Abstract

The pattern of nuclear DNA variations in 8 Hypochaeris species was established by flow cytometry. Phylogenetic analysis suggests the high probability that genome size evolved by losing DNA content in some lineages and by gain in another. No correlation between 2C DNA content and GC% was discovered, so neither GC nor AT fractions were preferentially associated with size in genome.

Abstract

The pattern of rDNA sequence distribution in 11 Hypochaeris species from South America and Europe was observed by in situ hybridization utilizing probes of 18S-5.8S-28S and 5S ribosomal RNA genes. The rDNA localization revealed that chromosomes rearrangements as centric shifts, centric fusions and inactivation of rDNA genes happened during the course of evolution of Hypocharies species from the two geographical areas.

Abstract

Three taxa belonging to the genus Hyoseris L. are compared from the caryologica! point of view. H. i-adiaia L. and H. scabra L. are regarded as "good species", whereas the taxonomic status of H. lucida L. is doubtful. This study is carried out on several populations collected in North Africa and the Iberian peninsula. Karyotypic differences occur in the morphology of chromosomes and in their banding patterns with the fluorochromes Chromomycin A, and Hoechst 33258, which settle on DNA rich in GC and AT bases respectively. Chromosome morphology differentiates H. scabra from both other taxa, which can be distinguished from each other by the number of GC-rich centromeric bands. The length of the heterochrornatic part of chromosome pair 3 displays a variation within H. scabra. Ribosomal RNA genes are located by in situ hybridization on chromosome pair 3 in H. scabra and provide additional evolutionary markers.
