1. Unraveling gluten protein compositions of einkorn, emmer, and spelt grown in the Lyon region in France.

Abstract

Background and Objectives: Minor species of wheat, including einkorn (Ei), emmer (Em), and spelt (Sp), are believed to have different gluten compositions compared to common wheat (CW). Given the limited research on these resources, and as a result of a collaboration with a collective of farmers, we aimed to provide insights into their gluten compositions. We focused on varieties readily accessible to farmers and bakers within the region of Lyon, France. Findings: We compared gluten compositions in 31 wheat varieties, revealing significantly lower glutenin and protein aggregate contents in Ei and Em than in CW. Similar trends, though milder, were observed for Sp. Additionally, we observed intra‐species variability, especially for Em, and identified varieties within each species with noteworthy protein profiles. Conclusions: Each species showed characteristic protein contents, yet with intraspecies variability. Utilizing an innovative approach to analyze the full information provided by chromatograms, we successfully identified varieties with unusual characteristics. Significance and Novelty: The specific protein compositions of Ei and Em, and to a lower extent Sp, may impact their processability and digestibility. Our study also highlights that considering complete chromatographic profiles can reveal differences that remain unnoticed when using traditional analytical approaches. [ABSTRACT FROM AUTHOR]

21. Fluorescence Anisotropy of Hydrophobic Probes in Poly(N-decylacrylamide)-block-poly(N,N-diethylacrylamide) Block Copolymer Aqueous Solutions: Evidence of Premicellar Aggregates

Abstract

Fluorescent probes, coumarin 153 (C153) and octadecylrhodamine B (ORB), were used to study the self-assembly in water of poly(N-decylacrylamide)-block-poly(N,N-diethylacrylamide), (PDcA11-block-PDEA295; Mn= 40 300 g mol−1; Mw/Mn= 1.01). From the variation of both the fluorescence intensity and the solvatochromic shifts of C153 with polymer concentration, the critical micelle concentration (CMC) was determined as 1.8 ± 0.1 μM. On the other hand, steady-state anisotropy measurements showed the presence of premicellar aggregates below the CMC. Time-resolved fluorescence anisotropy evidenced that ORB is located in the premicellar aggregates and the micelle core, while C153 is partitioned between the aggregates and the water phase. The micelle core contains both semicrystalline and amorphous regions. In the semicrystalline regions the probes cannot rotate, while in the amorphous regions the rotational correlation times correlate well with the hydrodynamic volume of the probes. The amorphous region of the micelle core is relatively fluid, reflecting the large free-volume accessible to the probes.

Abstract

Phenanthrene -end-labeled poly(N-decylacrylamide)n-b-poly(N,N-diethylacrylamide)m (PDcAn-b-PDEAm) block copolymers consisting in a highly hydrophobic block (n=11) and a thermoresponsive block with variable length (79 ≤m≤ 468) were synthesized by reversible addition-fragmentation chain transfer (RAFT) polymerization. A new phenanthrene-labeled chain transfer agent (CTA) was synthesized and used to control the RAFT polymerization of a hydrophobic acrylamide derivative, N-decylacrylamide (DcA). This first block was further used as macroCTA to polymerize N,N-diethylacrylamide (DEA) in order to prepare diblock copolymers with the same hydrophobic block of PDcA (number average molecular weight: Mn = 2720 g mol-1, polydispersity index: Mw/Mn = 1.13) and various PDEA blocks of several lengths (Mn = 10000 to 60000 g mol-1) with a very high blocking efficiency. The resulting copolymers self-assemble in water forming thermoresponsive micelles. The critical micelle concentration (CMC) was determined using the Förster resonance energy transfer (FRET) between phenanthrene linked at the end of the PDcA block and anthracene added to the solution at a low concentration (10-5 M), based on the fact that energy transfer only occurs when phenanthrene and anthracene are located in the core of the micelle. The CMC (~ 2 M) was obtained at the polymer concentration where the anthracene fluorescence intensity starts to increase. The size of the polymer micelles decreases with temperature increase around the lower critical solution temperature of PDEA in water (LCST~32 ºC) owing to the thermoresponsiveness of the PDEA shell.

Abstract

The thermoresponsive behavior of the rhodamine B end-labeled double hydrophilic block copolymer (DHBC) poly(N,N-dimethylacrylamide)-b-poly(N,N-diethylacrylamide) (RhB-PDMA207-b-PDEA177) and the 1:1 segmental mixture of PDEA and rhodamine B end-labeled PDMA homopolymers was studied over the range of 10-40°C at the air-water interface. The increase in collapse surface pressure (second plateau regime) of the DHBC with temperature confirms the thermoresponsiveness of PDEA at the interface. The sum of the π-A isotherms of the two single homopolymers weighted by composition closely follows the π-A isotherm of the DHBC, suggesting that the behavior of each block of the DHBC is not influenced by the presence of the other block. Langmuir-Blodgett monolayers of DHBC deposited on glass substrates were analyzed by laser scanning confocal fluorescence microscopy (LSCFM), showing schizophrenic behavior: at low temperature, the RhB-PDMA block dominates the inside of bright (core) microdomains, switching to the outside (shell) at temperatures above the lower critical solution temperature (LCST) of PDEA. This core-shell inversion triggered by the temperature increase was not detected in the homopolymer mixture. The present results suggest that both the covalent bond between the two blocks of the DHBC and the tendency of rhodamine B to aggregate play a role in the formation of the bright cores at low temperature whereas PDEA thermoaggregation is responsible for the formation of the dark cores above the LCST of PDEA.

Abstract

International audience; Biotinylated gradient glycopolymers have been synthesized via RAFT copolymerization of an acrylamide derivative of galactose with N-acryloylmorpholine in the presence of a biotin CTA. The polymerization was controlled with a linear increase in molecular weights up to 80% conversion. Copolymer chains have a gradient microstructure with an increasing proportion of galactose units towards the chain end. The presence of the biotin ligand at the end of the chains was confirmed by 1H NMR and MALDI-ToF MS. This strategy based on the use of a biotin-CTA instead of a post-polymerization labelling of the chains resulted in a high percentage of -functionalized chains (92-95%). Such -end-functionalized glycopolymer chains may interact with streptavidin-modified surfaces.

Abstract

1H DOSY NMR provides a simple and convenient method to determine both the molecular weight and the solution properties of poly( N-acryloylmorpholine) (PNAM) in various organic and aqueous solvents. This technique is used to measure the average diffusion coefficient DPNAM of a library of PNAM samples of increasing molecular weights synthesized by reversible addition fragmentation chain transfer controlled radical polymerization. In all solvents, the highly linear relationship between Log DPNAM and Log Mw yields calibration curves that can be used for the reliable determination of the molecular weight of various types of PNAM samples.
