Dyslipidemia, a major risk factor for cardiovascular diseases (CVDs), is modifiable by diet and lifestyle changes. A large population with mild to moderate dyslipidemia is at risk of developing CVDs, and early initiation of preventive measures can avert advancing into severe medical conditions. Studies suggest increasing slowly digestible starch (SDS) in diets can help lower blood lipids. We processed dehulled adlay, a cereal rich in bioactive compounds, such as polyphenols and phytosterols, into an instant meal by extrusion and milling and then assessed its starch composition and in vitro digestibility. The dehulled adlay was found to consist of 32% SDS and resistant starch combined. Then, eligible subjects with dyslipidemia were recruited to explore the adlay’s hypolipidemic potential, safety, and acceptability. Subjects consumed the dehulled adlay as the sole carbohydrate source in their breakfast, without changing other components in the diet or lifestyle, for 12 weeks. After intervention, serum total cholesterol (TC) decreased significantly in subjects with hypercholesterolemia. In addition, both TC and triglyceride levels decreased significantly in those above 50 years old. In conclusion, the extruded dehulled adlay displays potential for favorably modulating blood lipids, and the effect is more pronounced in the middle-aged population.

The results presented differences between different types of impulse buying. The findings contribute to the future study of consumer behavior and provide implications for commercial providers.

Abstract The most widely used method for intracellular RNA fluorescence labeling is MS2 labeling, which generally relies on the use of multiple protein labels targeted to multiple RNA (MS2) hairpin structures installed on the RNA of interest (ROI). While effective and conveniently applied in cell biology labs, the protein labels add significant mass to the bound RNA, which potentially impacts steric accessibility and native RNA biology. We have previously demonstrated that internal, genetically encoded, uridine-rich internal loops (URILs) comprised of four contiguous UU pairs (8 nt) in RNA may be targeted with minimal structural perturbation by triplex hybridization with 1 kD bifacial peptide nucleic acids (bPNAs). A URIL-targeting strategy for RNA and DNA tracking would avoid the use of cumbersome protein fusion labels and minimize structural alterations to the RNA of interest. Here we show that URIL-targeting fluorogenic bPNA probes in cell media can penetrate cell membranes and effectively label RNAs and RNPs in fixed and live cells. This method, which we call fluorogenic U-rich internal loop (FLURIL) tagging, was internally validated through the use of RNAs bearing both URIL and MS2 labeling sites. Notably, a direct comparison of CRISPR-dCas labeled genomic loci in live U2OS cells revealed that FLURIL-tagged gRNA yielded loci with signal to background up to 7X greater than loci targeted by guide RNA modified with an array of eight MS2 hairpins. Together, these data show that FLURIL tagging provides a versatile scope of intracellular RNA and DNA tracking while maintaining a light molecular footprint and compatibility with existing methods.

To increase the knowledge of bus evacuation in tunnel fire scenarios, we investigated the bus evacuation flow rate in the event of a tunnel fire, starting with a video analysis of the 2012 Hsuehshan tunnel fire in Taiwan, to gain a preliminary understanding of an actual bus evacuation flow rate. Two evacuation situations were manifested with different flow rates: the first one in which bus passengers non-urgently evacuated as in the case of a daily bus experience (the first seven evacuees, 0.28 PPS (Person Per Second)) and another situation in which passengers feel the urgency to evacuate (the next twelve evacuees, 0.67 PPS). Based on the investigation of the real fire incident, we conducted a bus evacuation experiment and a bus alighting observation. And the results of flow rates of normal exit were within 0.69–0.88 and 0.21–0.48 PPS, respectively. These results were similar to the actual flow rates in the two assumed evacuation situations in the Hsuehshan tunnel fire incident (nonurgent and urgent), respectively. Our approach can provide a viable approach to describe evacuation flow rates during tunnel fires. Moreover, using the flow rate of the normal alighting situation was preferable for highly reliable tunnel fire safety evaluation.

Nuclear chromosome compaction is non-random and dynamic. The spatial distance among genomic elements instantly modulates transcription. Visualization of the genome organization in the cell nucleus is essential to understand nuclear function. In addition to cell type-dependent organization, high-resolution 3D imaging shows heterogeneous compaction of chromatin organization among the same cell type. Questions remain to be answered if these structural variations were the snapshots of dynamic organization at different time points and if they are functionally different. Live-cell imaging has provided unique insights into dynamic genome organization at short (milliseconds) and long (hours) time scales. The recent development of CRISPR-based imaging opened windows for studying dynamic chromatin organization in single cells in real time. Here we highlight these CRISPR-based imaging techniques and discuss their advances and challenges as a powerful live-cell imaging method that poses high potential to generate paradigm-shifting discoveries and reveal functional implications of dynamic chromatin organization.

Abstract Background Cholangitis may affect liver failure of biliary atresia (BA) patients after Kasai portoenterostomy (KP). We examined whether the number of cholangitis episodes could be a prognostic marker for liver transplant (LT) in children with BA after Kasai portoenterostomy (KP). Methods Data for BA patients born after 1998 and undergoing KP were obtained from National Health Insurance Research Database (NHIRD), Taiwan. Patients were followed up until the end of 2011. An optical fiber sensing system integrating a fiber Bragg grating (FBG) sensor, a long-period fiber grating (LPFG) optical filter and a photodetector is presented to monitor the dynamic response of a structure subjected to base excitation and impact loading. The FBG sensor is attached to a test specimen and connected to an LPFG filter. As the light reflected from the FBG sensor is transmitted through the long-period fiber grating filter, the intensity of the light is modulated by the wavelength, which is affected by the strain of the FBG. By measuring the intensity of the light using a photodetector, the wavelength reflected from the FBG sensor can be demodulated, thus leading to the determination of the strain in the structure. To demonstrate its effectiveness, the proposed sensing system was employed to measure the dynamic strain of a beam subjected to mechanical testing. The mechanical tests comprised three load scenarios: base excitation by a shaker at resonant frequency, impact loading by a hammer and shock test on a drop table. To monitor the dynamic strain during the test and validate the accuracy of the measurement of the FBG sensor, strain gauge was used as reference. Experimental results show good correlation between the measurements of FBG sensor and strain gauge. The present work provides a fast response and easy-to-implement optical fiber sensing system for structural health monitoring based on real-time dynamic strain measurements.

Pumpkin (Cucurbita moschata) is a popular and nutritious vegetable consumed worldwide. The overall purpose of this study was to evaluate the effects of C. moschata fruit extract (CME) on anti-fatigue and ergogenic functions following physiological challenges. Male ICR mice from four groups designated vehicle, CME-50, CME-100 and CME-250, respectively (n = 8 per group in each test) were orally administered CME for 14 days at 0, 50, 100 and 250 mg/kg/day. The anti-fatigue activity and exercise performance were evaluated using exhaustive swimming time, forelimb grip strength, as well as levels of plasma lactate, ammonia, glucose, and creatine kinase after an acute swimming exercise. The resting muscular and hepatic glycogen was also analyzed after 14-day supplementation with CME. Trend analysis revealed that CME treatments increased grip strength. CME dose-dependently increased 5% body weight loaded swimming time, blood glucose, and muscular and hepatic glycogen levels. CME dose-dependently decreased plasma lactate and ammonia levels and creatine kinase activity after a 15-min swimming test. The mechanism was relevant to the increase in energy storage (as glycogen) and release (as blood glucose), and the decrease of plasma levels of lactate, ammonia, and creatine kinase. Therefore, CME may be potential for the pharmacological effect of anti-fatigue.
