13. Research on Motion Characteristics in Opening Process of Aircraft Deformed Wing

Abstract

Deformed wing mechanism is a developed system of aircraft for miniaturization, which is convenient for box (cylinder) storage, transportation and launch, thus can save the storage and transportation space of aircraft and can improve the space utilization performance of flight system. In order to study wing dynamic opening motion characteristics during aircraft flight, based on gas theory and structural dynamics, this paper studies the dynamic process of the rotary opening movement of deformed wing system caused by the pressure acting on the piston because of propellant combustion with different charge amounts in the chamber, and analyzes relationship between the opening angle of inner and outer wings, angular velocity with time under different gas pressure. The results show that the opening motion of the deformed wing is sensitive to the initial conditions, and different motion characteristics is reflected at different gas pressures.

Abstract

Traditional Chinese medicine (TCM), as a significant complementary and alternative approach, has received increasing attention from medical practitioners worldwide and is now regarded as an important source for modern drug discovery owing to its special therapeutic efficiency and few side effects. In this work, a popular TCM, Ganoderma lingzhi (G. lingzhi), was used as a test drug to show how Raman spectroscopy combined with traditional chemical and biological assays could be used to monitor alternations in molecular events in G. lingzhi extract-induced cells with high sensitivity. The observations indicated that the Raman fingerprint of MDA-MB-231 cells after G. lingzhi triterpene treatment was significantly different in contrast to that of the control, and changes in the Raman bands showed that some important molecular events could be discerned, including protein denaturation, DNA fragmentation, and lipid metabolism disorder. Network pharmacology analysis further supplemented the drug's target and signaling pathways. Our work proposes an effective way for screening potential new drug products early from the complex chemical compositions of TCM, as well as for accelerating preclinical medicinal chemistry optimization cycles, playing a significant role in the development of new drugs. The complex physiological environment has brought significant challenges to the field of antitumor micelles. The ideal micelles must not only have an invisible surface to extend the circulation time but must also enhance the retention of drugs and cellular internalization at the tumor.Methods: A graded response micelle (RPPssD@IR780/DOC) was designed to self-assemble by cRGD-poly(β-amino esters)-polyethylene glycol-ss-distearoyl phosphatidylethanolamine (cRGD-PBAE-PEG-ss-DSPE) loaded with docetaxel (DOC) and IR-780 iodide (IR780). The micelles were designed to allow the PEG shell to prolong the blood circulation time in the body and effectively accumulate in the tumor. Subsequently, the acidic microenvironment of the tumor could transform the PBAE to hydrophilic, thereby increasing the size of micelles and exposing cyclic Arg-Gly-Asp (cRGD) peptides to increase the retention and cellular internalization of micelles in the tumor. After tumor cells had captured micelles, the high expression of glutathione in the cells prompted the release of DOC and IR780. Subsequently, the IR780 was stimulated by an 808-nm laser to generate local heat and reactive oxygen species (ROS) to synergize with DOC to treat the tumor.Results: In vitro and in vivo experimental results suggested that RPPssD@IR780/DOC was a potential photochemical effective for the treatment of tumors with non-negligible antitumor activity and good biocompatibility.Conclusion: A dual-response pH/redox delivery system with on-demand RGD exposure was designed to achieve photochemotherapy of tumors with good biosafety and antitumor effects.Graphical Abstract: Keywords: micelles, drug delivery, ligand hidden, size variable, photo-chemotherapy

Platelets are now known to maintain tumor vascular integrity. Therefore, inhibition of tumor-associated platelets may be an effective method to increase T cell infiltration and drug accumulation at tumor sites. Herein, we designed an ultrasound-responsive nitric oxide (NO) release nanosystem, SNO-HSA-PTX, which can release NO in response to ultrasound (US) irradiation, thereby inhibiting platelet function and opening the tumor vascular barrier, promoting drug accumulation and T cell infiltration.Methods: We evaluated the ability of SNO-HSA-PTX to release NO in response to US irradiation. We also tested the effect of SNO-HSA-PTX on platelet function. Plenty of studies including cytotoxicity, pharmacokinetics study, biodistribution, blood perfusion, T cell infiltration, in vivo antitumor efficacy and safety assessment were conducted to investigate the antitumor effect of SNO-HSA-PTX.Results: SNO-HSA-PTX with US irradiation inhibited tumor-associated platelets activation and induced openings in the tumor vascular barriers, which promoted the accumulation of SNO-HSA-PTX nanoparticles to the tumor sites. Meanwhile, the damaged vascular barriers allowed oxygen-carrying hemoglobin to infiltrate tumor regions, alleviating hypoxia of the tumor microenvironment. In addition, the intratumoral T cell infiltration was augmented, together with chemotherapy and NO therapy, which greatly inhibited tumor growth.Discus
