Abstract

The Ewing sarcoma family of tumors (EFT) is a group of highly malignant small round blue cell tumors occurring in children and young adults. We report here the largest genomic survey to date of 101 EFT (65 tumors and 36 cell lines). Using a combination of whole genome sequencing and targeted sequencing approaches, we discover that EFT has a very low mutational burden (0.15 mutations/Mb) but frequent deleterious mutations in the cohesin complex subunit STAG2 (21.5% tumors, 44.4% cell lines), homozygous deletion of CDKN2A (13.8% and 50%) and mutations of TP53 (6.2% and 71.9%). We additionally note an increased prevalence of the BRCA2 K3326X polymorphism in EFT patient samples (7.3%) compared to population data (OR 7.1, p = 0.006). Using whole transcriptome sequencing, we find that 11% of tumors pathologically diagnosed as EFT lack a typical EWSR1 fusion oncogene and that these tumors do not have a characteristic Ewing sarcoma gene expression signature. We identify samples harboring novel fusion genes including FUS-NCATc2 and CIC-FOXO4 that may represent distinct small round blue cell tumor variants. In an independent EFT tissue microarray cohort, we show that STAG2 loss as detected by immunohistochemistry may be associated with more advanced disease (p = 0.15) and a modest decrease in overall survival (p = 0.10). These results significantly advance our understanding of the genomic and molecular underpinnings of Ewing sarcoma and provide a foundation towards further efforts to improve diagnosis, prognosis, and precision therapeutics testing.

Abstract

This thesis aims to study the pattern of settlement of the Chinese Community in the United Kingdom: taking samples mainly from the Chinese community in Tower Hamlets in East London. It examines in details the behavioural pattern and traditional values of the Chinese culture and the various sociological factors that may have influenced the Chinese people in their choice of places to settle. Various concepts and theories on ethnic minorities and the sociological issues of the Chinese community in London have been explored. I believe that it is impossible for us, as town planners, to plan for a society without a thorough understanding of the behaviour of its parts. The increased concern of planning (in housing and the local government generally) with ethnic minorities can be disastrous and dangerous if it is based on ill-defined stereotypes ignoring the great diversity of the various groups identified in the 1991 Census. One aim of the thesis is thus to explain some of the key features of one particular minority group - the Chinese. I believe it is an essential basis for better planning. I am interested in the Chinese community because with a population of around 57,000, it is the smallest ethnic minority group in the UK; hence very little attention has been paid to them. The reason for choosing Tower Hamlets is because it used to be home to the first Chinese immigrants. The London dock area around Tower Hamlets (as it is called today) was the first point of entry for a lot of the earlier Chinese settlers in the UK as many of them jumped ship and settled there.

Abstract

The effectiveness of the content-based image retrieval can be enhanced using heterogeneous features embedded in the images. However, since the features in texture, color, and shape are generated using different computation methods and thus may require different similarity measurements, the integration of the retrievals on heterogeneous features is a nontrivial task. In this paper, we present a semantics-based clustering and indexing approach, termed SemQuery, to support visual queries on heterogeneous features of images. Using this approach, the database images are classified based on their heterogeneous features. Each semantic image cluster contains a set of subclusters that are represented by the heterogeneous features that the images contain. An image is included in a semantic cluster if it falls within the scope of all the heterogeneous clusters of the semantic cluster. We also design a neural network model to merge the results of basic queries on individual features. A query processing strategy is then presented to support visual queries on heterogeneous features. An experimental analysis is conducted and presented to demonstrate the effectiveness and efficiency of the proposed approach. En los últimos años, Perú ha destacado como líder en la región por encima de Chile, Colombia y México (según las estimaciones del Banco Mundial) por su crecimiento sostenido basado en los buenos fundamentos económicos. Sin embargo, este crecimiento no se traduce en su Mercado de Capitales local, debido a distintos factores como son los culturales, altos costos, regulación inadecuada, poca profundidad, pocos emisores, etc. La Bolsa de Lima (única Bolsa de Valores en Perú) es principalmente minera y maneja bajos volúmenes de negociación, ello hace que sea muy volátil y que esté expuesta a algunos eventos críticos como por ejemplo: la caída de precios de metales o especulaciones de una posible reclasificación de mercado (de emergente a frontera). Por lo tanto, es importante que la gestión de una empresa de este sector gestione los recursos de forma eficiente para mitigar el impacto de situaciones críticas. El plan estratégico propuesto para Credicorp Capital Sociedad Agente de Bolsa S.A., al ser el principal intermediario de valores en la Bolsa de Valores de Lima, permitió analizar las principales dificultades para incrementar la participación de mercado, adquirir y desarrollar sistemas de tecnología de información para captar a nuevos clientes potenciales y retener el talento humano en la empresa. En la presente tesis se muestran los resultados del análisis externo e interno de Credicorp Capital Sociedad Agente de Bolsa S.A., además de la propuesta de objetivos a corto y largo plazo en función de la Visión y Misión formuladas en éste estudio. Finalmente, se concluye que los objetivos planteados son viables y se recomienda dar prioridad a los planes de acción de cambio tecnológico y reducción de costos en servicios tercerizados con la implementación de áreas propias o contratando a empresas de menor costo. In recent years, Peru has stood out as a leader in the region over Chile, Colombia and Mexico (according to World Bank estimates) for its sustained growth based on the good economic fundamentals. However, this growth does not translate into your local Capital Market, due to different factors such as cultural, high costs, inadequate regulation, shallow depth, few issuers, etc. The Lima Stock Exchange (the only Stock Exchange in Peru) is mainly mining and handles low trading volumes, which makes it very volatile and exposed to some critical events such as: the fall in metal prices or speculation of a possible market reclassification (from emerging to frontier). Therefore, it is important that the management of a company in this sector manages resources efficiently to mitigate the impact of critical situations. The proposed strategic plan for Credicorp Capital Sociedad Agent de Bolsa SA, being the main securities intermediary in the Lima Stock Exchange, allowed analyzing the main difficulties to increase market share, acquire and develop information technology systems to capture to new potential customers and retain human talent in the company. This thesis shows the results of the external and internal analysis of Credicorp Capital Sociedad Agent de Bolsa S.A., in addition to the proposal of short and long-term objectives based on the Vision and Mission formulated in this study. Finally, it is concluded that the proposed objectives are viable and it is recommended to give priority to action plans for technological change and cost reduction in outsourced services with the implementation of own areas or hiring lower cost companies. Abstract

We undertook a multidimensional clinical genomics study of children and adolescent young adults with relapsed and refractory cancers to determine the feasibility of genome-guided precision therapy.Patients with non-central nervous system solid tumors underwent a combination of whole exome sequencing (WES), whole transcriptome sequencing (WTS), and high-density single-nucleotide polymorphism array analysis of the tumor, with WES of matched germline DNA. Clinically actionable alterations were identified as a reportable germline mutation, a diagnosis change, or a somatic event (including a single nucleotide variant, an indel, an amplification, a deletion, or a fusion gene), which could be targeted with drugs in existing clinical trials or with FDA-approved drugs.Fifty-nine patients in 20 diagnostic categories were enrolled from 2010 to 2014. Ages ranged from 7 months to 25 years old. Seventy-three percent of the patients had prior chemotherapy, and the tumors from these patients with relapsed or refractory cancers had a higher mutational burden than that reported in the literature. Thirty patients (51% of total) had clinically actionable mutations, of which 24 (41%) had a mutation that was currently targetable in a clinical trial setting, 4 patients (7%) had a change in diagnosis, and 7 patients (12%) had a reportable germline mutation.We found a remarkably high number of clinically actionable mutations in 51% of the patients, and 12% with significant germline mutations.
