Background: The analysis of phenotypic characteristics on Mycobacterium tuberculosis (MTB)-specific T cells is a promising approach for the diagnosis of active tuberculosis (aTB) and for monitoring treatment success. We therefore studied phenotypic changes on MTB-specific CD4 T cells upon anti-tuberculosis treatment initiation in relation to the treatment response as determined by sputum culture.Methods: Peripheral blood mononuclear cells from subjects with latent MTB infection (n = 16) and aTB (n = 39) at baseline, weeks 9, 12, and 26 (end of treatment) were analyzed after intracellular interferon gamma staining and overnight stimulation with tuberculin. Liquid sputum cultures were performed weekly until week 12 and during 4 visits until week 26.Results: T cell activation marker expression on MTB-specific CD4 T cells differed significantly between subjects with aTB and latent MTB infection with no overlap for the frequencies of CD38pos and Ki67pos cells (both p < 0.0001). At 9 weeks after anti-TB treatment initiation the frequencies of activation marker (CD38, HLA-DR, Ki67) positive MTB-specific, but not total CD4 T cells, were significantly reduced (p < 0.0001). Treatment induced phenotypic changes from baseline until week 9 and until week 12 differed substantially between individual aTB patients and correlated with an individual's time to stable sputum culture conversion for expression of CD38 and HLA-DR (both p < 0.05). In contrast, the frequencies of maturation marker CD27 positive MTB-specific CD4 T cells remained largely unchanged until week 26 and significantly differed between subjects with treated TB disease and latent MTB infection (p = 0.0003).Discussion: Phenotypic changes of MTB-specific T cells are potential surrogate markers for tuberculosis treatment efficacy and can help to discriminate between aTB (profile: CD38pos, CD27low), treated TB (CD38neg, CD27low), and latent MTB infection (CD38neg, CD27high).

Preharvest application of hormetic doses of ultraviolet-C (UV-C) generates beneficial effects in plants. In this study, within 1 week, four UV-C treatments of 0.4 kJ/m2 were applied to 3-week-old lettuce seedlings. The leaves were inoculated with a virulent strain of Xanthomonas campestris pv. vitians ( Xcv ) 48 h after the last UV-C application. The extent of the disease was tracked over time and a transcriptomic analysis was performed on lettuce leaf samples. Samples of lettuce leaves, from both control and treated groups, were taken at two different times corresponding to T2, 48 h after the last UV-C treatment and T3, 24 h after inoculation (i.e., 72 h after the last UV-C treatment). A significant decrease in disease severity between the UV-C treated lettuce and the control was observed on days 4, 8, and 14 after pathogen inoculation. Data from the transcriptomic study revealed, that in response to the effect of UV-C alone and/or UV-C + Xcv, a total of 3828 genes were differentially regulated with fold change (|log2-FC|) > 1.5 and false discovery rate (FDR) < 0.05. Among these, of the 2270 genes of known function 1556 were upregulated and 714 were downregulated. A total of 10 candidate genes were verified by qPCR and were generally consistent with the transcriptomic results. The differentially expressed genes observed in lettuce under the conditions of the present study were associated with 14 different biological processes in the plant. These genes are involved in a series of metabolic pathways associated with the ability of lettuce treated with hormetic doses of UV-C to resume normal growth and to defend themselves against potential stressors. For centuries, some Indigenous Peoples of the Americas have planted corn, beans and squash or pumpkins together in mounds, in an intercropping complex known as the Three Sisters. Agriculturally, nutritionally and culturally, these three crops are complementary. This literature review aims to compile historical foods prepared from the products of the Three Sisters planting system used in Indigenous communities in the region encompassing southern Quebec and Ontario in Canada, and northeastern USA. The review does not discuss cultural aspects of the Three Sisters cropping system or describe foods specific to any one Indigenous group, but rather, gives an overview of the historical foods stemming from this intercropping system, many foods of which are common or similar from one group to another. Some of the methods of food preparation used have continued over generations, some of the historical foods prepared are the foundation for foods we eat today, and some of both the methods and foods are finding revival.

As an environmentally friendly approach for fruit quality improvement, the effect of preharvest UV-C on the physiology of strawberry fruit during postharvest storage remains to be assessed. Strawberry fruit developed with supplementary UV-C were stored at room temperature for 2 weeks. Preharvest UV-C attenuated fruit postharvest senescence and altered phytochemicals composition. Higher ester titer was found in the treated fruit at harvest, whereas higher terpene and furanone contents were detected after 72 h of storage. At harvest, polyphenolics accumulated to a higher level in UV-C group, but the difference disappeared after 24 h of storage. Meanwhile, the intrinsic level of abscisic acid and the expressions of FaPYR1, SnRK2, and FaASR in the UV-C-treated fruit was enhanced at harvest but returned to a lower level as storage proceeded. This study highlights the time-dependent effect of preharvest UV-C on strawberry fruit postharvest biochemical indexes and the possible involvement of abscisic acid signaling factors., (Crown Copyright © 2019. Bacterial leaf spot (BLS) caused by Xanthomonas campestris pv. vitians ( Xcv ) places a major constraint on lettuce production worldwide. The most sustainable strategy known to date for controlling BLS is the use of resistant cultivars. The nutrient elemental signature (ionome) of ten lettuce cultivars with three levels of resistance was analyzed by inductively coupled plasma optical emission spectroscopy (ICP-OES) to determine which nutrient balances are linked to resistance to BLS, and to assess the effect of Xcv infection on the ionome. The elemental concentrations were preprocessed with isometric log-ratios to define nutrient balances. Using this approach, 4 out of 11 univariate nutrient balances were found to significantly influence the resistance of lettuce cultivars to BLS ( P < 0.05). These significant balances were the overall nutritional status balancing all measured nutrients with their complementary in the dry mass, as well as balances [Mn | Zn,Cu], [Zn | Cu], and [S,N | P]. Moreover, the infection of lettuce cultivars mostly affected the lettuce ionome on the [N,S | P] balance, where infection tended to lean the balance toward the N,S part relatively to P. This study shows that nutrient uptake in lettuce can be affected by BLS infection and that nutrient status influences resistance to BLS infection.

Ultraviolet-C (UV-C) radiation has been reported to induce defence responses to pathogens in growing crops and described as a new environmentally friendly method for disease control. However, whether the effect of the induced defence mechanisms will persist after the stress imposed by UV-C is alleviated and how these mechanisms interact with pathogen elicitors upon infection have not yet been investigated. Thus, we inoculated strawberry plants with Mycosphaerella fragariae, the causal agent of leaf spot disease, after 5 weeks of repeated UV-C irradiation treatment (cumulative dose of 10.2 kJ m -2 ) and investigated the alteration of gene expression and biochemical phenotypes. The results revealed that UV-C treatment had a significant impact on gene expression in strawberry leaves and led to the overexpression of a set of genes involved in plant-pathogen interaction. UV-C-treated leaves displayed a stronger response to infection after inoculation, with reduced symptoms and increases in accumulation of total phenolics and volatile terpenes, higher expression of pathogenesis-related proteins and the activity of several defence enzymes. Recent studies presented preharvest ultraviolet C (UV-C) as an environmentally friendly approach for the management of horticultural crop diseases. The effect of this approach on quality preservation during postharvest storage has not yet been investigated. Strawberry fruit harvested from plants grown with supplemental UV-C were stored at room temperature for 72 h, and their postharvest shelf-life biochemical indicators were evaluated. The involvement of microRNAs (miRNAs) in the activation of UV-C-induced antioxidant systems was investigated. Preharvest UV-C contributed to the preservation of sugar and organic acid and reduced overall lipid peroxidation in strawberry fruit during storage. We found that miR159 and miR398 were downregulated by preharvest UV-C and that their respective targets were upregulated at the early stage of storage with enhancement of the activity of antioxidant enzymes. The initial burst of H 2 O 2 and O 2 •   - suggested that preharvest UV-C primed the fruit in an antioxidative activated state via reactive-oxygen-species-mediated feedback control with post-transcriptional involvement of miRNAs.

Preharvest ultraviolet C (UV-C) irradiation is an innovative approach for increasing the bioactive phytochemical content of strawberries to increase the disease resistance and nutritional value. This study investigated the changes in individual flavonoids in strawberry developed with three different cumulative doses of preharvest UV-C treatment (low, 9.6 kJ m -2 ; middle, 15 kJ m -2 ; and high , 29.4 kJ m -2 ). Significant accumulation (p < 0.05) of phenolics (25-75% increase), namely, cyanidin 3-glucoside, pelargonidin 3-glucoside/rutinoside, glucoside and glucuronide of quercetin and kaempferol, and ellagic acid, was found in the fruit subjected to low and middle supplemental doses of UV-C radiation. The expression of the flavonoid pathway structural genes, i.e., FaCHS1, FaCHI, FaFHT, FaDFR, FaFLS, and FaFGT, was upregulated in the low- and middle-dose groups, while the early stage genes were not affected by the high dose. FaMYB1 was also relatively enhanced in the low- and middle-dose groups, while FaASR was upregulated in only the low-dose group. Hormetic preharvest UV-C dose ranges for enhancing the polyphenol content of strawberries were established for the first time.

Recent studies have highlighted the biological and physiological effects of pre-harvest ultraviolet (UV)-C treatment on growing plants. However, little is known about the involvement of volatile organic compounds (VOCs) and their response to this treatment. In this study, strawberry plants were exposed to three different doses of UV-C radiation for seven weeks (a low dose: 9.6kJm -2 ; a medium dose: 15kJm -2 ; and a high-dose: 29.4kJm -2 ). Changes in VOC profiles were investigated and an attempt was made to identify factors that may be involved in the regulation of these alterations. Principle compounds analysis revealed that VOC profiles of UV-C treated samples were significantly altered with 26 VOCs being the major contributors to segregation. Among them, 18 fatty acid-derived VOCs accumulated in plants that received high and medium dose of UV-C treatments with higher lipoxygenase and alcohol dehydrogenase activities. In treated samples, the activity of the antioxidant enzymes catalase and peroxidase was inhibited, resulting in a reduced antioxidant capacity and higher lipid peroxidation. Simultaneously, jasmonic acid level was 74% higher in the high-dose group while abscisic acid content was more than 12% lower in both the medium and high-dose UV-C treated samples. These results indicated that pre-harvest UV-C treatment stimulated the biosynthesis of fatty acid-derived VOCs in strawberry leaf tissue by upregulating the activity of enzymes of the LOX biosynthetic pathway and downregulating antioxidant enzyme activities. It is further suggested that the mechanisms underlying fatty acid-derived VOCs biosynthesis in UV-C treated strawberry leaves are associated with UV-C-induced changes in phytohormone profiles., (Crown Copyright © 2017. Preharvest ultraviolet-C (UV-C) treatment of strawberry is a very new approach, and little information is available on the effect of this treatment on plant growth regulators. In this study, the effect of preharvest UV-C irradiations at three different doses on strawberry yield, fruit quality parameters and endogenous phytohormones was investigated simultaneously. The overall marketable yield of strawberry was not affected by the preharvest UV-C treatments, although more aborted and misshapen fruits were found in UV-C treated groups than in the untreated control. The fruits in the high dose group were firmer and had approximately 20% higher sucrose content and 15% higher ascorbic acid content than the control, while fruits from the middle and low dose groups showed no significant changes in these parameters. The lower abscisic acid (ABA) content found in the fruits in the high UV-C group may be associated with those quality changes. The citric acid content decreased only in the low dose group (reduction of 5.8%), with a concomitant 37% reduction in jasmonic acid (JA) content, compared to the control. The antioxidant status of fruits that received preharvest UV-C treatment was considered enhanced based on their oxygen radical absorbance capacity (ORAC) and malondialdehyde (MDA) content. In terms of aroma, three volatile alcohols differed significantly among the various treatments with obvious activation of alcohol acyltransferase (AAT) activity. Postharvest ultraviolet-C (UV-C) hormesis has been shown effective for the treatment of the edible part of several horticultural crops such as strawberry fruit; however, there is a lack of information on its potential preharvest impact. In the present study three strawberry cultivars (Fragaria × ananassa Duch. 'Albion', 'Charlotte' and 'Seascape') were exposed to UV-C during two growth seasons for a period of three weeks. Treatment begins when the first flowers were wide open and fruits at commercial maturity were harvested within one week after UV treatment. The physicochemical quality parameters of the fruits harvested from the treated plants were compared to those of the fruits of the untreated control plants. Preharvest UV-C treatment tended to increase fruit firmness in all cultivars with significant differences declared only for 'Albion' and 'Seascape' in season 2. Fruits from treated plants were generally redder but a significant difference was observed only for cultivar 'Charlotte' in the second growing season. Other color attributes were not affected by UV-C, neither were organic acids, simple sugars, soluble solids content (SSC), titratable acidity (TA) and pH, although in most cases slight decreases were noticed. Cultivar and growing season were the factors that mostly influenced on the parameters under study. Background: Ultraviolet-C (UV-C) has proven effective in extending shelf-life, reducing disease incidence and increasing the levels of health-promoting compounds in several crops. While most studies were conducted at the postharvest stage, our study examined the effect of preharvest UV-C application in three strawberry cultivars (Fragaria × ananassa Duch. 'Albion', 'Charlotte' and 'Seascape'). UV-C treatment was applied from the onset of flowering until the fruits reached commercial maturity on plants grown for two consecutive seasons under greenhouse conditions. The phytochemical profiles and antioxidant capacity of the fruits were assessed at harvest., Results: The ellagic acid and kaempferol-3-glucuronide contents were significantly increased only in fruits of the cultivar 'Albion' collected from UV-C-treated plants in season 1. UV-C did not consistently affect the other phenolic compounds that were measured. Based on the results of the ferric-reducing antioxidant power, oxygen radical absorbance capacity and total phenolic content assays, the antioxidant capacity of the three strawberry cultivars was not affected by UV-C. In controlled conditions, the effect of leaf infection by Mycosphaerella fragariae on total superoxide dismutase (SOD, EC 1.15.1.1) activity and induction of SOD isozymes was studied in three different strawberry cultivars, i.e. "Joliette" (resistant), "Honeoye" (partially resistant) and "Kent" (susceptible). Infection of the strawberry leaves with M. fragariae resulted in increase in SOD activities in all three cv. Total SOD increased 1d after inoculation in Joliette and Kent, and 2d after inoculation in Honeoye and reached the highest level in all three cv, at the 2nd day after inoculation, then slowly declined afterward. Total SODs in Joliette and Honeoye at the 2nd day after inoculation were 4516 and 4947Ug(-1) FW, respectively, which were significantly higher than that of Kent (3255Ug(-1)FW). Banding pattern of SOD isozymes in all three cv was also affected by infection. Electrophoresis profile of infected cv revealed two newly synthesized isozymes in Joliette and Honeoye, in which one band, i.e. R(f) = 0.53 was observed exclusively in inoculated Joliette and Honeoye. Therefore, it is considered to be associated with leaf spot resistance.

The effect of colonization with the vesicular-arbuscular mycorrhizal fungus Glomus etunicatum on the content of rishitin and solavetivone was determined in potato plants cv. Goldrush challenged with Rhizoctonia solani. Mycorrhization stimulated significantly the accumulation of both phytoalexins in roots of plantlets challenged with R. solani but did not influence phytoalexin levels in non-challenged plantlet roots. No accumulation of solavetivone or rishitin was detected in shoots. In Petri dish bioassays, rishitin and solavetivone inhibited mycelial growth of R. solani.

ABSTRACT The induction of resistance to Botrytis cinerea in carrot roots by UV radiation, a possible means for controlling storage diseases, was compared with systemic resistance induced by inoculation with the pathogen. UV radiation did not have any systemic effect, and disease resistance was induced only in tissues directly exposed to the radiation. Although UV radiation induced a local accumulation of 6-methoxymellein (6-MM), inoculation with B. cinerea caused 6-MM to accumulate systemically, away from the inoculation site. Because of amounts near or higher than the ED(50) (50% effective dose) for inhibiting B. cinerea found in both UV-treated and preinoculated roots at the time of challenge, 6-MM could be involved in both types of resistance. Freshly harvested carrots had a number of constitutive chitinases and beta-1,3-glucanases, which were not affected by UV radiation or inoculation. When challenged with B. cinerea, the induction of a 24-kDa chitinase was enhanced in UV-treated and preinoculated roots. Again, UV radiation had only a local effect in priming this chitinase response. Although UV- and pathogen-induced resistance in carrot may involve the same defenses, the responses are probably mediated differently, because UV radiation has an essentially local effect.
