupling between the divertor and the main chamber and allow for detailed investigations on the role of fuelling in general and, together with upgraded boundary diagnostics, test divertor and edge models in particular. The increased heating capabilities broaden the operational regime to include T (e)/T (i) similar to 1 and have stimulated refocussing studies from L-mode to H-mode across a range of research topics. ITER baseline parameters were reached in type-I ELMy H-modes and alternative regimes with `small' (or no) ELMs explored. Most prominently, negative triangularity was investigated in detail and confirmed as an attractive scenario with H-mode level core confinement but an L-mode edge. Emphasis was also placed on control, where an increased number of observers, actuators and control solutions became available and are now integrated into a generic control framework as will be needed in future devices. The quantity and quality of results of the 2019-20 TCV campaign are a testament to its successful integration within the European research effort alongside a vibrant domestic programme and international collaborations.

Construction of the JT-60SA tokamak was completed on schedule in March 2020. Manufacture and assembly of all the main tokamak components satisfied technical requirements, including dimensional accuracy and functional performances. Development of the plasma heating systems and diagnostics have also progressed, including the demonstration of the favourable electron cyclotron range of frequency (ECRF) transmission at multiple frequencies and the achievement of long sustainment of a high-energy intense negative ion beam. Development of all the tokamak operation control systems has been completed, together with an improved plasma equilibrium control scheme suitable for superconducting tokamaks including ITER. For preparation of the tokamak operation, plasma discharge scenarios have been established using this advanced equilibrium controller. Individual commissioning of the cryogenic system and the power supply system confirmed that these systems satisfy design requirements including operational schemes contributing directly to ITER, such as active control of heat load fluctuation of the cryoplant, which is essential for dynamic operation in superconducting tokamaks. The integrated commissioning (IC) is started by vacuum pumping of the vacuum vessel and cryostat, and then moved to cool-down of the tokamak and coil excitation tests. Transition to the super-conducting state was confirmed for all the TF, EF and CS coils. The TF coil current successfully reached 25.7 kA, which is the nominal operating current of the TF coil. For this nominal toroidal field of 2.25 T, ECRF was applied and an ECRF plasma was created. The IC was, however, suspended by an incident of over current of one of the superconducting equilibrium field coil and He leakage caused by insufficient voltage holding capability at a terminal joint of the coil. The unique importance of JT-60SA for H-mode and high-β steady-state plasma research has been confirmed using advanced integrated modellings. These experiences of assembly, IC and plasma operation of JT-60SA contribute to ITER risk mitigation and efficient implementation of ITER operation.

The COMPASS tokamak provided unique conditions to study Runaway Electrons (RE) physics in the ITER-like geometry. Twelve experimental campaigns were dedicated to identify limiting thresholds for safe termination or efficient suppression of RE beam [1] and to thoroughly explore the possible advantages of alternative mitigation strategies. High flexibility and reproducibility of COMPASS RE scenarios allowed the execution of extensive scans devoted, for example, to decoupling effects of individual factors acting on RE dynamics. The studied factors include resonant magnetic field perturbations (RMP), radiation drag, instabilities, and the newly developed unique RE radial position feedback [2]. The main principle of the RE radial feedback was successfully used as a fast and robust method to obtain information about the RE energy in parallel to HXR radiation and unique calorimetry measurements. These methods are used to show the impact of different mitigation materials, externally triggered RMPs and the feedback itself on the RE beam energy and its current decay rate. The experimentally observed effects of RMPs are supported by a full orbit RE tracer developed at COMPASS, MARS-F [3] and ORBIT [4] codes. First tests with an active launch of low power 0.5 GHz waves into a RE populated low density plasma were performed. Preliminary results from measurements of electromagnetic waves in the 0.1-1.5 GHz range during this scenario are presented.

In the last runaway electron campaigns that were performed in 2020, broad range of experi- mental results were obtained in several areas of runaway electron physics. Special diagnostic tools and methods were applied including HXR spectrometry, limiter calorimetry, estimates of energy based on runaway electron equilibrium, synchrotron radiation diagnostics (REIS2), matrix SXR detectors and passive and active high frequency antennas. The measurements of RE beam average energy based on the relativistic pressure and equilibrium properties seem to present a fast and efficient method that can be compared well with the methods based on the hard X-ray bremsstralung and synchrotron radiation diagnostics. The comparison of RE beams generated by injections of different gas species and amounts shows that the least dangerous terminations of the RE beam can be achieved via Ne injection or impurity injection followed by a D2 secondary injection. The D2 secondary injection beneficial effects tend to be consistent with DIII-D and JET results. The RMP experiments have also provided many interesting results including significant reduction of HXRs and impact energy of the RE beam with application of RMPs during the RE beam generation. Measurements with high frequency antennas, which will be briefly introduced, also showed presence of wide range of high frequency kinetic instabilities including the whistler-like and various chirping phenomena. Last but not least the room temperature solid state pellet injector has been implemented and used for RE beam generation as well as for enhancing the RE dissipation.

Runaway electrons (RE) still present a serious for the tokamak operation. Furthermore, the physics phenomenon itself is complicated and extrapolation to the large devices is difficult. In order to understand the RE phenomenon, it is necessary to optimise the diagnostics of the runaway electrons. Some of the properties of RE can be measured using the standard tokamak diagnostics directly (e.g. plasma or runaway current measurement by magnetic diagnostics). In some cases, it is necessary to improve the understanding of the effect of the RE on the diagnos- tics (e.g. electron cyclotron emission measurement by radiometer) by modelling and analytical approximations. However, often it is necessary to introduce a completely new diagnostics, that can be used specifically for RE (e.g. local loss measurement using a Cerenkov detector). In this contribution a new application of magnetic measurements for estimation of the RE energy is dis- cussed for two European machines with extensive RE experimental program.

Mycobacteria share with other actinomycetes the ability to produce large quantities of triacylglycerol (TAG), which accumulate as intracytoplasmic lipid inclusions (ILI) also known as lipid droplets (LD). Mycobacterium tuberculosis (M. tb), the etiologic agent of tuberculosis, acquires fatty acids from the human host which are utilized to synthesize TAG, subsequently stored in the form of ILI to meet the carbon and nutrient requirements of the bacterium during long periods of persistence. However, environmental factors governing mycobacterial ILI formation and degradation remain poorly understood. Herein, we demonstrated that in the absence of host cells, carbon excess and nitrogen starvation promote TAG accumulation in the form of ILI in M. smegmatis and M. abscessus, used as surrogate species of M. tb. Based on these findings, we developed a simple and reversible in vitro model to regulate ILI biosynthesis and hydrolysis in mycobacteria. We also showed that TAG formation is tgs1 dependent and that lipolytic enzymes mediate TAG breakdown. Moreover, we confirmed that the nitrogen-deprived and ILI-rich phenotype was associated with an increased tolerance towards several drugs used for treating mycobacterial infections. Importantly, we showed that the presence of ILI substantially enhanced the bacterial burden and granuloma abundance in zebrafish embryos infected with lipid-rich M. abscessus as compared to embryos infected with lipid-poor M. abscessus, suggesting that ILI are actively contributing to mycobacterial virulence and pathogenesis.

In the quest for new antibacterial agents, a series of novel long- and medium-chain mono- and disubstituted β-lactones was developed. Their activity against three pathogenic mycobacteria—M. abscessus, M. marinum, and M. tuberculosis—was assessed by the resazurin microtiter assay (REMA). Among the 16 β-lactones synthesized, only 3-hexadecyloxetan-2-one (VM005) exhibited promising activity against M. abscessus, whereas most of the β-lactones showed interesting activities against M. marinum, similar to that of the classical antibiotic, isoniazid. Regarding M. tuberculosis, six compounds were found to be active against this mycobacterium, with β-lactone VM008 [trans-(Z)-3-(hexadec-7-en-1-yl)-4-propyloxetan-2-one] being the best growth inhibitor. The promising antibacterial activities of the best compounds in this series suggest that these molecules may serve as leads for the development of much more efficient antimycobacterial agents. © 2019 Wiley-VCH Verlag GmbH & Co. International audience; This paper summarizes the status of the COMPASS tokamak, its comprehensive diagnostic equipment and plasma scenarios as a baseline for the future studies. The former COMPASS-D tokamak was in operation at UKAEA Culham, UK in 1992-2002. Later, the device was transferred to the Institute of Plasma Physics of the Academy of Sciences of the Czech Republic (IPP AS CR), where it was installed during 2006-2011. Since 2012 the device has been in a full operation with Type-I and Type-III ELMy H-modes as a base scenario. This enables together with the ITER-like plasma shape and flexible NBI heating system (two injectors enabling co- or balanced injection) to perform ITER relevant studies in different parameter range to the other tokamaks (ASDEX-Upgrade, DIII-D, JET) and to contribute to the ITER scallings. In addition to the description of the device, current status and the main diagnostic equipment, the paper focuses on the characterization of the Ohmic as well as NBI-assisted H-modes. Moreover, Edge Localized Modes (ELMs) are categorized based on their frequency dependence on power density flowing across separatrix. The filamentary structure of ELMs is studied and the parallel heat flux in individual filaments is measured by probes on the outer mid-plane and in the divertor. The measurements are supported by observation of ELM and inter-ELM filaments by an ultra-fast camera.
