1. In-silico analysis of mt-CO1 gene of Taenia hydatigena sheep isolates

Abstract

Taenia hydatigena is a tapeworm that affects herbivores in different regions of the world. Cysticercus tenuicollis (larvae of T. hydatigena), is prevalent in ruminants and pigs. In the current study, phylogenetic analysis of the published mt-CO1 gene of C. tenuicollis sheep isolates was analyzed using in-silico method and vertical and horizontal transmission at the global level by using a meta-analysis approach. A total of 82 mt-CO1 nucleotide sequences (339 bp) of C. tenuicollis sheep isolates from the NCBI database (Italy -Sardinia-, Iran, Palestine, Iraq, Finland, India and China) were used to investigate haplotype and genetic relationships. Tajima’s D (-2,2984) value was negative for the mt-CO1 sequences signifying the population expansion and/or purifying selection. The highly negative Fu’s Fs (-60,528) values determined for the sequences reflecting the existence of uncommon haplotypes. The mt-CO1 of C. tenuicollis haplotype network had 47 haplotypes arranged within a star-like configuration with a main haplotype, which encompassed 25.6 % of the total isolates. In the mt-CO1 haplotype network analyzed, there were 80.5 % unique single haplotype and highest ratio was observed in C. tenuicollis from sheep originating from Iran, followed by Sardinia, Palestine and Finland. If the current condition continues, genetic differences in T. hydatigena will be able to rise, and possible new strains and/or genotypes that may influence the host adaptation and life cycle of the parasite may emerge.

Abstract

Background Headaches are frequent neurological disorders that are yet to be unveiled and treated comprehensively worldwide. Bearing in mind that the distribution of headache subtypes in neurology clinics (NC) is essential for planning appropriate diagnostic and therapeutic approaches, the primary goals of this multi-centric study are to carry out inter-regional comparisons by using current diagnostic criteria with evaluations of neurologists to delineate headache burden. Methods A cross-sectional study between April 1 and May 16, 2022 was conducted with the participation of 13 countries from the Middle East, Asia, and Africa. Patients were included in the study on a specific day each week during five consecutive weeks. All volunteers over the age of 18 and whose primary cause for admission was headache were examined. The patients admitted to NC or referred from emergency services/other services were evaluated by neurologists by means of the International Classification of Headache Disorders (ICHD-3) criteria. Results Among the 13,794 patients encountered in NC, headache was the primary complaint in 30.04%. The headache patients’ mean age was 42.85 ± 14.89 (18–95 years), and 74.3% were female. According to the ICHD-3 criteria, 86.7% of the main group had primary headache disorders, 33.5% had secondary headaches, 4% had painful cranial neuropathies along with other facial and headaches, and 5.2% had headaches included in the appendix part showing some overlapping conditions. While the most common primary headache was migraine without aura (36.8%), the most common secondary headache was medication-overuse headache (MOH) (9.8%). Headaches attributed to COVID-19, its secondary complications, or vaccines continue to occur at rates of 1.2%-3.5% in current neurology practice. Pain severity was significantly lower in Ivory Coast and Sudan than in Türkiye, Turkish Republic of Northern Cyprus, Iran, Egypt, Senegal, Tatarstan, and Azerbaijan (p Conclusions The study showed that migraine is still the most common motive for admissions to NC in different regions. Item does not contain fulltext, PURPOSE: MYC gene rearrangements in diffuse large B-cell lymphoma (DLBCL) patients are associated with poor prognosis. Our aim was to compare patterns of 2[(18)F]fluoro-2-deoxy-D-glucose positron emission tomography computed tomography (PET/CT) response in MYC + and MYC- DLBCL patients. METHODS: Interim PET/CT (I-PET) and end of treatment PET/CT (EoT-PET) scans of 81 MYC + and 129 MYC- DLBCL patients from 2 HOVON trials were reviewed using the Deauville 5-point scale (DS). DS1-3 was regarded as negative and DS4-5 as positive. Standardized uptake values (SUV) and metabolic tumor volume (MTV) were quantified at baseline, I-PET, and EoT-PET. Negative (NPV) and positive predictive values (PPV) were calculated using 2-year overall survival. RESULTS: MYC + DLBCL patients had significantly more positive EoT-PET scans than MYC- patients (32.5 vs 15.7%, p = 0.004). I-PET positivity rates were comparable (28.8 vs 23.8%). In MYC + patients 23.2% of the I-PET negative patients converted to positive at EoT-PET, vs only 2% for the MYC- patients (p = 0.002). Nine (34.6%) MYC + DLBCL showed initially uninvolved localizations at EoT-PET, compared to one (5.3%) MYC- patient. A total of 80.8% of EoT-PET positive MYC + patients showed both increased lesional SUV and MTV compared to I-PET. In MYC- patients, 31.6% showed increased SUV and 42.1% showed increased MTV. NPV of I-PET and EoT-PET was high for both MYC subgroups (81.8-94.1%). PPV was highest at EoT-PET for MYC + patients (61.5%). CONCLUSION: MYC + DLBCL patients demonstrate aberrant PET response patterns compared to MYC- patients with more frequent progression during treatment after I-PET negative assessment and new lesions at sites that were not initially involved. Abstract

Saffron is an important medicine, spice, dye, and cosmetic plant, and weeds limit its production and increase the related costs of management. This study aimed to determine effective control methods against weeds in saffron cultivation and find the effects of weed control on crown development, stigma yield, and daughter corm yield and quality. The study was established in the production periods of 2019-2021, at Hatay Olive Research Institute, Hassa Station, Turkey, based on completely randomized blocks design with 14 treatments and 3 replications. Results indicated that the highest effect (100%) on weeds was recorded for U5 (pine sawdust+benfluralin) and U6 (textile mulch) applications and the lowest effect was obtained from U13 (2,4-D amine) application at the first and second year of the study. The best quality criteria of saffron were achieved in U6 and U4 (pine sawdust) applications for corm production, and U6, U5, and U4 applications for quality daughter corm production. Abstract

This study concluded the results of a research on the features of cement based permeation grout, based on some important grout parameters, such as the rheological properties (yield stress and viscosity), coefficient of permeability to grout (kG) and the inject ability of cement grout (N and Nc assessment), which govern the performance of cement based permeation grouting in porous media. Due to the limited knowledge of these important grout parameters and other influencing factors (filtration pressure, rate and time of injection and the grout volume) used in the field work, the application of cement based permeation grouting is still largely a trial and error process in the current practice, especially in the local construction industry. It is seen possible to use simple formulas in order to select the injection parameters and to evaluate their inter-relationship, as well as to optimize injection spacing and times with respect to injection source dimensions and in-situ permeability. The validity of spherical and cylindrical flow model was not verified by any past research works covered in the literature review. Therefore, a theoretical investigation including grout flow models and significant grout parameters for the design of permeation grouting was conducted in this study. This two grout flow models were applied for three grout mixes prepared for w/c=0.75, w/c=1.00 and w/c=1.25 in this study. The relations between injection times, radius, pump pressure and flow rate for both flow models were investigated and the results were presented. Furthermore, in order to investigate these two flow model, some rheological properties of the grout mixes, particle size distribution of the cement used in this study and some geotechnical properties of the sand used in this work were defined and presented. © 2019 Techno-Press, Ltd.

22. Does early enteral feeding of very low birth weight infants increase the risk of necrotizing enterocolitis?

Abstract

Background/Objective: In this retrospective study, we intended to test whether early enteral feeding (EEF) of very low birth weight (VLBW) preterm babies increases the risk of necrotizing enterocolitis (NEC) or not. Subjects and Methods: Overall, 297 VLBW preterm babies admitted to the neonatal intensive care unit (NICU) between April 2003 and April 2006 were included. The study consisted of two periods: the first period was between April 2003 and October 2004, when babies were not fed enterally until they were extubated (167 preterm VLBWs). The second period was between November 2004 and April 2006, when babies were fed even when they were intubated, starting preferably on the first day of life (130 preterm VLBWs). Criteria for withholding enteral feeding in both periods were hypotension necessitating vasopressor agent use, abdominal distention, abdominal tenderness and suspected or proven NEC. Possible risk factors for NEC were also recorded. Results: The overall incidence of NEC in VLBW preterm babies was 6.7% and did not differ between the two study periods: 7.2% in the late and 6.2% in the EEF regimens. Abstract

ERRATIONS; RECEPTORS; RISK; CNS Autism Spectrum Disorder (ASD) is a complex neurodevelopmental disorder characterized by impairments in communication and social interaction as well as restricted interests and repetitive behaviors. The pathogenesis of ASD is not completely understood, but a growing body of research has demonstrated that the immune response may be a contributing factor in the etiology and/or ontogeny of ASD. The aim of this study was to determine the expression levels of IL-1 beta, IL-1 alpha, IL-4, IL-6, IL-17, TNF-alpha and TGF-beta in peripheral blood mononuclear cells of children with ASD and healthy controls in order to determine the contributions of cytokines to ASD. Within the study timeframe, 195 children with ASDs (80.5% male) and 162 controls (73.6% male) were enrolled. Most children with ASD had a comorbid disorder (n = 114, 58.5%), with the most common diagnoses as Intellectual Developmental Disorder (IDD, n = 64, 32.8%) and ADHD (n = 64, 32.8%). The majority of children with ASD had severe autistic symptoms as evaluated via Childhood Autism Rating Scale (CARS, n = 130, 64.6%). The mean CARS score in the ASD sample was 40.8 (S.D. = 7.6). The patients with ASD were found to have significantly higher levels of IL-6 (p < 0.001) and significantly lower levels of IL-17 (p < 0.05, all Bonferroni corrected). Treatment tended to affect IL-4 levels. Lastly, discriminant function analysis (DFA) revealed that a combination of IL-6, IL-17 and IL-1 alpha correctly classified 56.6% of cases. Despite extensive immunological evidence suggesting immune system aberrations, further research is required to clarify the relationship between immune profiles and ASD symptoms. Abstract

Immune dysregulation may be important in the etiology of obsessive-compulsive and related disordersandbody-focusedrepetitivebehaviors, such as Trichotillomania (TTM). The role of inflammation and inflammatory markers in TTM has received relatively little attention. This study was aimed to determine the expression levels of inflammatory markers (i.e. IL-1 beta, IL-1 alpha, IL-4, IL-6, IL-17, TNF-alpha and TGF-5) in peripheral blood mononuclear cells of children with TTM and healthy controls and to evaluate their association with clinical variables. Seventy-seven patients with TTM and 107 healthy controls were enrolled in the study. Peripheral blood was collected in standardized conditions. The mean age of patients and controls did not differ significantly (10.8 +/- 4.4 and 12.0 +/- 3.2 years; respectively). The majority of patients with TTM and controls were females (n = 55, 71.4 % and n = 55, 51.4 %; respectively); with a greater preponderance of females among TTM. Patients with TTM had significantly elevated expression levels of TNF-alpha, IL-6 and IL-17 compared to controls. However, the expression level of IL-4 was significantly reduced in TTM patients compared to controls. Accordingly, we found a proinflammatory state in TTM and those findings may suggest novel treatment options for TTM and further, crossdisciplinary studies focusing on neuro- inflammation in TTM conducted on larger samples are needed. Item does not contain fulltext, Aim: Timing of radium-223 (Ra-223) in metastatic castration-resistant prostate cancer (mCRPC) remains challenging due to alternative options and short window of opportunity. Methods: Ra-223 treated patients in the CAPRI-registry were included. Outcomes were evaluated based on treatment line of Ra-223. Results: Out of 285 patients, 49% received Ra-223 in line >/=3. 51% completed six Ra-223 injections and 34% had a symptomatic skeletal event after first Ra-223 without differences between subgroups. After correction of known prognostic factors Ra-223 in line >/=3 (HR: 3.267; 95% CI: 1.689-6.317; p < 0.01) remained associated with worse OS. Conclusion: In the Netherlands, Ra-223 was mainly started as second or third mCRPC-treatment in 2014-2018. Later timing of Ra-223 did affect OS, but not treatment completion and occurrence of symptomatic skeletal events.

Contains fulltext : 229836.pdf (Publisher’s version ) (Open Access), The ALSYMPCA study established a 3.6 month Overall Survival (OS) benefit in metastatic Castration Resistant Prostate Cancer (mCRPC) patients treated with Radium‐223 dichloride (Ra‐223) over placebo. Here we report clinical outcomes of Ra‐223 treatment in a nonstudy population. In this prospective registry, patients from 20 Dutch hospitals were included prior to Ra‐223 treatment. Clinical parameters collected included previous treatments and Adverse Events. Primary outcome was 6 months Symptomatic Skeletal Event (SSE)‐free survival, while secondary outcomes included Progression‐Free Survival (PFS) and Overall Survival (OS). Of the 305 patients included, 300 were evaluable. The mean age was 73.6 years, 90% had ≥6 bone metastases and 74.1% were pretreated with Docetaxel, 19.5% with Cabazitaxel and 80.5% with Abiraterone and/or Enzalutamide. Of all patients, 96.7% were treated with Ra‐223 and received a median of 5 cycles. After a median follow‐up of 13.2 months, 6 months SSE‐free survival rate was 83%, median PFS was 5.1 months and median OS was 15.2 months. Six months SSE‐free survival rate and OS were comparable with those reported in ALSYMPCA. “Previous Cabazitaxel treatment” and “bone‐only metastases” were independent predictors of a shorter and longer PFS, respectively, while above‐median LDH and “bone‐only metastases” were independent predictors of shorter and longer OS, respectively. Toxicity was similar as reported in the ALSYMPCA trial. These results suggest that in a nonstudy population, Ra‐223 treatment is well‐tolerated, equally effective as in the ALSYMPCA population and that patients not previously treated with Cabazitaxel benefit most from Ra‐223. 31. The role of neutrophil albumin ratio in predicting the stage of non-small cell lung cancer.

Abstract

OBJECTIVE: Inflammation forms the basis of cancer development and progression. It causes changes in complete blood count parameters, such as neutrophil counts. Low albumin levels are associated with poor prognosis in cancer patients. We aimed to investigate the association between neutrophil to albumin ratio (NAR) and the stage of non-small cell lung cancer (NSCLC). PATIENTS AND METHODS: 2 57 N SCLC p atients (24 females and 198 males) were included in the study. Patients were divided into two groups. Group 1 (n=61) included patients with early stage cancer (stage 1 and 2), while group 2 (n=196) included those with advanced stage cancer (stage 3 and 4). Demographic data, neutrophil, lymphocyte, platelet, white blood cell counts (WBC), C-reactive protein (CRP), erythrocyte sedimentation rate (ESR), ferritin and albumin levels at the time of diagnosis were recorded. The NAR of 2 groups were compared. RESULTS: There w ere no s ignificant d ifferences between the lymphocyte count (2.0 vs. 2.0 10³/mm³) and platelet count (291 vs. 311 10³/mm³) of the two groups (p > 0.05). ESR (38.8 vs. 57.5 mm/h), CRP (158 vs. 57 mg/l), ferritin (85 vs. 261 ng/ml), WBC count (8.6 vs. 10.6 10³/mm³), neutrophil count (5.6 vs. 7.5 10³/mm³), albumin values (2.9 vs. 3.7 gr/dl), and (p < 0.05) NAR levels (1.6 vs. 2.3) (p < 0.05) were significantly higher in group 2. Abstract

Today, with the faster development of science and technology, people have started to pay more attention to healthy living and consuming foods that are beneficial for human health. In this context, the value given to fruit species has become increasingly important and the importance of plants whose seeds are consumed has increased, as well as colourful fruits and vegetables. Pomegranate is a fruit grown in many countries in the world, especially in the Mediterranean climate, and its economic value has been increasing in recent years. This fruit has gained great nutritional importance not only because of its flavour but also because it contains many antioxidant substances and phenolic compounds that are beneficial to human health. There are nearly fifty pomegranate varieties registered in our country. These pomegranate varieties range from sweet to sour, from small-in size to large in size, and from hard-seeded to soft-seeded in terms of skin colour and fruit colour. In recent years, the use of pomegranate parts such as peel, seeds, and flowers in cosmetics and food industry has become widespread and studies on the antioxidant properties of different parts of pomegranate have attracted interest. The aim of this review is to investigate the health effects of pomegranate fruit, different parts of pomegranate, and products made from pomegranate.

Abstract

Anatolia is the gene center of many fruit species grown in the world and is also very rich in terms of fruit species and varieties. Diospyrus kaki L., known as persimmon, is one of these fruit species. Considering Türkiye’s ecological conditions, persimmon has a great potential for production. In recent studies, the potential health effects of persimmon have been cited as an important factor, increasing the popularity and consumption of persimmon. In addition, persimmon has become the center of attention in recent years due to the attractive orange color of the fruit, its distinctive taste and structure, rich in antioxidants and phenolic compounds. In this review study, it was aimed to increase awareness of the production potential of persimmon grown in Turkiye by revealing its production potential, and to shed light on decision-makers in future production planning by providing information in terms of economic importance and health.

Abstract

Objective: The ABCB1/MDR1 is a 170-kDa transporter protein called P-glycoprotein (P-gp) which has been associated with the transport of cellular lipids and drugs. Recent studies focused on MDR1, and its effects on lipid transport, show that the constitutive expression of P-gp transporter in normal tissues plays an important role in drug disposition and response. In addition, it is known that some polymorphisms on the MDR1 gene alter the expression of the P-gp. This study aims to investigate the effects of the MDR1 C3435T and the C1236T gene polymorphisms on the dyslipidemia of diabetic patients. Material and methods: The PCR-RFLP method has been used on 77 patients and 75 controls in order to determine the MDR1 genotype. Results: There are no statistical differences for the MDR1 C3435T and the C1236T genotype ranges among between our inspected groups. However, the C1236T mutant type T allele ratio increases statistically in the patient group (p = 0.026 OR: 1.679 % 95 CI: 1.062 - 2.652). A weak connection has been observed between the MDR1 C3435T C and the C1236T C alleles, according to the linkage disequilibrium analysis. Conclusion: This study is one of the preceding studies, which examines the relationship between MDR1 polymorphisms and type 2 diabetes. Studies on MDR1 genotypes, and their effects on lipid levels are new in literature, hence our study proves to be unique when it comes to both the C3435T and the C1236T polymorphisms in type 2 diabetes patients; however, further research is still needed for the confirmation of our findings.
