The shells of the Pinnidae family are based on a double layer of single-crystal-like calcitic prisms and inner aragonitic nacre, a structure known for its outstanding mechanical performance. However, on the posterior side, shells are missing the nacreous layer, which raises the question of whether there can be any functional role in giving up this mechanical performance. Here, it is demonstrated that the prismatic part of the Pinna nobilis shell exhibits unusual optical properties, whereby each prism acts as an individual optical fiber guiding the ambient light to the inner shell cavity by total internal reflection. This pixelated light channeling enhances both spatial resolution and contrast while reducing angular blurring, an apt combination for acute tracking of a moving object. These findings offer insights into the evolutionary aspects of light-sensing and imaging and demonstrate how an architectured optical system for efficient light-tracking can be based on birefringent ceramics., (© 2023 The Authors. Swimming microorganisms often experience complex environments in their natural habitat. The same is true for microswimmers in envisioned biomedical applications. The simple aqueous conditions typically studied in the lab differ strongly from those found in these environments and often exclude the effects of small volume confinement or the influence that external fields have on their motion. In this work, we investigate magnetically steerable microswimmers, specifically magnetotactic bacteria, in strong spatial confinement and under the influence of an external magnetic field. We trap single cells in micrometer-sized microfluidic chambers and track and analyze their motion, which shows a variety of different trajectories, depending on the chamber size and the strength of the magnetic field. Combining these experimental observations with simulations using a variant of an active Brownian particle model, we explain the variety of trajectories by the interplay between the wall interactions and the magnetic torque. We also analyze the pronounced cell-to-cell heterogeneity, which makes single-cell tracking essential for an understanding of the motility patterns. In this paper, we synthesized superparamagnetic iron oxide nanoparticles (NPs) functionalized with (3-aminopropyl)triethoxysilane (Fe 3 O 4 @APTES). The synthesized NPs were coated with succinic anhydride (Fe 3 O 4 @COOH) in the next step. Half the surface of the NPs was shielded with wax microparticles via the Pickering emulsion technique, and the unshielded side was covered with poly(ethylene glycol) methyl ether. Platinum nanoparticles (Pt NPs) were deposited between PEG chains by the oxidation-reduction method through an in situ procedure to obtain a metal-polymer composite. These deposited Pt NPs have the potential to catalyze the decomposition of hydrogen peroxide at the surface of Janus nanomotors (JNMs). After de-waxing of the NPs, Irgacure 2959 (as the initiator) was reacted with the bare side of the NPs to provide the opportunity to grow poly(ε-caprolactone) (PCL) chains on the surface of the nanomotors through the "grafting from" method. The diffusion coefficient and velocity of the JNMs (before and after the PCL reaction) in the aqueous solution of 1, 2, 3, 5, and 10% (w/w) hydrogen peroxide and in the presence of different concentrations of NaCl solutions (0, 5, and 10% (w/v)) were investigated by mean square displacement analysis for single-particle or collective motions of JNMs. In addition, the simultaneous effect of an external magnetic field and the NaCl concentration on the movement direction of JNMs was also evaluated in the presence of hydrogen peroxide (10%). Increasing the ionic strength through NaCl addition permits the JNMs to move with relatively lower amounts of fuel [ i.e. , 2% (w/w)]. The collective motion investigation of the JNMs showed the highest speed in the media with 10% (w/w) hydrogen peroxide and 5% (w/v) NaCl solution (about 1215.78 μm 2 /s) due to the surfactant effect of the Janus architecture.

Imaging of cells is a challenging problem as they do not appreciably change the intensity of the illuminating light. Interferometry-based methods to do this task suffer from high sensitivity to environmental vibrations. We introduce scanning diffractometry as a simple non-contact and vibration-immune methodology for quantitative phase imaging. Fresnel diffractometry by a phase step has led to several applications such as high-precision measurements of displacement. Additional scanning may lead to 3D imaging straightforwardly. We apply the technique to acquire 3D images of holographic grating, red blood cell, neuron, and sperm cell. Either visibility of the diffraction fringes or the positions of extrema may be used for phase change detection. The theoretical analysis through the Fresnel diffraction from one-dimensional phase step is presented and the experimental results are validated with digital holographic microscopy. The presented technique can be suggested to serve as a robust device for 3D phase imaging and biomedical measurements.

Bacteria propel and change direction by rotating long, helical filaments, called flagella. The number of flagella, their arrangement on the cell body and their sense of rotation hypothetically determine the locomotion characteristics of a species. The movement of the most rapid microorganisms has in particular remained unexplored because of additional experimental limitations. We show that magnetotactic cocci with two flagella bundles on one pole swim faster than 500 µm·s -1 along a double helical path, making them one of the fastest natural microswimmers. We additionally reveal that the cells reorient in less than 5 ms, an order of magnitude faster than reported so far for any other bacteria. Using hydrodynamic modeling, we demonstrate that a mode where a pushing and a pulling bundle cooperate is the only possibility to enable both helical tracks and fast reorientations. In this paper, we measure the proximity effect of a flat wall on the sedimentation of colloidal particles by the use of digital holographic microscopy (DHM). DHM provides quantitative phase contrast and three-dimensional (3D) imaging in arbitrary time scales that are suitable to investigate various phenomena, including dynamic behavior of colloids. 3D information is obtained by post-processing of the recorded digital holograms through the angular spectrum propagation approach for numerical reconstruction. We have measured the trajectory and velocity of spherical colloids in various distances from a flat wall while they sediment under gravity. The results are in agreement with theoretical studies of Goldman et al. [Chem. Eng. Sci.22, 637 (1967)CESCAC0009-250910.1016/0009-2509(67)80047-2]. We observed an increase in the drag force due to the proximity to the flat wall, which was effective for distances up to several particle sizes. The presented method is capable of being used for different applications in study of colloidal dynamics where 3D information in real time is required.

The aim of this paper is to introduce digital holographic microscopy (DHM) as a non-contact, inexpensive, and non-abrasive method for 3D surface characterization of polymeric nanocomposites. A common-path and vibration-immune Mirau system with a microsphere-assisted arrangement is utilized to increase the lateral resolution of the images. The characterization is performed through the measurement of roughness parameters of the surfaces, which are derived from the recorded holograms. Pure poly(triazole-amide-imide) (PTAI) and PTAI nanocomposite reinforeced with surface modified TiO 2 nanoparticles (MN-TiO 2 ) are used and compared. The experimental results show the potential of the presented method to serve as an alternative for expensive surface measurement devices such as stylus profiler and atomic force microscope (AFM) for polymeric surface characterization., (Copyright © 2017 Elsevier B.V. Using particle-tracking techniques, the translational and rotational diffusion of paralyzed E. coli with and without flagella are studied experimentally. The position and orientation of the bacteria are tracked in the lab frame and their corresponding mean-square displacements are analyzed in the lab frame and in the body frame to extract the intrinsic anisotropic translational diffusion coefficients as well as the rotational diffusion coefficient for both strains. The deflagellated strain is found to show an anisotropic translational diffusion, with diffusion coefficients that are compatible with theoretical estimates based on its measured geometrical features. The corresponding translational diffusion coefficients of the flagellated strain have been found to be reduced as compared to those of the deflagellated counterpart. Similar results have also been found for the rotational diffusion coefficients of the two strains. Our results suggest that the presence of flagella --even as a passive component-- has a significant role in the dynamics of E. coli, and should be taken into account in theoretical studies of its motion.

Maximum trapping efficiency in optical tweezers occurs close to the coverslip because spherical aberration owing to a mismatch in the refractive indices of the specimen (water) and the immersion oil dramatically decreases the trap efficiency as the trap depth increases. Measuring the axial trap efficiency at various tube lengths by use of an oil-immersion objective has shown that such an aberration can be balanced by another source of spherical aberration, leading to a shift in the position of the maximum efficiency in the Z direction. For a 1.1 microm polystyrene bead we could achieve the maximal efficiency at a depth of 70 microm, whereas the trap was stable up to a depth of 100 microm.
