Abstract

International audience; Production of in planta haploid embryos that inherit chromosomes from only one parent can greatly increase breeding efficiency via quickly generating homozygous plants, called doubled haploid. One of the main players of in planta haploid induction is a pollen-specific phospholipase A, which is able, when mutated, to induce in vivo haploid induction in numerous monocots. However, no functional orthologous gene has been identified in dicots plants. Here, we show that loss-of-function of gynoecium-expressed phospholipase AII (pPLAIIγ) triggers maternal haploid plants in Arabidopsis, at an average rate of 1.07%. Reciprocal crosses demonstrate that haploid plants are triggered from the female side and not from the pollen, and the haploid plants carry the maternal genome. Promoter activity of pPLAIIγ shows enriched expression in the funiculus of flower development stages 13 and 18, and pPLAIIγ fused to yellow fluorescent protein reveals a plasma-membrane localization Interestingly, the polar localized PIN1 at the basal plasma membrane of the funiculus was all internalized in pplaIIγ mutants, suggesting that altered PIN1 localization in female organ could play a role in maternal haploid induction.

Abstract

Pollen-specific phospholipase A-mediated in vivo haploid induction system was functionally well conserved in numerous monocots. However, no functional orthologous gene has been identified in dicots plants. Here, we show that loss-of-function of gynoecium-expressed phospholipase AII (pPLAIIγ) triggers haploid plants in Arabidopsis, at an average rate of 1.46%. Reciprocal crosses demonstrate that haploid plants are triggered from the female side and not from the pollen, and the haploid plants carry the maternal genome.

Abstract

International audience; Petrophysical properties of volcanic rocks are controlled by lithology and subsequent modification by alteration processes. Investigating the linkages, using a range of different techniques, are important to establish how petrophysical properties can inform about the alteration state of volcanic rocks. Here, we compile petrophysical data and correlate these with geochemical and mineralogical analyses acquired from a volcanic sequence on the Naturaliste Plateau, offshore southwest Australia (IODP Site U1513). The sequence consists of alternating basalt lava flows and volcaniclastic deposits, intruded by multiple dolerite dikes. Variable alteration intensities from fresh-slight to strong are quantified using Chemical Index of Alteration. Intervals of slightly altered dikes exhibit low porosity and high values of bulk density, P-wave velocity and thermal conductivity. The increase of alteration intensity corresponds to decreases in bulk density to ∼2 g/cm3, P-wave velocity to ∼2000 m/s, thermal conductivity to ∼1.2 W/(m·K) and an increase in porosity up to 50%. Natural Gamma Ray and magnetic susceptibility vary downhole with rock composition and at lithologic boundaries. The distinct variations exhibit a good correlation with primary lithologic characteristics and secondary mineralogical and textural changes attributed to alteration processes. We provide synthesis models of petrophysical variation with alteration intensity. Although differences in primary lithology and alteration type introduce limitations and uncertainties, there is a reasonable applicability of our results to rapidly characterize the alteration intensity and volcanic stratigraphy in volcanic sequences and to calibrate wireline log-based determinations. This will help others to develop strategies for exploration, drilling, and geophysical research of volcanic rocks.

Abstract

International audience; High temperatures during the crop reproductive stage impact seed yield and quality. The changing climate will require consideration of the effects of high temperature events that differ from their intensity, their duration and their frequency over the seed quality-building stages. The impact of these features deserve to be investigated at the light of induced thermo-sensitization which can lead to alleviate expected negative impacts. In our work, maturing seeds of the sulphur-demanding crop, oilseed rape, were exposed to several temperature sequences that varied in intensity, duration and frequency at the onset of seed maturation. Results-measured in seeds that were at the onset of maturation when the temperature stress occurred-indicated that (i) the longer the cumulated duration of the temperature stress, the more negatively impacted the quality criteria with decreased fatty acids (FAs) concentration, increased omega 6:omega 3 ratio, lower seed membrane integrity and increased seed dormancy and (ii) a mild stress event prior to heat peaks had an alleviating effect on the negative impact of the later heat peaks (priming effect) on seed nitrogen, desiccation tolerance and the phytohormones involved in thermoinhibition. Sulphur restriction was positive on FAs, protein concentration and negative on breaking dormancy. In addition, sulphur supply interfered with temperature modality, features such that positive impact of sulphur limitation on boosting oxidative response were cancelled with intense late heat peaks. This work provides insights to define thermopriming protocols in relation to the timing of quality building processes, their respective optimal temperature and adequate sulphur supply.

Abstract

Objectives This study sought to establish worldwide and regional diagnostic reference levels (DRLs) and achievable administered activities (AAAs) for single-photon emission computed tomography (SPECT) myocardial perfusion imaging (MPI). Background Reference levels serve as radiation dose benchmarks to compare individual laboratories against aggregated data, helping to identify sites in greatest need of dose reduction interventions. DRLs for SPECT MPI have previously been derived from national or regional registries. To date there have been no multiregional reports of DRLs for SPECT MPI from a single standardized dataset. Methods Data were submitted voluntarily to the INCAPS (International Atomic Energy Agency Nuclear Cardiology Protocols Study), a cross-sectional, multinational registry of MPI protocols. A total of 7,103 studies were included. DRLs and AAAs were calculated by protocol for each world region and for aggregated worldwide data. Results The aggregated worldwide DRLs for rest-stress or stress-rest studies employing technetium Tc 99m–labeled radiopharmaceuticals were 11.2 mCi (first dose) and 32.0 mCi (second dose) for 1-day protocols, and 23.0 mCi (first dose) and 24.0 mCi (second dose) for multiday protocols. Corresponding AAAs were 10.1 mCi (first dose) and 28.0 mCi (second dose) for 1-day protocols, and 17.8 mCi (first dose) and 18.7 mCi (second dose) for multiday protocols. For stress-only technetium Tc 99m studies, the worldwide DRL and AAA were 18.0 mCi and 12.5 mCi, respectively. Stress-first imaging was used in 26% to 92% of regional studies except in North America where it was used in just 7% of cases. Significant differences in DRLs and AAAs were observed between regions. Conclusions This study reports reference levels for SPECT MPI for each major world region from one of the largest international registries of clinical MPI studies. Regional DRLs may be useful in establishing or revising guidelines or simply comparing individual laboratory protocols to regional trends. Organizations should continue to focus on establishing standardized reporting methods to improve the validity and comparability of regional DRLs.

Abstract

BackgroundElevated systolic blood pressure (SBP) after mechanical thrombectomy (MT) correlates with worse outcome. However, the association between SBP reduction (SBPr) and outcome after successful reperfusion with MT is not well established.ObjectiveTo investigate the association between SBPr in the first 24 hours after successful reperfusion and the functional and safety outcomes of MT.MethodsA multicenter retrospective study, which included 10 comprehensive stroke centers, was carried out. Patients with acute ischemic stroke and anterior circulation large vessel occlusions who achieved successful reperfusion via MT were included. SBPr was calculated using the formula 100×([admission SBP−mean SBP]/admission SBP). Poor outcome was defined as a modified Rankin Scale (mRS) score of 3–6 at 90 days. Safety endpoints included symptomatic intracerebral hemorrhage, mortality, and requirement for hemicraniectomy during admission. A generalized mixed linear model was used to study the association between SBPr and outcomes.ResultsA total of 1361 patients were included in the final analysis. SBPr as a continuous variable was inversely associated with poor outcome (OR=0.97; 95% CI 0.95 to 0.98; p20%), the rate of poor outcome was highest in the first group.ConclusionSBPr in the first 24 hours after successful reperfusion was inversely associated with poor outcome. No association between SBPr and safety outcome was found.

Abstract

Dinoflagellates are a major component of the modern plankton. Of the 2192 species of marine free-living dinoflagellates presently described, an increasing number are being shown to produce resting cysts (probably hypnozygotes) within their life cycle. With rare exception, only the resting cysts fossilize, so they are of central importance in tracing the history of dinoflagellates through geological time. Cysts of many of the more common dinoflagellate species have distinctive morphologies allowing their geographic and stratigraphic occurrences to be traced. An ever-increasing number of taxa are also being shown to produce distinctive and geologically preservable cysts, potentially enhancing our knowledge of the diverse representation of dinoflagellates through time. Here the organic-walled cysts of 73 rare, poorly known or morphologically problematic marine dinoflagellate cyst species belonging to the orders Gymnodiniales (nine species) and Peridiniales (64 species) are reviewed, described and illustrated, and their stratigraphic ranges assessed. The names Echinidinium aculeatum and Echinidinium transparantum are validated herein. © 2019 Elsevier B.V.

Abstract

The relationships between subjective status and perceived legitimacy are important for understanding the extent to which people with low status are complicit in their oppression. We use novel data from 66 samples and 30 countries (N = 12,788) and find that people with higher status see the social system as more legitimate than those with lower status, but there is variation across people and countries. The association between subjective status and perceived legitimacy was never negative at any levels of eight moderator variables, although the positive association was sometimes reduced. Although not always consistent with hypotheses, group identification, selfesteem, and beliefs in social mobility were all associated with perceived legitimacy among people who have low subjective status. Abstract

International audience; The Naturaliste Plateau is a submarine continental ribbon rifted from the southwest Australian margin during the Early Cretaceous breakup of East Gondwana. It occupied a key position near the juncture of Greater India and the boundary between Australia and Antarctica. However, details of the Early Cretaceous evolution of the plateau are not well known because of limited data. Drilling at Site U1513 during IODP Expedition 369 recovered the first complete Lower Cretaceous succession on the eastern Naturaliste Plateau. The succession includes syn-rift volcanic rocks, Hauterivian to early Aptian volcaniclastic-rich sedimentary rocks, and Albian claystone strata. The 235-m thick volcaniclastic-rich sequence represents the missing post-breakup record in the southwest Australian rifted margin. It spans the transition from syn- to post-rift phase during the final stages of breakup between Greater India and Australia-Antarctica. We report the lithological, petrophysical, geochemical, paleontological, and paleomagnetic characteristics of the sequence, and then synthesize the results to define the Early Cretaceous depositional environment and subsidence history of the Naturaliste Plateau. From the early Hauterivian, weathered volcanic products were eroded and re-deposited locally as a volcaniclastic-rich sequence, with a major contribution from the southern Naturaliste Plateau. The depositional environment evolved from a shelf to upper bathyal condition during the Hauterivian through early Barremian with a decreasing sedimentation rate. This period is defined as a late syn-rift subsidence phase by NW-SE trending extension. After the final breakup with Greater India, the plateau remained at upper bathyal depths with little deposition until the early Aptian. Mid–lower bathyal depths inferred from the Albian claystone strata suggest that the post-rift thermal subsidence commenced during the late Aptian. This two-phase post-rift subsidence reflects the proximity or high temperature of mantle plume, possibly the Kerguelen plume, and its westward migration relative to the southwest Australian rifted margin.

Abstract

Background The objective of this study was to assess regional and ethnic differences in an unselected patient population treated with polymer-free sirolimus-eluting stents (PF-SES) in Asia and Europe. Methods Two all-comers observational studies based on the same protocol (ClinicalTrials.gov Identifiers: NCT02629575 and NCT02905214) were combined for data analysis to assure sufficient statistical power. The primary endpoint was the accumulated target lesion revascularization (TLR) rate at 9-12 months. Results Of the total population of 7243 patients, 44.0% (3186) were recruited in the Mediterranean region and 32.0% (2317) in central Europe. The most prominent Asian region was South Korea (17.6%, 1274) followed by Malaysia (5.7%, 413). Major cardiovascular risk factors varied significantly across regions. The overall rates for accumulated TLR and MACE were low with 2.2% (140/6374) and 4.4% (279/6374), respectively. In ACS patients, there were no differences in terms of MACE, TLR, MI and accumulated mortality between the investigated regions. Moreover, dual antiplatelet therapy (DAPT) regimens were substantially longer in Asian countries even in patients with stable coronary artery disease as compared to those in Europe. Conclusions PF-SES angioplasty is associated with low clinical event rates in all regions. Further reductions in clinical event rates seem to be associated with longer DAPT regimens.

Abstract

International audience; Background and Purpose— Successful reperfusion can be achieved in more than two-thirds of patients treated with mechanical thrombectomy. Therefore, it is important to understand the effect of blood pressure (BP) on clinical outcomes after successful reperfusion. In this study, we investigated the relationship between BP on admission and during the first 24 hours after successful reperfusion with clinical outcomes. Methods— This was a multicenter study from 10 comprehensive stroke centers. To ensure homogeneity of the studied cohort, we included only patients with anterior circulation who achieved successful recanalization at the end of procedure. Clinical outcomes included 90-day modified Rankin Scale, symptomatic intracerebral hemorrhage (sICH), mortality, and hemicraniectomy. Results— A total of 1245 patients were included in the study. Mean age was 69±14 years, and 51% of patients were female. Forty-nine percent of patients had good functional outcome at 90-days, and 4.7% suffered sICH. Admission systolic BP (SBP), mean SBP, maximum SBP, SBP SD, and SBP range were associated with higher risk of sICH. In addition, patients in the higher mean SBP groups had higher rates of sICH. Similar results were found for hemicraniectomy. With respect to functional outcome, mean SBP, maximum SBP, and SBP range were inversely associated with the good outcome (modified Rankin Scale score, 0–2). However, the difference in SBP parameters between the poor and good outcome groups was modest. Conclusions— Higher BP within the first 24 hours after successful mechanical thrombectomy was associated with a higher likelihood of sICH, mortality, and requiring hemicraniectomy.
