Multiple myeloma (MM) is a cancer of terminally differentiated plasma cells. MM remains incurable, but overall survival of patients has progressively increased over the past two decades largely due to novel agents such as proteasome inhibitors (PI) and the immunomodulatory agents. While these therapies are highly effective, MM patients can be de novo resistant and acquired resistance with prolonged treatment is inevitable. There is growing interest in early, accurate identification of responsive versus non-responsive patients; however, limited sample availability and need for rapid assays are limiting factors. Here, we test dry mass and volume as label-free biomarkers to monitor early response of MM cells to treatment with bortezomib, doxorubicin, and ultraviolet light. For the dry mass measurement, we use two types of phase-sensitive optical microscopy techniques: digital holographic tomography and computationally enhanced quantitative phase microscopy. We show that human MM cell lines (RPMI8226, MM.1S, KMS20, and AMO1) increase dry mass upon bortezomib treatment. This dry mass increase after bortezomib treatment occurs as early as 1 h for sensitive cells and 4 h for all tested cells. We further confirm this observation using primary multiple myeloma cells derived from patients and show that a correlation exists between increase in dry mass and sensitivity to bortezomib, supporting the use of dry mass as a biomarker. The volume measurement using Coulter counter shows a more complex behavior; RPMI8226 cells increase the volume at an early stage of apoptosis, but MM.1S cells show the volume decrease typically observed with apoptotic cells. Altogether, this cell study presents complex kinetics of dry mass and volume at an early stage of apoptosis, which may serve as a basis for the detection and treatment of MM cells.

A YOLOv5-based YOLOv5-KE unmanned aerial vehicle (UAV) image detection algorithm is proposed to address the low detection accuracy caused by the small size, high density, and overlapping leaves of Ambrosia trifida targets in UAV images. The YOLOv5-KE algorithm builds upon the YOLOv5 algorithm by adding a micro-scale detection layer, adjusting the hierarchical detection settings based on k-Means for Anchor Box, improving the loss function of CIoU, reselecting and improving the detection box fusion algorithm. Comparative validation experiments of the YOLOv5-KE algorithm for Ambrosia trifida recognition were conducted using a self-built dataset. The experimental results show that the best detection accuracy of Ambrosia trifida in UAV images is 93.9%, which is 15.2% higher than the original YOLOv5. Furthermore, this algorithm also outperforms other existing object detection algorithms such as YOLOv7, DC-YOLOv8, YOLO-NAS, RT-DETR, Faster RCNN, SSD, and Retina Net. Therefore, YOLOv5-KE is a practical algorithm for detecting Ambrosia trifida under complex field conditions. This algorithm shows good potential in detecting weeds of small, high-density, and overlapping leafy targets in UAV images, it could provide technical reference for the detection of similar plants.

Bromine chemistry is responsible for the catalytic ozone destruction in the atmosphere. The heterogeneous reactions of sea‐salt aerosols are the main abiotic sources of reactive bromine in the atmosphere. Here, we present a novel mechanism for the activation of bromide ions (Br−) by O2 and H2O in the absence of additional oxidants. The laboratory and theoretical calculation results demonstrated that under dark conditions, Br−, O2 and H3O+ could spontaneously generate Br and HO2 radicals through a proton–electron transfer process at the air–water interface and in the liquid phase. Our results also showed that light and acidity could significantly promote the activation of Br− and the production of Br2. The estimated gaseous Br2 production rate was up to 1.55×1010 molecules cm−2 ⋅ s−1 under light and acidic conditions; these results showed a significant contribution to the atmospheric reactive bromine budget. The reactive oxygen species (ROS) generated during Br− activation could promote the multiphase oxidation of SO2 to produce sulfuric acid, while the increase in acidity had a positive feedback effect on Br− activation. Our findings highlight the crucial role of the proton‐electron transfer process in Br2 production; here, H3O+ facilitates the activation of Br− by O2, serves as a significant source of atmospheric reactive bromine and exerts a profound impact on the atmospheric oxidation capacity. Substantial NOx emission mitigation is crucial for the synergistic reduction of particulate matter and ozone (O3) pollution in China. The traditional air quality model does not consider heterogeneous HONO chemistry, leading to uncertainties in estimating the benefits of NOx control. Previous studies have shown that the parameterization of heterogeneous HONO formation increases both the simulated value of sulfate–nitrate–ammonium (SNA) and that of O3, thus adding the heterogeneous reactions of HONO into air quality models inevitably leads to changes in the estimated benefits of NOx abatement. Here we investigated the changes in SNA and O3 concentrations from NOx emission reduction before and after adding heterogeneous HONO reactions in the Community Multi-Scale Air Quality (CMAQ) model. Including heterogeneous HONO reactions in the simulation improved the benefits of NOx reduction in terms of SNA control in winter. With 80% NOx reduction, the reduction in SNA increased from 36.9% without considering heterogeneous HONO reactions to 42.8% with heterogeneous HONO chemistry. The reduction in the maximum daily 8h average (MDA8) O3 in summer caused by NOx reduction increased slightly from 4.7% to 5.2% after adding heterogeneous HONO reactions. The results in this study highlight the enhanced effectiveness of NOx controls for the reduction of SNA and O3 after considering heterogeneous HONO formation in a complex chemical ambient, demonstrating the importance of NOx controls in reducing PM2.5 and O3 pollution in China. Nitrogen dioxide (NO2) is a highly photochemically reactive gas, has a lifetime of only a few hours, and at high concentrations is harmful to human beings. Therefore, it is important to monitor NO2 with high-precision, time-resolved instruments. To this end, a Pandora spectrometer has been installed on the roof of the laboratory building of the Aerospace Information Research Institute of the Chinese Academy of Sciences in the Olympic Park, Beijing, China. The concentrations of trace gases (including NO2 , HCHO , O3) measured with Pandora are made available through the open-access Pandora database (https://data.pandonia-global-network.org/Beijing-RADI/Pandora171s1/ , last access: 11 July 2023). In this paper, an overview is presented of the Pandora total and tropospheric NO2 vertical column densities (VCDs) and surface concentrations collected during the first year of operation, i.e., from August 2021 to July 2022. The data show that NO2 concentrations were high in the winter and low in the summer, with a diurnal cycle where the concentrations reached a minimum during the daytime. The concentrations were significantly lower during the 2022 Winter Olympics in Beijing, showing the effectiveness of the emission control measures during that period. The Pandora observations show that during northerly winds, clean air is transported to Beijing with low NO2 concentrations, whereas during southerly winds, pollution from surrounding areas is transported to Beijing and NO2 concentrations are high. The contribution of tropospheric NO2 to the total NO2 VCD varies significantly on daily to seasonal timescales; i.e., monthly averages vary between 50 % and 60 % in the winter and between 60 % and 70 % in the spring and autumn. A comparison of Pandora-measured surface concentrations with collocated in situ measurements using a Thermo Scientific 42i-TL analyzer shows that the Pandora data are low and that the relationship between Pandora-derived surface concentrations and in situ measurements is different for low and high NO2 concentrations. Explanations for these differences are offered in terms of measurement techniques and physical (transport) phenomena. The use of Pandora total and tropospheric NO2 VCDs for validation of collocated TROPOspheric Monitoring Instrument (TROPOMI) data, resampled to 100 m × 100 m , shows that although on average the TROPOMI VCDs are slightly lower, they are well within the expected error for TROPOMI of 0.5 Pmolec.cm-2 + (0.2 to 0.5) ⋅ VCDtrop (1 Pmolec.cm-2 = 1 × 10 15 moleccm-2). The location of the Pandora instrument within a sub-orbital TROPOMI pixel of 3.5 km × 5.5 km may result in an error in the TROPOMI-derived tropospheric NO2 VCD between 0.223 and 0.282 Pmolec.cm-2 , i.e., between 1.7 % and 2 %. Soot is a crucial component of aerosols in the atmosphere. Understanding the hygroscopicity of soot particles is important for studying their role as cloud condensation nuclei (CCN) as well as their chemical behavior and atmospheric lifetime. However, there is still a lack of comprehensive understanding regarding the factors that determine the hygroscopic properties of soot. In this work, the hygroscopic behavior of soot particles generated from different types of fuel combustion and aged with SO 2 for varying durations was measured by a vapor sorption analyzer. Various characterizations of soot were conducted to understand the key factors that influence the hygroscopic properties of soot. It was found that water-soluble substances in soot facilitate the completion of monolayer water adsorption at low relative humidity and increase the number of water adsorption layers at high relative humidity. On the other hand, soot prepared from fuel burning typically lacks water-soluble inorganic ions, and their hygroscopicity is primarily influenced by organic carbon (OC) and microstructure. Furthermore, the hygroscopicity of soot can be enhanced by the formation of sulfate due to heterogeneous oxidation of SO 2. These finding sheds light on the critical factors that affect soot hygroscopicity during water adsorption and allows for estimating the interaction between water molecules and soot particles in a humid atmosphere. Black carbon (BC) is a crucial component of aerosols in the atmosphere. Understanding the hygroscopicity of BC particles is important for studying their role as cloud condensation nuclei (CCN) and ice nuclei (IN), as well as their chemical behavior and atmospheric lifetime. However, there is still a lack of comprehensive understanding regarding the factors that determine the hygroscopic properties of fresh BC. N2O5 is an important intermediate in the atmospheric nitrogen cycle. Using a flow tube reactor, N2O5 was found to be released from the TiO2 surface during the photocatalytic oxidation of NO2, revealing a previously unreported source of N2O5. The rate of N2O5 release from TiO2 was dependent on the initial NO2 concentration, relative humidity, O2/N2 ratio, and irradiation intensity. Experimental evidence and quantum chemical calculations showed that NO2 can react with the surface hydroxyl groups and the generated electron holes on the TiO2, followed by combining with another NO2 molecule to form N2O5. The latter was physisorbed on TiO2 and had a low adsorption energy of −0.13 eV. Box model simulations indicated that the new source of N2O5 released from TiO2 can increase the daytime N2O5 concentration by up to 20 % in urban areas if abundant TiO2‐containing materials and high NOx concentrations were present.
