In this work, poly(N-isopropylacrylamide) (PNIPAM) based chemically cross-linked hydrogels are used as novel precipitation and stabilization media for solid lipid nanoparticles (SLNs) for the first time. The hydrogels and the hybrid thermoresponsive composite hydrogels with SLNs were characterized by SEM, DSC, DLS and rheometric analysis. The results showed that the SLNs obtained directly in the gel matrix by the newly devised method were well-dispersed and remained stable for one month. A remarkable advantage of this approach is that it yields the thermoresponsive nanocomposite hydrogels in a single step. This approach is a significant advancement in the preparation of hybrid thermoresponsive nanocomposite systems based on smart gels and SLNs for their use in biomedical applications.

Biowaste-derived substances isolated from green compost (BBS-GC) are environmentally friendly reactants similar to humic substances, which contain multiple functionalities, that are suitable for adsorbing different kinds of pollutants in wastewater. Herein, sodium alginate (derived from brown algae) cross-linked with both Ca2+ ions and BBS-GC in the form of hydrogels and dried films are proposed as green, easy-to-form, and handleable materials for tertiary water treatments. The results show that both hydrogels and films are mechanically stable and can effectively remove differently charged dyes through an adsorption mechanism that can be described by the Freundlich model. BBS-GC-containing gels always performed better than samples prepared without BBS-GC, revealing that such unconventional materials can integrate waste valorization and water decontamination, potentially providing social and environmental benefits.

Agar is an interesting and versatile biopolymer capable of forming hydrogels useful in different applications. In particular, in the field of Cultural Heritage agar gels are often used as tools for cleaning the surfaces of artifacts. Agar gels are thermoreversible, peelable materials, exhibiting high retention of water and an intrinsic cleaning capability that can be increased and modulated by loading different cleaning agents. Agar gels are able to solubilize and retain dirt, and allow a confined release of the liquid phase, which is especially important when they are applied on water sensitive substrates. A detailed knowledge of the role of water in agar gels and of water/agar interactions is important to fully understand the properties and performances of agar gels as cleaning materials. Here we report the results of a systematic study aimed at exploring the water state and the types of water (i.e. bound non-freezable, bound freezable and freezable bulk-like) in agar hydrogels prepared from four different types of commercial agar powders. The influence of agar concentration on the water behaviour in the hydrogels was also investigated. The total water content, the identification and quantification of the different types of water was carried out by gravimetric, thermogravimetric and calorimetric analyses, while water mobility and localization in the gel network were investigated by unilateral H-1 NMR. Moreover, H-1 NMR depth profiles of agar gels applied on Noto stone specimens were collected to monitor the gel deformation due to water release and to evaluate the ability of the gel in controlling the water release into a porous material. Effectiveness and selectivity are the main requirements for a proper cleaning of artworks. The complexity of the soiling and the limits of traditional cleaning methods make it necessary to increase the knowledge on the chemical and physical mechanisms involved in cleaning methods Agar gels have been successfully used in the restoration practise, but it is not yet clarified if their cleaning capability is due only to the solvent action or also to the polysaccharide structure. A multi-analytical procedure including elemental analyses, vibrational spectroscopies, thermogravimetric analysis and pyrolysis-gas chromatography/mass spectrometry is proposed to characterize agar powders in terms of morphology, composition and thermal stability, and to differentiate agar products intended for different applications, i.e. conservation of Cultural Heritage, biology and food industry. The multi-analytical approach proposed allows differentiating agar samples with slight compositional differences. In particular, agar products can be rapidly differentiated by a semi-quantitative analysis of pyrolysis-gas chromatography/mass spectrometry data. The results presented show that agar powders used for cleaning treatments contain impurities probably derived from cellulosic residues.

The possibility of successfully applying a common GC–MS procedure for identification in one step of all types of dyes from plants of unknown origin and from historical objects is particularly attractive due to the high separation efficiency of the capillary columns, the MS detection sensitivity and the reproducibility of results. In this work, GC–MS analysis, previously and successfully used for the characterization of anthraquinones, flavonoids and tannins from plant extracts and historical samples, has been tested on indigoid dyestuffs. An analytical procedure based on the silylating agent N,O-bis-(trimethylsilyl)trifluoroacetamide (BSTFA) with 1 % trimethylchlorosilane (TMCS) was applied to pure molecules of indigotin and indirubin and to plant extracts of Indigofera tinctoria L. and Isatis tinctoria L. Preliminary tests have been done to establish the chromatographic conditions and the derivatization amounts most suitable for the simultaneous detection of indigoid molecules and of the other natural compounds, such as fatty acids, carboxylic acids and sugars, contained within the plant extracts. In order to assess the capacity and the sensitivity of the analytical procedure in typical archaeometric applications, wool samples dyed in the laboratory with indigo were analysed by mimicking the sample amounts typically available with historical objects. The electron ionization (EI) spectra of the main silylated derivatives of indigoid molecules obtained in this way constitute the necessary data set for the characterization of natural extracts and historical works of art. Subsequently, the procedure has been applied to historical samples for the detection of indigo and of other dyestuffs eventually contained in samples. Additional information, useful for restoration and preservation of works of art, could be also obtained on the nature of stains and smudges present on the sampled textile material. The GC–MS method turns out to be an efficient and fast analytical tool also for the identification of natural indigo in plants and textile artefacts, providing results complementary to those from high-performance liquid chromatography (HPLC).

Agar is a natural polymer commonly used in various fields of application ranging from cosmetics to the food industry. In particular, for over forty years agar gels have been used in the field of conservation of Cultural Heritage where they are considered as one of the main well-performing tools in cleaning procedures. In the present work, the relation between the chemical composition and the mechanical strength of four different agar hydrogels was evaluated by comparing the results obtained via pyrolysis-gas chromatography/mass spectrometry and rheological characterization. Agar composition was studied by means of a pyrolysis-gas chromatography/mass spectrometry approach in order to differentiate the anhydrous, galactose and glucose units. Pristine agar gels, gels after double annealing, and gels with and without chelating agent were studied by means of amplitude, frequency and time sweep rheological tests to evaluate all the preparation approaches commonly used by conservators, also taking into account changes in the transparency via UV–vis spectroscopy. A high percentage of anhydrous units in the polymer backbone was found to provide superior mechanical stiffness to the pristine hydrogels, even if it did not seem to affect their long-term stability. The annealing process significantly improved the rheological response of galactose-rich agar hydrogels being able to promote the establishment of additional crosslinking points, whereas the additive presence showed to improve the hydrogel stiffness owing to a more structured polymer network. Moreover, the progressive reduction of the impurities and/or network defects within the hydrogels occurring due to the annealing process slightly increased the transparency of the hydrogels, which is an important aspect for applications in the conservation of Cultural Heritage.

Nine black powders found in Pompeii houses in three different types of bronze vessels (cylindrical theca atramentaria, unguentaries, and aryballoi) were characterized in order to assess a correspondence between the composition and the type of vessel and, possibly, to verify if these powders were inks or not. For the compositional characterization, a multi-analytical approach was adopted, which involved the use of scanning electron microscopy-energy dispersive X-ray, Fourier-transformed infrared spectroscopy, Raman, X-ray diffraction, electron paramagnetic resonance spectroscopy, thermogravimetric analysis, gas chromatography coupled with mass spectrometry (GC/MS), and pyrolysis GC/MS. Powders contained in cylindrical theca atramentaria form a homogeneous group, and their organic and inorganic compositions suggest that they were writing inks, while powders contained in unguentaries and aryballoi could have had several different uses, including writing inks and cosmetics. Furthermore, the composition profile of the powders found in cylindrical cases shows that, at 79 AD: , in Pompeii, carbon-based inks were still used for writing, and iron gall inks had not been introduced yet.

Plant gums are complex polysaccharides used in the field of cultural heritage especially as binding media. Classification of polysaccharides may be achieved on the basis of monosaccharides composition after cleavage of glycosidic bond. Characterization of plant gums in works of art is complicated by the necessity of to use a method minimally invasive and requiring a small mount of sample. Pyrolisys is an useful method to obtain polysaccharides decomposition and generally pyrolysis products can be identified by the use of gas chromatography–mass spectrometry. This paper describes a method where two plant gums, arabic and tragacanth, were pyrolized in presence of silylating agents (HMDS e BSTFA alone and with TMCS as catalyst) using an on-line Py-GC/MS apparatus. Some characteristic trimethylsilyl derivatives of monosaccharides were identified on the basis of mass spectra. The presence of characteristic pyrolysis products of sugars allows to distinguish the two gums.

The monomeric components of a series of lattices suitable as protective coatings for the safeguard of cultural heritage, and containing unfluorinated and partially fluorinated acrylic units were determined by pyrolysis-gas chromatography/mass spectrometry. Their identification was possible on the basis of a good knowledge of the degradative behavior, also gained through the direct study of different reference fluoropolymers, which enabled to distinguish between monomers and monomer-related products of degradation. All the samples were identified as copolymers of an acrylic unit, either partially fluorinated or unfluorinated, with a methacrylic unit. Only in two cases different amounts of styrene as monomeric component were also identified.

Classification of polysaccharides may be achieved on the basis of the monosaccharidic composition after cleavage of glycosidic bonds. Pyrolysis is a useful method to obtain polysaccharide decomposition and generally pyrolysis products can be identified by the use of gas chromatography/mass spectrometry. This paper describes an on-line silylation pyrolysis approach and compares the efficiency of two derivatization reagents, hexamethyldisilazane (HMDS) and N , O -bis(trimethylsilyl)trifluoroacetamide (BSTFA), used alone and in combination with trimethylchlorosilane (TMCS) as a catalyst. Some characteristic trimethylsilyl derivatives of monosaccharides have been identified on the basis of their mass spectra. The detected products include trimethylsilyl derivatives of anhydrosugars, cyclopentenones, furancarboxyaldehydes and, at the best silylating conditions, pertrimethylsilylated monosaccharides. The optimization of the derivatization process is under study, as well as the possibility to extend the use of this approach to unknown polysaccharidic samples, with particular attention to cultural heritage materials.

Poly(vinylacohol) (PVA) is a versatile polymer capable of forming hydrogels useful in a wide range of applications. Hydrogels based on cross-linking of PVA with borax, and able to incorporate organic solvents, have been used in the field of cultural heritage as tool for cleaning the surfaces. Poly(ethylenoxide) (PEO) has been introduced in the formulations of PVA-borax, with the aim of increasing the gel compatibility with organic solvents and of improving the mechanical properties of the materials. The formulations were investigated with rheological characterization, solvent loss measurements and study of the free water through the determination of the Free Water Index. The PEO molecules do not participate in the formation of the network but have been found to be responsible for a decrement of the storage moduli, explained by a spacing between crosslinks and the partial disentanglement of the PVA chains. At the same time PEO produces in the gel an increase of the apparent relaxation time, thus stabilizing the dispersions and enhancing their ability to maintain the shape. Cleaning tests for removal of an acrylic coating on limestone have been also successfully performed.

The method of Thermally assisted Hydrolysis and Methylation (THM) was applied to the analysis of plant gums that can be found in artistic and archaeological objects. The main products obtained by THM of gums arabic, cherry, tragacanth, ghatti and karaya are permethylated and partially methylated 3-deoxyaldonic acids, characteristic products obtained from alkaline hydrolysis of monosaccharides. These markers allowed only a preliminary distinction between the different gums because they are not representative of the entire monosaccharides profile. As classification on the basis of sugars quantification is impossible, multivariate data analysis was employed, using the peak areas of THM products as variables. Calculations were performed on several different sets of data, enhancing the capability of the method to differentiate gums, also when submitted to accelerated ageing treatments. A case study on samples coming from ancient Egyptian cartonnage funerary masks is presented.

Analytical pyrolysis with thermally assisted hydrolysis and methylation was employed to investigate ancient ointments collected from Spanish vessels coming from the sixteenth century pharmacies. The ointments were reproduced on the basis of historical recipes and characterization was made in comparison with real samples. Characteristic markers indicate the presence of beeswax, of animal and plant lipids, and of natural resins. Analyses of old samples are consistent with the modern reproductions and with the analysis of raw materials. Multivariate data analysis was used to discriminate between the different types of lipidic materials, also in connection with their relative amount in the samples.

Chemical analysis of ancient residues of pharmaceutical or cosmetic preparations such as balms or ointments is made problematic by the high complexity of these mixtures, composed of organic and inorganic materials. Consequently, a multi-analytical approach and special caution in the interpretation of the results are necessary. In order to contribute to the improvement of analytical strategies for the characterization of complex residues and to reconstruct ancient medical practices, a replica of a pharmaceutical formulation of the seventeenth century was prepared in the laboratory according to a historically documented recipe. In a round robin exercise, a portion of the preparation was analysed as a blind sample by 11 laboratories using various analytical techniques. These included spectroscopic, chromatographic and mass spectrometric methods. None of the laboratories was able to completely reconstruct the complex formulation, but each of them gave partial positive results. The round robin exercise has demonstrated that the application of a multi-analytical approach can permit a complete and reliable reconstruction of the composition. Finally, on the basis of the results, an analytical protocol for the study of residues of ancient medical and pharmaceutical preparations has been outlined. © 2011 Springer-Verlag.

Natural organic materials used to prepare pharmaceutical mixtures including ointments and balsams have been characterized by a combined non-destructive spectroscopic analytical approach. Three classes of materials which include vegetable oils (olive, almond and palm tree), gums (Arabic and Tragacanth) and beeswax are considered in this study according to their widespread use reported in ancient recipes. Micro-FTIR, micro-Raman and fluorescence spectroscopies have been applied to fresh and mildly thermally aged samples. Vibrational characterization of these organic compounds is reported together with tabulated frequencies, highlighting all spectral features and changes in spectra which occur following artificial aging. Synchronous fluorescence spectroscopy has been shown to be particularly useful for the assessment of changes in oils after aging; spectral difference between Tragacanth and Arabic gum could be due to variations in origin and processing of raw materials. Analysis of these materials using non-destructive spectroscopic techniques provided important analytical information which could be used to guide further study.

Plant gums are present in works of art as binding media for watercolours and adhesives for cellulosic substrates. Thermally assisted hydrolysis and methylation (THM) in combination with analytical pyrolysis coupled to GC/MS has been applied to the characterisation of plant gums typically used in artworks. THM products from standard samples of arabic gum, tragacanth gum and cherry gum were characterised. The main products identified are permethylated and partially methylated aldonic acids, characteristic of specific epimeric sugars. Aldonic acids were formed by alkaline hydrolysis of free reducing sugars and of reducing polysaccharide terminal groups, while methylation occurs during pyrolysis. The presence of these characteristic markers allows gum identification. A systematic analysis of all the parameters that can affect the marker yields was performed. In particular, the influence of pyrolysis temperature, reagent concentration and contact time between tetramethylammonium hydroxide and sample were studied, and different kinds of sample preparation procedures were tested. Some analyses on real watercolours were performed, and gum binders were classified using the peak area ratio of the main monosaccharide markers.
