Microalgae are booming as a sustainable protein source for human nutrition and animal feed. Nevertheless, certain strains were reported to have robust cell walls limiting protein digestibility. There are several disruption approaches to break down the cell integrity and increase digestive enzyme accessibility. This review's intent is to discuss the digestibility of microalgae proteins in intact cells and after their disruption. In intact single cells, the extent of protein digestibility is chiefly related to cell wall structural properties (differing among strains) as well as digestion method and when added to food or feed protein digestibility changes depending on the matrix's composition. The degree of effectiveness of the disruption method varies among studies, and it is complicated to compare them due to variabilities in digestibility models, strains, disruption method/conditions and their consequent impact on the microalgae cell structure. More exhaustive studies are still required to fill knowledge gaps on the structure of microalgal cell walls and to find efficient and cost-effective disruption technologies to increase proteins availability without hindering their quality. The green microalga Monoraphidium sp. has potential for biodiesel production since it grows fast and can accumulate high levels of lipids. However, there is a lack of information on the potential use in human nutrition. In this work Monoraphidium sp. was characterized in terms of macronutrients with a special focus on the fatty acid profile of the lipid fraction and on the amino acid profile of the proteins. Furthermore, aiming at nutrient valorization, various methods for lipid extraction that could be used in the production of high quality and safe foods were investigated. To this end, the focus was on cell disruption methods in order to optimize oil recovery. The Monoraphidium sp. biomass had a high protein concentration (44.5 %) and a relatively low lipid concentration (12.5 %) but was rich in ω-3 fatty acids demonstrating its high nutritive value. Regarding cell disruption, ultrasonication and high-speed homogenization were insufficient to disrupt cells under the conditions examined. On the other hand, the effectiveness of the ball milling was regulated via processing time and water-to-biomass ratio and complete cell disruption could be achieved by this method. For lipid extraction, solvent extraction using a mixture of food-grade hexane and ethanol resulted in an oil recovery of 70.4 %, whereas using ethanol alone as an environmentally friendly solvent resulted in an oil recovery equal to 54 %. Supercritical CO2 extraction resulted in a lower oil recovery (25 %), whereas ethanol addition, as a cosolvent to CO2, significantly increased the oil recovery (60 %). [ABSTRACT FROM AUTHOR]

Summary This study explores enhancing lipid recovery from the lipid‐rich marine microalgae Aurantiochytrium sp. SW1, known for its robust cell wall posing challenges, which necessitates energy‐intensive cell‐disruption for efficient intracellular lipid extraction. Despite advancements in other microalgae, there is lack of reports on efficient cell disruption for Aurantiochytrium sp. SW1 which is vital to reduce the energy costs and minimising the downstream process while maximising lipid yields. Three process intensification technologies (PIT) – microwave treatment, autoclave treatment and ultrasonication were employed on wet Aurantiochytrium sp. SW1 cells to improve the lipid extraction efficiency. Assessing the disruption effectiveness via lipid content, suspension turbidity and particle size distribution, alongside scanning electron microscopy (SEM) for cellular morphology changes post‐PIT treatments, this study indicates all three methods reduced particle size with prolonged treatment time, implying successful cell wall disruption and intracellular component release. Ultrasonication, especially at 50 W power for 10 min, exhibited the highest efficacy, yielding lipid content of 67.76 ± 0.03%. Microscopic analysis unveiled ultrasonication‐induced cell shrinkage and increased membrane permeability, supporting its effectiveness in enhancing solvent penetration for improved lipid recovery. ABSTRACT: Microalgae have potentially beneficial effects on animal health and nutritional value when added to feed. Crucial hereby is that intracellular bio-active molecules are released in the intestinal tract. Digestibility of Chlorella vulgaris and its impact on total digestibility of broiler feed is a first step in assessing its characteristics as feed supplement. Different methods could be used to increase the digestibility of the algae. Among other, pulsed electric field (PEF) and freezing to disrupt autotrophic (A) and heterotrophic (H) Chlorella vulgaris cells was assessed to increase their availability followed by in-vivo trials. In these trials effect of algae type (A and H) and effect of PEF-processing was evaluated on the apparent nutrient digestibility. Pulsed electric field showed to have a disruption efficiency of 83.90% and 79.20% for heterotrophic and autotrophic C. vulgaris respectively. Freezing C. vulgaris only showed efficiencies ranging from 3.86 to 11.58%. In the in-vivo trials, microscopic counting of intact C. vulgaris cells showed an increase in digested intact C. vulgaris cells of PEF-processed C. vulgaris compared to nonprocessed cells ranging from 12.16% to 15.20%. Autotrophic C. vulgaris had a higher digestibility compared to heterotrophic C. vulgaris, with an increase of 7.29, 9.44, and 17.29% in digestibility of C. vulgaris in the 1, 2, and 5% feed respectively. Feeds with PEF-processed C. vulgaris showed no significant increase in digestibility compared to nonprocessed C. vulgaris supplemented feeds. The 5% C. vulgaris feeds showed lower fat digestibility than the 1 and 2% and control feeds. Protein digestibility was lower for all C. vulgaris feeds compared to the control feed. There was a significant linear decreasing effect (P < 0.001) for all digestibility parameters. Except for crude ash digestibility, which first lowered for the 1 and 2% feeds, but then increased at 5% inclusion. Considering this study, including low dosages of 1 and 2% of C. vulgaris in broiler feed does not compromise its digestibility.

ABSTRACT Microalgal lipids exhibit physical-chemical characteristics similar to vegetable oils, representing alternative sources of long-chain fatty acids for food supplements. However, separating intracellular lipids from microalgae biomass is still challenging due to downstream processing technology limitations that depend on the particular microalgal species and its cultivation conditions. Thus, this work evaluated a multistep process for lipid extraction from Tetradesmus obliquus biomass instead of conventional single extraction methods to improve the lipid yield. The wet biomass was freeze-dried, the cells of the dried biomass were disrupted in a ball mill, and the milled biomass was suspended in water to obtain solid contents of 5 % and 10 % w v–1. Lipids were extracted from the aqueous suspension using the ethanol polar solvent and the hexane apolar solvent with ethanol:hexane ratios of 1:1, 1:2, and 3:5 v v–1. Tests were also carried out on the deproteinized biomass to evaluate the effect of proteins on emulsion formation since deproteinization can avoid emulsion formation. Thus, lipid downstream processing could be designed without an emulsion removal stage. The biomass was deproteinized before lipid extraction with NaOH at pH 10. The highest lipid content was verified for the 1:2 ethanol:hexane ratio, in which up to 42 % of the total lipids were extracted from the biomass. With the 3:5 ratio, the organic and aqueous phases emulsified due to the presence of lipoproteins, which hindered the lipid extraction process. Under the studied conditions, no increase in lipid extraction yield was observed with increases in the solid content or biomass deproteinization.

Background: Antimicrobial peptides are promising alternatives to antibiotics to treat bacterial and fungal infections, especially drug-resistant clinical pathogens. Methods: Antimicrobial peptides (AMPs) were synthesized and antimicrobial activity was assayed. The antibacterial mechanism, ATP production, ROS generation and molecular mechanism were determined. Biofilm inhibition assay was performed in planktonic bacterial cells and biofilm degradation assay was performed using mature biofilm. The synthesized AMP2 was subjected to in vitro and in vivo analysis to analyze the safety. Results: The synthesized peptides AMP1, AMP2, AMP3 and AMP4 exhibited antimicrobial activity against Gram-positive and Gram-negative bacteria. The MIC values ranged from 1.5 ± 0.25–12.5 ± 1.25 µM and the MFC values range from 2.25 ± 0.12–25 ± 1.25 µM. F. solani showed fewer MFC values than other fungal strains. Time kill assay was performed and the AMP2 killed about 70 % of Acinetobacter baumannii at 1 × MIC concentration within 10 min incubation and killed 97 % of bacteria at 1 × MBC concentration within 15 min. The antimicrobial peptide AMP2 was highly effective against planktonic A. baumannii and L. monocytogenes. The tested AMP2 showed less toxicity to cell lines and Zebrafish. Conclusions: Antimicrobial peptides have potential antimicrobial properties against Gram-positive and Gram-negative bacteria. The in silico studies of these antimicrobial peptides are useful for eradicating drug-resistant bacteria.

It has various activities, such as anti-inflammatory and antioxidant properties. And are potential for applications in food, cosmetics, and biomedicine. Improving their metabolic yield is of great interest. Microalgaes are one of the important sources of PBPs, with high growth rate and have the potential for large-scale production. The key to large-scale PBPs production depends on accumulation and recovery of massive productive alga in the upstream stage and the efficiency of microalgae cells breakup and extract PBPs in the downstream stage. Therefore, we reviewed the status quo in the research and development of PBPs production, summarized the advances in each stage and the feasibility of scaled-up production, and demonstrated challenges and future directions in this field.

Microalgae have been suggested as a promising feedstock for bioethanol production due to accumulating high levels of carbohydrates. One of the significant challenges to facilitating the release of carbohydrates from the microalgal cell is identifying an effective cell disruption method. The present study evaluates pretreatment methods such as sonication, high-pressure homogenization, microwave-acid, and autoclave-acid to disrupt the cell wall of two native microalgae, Chlorella S4 and Picochlorum D8. The efficiency of each treatment was evaluated in terms of the disruption rate (%) and carbohydrate release. Microscopic images showed the disintegrated cells of two isolates with a high quantity of cell debris after autoclave-acid treatment compared to typical spherical cells before each pretreatment. Based on the results, the acid-autoclave pretreatment (1.5% (v/v) of H2SO4, 121 °C for 20 min) with a disruption rate of 86% was selected as the most effective disruption method for Chlorella S4. A maximum carbohydrate concentration of 0.211 ± 0.028 g/g dry biomass was obtained. For Picochlorum D8, the carbohydrate release of 0.237 ± 0.002 g/g dry biomass was achieved using the same pretreatment method with a disruption rate of 84%. The significant results demonstrate the effectiveness of autoclave-acid pretreatment for a wide range of biomass samples and a possible pilot-scale operation to produce bioethanol. Apple pomace, a solid waste produced during industrial processing of apple juice or cider, is a rich source of high value-added compounds such as polyphenols. This review summarizes present studies on the qualitative and quantitative methods, including Folin–Ciocalteu colorimetric, high pressure liquid chromatography (HPLC) and fluorescence spectrum, as well as enhanced extraction methods of polyphenols in apple pomace by different traditional and novel technologies, including ultrasounds (US), microwave (MW), pulsed electric fields (PEF), high voltage electrical discharges (HVED) and enzyme. The principles and characteristics of different effective enhanced extraction technologies of polyphenols in apple pomace were compared. In addition, the different cell disruption analysis methods, such as destructive detection method (electrical conductivity disintegration index, Zc), image analysis method (including scanning electron microscopy, SEM, and confocal laser scanning microscopy, CLSM), and nondestructive method (such as magnetic resonance imaging, MRI) are presented in this review. The study proved that there was a correlation between destructive detection method and image analysis method. However, each of the technologies reviewed in this study has some disadvantages to overcome, and some mechanisms need to be further substantiated. There is a growing interest in applying natural products in agriculture to stimulate growth and/or resistance of plants, and ultimately to increase productivity without causing further environmental degradation. Microalgae synthesize a wide variety of high-value products, including phytostimulants. Even if applied to plants in small proportions, these substances can increase crop yield and protect the plants from biotic and abiotic stress. The microalga Scenedesmus sp. was grown in simulated treated wastewater to demonstrate its ability to bioremediate and to be a source of phytostimulants. Optimization of the cell disruption method is required to improve the extraction of biomolecules. Therefore, in this work, cell disruption methods, namely high-pressure homogenization (HPH), ultrasonication (US), and hydrolysis, were compared. To evaluate the biostimulant potential, extracts were evaluated using the germination index of watercress seeds, rooting bioassay in mung bean and cucumber, and cotyledon expansion in cucumber. There was a 44% increase in the germination index when watercress were treated with a combination of HPH and enzymatic hydrolysis disrupted extracts. In the mung bean bioassay, the best results were obtained with extracts prepared by HPH and enzymatic hydrolysis, achieving 160% rooting compared to 100% rooting in the control treated with distilled water. For rooting activity in cucumber, the best results were obtained after biomass pre-treatment by HPH, both with or without the enzymatic hydrolysis treatment, achieving maximum root development values of 275%. Differences in cotyledon expansion were not significant. This work demonstrated the biostimulant effect of Scenedesmus sp. biomass on three plant species. HPH and enzymatic hydrolysis increased the overall effect although their inclusion might not be economical. [ABSTRACT FROM AUTHOR]

Abstract The interest for oleaginous yeasts has grown significantly in the last three decades, mainly due to their potential use as a renewable source of microbial oil or single cell oils (SCOs). However, the methodologies for cell disruption to obtain the microbial oil are considered critical and determinant for a large-scale production. Therefore, this work aimed to evaluate different methods for cell wall disruption for the lipid extraction of Yarrowia lipolytica QU21 and Meyerozyma guilliermondii BI281A. The two strains were separately cultivated in 5 L batch fermenters for 120 hours, at 26 ºC and 400 rpm. Three different lipid extraction processes using Turrax homogenizer, Ultrasonicator and Braun homogenizer combined with bead milling were applied in wet, oven-dried, and freeze-dried biomass of both strains. The treatment with the highest percentage of disrupted cells and highest oil yield was the ultrasonication of oven-dried biomass (37-40% lipid content for both strains). The fact that our results point to one best extraction strategy for two different yeast strains, belonging to different species, is a great news towards the development of a unified technique that could be applied at industrial plants.

This paper aims to develop a numerical methodology to investigate the penetration process of nanoneedles into cells and the corresponding force and indentation length. The finite element approach via the explicit dynamic method handles convergence difficulties in the nonlinear phenomenon. The cell is modeled as an isotropic elastic hemiellipsoidal shell with a thickness of 200 nm, which represents the lipid membrane and actin cortex, encapsulating cytoplasm that is regarded as an Eulerian body because of its fluid‐type behavior. Nanoneedles with diameters 400, 200, and 50 nm are considered for model development based on available experimental data. The Von Mises strain failure criterion is used for rupture detection. A parameter study using 1, 2.5, 5, 7.5, and 10 kPa shows that Young's modulus of the HeLa cell membrane is about 5 kPa. Moreover, a failure strain of 1.2 chosen among 0.2, 0.4, 0.6, 0.8, 1, and 1.2 matches best the experimental data. In addition, a diameter study shows that the relations between force‐diameter and indentation length‐diameter are linear and polynomial, respectively. Furthermore, regarding the experimental data and by using contour of minimum principal stress around needle and an analytical equation for calculation of buckling force of a woven fabric, we proposed that for a given cell, membrane structural stability—a function of the coupled effect of Young's modulus and actin meshwork size—contributes directly to needle insertion success rate for that type of cell. In the modern era, macro-microalgae attract a strong interest across scientific disciplines, owing to the wide application of these cost-effective valuable bioresources in food, fuel, nutraceuticals, and pharmaceuticals etc. The practice of eco-friendly extraction techniques has led scientists to create alternative processes to the conventional methods, to enhance the extraction of the key valuable compounds from macro-microalgae. This review narrates the possible use of novel cell disruption techniques, including use of ionic liquid, deep eutectic solvent, surfactant, switchable solvents, high voltage electrical discharge, explosive decompression, compressional-puffing, plasma, and ozonation, which can enable the recovery of value added substances from macro-microalgae, complying with the principles of green chemistry and sustainability. The above-mentioned innovative techniques are reviewed with respect to their working principles, benefits, and possible applications for macro-microalgae bioactive compound recovery and biofuel. The benefits of these techniques compared to conventional extraction methods include shorter extraction time, improved yield, and reduced cost. Furthermore, various combinations of these innovative technologies are used for the extraction of thermolabile bioactive compounds. The challenges and prospects of the innovative extraction processes for the forthcoming improvement of environmentally and cost-effective macro-microalgal biorefineries are also explained in this review. Microalgae are an alternative source of renewable energy and high-value products for pharmaceutical, nutraceutical, etc., due to rich in carbohydrates, proteins, lipids, and high-density lipoproteins. Existing methods for cell disruption and extraction are costly and suffered from low proficiencies. Ionic liquids are proven to be an environmentally friendly substitute to conventional volatile organic solvents. They have been used in extracting different types of biomass, including microalgae. This article reviews the potential of ILs in extracting biomolecules, lipid, and omega-3, from microalgae biomass. The physicochemical properties of ILs, including viscosity, density, and melting point, their advantages and limitation, as well as toxicity and recyclability of ILs in lipid processing, are discussed. Natural carotenoids are secondary metabolites that exhibit antioxidant, anti-inflammatory, and anti-cancer properties. These types of compounds are highly demanded by pharmaceutical, cosmetic, nutraceutical, and food industries, leading to the search for new natural sources of carotenoids. In recent years, the production of carotenoids from bacteria has become of great interest for industrial applications. In addition to carotenoids with C40-skeletons, some bacteria have the ability to synthesize characteristic carotenoids with C30-skeletons. In this regard, a great variety of methodologies for the extraction and identification of bacterial carotenoids has been reported and this is the first review that condenses most of this information. To understand the diversity of carotenoids from bacteria, we present their biosynthetic origin in order to focus on the methodologies employed in their extraction and characterization. Special emphasis has been made on high-performance liquid chromatography-mass spectrometry (HPLC-MS) for the analysis and identification of bacterial carotenoids. We end up this review showing their potential commercial use. This review is proposed as a guide for the identification of these metabolites, which are frequently reported in new bacteria strains. Nutritional oils (mainly omega-3 fatty acids) are receiving increased attention as critical supplementary compounds for the improvement and maintenance of human health and wellbeing. However, the predominant sources of these oils have historically shown numerous limitations relating to desirability and sustainability; hence the crucial focus is now on developing smarter, greener, and more environmentally favourable alternatives. This study was undertaken to consider and assess the numerous prevailing and emerging techniques implicated across the stages of fatty acid downstream processing. A structured and critical comparison of the major classes of disruption methodology (physical, chemical, thermal, and biological) is presented, with discussion and consideration of the viability of new extraction techniques. Owing to a greater desire for sustainable industrial practices, and a desperate need to make nutritional oils more available; great emphasis has been placed on the discovery and adoption of highly sought-after ‘green’ alternatives, which demonstrate improved efficiency and reduced toxicity compared to conventional practices. Based on these findings, this review also advocates new forays into application of novel nanomaterials in fatty acid separation to improve the sustainability of nutritional oil downstream processing. In summary, this review provides a detailed overview of the current and developing landscape of nutritional oil; and concludes that adoption and refinement of these sustainable alternatives could promptly allow for development of a more complete ‘green’ process for nutritional oil extraction; allowing us to better meet worldwide needs without costing the environment.

Hydrodynamic cavitation (HC) is the process of bubbles formation, expansion, and violent collapse, which results in the generation of high pressures in the order of 100–5000 bar and temperatures in the range of 727–9727 °C for just a fraction of seconds. Increasing consumer demand for high-quality foods with higher nutritive values and fresh-like sensory attributes, food processors, scientists, and process engineers are pushed to develop innovative and effective non-thermal methods as an alternative to conventional heat treatments. Hydrodynamic cavitation can play a significant role in non-thermal food processing as it has the potential to destroy microbes and reduce enzyme activity while retaining essential nutritional and physicochemical properties. As hydrodynamic cavitation occurs in a flowing liquid, there is a decrease in local pressure followed by its recovery; hence it can be used for liquid foods. It can also be used to create stable emulsions and homogenize food constituents. Moreover, this technology can extract food constituents such as polyphenols, essential oils, pigments, etc., via biomass pretreatment, cell disruption for selective enzyme release, waste valorization, and beer brewing. Other applications related to food production include water treatment, biodiesel, and biogas production. The present review discusses the application of HC in the preservation, processing, and quality improvement of food and other related applications. The reviewed examples in this paper demonstrate the potential of hydrodynamic cavitation with further expansion toward the scaling up, which looks at commercialization as a driving force.

Traditional technology of cell disruption has become one of the bottlenecks restricting the industrialization of genetic engineering products due to its high cost and low efficiency. In this study, a novel bioprocess of phage lysis coupled with salting‐out extraction (SOE) was evaluated. The lysis effect of T7 phage on genetically engineered Escherichia coli expressing κ‐carrageenase was investigated at different multiplicity of infection (MOI), meanwhile the phage and enzyme released into the lysate were separated by SOE. It was found that T7 phage could lyse 99.9% of host cells at MOI = 1 and release more than 90.0% of enzyme within 90 min. After phage lysis, 87.1% of T7 phage and 71.2% of κ‐carrageenase could be distributed at the middle phase and the bottom phase, respectively, in the SOE system composed of 16% ammonium sulfate and 20% ethyl acetate (w/w). Furthermore, κ‐carrageenase in the bottom phase could be salted out by ammonium sulfate with a yield of 40.1%. Phage lysis exhibits some advantages, such as mild operation conditions and low cost. While SOE can efficiently separate phage and intracellular products. Therefore, phage lysis coupled with SOE is expected to become a viable alternative to the classical cell disruption and intracellular product recovery. [ABSTRACT FROM AUTHOR]

The environmental concerns about fossil energy and rising oil prices are regular global phenomena. In such an exigency where fossil fuel sources are dwindling, carbon‐neutral biodiesel derived from microalgae lipid seems to be an important renewable energy resource. In recent years, microalgae have become a lucrative feedstock for economically viability of commercial‐scale biofuel production owing to its high biomass growth and high oil yield compared with other crops. The screening and selection of promising microalgae strain should be based on their fatty acid profile and culture conditions. The laboratory to pilot scale culture of microalgae species requires systematic experimental designs to maximize microalgal growth using open or raceway ponds, closed photo bioreactors and hybrid systems. The pros and cons of each harvesting technique, including filtration, centrifugation, sedimentation and flocculation as well cell disruption by thermal, mechanical, chemical and biological techniques, are deliberated in details. Furthermore, the application of homogeneous, heterogeneous and enzyme catalysts in transesterification processes as well as purification technologies are emphasized to ensure high‐quality biodiesel production as per ASTM norms. Microalgae comprise a potential feedstock and biodiesel synthesis from them requires a series of complex up‐streaming and down‐stream processes which are inherently disadvantageous in technology transfer from laboratory to industrial scale. Abstract Traditional technology of cell disruption has become one of the bottlenecks restricting the industrialization of genetic engineering products due to its high cost and low efficiency. In this study, a novel bioprocess of phage lysis coupled with salting‐out extraction (SOE) was evaluated. The lysis effect of T7 phage on genetically engineered Escherichia coli expressing κ‐carrageenase was investigated at different multiplicity of infection (MOI), meanwhile the phage and enzyme released into the lysate were separated by SOE. It was found that T7 phage could lyse 99.9% of host cells at MOI = 1 and release more than 90.0% of enzyme within 90 min. After phage lysis, 87.1% of T7 phage and 71.2% of κ‐carrageenase could be distributed at the middle phase and the bottom phase, respectively, in the SOE system composed of 16% ammonium sulfate and 20% ethyl acetate (w/w). Furthermore, κ‐carrageenase in the bottom phase could be salted out by ammonium sulfate with a yield of 40.1%. Phage lysis exhibits some advantages, such as mild operation conditions and low cost. While SOE can efficiently separate phage and intracellular products. Therefore, phage lysis coupled with SOE is expected to become a viable alternative to the classical cell disruption and intracellular product recovery.

BACKGROUND:Cerebral microhemorrhages (CMH) are commonly found in the aging brain. CMH are also the neuropathological substrate of cerebral microbleeds (CMB), demonstrated on brain MRI. Recent studies demonstrate the importance of systemic inflammation in CMH development, but the relationships among inflammation, aging, and CMH development are not well-defined. In the current study, we hypothesized that the pathogenesis of inflammation-induced CMH in mice differs by age. METHODS:We studied young (3 months, n = 20) and old (18 months, n = 25) C57BL/6 mice injected with low-dose lipopolysaccharide (LPS; 1 mg/kg, i.p.) or saline at 0, 6, and 24 h. Seven days after the first LPS/saline injection, brains were harvested, sectioned, and stained with hematoxylin and eosin (H&E) and Prussian blue (PB) to estimate acute/fresh and sub-acute CMH development, respectively. The relationships between microglial/macrophage activation (ionized calcium-binding adapter molecule-1), astrocyte activation (glial fibrillary acidic protein), blood-brain barrier (BBB) disruption (brain immunoglobulin G), aging, and CMH development were examined using immunohistochemistry. RESULTS:Aging alone did not increase spontaneous H&E-positive CMH development but significantly increased the number, size, and total area of LPS-induced H&E-positive CMH in mice. LPS- and saline-treated aged mice had significantly larger PB-positive CMH compared with young mice, but the total area of PB-positive CMH was increased only in LPS-treated aged mice. Aged mice had significantly increased microglial/macrophage activation, which correlated with H&E- and PB-positive CMH development. Aged mice treated with LPS had significantly increased astrocyte activation and BBB disruption compared with young LPS-treated mice. CONCLUSIONS:Aging makes the brain more susceptible to inflammation-induced CMH in mice, and this increase in CMH with aging is associated with microglial/macrophage activation.

Microalgae are potential biomass source for biodiesel production. However, their strong cell walls make efficient lipid extraction problematic. Disrupting the cell wall is a key point in enhancing lipid yield from microalgae biomass. A new type of ionic liquid (IL) has been suggested in this work as a potentially viable solvent to permeabilize the strong microalgae cell structure for the efficient extraction of lipids. Morphological changes in microalgae cells were studied before and after ionic liquid permeabilization to understand the mechanism of ionic liquid treatment. Among the three selected CO2-based alkyl carbamate ionic liquids, DIMCARP performed with the best extraction efficiency. The effects of extraction variables (temperature, time, ratio ionic liquid/Methanol, and solvent to biomass) on lipid extraction were examined via single-factor experiments coupled with response surface methodology (RSM) using a Box-Behnken design (BBD). The highest lipid yield (16.40%) was obtained after 45 min of extraction at 45 °C using a 9:1 ionic liquid to methanol and 7 mL of solvent to biomass ratio. Transesterification of lipids to make fatty acid methyl esters found that the most common fatty acids were C16:0, C18:2, and C18:3 (19.50%). The quality of the biodiesel made meets European and US standards.

This research aimed to develop blends of corn starch and poly(butylene adipate‐co‐terephthalate) (PBAT) films by blown film extrusion with the addition of biomass of Chlorella pyrenoidosa (CP). Two ultrasound tracts disrupted CP biomass: ultrasound bath and ultrasonic probe. The influence of CP biomass and disrupted CP biomass on the physical, mechanical, barrier, structural, and thermal properties of starch/PBAT films was evaluated. The film with higher content (5.0%) of CP biomass showed better values in tensile strength (TS) (4.37 MPa) and elongation (88.43%), and lower water vapor permeability (WVP) (5.19 × 10−11 g m−1 s−1 Pa−1) than the film incorporated with disrupted CP biomass. The ultrasound treatment applied to the cell disruption of the microalgae did not present improvements in the properties of the films. In this context, the cell disruption of CP biomass by ultrasound for incorporation into biodegradable films was not feasible. However, it was possible to produce starch/PBAT/microalgae films with potential application as food packaging due to natural bio‐pigmentation's protection system against ultraviolet light. Practical Applications: Films produced by blown extrusion from starch, PBAT, and CP microalgae biomass have the technological potential to be used as packaging for food products. Starch and PBAT blends are widely studied. The incorporation of PBAT is necessary to confer adequate mechanical properties to the extruded material. Phycoerythrin (PE) is a phycobiliprotein holding great potential as a high-value food colorant and medicine. Deep eutectic solvent (DES)-based ultrasound-assisted extraction (UAE) was applied to extract B-PE by disrupting the resistant polysaccharide cell wall of Porphyridium purpureum. The solubility of cell wall monomers in 31 DESs was predicted using COSMO-RS. Five glycerol-based DESs were tested for extraction, all of which showed significantly higher B-PE yields by up to 13.5 folds than water. The DES-dependent B-PE extraction efficiencies were proposedly associated with different cell disrupting capabilities and protein stabilizing effects of DESs. The DES-based UAE method could be considered green according to a metric assessment tool, AGREEprep. The crude extract containing DES was further subjected to aqueous two-phase system, two-step ammonium sulfate precipitation, and ultrafiltration processes. The slow breakdown of sludge is the primary obstacle hindering the conversion of waste-activated sludge to short-chain fatty acids (SCFAs) by anaerobic fermentation. This study proposed a novel method incorporating sodium pyrophosphate and thermal hydrolysis (SP-TH) for sludge pretreatment and evaluated its effectiveness regarding SCFA production. The combined pretreatment of SP at 0.4 g/g of total suspended solids and TH at 140 °C enhanced SCFA production from 2,169 ± 208 to 4,388 ± 184 mg chemical oxygen demand/L. SP strips extracellular polymeric substances, and the subsequent TH decomposes cells in the sludge, thus promoting sludge hydrolysis. SP-TH pretreatment promoted SCFA accumulation by enhancing enzyme activity and enriching acidifying bacteria. This study demonstrated that SP-TH pretreatment can effectively promote acid production from sludge, providing a new avenue for organic matter recovery through sludge anaerobic fermentation pretreatment. Microalgae and cyanobacteria are photosynthetic and unicellular organisms that contain considerable amounts of proteins, lipids, carbohydrates, and polyunsaturated fatty acids, among others, with applications in the cosmetic, pharmaceutical, and food industries. These microorganisms can accumulate protein up to 70 % of total biomass depending on the microalgal strain, hence they have been regarded as an alternative protein source for the future. Microalgal proteins have important applications such as emulsifying, foaming, and gelation properties, which are important for the determination of quality and texture of foods. Some microalgal peptides possess important bioactivity with many health-benefit effects. Therefore, to maximize the production of proteins from microalgae and cyanobacteria, many protein extraction procedures have been studied to increase the economic return. They have been tested towards higher protein yields at low energy cost, the preservation of protein native properties, and lower cell debris. This later is fundamental to facilitate the subsequent purification processes so that the overall cost can be reduced. The aim of this work is to review some cell disruption processes for the extraction of protein from microalgae and cyanobacteria, considering that this step is crucial for the overall process due to the high rigidness of microalgal cell covering, which can hamper the release of proteins. It also aims at reviewing the purification techniques after cellular disruption, from conventional to more recent approaches, and finally addresses the antioxidant, antidiabetic, antihypertensive, antibacterial and other bioactive properties of microalgal protein hydrolysates and peptides. Microalgae are a potential source of numerous nutritional products and biofuels. Their applications range from the food industry to the medical and fuel sectors and beyond. Recently, the conversion of biomass into biodiesel and other biofuels has received a lot of positive attention within the fossil fuel arena. The objective of biorefineries is to focus on utilising biomass efficiently to produce quality biofuel products by minimising the input as well as to reduce the use of chemical or thermal pre-treatments. Pre-treatment processes in biorefineries involve cell disruption to obtain lipids. Cell disruption is a crucial part of bioconversion, as the structure and nature of microalgae cell walls are complex. In recent years, many research papers have shown various pre-treatment methods and their advantages. The objective of this paper was to provide a comprehensive in-depth review of various recent pre-treatment techniques that have been used for microalgal biodiesel production and to discuss their advantages, disadvantages, and how they are applied in algal biorefineries. Converting microalgal lipids into biodiesel is considered a promising route in the field of biofuel production. However, the cost of microalgae-based biodiesel is still too high to be economically feasible, especially economical and effective methods for microalgae cell wall breaking are still lacking. In this study, five algicidal bacteria and their cell-free supernatant were evaluated by the percentage of cell disruption and lipid extraction of Chlorella sp.. Results showed that both bacteria and supernatant could induce cell disruption of Chlorella sp. to release lipids. Among them, the supernatant of Aeromonas hydrophila showed the best effects both in terms of the percentage of cell disruption and lipid extraction, and the cell disruption percentage increased with the extension of co-culture time with A. hydrophila supernatant. The inoculation volume of co-culture (A. hydrophila cell-free supernatant and Chlorella sp.) was explored, indicating that 10% volume ratio of A. hydrophila supernatant was identified as optimal proportion to improve lipid extraction. In addition, A. hydrophila supernatant showed obvious effects on the composition distribution of lipids extracted from Chlorella sp. that the fraction of unsaturated fatty acids decreased apparently, which would be beneficial to biodiesel production. The mechanism of microalgal cell disruption by A. hydrophila supernatant was further studied by scanning electron microscopy, ROS levels and antioxidant responses. This indicated that the cell morphology of Chlorella sp. changed dramatically with the time of co-culture with A. hydrophila supernatant.. These findings implied that algicidal properties of bacteria components are a promising tool for microalga cell disruption and lipid recovery, providing a new insight into the method for biofuel production. While microalgae are promising biomass candidates for biodiesel production, efficient lipid extraction is hindered by the strong cell walls. Cell wall disruption is an essential processing step for improving lipid yields from microalgae biomass. Conventional methods use toxic organic solvents (e.g., mixtures of chloroform and methanol or hexane) to extract microalgal lipids. This study with Chlorella pyrenoidosa investigates a novel microalgal pretreatment by a deep eutectic solvent (choline chloride:acetic acid) to increase cell wall disruption prior to lipid extraction. Deep eutectic solvent and ultrasonic homogenization should be best performed after 5 min of pretreatment. Biomass was characterized before and after the pretreatment to understand the mechanism. To optimize the lipid extraction yield, the effect of various variables (methanol to n-butanol, solvent to biomass ratio, time, and temperature) on lipid extraction yield was investigated using a single-factor experiment. The maximum lipid yield (19.25%) was extracted at 1:4 methanol to n-butanol and a 30 (v/w) solvent to biomass ratio over 120 min at 60 °C. Fatty acid methyl esters were obtained through transesterification reactions from extracted lipid of optimized condition and biodiesel quality was studied in comparison with European and US standards. The dominant fatty acids were C16:0 (29.54%), C18:2 (18.42%), and C18:3 (19.50%) and properties of biodiesel are within the European and US standards. This novel pretreatment method, coupled with lipid yields and characterization, documents the significant potential for microalgae biodiesel as an alternative, sustainable energy source. Microalgae have evolved into a promising sustainable source of a wide range of compounds, including protein, carbohydrates, biomass, vitamins, animal feed, and cosmetic products. The process of extraction of intracellular composites in the microalgae industry is largely determined by the microalgal species, cultivation methods, cell wall disruption techniques, and extraction strategies. Various techniques have been applied to disrupt the cell wall and recover the intracellular molecules from microalgae, including non-mechanical, mechanical, and combined methods. A comprehensive understanding of the cell disruption processes in each method is essential to improve the efficiency of current technologies and further development of new methods in this field. In this review, an overview of microalgal cell disruption techniques and an analysis of their performance and challenges are provided. A number of studies on cell disruption and microalgae extraction are examined in order to highlight the key challenges facing the field of microalgae and their future prospects. In addition, the amount of product recovery for each species of microalgae and the important parameters for each technique are discussed. Finally, pulsed electric field (PEF)-assisted treatments, which are becoming an attractive option due to their simplicity and effectiveness in extracting microalgae compounds, are discussed in detail. [Display omitted] Whereas limited amount of precious metal adsorbed by bacteria conflicting the needs of high loadings for better catalytic performances, cell disruption technology was adopted to smash Shewanella cells in this work, releasing abundant oxygen functional groups inside the cells for better adsorption of palladium ion. Then palladium catalysts were synthesized in two ways: 1) Pd catalyst supported on carbonized-broken-bacterial (Pd/FHNC) was obtained after direct carbonization and reduction; 2) Electrospinning technology was used to spin the broken Shewanella into fibers, and Pd nanoparticles supported on nitrogen-doped carbon nanofiber (Pd/NCNF) was prepared following carbonization and hydrogen reduction. The as-prepared catalysts exhibit excellent oxygen reduction reaction (ORR) electrocatalytic performance in the acid medium. The mass specific activities at 0.7 V of Pd/FHNC and Pd/NCNF were 0.213 A mg−1 and 0.121 A mg−1 which were 5.92 and 3.36 times than those of commercial Pd/C(0.036 A mg−1) respectively, and they also displayed higher stability than Pd/C. Furthermore, the Pd loadings of Pd/FHNC and Pd/NCNF were 21.52% and 17.13% respectively. An explanation for the improved performance is the co-doping of nitrogen and phosphorus, also the tight integration of Pd and broken-bacterial. Alternative nutrient sources for fish feed are currently of high interest. Feed ingredients with suitable lipid contents and valuable amounts of essential fatty acids are especially needed to relieve the usage of fish oil and maintain high product quality of fish from aquaculture. Microalgae contain promising nutrient contents and suitable fatty acid profiles that cover dietary requirements of fish. Nutrient digestibility, however, can be hindered by their rigid cell wall. Therefore, in the presented study, the microalgae species Isochrysis galbana and Tetraselmis chui were processed by freeze-drying and centrifugal milling to evaluate their nutrient and fatty acid digestibility in fish nutrition. For T. chui , an additional treatment with enzyme supplementation was tested. Test diets contained 70% of a fish meal based reference diet and 30% of the respective microalgae meal, as well as titanium dioxide as an inert marker to calculate apparent digestibility coefficients (ADC). Rainbow trout (Oncorhynchus mykiss) were fed for 14 days and faeces were collected via manual stripping. The ADCs of Kjeldahl nitrogen (∼ 64–79%), nitrogen-free extracts (∼ 40–54%) and energy (∼ 52–66%) were not significantly different between the algae species, but I. galbana inclusion resulted in reduced lipid digestibility (∼ 57% to 77%), which was linked to the high content of saturated fatty acids. The fatty acid ADCs were higher for T. chui except for arachidonic acid (20:4 n-6) and lauric acid (12:0). Mechanical or enzyme-treating processing did not increase nutrient and fatty acid digestibility for the microalgae used in this trial, and sole freeze-drying sufficiently ensured nutrient availability. The high digestibility of nutrients and, particularly, polyunsaturated fatty acids of I. galbana and T. chui underscore their potential as fatty acid and nutrient sources in fish feed. • Isochrysis galbana and Tetraselmis chui are promising fatty acid sources for fish. • Lipid and fatty acid digestibility is lower for I. galbana than for T. chui. • Freeze-drying sufficiently provided nutrient availability for these microalgae. High-intensity ultrasonication is an emerging technology for plant protein isolation and modification. In this study, the potential of temperature-controlled ultrasonication to enhance the recovery of functional proteins from potato trimmings was assessed. Different ultrasound energy levels [2000–40,000 J/g fresh weight (FW)] were applied during protein extraction at pH 9.0. True protein yields after ultrasonication significantly increased (up to 91%) compared to conventional extraction (33%). Microstructural analysis of the extraction residues showed more disrupted cells as ultrasonication time increased. Ultrasound treatments (10,000 and 20,000 J/g FW) increased the protein yield without affecting the foaming and air-water interfacial properties of protein isolates obtained after isoelectric precipitation (pH 4.0). However, proteins obtained after extended ultrasonication (40,000 J/g FW) had significantly slower early-stage adsorption kinetics. This was attributed to ultrasound-induced aggregation of the protease inhibitor fraction. In conclusion, ultrasonication shows potential to help overcome some challenges associated with plant protein extraction. Microalgae are a promising feedstock for the production of biofuels, nutraceuticals, pharmaceuticals and cosmetics, due to their superior capability of converting solar energy and CO2 into lipids, proteins, and other valuable bioactive compounds. To facilitate the release of these important biomolecules from microalgae, effective cell disruption is usually necessary, where the use of ultrasound has gained tremendous interests as an alternative to traditional methods. This review not only summarizes the mechanisms of and operation parameters affecting cell disruption, but also takes an insight into measuring techniques, synergistic integration with other disruption methods, and challenges of ultrasonication for microalgal biorefining. Optimal conditions including ultrasonic frequency, intensity, and duration, and liquid viscosity and sonochemical reactor are the key factors for maximizing the disruption and extraction efficiency. A combination of ultrasound with other disruption methods such as ozonation, microwave, homogenization, enzymatic lysis, and solvents facilitates cell disruption and release of target compounds, thus provides powerful solutions to commercial scale-up of ultrasound extraction for microalgal biorefining. It is concluded that ultrasonication is a sustainable “green” process, but more research and work are needed to upscale this process without sacrificing performance or consuming more energy.

Microalgae have potentially beneficial effects on animal health and nutritional value when added to feed. Crucial hereby is that intracellular bio-active molecules are released in the intestinal tract. Digestibility of Chlorella vulgaris and its impact on total digestibility of broiler feed is a first step in assessing its characteristics as feed supplement. Different methods could be used to increase the digestibility of the algae. Among other, pulsed electric field (PEF) and freezing to disrupt autotrophic (A) and heterotrophic (H) Chlorella vulgaris cells was assessed to increase their availability followed by in-vivo trials. In these trials effect of algae type (A and H) and effect of PEF-processing was evaluated on the apparent nutrient digestibility. Pulsed electric field showed to have a disruption efficiency of 83.90% and 79.20% for heterotrophic and autotrophic C. vulgaris respectively. Freezing C. vulgaris only showed efficiencies ranging from 3.86 to 11.58%. In the in-vivo trials, microscopic counting of intact C. vulgaris cells showed an increase in digested intact C. vulgaris cells of PEF-processed C. vulgaris compared to nonprocessed cells ranging from 12.16% to 15.20%. Autotrophic C. vulgaris had a higher digestibility compared to heterotrophic C. vulgaris , with an increase of 7.29, 9.44, and 17.29% in digestibility of C. vulgaris in the 1, 2, and 5% feed respectively. Feeds with PEF-processed C. vulgaris showed no significant increase in digestibility compared to nonprocessed C. vulgaris supplemented feeds. The 5% C. vulgaris feeds showed lower fat digestibility than the 1 and 2% and control feeds. Protein digestibility was lower for all C. vulgaris feeds compared to the control feed. There was a significant linear decreasing effect (P < 0.001) for all digestibility parameters. Except for crude ash digestibility, which first lowered for the 1 and 2% feeds, but then increased at 5% inclusion. The world will face a significant protein demand in the next few decades, and due to the environmental concerns linked to animal protein, new sustainable protein sources must be found. In this regard, microalgae stand as an outstanding high-quality protein source. However, different steps are needed to separate the proteins from the microalgae biomass and other biocompounds. The protein recovery from the disrupted biomass is usually the bottleneck of the process, and it typically employs organic solvents or harsh conditions, which are both detrimental to protein stability and planet health. Different techniques and methods are applied for protein recovery from various matrices, such as precipitation, filtration, chromatography, electrophoresis, and solvent extraction. Those methods will be reviewed in this work, discussing their advantages, drawbacks, and applicability to the microalgae biorefinery process. Special attention will be paid to solvent extraction performed with ionic liquids (ILs) and deep eutectic solvents (DESs), which stand as promising solvents to perform efficient protein separations with reduced environmental costs compared to classical alternatives. Ionic liquids (ILs) are new green solvents, which are widely used in lignocellulosic and microalgal biorefineries. In this study, we report the technical feasibility of room-temperature astaxanthin extraction from Haematococcus lacustris cysts with a thick and complex cell wall structure, by combining ultrathin α-quartz nanoplates (NPLs) with ethyl-3-methylimidazolium ([Emim])-based ILs. When four different [Emim]-based ILs with thiocyanate (SCN), diethylphosphate (DEP), HSO4, and Cl anions were applied to 90-day-old H. lacustris cysts at room temperature (~28 °C), the astaxanthin extraction efficiency was as low as 9.6–14.2%. Under sonication, α-quartz NPLs disrupted the cyst cell wall for a short duration (5 min). The astaxanthin extraction efficacies of a subsequent IL treatment improved significantly to 49.8% for [Emim] SCN, 60.0% for [Emim] DEP, 80.7% for [Emim] HSO4, and 74.3% for [Emim] Cl ions, which were 4.4, 6.1, 8.4, and 5.2 times higher than the extraction efficacy of only ILs, respectively. This finding suggests that α-quartz NPLs can serve as powerful cell-wall-disrupting agents for the room-temperature IL-mediated extraction of astaxanthin from robust algal cyst cells.
