The appropriate flowering time is of great significance for crop yield. In the cropping pattern of “rice-rice-oil” in southern region, researching on the flowering period of Brassica napus can provide a theoretical basis for the breeding of early maturing rape varieties. The AP2 family transcription factors in rape are widely involved in the growth and development and play an important role during flower development. However, there are few studies exploring the regulation of AP2 at the microRNA level. In order to investigate the regulatory functions of the miR172 precursor (pre-miR72) and mature body (miR172) on AP2 gene in rape, the regulatory elements of miR172 and AP2 promoters were predicted based on bioinformatics, then the evolutionary relationship of six rape AP2 genes and the targeting relationship between miR172 and AP2 were analyzed, and the expression patterns of AP2, miR172 and pre-miR172 in different tissues of early and late maturing rape were detected by qRT-PCR. Finally, the correlation between miR172 abundance and AP2 expression level was studied, as well as the correlation between pre-miR172 and miR172. The results were as follows: (1) Both miR172 and AP2 promoter regions had cis-elements that regulated flower development. (2) The six AP2sequences holded the strong purification selection, and they were the target genes of miR172 based on their binding sites for miR172. (3) miR172 family could promote the flowering of early maturing rape by increasing AP2 expression levels, except for miR172d. In late maturing rape, miR172a and miR172c performed weakly in flowering, while miR172b and miR172d worked together to reduce the expression level of AP2 to inhibit flowering. (4) The pre-miR172 family had a promoting effect on the expression level of miR172 family in early maturing rape; in late maturing rape, pre-miR172a and pre-miR172b exerted positive regulation on the formation of their mature bodies, while pre-miR172c and pre-miR172d exerted the opposite effects. After overexpression of pre-miR172, the expression patterns of miR172 and AP2 remained consistent with the above results, confirming the regulatory function of pre-miR172 on miR172 and AP2. The results of this study enrich the functional regulation pathway of rape AP2 gene, and provide new ideas for the study of gene regulatory function.

Hippo signaling has been recognized as a key tumor suppressor pathway. Here, we perform a comprehensive molecular characterization of 19 Hippo core genes in 9,125 tumor samples across 33 cancer types using multidimensional "omic" data from The Cancer Genome Atlas. We identify somatic drivers among Hippo genes and the related microRNA (miRNA) regulators, and using functional genomic approaches, we experimentally characterize YAP and TAZ mutation effects and miR-590 and miR-200a regulation for TAZ. Hippo pathway activity is best characterized by a YAP/TAZ transcriptional target signature of 22 genes, which shows robust prognostic power across cancer types. Our elastic-net integrated modeling further reveals cancer-type-specific pathway regulators and associated cancer drivers. Our results highlight the importance of Hippo signaling in squamous cell cancers, characterized by frequent amplification of YAP/TAZ, high expression heterogeneity, and significant prognostic patterns. This study represents a systems-biology approach to characterizing key cancer signaling pathways in the post-genomic era.

BACKGROUND: The pickling process with NaCl is an essential step for pork preservation. This study aimed to investigate the effect of different ultrasonic intensities of tri‐frequency simultaneous ultrasound (TSIU) pickling on the NaCl content and quality of pork (longissimus dorsi). After 30 min pickling, the NaCl content, moisture content, pickling yield, cooking loss, textural properties, color, pH, moisture migration and distribution as well as microstructure of pork were assessed. RESULTS: Results showed that among all the ultrasonic treatment intensities (85–150 W L−1), the NaCl content of the sample pickled by an intensity of 101.3 W L−1 was higher than that of other intensities. TSIU 101.3 W L−1 showed 59.95% higher NaCl content than the control sample. In addition, the sample treated with TSIU of 101.3 W L−1 had higher pickling yield and moisture content, better textural properties of pork (including hardness and chewiness), and less cooking loss. The results of the low‐field nuclear magnetic resonance showed that, compared with the control group, the relaxation time T21 of the ultrasound‐assisted pickling samples increased, while the proportion of T22 (A22) reduction ranged from 175.0% to 379.9%. The microstructure designated that the ultrasonic treatment could facilitate changes in meat texture. CONCLUSION: Ultrasound marination of different intensities promoted the diffusion of NaCl and affected the quality of pork tenderloins. The TSIU at 101.3 W L−1 could better accelerate NaCl transport and homogeneous distribution on meat, thereby improving the sample quality. © 2024 Society of Chemical Industry. [ABSTRACT FROM AUTHOR]

This paper addresses the active target protection problem in a three-player (Target–Attacker–Defender, TAD) conflict by proposing a cooperative integrated guidance and control (IGC) strategy. Unlike previous studies that have designed guidance and control loops separately, this work establishes an IGC model by linearizing both the translational motion and the rotational motion of the vehicles, thereby generating actuator commands directly. This model integrates the kinematics and short-period dynamics, providing a more comprehensive and accurate representation of the vehicles' characteristics. Based on the linearization and order reduction, differential game theory and the sweep method are employed to derive and analytically solve the Riccati differential equation, yielding an optimal control strategy with an explicit expression. The theoretical rigor of the proposed approach is ensured through a proof of optimality sufficiency. Furthermore, factors influencing the computational accuracy of the Riccati equation solution, including the singular values of the control matrix and condition numbers of the solution matrix, are analyzed. Taking into account the dynamic response and limitations of the actuators, numerical simulations demonstrate the effectiveness and superiority of the proposed IGC strategy in intercepting the attacker and protecting the target compared to traditional separated guidance and control designs. Protein ubiquitination is a dynamic and reversible process of adding single ubiquitin molecules or various ubiquitin chains to target proteins. Here, using multidimensional omic data of 9,125 tumor samples across 33 cancer types from The Cancer Genome Atlas, we perform comprehensive molecular characterization of 929 ubiquitin-related genes and 95 deubiquitinase genes. Among them, we systematically identify top somatic driver candidates, including mutated FBXW7 with cancer-type-specific patterns and amplified MDM2 showing a mutually exclusive pattern with BRAF mutations. Ubiquitin pathway genes tend to be upregulated in cancer mediated by diverse mechanisms. By integrating pan-cancer multiomic data, we identify a group of tumor samples that exhibit worse prognosis. These samples are consistently associated with the upregulation of cell-cycle and DNA repair pathways, characterized by mutated TP53, MYC/TERT amplification, and APC/PTEN deletion. Our analysis highlights the importance of the ubiquitin pathway in cancer development and lays a foundation for developing relevant therapeutic strategies.

Metabolic reprogramming provides critical information for clinical oncology. Using molecular data of 9,125 patient samples from The Cancer Genome Atlas, we identified tumor subtypes in 33 cancer types based on mRNA expression patterns of seven major metabolic processes and assessed their clinical relevance. Our metabolic expression subtypes correlated extensively with clinical outcome: subtypes with upregulated carbohydrate, nucleotide, and vitamin/cofactor metabolism most consistently correlated with worse prognosis, whereas subtypes with upregulated lipid metabolism showed the opposite. Metabolic subtypes correlated with diverse somatic drivers but exhibited effects convergent on cancer hallmark pathways and were modulated by highly recurrent master regulators across cancer types. As a proof-of-concept example, we demonstrated that knockdown of SNAI1 or RUNX1-master regulators of carbohydrate metabolic subtypes-modulates metabolic activity and drug sensitivity. Our study provides a system-level view of metabolic heterogeneity within and across cancer types and identifies pathway cross-talk, suggesting related prognostic, therapeutic, and predictive utility.
