Objective To analyze the effect of the standing time of density gradient solution on the recovery rate of semen optimized by discontinuous density gradient centrifugation. Methods Semen samples from 30 patients were collected. According to the experimental design, SpermGrad bilayer gradient solution(1.5 mL of 40% + 1.5 mL 80%) was prepared, and then placed for 2 hours, 1 hour, 30 minutes, 20 minutes, 10 minutes, and 5 minutes, respectively. Afterwards, 1 mL of the same patient’s semen was simultaneously transferred to the gradient solution mentioned above for optimization centrifugation treatment, and their PR recovery rates were calculated and compared statistically. Results The PR recovery rates of semen were(37.21±3.89)%、(39.62±4.30)%、(43.09±4.11)%、(45.89±4.71)%、(46.65±4.65)%、(46.87±4.84)%, after the gradient solutions were placed for 2 hours, 1 hour, 30 minutes, 20 minutes, 10 minutes, and 5 minutes. There was no significant difference in PR recovery rate between 20min, 10 min and 5 min(P>0.05), However, the PR recovery rates of the groups with the standing time over 20 minutes were significantly different from that of the group with standing time for 5 min(P<0.05). Conclusion The PR recovery rate is significantly decreased when the standing time of gradient solution is more than 20 minutes. It is suggested to perform gradient centrifugation within 20 minutes after the gradient solution preparation for improving the PR recovery rate. [ABSTRACT FROM AUTHOR]
