The Singapore Preconception Study of Long-Term Maternal and Child Outcomes (S-PRESTO) is a preconception, longitudinal cohort study that aims to study the effects of nutrition, lifestyle, and maternal mood prior to and during pregnancy on the epigenome of the offspring and clinically important outcomes including duration of gestation, fetal growth, metabolic and neural phenotypes in the offspring. Between February 2015 and October 2017, the S-PRESTO study recruited 1039 Chinese, Malay or Indian (or any combinations thereof) women aged 18 to 45 years and who intended to get pregnant and deliver in Singapore, resulting in 1032 unique participants and 373 children born in the cohort. The participants were followed up for 3 visits during the preconception phase and censored at 12 months of follow up if pregnancy was not achieved (N=557 censored). Women who successfully conceived (N=475) were characterised at gestational weeks 6-8, 11-13, 18-21, 24-26, 27-28 and 34-36. Follow up of their index offspring (N=373 singletons) is on-going at birth, 1, 3 and 6 weeks, 3, 6, 12, 18, 24 and 36 months and beyond. Women are also being followed up post-delivery. Data is collected via interviewer-administered questionnaires, metabolic imaging (magnetic resonance imaging), standardized anthropometric measurements and collection of diverse specimens, i.e. blood, urine, buccal smear, stool, skin tapes, epithelial swabs at numerous timepoints. S-PRESTO has extensive repeated phenotypic data collected which include genetic and epigenetic sampling from preconception which is unique in mother-offspring epidemiological cohorts This enables prospective assessment of a wide array of potential determinants of future health outcomes in women from preconception to post-delivery and in their offspring across the earliest development from embryonic stages into early childhood. In addition the S-PRESTO study, draws from the three major Asian ethnic groups that represent 50% of the global population, increasing the relevance of its findings to global efforts to address non-communicable diseases.

Background: Infants with atopic dermatitis (AD) are reported to be at higher risk of early childhood caries (ECC) at 3-years, but the clinical validity of the reported link remains unknown. We investigated if clinical severity of AD in young children is associated with increased ECC risk at 3-years. Methods: In Growing Up in Singapore Towards healthy Outcomes (GUSTO) mother-offspring cohort, AD was diagnosed by trained physicians using Hanifin and Rajka criteria at 18-month and 3-year clinic visits (n=837). Rapid progress in biological sensing and imaging technologies offers new and exciting opportunities to identify biochemical changes fundamental to preclinical research that may develop into the next generation of point-of-care devices. Small upconverting fluorescent nanoparticles (UCNPs) are a special class of lanthanide-doped, optically active, microscopic particles that fluoresce upon near-infrared light activation. With the additional advantages of deep penetration, minimal photodamage, and low background fluorescence in biological tissues, biosensors incorporating these lanthanide nanoparticles are rapidly emerging as serious contenders to rival the traditional downconversion-based fluorescence nanoparticles. Emphasizing on two application areas, namely biosensing and bioimaging, the recent developments in lanthanide-based UCNP design for optimum upconversion efficiency are reviewed. The article discusses the use of UCNPs in biosensing and bioimaging, highlights the challenges that hamper further applications, and concludes with future directions.

Studies have suggested that maternal PUFA status during pregnancy may influence early childhood allergic diseases, although findings are inconsistent. We examined the relationship between maternal PUFA status and risk of allergic diseases in early childhood in an Asian cohort. Maternal plasma samples from the Growing Up in Singapore Towards Healthy Outcomes mother–offspring cohort were assayed at 26–28 weeks of gestation for relative abundance of PUFA. Offspring (n 960) were followed up from 3 weeks to 18 months of age, and clinical outcomes of potential allergic diseases (rhinitis, eczema and wheezing) were assessed by repeated questionnaires. Skin prick testing (SPT) was also performed at the age of 18 months. Any allergic disease with positive SPT was defined as having any one of the clinical outcomes plus a positive SPT. The prevalence of a positive SPT, rhinitis, eczema, wheezing and any allergic disease with positive SPT was 14·1 % (103/728), 26·5 % (214/808), 17·6 % (147/833), 10·9 % (94/859) and 9·4 % (62/657), respectively. After adjustment for confounders, maternal total n-3, n-6 PUFA status and the n-6:n-3 PUFA ratio were not significantly associated with offspring rhinitis, eczema, wheezing, a positive SPT and having any allergic disease with positive SPT in the offspring (P>0·01 for all). A weak trend of higher maternal n-3 PUFA being associated with higher risk of allergic diseases with positive SPT in offspring was observed. These findings do not support the hypothesis that the risk of early childhood allergic diseases is modified by variation in maternal n-3 and n-6 PUFA status during pregnancy in an Asian population.

We report the implementation of a unique multimodal nonlinear optical microscopy (i.e., coherent anti-Stokes Raman scattering (CARS), second harmonic generation (SHG), third harmonic generation (THG) and two photon excitation fluorescence (TPEF)) platform for label-free imaging of dentin. A picosecond tunable laser together with an OPO is used as the excitation source for simultaneously multimodal imaging. CARS shows similar information as TPEF in dentin, but it has a higher sectioning performance than TPEF and thus it is a good alternative for TPEF. Microtubule structure is revealed nearby dentin enamel junction (DEJ) from the multimodal images. This work demonstrates that combining different nonlinear optical imaging modalities can provide new insights into the understanding of morphological structures and biochemical/biomolecular distributions of the dentine without the need of labeling.

In this study, we are using two-photon (2-p) excited autofluorescence and second harmonic (SH) as imaging modalities to investigate dental sections that contains the enamel and the dentin. The use of near-infrared wavelengths for multiphoton excitation greatly facilitates the observation of these sections due to the hard tissue's larger index of refraction and highly scattering nature. Clear imaging can be achieved without feature altering preparation procedures of the samples. Specifically, we perform polarization resolving on SH and lifetime analysis on autofluorescence. Polarization resolved SH reflects the preferred orientation of collagen while very different autofluorescence lifetimes are observed from the dentin and the enamel. The origin of 2-p autofluorescence and SH signals are attributed to hydroxyapatite crystals and collagen fibrils, respectively. Hydroxyapatite is found to be present throughout the sections while collagen fibrils exist only in the dentin and dentinoenamel junctions.
