ABSTRACT The Hadal Zone is acknowledged for its extreme environmental conditions, especially high hydrostatic pressures. The dominant scavengers in the Hadal Zone, Hadal amphipods, fulfill vital roles in the Hadal food web and ecological niches. However, research on the gut microbiota of amphipods related to ecological functions and environmental adaptation is still limited. Here, we used 16S rRNA sequencing technology and a culture-dependent method to analyze the composition of the gut microbiota in Amphipoda living in the Mariana Trench. A total of 16 bacterial genera were identified. Among them, Firmicutes and Proteobacteria were the predominant phyla. The adaptability of gut probiotics to the environment was investigated. Pediococcus pentosaceus XY62 was picked up as the representative strain to elucidate the ecological functions of gut microbes in amphipods. The ProBio database and the K-B agar diffusion method indicated that P. pentosaceus XY62 exhibited the highest probiotic activity compared with all other isolated strains. Specific metabolic pathways and transporter systems that contribute to a range of environmental adaptation strategies have been revealed by genomic analysis of P. pentosaceus XY62. The environmental response genes and a specialized KDP transport system allow it to adapt to the challenging conditions of the Hadal Zone. In addition, the presence of antibacterial compounds and antibiotic resistance genes, as well as the ability to form a biofilm, facilitated the successful colonization of P. pentosaceus XY62 in the gut environment.IMPORTANCEAmphipods are widely distributed in the Hadal trenches, and the study of their gut microbes has garnered considerable scientific interest. Our research breaks away from traditional omics approaches, innovatively combining sequencing technologies with culture-dependent methods to analyze the gut microbiome structure of amphipods from the Mariana Trench. This not only complements the current omics-dominated field but also paves the way for future resource development of extreme microbes. Furthermore, by conducting genomic analyses and functional validations on a representative strain, we have uncovered its probiotic effects and strategies for adapting to extreme environments. This provides new insights into the theoretical study of the ecological functions of deep-sea bacteria. Overall, our findings offer a fresh perspective on the microbial community structure and environmental adaptation strategies of gut microorganisms in the Hadal Zone.

The extreme environment shapes fungi in deep-sea sediments with novel metabolic capabilities. The ubiquity of fungi in deep-sea habitats supports their significant roles in these ecosystems. However, there is limited research on the metabolic activities and adaptive mechanisms of filamentous fungi in deep-sea ecosystems. In this study, we investigated the biological activities, including antibacterial, antitumor and nematicidal activity of Purpureocillium lilacinum FDZ8Y1, isolated from sediments of the Mariana Trench. A key feature of P. lilacinum FDZ8Y1 was its tolerance to high hydrostatic pressure (HHP), up to 110 MPa. We showed that HHP affected its vegetative growth, development, and production of secondary metabolites, indicating the potential for discovering novel natural products from hadal fungi. Whole-genome sequencing of P. lilacinum FDZ8Y1 revealed the metabolic potential of this piezotolerant fungus in carbon (carbohydrate metabolism), nitrogen (assimilatory nitrate reduction and protein degradation) and sulfur cycling processes (assimilatory sulfate reduction). Transcriptomic analysis under elevated HHP showed that P. lilacinum FDZ8Y1 may activate several metabolic pathways and stress proteins to cope with HHP, including fatty acid metabolism, the antioxidant defense system, the biosynthetic pathway for secondary metabolites, extracellular enzymes and membrane transporters. This study provides valuable insights into the metabolic potential and adaptation mechanisms of hadal fungi to the challenging conditions of the hadal environment.

IntroductionNumerous observational studies have indicated that smoking is a substantial risk factor for esophageal cancer. However, there is a shortage of research that delves into the specific causal relationship and potential mediators between the two. Our study aims to validate the correlation between smoking-related traits and esophageal cancer while exploring the possible mediating effects of immune factors.MethodsInitially, we conducted bidirectional univariate Mendelian Randomization (MR) analyses to forecast the causal effects linking smoking-related traits and esophageal cancer. Subsequently, we employed a two-step MR analysis to scrutinize immune cell phenotypes that could mediate these effects. Finally, the coefficient product method was employed to determine the precise mediating impact. Additionally, we have refined our sensitivity analysis to ensure the reliability of the outcomes.ResultsAfter analysis, Smoking status: Never had a significant negative association with the incidence of esophageal cancer (inverse-variance weighted (IVW) method, p=1.82e-05, OR=0.10, 95%CI=0.04~0.29). Ever smoked (IVW, p=1.49e-02, OR=4.31, 95%CI=1.33~13.94) and Current tobacco smoking (IVW, p=1.49e-02, OR=4.31, 95%CI=1.33~13.94) showed the promoting effect on the pathogenesis of esophageal cancer. Through further examination, researchers discovered 21 immune cell phenotypes that have a causal relationship with esophageal cancer. After careful screening, two immune cell phenotypes were found to have potential mediating effects. In particular, it was observed that in the case of the preventive effect of Smoking status: Never on esophageal cancer, the absolute count of CD62L plasmacytoid dendritic cells mediated a reduction of 4.21%, while the mediating effect of CD27 in CD20-CD38-B cells was -4.12%. In addition, sensitivity analyses did not reveal significant heterogeneity or level pleiotropy.ConclusionThe study provides new evidence for the causal relationship between smoking-related features and esophageal cancer and proposes immune factors with potential mediating effects. However, this finding needs to be further demonstrated by more extensive clinical studies.

Abstract With the explosive growth of online services, the Internet Data Center (IDC) has become an emerging load demand . The power consumption of IDCs are believed suitable for demand response (DR) purposes due to its temporal‐spatial flexibility properties. To what extent IDCs could be used as candidate DR resources to provide capacity support are dependent both on the technical property and the willingness of data end‐users . To address the above issue, this paper presents a methodological framework for quantifying the potential value of IDC‐based DR in distribution grids. To achieve this, the concept of capacity credit (CC) is introduced and extended into the IDC scenario. The proposed framework explicitly considered the impacts of uncertainty of user participation intention and the reliability of basic information in the DR process. A novel Z‐number based price elasticity uncertainty model is introduced, and a more realistic evaluation of IDC capacity credit is obtained. Finally, the effectiveness of the proposed model and method is illustrated on a modified IEEE‐33 node network, and the obtained results verify the significance of IDC‐based DR in enhancing the adequacy of supply in distribution grids.

Successful gene therapy for brain tumors are often limited by two important factors, the existence of blood brain barrier (BBB) and inefficient transfection of brain tumor cells. In this study, we designed a series of peptide-based gene delivery vectors decorated with T7 segment for binding the transferrin (Tf) receptors which were highly expressed on brain tumor cells, and evaluated their ability of gene delivery. The physicochemical properties of peptide vectors or peptide/DNA complexes were studied as well. The in vitro transfection efficiency was investigated in normal and glioma cell lines. Among these complexes, PT-02/DNA complexes showed the highest transfection efficiency in glioma cells and low cytotoxicity in normal cell lines, and it could transport DNA across the BBB model in vitro. Furthermore, PT-02/DNA could deliver pIRES2-EGFP into the brain site of zebrafish in vivo. The designed peptide vectors offered a promising way for glioma gene therapy.

Arctium lappa L. is a prevalent medicinal herb and a health supplement that is commonly used in Asia. Over the last few decades, the bioactive component arctigenin has attracted the attention of researchers because of its anti-inflammatory, antioxidant, immunomodulatory, multiple sclerosis fighting, antitumor, and anti-leukemia properties. After summarising the research and literature on arctigenin, this study outlines the current status of research on pharmacological activity, total synthesis, and structural modification of arctigenin. The purpose of this study is to assist academics in obtaining a more comprehensive understanding of the research progress on arctigenin and to provide constructive suggestions for further investigation of this useful molecule.

Abstract Background X-linked hyper-immunoglobulin M (XHIGM), a primary immunodeficiency syndrome caused by mutations in the CD40 ligand gene(CD40LG), presents with recurrent respiratory infections in pediatric patients. We aimed to evaluate the spectrum of clinical features and respiratory pathogens in pediatric patients with XHIGM in China. Methods We retrospectively reviewed seven pediatric patients who were diagnosed with XHIGM and received follow-up treatment at the Guangzhou Women and Children’s Medical Center between January 2010 and January 2021. We determined their clinical characteristics, causative pathogens, and prognosis by performing peripheral immunological and genetic tests. Results There were seven boys with age ranging from 4–20 months (median age, 13 months). Four of the seven respiratory infections were caused by Talaromyces marneffei(T. marneffei). Two patients had viral infections caused by cytomegalovirus (CMV) and human adenovirus respectively. One patient had a mixed infection caused by Pneumocystis carinii and CMV. Except for one child who died of respiratory failure, one patient received hematopoietic stem cell transplantation (HSCT) and recovered well, the other five patients survived with regular infusions of intravenous immunoglobulin (IVIg) during the follow-up period. Six patients had reduced antibody levels, especially IgG, IgA, and IgE levels. Increased serum IgM levels were detected in four cases, and three cases presented normal IgM levels at onset. All children were diagnosed with XHIGM with CD40LG variation. Three novel mutations were identified in the present study. Conclusions Our study suggests that respiratory infections usually begin within 2 years old, fungi and viruses are important pathogens causing respiratory infections in children with XHIGM. In endemic areas, T. marneffei is the common pathogen of respiratory tract infection in children with the disease.

The Altyn Tagh fault has been a crucial tectonic boundary of the Tibetan Plateau during the Cenozoic India-Eurasia collision. However, issues have not been addressed regarding the Cenozoic evolution of the kinematic transformation from the eastern Altyn Tagh fault to the Qilian Shan. Here we focus on the kinematics at a crucial point, the Subei triple junction, along the Altyn Tagh fault, which was recorded by faulting in the Suganhu basin to the south of the junction. We reconstructed the structural pattern of faults and thickness distribution of the Cenozoic strata in the Suganhu basin by integrating seismic profiles, well logging, and topographic data. We inferred that only crustal shortening and thickening in the Danghenan Shan, a prominent topographic high, absorbed the strike-slip displacement along the Altyn Tagh fault during the early Cenozoic. Since the mid-Miocene, strike-slip fault belts within the Suganhu basin were initiated, based on the fault geometry and uneven thickness distribution across the fault belts. We thus proposed a mid-Miocene kinematic transformation realized by blocks extruding southeastward, as well as the crustal shortening and thickening in the entire Qilian Shan. Those blocks are bounded by preexisting weaknesses with lateral movements, and lithospheric heterogeneity played an essential role in the block-scale extrusion.

Deep-sea fungi have evolved extreme environmental adaptation and possess huge biosynthetic potential of bioactive compounds. However, not much is known about the biosynthesis and regulation of secondary metabolites of deep-sea fungi under extreme environments. Here, we presented the isolation of 15 individual fungal strains from the sediments of the Mariana Trench, which were identified by internal transcribed spacer (ITS) sequence analysis as belonging to 8 different fungal species. High hydrostatic pressure (HHP) assays were performed to identify the piezo-tolerance of the hadal fungi. Among these fungi, Aspergillus sydowii SYX6 was selected as the representative due to the excellent tolerance of HHP and biosynthetic potential of antimicrobial compounds. Vegetative growth and sporulation of A. sydowii SYX6 were affected by HHP. Natural product analysis with different pressure conditions was also performed. Based on bioactivity-guided fractionation, diorcinol was purified and characterized as the bioactive compound, showing significant antimicrobial and antitumor activity. The core functional gene associated with the biosynthetic gene cluster (BGC) of diorcinol was identified in A. sydowii SYX6, named as AspksD. The expression of AspksD was apparently regulated by the HHP treatment, correlated with the regulation of diorcinol production. Based on the effect of the HHP tested here, high pressure affected the fungal development and metabolite production, as well as the expression level of biosynthetic genes which revealed the adaptive relationship between the metabolic pathway and the high-pressure environment at the molecular level.

Objective To compare the clinical efficacy and radioactivity of the bridge‐type zero‐profile anchored spacer (ROI‐C) interbody fusion cage and anterior cervical discectomy and fusion with plating and cage system (ACDF) for cervical spondylotic myelopathy (CSM). Methods This is a retrospective contrastive study. We recruited 35 patients who received ROI‐C (ROI‐C group) and 34 patients who received ACDF (ACDF group), between January 2014 to January 2019, at our treatment center. The ROI‐C group comprised of 11 males and 24 females with a mean age of 61.59 ± 8.21 years (range, 51–71 years). The ACDF group comprised of 12 males and 22 females with a mean age of 60.15 ± 7.52 years (range, 52–74 years). Neck Disability Index (NDI), Japanese Orthopaedic Association score (JOA), Odom's score, cervical Cobb angle, fusion rate, adjoining ossification, and dysphagia. Results A total of 69 patients met the inclusion criteria, and these patients received more than two years of follow‐up. There were significant differences in surgical duration (101 ± 22 min vs. 118 ± 29 min) and blood loss (102 ± 46 ml vs. 145 ± 58 ml) between two groups (P 0.05). Conclusion Both ROI‐C and ACDF achieved good therapeutic effects. However, ROI‐C can reduce the operation time and postoperative complications.

As space resources become increasingly constrained, the major space-faring nations are establishing large space target monitoring systems. There is a demand for both the number and the detection capability of space-based optical monitoring equipment. The detection range (i.e., field of view) and parasitic capability (lightweight and small size) of a single optical payload will largely reduce the scale and cost of the monitoring system. Therefore, in this paper, the optic-mechanical system of an ultra-lightweight and ultra-compact space camera and the optical alignment method are investigated around a fully freeform off-axis triple-reversal large field of view (FOV) optical system. The optic-mechanical system optimisation design is completed by adopting the optic-mechanical integration analysis method, and the weight of the whole camera is less than 10 kg. In addition, to address the mounting problems caused by the special characteristics of the freeform surface optical system, a dual CGH coreference alignment method is innovatively proposed. The feasibility of the method is verified by the mounting and testing test, and the test results show that the system wavefront difference is better than 1/10 λ. The imaging test of the space camera and the magnitude test results meet the design requirements of the optical system. The optic-mechanical system design method and alignment method proposed in this paper are instructive for the design and engineering of large field of view full freeform optical loads.

Abstract In this research the interlayer contact condition was considered between the adjacent layers of orthotropic steel deck pavement, and an interface contact bonding model was applied to simulate the interlayer bonding condition and evaluate the response of deck pavement under vehicle loads. An advantage of this model is that it can simulate not only the full-bond condition but also the debonding condition at somewhere between adjacent layers. The responses of the orthotropic steel deck pavement were calculated and analyzed by the model, and it found that this model is reasonable and credible to evaluate the responses of the deck pavement comparing with the previous researches. The full-bond condition was an ideal condition between adjacent layers, which was prone to underestimate the responses and deformation of the deck pavement. Moreover, the position and size of the disengaging area have a notable influence on the tensile strain at the top of SMA layer and the bottom of GA layer, and the tensile strain of them also increase with the increase of the disengaging area. Finally, the responses of the steel deck pavement changed obviously when the vehicle speed increase, so the suitable speed limit may reduce the responses and deformation for prolonging the service life of the orthotropic steel deck pavement.

Abstract Background To investigate the expression and clinical significance of EFNA1 in broad-spectrum tumors, and to evaluate its relationship with prognosis and biological functions of esophageal carcinoma (ESCA). Methods EFNA1 expression in various cancers was analyzed according to the data in the TCGA database. The clinical data were integrated, to analyze the relationship with ESCA clinical parameters and prognosis, and EFNA1 expression in ESCA tissue samples was detected by immunohistochemistry (IHC). Based on bioinformatics, the functional background of EFNA1 overexpression was analyzed. EFNA1 knockout cell model was established by EFNA1-shRNA transfecting ESCA cells, and the effect of knocking down EFNA1 on the proliferation of ESCA cells was detected by MTT. Results Among 7563 samples from TCGA, the EFNA1 gene highly expressed in 15 samples with common cancers and endangered the prognosis of patients with tumors. Its overexpression in ESCA and its influence on the prognosis were most significant. EFNA1 expression in 80 samples with ESCA and their paired samples was tested by IHC to verify its high expression (paired t test, P < 0.001) in ESCA tissues. It was found that EFNA1 expression was related to clinical factors (TNM staging, P = 0.031; lymph node metastasis, P = 0.043; infiltration, P = 0.016). Meanwhile, EFNA1 was found to be an independent risk factor based on the COX multi-factor analysis. And to further explore the importance of EFNA1 in tumors, EC-9706 and ECA109 cells were screened from 8 ESCA-related cell lines to build EFNA1 knockdown cell models. The results showed that EFNA1 knockdown significantly inhibited the proliferation of tumor cells (P < 0.05). In terms of molecular mechanism, EFNA1 related genes were significantly enriched in the proliferative pathway according to the pathway enrichment analysis. It was found that knocking down EFNA1 did inhibit cell proliferation based on cell experiments. Conclusions EFNA1 overexpression in ESCA tissue is related to the prognosis of patients. Knocking down EFNA1 can significantly inhibit the proliferation of ESCA cells.

With the continuous improvement of the imaging quality requirement of the space optical system, the large-aperture mirror becomes the research focus. However, the increase of the aperture will increase the whole weight which results in high launch cost and degrades the mirror surface figure accuracy. Therefore, the lightweight design method of the mirror structure is of great importance. In recent years, many space telescope system schemes have demonstrated the progress of the structural lightweight design of mirrors, such as Spitzer, SOFIA, JWST, etc. This article reviews the main content and innovations of the research on the structural designs of mirrors including conventional machining designs and topology optimization structures. Meanwhile, some emerging designs (e.g., lattices and Voronoi structures) considering additive manufacturing (AM) are also introduced. Several key elements of different structural design approaches for lightweight mirrors are discussed and compared, such as material, lightweight ratio, design methods, surface figure, etc. Finally, future challenges, trends, and prospects of lightweight design for mirrors are discussed. This article provides a reference for further related research and engineering applications.

Despite an increasing appreciation in the importance of host–microbe interactions in ecological and evolutionary processes, information on the gut microbial communities of some marine mammals is still lacking. Moreover, whether diet, environment, or host phylogeny has the greatest impact on microbial community structure is still unknown. To fill part of this knowledge gap, we exploited a natural experiment provided by an aquarium with belugas (Delphinapterus leucas) affiliated with family Monodontidae, Pacific white-sided dolphins (Lagenorhynchus obliquidens) and common bottlenose dolphin (Tursiops truncatus) affiliated with family Delphinidae, and Cape fur seals (Arctocephalus pusillus pusillus) affiliated with family Otariidae. Results show significant differences in microbial community composition of whales, dolphins, and fur seals and indicate that host phylogeny (family level) plays the most important role in shaping the microbial communities, rather than food and environment. In general, the gut microbial communities of dolphins had significantly lower diversity compared to that of whales and fur seals. Overall, the gut microbial communities were mainly composed of Firmicutes and Gammaproteobacteria, together with some from Bacteroidetes, Fusobacteria, and Epsilonbacteraeota. However, specific bacterial lineages were differentially distributed among the marine mammal groups. For instance, Lachnospiraceae, Ruminococcaceae, and Peptostreptococcaceae were the dominant bacterial lineages in the gut of belugas, while for Cape fur seals, Moraxellaceae and Bacteroidaceae were the main bacterial lineages. Moreover, gut microbial communities in both Pacific white-sided dolphins and common bottlenose dolphins were dominated by a number of pathogenic bacteria, including Clostridium perfringens, Vibrio fluvialis, and Morganella morganii, reflecting the poor health condition of these animals. Although there is a growing recognition of the role microorganisms play in the gut of marine mammals, current knowledge about these microbial communities is still severely lacking. Large-scale research studies should be undertaken to reveal the roles played by the gut microbiota of different marine mammal species.

Trabecular bone is natural material with heterogeneous tissue properties. The effect of tissue heterogeneity on the micromechanical behavior of trabecular bone is commonly evaluated by microCT-based finite element (microFE) analysis. Results from prior work remain inconclusive and lack of experimental validation. To address these issues, we combined microFE analysis with mechanical testing and microCT-based digital volume correlation (DVC), as a validation for the microFE approach. Porcine trabecular specimens were tested in compression as sequential microCT scans were taken. DVC was performed to extract “realistic” boundary conditions that were applied to microFE models, and to measure microstructural deformation and strain of the trabecular specimens. Heterogeneous and homogeneous microFE models of each trabecular specimen were created and compared with the experimentally measured microstructural displacement and strains. Results showed strong correlations between DVC-measured and microFE-predicted trabecular displacement and strain fields (R2 ​> ​0.9, p ​

Background: Asthma can be exacerbated by many triggers, and the heterogeneity of asthma triggers is clear among children with asthma. This study describes asthma triggers using a large-scale electronic dataset from the smartphone-based Chinese Children's Asthma Action Plan (CCAAP) app and aims to examine the difference in asthma triggers among different subgroups of children with asthma.Methods: Data from the smartphone-based CCAAP app between February 22, 2017, and November 23, 2020, were reviewed, and children with asthma who reported their asthma triggers were enrolled. Eight common asthma triggers were listed in the software: upper respiratory infection (URI), allergen sensitization, exercise, emotional disturbances, pungent odors, air pollution/smog, weather change, and tobacco smoke. Business travel planning decisions based on big data analysis are an important part of controlling travel dynamics and developing travel specifications.Based on the information of the national network business travel information,aiming at the optimization of the travel mode during the travel process and the personal preference of hotel accommodation,a user portrait framework technology was constructed to realize the rapid and accurate identification of sensitive customer groups.Firstly,the user's sensitivity was predicted by two-channel modeling for different characteristics of users.Secondly,users were characterized by business approval,travel control,hotel evaluation,time characteristics,numerical characteristics,etc.,and users’ multi-source feature system was constructed.Finally,business travel data was fully utilized.Multi-source,create a multi-view fusion model based on double-layer XGBoost,improve classification accuracy,and verify the effectiveness of the method by comparing experiments.

A general coupled KdV equation, which describes the interactions of two long waves with different dispersion relation, is considered. By employing the Hirota’s bilinear method, the bilinear form is obtained, and the one-soliton solution and two-soliton solution are constructed. Moreover, the elasticity of the collision between two solitons is proved by analyzing the asymptotic behavior of the two-soliton solution. Some figures are displayed to illustrate the process of elastic collision.

In this paper, a novel methodological framework for energy hub (EH) planning, considering the correlation between renewable energy source (RES) and demand response (DR) uncertainties, is proposed. Unlike other existing works, our study explicitly considers the potential correlation between the uncertainty of integrated energy system operations (i.e., wind speed, light intensity, and demand response). Firstly, an EH single-objective interval optimization model is established, which aims at minimizing investment and operation costs. The model fully considers the correlation between various uncertain parameters. Secondly, the correlation between uncertainties is dealt with by the interval models of multidimensional parallelism and affine coordinate transformation, which are transformed into a deterministic optimization problem by the interval order relationship and probability algorithm, and then solved by a genetic algorithm. Finally, an experimental case is analyzed, and the results show that the research method in this paper has good engineering practicability. At the same time, different correlations among uncertainties have different influences on integrated energy system planning. Correlation and influence are positively correlated.

This paper proposes a comprehensive methodological framework to investigate the potential role of the grid-connected battery swapping station (BSS) with vehicle-to-grid (V2G) capability in improving the reliability of supply in future distribution networks. For this aim, we first develop an empirical model for describing the energy demand of electric vehicles (EVs) and their resultant available generation capacity (AGC) that can be utilized for BSS operation. Then, on this basis, a quantitative method to quantify the effect of grid-connected BSS on distribution system reliability is proposed. In order to capture the uncertainties associated with EV users’ behaviors, Latin Hypercube Sampling (LHS) methods were utilized to obtain the time series of the BSS traffic flow and initial State of Charge (SOC) of each EV battery, according to the probability distribution of corresponding uncertain factors whose statistics are obtained from real-world historical data. Compared with existing works in this research field, the main contributions of this paper are threefold. (i) A comprehensive and efficient method to assess the reliability benefits of BSS with an explicit consideration of BSS characteristics (including physical structure, charging strategy, and swapping model) is proposed, which is in contrast to most of the extant studies that only focus on the EV fast-charging paradigm and thus provide a practical tool to analyze the potential value of BSS resources in future distribution systems. (ii) The randomness of EV user behaviors in BSS operation is explicitly modeled and considered. (iii) The LHS-based sequential simulation is used to improve the accuracy and convergence performance of the evaluation, as compared to the traditional Sequential Monte Carlo Simulation (SMCS) method. To verify the effectiveness of the proposed approach, numerical studies are conducted based on a modified IEEE 33-bus distribution network. The simulation results show that with V2G capabilities, BSS can improve reliability to a certain extent and reduce the adverse impact on the reliability of the distribution network. In addition, EV resources should be orderly managed and exploited; otherwise, uncoordinated charging activities could impose a negative impact on the reliability performance of distribution networks. Finally, it is also shown that under the same sampling time, LHS-based sequential simulation could be better than SMCS in the accuracy and convergence speed of the procedure.

Topology optimization has developed rapidly in the past three decades; as a creative and efficient optimization technique, it has been applied in engineering fields of aerospace and mechanical. However, there are a few attempts in bridge form design. In this paper, the parametric level set method is utilized to solve the form finding of arch bridges. The optimization model for minimizing the structural compliance under the volume constraint is built. Three numerical examples of form finding of arch bridges are studied. Results show that the optimal structures which have well-distributed stress and smooth force transmission are almost identical with the actual forms of arch bridges. The optimal forms can be treated as alternatives in the preliminary design stage, and topology optimization has a bright prospect in form finding of arch bridges.
