The objective of the research is to create a rapid, uncomplicated, accurate, and cost-effective RPHPLC technique for quantifying the quantities of Grazoprevir and Elbasvir concurrently in both pharmaceutical products and bulk materials. This approach has successfully accomplished the separation of Grazoprevir and Elbasvir in significant quantities. The separation was conducted with a Waters C18 (250 x 4.6 mm x 5 μ particle size) analytical column, with detection at a wavelength of 260 nm. The mobile phase included a blend of Methanol and Phosphate Buffer pH 4.0 at a volumetric ratio of 60:40. The separation was conducted using isocratic elution mode at a flow rate of 1.0 ml/min. Grazoprevir had a retention time of 2.400 minutes, whereas Elbasvir showed a retention length of 3.016 minutes. The quantification of Grazoprevir and Elbasvir was performed by utilizing PDA detection at a wavelength of 260 nm, employing a linear calibration curve. For accurate quantification, the concentration ranges of 20-100 μg/ml (with a correlation value of 0.9999) and 10-50 μg/ml (with a correlation coefficient of 0.999) were used. The suggested approach is well-suited for use in quality-control labs for the quantitative examination of medicines, both when employed separately or in combination, particularly in bulk quantities. An innovative, cost-effective reverse phase high performance liquid chromatography (RPHPLC) approach has been devised to accurately and precisely determine the quantity of Nelaribine in both bulk and sterile dosage forms. The separation process was conducted using an Inertsil C18-ODS 3V column with dimensions of 250×4.6 mm and a particle size of 5 µm. Isocratic mode was employed, with a mobile phase consisting of a mixture of acetic acid pH-4.0 buffer and methanol in a ratio of 75:25 (v/v). The mobile phase was pumped into the column at a flow rate of 1.5 mL min-1. The eluent from the column was detected using a UV detector set at 260 nm. The whole duration of the experiment was 8 minutes, during which the column was kept at the ambient temperature. The observed retention time for Nelaribine was determined to be 3.99 minutes. The linearity of the standard curves was seen within the concentration range of 20-100 µg/ml, exhibiting a high coefficient of determination (R2 = 0.9998). The results of the experiment indicated a percentage recovery ranging from 100.0272% to 101.8%. Additionally, the relative standard deviation (RSD) was determined to be 0.3693%. The measured percentage content of a commercially available Nelaribine formulation was determined to be 100.20 %. The methodology was verified in accordance with the International Council for Harmonisation of Technical Requirements for Pharmaceuticals for Human Use (ICH) recommendations. The suggested RP-HPLC technique has been validated by investigations, which have shown that it has characteristics of simplicity, specificity, rapidity, reliability, and reproducibility. The ketogenic diet is gaining popularity among the scientific community for minimizing complications of elevated postprandial glucose and obesity. Caloric restrictions have proven their potential for minimizing oxidative damage. Short-term usage of the Keto diet has shown promising results; but in long term, there found many complications such as increased postprandial glucose, glucose intolerance, hepatic steatosis, and systemic inflammation as well. Carbohydrate and lipid; are both energy sources that are of utmost importance and inseparable in metabolism. Lipids produce significantly high ATPs from their equivalent amount of carbohydrates, but at the same time make cells more vulnerable as they are the easiest target of ROS. Thus, it creates confusion about whether to go for the keto diet or not? When, we compare normal diet and ketogenic diet with respect to the variability of carbohydrate component and how much it costs in terms of peroxidative damage, it has been observed that low sugars gave more protection while their capacity can be synergically boosted when used with antioxidants like ascorbic acid and alpha-tocopherol. The diterpene steviol glycoside, rebaudioside A, is a natural high potency non-caloric sweetener extracted from the leaves of Stevia rebaudiana. This compound shows a parabolic change in sweet taste intensity with temperature which contrasts with the general finding for other synthetic or natural sweeteners whose sweet taste increases with temperature. The nonmonotonic taste behavior was determined by sensory analysis using large taste panels. The conformational landscape of rebaudioside A was established at a range of temperatures by means of nuclear magnetic resonance and molecular dynamics simulation.
