Oral bacterial infections are a great health concern worldwide especially in diabetic patients. Emergence of antimicrobial resistance with reference to biofilms in oral cavity is of great concern. We investigated antibiotics combination with proton pump inhibitors against oral clinical isolates. The strains were identified as Staphylococcus epidermidis and Staphylococcus aureus by the 16S rRNA gene sequencing. In molecular docking, ciprofloxacin, levofloxacin, and omeprazole best fit to active pockets of transcriptional regulators 4BXI and 3QP1. None of the proton pump inhibitors were active against S. epidermidis, whereas omeprazole showed significant inhibition (MIC 3.9 μg/ml). Fluoroquinolones were active against both S. epidermidis and S. aureus. In combination analysis, a marked decrease in minimum inhibitory concentration was noticed with omeprazole (MIC 0.12 μg/ml). In antiquorum sensing experiments, a significant inhibitory zone was shown for all fluoroquinolones (14–20 mm), whereas among proton pump inhibitors, only omeprazole (12 ± 0.12 mm) was active against Chromobacterium violaceum. In combination analysis, a moderate increase in antiquorum sensing activity was recorded for ciprofloxacin, ofloxacin, and proton pump inhibitors. Further, significant S. aureus biofilm eradication was recorded using of ciprofloxacin, levofloxacin, and omeprazole combination (78 ± 2.1%). The time-kill kinetic studies indicated a bactericidal effect by ciprofloxacin: levofloxacin: omeprazole combination over 24 hrs. It was concluded that fluoroquinolone combined with omeprazole could be an effective treatment option for eradicating oral bacterial biofilms.

Background: Amid growing concern about antimicrobial resistance due to the irrational use of antibiotics in treating common poultry diseases, particularly Salmonella which is a foodborne pathogen in humans. This study investigates the effects of ethnoveterinary supplementation of Rauwolfia serpentina (L. Benth. ex Kurz) powder (RSP) on three key immune-related genes; Suppressor of cytokine signaling 3 (SOCS3), the quiescence-related gene P20K (P20K), and the major histocompatibility complex Class IIβ (MHC class IIβ), gut morphology and growth performance of broiler chicks infected with Salmonella Gallinarum. Methods: Two hundred and forty day-old Hubbard classic chickens were randomly assigned to four groups: non-challenged control (NC), and Salmonella Gallinarum challenge group (SGC), and two treatment groups fed a basic diet supplemented with 1.5% Rauwolfia serpentina powder (RSP) with SGC (RSP-1) and 3% RSP with SGC (RSP-2), respectively, from day 3 till 28 days of age. Each treatment was replicated 4 times with 15 bird/replicate pen. On day 7, all the birds in the RSP-1, RSP-2 and SGC groups received 1 ml of BHI broth containing 2 × 108 CFU of Salmonella Gallinarum via oral gavage. While control birds received an equivalent volume of sterile BHI broth. Gene expression analysis was conducted using real-time PCR to measure the expression of key immune-related genes: SOCS3, P20K, and MHC Class IIβ in spleen, liver, and caeca. Additionally, histopathological assessments of gut and growth performance parameters including feed intake, body weight gain, and feed conversion ratio (FCR) were monitored throughout the experimental period. Result: The gene expression analysis at 3 and 21 days post-challenge revealed that SGC birds had significantly higher SOCS3, P20K, and lower MHC class IIβ expression (p < 0.001) in the caecum, liver, and spleen of broiler chickens. In contrast, the RSP-1 and RSP-2 groups showed significantly lower SOCS3 and P20K expression (p < 0.001), alongside improved gut morphology, weight gain, and FCR compared to the SGC group, with these benefits increasing over time. Conclusion: In conclusion, these findings suggest that Rauwolfia serpentina supplementation modulates key immune-related gene expression (SOCS3, P20K, and MHC class IIβ), enhances intestinal health, and improves growth performance in broilers challenged with Salmonella Gallinarum. Mangifera indica peels are a rich source of diverse flavonoids and xanthonoids; however, generally these are discarded. Computational studies revealed that mangiferin significantly interacts with amino acid residues of transcriptional regulators 1IK3, 3TOP, and 4f5S. The methanolic extract of Langra variety of mangoes contained the least phenol concentrations (22.6 ± 0.32 mg/gGAE [gallic acid equivalent]) compared to the chloroform (214.8 ± 0.12 mg/gGAE) and ethyl acetate fractions (195.6 ± 0.14 mg/gGAE). Similarly, the methanolic extract of Sindhri variety contained lower phenol concentrations (42.3 ± 0.13 mg/gRUE [relative utilization efficiency]) compared with the chloroform (85.6 ± 0.15 mg/gGAE) and ethyl acetate (76.1 ± 0.32 mg/gGAE) fractions. Langra extract exhibited significant α‐glucosidase inhibition (IC50 0.06 mg/mL), whereas the ethyl acetate fraction was highly active (IC50 0.12 mg/mL) in Sindhri variety. Mangiferin exhibited significant inhibition (IC50 0.026 mg/mL). A moderate inhibition of 15‐LOX was observed in all samples, whereas mangiferin was least active. In advanced glycation end product inhibition assay, the chloroform fraction of Langra variety exhibited significant inhibition in nonoxidative (IC50 64.4 μg/mL) and oxidative modes (IC50 54.7 μg/mL). It was concluded that both Langra and Sindhri peel extracts and fractions possess significant antidiabetic activities. Purpose: To determine the cytotoxic and antiviral potentials of Euphorbia milii var. splendens leaf against Peste des petits ruminants virus (PPRV). Methods: The methanol extract, as well as n-hexane, chloroform, ethyl acetate, and n-butanol fractions of E. milii leaves were screened for cytotoxic and antiviral activities against PPRV using Vero cell line and 3-(4,5-dimethylthiazol-2-yl)-2, diphenyltetrazolium bromide (MTT) assay. Results: Non-cytotoxic concentrations with cell survival rate (CSP) greater than 50 % were considered virucidal. Methanol extract and fractions produced significant (p < 0.05) effects at all test concentrations against PPRV. The 50 % cytotoxic concentration (CC50) calculated was ≤ 25 µg/mL for extract and fractions. In antiviral assay, ethyl acetate, n-hexane, and n-butanol fractions at all test concentrations ranging from 1.56 - 800 µg/mL were non-virucidal; even at their non-cytotoxic concentrations these fractions did not show antiviral activities. However, the methanol extract and its chloroform fractions showed significant (p < 0.05) virucidal potential. This study scrutinized the phytochemical composition, curative, hepato and nephro protective effect of different doses of lyophilized juice of Citrus reticulata fruit against paracetamol induced toxicity. Phytochemical screening and RP-HPLC analysis were conducted to quantify total polyphenols and flavonoids respectively. For evaluation of in vivo curative and protective effects, thirty six rats were randomly divided into six groups. In first four groups 1, II, III and IV paracetamol 75mg/kg, i.p, 150mg/kg, 250mg/kg and 500mg/kg p.o doses of lyophilized juice were administered to rats respectively. Blood samples were withdrawn at 0, 24, 48 and 72 hours in paracetamol treated rats. For screening of hepato and nephro protective effect Group V and VI were fed on lyophilized juice (250mg/kg and 500mg/kg p.o) for seven days and on 8 th day blood samples were collected at 0,24,48 and 72 hours. Hepatic and renal biomarkers were monitored. Phytochemical analysis revealed the presence of total polyphenols (20.7±0.3GAEmg/g) and flavonoid contents (21.2±0.4QE mg/g). RP-HPLC also confirmed the presence of Myricetin, Quercetin and Kaempferol in fruit juice. The lyophilized juice at 500mg/kg dose have shown profound decrease in paracetamol induced elevated serum levels of liver and kidney functions, which suggests a possible therapeutic role of its constituents in hepatic and kidney malfunctions.
