Near-infrared (NIR)-light-modulated photothermal thrombolysis has been investigated to overcome the hemorrhage danger posed by clinical clot-busting substances. A long-standing issue in thrombosis fibrinolytics is the lack of lesion-specific therapy, which should not be ignored. Herein, a novel thrombolysis therapy using photothermal disintegration of a fibrin clot was explored through dual-targeting glycol chitosan/heparin-decorated polypyrrole nanoparticles (GCS-PPY-H NPs) to enhance thrombus delivery and thrombolytic therapeutic efficacy. GCS-PPY-H NPs can target acidic/P-selectin high-expression inflammatory endothelial cells/thrombus sites for initiating lesion-site-specific thrombolysis by hyperthermia using NIR irradiation. A significant fibrin clot-clearance rate was achieved with thrombolysis using dual-targeting/modality photothermal clot disintegration in vivo. The molecular level mechanisms of the developed nanoformulations and interface properties were determined using multiple surface specific analytical techniques, such as particle size distribution, zeta potential, electron microscopy, Fourier-transform infrared spectroscopy (FTIR), wavelength absorbance, photothermal, immunofluorescence, and histology. Owing to the augmented thrombus delivery of GCS-PPY-H NPs and swift treatment time, dual-targeting photothermal clot disintegration as a systematic treatment using GCS-PPY-H NPs can be effectively applied in thrombolysis. This novel approach possesses a promising future for thrombolytic treatment.

Mycobacterium fortuitum group (M. fortuitum), also known as rapidly growing Mycobacteria, can cause pyogenic infections in human beings, most commonly in immunocompromised patients. Herein, we present a 40-year-old immunocompetent male patient who underwent planned excision of a sebaceous cyst in the abdominal wall. He suffered from tender erythematous lesions with purulent discharge around the healing wound that developed 2 weeks after surgery. Gram stain, bacterial and fungal culture results of the wound were negative. A diagnosis of non-tuberculous mycobacteria was made from a wound culture from the area of operative debridement, which was subsequently confirmed to be M. fortuitum group using PCR-restriction fragment length polymorphism analysis of the hsp65 gene. The patient received 4 weeks of parenteral imipenem/cilastatin 500 mg every 6 hours and amikacin 500 mg every 12 hours, plus oral clarithromycin 500 mg twice daily, and the wound recovered completely. He was discharged and followed up regularly at our outpatient clinic, and continued taking oral ciprofloxacin and clarithromycin 500 mg twice daily for 6 months. This case highlights the importance of strict aseptic precautions even during minor procedures, and also the characteristics of M. fortuitum infections in immunocompetent patients, which usually develop as localized postsurgical wound infections. We also share our experience in successfully treating a M. fortuitum complicated skin and soft tissue infection.

Enhancements in 630.0 nm airglow around midnight at equatorial latitudes were observed by many optical observations. Such features had been suggested as the signature of thermospheric midnight temperature maximum (MTM) effect, which was associated with temperature and meridional neutral winds. This study investigates the influence of neutral temperature and meridional neutral wind on the volume emission rates of the 630.0 nm nightglow. We utilize the SAMI2 model to simulate the charged and neutral species at the 630.0 nm nightglow emission layer under different temperatures with and without the effect of neutral wind. The results show that the neutral wind is more efficient than temperature variation in affecting the nightglow emission rates. However, the emission rate features a local maximum in its variation with the temperature. Two kinds of tendencies can be seen regarding the temperature that corresponds to the turning point, which is named the turning temperature (Tt) in this study: firstly, Tt decreases with the emission rate for the same altitude; secondly, for approximately the same emission rate, Tt increases with the altitude.

Anthrax is a disease caused by the bacterium Bacillus anthracis, which results in high mortality in animals and humans. Although some of the mechanisms are already known such as asphyxia, extensive knowledge of molecular pathogenesis of this disease is deficient and remains to be further investigated. Lethal toxin (LT) is a major virulence factor of B. anthracis and a specific inhibitor/protease of mitogen-activated protein kinase kinases (MAPKKs). Anthrax LT causes lethality and induces certain anthrax-like symptoms, such as anemia and hypoxia, in experimental mice. Mitogen-activated protein kinases (MAPKs) are the downstream pathways of MAPKKs, and are important for erythropoiesis. This prompted us to hypothesize that anemia and hypoxia may in part be exacerbated by erythropoietic dysfunction. As revealed by colony-forming cell assays in this study, LT challenges significantly reduced mouse erythroid progenitor cells. In addition, in a proteolytic activity-dependent manner, LT suppressed cell survival and differentiation of cord blood CD34(+)-derived erythroblasts in vitro. Suppression of cell numbers and the percentage of erythroblasts in the bone marrow were detected in LT-challenged C57BL/6J mice. In contrast, erythropoiesis was provoked through treatments of erythropoietin, significantly ameliorating the anemia and reducing the mortality of LT-treated mice. These data suggested that suppressed erythropoiesis is part of the pathophysiology of LT-mediated intoxication. Because specific treatments to overcome LT-mediated pathogenesis are still lacking, these efforts may help the development of effective treatments against anthrax.

This study investigates the distributions and occurrence mechanisms of the global local-midnight airglow brightness through FORMOSAT-2/ISUAL satellite imaging observations. We focus on the OI 630.0 nm nightglow emission at altitudes of ~250 km along equatorial space. The database used in this study included data from 2007 to 2008 under solar minimum conditions. The data were classified into four specified types in the statistical study. We found that the occurrence of equatorial brightness was often in the vicinity of the geographic equator and mostly at equinoxes with a tendency to move toward the summer hemisphere as the season changes. Conjugate brightness occurring simultaneously on both sides of the geomagnetic equator was observed predominantly in the northern winter. Furthermore, midnight brightness appeared to have lower luminosity from May to July. We suggest that the global midnight brightness associated with the locations and seasons was the result of several effects which include the influence of the thermospheric midnight temperature maximum (MTM), summer-to-winter neutral wind, and ionospheric anomalies.

In the past 6 years, the ISUAL payload onboard the Taiwanese FORMOSAT-2 satellite has successfully carried out the primary mission of performing a global survey of transient luminous events (TLE) and lightning activities. The observation data have been used to construct the first global distribution map of TLEs and to infer their occurrence rates. To register dim TLE emissions at night, ISUAL employs electron-multiplying devices like a photomultiplier, multi-anode array, and micro-channel plate to intensify the photons emitted by transient events. These devices, however, will degrade gradually with age. The degradation rate and the performance change were monitored through periodic calibration observations and are investigated carefully in order to achieve precise photometry and accurate statistics. The annual degradation rates up to 14% were identified for some of the sensor elements. This paper presents the variations of instrument efficiencies and overall detection capabilities, before and after applying a gain-adjustment to compensate for sensor degradation. The resulting stable trend of the ISUAL trigger and lightning rates suggests that the bias from instrument degradation and gain adjustments is minimal. This point is also supported by the near constant sprite and halo detection rates deduced from different statistical temporal intervals. Hence, the anomalies in the ISUAL elve detection rate for 2008 ~ 2009 likely are due to the El Niño and Southern Oscillation (ENSO) events. Forecast based on the ISUAL performance trend also indicates that the ISUAL sensors have sufficient sensitivity margins to operate efficiently until the end of 2013, or even beyond.

[1] A previous study of lightning-induced transient emissions in and below the OH airglow layer using observations by the Imager of Sprites and Upper Atmospheric Lightning (ISUAL) CCD camera onboard the FORMOSAT-II satellite showed that intensity enhancements occurred more frequently in the OH airglow layer. Here we show the results of new observations made in December 2009 and January 2010 using a narrowband 630 nm filter and spectrophotometer and present further analysis. We estimated the N 2 1P intensity enhancements to be ~65% and 53% of the total intensity enhancements for the two events we analyzed using ISUAL and the spectrophotometer data in conjunction with a model for emissions of light and VLF perturbations from electromagnetic pulse sources (elves). Our analysis indicates that there is still somewhat considerable intensity enhancement (~1.25 kR) unaccounted for after the N 2 1 P contribution has been removed. Our study suggests that there might be OH emissions in elves and that OH species might also be involved in the lightning-induced process and contribute to the intensity enhancements that we observed.

[1] This is the first report of the most comprehensive 630.0 nm airglow limb images taken using Imager of Sprites and Upper Atmospheric Lightnings (ISUAL) onboard FORMOSAT-2. The limb scans reveal two distinct airglow layers: the upper one corresponds to the thermospheric O(1D) emission and the lower one corresponds to the OH (9–3) emissions. Sequences of such observations are combined to generate altitude-latitude maps of the emissions, which reveal intensity enhancements of both the layers at certain locations where they often appear to be linked/joined vertically. A detailed analysis of the location and occurrence of the enhancements in the entire observations, together with simulations of the emissions suiting the ISUAL limb geometry are carried out to understand the causes and related processes.
