Abstract Objective The mainstream view in trait aggression research has regarded the structure as representing the latent cause of the cognitions, emotions, and behaviors that supposedly reflect its nature. Under network perspective, trait aggression is not a latent cause of its features but a dynamic system of interacting elements. The current study uses network theory to explain the structure of relationships between trait aggression features in juvenile offenders and their peers. Methods Network analysis was applied to investigate the dynamic system of trait aggression operationalized by the Buss-Perry Aggression Questionnaire in a sample of community youths (Mage = 17.46, N = 715) and juvenile offenders (Mage = 18.36, N = 834). Results The facet level networks showed that anger is a particularly effective mechanism for activating all other traits. In addition, anger was more strongly associated with physical aggression and the overall network strength was greater in juvenile delinquency networks than in their peers. The item level networks revealed that A4 and A6 exhibited the highest predictability and strength centrality in both samples. Also, the Bayesian network indicated that these two items were positioned at the highest level in the model. There are similarities and differences between juvenile delinquents and community adolescents in trait aggression. Conclusion Trait aggression was primarily activated by difficulty controlling one’s temper and feeling like a powder keg.

Abstract Boron-doped polycyclic aromatic hydrocarbons exhibit excellent optical properties, and regulating their photophysical processes is a powerful strategy to understand the luminescence mechanism and develop new materials and applications. Herein, an electrochemically responsive B–O dynamic coordination bond is proposed, and used to regulate the photophysical processes of boron-nitrogen-doped polyaromatic hydrocarbons. The formation of the B–O coordination bond under a suitable voltage is confirmed by experiments and theoretical calculations, and B–O coordination bond can be broken back to the initial state under opposite voltage. The whole process is accompanied by reversible changes in photophysical properties. Further, electrofluorochromic devices are successfully prepared based on the above electrochemically responsive coordination bond. The success and harvest of this exploration are beneficial to understand the luminescence mechanism of boron-nitrogen-doped polyaromatic hydrocarbons, and provide ideas for design of dynamic covalent bonds and broaden material types and applications.

Abstract Background Insulin resistance (IR) is indicated to be linked with adverse outcomes of acute myocardial infarction (AMI), for its pro-inflammatory and pro-thromboplastic function. The triglyceride-glucose (TyG) index is a newly developed substitute marker for IR. The aim of this pooled analysis was to provide a summary of the relationship of TyG index with occurrences of major adverse cardiovascular and cerebrovascular events (MACCEs) among populations suffering from AMI. Methods Cohorts reporting multivariate-adjusted hazard ratios of TyG index with MACCEs or its independent events were identified through systematically searching PubMed, MEDLINE, Web of science, Embase and Cochrane databases. Results were combined using a random-effects model. Results 21 cohorts comprising 20403 individuals were included. Compared to individuals in the lowest TyG category, patients in the highest TyG category exhibited elevated risks of both MACCEs (P < 0.00001) and all-cause death (P < 0.00001). These findings were in line with the results as TyG analyzed as continuous variables (MACCEs: P = 0.006; all-cause death: P < 0.00001). Subgroup analysis demonstrated that diabetic status, type of AMI, nor the reperfusion therapy did not destruct this correlation (for subgroups, all P < 0.05). Conclusion All these indicated that higher TyG index could potentially predict MACCEs and all-cause death in patients with AMI as an independent indicator.

The Lower Cambrian Longwangmiao Formation in the Penglai gas area has promising carbonate reservoirs and exhibit strong gas indications, making itself a pivotal exploration zone following the Anyue gas field in the central Sichuan region. However, the lack of clarity regarding reservoir development patterns has constrained exploration deployment. Through comprehensive analysis of the most recent drilling and testing data, this study systematically examines the reservoir characteristics and primary controlling factors of the Longwangmiao Formation in Penglai gas area. Results reveal a mixed sedimentary background of terrigenous debris and carbonate, with reservoirs predominantly situated in the middle and upper strata of the Longwangmiao Formation. The reservoir rock types are predominantly (residual) granular dolomite and crystalline dolomite. Main types of reservoir space include intergranular dissolved pores, intragranular dissolved pores, intercrystalline dissolved pores, and micro-fractures. The reservoir is characterized by low porosity and permeability, with thickness typically ranging between 10-42 m. Reservoir development is controlled by sequence stratigraphy, lithology and diagenesis, particularly evident in the granular dolomite and crystalline dolomite within the middle and upper sections of the progradational parasequence set. Key constructive diagenetic processes include atmospheric freshwater dissolution, oil and gas dissolution and fracture. A comparison with the Moxi-Gaoshiti area in the main area of Anyue gas field suggests similar controlling factors, primarily revolving around granular dolomite, dissolution and fracturing. Nonetheless, the Longwangmiao Formation reservoir in Penglai gas area exhibit distinctive traits such as high ash content, elevated terrigenous clastic content and limited penecontemporaneous exposure. Consequently, identifying high-energy granular dolomite and fine-grained dolomite exposed during the penecontemporaneous period, as well as dolomite modified by supergene karst, emerges as imperative in targeting reservoirs within the Penglai gas area.

Abstract Fullerene derivatives are highly attractive materials in solar cells, organic thermoelectrics, and other devices. However, the intrinsic low electron mobility and electrical conductivity restrict their potential device performance, such as perovskite solar cells (PSCs). Herein, we successfully enhanced the electric properties and morphology of phenyl‐C61‐butyric acid methyl ester (PCBM) by n‐doping it with a benzimidazoline derivative, 9‐(1,3‐dimethyl‐2,3‐dihydro‐1H‐benzoimidazol‐2‐yl)‐julolidine (JLBI‐H) via a solution process. We found the n‐doping can not only improve the conductivity and optimize the band alignment but also enable the PCBM to have a constantly strong charge extraction ability in a wide temperature from 173 to 373 K, which guarantees a stable photovoltaic performance of the corresponding PSCs under a wide range of operating temperatures. With the JLBI‐H‐doped PCBM, we improved the efficiency from 17.9% to 19.8%, along with enhanced stability of the nonencapsulated devices following the aging protocol of ISOS‐D‐1.

Background: Associations between napping and incident atrial fibrillation (AF) remain unknown, and few studies have accounted for dynamic transitions between AF and dementia. Objectives: The purpose of this study was to evaluate associations between napping with incident AF and the dynamic transitions of AF and dementia, as well as the mediation pathway of left ventricular (LV) size and function. Methods: A total of 476,588 participants from UK Biobank were included. Napping frequency and other sleep behaviors were evaluated. Incident AF, dementia, and mortality were ascertained via linkage to external registry databases. LV size and function indices were obtained from cardiovascular magnetic resonance imaging phenotypes. A multistate survival analysis was conducted to examine daytime napping in relation to dynamic transitions. Weighed AF genetic risk score was calculated. Results: Frequent daytime napping, compared to never/rarely napping, was associated with a 1.17-fold AF risk (HR: 1.17; 95% CI: 1.12-1.22), which persisted after controlling for other sleep behaviors. The successful application of new technologies such as remotely piloted aircraft systems, distributed electric propulsion systems, and automatic control systems on electric vertical take-off and landing(eVTOL) aircraft has prompted Urban Air Mobility (UAM) to be mentioned frequently. UAM is a newly raised transport mode of using eVTOL aircraft to transport people and cargo in urban areas, which is thought to share some of the traffic on the ground. One of the prerequisites for UAM to operate on a regular basis is that its demand can support the operating costs, so forecasting UAM demand is necessary. We conduct UAM demand forecasting based on the four-step method, focusing on improving the third-step modal split, and propose a demand forecasting model based on the logit model. The model combines a nested logit (NL) model with a multinomial logit (MNL) model to solve the problem of non-existent UAM sharing rates. We use Chengdu, China as an example, and focus on forecasting the UAM traffic demand in 2030 with the help of the four-step method. The results show that UAM is suitable for shared operation during the early stages. With a fully shared operation, the UAM share rate increases by 0.73% for every kilometer increase in distance. Moreover, UAM is more competitive than other modes for delivery distances exceeding 15 ​km. Finally, using the distributions of the share rate and traffic flow pattern from the simulation, we propose the routes that can be prioritized for UAM operations in Chengdu.

Automatic creation of real-time, emotion-based piano music pieces remains a challenge for deep learning models. While Generative Adversarial Networks (GANs) have shown promise, existing methods can struggle with generating musically coherent pieces and often require complex manual configuration. This paper proposes a novel model called Learning Automata-based Self-Attention Generative Adversarial Network (LA-SAGAN) to address these limitations. The proposed model uses a Generative Adversarial Network (GAN), combined with Self-Attention (SA) mechanism to reach this goal. The benefits of using SA modules in GAN architecture is twofold: First, SA mechanism results in generating music pieces with homogenous structure, which means long-distance dependencies in generated outputs are considered. Second, the SA mechanism utilizes the emotional features of the input to produce output pieces. This results in generating music pieces with desired genre or theme. In order to control the complexity of the proposed model, and optimize its structure, a set of Learning Automata (LA) models have been used to determine the activity state of each SA module. To do this, an iterative algorithm based on cooperation of LAs is introduced which optimizes the model by deactivating unnecessary SA modules. The efficiency of the proposed model in generating piano music has been evaluated. Evaluations demonstrate LA-SAGAN’s effectiveness: at least 14.47% improvement in entropy (diversity) and improvements in precision (at least 2.47%) and recall (at least 2.13%). Moreover, human evaluation confirms superior musical coherence and adherence to emotional cues.

Abstract Advanced photodetectors with intelligent functions are expected to take an important role in future technology. However, completing complex detection tasks within a limited number of pixels is still challenging. Here, we report a differential perovskite hemispherical photodetector serving as a smart locator for intelligent imaging and location tracking. The high external quantum efficiency (~1000%) and low noise (10−13 A Hz−0.5) of perovskite hemispherical photodetector enable stable and large variations in signal response. Analysing the differential light response of only 8 pixels with the computer algorithm can realize the capability of colorful imaging and a computational spectral resolution of 4.7 nm in a low-cost and lensless device geometry. Through machine learning to mimic the differential current signal under different applied biases, one more dimensional detection information can be recorded, for dynamically tracking the running trajectory of an object in a three-dimensional space or two-dimensional plane with a color classification function.

Abstract Background and aim Two oral antivirals (Nirmatrelvir- ritonavir and Azvudine) are widely used in China practice during the Omicron wave of the pandemic. However, little evidence regarding the real-world effectiveness of these two oral antivirals in in-hospital patients. We aimed to evaluate the clinical effectiveness of nirmatrelvir-ritonavir versus azvudine among adult hospitalized patients with COVID-19. Methods This retrospective cohort study used data from three Chinese PLA General Hospital medical centres. Hospitalized patients with COVID-19 treated with azvudine or nirmatrelvir-ritonavir from Dec 10, 2022, to February 20, 2023, and did not require invasive ventilation support on admission were eligible for inclusion. Results After exclusions and propensity-score matching, the final analysis included 486 azvudine recipients and 486 nirmatrelvir-ritonavir recipients. By 28 days of initiation of the antivirus treatment, the crude incidence rate of all-cause death was similar in both types of antivirus treatment (nirmatrelvir-ritonavir group 2.8 events 1000 person-days [95% CI, 2.1–3.6] vs azvudine group 3.4 events/1000 person-days [95% CI, 2.6–4.3], P = 0.38). Landmark analysis showed that all-cause death was lower in the nirmatrelvir-ritonavir (3.5%) group than the azvudine (6.8%, P = 0.029) within the initial 10-day admission period, while no significant difference was observed for results between 10 and 28 days follow-up. There was no significant difference between the nirmatrelvir-ritonavir group and the azvudine group in cumulative incidence of the composite disease progression event (8.6% with nirmatrelvir-ritonavir vs. 10.1% with azvudine, HR, 1.22; 95% CI 0.80–1.86, P = 0.43). Conclusion Among patients hospitalized with COVID-19 during the omicron wave in Beijing, similar in-hospital clinical outcomes on 28 days were observed between patients receiving nirmatrelvir-ritonavir and azvudine. However, it is worth noticing that nirmatrelvir-ritonavir appears to hold an advantage over azvudine in reducing early mortality. Further randomized controlled trials are needed to verify the efficacy of those two antivirus medications especially in early treatment.

Reef limestone is a biogenic sedimentary rock widely distributed in coral reef areas, acting as an important foundation for coast construction. Due to its special biogenic origin, reef limestone is different from conventional rocks both in terms of rock structure and mechanical properties. In this study, mesoscale uniaxial compression experiments with five different loading directions were conducted on two kinds of reef limestones from the Maldives Islands and the South China Sea, respectively. The real-time high-resolution videos and images of failure processes were recorded simultaneously to investigate the fracture evolution and fracture surface roughness of reef limestones. It demonstrated that the reef limestones belonged to extremely soft to soft rocks, and their uniaxial compressive strength (UCS) values fluctuated with high discreteness. The mesoscale mechanical properties of reef limestones were highly anisotropic and mainly controlled by pore structure. The occurrence of dissolution pores in reef limestone tended to intensify mechanical anisotropy. With the integration of the fracture initiation and propagation features of reef limestones, it is supposed that the intrinsic mechanism of anisotropy was probably attributed to the differences in coral growth direction and dissolution. Furthermore, the quantified fracture surface roughness was revealed to have a good consistency with density and UCS for the reef limestones from the South China Sea. The findings are helpful for providing theoretical and experimental references for engineering construction in coral reef areas.

AbstractRheumatoid arthritis (RA) is a systemic autoimmune disease (AD) caused by the aberrant attack of the immune system on its own joint tissues. Genetic and environmental factors are the main reasons of immune system impairment and high incidence of RA. Although there are medications on the market that lessen disease activity, there is no known cure for RA, and patients are at risk in varying degrees of systemic immunosuppression. By transporting (encapsulating or surface binding) RA-related self-antigens, nucleic acids, immunomodulators, or cytokines, tolerogenic nanoparticles—also known as immunomodulatory nano-preparations—have the potential to gently regulate local immune responses and ultimately induce antigen-specific immune tolerance. We review the recent advances in immunomodulatory nano-preparations for delivering self-antigen or self-antigen plus immunomodulator, simulating apoptotic cell avatars in vivo, acting as artificial antigen-presenting cells, and based on scaffolds and gels, to provide a reference for developing new immunotherapies for RA.

Human varicose veins are commonly claimed to be responsible for lower limb symptoms. Mutation in KRAS gene has been implicated in various diseases, including cancers and vascular diseases. While little known about the novel mutation in KRAS gene and its contribution to the development of varicose veins. Here, we have generated human induced pluripotent stem cell (iPSC) line, which harboured a novel mutation in KRAS (c.209A>T) gene. This cell line provided a novel tool for understanding the mechanism of KRAS mutation in the pathogenesis of varicose veins.

Abstract Shear strength of damage zone, representing the stress threshold for rupture initiation, is a critical parameter in faulting mechanics. Despite its significance, the damage‐zone's shear strength has not been estimated in natural earthquake ruptures. Here we employed coseismic deformation and strain, kinematic slip model, and finite element modeling to determine the elastic properties and peak shear stress of coseismic damage zones along the 2021 Mw 7.4 Maduo earthquake. Through the analysis of the lowest shear stress resulting in surface ruptures and the highest stress without surface rupture, we constrained the strength within a range of 7–17 MPa. Our result is consistent with strength (5–16 MPa) of sandstone samples from laboratory tests, demonstrating the validity of this estimation. Although factors such as fault maturity and confining pressure influence strength variation, the strength can directly reflect the stress threshold required for macroscopic surface rupture formation in fault damage zones dominated by sandstone.

Composite salt formation is a high-quality caprock for oil and gas resources. The accident encountered in composite salt formation drilling is a big problem to be solved in the drilling process. According to statistics, nearly 50% of drilling accidents occur in composite salt formations. The composite salt formation is mainly composed of salt, gypsum, and interbed mud, and the lithology is complex and changeable. Therefore, it is very important to study the deformation mechanism and leading influencing factors of composite salt formation in view of the problem of frequent accidents in the drilling process. In this article, the creep parameters based on the constitutive equation of creep of salt rock are obtained by combining theoretical with experimental research. A three-dimensional directional wellbore mechanical model is established to analyze the influence of inclination on borehole shrinkage.

Abstract The February 2023 Turkey‐Syria Earthquake doublet ruptured multiple segments of the East Anatolian Fault (EAF) Zone. Dominating seismicity focal mechanism shifted dramatically from strike‐slip to normal‐faulting after the doublet. To better understand this shift, here we derived a comprehensive 3D co‐seismic displacement field and performed the stress analysis. Abundant space geodetic data were used to generate high‐resolution 3D surface displacement, which provide tight constraints on fault geometry, slip distribution and stress field. Together with stress inversion from aftershock focal mechanisms, we show that the principal stress direction rotation in the region with the most normal‐faulting aftershocks is the staggering 29°. The induced heterogenous stress may explain the shift of the dominant focal mechanism toward normal faulting. We suggest that the extensional horsetail splay faults, likely formed through geologic time scale related to the releasing bend on the EAF, are the hosts of most of the normal faulting aftershocks.

Abstract Background Limited data are available on renal complications in patients with acute fulminant myocarditis (AFM) receiving venoarterial extracorporeal membrane oxygenation (VA-ECMO) support in China. To evaluate the impact of renal complications on outcomes in adult patients with AFM supported with VA-ECMO. Methods Data were extracted from Chinese Society of ExtraCorporeal Life Support (CSECLS) Registry database. Adult patients who were diagnosed with AFM receiving VA-ECMO support in the database were included. The primary outcome was 30-day mortality in patients with AFM supported with VA-ECMO. Logistic regression model was used to examine the impact of renal complications on 30-day mortality by adjusting confounders. Results A total of 202 patients were included. The median age was 38 years (IQR 29–48) and males (n = 103) represented 51.0% of the total accounted patients. The median ECMO duration was 142.9 h (IQR 112.1–188.8 h). 178 (88.1%) patients weaned from ECMO and 156 (71.9%) patients survived. 94(46.5%) patients developed renal complications while on ECMO course. Abstract Bladder cancer (BCa) is one of the most common malignancies worldwide. However, the lack of accurate and effective targeted drugs has become a major problem in current clinical treatment of BCa. Studies have demonstrated that squalene epoxidase (SQLE), as a key rate-limiting enzyme in cholesterol biosynthesis, is involved in cancer development. In this study, our analysis of The Cancer Genome Atlas, The Genotype-Tissue Expression, and Gene Expression Omnibus databases showed that SQLE expression was significantly higher in cancer tissues than it was in adjacent normal tissues, and BCa tissues with a high SQLE expression displayed a poor prognosis. We then confirmed this result in qRT-PCR and immunohistochemical staining experiments, and our vitro studies demonstrated that SQLE knockdown inhibited tumor cell proliferation and metastasis through the PTEN/AKT/GSK3β signaling pathway. By means of rescue experiments, we proved that that P53 is a key molecule in SQLE-mediated regulation of the PTEN/AKT/GSK3β signaling pathway. Simultaneously, we verified the above findings through a tumorigenesis experiment in nude mice. In conclusion, our study shows that SQLE promotes BCa growth through the P53/PTEN/AKT/GSK3β axis, which may serve as a therapeutic biological target for BCa.

Objective Mechanical stress is an important risk factor for intervertebral disc degeneration (IVDD). Angiopoietin‐2 (ANG‐2) is regulated by mechanical stress and is widely involved in the regulation of extracellular matrix metabolism. In addition, the signaling cascade between HIF‐1α and NF‐κB is critical in matrix degradation. This study aims to investigate the role and molecular mechanism of ANG‐2 in regulating the degeneration of annulus fibrosus (AF) through the HIF‐1α/NF‐κB signaling pathway. Methods The bipedal standing mice IVDD model was constructed, and histological experiments were used to evaluate the degree of IVDD and the expression of ANG‐2 in the AF. Mouse primary AF cells were extracted in vitro and subjected to mechanical stretching experiments. Western blot assay was used to detect the effect of mechanical stress on ANG‐2, and the role of the ANG‐2‐mediated HIF‐1α/NF‐κB pathway in matrix degradation. In addition, the effect of inhibiting ANG‐2 expression by siRNA or monoclonal antibody on delaying IVDD was investigated at in vitro and in vivo levels. One‐way ANOVA with the least significant difference method was used for pairwise comparison of the groups with homogeneous variance, and Dunnett's method was used to compare the groups with heterogeneous variance. Results In IVDD, the expressions of catabolic biomarkers (mmp‐13, ADAMTS‐4) and ANG‐2 were significantly increased in AF. In addition, p65 expression was increased while HIF‐1α expression was significantly decreased. The results of western blot assay showed mechanical stress significantly up‐regulated the expression of ANG‐2 in AF cells, and promoted matrix degradation by regulating the activity of HIF‐1α/NF‐κB pathway. Exogenous addition of Bay117082 and CoCl2 inhibited matrix degradation caused by mechanical stress. Moreover, injection of neutralizing antibody or treatment with siRNA to inhibit the expression of ANG‐2 improved the matrix metabolism of AF and inhibited IVDD progression by regulating the HIF‐1α/NF‐κB signaling pathway. Conclusion In IVDD, mechanical stress could regulate the HIF‐1α/NF‐κB signaling pathway and matrix degradation by mediating ANG‐2 expression in AF degeneration.

Insulators play a pivotal role in the reliability of power distribution networks, necessitating precise defect detection. However, compared with aerial insulator images of transmission network, insulator images of power distribution network contain more complex backgrounds and subtle insulator defects, it leads to high false detection rates and omission rates in current mainstream detection algorithms. In response, this study presents ID-YOLOv7, a tailored convolutional neural network. First, we design a novel Edge Detailed Shape Data Augmentation (EDSDA) method to enhance the model's sensitivity to insulator's edge shapes. Meanwhile, a Cross-Channel and Spatial Multi-Scale Attention (CCSMA) module is proposed, which can interactively model across different channels and spatial domains, to augment the network's attention to high-level insulator defect features. Second, we design a Re-BiC module to fuse multi-scale contextual features and reconstruct the Neck component, alleviating the issue of critical feature loss during inter-feature layer interaction in traditional FPN structures. Finally, we utilize the MPDIoU function to calculate the model's localization loss, effectively reducing redundant computational costs. We perform comprehensive experiments using the Su22kV_broken and PASCAL VOC 2007 datasets to validate our algorithm's effectiveness. On the Su22kV_broken dataset, our approach attains an 85.7% mAP on a single NVIDIA RTX 2080ti graphics card, marking a 7.2% increase over the original YOLOv7. On the PASCAL VOC 2007 dataset, we achieve an impressive 90.3% mAP at a processing speed of 53 FPS, showing a 2.9% improvement compared to the original YOLOv7.

Background: Clear cell renal cell carcinoma (ccRCC) is one of the most common malignant tumors that can be highly aggressive. Despite advances in the exploration of its underlying molecular biology, the clinical outcome for advanced ccRCC is still unsatisfied. Recently, more attention was paid to the functions of Kinesin family member 2C (KIF2C) in cancer progression, while the specific function of KIF2C in ccRCC has not been sufficiently elucidated. The present study aims to investigate the role of KIF2C in the progression of ccRCC and reveal potential mechanisms. Methods: Expression of KIF2C in ccRCC tissues and adjacent normal tissue was compared and the association of KIF2C expression level with tumor grade, stage, and metastasis were analyzed using online web tool. Kaplan-Meier survival was performed to detect the association of KIF2C expression and patient’ prognosis. Stably cell lines with KIF2C knockdown or overexpression were constructed by lentivirus infection. CCK-8, colony formation, scratch healing, and transwell invasion assays were carried out to explore the effect of KIF2C knockdown or overexpression on the proliferation, migration, and invasion of ccRCC cells. Gene set enrichment analysis (GSEA) was conducted to reveal signaling pathways associated with KIF2C expression. The effect of KIF2C on JAK2/STAT3 signaling pathway were explored by western blot assay. Results: KIF2C expression was significantly upregulated in ccRCC tissues and was higher with the increase of tumor grade, stage, and metastasis. Higher expression of KIF2C was correlated with worse overall survival and diseases free survival in ccRCC patients. Silence of KIF2C inhibited proliferation, migration, and invasion in ccRCC cells. Conversely, overexpression of KIF2C had the opposite effect. GSEA results showed that JAK/STAT signaling pathway was markedly enriched in KIF2Chigh group. Pearson’ correlation revealed that KIF2C expression was significantly associated with genes in JAK2/STAT3 signaling. Western blot results showed that KIF2C knockdown decreased protein expression of p-JAK2 and p-STAT3, and KIF2C overexpression increased the phosphorylation of JAK2 and STAT3. AG490, a JAK2/STAT3 signaling inhibitor, could partly impair the tumor-promoting effects of KIF2C in ccRCC. Conclusion: KIF2C expression was significantly upregulated in ccRCC and correlated with tumor grade, stage, metastasis, and patients’ prognosis. KIF2C promoted ccRCC progression via activating JAK2/STAT3 signaling pathway, and KIF2C might be a novel target in ccRCC therapy.

Background The association of age at coronary heart disease (CHD) onset with incident dementia remains unexplored. This study aimed to examine whether younger onset age of CHD is associated with a higher risk of incident dementia. Methods and Results Data were obtained from the UK Biobank. Information on the diagnosis of CHD and dementia was collected at baseline and follow‐ups. Propensity score matching method and Cox proportional hazards models were used to evaluate the association between different ages at CHD onset and incident dementia. A total of 432 667 adults (mean±SD age, 56.9±8.1 years) were included, of whom 11.7% had CHD. Compared with participants without CHD, participants with CHD exhibited higher risks of developing all‐cause dementia, Alzheimer's disease, and vascular dementia. Abstract Background Percutaneous cannulation is now accepted as the first-line strategy for extracorporeal cardiopulmonary resuscitation (ECPR) in adults. However, previous studies comparing percutaneous cannulation to surgical cannulation have been limited by small sample size and single-center settings. This study aimed to compare in-hospital outcomes in cardiac arrest (CA) patients who received femoro-femoral ECPR with percutaneous vs surgical cannulation. Methods Adults with refractory CA treated with percutaneous (percutaneous group) or surgical (surgical group) femoro-femoral ECPR between January 2008 and December 2019 were extracted from the international Extracorporeal Life Support Organization registry. The primary outcome was severe neurological complication. Multivariable logistic regression analyses were performed to assess the association between percutaneous cannulation and in-hospital outcomes. Synthetic microbial community has widely concerned in the fields of agriculture, food and environment over the past few years. However, there is little consensus on the method to synthetic microbial community from construction to functional verification. Here, we review the concept, characteristics, history and applications of synthetic microbial community, summarizing several methods for synthetic microbial community construction, such as isolation culture, core microbiome mining, automated design, and gene editing. In addition, we also systematically summarized the design concepts, technological thresholds, and applicable scenarios of various construction methods, and highlighted their advantages and limitations. Ultimately, this review provides four efficient, detailed, easy-to-understand and -follow steps for synthetic microbial community construction, with major implications for agricultural practices, food production, and environmental governance.

Welded tuffs have a wide range of welding degrees and show significant variability in mechanical behavior. However, the detailed influence of welding degree on the meso-mechanical behavior of welded tuffs remains unclear. Based on petrographic and pore-structure analysis, we conducted a series of meso-mechanical experiments on weakly to strongly welded tuffs by utilizing a mesoscale real-time loading-observation-acquisition system. The results indicated that the strongly and weakly welded tuffs showed a small range in mineralogical composition and porosity, while the meso-mechanical behavior exhibited significant variability. Strongly welded tuffs showed lower uniaxial compression strength, weaker mechanical anisotropy, and smaller fracture surface roughness. In contrast, weakly welded tuffs exhibited higher uniaxial compression strength, stronger mechanical anisotropy, and rougher fracture surface roughness. Welded tuffs with strong packing and welding of glass shards tended to have fractures propagating along the maximum principal direction, while those with weak packing and welding of glass shards may have had failure along the alignment of glass shards. The influence of welding degree on the meso-mechanical behavior of welded tuffs probably originates from their diagenesis environments, mainly depending on the combined effect of the pyroclastic properties and pseudo-rhyolitic structure. The findings reveal the meso-mechanical differences of welded tuffs and shed light on improving tuffs for stable and durable construction.

In this paper, we present a novel localization scheme, location-aware ranging correction (LARC), to correct ranging estimates from ultra wideband (UWB) signals. Existing solutions to calculate ranging corrections rely solely on channel information features (e.g., signal energy, maximum amplitude, estimated range). We propose to incorporate a preliminary location estimate into a localization chain, such that location-based features can be calculated as inputs to a range-error prediction model. This way, we can add information to range-only measurements without relying on additional hardware such as an inertial measurement unit (IMU). This improves performance and reduces overfitting behavior. We demonstrate our LARC method using an open-access measurement dataset with distances up to 20 m, using a simple regression model that can run purely on the CPU in real-time. The inclusion of the proposed features for range-error mitigation decreases the ranging error 90th percentile (P90) by 58% to 15 cm (compared to the uncorrected range error), for an unseen trajectory. The 2D localization P90 error is improved by 21% to 18 cm. We show the robustness of our approach by comparing results to a changed environment, where metallic objects have been moved around the room. In this modified environment, we obtain a 56% better P90 ranging performance of 16 cm. The 2D localization P90 error improves as much as for the unchanged environment, by 17% to 18 cm, showing the robustness of our method. This method evolved from the first-ranking solution of the 2021 and 2022 International Conference on Indoor Position and Indoor Navigation (IPIN) Competition.

Surface-coupled transition metal oxyhydroxide (TMOOH) on semiconductor (SC)-based photoanodes are effective strategies for improving photoelectrochemical (PEC) performance. However, there is a substantial difference between the current density and theoretical value due to the inevitable interfacial charge recombination of SC/TMOOH. Here, we employ BiVO4/FeNiOOH as a model, constructing the BiVO4/MnOx/CoOx/FeNiOOH integrated system by introducing a novel hetero-interface regulation unit, i.e., MnOx/CoOx. As expected, the optimized integrated system demonstrates a photocurrent density as high as 5.0 mA/cm2 at 1.23 V versus the reversible hydrogen electrode (RHE) under 1 sun AM 1.5G illumination, accompanied by 12-h stability. The detailed electrochemical analysis and intensity modulated photocurrent spectroscopy (IMPS) have confirmed that the high PEC performance mainly originates from the hetero-interface structure, which not only suppresses the interfacial charge recombination by accelerating the photogenerated hole transfer kinetics from BiVO4 to FeNiOOH but promotes the kinetics of surface oxygen evolution reaction (OER). Notably, these findings can also be extended to other structures (CeOx/CoOx), reflecting its universality. This finding has provided a new insight into the highly efficient solar energy conversion in the SC/TMOOH system.

In this research, an ammonium perchlorate/polydopamine (AP/PDA) core–shell composite was prepared in a non-aqueous solution to reduce the mechanical sensitivity of ammonium perchlorate (AP). The result showed that the AP/PDA core–shell composite could be successfully constructed in ethyl acetate solution with an AP recovery rate that reached 86%. The mechanical sensitivity of the obtained AP/PDA core–shell composite was significantly reduced with a PDA content of only 0.76%. The DSC and TG also indicated that the coating of PDA showed catalytic activity in the thermal decomposition of AP with a lower decomposition temperature and a decreased Ea value of AP. Thus, this study proposed a simple strategy for achieving a good balanced between harnessing the energy and ensuring the safety of ammonium perchlorate by significantly reducing its mechanical sensitivity by using a very low polydopamine coating layer content, and this shows great potential for the design and fabrication of insensitive energetic composites for use in propellants.

This research used fly ash and slag to create geopolymer foam concrete. They were activated with an alkali, resulting in a chemical reaction that produced a gel that strengthened the concrete’s structural integrity. The experimental approach involved varying the fly ash content in the precursors at incremental percentages (10%, 30%, 50%, 70% and 90%) and subjecting the fly ash to mechanical activation through a planetary ball mill at distinct rotational speeds (380, 400, 420 and 440 rpm). The investigation discerned that the fly ash content and particle structure exert a discernible influence on macroscopic properties, including flowability, air generation height, compressive strength, dry density and microstructural characteristics such as pore distribution and hydration product arrangement in the geopolymer foam concrete. Employing analytical techniques such as X-ray diffraction (XRD) and scanning electron microscopy (SEM), it was deduced that diminishing the fly ash content correlates with an enhancement in compressive strength. Furthermore, the specific strength of the geopolymer foam concrete reached a peak of 0.041 when the activated fly ash in the planetary ball mill rotated at 420 rpm, manifesting a lightweight and high-strength outcome.

Drones equipped with visible and infrared sensors play a vital role in urban road supervision. However, conventional methods using RGB-IR image pairs often struggle to extract effective features. These methods treat these spectra independently, missing the potential benefits of their interaction and complementary information. To address these challenges, we designed the Multispectral Feature Mutual Guidance Network (MFMG-Net). To prevent learning bias between spectra, we have developed a Data Augmentation (DA) technique based on the mask strategy. The MFMG module is embedded between two backbone networks, promoting the exchange of feature information between spectra to enhance extraction. We also designed a Dual-Branch Feature Fusion (DBFF) module based on attention mechanisms, enabling deep feature fusion by emphasizing correlations between the two spectra in both the feature channel and space dimensions. Finally, the fused features feed into the neck network and detection head, yielding ultimate inference results. Our experiments, conducted on the Aerial Imagery (VEDAI) dataset and two other public datasets (M3FD and LLVIP), showcase the superior performance of our method and the effectiveness of MFMG in enhancing multispectral feature extraction for drone ground detection.

Abstract Background Hepatocellular carcinoma (HCC) is aggressive liver cancer. Despite advanced imaging and other diagnostic measures, HCC in a significant portion of patients had reached the advanced stage at the first diagnosis. Unfortunately, there is no cure for advanced HCC. As a result, HCC is still a leading cause of cancer death, and there is a pressing need for new diagnostic markers and therapeutic targets. Methods We investigated sulfotransferase 1C2 (SUTL1C2), which we recently showed was overexpressed in human HCC cancerous tissues. Specifically, we analyzed the effects of SULT1C2 knockdown on the growth, survival, migration, and invasiveness of two HCC cell lines, i.e., HepG2 and Huh7 cells. We also studied the transcriptomes and metabolomes in the two HCC cell lines before and after SULT1C2 knockdown. Based on the transcriptome and metabolome data, we further investigated the SULT1C2 knockdown‐mediated shared changes, i.e., glycolysis and fatty acid metabolism, in the two HCC cell lines. Finally, we performed rescue experiments to determine whether the inhibitory effects of SULT1C2 knockdown could be rescued via overexpression. Results We showed that SULT1C2 overexpression promoted the growth, survival, migration, and invasiveness of HCC cells. In addition, SULT1C2 knockdown resulted in a wide range of gene expression and metabolome changes in HCC cells. Moreover, analysis of shared alterations showed that SULT1C2 knockdown significantly suppressed glycolysis and fatty acid metabolism, which could be rescued via SULT1C2 overexpression. Conclusions Our data suggest that SULT1C2 is a potential diagnostic marker and therapeutic target for human HCC.

Abstract Lung cancer is the most lethal malignancies with high aggressive and poor prognosis. Until now, the five-year survival rate has not been improved which brings serious challenge to human health. Lung cancer stem cells (LCSCs) serve as the root of cancer occurrence, progression, recurrence, and drug resistance. Therefore, effective anti-cancer agents and molecular mechanisms which could specifically eliminate LCSCs are urgently needed for drug design. In this article, we discovered Olig2 was overexpressed in clinical lung cancer tissues and acted as a transcription factor to regulate cancer stemness by regulating CD133 gene transcription. The results suggested Olig2 could be a promising target in anti-LCSCs therapy and new drugs targeted Olig2 may exhibit excellent clinical results. Furthermore, we verified ACT001, a guaianolide sesquiterpene lactone in phase II clinical trial with excellent glioma remission, inhibited cancer stemness by directly binding to Olig2 protein, inducing Olig2 ubiquitination degradation and inhibiting CD133 gene transcription. All these results suggested that Olig2 could be an excellent druggable target in anti-LCSCs therapy and lay a foundation for the further application of ACT001 in the treatment of lung cancer in clinical.

Neutrophils play a critical role in the immune response to infection and tissue injury. However, recent studies have shown that neutrophils are a heterogeneous population with distinct subtypes that differ in their functional properties. Moreover, aging can alter neutrophil function and exacerbate immune dysregulation. In this review, we discuss the concept of neutrophil heterogeneity and how it may be affected by aging. We then examine the implications of neutrophil heterogeneity and aging for COVID-19 pathogenesis and wound healing. Specifically, we summarize the evidence for neutrophil involvement in COVID-19 and the potential mechanisms underlying neutrophil recruitment and activation in this disease. We also review the literature on the role of neutrophils in the wound healing process and how aging and neutrophil heterogeneity may impact wound healing outcomes. Finally, we discuss the potential for neutrophil-targeted therapies to improve clinical outcomes in COVID-19 and wound healing.

Introduction: Acorus tatarinowii Schott is a traditional Chinese medicine that aromatizes dampness, opens the body and pacifies the mind, and is used medicinally as the dried rhizome of Acorus tatarinowii, family Tenaxaceae. Research has confirmed that it has a strong anti-inflammatory activity, however, its mechanism of anti-inflammatory is still unclear, and we continue to investigate its chemical composition and anti-inflammatory mechanism. Methods: The separation and purification of the EtOH extract was carried out by preparative thin layer chromatography and column chromatography. The structures of the isolated compounds were identified by NMR spectroscopy and comparing their spectral data with those previously reported in literature. CCK-8 method was used to evaluate the anti-inflammatory activity of all compounds. The MDA, NO, SOD, IL-10, IL-1β, IL-6 and TNF-α levels were analyzed using ELISA kits. TNF-α and Beclin-1 expression were examined by western blot and immunofluorescence. Results: Ten compounds, named cinnamic acid(1), sinapic acid(2), isoferulic acid(3), α-asarone(4), ferulic acid(5), arbutin (6), caffeic acid (7), 3-(3-hydroxyl phenyl)propanol(8), ethyl caffeate(9) and β-asarone(10) were isolated from the rhizome of Acorus tatarinowii Schott. Their structures were elucidated on the basis of extensive spectroscopic analyses, as well as by comparison with literature data. The anti-inflammatory effects of the all compounds were evaluated in lipopolysaccharide(LPS)-induced PC12 cell models in our study. The results of activity test showed that compounds 3 and 6 had good inhibitory effects on 5 μg·mL−1 LPS-induced PC12 cells. The IC50 of compounds 3 and 6 against PC12 cells were 27.41 ± 0.09 µmol·L−1 and 30.19 ± 0.10 µmol·L−1, respectively. Moreover, compounds 3 and 6 showed a significant protective effect on inflammation by increasing the IL-10 and SOD levels, and reducing the TNF-α, IL-6, IL-1β, MDA and NO levels. The most important point is that their anti-inflammatory effect is related to the autophagy factor Beclin-1. Conclusion: These findings imply that compounds 3 and 6 exerts anti-inflammatory effects by regulating TNF-α/Beclin-1 pathway, which provide a new basis for the anti-inflammatory activity of Acorus tatarinowii Schott.

Abstract Background Major concerns about the adverse mental health impact of the rapidly spread COVID-19 pandemic have been raised. Previous studies on changes of depressive symptoms during the COVID-19 pandemic have yielded inconsistent results regarding the sex differences. Since women have higher depressive symptoms even without the pandemic, it is essential to consider the pre-existing change of depressive symptoms of a similar period to discern the effect of the pandemic on depression. This study aimed to evaluate sex differences in depressive symptoms before and during the pandemic. Methods Data from the Health and Retirement Study (HRS; waves 13 to 15) and the English Longitudinal Study of Ageing (ELSA; wave 8 to COVID-19 wave 2) were analyzed. Depressive symptoms were assessed by the 8-item Center for Epidemiological Studies Depression (CES-D) scale. According to the time of COVID-19 outbreak in the US and the UK, the intervals from waves 13 to 14 surveys of the HRS and from waves 8 to 9 surveys of the ELSA were employed as pre-pandemic periods to control for the pre-existing depressive symptoms, respectively. Changes of CES-D scores during the pre-pandemic and pandemic periods were assessed by linear mixed models. Results Nine thousand, seven hundred thirty-seven participants (mean age: 66.7 ± 10.7 years) from the HRS and 5,098 participants (mean age: 68.7 ± 10.0 years) from the ELSA were included. CES-D scores among women were significantly higher than those among men at all waves in both cohorts. During the pre-pandemic period, no significant sex difference on changes of CES-D scores was detected in either the HRS or the ELSA. During the pandemic period, CES-D scores were increased in both men and women and the sex differences in CES-D increments of the two cohorts were both significant. Enlarged sex differences were demonstrated in increments of CES-D scores during the pandemic period. Conclusions Our results suggest women suffered from worse depressive symptoms in response to the pandemic, although the changes of depression were similar between men and women before the pandemic. These findings underscore the necessity to support the vulnerable populations, especially women, to manage the distress brought by the pandemic and maintain optimal mental health status.

Abstract Background Frailty is a dynamic process that increases with ageing, while it remains unclear whether cardiovascular disease (CVD) risk algorithm could predict life course dynamic frailty trajectories, for example, the longitudinal patterns of how frailty evolves with time. We intended to examine the predictive utility of the Systemic Coronary Risk Estimation 2 (SCORE2) algorithm for life course accelerated frailty and physical function decline, in comparison with the precedent SCORE algorithm. Methods Longitudinal data regarding accumulation of deficits frailty index (FI) and physical function (grip strength, gait speed, peak expiratory flow and timed chair rises) were drawn from the English Longitudinal Study of Ageing (ELSA) and Health and Retirement Study (HRS), two nationally representative cohorts with community‐dwelling adults aged ≥50 years. SCORE and SCORE2 were calculated at baselines following European Society of Cardiology guidelines. A group‐based trajectory modelling approach was used for identifying potential life course frailty trajectories, based on 14‐ and 12‐year FI data in the ELSA and HRS. Modified Poisson regression and linear mixed model were applied for analysing associations between SCORE2 with accelerated frailty trajectory and physical function decline, respectively. Receiver operating characteristic curve (ROC) analysis was conducted to evaluate predictive utility for accelerated frailty increase trajectory of SCORE and SCORE2, with the area under the curve (AUC) compared using the paired DeLong's test. Results A total of 4834 participants from the ELSA and 7815 participants from the HRS were included (mean age: 64.0 ± 9.2 and 65.4 ± 9.9 years; men: 44.3% and 41.4%, respectively). Three frailty trajectories were consistently identified in both cohorts: (1) stable frailty increase (n = 3026 in ELSA and 4004 in HRS); (2) moderate frailty increase (n = 1325 in ELSA and 2955 in HRS); (3) accelerated frailty increase (n = 483 in ELSA and 856 in HRS). Abstract Polycystic ovary syndrome (PCOS), a common reproductive endocrine disease, has clinically heterogeneous characteristics. Recently, cuproptosis causes several diseases by killing cells. Hence, we aimed to explore cuproptosis-related molecular clusters in PCOS and construct a prediction model. Based on the GSE5090, GSE43264, GSE98421, and GSE124226 datasets, an analysis of cuproptosis regulators and immune features in PCOS was conducted. In 25 cases of PCOS, the molecular clusters of cuproptosis-related genes and the immune cell infiltration associated with PCOS were investigated. Weighted gene co-expression network analysis was used to identify differentially expressed genes within clusters. Next, we compared the performance of the random forest model, support vector machine model, generalized linear model, and eXtreme Gradient Boosting for deciding the optimum machine model. Validation of the predictive effectiveness was accomplished through nomogram, calibration curve, decision curve analysis, and using other two datasets. PCOS and non-PCOS controls differed in the dysregulation of cuproptosis-related genes and the activation of immunoreaction. Two cuproptosis-related molecular clusters associated with PCOS were identified. Significant heterogeneity was noted in immunity between the two clusters based on the analysis of immune infiltration. The immune-related pathways related to cluster-specific differentially expressed genes in Cluster1 were revealed by functional analysis. With a relatively low residual error and root mean square error and a higher area under the curve (1.000), the support vector machine model demonstrated optimal discriminative performance. An ultimate 5-gene-based support vector machine model was noted to perform satisfactorily in the other two validation datasets (area under the curve = 1.000 for both). Moreover, the nomogram, calibration curve, and decision curve analysis showed that PCOS subtypes can be accurately predicted. Our study results helped demonstrate a comprehensive understanding of the complex relationship between cuproptosis and PCOS and establish a promising prediction model for assessing the risk of cuproptosis in patients with PCOS.

Gesture fingertip detection plays a vital role in human-computer interaction applications such as VR and robot control. To improve the accuracy of fingertip detection in complex background, this paper proposes a fingertip detection algorithm based on a novel maximum discrimination HOG feature. Firstly, the Holistically-nested edge detection algorithm is used to detect the edge of hand images and non-hand images, and the HOG features of the contour edge are extracted to obtain the training set of positive and negative samples to reduce the influence of illumination, color, and texture on fingertip detection. Secondly, the maximum discrimination features are filtered from the positive sample set by a custom maximum discrimination feature filter and stored as a feature dictionary. The filtered maximum discrimination features and negative sample set are input into the XGBoost classifier for training and the voting rights classifier is obtained. Filtering maximum discrimination features can reduce the interference of irrelevant features and improve the performance of the classifier. Then, the KNN algorithm is used to find the best match in the dictionary, and the final fingertip position is obtained by the Meanshift algorithm. Finally, the algorithm in this paper is tested. The test results show that the accuracy of fingertip detection in this paper can reach 99 %, and the detected RMSE is kept in the range of 5 pixels, which is higher than that based on YCbCr skin color segmentation, YOLO target detection, and YOLO-YCbCr.

Fermatean fuzzy sets (FFSs) serve as a nascent yet potent approach for coping with fuzziness, with their efficacy recently being demonstrated across a spectrum of practical contexts. Nevertheless, the scholarly literature remains limited in exploring the similarity and distance measures tailored for FFSs. The limited existing measures on FFSs sometimes yield counter-intuitive outcomes, which can obfuscate the accurate quantification of similarity and difference among FFSs. This paper introduces a suite of similarity and distance measures tailored for FFSs, drawing inspiration from the Tanimoto measure. We delve into the characteristics of these novel measures and offer some comparative studies with existing FFSs measures, highlighting their superior efficacy in the processing of fuzzy data from FFSs. Our proposed measures effectively rectify the counter-intuitive situations encountered with many existing measures and demonstrate a significant enhancement in differentiating between diverse FFSs. Moreover, we showcase the real-world applicability of our proposed measures through case studies in pattern recognition, medical diagnostics, and multi-attribute decision-making.

Among renewable energy sources, wind energy is regarded as one of the fastest-growing segments, which plays a key role in enhancing environmental quality. Wind turbines are generally located in remote and harsh environments. Bearings are a crucial component in wind turbines, and their failure is one of the most frequent reasons for system breakdown. Wind turbine bearing faults are usually very localized during their early stages which is precisely when they need to be detected. Hence, the early diagnosis of bearing faults holds paramount practical significance. In order to solve the problem of weak pulses being masked by noise in early failure signals of rolling element bearings, a novel fault diagnosis method is proposed based on the combination of complete ensemble empirical mode decomposition with adaptive noise (CEEMDAN) and an improved TFR demodulation method. Initially, the decomposition of vibration signals using CEEMDAN is carried out to obtain several intrinsic mode functions (IMFs). Subsequently, a novel KC indicator that combines kurtosis and the correlation function is designed to select the effective components for signal reconstruction. Finally, an innovative approach based on the continuous wavelet transform (CWT) for multi-scale demodulation analysis in the domain of time–frequency representation (TFR) is also introduced to extract the envelope spectrum. Further fault diagnosis can be achieved by the identification of the fault characteristic frequency (FCF). This study focuses on the theoretical exploration of bearing faults diagnosis algorithms, employing modeling and simulation techniques. The effectiveness and feasibility of the proposed method are validated through the analysis of simulated signals and experimental signals provided by the Center for Intelligent Maintenance Systems (IMS) of the University of Cincinnati and the Case Western Reserve University (CWRU) Bearing Data Center. The method demonstrates the capability to identify various types of bearing faults, including outer race and inner race faults, with a high degree of computational efficiency. Comparative analysis indicates a significant enhancement in fault diagnostic performance when compared to existing methods. This research contributes to the advancement of effective bearing fault diagnosis methodologies for wind turbines, thereby ensuring their reliable operation.

Abstract The 2021 Mw 7.4 Maduo earthquake (eastern Tibetan Plateau), with a surface deformation zone that is characterized by multiple distinct surface rupture segments separated by segments with no surface ruptures, provides an excellent opportunity to constrain the strain threshold (minimum strain) for the formation of surface ruptures. Through sub‐pixel correlation of pre‐ and post‐event SPOT‐6/7 satellite images (pixel = 1.5 m), we derive surface displacement and shear‐strain fields of the Maduo earthquake. By quantifying the minimum shear strain along the surface rupture segments and the maximum shear strain along the segments with no surface ruptures, we estimate a rupture strain threshold, which ranges from 0.8% to 1.8% and appears to decrease with structural maturity of the causative fault. This threshold is generally consistent with a rupture strain limit (0.5%–1.5%) of intact rocks from laboratory measurements, and is higher than a commonly‐assumed strain threshold (0.5%) for inelastic deformation.

Allium victorialis L. (Family: Liliaceae) is an herb with nutritional and medicinal properties. In Jilin Province, China, A. victorialis is mainly distributed in the Changbai Mountains and grows in various habitat conditions. However, the relationship between habitat factors and the nutritional quality of A. victorialis in the Changbai Mountains has not yet been examined. We assessed the nutritional quality of five A. victorialis populations growing in five different habitats in the Changbai Mountains and analyzed the relationship between nutritional quality and habitat factors. Allium victorialis populations in this region were primarily found in the undergrowth at elevations above 500 m and within specific ranges of air temperature, air relative humidity, soil temperature, and soil water content. Among the habitat factors investigated, canopy density significantly affected several nutritional components of A. victorialis; however, elevation had a significant effect only on the total flavonoid content, and the vitamin C content was not strongly associated with the main habitat factors in this study. During germplasm selection and artificial cultivation, it is important to simulate the growth conditions of the original habitat. Our results provide useful information for site selection and environmental condition optimization for the artificial cultivation of A. victorialis.

AbstractBackground The effectiveness of nirmatrelvir–ritonavir has mainly been shown in non-hospitalized patients with mild-to-moderate coronavirus disease 2019 (COVID-19). The real-world effectiveness of nirmatrelvir-ritonavir urgently needs to be determined using representative in-hospital patients with COVID-19 during the Omicron wave of the pandemic.Methods We performed a multicentre, retrospective study in five Chinese PLA General Hospital medical centers in Beijing, China. Patients hospitalized with COVID-19 from 10 December 2022 to 20 February 2023 were eligible for inclusion. A 1:1 propensity score matching was performed between the nirmatrelvir-ritonavir group and the control group.Results 1010 recipients of nirmatrelvir-ritonavir and 1010 matched controls were finally analyzed after matching. Compared with matched controls, the nirmatrelvir-ritonavir group had a lower incidence rate of all-cause death (4.6/1000 vs. 6.3/1000 person-days, p = 0.013) and a higher incidence rate of clinical improvement (47.6/1000 vs. 45.8/1000 person-days, p = 0.012). Nirmatrelvir-ritonavir was associated with a 22% lower all-cause mortality and a 14% higher incidence of clinical improvement. Initiation of nirmatrelvir-ritonavir within 5 days after symptom onset was associated with a 50% lower mortality and a 26% higher clinical improvement rate. By contrast, no significant associations were identified among patients receiving nirmatrelvir-ritonavir treatment more than 5 days after symptom onset. Nirmatrelvir-ritonavir was also associated with a 50% increase in survival days and a 12% decrease in days to clinical improvement.Conclusion Among hospitalized patients with COVID-19 during the Omicron wave in Beijing, China, the early initiation of nirmatrelvir-ritonavir was associated with clinical benefits of lowering mortality and improving clinical recovery.

Abstract Background Cerebral vasospasm (CV) can contribute to significant morbidity in subarachnoid hemorrhage (SAH) patients. A key unknown is how CV induction is triggered following SAH. Methods Human aneurysmal blood and cerebral spinal fluid were collected for evaluation. To confirm mechanism, c57/bl6 wild type and c57/bl6 IL-6 female knockout (KO) mice were utilized with groups: saline injected, SAH, SAH + IL-6 blockade, SAH IL-6 KO, SAH IL-6 KO + IL-6 administration, SAH + p-STAT3 inhibition. Dual-labeled microglia/myeloid mice were used to show myeloid diapedesis. For SAH, 50 μm blood was collected from tail puncture and administered into basal cisterns. IL-6 blockade was given at various time points. Various markers of neuroinflammation were measured with western blot and immunohistochemistry. Cerebral blood flow was also measured. Background: As a class of promising bone augmentation materials, bone cements have attracted particular attention. Due to various limitations, the current bone cements are still imperfect. In this study, an injectable pH neutral bioactive bone cement (PSC/CSC) was developed by mixing phosphosilicate bioactive glass (PSC) and α-calcium sulfate hemihydrate (CSH), with the goal of optimizing bone defects repairs. Methods: A range of compositions (PSC/CSC: 10P/90C, 30P/70C, 50P/50C) were developed and their physicochemical properties evaluated. Their bone regeneration ability was compared to those of two widely used bone cements as controls (calcium phosphate cement (CPC) and Genex®) in rabbit femoral condyle bone defect models for 4, 8 and 12 weeks. Based on physicochemical properties and in vivo bone regeneration ability, the PSC/CSC exhibited the best outcomes was selected. Then, in vitro, the effects of selected PSC/CSC, CPC and Genex® extracts on MC3T3-E1 cell proliferation, migration and osteogenesis as well as angiogenesis of HUVECs were examined. Results: Based on physicochemical properties, the 30P/70C formula exhibited suitable operability and compressive strength (3.5 ± 0.3 MPa), which fulfilled the requirements for cancellous bone substitutes. In vivo, findings from micro-CT and histological analyses showed that the 30P/70C formula better promoted bone regeneration, compared to 10P/90C, 50P/50C, CPC and Genex®. Hence, 30P/70C was selected as the ideal PSC-based cement. In vitro, the 30P/70C extracts showed better promotion of cell viability, alkaline phosphatase (ALP) activity, calcium mineral deposition, mRNA and protein expression levels of osteogenesis in MC3T3-E1 cells, further supporting its superiority. Meanwhile, the 30P/70C extracts also showed better stimulation of HUVECs proliferation and angiogenesis. Conclusion: The new composite cement, 30P/70C, is a favorable bioactive glass-based bone cement with suitable operability, compressive strength and bone regeneration ability. The translational potential of this article: Clinically, treatment of large bone defects is still a major challenge for orthopaedic trauma. We showed that 30P/70C has the potential to be clinically used as an injectable cement for rapid bone repairs and reconstruction of critical sized bone defects.

Abstract Pancreatic acinar cells display a remarkable degree of plasticity and can dedifferentiate into ductal-like progenitor cells by a process known as acinar ductal metaplasia (ADM). ADM is believed to be one of the earliest precursor lesions toward the development of pancreatic ductal adenocarcinoma and maintaining the pancreatic acinar cell phenotype suppresses tumor formation. The effects of a novel pStat3 inhibitor (LLL12B) and the histone deacetylase (HDAC) inhibitor trichostatin A (TSA) were investigated using 3-D cultures from p48Cre/+ and p48Cre/+LSL-KrasG12D/+ (KC) mice. LLL12B and TSA inhibited ADM in both KC and p48Cre/+ mouse pancreatic organoids. Furthermore, treatment with LLL12B or TSA on dedifferentiated acini from p48Cre/+ and KC mice that had undergone ADM produced morphologic and gene expression changes that suggest a reversal of ADM. Validation experiments using qRT-PCR (p48Cre/+ and KC) and RNA sequencing (KC) of the LLL12B and TSA treated cultures showed that the ADM reversal was more robust for the TSA treatments. Pathway analysis showed that TSA inhibited Spink1 and PI3K/AKT signaling during ADM reversal. The ability of TSA to reverse ADM was also observed in primary human acinar cultures. We report that pStat3 and HDAC inhibition can attenuate ADM in vitro and reverse ADM in the context of wild-type Kras. Our findings suggest that pharmacological inhibition or reversal of pancreatic ADM represents a potential therapeutic strategy for blocking aberrant ductal reprogramming of acinar cells.

Abstract Background In critically ill patients requiring extracorporeal membrane oxygenation (ECMO) therapy, early initiation of continuous renal replacement therapy (CRRT) and beta-blockade of catecholamine-induced inotropic effects may improve outcomes. Methods A 2 × 2 partial factorial randomized controlled trial in eligible ECMO patients without a clear indication or contraindication to either intervention is centrally randomly assigned to (A) early or conventional-indicated CRRT and/or (B) beta-blocker or usual care. The primary outcome is all-cause mortality at 30 days for both arms. A total of 496 participants provides 80% power to determine a 20% risk reduction in mortality at 30 days with 5% type I error. Discussion This trial will help define the role of early CRRT and beta-blockade in ECMO patients. There have been 89 patients enrolled at 10 hospitals in study A and is ongoing. However, study B was stopped in August 2019 in the absence of any patients being enrolled. Trial registration ClinicalTrials.gov NCT03549923 . Registered on 8 June 2018.
