The immune system continuously interacts with tumors, possibly leading to systemic alterations in circulating immune cells. However, the potential of these cancer-associated changes for diagnostic purposes remains poorly explored. To investigate this, we conducted a comprehensive flow cytometric analysis of 452 peripheral blood mononuclear cell (PBMC) samples from 206 non-small-cell lung cancer (NSCLC) patients, 100 small-cell lung cancer (SCLC) patients, 94 healthy individuals, and 52 benign lung disease (BLD) patients. We focused specifically on circulating T cells, given their close interaction with tumors, and initially assessed 93 T-cell features from the flow cytometric analysis. Using a feature selection protocol, we identified five T-cell features in peripheral blood with strong diagnostic relevance. Notably, while individual alterations in these features lacked cancer specificity, simultaneous alterations were uniquely indicative of lung cancer. To comprehensively analyze these features, we developed a scoring model, IMmunoPhenotypic Analysis for Cancer deTection (IMPACT). Comprehensive analysis using the five features (IMPACT-5) demonstrated high cancer specificity and biomarker efficacy, as evidenced by the high area under the receiver operating characteristic curve values for lung cancer patients (0.9187, 0.9277, and 0.9363 for stage I NSCLC, stage IV NSCLC, and SCLC patients, respectively), in stark contrast to BLD patients (0.5212). These findings suggest that comprehensive analysis of cancer-associated changes in circulating T cells can effectively detect lung cancer from its early stages, proposing immunophenotypic analysis of circulating T cells as an innovative liquid biopsy-based diagnostic biomarker.

There are mounting concerns about the adverse effects of social media on the public understanding of the COVID-19 pandemic and its potential effects on vaccination coverage. Yet early studies have focused on generic social media use and been based on cross-sectional data limiting any causal inferences. This study is among the first to provide causal support for the speculation that social media news use leads to vaccine hesitancy among US citizens. This two-wave survey study was conducted in the US using Qualtrics online panel-based recruitment. We employ mediation and moderated mediation analyses to test our assumptions. The results suggest that using social media to consume news content can translate into vaccine hesitancy by increasing citizens' skepticism regarding the efficacy of vaccines. However, these effects are contingent upon the news literacy of users, as the effects on vaccine hesitancy are more substantial among those with lower news literacy. The current study recommends to public policymakers and vaccine communication strategists that any attempt to reduce vaccine hesitancy in society should factor in the adverse effects of social media news use that can increase vaccine safety concerns.

Capicua (CIC) is a transcriptional repressor that regulates several developmental processes. CIC deficiency results in lymphoproliferative autoimmunity accompanied by expansion of CD44hiCD62Llo effector/memory and follicular Th cell populations. Deletion of Cic alleles in hematopoietic stem cells (Vav1-Cre-mediated knockout of Cic) causes more severe autoimmunity than that caused by the knockout of Cic in CD4+CD8+ double positive thymocytes (Cd4-Cre-mediated knockout of Cic). In this study, we compared splenic CD4+ T cell activation and proliferation between whole immune cell-specific Cic-null (Cicf/f;Vav1-Cre) and T cell-specific Cic-null (Cicf/f;Cd4-Cre) mice. Hyperactivation and hyperproliferation of CD4+ T cells were more apparent in Cicf/f;Vav1-Cre mice than in Cicf/f;Cd4-Cre mice. Cicf/f;Vav1-Cre CD4+ T cells more rapidly proliferated and secreted larger amounts of IL-2 upon TCR stimulation than did Cicf/f;Cd4-Cre CD4+ T cells, while the TCR stimulation-induced activation of the TCR signaling cascade and calcium flux were comparable between them. Mixed wild-type and Cicf/f;Vav1-Cre bone marrow chimeras also exhibited more apparent hyperactivation and hyperproliferation of Cic-deficient CD4+ T cells than did mixed wild-type and Cicf/f;Cd4-Cre bone marrow chimeras. Taken together, our data demonstrate that CIC deficiency at the beginning of T cell development endows peripheral CD4+ T cells with enhanced T cell activation and proliferative capability.

This study examines algorithm effects on user opinion, utilizing a real-world recommender algorithm of a highly popular video-sharing platform, YouTube. We experimentally manipulate user search/watch history by our custom programming. A controlled laboratory experiment is then conducted to examine whether exposure to algorithmically recommended content reinforces and polarizes political opinions. Results suggest that political self-reinforcement, as indicated by the political emotion-ideology alignment, and affective polarization are heightened by political videos–selected by the YouTube recommender algorithm–based on participants’ own search preferences. Suggestions for how to reduce algorithm-induced political polarization and implications of algorithmic personalization for democracy are discussed.

While the online sphere is believed to expose individuals to a wider array of viewpoints, a worry about self-reinforcing political echo chambers also persists. We join this scholarly debate by focusing on individual motives for political discussion and dyadic- and structural-level mechanisms that can drive one’s message-selection decision in online discussion settings. Using unobtrusively logged behavioral data matched with panel survey responses, our temporal exponential random graph model (TERGM) analysis indicates that message selection in online discussion settings is largely driven by the similarity of one’s candidate evaluative criteria and various endogenous structural factors, whereas the impact of overt partisan preference in shaping message selection is much more limited than is often assumed.

This study investigates the role of the Internet in civic participation inequality across 108 countries. Merging individual-level survey data from the 2016 Gallup World Poll with country-level indices, we conduct multilevel analyses to answer three broader sets of questions: (1) Does access to the Internet increase the likelihood of civic participation? (2) Does Internet access amplify or lessen socioeconomic stratification in civic participation? (3) Do press freedom and government intervention as contextual factors shape the role of the Internet in civic participation inequality? The findings suggest that Internet access increases the likelihood of civic participation while it also deepens socioeconomic stratification in participation. Cross-level interactions unveil that the intervening role of the Internet remains unaffected by press freedom, but government intervention through the promotion of ICT use can help control the growing inequality. We discuss the theoretical implications of these findings for political inequality research and the applied global significance.

This study attempts to understand how geopolitical proximity influences framing of social conflicts in news coverage and social media discussions. Within the context of 2013 Little India riot in Singapore, a manual content and automated linguistic analyses are conducted on 227 news articles and 4,495 tweets. A multinational comparison suggests that news media follow the traditional hypothesis of geopolitical proximity and international news coverage. However, Twitter seems less constrained by geopolitical boundaries of news making allowing citizens to bypass press censorship in an alternate information system. The reasons for framing differences across mediums and between countries are explored. Implications of these findings and limitations of the study are discussed.

Since its introduction, weight-bearing computed tomography (WBCT) has gained prominence due to its ability to produce accurate three-dimensional images under natural loading conditions, making it particularly useful for assessing complex foot deformities. This review aimed to focus on the diseases of the foot and categorized the pathological conditions into forefoot disease (hallux valgus), midfoot disease (Lisfranc injuries and midfoot osteoarthritis), and progressive collapsing foot deformity. For each category, the authors detail how WBCT enhances diagnostic accuracy and informs treatment strategies. In hallux valgus, WBCT allows for more precise measurement of established parameters and reveals crucial information about metatarsal pronation and ray instability. For midfoot pathologies, WBCT's superiority in detecting subtle Lisfranc injuries and characterizing midfoot osteoarthritis is emphasized, highlighting the development of novel measurement techniques. The review extensively covers the application of WBCT in assessing the complex three-dimensional features of PCFD, including hindfoot valgus, midfoot/forefoot abduction, medial column instability, peritalar subluxation, and valgus tilting, presenting several WBCT-specific measurements and the use of distance mapping to quantify joint surface interaction. The authors conclude that WBCT, potentially enhanced through integration with artificial intelligence (AI), represents a significant advancement in foot and ankle care, promising improved diagnostic accuracy, streamlined treatment planning, and, ultimately, better patient outcomes. The relationship between risk-taking personality and health-risk behaviors has been widely established, where people who like to take risks are more likely to engage in risky sexual behaviors such as having multiple casual partners and having unprotected sex. Drawing on a national U.S. sample from the National Longitudinal Study of Adolescent to Adult Health, the present study examined the relationship between risk-taking personality and sexual experience among adults in early adulthood, and the role of family (parent-child) communication in moderating this relationship. Findings indicated that, for both males and females, the effect of risk taking on sexual experience through alcohol use dissipated at high levels of father-child communication. However, mother-child communication did not have such moderating effects. Implications for the way in which we study parent-child communication are discussed.

Online algorithms have received much blame for polarizing emotions during the 2016 U.S. presidential election. We use transfer entropy to measure directed information flows from human emotions to YouTube’s video recommendation engine, and back, from recommended videos to users’ emotions. We find that algorithmic recommendations communicate a statistically significant amount of positive and negative affect to humans. Joy is prevalent in emotional polarization, while sadness and fear play significant roles in emotional convergence. These findings can help to design more socially responsible algorithms by starting to focus on the emotional content of algorithmic recommendations. Employing a computational-experimental mixed method approach, the study serves as a demonstration of how the mathematical theory of communication can be used both to quantify human-machine communication, and to test hypotheses in the social sciences.

This study analyzes the political use of Twitter in the run-up to the 2013 Malaysian General Election. It follows a content and social network analysis approach to investigate the interplay of language and political partisanship in social media use, among Twitter users in Malaysia. In the period leading up to the 2013 elections, Twitter posts collected under the hashtag #GE13 reveal that communities that post in English versus the Malay language, differ in how they use Twitter and with whom they interact. As compared to English users, Malay users are more likely to seek political information and express their political opinion. In online discussions, we observe language-based homophily within the English and Malay language communities, but there are some cross-cutting interactions between opposing political communities. We discuss the implications of our findings for the political use of new communication technologies in multi-ethnic and multilingual societies.

Over the past decade, various online communication platforms have empowered citizens to express themselves politically. Although the political impact of online citizen expression has drawn considerable attention, research has largely focused on whether and how citizen-generated messages influence the public as an information alternative to traditional news outlets. The present study aims to provide a new perspective on understanding citizen expression by examining its political implications for the expressers themselves rather than those exposed to the expressed ideas. Data from a national survey and an online discussion forum study suggest that expressing oneself about politics provides self-reinforcing feedback. Political expressions on social media and the online forum were found to (a) reinforce the expressers’ partisan thought process and (b) harden their pre-existing political preferences. Implications for the role the Internet plays in democracy will be discussed.

In this study, it is theorized that the communicative affordances offered by social media platforms will enable politically under-resourced candidates to contest the marginalization they face in traditional media. Multivariate analyses were conducted of the tweets of 205 political candidates of the 2014 Indian general election. Findings reveal that fringe party candidates received the least media attention and tended to use Twitter more frequently than major party candidates, especially for interaction and mobilization. Minor party candidates also received less media attention, albeit their Twitter usage patterns were not significantly different than major party candidates. The results illustrate that social media platforms can help overcome resource inequality in politics. The larger implications of this study are discussed.

Simple Summary: This study investigates the optimal timing and effectiveness of radiotherapy in treating oligometastatic prostate cancer (OMPC), which is characterized by limited metastatic spread. We evaluated the impact of early intervention with radiotherapy, targeting both the primary tumor and metastatic sites, on patient survival. Our results show that early radiotherapy to oligometastatic sites, particularly when combined with primary tumor treatment, significantly enhances survival outcomes. These findings underscore the critical importance of timely and personalized treatment strategies in improving the management and care of patients with OMPC. Background/Objectives: Oligometastatic prostate cancer (OMPC) represents an early stage of metastatic disease characterized by a limited number of lesions. Recent advancements in imaging and treatment have revived interest in personalized therapies, including metastasis-directed radiotherapy (OMDRT) and primary prostate radiotherapy (PPR). This study evaluates the impact of OMDRT timing and the role of PPR on survival outcomes in OMPC patients; Methods: In this retrospective cohort study, 82 patients with OMPC who underwent OMDRT between 2010 and 2019 were analyzed. Patients were classified based on OMDRT timing (early vs. late) and disease type (synchronous vs. metachronous). Progression-free survival (PFS) and overall survival (OS) were the primary endpoints, assessed via Kaplan-Meier analysis and Cox proportional hazards models; Results: Among the patients, 36 (43.9%) had synchronous and 46 (56.1%) had metachronous OMD. With a median follow-up of 32 months, the 5-year PFS and OS rates were 77.5% and 88.5%, respectively. Early OMDRT significantly improved PFS (HR 0.461, 95% CI: 0.257–0.826, p = 0.009) and OS (HR 0.219, 95% CI: 0.080–0.603, p = 0.003). Subgroup analysis showed the most favorable outcomes for synchronous OMD patients receiving early OMDRT, with a median PFS of 22.2 months and a 5-year survival rate of 42.1%. The treatment of the primary prostate provided a survival benefit in the OS of synchronous OMD patients (5-year 83.1% vs. 50%, p = 0.025), and there was a further improvement in OS after PPR (5-year 87.7% vs. 50%, p = 0.015). Conclusions: Early OMDRT significantly enhances survival outcomes in OMPC, in both synchronous and metachronous cases. The integration of PPR can further improve results, emphasizing the importance of early intervention and personalized treatment strategies. To more definitively clarify our findings across various clinical situations, further studies with larger cohorts or prospective designs are necessary. [ABSTRACT FROM AUTHOR]

The intersection of consumer culture and civic life has long been a topic of academic discubion. This study revisits the relationship between consumption and civic engagement and investigates the moderators of this relationship. Specifically, we focus on news consumption and opinion leadership as intervening factors that condition the way consumption and civic life are interconnected. Our data reveal that both socially conscious consumption and status-oriented consumption are positively related with civic participation. The positive relationships become stronger when news consumption increases or when one's opinion leadership is strong. Implications for research on consumer culture and civic engagement are discubed.

Myeloid cell leukemia-1 (MCL-1), which belongs to the anti-apoptotic B cell lymphoma-2 family protein, is overexpressed in various cancers and is associated with cell immortality, malignant transformation, chemoresistance, and poor prognosis in humans. However, the significance of MCL-1 in canine mammary gland tumors (MGTs) remains unknown. This study aimed to examine MCL-1 expression in normal canine mammary glands and tumors and to assess its correlation with clinical and histologic variables. In total, 111 samples were examined, including 12 normal mammary gland tissues, 51 benign MGTs, and 48 malignant MGTs. Immunohistochemistry revealed that 53% of benign tumors and 75% of malignant tumors exhibited high MCL-1 expression, whereas only 8% of normal mammary glands exhibited high MCL-1 expression. High MCL-1 expression correlated with tumor malignancy (p < 0.001), large tumor size (> 3 cm) (p = 0.005), high Ki-67 expression (p = 0.046), and metastasis (p = 0.027). Survival curve analysis of dogs with malignant MGTs demonstrated a significant association between high MCL-1 expression and shorter median overall survival (p = 0.027) and progression-free survival (p = 0.014).
