31. N-acetyl-serotonin reduces copper (I) ion-induced lipid peroxidation in bovine retinal homogenates

Abstract

.Purpose:
Chalcosis is an ocular condition caused by penetration injury of copper or its alloy, which leads to extensive ocular inflammation. N-acetyl-serotonin has recently been identified as a potent antioxidant against free radical stress. In this study, we determined the efficacy of N-acetyl-serotonin against the copper (I)-induced retinal lipid peroxidation.
Methods:
Copper (I)-treated (100 μM) bovine retinal homogenates were incubated with 6 different concentrations (i.e. 0.00, 0.25, 0.50, 1.00, 2.00 and 4.00 mM) of N-acetyl-serotonin or vitamin E. The malondialdehyde level was measured as an index of lipid peroxidation.
Results:
Copper (I) ions induced a significant dose-dependent increase in malondialdehyde (p=0.007). Co-incubation with N-acetyl-serotonin or vitamin E significantly suppressed the copper (I)-induced malondialdehyde production (p<0.0001). The concentration to inhibit 50% of damage for N-acetyl-serotonin and vitamin E were found to be 1.54 mM and 0.45 mM, respectively.
Conclusion:
Although N-acetyl-serotonin is only 29% as effective as vitamin E in suppressing the copper (I)-induced lipid peroxidation, the present study supports a pharmacological potential of N-acetyl-serotonin combating free radical oxidative damages in the ocular tissues.

Abstract

While SABR is known for its overall low toxicity and safety, there remains a research gap regarding its combined use with specific systemic therapies. This study aims to evaluate the toxicity of SABR in combination with various systemic therapies. The hypothesis is that certain systemic therapies would significantly increase the risk of Grade 2+ and Grade 3+ radiation therapy-related toxicities when used concurrently with Stereotactic Ablative Radiotherapy (SABR). A secondary analysis of the SABR-5 trial compared grade 2+ and 3+ toxicities associated with SABR until the last follow-up in patients receiving high-risk or non-high-risk systemic therapy at intervals of 3 months, 2 weeks, 1 week, and concurrently with SABR. High-risk systemic therapy was a priori defined, based on previous literature, as drugs that, when given close to SABR, may increase treatment toxicity. This category encompasses cytotoxic chemotherapy drugs, multi-targeted tyrosine kinase inhibitors, cyclin-dependent kinase 4/6 inhibitors, epidermal growth factor receptor inhibitors, anti-vascular endothelial growth factor agents, and anti-cytotoxic T-lymphocyte-associated protein 4 agents. Among the 381 patients, the actuarial rates of grade 2+ and 3+ toxic effects were as follows: for patients not on systemic therapy 3 months prior to SABR (n = 202), the rates were 17.3% and 3.5%, respectively; for patients on non-high-risk systemic therapy concurrent with SABR (n = 102), the rates were 18.6% and 3.9%, respectively; and for patients on high-risk systemic therapy concurrent with SABR (n = 5), the rates were notably higher at 60% and 40%, respectively. On multivariable analysis, concurrent use of high-risk systemic therapy was associated with a higher risk of grade 2+ (OR = 7.15, P = 0.043) or 3+ toxic effects (OR = 13.9, P = 0.015). Significance was not observed when high-risk drugs were used only within 1 week, 2 weeks, or 3 months of SABR, nor with the use of any non-high-risk drugs. A second adverse factor included increased tumor diameter (per 1 cm increment; G2+ OR = 1.25, P < 0.001; G3+ OR = 1.27, P = 0.015). High-risk drugs have demonstrated a potential of increased SABR-related toxicity, warranting caution in their concurrent use with SABR. In contrast, the combination of non-high-risk drugs with SABR may be safe. Ongoing efforts are essential to identify potential risks and uncertainties associated with this therapeutic combination. 35. Antibodies against deamidated gliadin as a novel serologic marker for celiac disease

Abstract

Introduction Recently it was shown that antibodies recognizing partially deamidated gliadin have higher sensitivity and specificity for celiac disease (CD) in comparison to antibodies recognizing native gliadin. The aim of the study was to assess the diagnostic performance of new ELISA test in which the antigen represents a repetitive motive of deamidated gliadin peptides (GAF-3X) and to compare it with anti-tTG test. Materials and methods Study included 56 pediatric patients (F/M=37/19 ; median age 11 years (range 7-15), for whom CD serology was requested. Anti-GAF-3X IgA and IgG together with anti-tTG IgA antibodies were measured in all sera using ELISA assays (Euroimmun, Lübeck, Germany). Manufacturer proposed cut off values were 20 RU/mL for anti- tTG and 25 RU/ml for GAF-3X IgA and IgG. Results Out of 56 patients 8 were positive for all 3 autoantibodies type, 1 for GAF-3X IgA and anti- tTG, 13 for anti-tTG and 1 for anti-GAF-3X IgA only. CD was biopsy confirmed in 14 patients of whom 7 were positive for all 3 autoantibodies type, 1 for anti-tTG and GAF-3X IgA and 2 for anti-tTG only. The residual 4 CD confirmed patients were on gluten free diet and 3 of them were seronegative while anti-tTG was positive in one. With cut off values proposed by the manufacturer sensitivity and specificity was 100% and 73, 8% for anti-tTG, 80, 0% and 95, 2% for anti-GAF-3X IgA and 70.0% and 97.6% for anti-GAF- 3X IgG. According to ROC analysis, sensitivity and specificity for anti-tTG IgA was 100% and 95.2%, for anti-GAF-3X IgA 90.0% and 92.9%, and for anti-GAF-3X IgG 70.0% and 97.6% RU/mL, respectively. Optimal cut-off value for tTG-IgA was 48.6 RU/mL, and 22 RU/mL for both anti-GAF-3X IgA and anti-GAF-3X IgG.
