Actualmente, los consumidores son más conscientes de la relación entre la dieta y la salud, buscando alimentos más saludables como los alimentos probióticos. Sin embargo, la mayoría de estos productos comerciales son productos lácteos fermentados. Este trabajo tiene como objetivo evaluar la viabilidad de los probióticos Lactobacillus plantarum y Bifidobacterium longum en una bebida de quinoa roja (Chenopodium quinoa, L.) tras diferentes tiempos de fermentación en esta matriz. Para ello, se determinó el número de células viables, las propiedades antioxidantes y la concentración de polifenoles totales en diferentes tiempos de fermentación, así como tras la digestión gastrointestinal in vitro, junto con parámetros físico-químicos y la resistencia microbiológica a distintos antibióticos. Los resultados mostraron que el tiempo de fermentación influyó positivamente sobre la viabilidad de los probióticos, siendo ésta superior en la bebida con B. longum después de 24 horas de fermentación. Tras la digestión in vitro de las bebidas de quinoa roja, la viabilidad de ambos probióticos fue muy elevada. Por lo tanto, las bebidas a base de este pseudocereal son un buen vehículo para mantener la viabilidad de L. plantarum y B. longum por encima del mínimo recomendado (106-107 UFC/mL) de modo que puedan ejercer sus efectos beneficiosos a nivel intestinal. The objective of this work is to evaluate the viability of the probiotics Lactobacillus plantarum and Bifidobacterium longum in a red quinoa (Chenopodium quinoa, L.) beverage at different fermentation times in this matrix. Thus, the number of viable cells, the antioxidant properties and the concentration of total polyphenols at different fermentation times were determined, as well as after the in vitro gastrointestinal digestion, together with physico-chemical parameters and the microbial resistance to different antibiotics. In conclusion, the results showed that the fermentation time had a positive influence in the probiotic viability being higher in the beverage with B. longum after 24 hours of fermentation. The viability of both probiotics was very high in the red quinoa beverages after in vitro digestion. Therefore this pseudocereal is a good carrier to maintain the viability of L. plantarum and B. longum above the minimum recommended (106-107 CFU/mL) so that they could exert their beneficial effects in the gut

El gen INCURVATA11 (ICU11) codifica un componente de la maquinaria epigenética de Arabidopsis thaliana cuya función molecular concreta se desconoce. En un escrutinio de interactores de ICU11, basado en el ensayo del doble híbrido de la levadura, se identificaron en el laboratorio de José Luis Micol tres proteínas que participan en la ruta de la metilación dependiente de la S-adenosil-L-metionina (SAM): ADENOSINE KINASE 2 (ADK2), COBALAMIN-INDEPENDENT METHIONINE SYNTHASE (CIMS) y DEREPRESSED FOR RIBOSOMAL PROTEIN S14 EXPRESSION 2 (DRE2). Con el fin de confirmar estos resultados, hemos estudiado en este Trabajo de Fin de Grado las interacciones genéticas de ICU11 con ADK2, DRE2 y CIMS, así como con otros dos genes relacionados con el ciclo de la SAM: S-ADENOSYL-L-HOMOCYSTEINE HYDROLASE 1 (SAHH1) y ADK1, un parálogo de ADK2. Hemos obtenido alelos mutantes de estos genes y sus combinaciones dobles con icu11-1, y caracterizado sus fenotipos. También hemos generado transgenes productores de microARN artificiales y pequeños ARN interferentes sintéticos que actúan en trans, a fin de determinar los efectos fenotípicos de la pérdida de función parcial de los genes a estudio, en plantas icu11-1 y del tipo silvestre Col-0. The INCURVATA11 (ICU11) gene encodes a component of the epigenetic machinery of Arabidopsis thaliana, whose specific molecular function is unknown. In a screen for ICU11 interactors, based on the yeast two-hybrid assay, three proteins were identified in the laboratory of José Luis Micol, which participate in the S-adenosyl-L-methionine (SAM)-dependent methylation pathway: ADENOSINE KINASE 2 (ADK2), DEREPRESSED FOR RIBOSOMAL PROTEIN S14 EXPRESSION 2 (DRE2), and COBALAMIN-INDEPENDENT METHIONINE SYNTHASE (CIMS). In this End of Degree Assignment, we studied the genetic interactions of ICU11 with ADK2, DRE2 and CIMS, as well as with two other genes related to the SAM cycle: S-ADENOSYL-L-HOMOCYSTEINE HYDROLASE 1 (SAHH1) and ADK1, an ADK2 paralog. We obtained mutant alleles of these genes and their double mutant combinations with icu11-1, and characterized their phenotypes. We also generated transgenes producing artificial microRNAs and synthetic trans-acting small interfering RNAs, to ascertain the phenotypic effects of the partial loss of function of the genes under study in icu11-1 and wild-type genetic backgrounds.
