To better understand and observe craniofacial growth, researchers carry out longitudinal cephalometric studies on animals. The authors constructed a cephalostat specially designed to study craniofacial growth in rats. They describe and validate a cephalometric technique using this cephalostat. They carried out cephalometric radiography and analysis of 62 Wistar rats. By using this method, the authors were able to take repeated, reproducible intracranial, intramandibular and craniomandibular measurements of the rats. The proposed method is a useful tool for studying the craniofacial growth process.

Background and Objective: There is a strong relationship between neuromuscular activity and facial skeletal morphogenesis. The systemic use of anabolic steroid accelerates craniofacial growth and induces high functional activity of rats' masticatory muscles. The present study examines the effect of an anabolic steroid locally applied in the masseter muscle of growing rats with the purpose of verifying possible craniofacial changes with minimum drug side effects, using cephalometric analysis., Materials and Methods: Forty-three 3-week-old Wistar rats were evaluated and divided into two groups: (1) NAN group (local bilateral masseter injection of nandrolone decanoate - 0.05 microg/3 microl/bi-weekly) and (2) CON group (local bilateral masseter injection of sterile oil - 3 microl). Tukey and Wilcoxon tests were used., Results: In NAN group the cephalometric measures showed horizontalized facial growth (reduce of FMA angle -p<0.01), increase of posterior facial height, decrease of anterior facial height, reduction of gonial angle (p<0.05), increase of mandibular height (p<0.01). There was no variation in the length of the mandible, in the skull and animals' weight gain, thus demonstrating that no important systemic effect occurred., Conclusion: The local injection of nandrolone in the masseter muscle of rats was capable to alter the growth direction and the morphology of the craniofacial complex in Wistar rats.

Cardiovascular effects of felypressin (FEL) were studied in Wistar rats. Heart rate and mean arterial pressure measurements were taken in awake rats treated with vasopressin (AVP), FEL, or epinephrine (EPI). Each group received either an intravenous (IV) or an intracerebroventricular V1 receptor antagonist, saline, area postrema removal, or sham surgery. Analysis of variance and Student-Newman-Keuls (P < .05) were applied. Felypressin and AVP induced a pressor effect, and bradycardia was inhibited by IV V1 antagonist. Intracerebroventricular V1 antagonist and area postrema removal enhanced their pressor effects. Epinephrine induced a higher pressor effect and a similar bradycardia that was not affected by the treatments. It was concluded that FEL depends on V1 receptors to induce pressor and bradycardic effects, and that it produces a high relationship between bradycardia and mean arterial pressure variation depending on area postrema and central V1 receptors. These effects are potentially less harmful to the cardiovascular system than the effects of EPI.

Central cholinergic activation by pilocarpine induces salivation dependent on the integrity of forebrain areas. The present work investigates the autonomic mediation of this salivation. Pilocarpine (500 nmol/rat) was injected into the lateral ventricle (LV) of tribromoethanol-anesthetized adult male rats. Preweighed cotton balls were inserted into the oral cavity and weighed again 7 min later. Alpha-adrenoceptor antagonists (3-50 micromol/kg) prazosin (alpha1), yohimbine (alpha2) or propranolol (beta) injected intraperitoneally (i.p.) produced, 80%, 20% and 0% inhibition respectively of the LV pilocarpine-induced salivation. Intracerebroventricular injections (160 nmol) of the antagonists did not alter the effects of pilocarpine injected into the LV. Bilateral section of chorda tympani nerve or bilateral sympathetic cervical ganglionectomy produced 0% and 40% inhibition of pilocarpine-induced salivation, respectively. Ganglionectomy did not alter salivation induced by i.p. injection of pilocarpine (4 micromol/kg). The results indicate that there is a large sympathetic contribution to the salivation induced by central cholinergic activation.
