Abstract

In vivo metabolite quantification by short echo time MR spectroscopy is a challenge for which various methods have been proposed. In this study, the reproducibility of quantification outcomes is questioned at three distinct levels: (i) between-software (LCModel and cQUEST), (ii) withinsoftware (with different parameter sets), and (iii) across software executions (when the fitting algorithm uses random seeds, like cQUEST). After running multiple quantification tasks on a dedicated platform (VIP), metrics from Bland-Altman analysis were used to assess the variability of outcomes in signals acquired on a lysolecithin rat model, from a study on demyelination. Results show substantial variations at the three levels, allowing for more potent analyses than from a single parameter set / single software point of view.

12. PET imaging of neuroinflammation: any credible alternatives to TSPO yet?

Abstract

Over the last decades, the role of neuroinflammation in neuropsychiatric conditions has attracted an exponentially growing interest. A key driver for this trend was the ability to image brain inflammation in vivo using PET radioligands targeting the Translocator Protein 18 kDa (TSPO), which is known to be expressed in activated microglia and astrocytes upon inflammatory events as well as constitutively in endothelial cells. TSPO is a mitochondrial protein that is expressed mostly by microglial cells upon activation but is also expressed by astrocytes in some conditions and constitutively by endothelial cells. Therefore, our current understanding of neuroinflammation dynamics is hampered by the lack of alternative targets available for PET imaging. We performed a systematic search and review on radiotracers developed for neuroinflammation PET imaging apart from TSPO. The following targets of interest were identified through literature screening (including previous narrative reviews): P2Y12R, P2X7R, CSF1R, COX (microglial targets), MAO-B, I2BS (astrocytic targets), CB2R & S1PRs (not specific of a single cell type). We determined the level of development and provided a scoping review for each target. Strikingly, astrocytic biomarker MAO-B has progressed in clinical investigations the furthest, while few radiotracers (notably targeting S1P1Rs, CSF1R) are being implemented in clinical investigations. Other targets such as CB2R and P2X7R have proven disappointing in clinical studies (e.g. poor signal, lack of changes in disease conditions, etc.). While astrocytic targets are promising, development of new biomarkers and tracers specific for microglial activation has proven challenging.

Abstract

To improve our understanding of the brain microstructure, high-resolution 3D imaging is used to complement classical 2D histological assessment techniques. X-ray computed tomography allows high-resolution 3D imaging, but requires methods for enhancing contrast of soft tissues. Applying contrast-enhancing staining agents (CESAs) ameliorates the X-ray attenuating properties of soft tissue constituents and is referred to as contrast-enhanced computed tomography (CECT). Despite the large number of chemical compounds that have successfully been applied as CESAs for imaging brain, they are often toxic for the researcher, destructive for the tissue and without proper characterization of affinity mechanisms. We evaluated two sets of chemically related CESAs (organic, iodinated: Hexabrix and CA4+ and inorganic polyoxometalates: 1:2 hafnium-substituted Wells-Dawson phosphotungstate and Preyssler anion), for CECT imaging of healthy murine hemispheres. We then selected the CESA (Hexabrix) that provided the highest contrast between gray and white matter and applied it to a cuprizone-induced demyelination model. Differences in the penetration rate, effect on tissue integrity and affinity for tissue constituents have been observed for the evaluated CESAs. Cuprizone-induced demyelination could be visualized and quantified after Hexabrix staining. Four new non-toxic and non-destructive CESAs to the field of brain CECT imaging were introduced. The added value of CECT was shown by successfully applying it to a cuprizone-induced demyelination model. Abstract

Neurological outcome after ischemic stroke depends on residual salvageable brain tissue at the time of recanalization. Head down tilt 15° (HDT15) was proven effective in reducing infarct size and improving functional outcome in rats with transient middle cerebral artery occlusion (t‐MCAO) by increasing cerebral perfusion within the ischemic penumbra. In this pooled analysis, individual animal‐level data from three experimental series were combined in a study population of 104 t‐MCAO rats (45 in HDT15 group and 59 in flat position group). Co‐primary outcomes were infarct size and functional outcome at 24 h in both groups. The secondary outcome was hemodynamic change induced by HDT15 in ischemic and non‐ischemic hemispheres in a subgroup of animals. Infarct size at 24 h was smaller in HDT15 group than in flat position group (absolute mean difference 31.69 mm3, 95% CI 9.1–54.2, Cohen's d 0.56, p = 0.006). Functional outcome at 24 h was better in HDT15 group than in flat position group (median [IQR]: 13[10–16] vs. 11), with a shift in the distribution of the neurobehavioural scores in favour of HDT15. Mean cerebral perfusion in the ischemic hemisphere was higher during HDT15 than before its application (Perfusion Unit [P.U.], mean ± SD: 52.5 ± 19.52 P.U. vs. 41.25 ± 14.54 P.U., mean of differences 13.36, 95% CI 7.5–19.18, p = 0.0002). Mean cerebral perfusion in the non‐ischemic hemisphere before and during HDT15 was unchanged (P.U., mean ± SD: 94.1 ± 33.8 P.U. vs. 100.25 ± 25.34 P.U., mean of differences 3.95, 95%, CI −1.9 to 9.6, p = 0.1576). This study confirmed that HDT15 improves the outcome in t‐MCAO rats by promoting cerebral perfusion in the ischemic territory, without disrupting hemodynamics in non‐ischemic areas. Abstract

PURPOSE: The prototypical TSPO radiotracer (R)-[11C]PK11195 has been used in humans for more than thirty years to visualize neuroinflammation in several pathologies. Alternative radiotracers have been developed to improve signal-to-noise ratio and started to be tested clinically in 2008. Here we examined the scientific value of these "(R)-[11C]PK11195 challengers" in clinical research to determine if they could supersede (R)-[11C]PK11195.METHODS: A systematic MEDLINE (PubMed) search was performed (up to end of year 2020) to extract publications reporting TSPO PET in patients with identified pathologies, excluding studies in healthy subjects and methodological studies.RESULTS: Of the 288 publications selected, 152 used 13 challengers, and 142 used (R)-[11C]PK11195. Over the last 20 years, the number of (R)-[11C]PK11195 studies remained stable (6 ± 3 per year), but was surpassed by the total number of challenger studies for the last 6 years. In total, 3914 patients underwent a TSPO PET scan, and 47% (1851 patients) received (R)-[11C]PK11195. The 2 main challengers were [11C]PBR28 (24%-938 patients) and [18F]FEPPA (11%-429 patients). Only one-in-ten patients (11%-447) underwent 2 TSPO scans, among whom 40 (1%) were scanned with 2 different TSPO radiotracers.CONCLUSIONS: Generally, challengers confirmed disease-specific initial (R)-[11C]PK11195 findings. However, while their better signal-to-noise ratio seems particularly useful in diseases with moderate and widespread neuroinflammation, most challengers present an allelic-dependent (Ala147Thr polymorphism) TSPO binding and genetic stratification is hindering their clinical implementation. As new challengers, insensitive to TSPO human polymorphism, are about to enter clinical evaluation, we propose this systematic review to be regularly updated (living review).

Abstract

The purpose of this study is to propose and validate a preclinical in vivo magnetic resonance imaging (MRI) tool to monitor neuroinflammation following ischemic stroke, based on injection of a novel multimodal nanoprobe, NanoGd, specifically designed for internalization by phagocytic cells. First, it is verified that NanoGd is efficiently internalized by microglia in vitro. In vivo MRI coupled with intravenous injection of NanoGd in a permanent middle cerebral artery occlusion mouse model results in hypointense signals in the ischemic lesion. In these mice, longitudinal two‐photon intravital microscopy shows NanoGd internalization by activated CX3CR1‐GFP/+ cells. Ex vivo analysis, including phase contrast imaging with synchrotron X‐ray, histochemistry, and transmission electron microscopy corroborate NanoGd accumulation within the ischemic lesion and uptake by immune phagocytic cells. Taken together, these results confirm the potential of NanoGd‐enhanced MRI as an imaging biomarker of neuroinflammation at the subacute stage of ischemic stroke. As far as it is known, this work is the first to decipher the working mechanism of MR signals induced by a nanoparticle passively targeted at phagocytic cells by performing intravital microscopy back‐to‐back with MRI. Furthermore, using a gadolinium‐based rather than an iron‐based contrast agent raises future perspectives for the development of molecular imaging with emerging computed tomography technologies., Neuroinflammation at the subacute stage of stroke is challenging to monitor in vivo. Here, a novel gadolinium‐based fluorophore‐grafted nanoprobe named NanoGd, specifically designed for internalization by phagocytic cells, is worked with. An in vivo magnetic resonance imaging (MRI) tool is proposed to monitor neuroinflammation following ischemic stroke in mice, which is validated using two‐photon intravital imaging back‐to‐back with MRI for the first time.

Abstract

Preregistration In 2016, more than 1.9 billion adults were overweight. Estimations based on current trends suggest that approximatively 25 % of the population will be obese in 2025. Better understanding of obesity pathophysiology will help to develop future therapeutics. Using antagonists, agonists or knock-out animals, serotonin subtype 6 receptor (5-HT6), have shown to be critically involved in appetite reduction and weight loss. However, it is not known if the pathological cascade triggered by obesity modifies 5-HT6 receptor density in the brain. In this study, we aim to explore the influence of a diet-induced obesity (DIO) rat model on the 5-HT6 receptor expression using the PET radiotracer [18F]2FNQ1P. The primary goal is to detect in-vivo changes in the cerebral uptake of the 5-HT6 radiotracer changes before and after a 10-week DIO, while measuring whole-body fat accumulation with MRI. Using control animals under normal diet, and a genetic obesity model (Zucker rats), the secondary goal is to compare final 5-HT6 receptor levels between the three groups (DIO, genetic obesity and control diet). In this stage 1 registered report, we provide preliminary results showing the feasibility of our study: i) DIO model was tested and led to 25.5 % increase in fat measured on MRI, ii) regional 5-HT6 receptor densities were reliably quantified in PET test-retests following tariquidar pre-injection to increase blood-brain barrier passage of the radiotracer [18F]2FNQ1P. This multimodal imaging study is expected to yield original results for the understanding of the physiopathology of obesity and the implication of 5-HT6 receptors in this disease.

Abstract

International audience; Choroid plexus (ChPs) are involved in the early inflammatory response that occurs in many brain disorders. However, the activation of immune cells within the ChPs in response to neuroinflammation is still largely unexplored in-vivo. There is therefore a crucial need for developing imaging tool that would allow the non-invasive monitoring of ChP involvement in these diseases. Magnetic resonance imaging (MRI) coupled with superparamagnetic particles of iron oxide (SPIO) is a minimally invasive technique allowing to track phagocytic cells in inflammatory diseases. Our aim was to investigate the potential of ultrasmall SPIO (USPIO)-enhanced MRI to monitor ChP involvement in-vivo in a mouse model of neuroinflammation obtained by intraperitoneal administration of lipopolysaccharide. Using high resolution MRI, we identified marked USPIO-related signal drops in the ChPs of animals with neuroinflammation compared to controls. We confirmed these results quantitatively using a 4-points grading system. Ex-vivo analysis confirmed USPIO accumulation within the ChP stroma and their uptake by immune cells. We validated the translational potential of our approach using the clinically-applicable USPIO Ferumoxytol. MR imaging of USPIO accumulation within the ChPs may serve as an imaging biomarker to study ChP involvement in neuroinflammatory disorders that could be applied in a straightforward way in clinical practice.

Abstract

White-matter injury leads to severe functional loss in many neurological diseases. Myelin staining on histological samples is the most common technique to investigate white-matter fibers. However, tissue processing and sectioning may affect the reliability of 3D volumetric assessments. The purpose of this study was to propose an approach that enables myelin fibers to be mapped in the whole rodent brain with microscopic resolution and without the need for strenuous staining. With this aim, we coupled inline (propagation-based) X-ray phase-contrast tomography (XPCT) to ethanol-induced brain sample dehydration. We here provide the proof-of-concept that this approach enhances myelinated axons in rodent and human brain tissue. In addition, we demonstrated that white-matter injuries could be detected and quantified with this approach, using three animal models: ischemic stroke, premature birth and multiple sclerosis. Furthermore, in analogy to diffusion tensor imaging (DTI), we retrieved fiber directions and DTI-like diffusion metrics from our XPCT data to quantitatively characterize white-matter microstructure. Finally, we showed that this non-destructive approach was compatible with subsequent complementary brain sample analysis by conventional histology. In-line XPCT might thus become a novel gold-standard for investigating white-matter injury in the intact brain. 35. Die Morphologie und räumliche Verteilung der Karstformen Niederösterreichs

Abstract

Der alpine Raum Niederösterreichs (NÖ), insbesondere die Nördlichen Kalkalpen und die Semmering Wechsel Region sind durch verschiedene Karstformen geprägt. Dolinen stellen die markantesten und häufigsten Karstformen dar. Bislang gibt es keine Synthese aller bekannten Karstformen in NÖ und keine Zuordnung zu den geologischen Einheiten. Neben dem wissenschaftlichen Aspekt, soll diese Arbeit als Hilfsmittel bei der Gefahrenabschätzung von Landnutzung, beispielsweise landwirtschaftlicher Natur, diverser Bauvorhaben aber auch zum Schutz der Karstlandschaften dienen. Ziel ist einerseits die Korrelation zwischen den stratigraphischen Einheiten und deren Verkarstungseigenschaften. Andererseits werden Morphologie und räumliche Verteilung der Karstformen empirisch ermittelt, beziehungsweise überprüft, ob diese bevorzugt an gewisse Lithologien gebunden sind. Es wird ein Überblick über das Ausmaß der Karstformen im alpinen Raum Niederösterreichs gegeben. Der Flächenanteil des verkarstungsfähigen Gesteins entspricht 12 % der Fläche Niederösterreichs, sowie 57 % des Untersuchungsgebietes. Methodisch wurden Daten über die diversen Karstformen im Rahmen von Geländebegehungen erhoben. Zudem wurden Daten aus sämtlichen weiteren publizierten Karten zusammengetragen sowie mittels Fernerkundung ermittelt. Der resultierende Datensatz wird visualisiert und im Anschluss eine Reihe statistischer Auswertungen an Dolinen durchgeführt. Die räumlichen Verteilungen der Dolinen sowie Dolinenvolumina wurden mittels Kernel Function ermittelt. Insgesamt werden 15.459 Dolinen, zwölf Glaziokarst-Hohlformen, fünf Poljen, 248 Ponore und 900 Karrenvorkommen ausgewertet. Die Gesamtfläche aller Dolinen beträgt 6,99 km², was 0,2 % der Fläche des Untersuchungsgebiets entspricht. Die statistischen Analysen der Dolinen bezüglich deren Tiefe, Fläche und Volumen zeigen eine linkssteile Verteilungskurve mit der Tendenz zur Kleinmaßstäbigkeit. Im Durchschnitt weisen die Dolinen eine Tiefe von 2,7 m, eine Fläche von 452 m² und ein Volumen von 2192 m³ auf. Die tiefste und größte Doline NÖs ist das Grünloch am Dürrenstein-Plateau, das 50 m Tiefe, 0,38 km² Fläche und 6,3 Mio. m³ Volumen aufweist. Die dominierenden Lithologien im Untersuchungsgebiet sind mit 55 % der Gesamtfläche aller verkarstungsfähigen Lithologien Dolomitgesteine. Die Kalkformationen nehmen nur etwa 28 % und die Gipse nur etwa 3 % ein. Die Anzahl an Dolinen ist mit 71 % in den Kalken deutlich höher als in den Dolomiten, in denen lediglich 13 % aller Dolinen ausgebildet sind. In den kaum verbreiteten Gipsen des Haselgebirges und der Werfen-Formation sind 9 % der Dolinen kartiert. Die Unterscheidung der verschiedenen Kalkformationen zeigt, dass mit 38 % die meisten Dolinen im Dachsteinkalk kartiert sind. Die Dichten der Dolinenvolumina unterscheiden sich lokal deutlich, wobei vor allem Gebiete in welchen Haselgebirge und Werfen-Formation, beziehungsweise Dachsteinkalk kartiert sind, hohe Dichten zeigen. Die meisten Dolinen sind in 1400 bis 1600 m Seehöhe ausgebildet, was mit dem Anstieg der durchschnittlichen Niederschlagsmenge mit steigender Seehöhe sowie vermutlich auch glazialer Überprägung korreliert. In einer semiquantitativen Tabelle wird die Verkarstungsfähigkeit der unterschiedlichen Lithologien klassifiziert. Aus dieser Quantifizierung der Oberflächenkarstformen in NÖ lässt sich ableiten, dass Kalke eine bessere Verkarstungsfähigkeit als Dolomite aufweisen und die meisten Karstformen im Dachsteinkalk ausgebildet sind. Neben den Kalken sind die Gipse des Haselgebirges und der Werfen-Formation extrem verkarstungsfähig., The Alpine area of Lower Austria, especially the Northern Calcareous Alps and the Semmering Wechsel region are characterized by numerous karst features. Dolines are the most prominent ones. So far, there is no other study, which merges all karst features in Lower Austria and allocates them to the geological units. Besides scientific aspects this thesis should provide authorities to assess potential hazard of land use, especially regarding agricultural use diverse infrastructural projects as well as the protection of karst landscapes. The aim of this thesis is the correlation between stratigraphical units and their karstification properties. Morphology and spatial distribution of karst features are empirically determined and checked if they are bound to specific lithologies. A land wide overview of the amount of karst features is given, focusing on the Alpine area of Lower Austria. The total area of soluble lithologies corresponds to 12 % of the total area of Lower Austria and 57 % of the study area. Several methods were used to gain data of different karst features. Data was collected during field work in the course of this thesis and the NÖ-Karst-Project. Further data was compiled from diverse published maps and from Airborne-Laser-Scanning calculations. The resulting dataset is visualized and a number of statistical analyses on dolines are performed. Kernel function is used to identify the morphology and spatial distribution of dolines and doline volumes. In total 15.459 dolines, twelve glaziokarstic depressions, five poljes, 248 ponors and 900 Karren features are analyzed. The distribution curve of statistical analyses of dolines regarding their depth, shape area and their volume point at the dominance of small-scale features. On average dolines are 2,7 m deep, have an area of 452 m² and a volume of 2192 m³. The Grünloch on the Dürrenstein-Plateau with a depth of about 50 m, an area of 0,38 km² and a volume of about 6,3 Mio. m³ represents the deepest and biggest doline in the study area. With an amount of 55 % dolomite dominated stratigraphic units are the most common karstic lithologies in the Alpine area of Lower Austria. Limestones occupy 28 %, gypsum units 3 % of the study area. The amount of mapped dolines is highest in limestone units (71 %), far less karst features are mapped in doline units (13 %). Even though gypsums of the Haselgebirge and Werfen-Formation unit are only mapped in small areas of Lower Austria, a significant number of dolines (9 %) is mapped in this unit. The comparison of different limestone formations shows, that Dachsteinkalk represents the most dominant lithological unit regarding the number of dolines. Doline volume densities significantly differ locally. High densities occur especially in regions where the Haselgebirge and Werfen-Formation or Dachsteinkalk are mapped. The height distribution of dolines underlines, that most dolines are formed between 1400 to 1600 m above see level, which can be correlated to the average rise of the amount of precipitation with rising altitude as well as glacial overprint during Pleistocene. The capacity of karstification of different lithological units has been classified in a semiquantitative table. This quantification of surface karst features in Lower Austria leads to the conclusion that the karstification properties are better in limestone dominated stratigraphic units, such as the Dachsteinkalk, than in dolomite dominated lithologies. Beside limestones also the gypsum bearing units Haselgebirge and Werfen-Formation have been identified as extremely karstifed.

Abstract

International audience; Several biological processes are involved in dementia, and fibrillar aggregation of misshaped endogenous proteins appears to be an early hallmark of neurodegenerative disease. A recently developed means of studying neurodegenerative diseases is magnetic resonance elastography (MRE), an imaging technique investigating the mechanical properties of tissues. Although mechanical changes associated with these diseases have been detected, the specific signal of fibrils has not yet been isolated in clinical or preclinical studies. The current study aims to exploit the fractal‐like properties of fibrils to separate them from nonaggregated proteins using a multi‐frequency MRE power law exponent in a phantom study. Two types of fibril, α‐synuclein (α‐Syn) and amyloid‐β (Aβ), and a nonaggregated protein, bovine serum albumin, used as control, were incorporated in a dedicated nondispersive agarose phantom. Elastography was performed at multiple frequencies between 400 and 1200 Hz. After 3D‐direct inversion, storage modulus (G'), phase angle (ϕ), wave speed and the power law exponent (y) were computed. No significant changes in G' and ϕ were detected. Both α‐Syn and Aβ inclusions showed significantly higher y values than control inclusions (P = 0.005) but did not differ between each other. The current phantom study highlighted a specific biomechanical effect of α‐Syn and Aβ aggregates, which was better captured with the power law exponent derived from multi‐frequency MRE than with single frequency‐derived parameters.
