27. Subacute electroacupuncture at Baihui (GV 20) and Dazhui (GV 14) promotes post-stroke functional recovery via neurogenesis and astrogliosis in a photothrombotic stroke mouse model

Abstract

To investigate the optimal timing and underlying mechanism of electroacupuncture (EA) at Baihui (GV 20) and Dazhui (GV 14) for improved long-term functional recovery after focal cerebral ischemia in a photothrombotic stroke mouse model.Totally 50 adult male C57BL/6J mice were assigned into 5 groups: (a) the control group, sham-operated mice (n = 10); (b) the vehicle group, focal cerebral ischemia induction without EA (n = 10); (c) the acute EA group, mice received EA immediately post-ischemia, followed by once-daily treatments for 7 consecutive days (n = 10); (d) the subacute EA group, mice received EA 4 days post-ischemia, followed by once-daily treatments for 7 consecutive days (n = 10); (e) the delayed EA group. EA stimulation (2 Hz, 2 V for 20 min) was applied to acupuncture points (acupoints), Baihui (GV 20) and Dazhui (GV 14), once a day for 7 consecutive days beginning immediately (acute treatment), 4 d (subacute treatment) and 10 d (delayed treatment) after focal cerebral ischemia in C57BL/6J mice. Behavioral assessments were conducted 21 and 28 d post-ischemia and histopathological analyses were performed 28 days post-ischemia.The subacute EA treatment at Baihui (GV 20) and Dazhui (GV 14) significantly improved functional recovery compared to the vehicle group 28 d after ischemic brain injury, although brain atrophy was not reduced. The number of NeuN+ and NeuN+/BrdU+ cells as well as GFAP intensity in the ipsilateral cortex were significantly increased in the subacute group compared to the vehicle group 28 d post-ischemia. We concluded that EA stimulation 4 d post-ischemia (subacute treatment) enhanced neurogenesis and astrogliosis, likely contributing to long-term functional recovery following focal cerebral ischemia.Our findings suggest that the timing of the EA therapy at Baihui (GV 20) and Dazhui (GV 14) determines the therapeutic effects in mice with focal cerebral ischemia induced by photothrombotic occlusion.

45. Electroacupuncture preconditioning reduces cerebral ischemic injury via BDNF and SDF-1α in mice

Abstract

Abstract Background This study was designed to determine if electroacupuncture (EA) preconditioning improves tissue outcome and functional outcome following experimentally induced cerebral ischemia in mice. In addition, we investigated whether the expression of brain-derived neurotrophic factor (BDNF) and stromal cell derived factor-1α (SDF-1α) and infarct volume were related with improvement in neurological and motor function by interventions in this study. Methods After treatment with EA at the acupoints ‘Baihui (GV20)’ and ‘Dazhui (GV14)’ for 20 min, BDNF was assessed in the cortical tissues based on Western blot and the SDF-1α and vascular endothelial growth factor (VEGF) levels in the plasma determined by ELISA. To assess the protective effects of EA against ischemic injury, the mice received once a day 20 min EA preconditioning for three days prior to the ischemic event. Focal cerebral ischemia was then induced by photothrombotic cortical ischemia. Infarct volumes, neurobehavioral deficit and motor deficit were evaluated 24 h after focal cerebral ischemia. Results The expression of BDNF protein increased significantly from 6 h, reaching a plateau at 12 h after the end of EA treatment in the cerebral cortex. Furthermore, SDF-1α, not VEGF, increased singnificantly from 12 h to 48 h after EA stimulation in the plasma. Moreover, EA preconditioning reduced the infarct volume by 43.5% when compared to control mice at 24 h after photothrombotic cortical ischemia. Consistent with a smaller infarct size, EA preconditioning showed prominent improvement of neurological function and motor function such as vestibule-motor function, sensori-motor function and asymmetric forelimb use. The expression of BDNF colocalized within neurons and SDF-1α colocalized within the cerebral vascular endothelium was observed throughout the ischemic cortex by EA. Conclusions Pretreatment with EA increased the production of BDNF and SDF-1α, which elicited protective effects against focal cerebral ischemia. These results suggest a novel mechanism of EA pretreatment-induced tolerance against cerebral ischemic injury.

Abstract

We propose a new frame rate up-conversion algorithm for high quality video. In the proposed scheme, bi-directional motion estimation (ME) is performed to construct the motion vector (MV) field for the frame to be interpolated. Unlike conventional motion-compensated interpolation (MCI) algorithms, the proposed technique does not produce any overlapped pixel and hole region in the interpolated frame, and thus can utilize the overlapped block motion compensation technique to reduce the blocking artifacts. The proposed algoritm is very simple to implement on consumer products when compared to conventional MCI methods. Computer simulation shows a high visual performance of the proposed frame rate up-conversion algorithm. 47. An effective de-interlacing technique using motion compensated interpolation

Abstract

In this paper, we propose a new de-interlacing algorithm using motion compensated interpolation. In the proposed scheme, motion estimation is first performed between the same parity fields, i.e., the previous and the next field, and then the motion vector is refined in the interpolated field. In conventional motion compensated schemes, a pre-filter such as line averaging is applied to interpolate missing lines before the motion estimation between the opposite parity fields. The proposed scheme does not require the pre-filter because block matching is performed between the same parity fields. For further improvement, a five-point median filter with temporal emphasis is used to reduce the interpolation error caused by incorrect motion. Computer simulation results indicate that the proposed scheme provides a better visual performance than conventional deinterlacing algorithms.
