Abstract Background Citrus reticulata Blanco ‘Orah’ is one of the most widely grown citrus varieties in southern China. Objective: This study investigates the effects of dwarfing treatment on the appearance and postharvest quality of banana fruits. Methods: The experimental material used in this study was 'Guijiao No.9'. A dwarfing agent, primarily composed of uniconazole and potassium dihydrogen phosphate, was dissolved in water and applied to the roots. The banana seedlings undergo their first treatment with the dwarfing agent when they reach the 18th leaf, followed by a second treatment when they reach the 23rd leaf. The fruits were harvested once they reach the desired level of commercial maturity. Fruit appearance (peduncle length, fruit finger circumference) and postharvest quality, including peel color (value L*, a*, b*, C*, h*), fruit hardness, respiration rate, ethylene release, soluble sugar content (fructose, sucrose, glucose), and organic acid content (oxalic acid, quinine acid, malic acid, succinic acid, VC, citric acid, fumaric acid) in the flesh were taken as reference indicators to study the effects of dwarfing treatment on the storage quality of banana fruits. Abstract Background The epidemiological distribution of functional mitral regurgitation (FMR) in heart failure (HF) and mildly reduced ejection fraction (HFmrEF) patients and its impact on outcomes remains unclear. We attempt to investigate the prognosis of FMR in patients with HFmrEF. Methods The HF center registry study is a prospective, single, observational study conducted at the Second Affiliated Hospital of Shenzhen University, where 2330 patients with acute HF (AHF) were enrolled and 890 HFmrEF patients were included in the analysis. The patients were stratified into three categories based on the severity of FMR: none/mild, moderate, and moderate-to-severe/severe groups. Subsequently, a comparison of the clinical characteristics among these groups was conducted, along with an assessment of the incidence of the primary endpoint (comprising all-cause mortality and readmission for HF) during a one-year follow-up period. Results The one-year follow-up results indicated that the primary composite endpoint occurrence rates in the three groups were 23.5%, 32.9%, and 36.5%, respectively. The all-cause mortality rates in the three groups were 9.3%, 13.7%, and 16.4% respectively. Abstract Background The gibberellic acid (GA) inhibitor, uniconazole, is a plant growth regulator commonly used in banana cultivation to promote dwarfing but also enhances the cold resistance in plants. However, the mechanism of this induced cold resistance remains unclear. Results We confirmed that uniconazole induced cold tolerance in bananas and that the activities of Superoxide dismutase and Peroxidase were increased in the uniconazole-treated bananas under cold stress when compared with the control groups. The transcriptome and metabolome of bananas treated with or without uniconazole were analyzed at different time points under cold stress. Compared to the control group, differentially expressed genes (DEGs) between adjacent time points in each uniconazole-treated group were enriched in plant-pathogen interactions, MAPK signaling pathway, and plant hormone signal transduction, which were closely related to stimulus-functional responses. Furthermore, the differentially abundant metabolites (DAMs) between adjacent time points were enriched in flavone and flavonol biosynthesis and linoleic acid metabolism pathways in the uniconazole-treated group than those in the control group. Temporal analysis of DEGs and DAMs in uniconazole-treated and control groups during cold stress showed that the different expression patterns in the two groups were enriched in the linoleic acid metabolism pathway. In addition to strengthening the antioxidant system and complex hormonal changes caused by GA inhibition, an enhanced linoleic acid metabolism can protect cell membrane stability, which may also be an important part of the cold resistance mechanism of uniconazole treatment in banana plants. Conclusions This study provides information for understanding the mechanisms underlying inducible cold resistance in banana, which will benefit the production of this economically important crop.

【Objective】The study was conducted to screen plant growth regulators that could shorten the plant height of banana without affecting the yield, and explore the dwarfing effects of their application times on banana plants. Furthermore, appropriate concentrations of uniconazole were detected for identifying the changes in the activities of several defensive antioxidant enzymes, and the contents of soluble protein, potassium, calcium, magnesium and phosphorus in banana leaves, to further analyze the positive auxiliary effect of uniconazole on banana plant growth in addition to dwarfing effect, which would provide references for the safe and rational use of plant growth regulators in banana production.【Method】The dwarfing effects of several plant growth regulators including uniconazole (XX), paclobutrazol (DX), chlormequat chloride (AZ), mepiquat (JP), compound agents 0802 and 0501 on banana plants were evaluated by root spraying method. The contents of polyphenol oxidase (PPO), superoxide dismutase (SOD), catalase (CAT), phenylalanine ammonia lyase (PAL), soluble protein, potassium, calcium, magnesium and phosphorus in banana leaves were determined after 15, 20 and 25 days of 0.3 g/plant XX treatment.【Result】According to the evaluation standard of dwarfing effect, banana plant could grow normally in the 3-5 level of internode distance after dwarfing. Under one dwarfing treatment, the banana plant heights of XX1, DX1, 0501, XX3, JP2, 0802, AZ1 and JP4 treatments were decreased by 11.67-24.00 cm. The banana plant heights of JP2, DX1, JP4, 0802, AZ1, 0501 and XX3 under two dwarfing treatments were decreased by 29.67-39.00 cm with normal growth. It found that the banana plant height was shortened with the increase of the concentrations and application times of these grow regulators. XX3, 0802 and 0501 treatments could promote the early bud shooting of banana, and the duration periods were 8-19 days earlier than that of the control; The plant shooting was neat. The shooting rate of XX3 under two drarfing and 0802 treatment was 62.01% and 89.32%. The yield per plant showed decreasing trends with the increase of XX concentration. The plant yields of banana were 27.26, 25.62 and 23.45 kg for the one time treatment of XX1, XX3 and XX5, respectively. Of which, the yields per plant of XX3 and XX5 treatment were significantly decreased compared with that of the control (27.45 kg). The yields per plant of banana treated with 0802 and 0501 were 27.37 and 26.99 kg, respectively, and there was no significant difference with that of the control. It was found that the yield performance of banana plants was similar under the two dwarfing treatments of different plant grow regulators. Under the treatment with low concentrations of chlormequat chloride and paclobutrazol, the early shooting of banana could be promoted. Whereas the shooting of banana was delayed when the treating concentration was above 0.2 g per plant, and the plant yield was greatly affected or even reduced. The uniconazole treatment could increase the activities of PPO, CAT and PAL in banana leaves as well as the contents of soluble protein, calcium, magnesium, phosphorus and potassium, and enhance plant resistance.【Conclusion】It is recommended to use uniconazole, compound agents 0802 and 0501 as banana dwarfing agents with concentration ranging of 0.2-0.3 g/plant in banana production, and the newly planted banana could be dwarfed once. Banana is more susceptible to paclobutrazol (DX) and chlormequat chloride (AZ), which are not recommended.

Background Fusarium wilt, caused by Fusarium oxysporum f. sp. cubense race 4 (Foc4), is the most lethal disease of bananas in Asia. Methods To better understand the defense response of banana to Fusarium wilt, the transcriptome and metabolome profiles of the roots from resistant and susceptible bananas inoculated with Foc4 were compared. Results After Foc4 inoculation, there were 172 and 1,856 differentially expressed genes (DEGs) in the Foc4-susceptible variety (G1) and Foc4-resistant variety (G9), respectively. In addition, a total of 800 DEGs were identified between G1 and G9, which were mainly involved in the oxidation-reduction process, cell wall organization, phenylpropanoid biosynthesis, and lipid and nitrogen metabolism, especially the DEGs of Macma4_08_g22610, Macma4_11_g19760, and Macma4_03_g06480, encoding non-classical arabinogalactan protein; GDSL-like lipase; and peroxidase. In our study, G9 showed a stronger and earlier response to Foc4 than G1. As the results of metabolomics, lipids, phenylpropanoids and polyketides, organic acids, and derivatives played an important function in response to Fusarium wilt. More importantly, Macma4_11_g19760 might be one of the key genes that gave G9 more resistance to Foc4 by a lowered expression and negative regulation of lipid metabolism. This study illustrated the difference between the transcriptomic and metabolomic profiles of resistant and susceptible bananas. These results improved the current understanding of host-pathogen interactions and will contribute to the breeding of resistant banana plants.

Abstract Background Uniconazole is an effective plant growth regulator that can be used in banana cultivation to promote dwarfing and enhance lodging resistance. However, the mechanisms underlying banana dwarfing induced by uniconazole are unknown. In uniconazole-treated bananas, gibberellin (GA) was downregulated compared to the control groups. An integrative analysis of transcriptomes and metabolomes was performed on dwarf bananas induced by uniconazole and control groups. The key pathways involved in uniconazole-induced dwarfism in banana were determined according to the overlap of KEGG annotation of differentially expressed genes and (DEGs) differential abundant metabolites (DAMs). Results Compared with the control groups, the levels of some flavonoids, tannins, and alkaloids increased, and those of most lipids, amino acids and derivatives, organic acids, nucleotides and derivatives, and terpenoids decreased in uniconazole-treated bananas. Metabolome analysis revealed the significant changes of flavonoids in uniconazole-treated bananas compared to control samples at both 15 days and 25 days post treatment. Transcriptome analysis shows that the DEGs between the treatment and control groups were related to a series of metabolic pathways, including lignin biosynthesis, phenylpropanoid metabolism, and peroxidase activity. Comprehensive analysis of the key pathways of co-enrichment of DEGs and DAMs from 15 d to 25 d after uniconazole treatment shows that flavonoid biosynthesis was upregulated. Conclusions In addition to the decrease in GA, the increase in tannin procyanidin B1 may contribute to dwarfing of banana plants by inhibiting the activity of GA. The increased of flavonoid biosynthesis and the change of lignin biosynthesis may lead to dwarfing phenotype of banana plants. This study expands our understanding of the mechanisms underlying uniconazole-induced banana dwarfing.

BackgroundThe incidence of nephrolithiasis is increasing rapidly worldwide. Calcium oxalate is the most common constituent, contributing to approximately 80% of all kidney stones. The gut microbiome, through its oxalate-degrading ability, may play a role in decreasing morbidity due to urinary calculus. Fecal microbiome transplantation (FMT) has been reported to be effective in restoring the gastrointestinal microbial community in different conditions. The transplantation of whole communities that have oxalate-degrading function may be a more effective strategy than the transplantation of isolated strains.MethodsFMT was carried out in male guinea pigs and male Sprague–Dawley laboratory rats (SDRs). Fresh feces were collected from guinea pigs housed in metabolic cages. SDRs were divided into four groups: two groups received standard rat chow (SC) (groups SC and SC + FMT), and two groups were fed a 5% potassium oxalate diet (OD) (groups OD + phosphate-buffered saline (PBS) and OD + FMT). On day 14, groups OD + PBS, OD + FMT, and SC + FMT received either PBS or guinea pig feces by esophageal gavage. The composition of the microbiota of guinea pigs and SDRs was analyzed using a 16S rRNA gene sequencing approach. Biochemical analysis of urine samples from SDRs revealed the presence of calcium oxalate (CaOx) crystals, which were presumed to originate from kidney stones. Renal function was examined using real-time PCR analysis and immunohistochemical staining for renin, angiotensin-converting enzyme, and osteopontin (OPN) expression.ResultsFMT resulted in a gut microbiota that was a mixture of guinea pig and SDR bacteria. A microbial network involving Muribaculaceae, Lactobacillus, and Bifidobacterium was activated by FMT in group OD + FMT. As a result, urinary oxalate, calcium, uric acid, creatinine and urea in urine samples were reduced significantly. Similarly, significant reduction of uric acid and blood urea nitrogen to creatinine ratio in serum samples was observed (p < 0.05). Microscopic observations revealed a high CaOx crystal score (4+) in the kidneys of rats in group OD + PBS, whereas a lower score (2+) was observed in the rats in group OD + FMT. Up-regulation of OPN and down-regulation of renin were also associated with FMT.ConclusionA microbial network involving Muribaculaceae and other oxalate-degrading bacteria achieved by FMT was capable of reducing urinary oxalate excretion and CaOx crystal deposition in the kidney through increasing intestinal oxalate degradation. FMT may exert a renoprotective function in oxalate-related kidney stones.

Abstract Background The present study aimed to investigate the inhibitory effects of aminoguanidine (AG) and 2-bromoethylamine (2-BEA) on the semicarbazide-sensitive amine oxidase (SSAO) activity both in vitro and in vivo, and the prevention role of AG and 2-BEA in the morphology of aorta and kidney in diabetic rats. Methods The aortic homogenates isolated from Sprague-Dawley (SD) rats were treated with different concentrations of AG or 2-BEA to investigate the inhibitory effects on the SSAO activity in vitro, using benzylamine as the substrate. In addition, 65 male SD rats were randomly assigned into normal control (NC) (n = 10), NC + AG (n = 10), NC + 2-BEA (n = 10) and diabetes mellitus (DM) model groups (n = 35). Type 1 diabetic rat model was induced by intraperitoneal injection of 1% streptozotocin-sodium citrate buffer 55 mg/kg. After establishing the diabetic rat model by a single intraperitoneal injection of streptozotocin. Except those failed in modeling, 30 rats in the DM model group were further randomly divided into the DM, DM + AG, DM + 2-BEA groups (n = 10 in each). Rats in the DM + AG and NC + AG group were intraperitoneally injected with AG (25 mg/kg),those in the DM + 2-BEA and NC + 2-BEA group were administered with 2-BEA (20 mg/kg) daily for eight weeks. After eight weeks of treatment, the SSAO activity in the plasma and aorta, and plasma levels of formaldehyde (FA) and methylamine (MA) were measured by high performance liquid chromatograph. Radioimmunoassay was used to determine the plasma endothelin-1 (ET-1) concentration, while nitric acid deoxidized enzyme method was performed to detect the plasma nitrate/nitrite (NO(x)-) level. Besides, the morphological changes of aorta and kidney tissues were examined by optical and electron microscopes. Results Both AG and 2-BEA exerted strong inhibitory effect on the aortic SSAO activity in vitro, with the IC50 values of 12.76 μmol/L and 3.83 μmol/L, respectively. Abstract The application of endophytic bacteria, particularly members of the genus Bacillus, offers a promising strategy for the biocontrol of plant fungal diseases, owing to their sustainability and ecological safety. Although multiple secondary metabolites that demonstrate antifungal capacity have been identified in diverse endophytic bacteria, the regulatory mechanisms of their biosynthesis remain largely unknown. To elucidate this, we sequenced the entire genome of Bacillus amyloliquefaciens GKT04, a strain isolated from banana root, which showed high inhibitory activity against Fusarium oxysporum f. sp. cubense race 4 (FOC4). The GKT04 genome consists of a circular chromosome and a circular plasmid, which harbors 4,087 protein‐coding genes and 113 RNA genes. Eight gene clusters that could potentially encode antifungal components were identified. We further applied RNA‐Seq analysis to survey genome‐wide changes in the gene expression of strain GKT04 during its inhibition of FOC4. In total, 575 upregulated and 242 downregulated genes enriched in several amino acid and carbohydrate metabolism pathways were identified. Specifically, gene clusters associated with difficidin, bacillibactin, and bacilysin were significantly upregulated, and their gene regulatory networks were constructed. Our work thereby provides insights into the genomic features and gene expression patterns of this B. amyloliquefaciens strain, which presents an excellent potential for the biocontrol of Fusarium wilt.

Synaptic loss induced by beta-amyloid (Aβ) plays a critical role in the pathophysiology of Alzheimer's disease (AD), but the mechanisms underlying this process remain unknown. In this study, we found that oridonin (Ori) rescued synaptic loss induced by Aβ1-42 in vivo and in vitro and attenuated the alterations in dendritic structure and spine density observed in the hippocampus of AD mice. In addition, Ori increased the expression of PSD-95 and synaptophysin and promoted mitochondrial activity in the synaptosomes of AD mice. Ori also activated the BDNF/TrkB/CREB signaling pathway in the hippocampus of AD mice. Furthermore, in the Morris water maze test, Ori reduced latency and searching distance and increased the number of platform crosses in AD mice. These data suggest that Ori might prevent synaptic loss and improve behavioral symptoms in Aβ1-42-induced AD mice.

Calcitonin gene-related peptide (CGRP) monoclonal antibodies in the treatment of episodic and chronic migraine was invetigated. A comprehensive literature search was conducted in Ovid Medline, Web of Science and Embase databases from their inception until April 2024 for randomized controlled trials comparing CGRP monoclonal antibodies with placebo or other active treatments in adults with episodic or chronic migraine. The primary outcome assessed was the incidence of hypertension, and secondary outcomes were tolerability, acceptability and adverse events. Data analysis was performed using a random-effects model, and the strength of evidence was evaluated using the Grading of Recommendation, Assessment, Development and Evaluation (GRADE) approach. A total of eleven studies involving 9729 participants were found eligible and included for data analysis. The results revealed that the pooled odds ratio for the incidence of hypertension in patients receiving CGRP monoclonal antibodies compared to placebo was (95% confidence interval (CI): 0.60, 2.21; I² = 32%), suggesting no significant increase in hypertension risk. Moreover, no significant differences were observed in tolerability or acceptability between the CGRP monoclonal antibody and placebo groups. However, the overall risk of total adverse events was significantly higher in the CGRP monoclonal antibody group (odds ratio (OR): 1.13; 95% CI: 0.97, 1.33; I² = 56%; p = 0.01). These findings indicate that CGRP monoclonal antibodies are well-tolerated and present a generally safe option for treating episodic and chronic migraine. Although there was no significant increase in the incidence of hypertension, a slight rise in overall adverse events was observed. Consequently, CGRP monoclonal antibodies may be considered a viable treatment option for patients who have not found other treatments effective or tolerable, or who have contraindications to alternative therapies. Abstract Background The addition of wine lees to diets can make up for the deficiencies caused by traditional forages in beef cattle farming. However, the effects of different wine lees ratios on average daily weight, gastrointestinal microbial community structure and metabolites in Guanling crossbred cattle have been rarely studied. This study assessed the effects of feeds containing wine lees on weight gain, gastrointestinal microbial community structure, and metabolites in Guanling crossbred cattle and elucidated the metabolic responses induced by wine lees. Eighteen cows were randomly assigned to receive fed concentrate (C group), feed containing 15% wine lees (group A), or feed containing 30% wine lees (group B) for 60 days. Results The average daily weight gain of group A and group B increased by 76.75% and 57.65%, respectively, compared with group C. Microbial community analysis showed that wine lees increased the abundance of Prevotella_1 in the rumen, decreased the abundance of Ruminococcaceae UCG 011 and Lachnospiraceae_FCS020_group in the rumen, and increased the abundance of Tyzzerella_4, Family_Xlll_AD3011_group, Granulicella, and Eisenbergiella in the cecum. Metabolomics analyses showed that wine lees decreased the concentrations of indole-3-ethanol in the rumen, and complexity cecal metabolism. Notably, linoleic acid metabolism was significantly enriched in both the rumen and cecum. Mantel test analyses indicated that the adverse effects of WL were reduced by stimulating the metabolism of linoleic acid, α-linolenic acid, and tryptophan, and these changes were mediated by intestinal microorganisms. The Guanling cattle cecum was enriched for several unfavorable metabolic pathways when wine lees concentrations reached 30%, which increased the likelihood of intestinal lesions. Conclusion This study shows that WL supplementation alters gut microbiota and metabolic pathways, improving cattle growth and health. Moderate WL levels (15%) enhance gut health and beneficial pathways (e.g., linoleic and alpha-linolenic acid metabolism). However, higher WL inclusion (30%) may activate adverse pathways, raising the risk of intestinal damage. To maximize benefits and minimize risks, WL levels should be carefully managed.

Abstract Despite mounting evidence linking Acute Myocardial Infarction (AMI) to Alzheimer’s disease (AD), the shared mechanism of these two conditions’ occurrence remains unclear. This research aims to delve deeper into the molecular process of the occurrence of the two diseases. We retrieved the gene expression profiles of AD (GSE5281) and AMI (GSE66360) from the Gene Expression Omnibus database. Then, a total of 22 common differentially expressed genes (DEGs) including one downregulated gene and 21 upregulated genes were chosen for further analysis. Following the discovery of the common DEGs between AMI and AD, we performed protein–protein interaction analysis and hub gene identification analysis. Next, ten important hub genes were identified. Additionally, the key genes were identified by the least absolute shrinkage and selection operator and support vector machine‐recursive feature elimination and multivariable logistic regression analysis. The BCL6 was identified to be the most connected with AMI and AD. Finally, the BCL6 gene was validated in the GSE40680 (AMI) and GSE122063 (AD) datasets. Our research indicates that AMI and AD share a comparable pathophysiology. The Hub genes, especially BCL6, were essential in developing AMI and AD. In addition, these hub genes and shared pathways can offer fresh perspectives for additional mechanism investigation.

Lactic acid bacteria (LAB) inoculants are commonly used in silage production, yet their effects on silage containing antimicrobial components, such as those found in Leonurus japonicus, remain less explored. Herein, the harvested alfalfa were thoroughly mixed with dried Leonurus japonicus Houtt. (LJH) at a ratio of 9:1 on a fresh weight basis and treated without (CK) or with a lactic acid bacterial inoculant (L; Lentilactobacillus buchneri). The mixtures were stored under anaerobic conditions in vacuum-sealed polyethylene bags for 30 days at ambient temperature. The L-treated silage exhibited high levels of water-soluble carbohydrates (4.98% dry matter (DM)) and acid detergent fiber (27.88% DM). Compared to that of treatment CK, treatment with L increased the acetic acid content of the silage, as a result of increased (p < 0.05) bacterial dominance and decreased (p < 0.05) bacterial richness indices (e.g., Pielou’s E, Shannon, and Simpson) in the pre-storage period. However, these changes gradually reduced as the storage length increased. Treatment L reshaped the bacterial community structure of silage, by increasing the prevalence of Lactobacillus and reducing relative abundances of Enterococcus and Weissella. However, the principal coordinate and Bray–Curtis index analyses illustrated that samples from the L-treated silages exhibited similarities to the CK samples post-fermentation. Overall, the effect of LJH on LAB was only observed in the later stages of fermentation, which did not sufficiently change the silage quality. Hence, using LJH in silage is vital for clean livestock production without compromising the function of LAB when mixed with alfalfa silage.

Abstract Background Atherosclerosis is now the main cause of cardiac-cerebral vascular diseases around the world. Disturbances in lipid metabolism have an essential role in the development and progression of atherosclerosis. Thus, we aimed to investigate lipid metabolism-related molecular clusters and develop a diagnostic model for atherosclerosis. Methods First, we used the GSE100927 and GSE43292 datasets to screen differentially expressed lipid metabolism-related genes (LMRGs). Subsequent enrichment analysis of these key genes was performed using the Metascape database. Using 101 atherosclerosis samples, we investigated the LMRG-based molecular clusters and the corresponding immune cell infiltration. After that, a diagnostic model for atherosclerosis was constructed using the least absolute shrinkage and selection operator (LASSO) and multivariate logistic regression. Finally, a series of bioinformatics techniques, including CIBERSORT, gene set variation analysis, and single-cell data analysis, were used to analyze the potential mechanisms of the model genes in atherosclerosis. Results A total of 29 LMRGs were found to be differentially expressed between atherosclerosis and normal samples. Functional and DisGeNET enrichment analyses indicated that 29 LMRGs are primarily engaged in cholesterol and lipid metabolism, the PPAR signaling pathway, and regulation of the inflammatory response and are also closely associated with atherosclerotic lesions. Two LMRG-related molecular clusters with significant biological functional differences are defined in atherosclerosis. A three-gene diagnostic model containing ADCY7, SCD, and CD36 was subsequently constructed. Receiver operating characteristic curves, decision curves, and an external validation dataset showed that our model exhibits good predictive performance. In addition, three model genes were found to be closely associated with immune cell infiltration, especially macrophage infiltration. Conclusions Our study comprehensively highlighted the intricate association between lipid metabolism and atherosclerosis and created a three-gene model for future clinical diagnosis.

BackgroundPrevious studies have not established potential causal associations between coffee and caffeine consumption in endometrial cancer (EC) and its subgroups. Therefore, we used a two-sample MR method to assess the causal association between coffee and caffeine consumption and EC risk. We also evaluated the association between these genetically predicted exposures and EC prognosis.Materials and methodsThis study used 12 and two independent single-nucleotide polymorphisms (SNPs) associated with coffee and caffeine consumption as instrumental variables at a genome-wide significance level of p < 5 × 10–8. The EC Association Consortium (ECAC) performed a genome-wide association study (GWAS) analysis of 12,906 cases and 108,979 controls. FinnGen Consortium performed a GWAS analysis of 1,967 EC cases and 167,189 controls. The primary technique we employed was inverse-variance weighted, followed by the weighted median, MR-Egger regression, and MR robust adjusted profile score methods. We used the MR pleiotropy residual sum, Outlier test, and MR-Egger regression to assess Outlier and pleiotropic variants. We also conducted a sensitivity analysis through the leave-one-out method.ResultsGenetically predicted coffee consumption was not associated with EC and its subgroups in the ECAC, and the association was consistent in the FinnGen consortium. After excluding eight SNPs with confounding factors, the study performed sensitivity analyses, delivering consistent results. We also observed that caffeine consumption was not correlated with EC risk. As confirmed by MR analysis, selected SNPs determined that most do not significantly impact the likelihood of developing EC.ConclusionOur study indicated no convincing evidence supports coffee and caffeine consumption causing EC or impacting its prognosis. More studies are needed to validate the results.

BackgroundMetS is associated with greater morbidity and mortality in relation to a number of malignancies, but its association with ovarian cancer remains contested. The present study was a systematic review and meta-analysis of case-control and cohort studies examining the association between MetS and ovarian cancer risk.MethodsThe study was registered on the PROSPERO platform in January 2023 (CRD42023391830). Up until February 13, 2023, a complete search was undertaken in PubMed, EMBASE, Web of Science, the Cochrane Library, and ClinicalTrials. On the basis of inclusion and exclusion criteria, eligible studies for meta-analysis were screened to determine the association between MetS and ovarian cancer risk.ResultsFive studies were included in total, including three cohort studies and two case-control studies. BackgroundCoronary artery disease (CAD) is a complex illness with unknown pathophysiology. Peripheral biomarkers are a non-invasive method required to track the onset and progression of CAD and have unbeatable benefits in terms of early identification, prognostic assessment, and categorization of the diagnosis. This study aimed to identify and validate the diagnostic and therapeutic potential of differentially expressed immune-related genes (DE-IRGs) in CAD, which will aid in improving our knowledge on the etiology of CAD and in forming genetic predictions.MethodsFirst, we searched coronary heart disease in the Gene Expression Omnibus (GEO) database and identified GSE20680 (CAD = 87, Normal = 52) as the trial set and GSE20681 (CAD = 99, Normal = 99) as the validation set. Functional enrichment analysis using protein-protein interactions (PPIs), Gene Ontology (GO), and Kyoto Encyclopedia of Genes and Genomes (KEGG) was carried out on the identified differentially expressed genes. Optimal feature genes (OFGs) were generated using the support vector machine recursive feature elimination algorithm and the least absolute shrinkage and selection operator (LASSO) algorithm. Furthermore, immune infiltration in CAD patients and healthy controls was compared using CIBERSORT, and the relationship between immune cells and OFGs was examined. In addition, we constructed potential targeted drugs for this model through the Drug-Gene Interaction database (DGIdb) database. Finally, we verify the expression of S100A8-dominated OFGs in the GSE20681 dataset to confirm the universality of our study.ResultsWe identified the ten best OFGs for CAD from the DE-IRGs. Functional enrichment analysis showed that these marker genes are crucial for receptor-ligand activity, signaling receptor activator activity, and positive control of the response to stimuli from the outside world. Additionally, CIBERSORT revealed that S100A8 could be connected to alterations in the immune microenvironment in CAD patients. Furthermore, with the help of DGIdb and Cytoscape, a total of 64 medicines that target five marker genes were subsequently discovered. Finally, we verified the expression of the OFGs genes in the GSE20681 dataset between CAD patients and normal patients and found that there was also a significant difference in the expression of S100A8.ConclusionWe created a 10-gene immune-related prognostic model for CAD and confirmed its validity. The model can identify potential biomarkers for CAD prediction and more accurately gauge the progression of the disease.

The present study investigated the effects of Lentilactobacillus buchneri, Saccharomyces cerevisiae, and a mixture of the two on the cellulose degradation and microbial community of cellulase-treated Pennisetum sinese (CTPS) during biological pretreatment. The CTPS was stored without additives (CK) or with L. buchneri (L), yeast (Y, S. cerevisiae), and their mixture (LY) under anaerobic conditions for 60 days. All inoculants enhanced the anaerobic fermentation of CTPS. In relative to L, inoculations with Y and LY decreased the cellulose level of fermented-CTPS by 8.90 ~ 17.13%. Inoculation with L inhibited the growth of Weissella cibaria during anaerobic storage. However, inoculations with LY increased the relative abundance of the homofermentative bacterium Lactiplantibacillus plantarum by 6.04%. Therefore, inoculating S. cerevisiae reduced the adverse effects of L. buchneri-stimulated fermentation on cellulose degradation by altering the bacterial community during anaerobic storage of P. sinese. This work provides a new insight for the subsequent anaerobic digestion of P. sinese.

The present study evaluated the effect of dietary supplementation with fermented-Moutai distillers' grain (FMDG) on the growth performance, meat quality, amino acid composition and blood metabolites of finishing cattle. Thirty cattle (2 years old; 237.55 ± 10.72 kg) were randomly assigned to one of three dietary supplementations: 0% FMDG (control), 15% FMDG (R1) and 30% FMDG (R2) [dry matter (DM) basis]. After 60 days, the inclusion of FMDG had no significant (p > 0.05) effects on the growth performance indices (dry matter intake, average daily gain and feed efficiency), meat quality (cooking yield, shear force, L*, a*, and b* values) or bovine blood biochemical indicators (except albumin and immunoglobulin A). Cattle fed R1 had the lowest (p = 0.001) loin eye area. Supplementation with FMDG significantly (p < 0.05) increased the beef contents of various amino acids (except isoleucine and arginine) compared with the control diet. Specifically, R2 significantly increased (p < 0.05) the total amino acid, essential amino acid, non-essential amino acid and umami amino acid contents in beef, while the difference in bitter amino acid content between different treatments was not significant (p = 0.165). These results suggest that it is feasible to include FMDG at up to 30% DM without affecting the growth performance, meat quality or blood metabolites of finishing cattle.

Articular cartilage lesion is a common disease to be treated by arthroscopic surgery. It will eventually progress to osteoarthritis without proper management, which can affect patients’ work and daily life seriously. Although mechanical debridement and laser have been used clinically for its treatment, due to their respective drawbacks, radiofrequency has drawn increasing attention from clinicians as a new technique with more advantages. However, the safety and efficacy of radiofrequency have also been questioned. In this article, the scope of application of radiofrequency was reviewed following an introduction of its development history and mechanism, and the methods to ensure the safety and effectiveness of radiofrequency through power and temperature control were summarized.

To effectively use local grass resources to cover the winter feed shortage on the Qinghai-Tibetan Plateau, the silage fermentation and in vitro digestibility of perennial oat (Helictotrichonvirescens Henr.) were investigated. Perennial oat was harvested at the heading/flowering stage, wilted under sunny conditions, chopped, vacuumed in small bag silos, and stored at ambient temperatures (5–15 °C) for 60 days. The silages were treated without (CK) or with local lactic acid bacteria (LAB) inoculant (IN1), commercial LAB inoculant (IN2), and sodium benzoate (BL). Control silages of perennial oat at early heading stage showed higher (p < 0.05) lactate and acetate contents and lower (p < 0.05) final pH, butyrate, and ammonia-N contents than those at the flowering stage. High levels of dry matter recovery (DMR) and crude protein (CP) were observed in IN1- and BL-treated silages, with high in vitro gas production and dry matter digestibility. Compared to CK, additives increased (p < 0.05) aerobic stability by inhibiting yeasts, aerobic bacteria, and coliform bacteria during ensiling. In particular, the local LAB inoculant increased (p < 0.05) concentrations of lactate, acetate and propionate, and decreased concentrations of butyrate and ammonia-N in silages. This study confirmed that local LAB inoculant could improve the silage quality of perennial oat, and this could be a potential winter feed for animals such as yaks on the Qinghai Tibetan Plateau.

The deformation and fracture characteristics of shale in the Changning-Xingwen region were experimentally studied under triaxial cyclic loading with a controlled pore-water pressure. An RLW-2000M microcomputer-controlled coal-rock rheometer was used in the State key Laboratory of coal mine disaster dynamics and control in Chongqing University. These experimental results have indicated the following. (i) The shale softened after being saturated with water, while its failure strength decreased with the increase of axial strain. (ii) A complete cyclic loading–unloading process can be divided into four stages under the coupling action of axial cyclic loading and pore-water pressure; namely the slow or accelerated increasing of strain in the loading stage, and the slow or accelerated decreasing of strain in the unloading stage. (iii) The axial plastic deformation characteristics were similar when pore-water pressures were set to 2, 6 and 10 MPa. Nevertheless, the shale softened ostensibly and fatigue damage occurred during the circulation process when the pore-water pressure was set to 14 MPa. (iv) It has been observed that the mean strain and strain amplitude under axial cyclic are positively correlated with pore-water pressure, while the elastic modulus is negatively correlated with pore-water pressure. As the cycle progresses, the trends in these parameters vary, which indicates that the deformation and elastic characteristics of shale are controlled by pore-water pressure and cyclic loading conditions. (v) Evidenced via triaxial compression tests, it was predominantly shear failure that occurred in the shale specimens. In addition, axial cyclic loading caused the shale to generate complex secondary fractures, resulting in the specimens cracking along the bedding plane due to the effect of pore-water pressure. This study provides valuable insight into the understanding of the deformation and failure mechanisms of shale under complicated stress conditions.

To investigate the differences in volatile components during the processing of Fuling Zhacai, or pickled mustard tuber, with three stacked layers of mustard tuber and salt, gas chromatography-tandem mass spectrometry (GC-MS/MS)-based non-targeted metabolomics combined with multivariate statistical analysis was used to analyze the changes in volatile metabolites and significantly differential metabolic pathways during the spontaneous fermentation of Zhacai. The results showed that a total of 627 volatile compounds were identified from Zhacai samples at different pickling stages, including terpenoids, esters, heterocyclic compounds, terpenoids, aldehydes, ketones, alcohols, phenols, amines, sulfur compounds, acids, halogenated hydrocarbons, and nitrogen compounds. A significant difference was observed in volatile components among pickling stages. A total 157 unique differential metabolites in the 1st pickling stage vs. the control group, 28 in the 2nd vs. the 1st pickling stage, and one in the 3rd vs. 2nd pickling stage were identified. Kyoto Encyclopedia of Genes and Genomes (KEGG) pathway analysis showed that the differential metabolites were significantly enriched in metabolic pathways including secondary metabolite biosynthesis, diterpenoid biosynthesis, monoterpenoid biosynthesis, caffeine metabolism, and fatty acid biosynthesis. This study provides a theoretical basis for the standardized production of Fuling Zhacai.
