2. An up-scaled biotechnological approach for phosphorus-depleted rye bran as animal feed

Abstract Side streams from the milling industry offer excellent nutritional properties for animal feed; yet their use is constrained by the elevated phosphorus (P) content, mainly in the form of phytate. Biotechnological P recovery fosters sustainable P management, transforming these streams into P-depleted animal feed through enzymatic hydrolysis. The enzymatic P mobilization not only enables P recovery from milling by-products but also supports the valorization of these streams into P-depleted animal feeds. Our study presents the scalability and applicability of the process and characterizes the resulting P-depleted rye bran as animal feed component. Batch mode investigations were conducted to mobilize P from 100 g to 37.1 kg of rye bran using bioreactors up to 400 L. P reductions of 89% to 92% (reducing from 12.7 gP/kg to 1.41–1.28 gP/kg) were achieved. In addition, High Performance Ion Chromatography (HPIC) analysis showed complete depletion of phytate. The successful recovery of the enzymatically mobilized P from the process wastewater by precipitation as struvite and calcium hydrogen phosphate is presented as well, achieving up to 99% removal efficiency. Our study demonstrates a versatile process that is easily adaptable, allowing for a seamless implementation on a larger scale. Graphical Abstract

3. Dimension reduction and outlier detection of 3-D shapes derived from multi-organ CT images

Abstract Background Unsupervised clustering and outlier detection are important in medical research to understand the distributional composition of a collective of patients. A number of clustering methods exist, also for high-dimensional data after dimension reduction. Clustering and outlier detection may, however, become less robust or contradictory if multiple high-dimensional data sets per patient exist. Such a scenario is given when the focus is on 3-D data of multiple organs per patient, and a high-dimensional feature matrix per organ is extracted. Methods We use principal component analysis (PCA), t-distributed stochastic neighbor embedding (t-SNE) and multiple co-inertia analysis (MCIA) combined with bagplots to study the distribution of multi-organ 3-D data taken by computed tomography scans. After point-set registration of multiple organs from two public data sets, multiple hundred shape features are extracted per organ. While PCA and t-SNE can only be applied to each organ individually, MCIA can project the data of all organs into the same low-dimensional space. Results MCIA is the only approach, here, with which data of all organs can be projected into the same low-dimensional space. We studied how frequently (i.e., by how many organs) a patient was classified to belong to the inner or outer 50% of the population, or as an outlier. Outliers could only be detected with MCIA and PCA. MCIA and t-SNE were more robust in judging the distributional location of a patient in contrast to PCA. Conclusions MCIA is more appropriate and robust in judging the distributional location of a patient in the case of multiple high-dimensional data sets per patient. It is still recommendable to apply PCA or t-SNE in parallel to MCIA to study the location of individual organs.

4. Dietary L-carnitine supplementation modifies blood parameters of mid-lactating dairy cows during standardized lipopolysaccharide-induced inflammation

L-carnitine, available as feed additive, is essential for the beta-oxidation of free fatty acids in the mitochondrial matrix. It provides energy to immune cells and may positively impact the functionality of leukocytes during the acute phase response, a situation of high energy demand. To test this hypothesis, German Holstein cows were assigned to a control group (CON, n = 26) and an L-carnitine supplemented group (CAR, n = 27, rumen-protected L-carnitine product: 125 g/cow/d, corresponded to total L-carnitine intake: 25 g/cow/d, supplied with concentrate) and received an intravenous bolus injection of lipopolysaccharides (LPS, 0.5 µg/kg body weight, E. coli) on day 111 postpartum as a model of standardized systemic inflammation. Blood samples were collected from day 1 ante injectionem until day 14 post injectionem (pi), with frequent sampling through an indwelling venous catheter from 0.5 h pi to 12 h pi. All parameters of the white blood cell count responded significantly to LPS, while only a few parameters were affected by L-carnitine supplementation. The mean eosinophil count, as well as the percentage of basophils were significantly higher in CAR than in CON over time, which may be due to an increased membrane stability. However, phagocytosis and production of reactive oxygen species by leukocytes remained unchanged following L-carnitine supplementation. In conclusion, although supplementation with 25 g L-carnitine per cow and day resulted in increased proportions of specific leukocyte populations, it had only minor effects on the functional parameters studied in mid-lactating dairy cows during LPS-induced inflammation, and there was no evidence of direct improvement of immune functionality.

5. Assessing concordance between Campylobacter prevalence in broilers and human cases before and during the COVID-19 pandemic in Lower Saxony, Germany, considering fresh chicken meat consumption patterns

As the most common foodborne disease, number of campylobacteriosis decreased in Germany with the beginning of the COVID-19 pandemic in 2020. As the consumption of fresh chicken meat is a major risk factor for human infection, this study investigated the relationship between Campylobacter contamination levels on chicken carcasses and human cases in Lower Saxony, Germany and observed fresh chicken meat consumption patterns between 2018 and 2021 including the time of the COVID-19 pandemic. Campylobacter levels in broilers and human cases were classified based on the median and descriptively analysed per week using contingency tables. Before the COVID-19 pandemic (2018 and 2019), high Campylobacter contamination levels on neck samples and many human cases were more present, whereas with the beginning of the COVID-19 pandemic (2020 and 2021), low contamination levels on chicken carcasses and few human cases were more present. Lowest concordance between both parameters was shown in 2018 (Cohen’s cappa coefficient: 0.37) and 2020 (0.38). The highest concordance was examined in 2021 (0.69). The private consumption of fresh chicken meat in Lower Saxony increased significantly with the beginning of the COVID-19 pandemic in 2020 by 63.9 tonnes compared to 2019 to an average of 453.5 tonnes per week. Public health measures and a reduced number of medical treatments have undoubtedly had an impact on less reported human cases during the COVID-19 pandemic. However, number of human cases remained at a low level in Germany in 2023 while chicken meat consumption increased. Thus, further risk assessments regarding the risk of campyloabcteriosis due to chicken meat consumption should include the country of origin, as the level of contamination of chicken carcasses varies between European countries.

6. Analysis of the Broiler Chicken Dead-on-Arrival (DOA) Rate in Relation to Normal Transport Conditions in Practice in Germany

In total, around 631 million broilers were slaughtered in Germany in 2022. This evaluation included data of approx. 198 million broilers of different ages and breeds that were transported in Germany in 2022 (31% of all cases of broiler chicken transport in 2022). The aim of this study was to analyze German broiler chicken transport (n = 14,054) to the slaughterhouse between January 2022 and May 2023 with regard to the dead-on-arrival (DOA) rate and the possible influencing factors. Therefore, the relation between the total amount of animals per transport, the duration and distance of the transport, the planned stocking density in the transport cages, the average daily temperature and time of day and season of the transport as well as the DOA rate were statistically evaluated. The results showed a mean DOA rate of 0.09% (SD 0.09). Transport conducted at midday (11:00 to 17:00) showed higher DOA rates (p < 0.05) than transport at other times (day split into 6 h intervals). The highest mean DOA rate (0.10%) was found in the fall, followed by the winter, while transport in the spring and summer resulted in the lowest DOA rate (p < 0.05). All in all, the relatively low DOA rate (%) in Germany indicates the good standard of their broiler transport compared to available data from research in other European countries.

7. Nutrient Composition and Feed Hygiene of Alfalfa, Comparison of Feed Intake and Selected Metabolic Parameters in Horses Fed Alfalfa Haylage, Alfalfa Hay or Meadow Hay

The aim of this study was to examine the nutrient composition and feed hygiene of alfalfa as well as investigate the feed intake, blood, urine and faecal parameters of horses fed alfalfa haylage (AS) compared with alfalfa hay (AH) and meadow hay (MH). A total of 11 geldings were fed ad libitum (2.1% dry matter (DM) of body weight (BW)) with alfalfa haylage, alfalfa hay and meadow hay (MH) in a Latin square design. On days 0 and 21 of the feeding period, blood samples were analysed for kidney and liver parameters. Faecal samples were analysed for pH, DM and short-chain fatty acids (SCFAs). Spontaneous urine was collected during the feeding period to analyse selected parameters. Forage was analysed in terms of feed hygiene and crude nutrients. In several feed samples of AS, AH and MH, the microbial reference ranges were exceeded for product-typical and spoilage-indicating bacteria and fungi. Crude nutrient analyses revealed a median crude protein content of 139 (138/142) g/kg DM for AS, which was similar to that in AH (127–135 g/kg DM) and substantially higher than in MH (79.1–87.7 g/kg DM). The calcium level in AS (11.3 g/kg DM) was significantly higher than that in MH (4.00–4.95 g/kg DM) but not compared with that in AH (9.80–10.4 g/kg DM). All blood parameters were within the reference ranges. Fractional excretion (FE)Ca for AS-fed horses ranged from 8.13 to 22.0%; the FECa for AH-fed horses ranged from 6.48 to 24.8%; the FECa for MH-fed horses ranged from 6.69 to 53.2%. No significant differences were found in faecal pH or SCFA content in AS-fed horses compared with AH-fed and MH-fed horses. We concluded that alfalfa haylage provides an alternative forage for equine nutrition.

8. Biotechnological production of food-grade polyphosphate from deoiled seeds and bran

Agricultural products, have a high phytate content that has a hidden potential as a renewable source of phosphate. We present the first biotechnological route for the production of food-grade, organic polyphosphate (polyP) from deoiled seeds or bran, included in a vision for a circular phosphorus economy. The three-step production process includes phosphorus mobilization (e.g., 37 mg PO43−/g bran) using phytase enzymes. Non-genetically modified phosphate starved Saccharomyces cerevisiae is then fed with soluble P-extracts. The yeast intracellularly polymerizes the phosphate to polyP (≥ 30% mol polyP in yeast per mol PO43− in medium) and polyP-rich yeast extract or pure polyP is purified from the biomass. We demonstrate that the obtained polyP-rich yeast extract is an excellent green surrogate for polyP from fossil P-sources in meat manufacturing. The valorization of phytate–P while producing P-depleted biomass as a demanded feed in livestock production is shown. Our sustainable process enables the production of food-grade polyP from renewable resources and is contributing to the sustainable management of the dwindling nutrient phosphorus.

9. Survival persistence of the 3 common Salmonella enterica serotypes isolated from broilers' in different matrices

ABSTRACT: Broiler meat is the predominant source of Salmonella as a foodborne pathogen. Several control strategies have focused on the reduction of Salmonella spp. levels at different production stages. However, the persistence of Salmonella between consecutive flocks is still of great concern. This study was designed to understand the cause of reinfection in broiler flocks due to survival of Salmonella in feeding lines of related matrices. Salmonella (S.) Enteritidis, S. Infantis, and S. Typhimurium isolated from broiler farms in North-West Germany were used. Four types of matrices (phosphate buffer saline (PBS), dietary plant fat, fat with feed mixture, and feed) were applied to evaluate Salmonella survival (with the initial dose about 8.0 log10 CFU/mL) during a simulation of 4 production cycles. To evaluate the growth and survival status of Salmonella ISO 6579-1:2017 were performed (quantitatively by plate count method (PCM) and most probable number method (MPN)) and qualitatively) at 5 defined time points (−7, 0, 4, 7, and 35 d). In all matrices and for the 3 serovars, the Salmonella count decreased at the end of the fourth cycle in comparison to the beginning of the experimental infection, and was still cultivated except for fat matrix. The PBS matrices showed the highest survival level of Salmonella and did not decline drastically by the end of the fourth cycle (5.93 ± 0.00, 5.87 ± 0.02, 5.73 ± 0.05 log10 CFU/mL, respectively). However, the fat matrices showed the lowest survival level for the 3 isolates at d 35 since the first cycle (0 log10 CFU/mL using PCM). Regarding the fat-feed mixture, and feed matrices, there was a fluctuation in the survival rate of Salmonella (all serovars) within each cycle. For the qualitative method, the 3 serovars persisted in all matrices until the end of the fourth cycle except for fat matrices. The present study highlights the ability of Salmonella to survive for a long time in different temperatures and matrices despite efficient cleaning and disinfection processes in the feeding lines, which may influence reinfection with Salmonella in poultry houses.

From a cost and sustainability perspective, the use of by-products such as rye bran in sow diets is of particular interest. Rye bran has valuable ingredients that have potential benefits for the gut health of sows. The aim of this study was to investigate the effects of including 15% rye bran in the sows’ feed on the performance of sows and piglets. The feeding started one week before the farrowing date and ended at weaning. Performance was evaluated by measuring sow (n = 175) and piglet body weight (n = 1372) and sows’ backfat thickness (n = 80). These data were additionally used to calculate the colostrum intake of the suckling piglets and the sows’ milk production. 11. Process Hygiene Criterion for Campylobacter and Number of Campylobacter Enteritis Cases in Northwest Germany

Campylobacteriosis is the most commonly reported bacterial foodborne disease in the European Union. Its transmission is often associated with the consumption of poultry meat. In 2018, Regulation (EC) No. 2017/1495 introduced a process hygiene criterion and with this, the testing requirements for Campylobacter. The results of microbiological testing for Campylobacter of chicken carcass neck skin samples from several slaughter lines in Northwest Germany collected by the food business operators and contamination levels (cfu/g Campylobacter) of these samples were analysed from 2018 to 2021. Classification into three different categories was made based on contamination levels. The proportion of highly contaminated (category three) neck samples (>1000 cfu/g) decreased from 2018 to 2021. Our analysis showed a relationship between the number of neck samples with high Campylobacter contamination levels (>1000 cfu/g) and human cases in Northwest Germany. Spearman’s rank test (p < 0.01) showed a higher correlation in 2018 (0.66) and 2019 (0.58) compared to 2020 and 2021. Campylobacter enteritis cases in Northwest Germany stayed at a low level in 2020 and 2021. It remains unclear whether the decrease in reported Campylobacter enteritis cases is related to a decrease in Campylobacter levels on chicken carcasses or due to other reasons like underreporting during the COVID-19 pandemic, and therefore must be investigated in further analyses.

Abstract Campylobacter is one of the leading bacterial foodborne pathogens worldwide. Poultry is the host species with this pathogen with the highest clinical impact. Flocks become colonised with Campylobacter, which leads to contamination of product entering the food-chain. Rapid and reliable Campylobacter detection methods could support controls to minimize the risks of contamination within the food-chain, which would easier enable the implementation of a logistical slaughter schedule or other control options. The present study evaluates current and emerging C. jejuni detection technologies on air samples in a unique study set-up of pre-defined C. jejuni prevalences. Both non-invasive detection technologies on air samples by subsequent measuring of volatile organic compounds (VOCs) or by qPCR detected the C. jejuni presence and could additionally distinguish between the number of present C. jejuni-positive birds in the study set-up. Nevertheless, electrostatic air samplers diagnosed fewer birds as C. jejuni-positive compared to the cultivation-based method. By measuring the VOCs, it was possible to detect the presence of two positive birds in the room. This apparent high sensitivity still needs to be verified in field studies. Techniques, such as these promising methods, that can facilitate C. jejuni surveillance in poultry flocks are desirable to reduce the risk of infection for humans.

13. Effect of replacing whole wheat with broken rye as a sustainable grain in diets of fattening turkeys on growth performance, litter quality, and foot pad health

IntroductionRye is one of the most important cereal crops in Central Europe, thus attempts have been made to include it in the diets of birds to reduce production costs, since the cost of feed accounts for as much as 50 %−70 % thereof. Nevertheless, the use of rye has been limited to date, particularly in turkeys. This study aimed to test the effects of rye inclusion up to 10 % on growth, excreta, and/or litter dry matter, and foot pad health.MethodsFour trials were performed with a total of 4,322, 4,307, 4,256, and 4,280 female turkeys (BUT BIG 6, Aviagen) for trials 1, 2, 3, and 4, respectively. All birds were fed commercial starter diets for the dietary phases 1 and 2 (up to d 35 of life). Thereafter, at the start of the study, the control group received commercial supplementary feed with 5 % or 10 % wheat until the end of the fattening period. The experimental group was offered supplementary feed to which instead of wheat increasing levels of rye were added stepwise from 5 % to 10 %.ResultsUsing supplementary feed with rye showed no significant differences in the final body weight between the control and experimental groups (10.9 vs. 10.8 kg). The dry matter content of fresh excreta for turkeys during the experimental period did not show significant differences between both groups, except at weeks 10 and 14 of life. The feed type (either control diet or experimental diet) did not significantly affect litter dry matter content between the groups throughout the experimental period. No significant differences were noted in food pad dermatitis scoring between both groups throughout the experimental period, except at weeks 11 and 16 of life. Overall, this study showed that including proportions of rye up to 10% could replace conventional ingredients and may increase sustainability in poultry production regardless of the addition of supplementary feed.

The primary contaminants in poultry are Salmonella enterica, Campylobacter jejuni, Escherichia coli, and Staphylococcus aureus. Their pathogenicity together with the widespread of these bacteria, contributes to many economic losses and poses a threat to public health. With the increasing prevalence of bacterial pathogens being resistant to most conventional antibiotics, scientists have rekindled interest in using bacteriophages as antimicrobial agents. Bacteriophage treatments have also been investigated as an alternative to antibiotics in the poultry industry. Bacteriophages’ high specificity may allow them only to target a specific bacterial pathogen in the infected animal. However, a tailor-made sophisticated cocktail of different bacteriophages could broaden their antibacterial activity in typical situations with multiple clinical strains infections. Bacteriophages may not only be used in terms of reducing bacterial contamination in animals but also, under industrial conditions, they can be used as safe disinfectants to reduce contamination on food-contact surfaces or poultry carcasses. Nevertheless, bacteriophage therapies have not been developed sufficiently for widespread use. Problems with resistance, safety, specificity, and long-term stability must be addressed in particular. This review highlights the benefits, challenges, and current limitations of bacteriophage applications in the poultry industry.

Lawsonia (L.) intracellularis is a widespread, economically important bacterium causing the porcine proliferative enteropathy (PPE). In this study, we evaluated intestinal microbiota of naturally exposed L. intracellularis-positive pigs under standardized conditions. To obtain three independent repetitions, 27 L. intracellularis-infected pigs (19.0 ± 1.50 kg body weight) from one farm were divided into three groups at an age of 7 to 8 weeks (nine pigs/group). Pigs were either vaccinated against L. intracellularis via oral drenching on their 21st day of life (attenuated live vaccine) or non-vaccinated and selected according to clinical findings (pigs without deviating fecal consistency or with moderate to soft fecal consistency). Comparison of the clinically inconspicuous piglets that differed regarding their vaccination status showed fewer significant differences in fecal microbiota composition. The vaccination led to an overall enrichment of bacterial species belonging to the order Clostridiales, while species of the genus Collinsella and Prevotella were decreased. Several bacterial species belonging to the order Bacteroidales, mainly of the family Prevotellacecae, often closely matching Prevotella copri differed significantly between non-vaccinated clinically inconspicuous and conspicuous piglets. Whether those bacterial species play a role in mitigating the severity of an L. intracellularis infection remains to be defined.

Precision livestock farming can combine sensors and complex data to provide a simple score of meaningful productivity, pig welfare, and farm sustainability, which are the main drivers of modern pig production. Examples include using infrared thermography to monitor the temperature of sows to detect the early stages of the disease. To take account of these drivers, we assigned 697 hybrid (BHZP db. Viktoria) sows to four parity groups. In addition, by pooling clinical findings from every sow and their piglets, sows were classified into three groups for the annotation: healthy, clinically suspicious, and diseased. Besides, the udder was thermographed, and performance data were documented. Results showed that the piglets of diseased sows with eighth or higher parity had the lowest daily weight gain [healthy; 192 g ± 31.2, clinically suspicious; 191 g ± 31.3, diseased; 148 g ± 50.3 (p < 0.05)] and the highest number of stillborn piglets (healthy; 2.2 ± 2.39, clinically suspicious; 2.0 ± 1.62, diseased; 3.91 ± 4.93). Moreover, all diseased sows showed higher maximal skin temperatures by infrared thermography of the udder (p < 0.05). Thus, thermography coupled with Artificial Intelligence (AI) systems can help identify and orient the diagnosis of symptomatic animals to prompt adequate reaction at the earliest time.

17. Influences of exocrine pancreatic insufficiency on nutrient digestibility, growth parameters as well as anatomical and histological morphology of the intestine in a juvenile pig model

In a pig model, pancreatic duct ligation (PL) leads to a complete loss of exocrine function, causing an exocrine pancreatic insufficiency (EPI) without affecting endocrine function, allowing research of clinical effects and therapy options. This study aimed to investigate effects of experimentally induced EPI in juvenile pigs on digestion and intestinal morphology. Eight female juvenile cross-bred pigs (BW 54.8 kg at the start of the study) were included. Three animals were considered as a control (CON group), and in five animals the ductus pancreaticus accessorius was ligated (PL group). During the 10-week trial period, body weight and body measurements were recorded regularly. At the end of the trial, gastrointestinal tract (GIT) was investigated macroscopically and histologically and weight and digesta samples of individual segments were obtained. The pigs in the CON showed a significantly higher apparent total tract digestibility of crude protein and crude fat (87.8 and 79.9%, respectively) compared to PL (52.4 and 16.6%, respectively). Significant differences were noted in relative weights of duodenum, jejunum and colon (with and without digesta) and also in absolute weights of jejunum and colon. The mean number of nuclei in the transverse section in stratum circulare were significantly higher in all intestinal segments in CON compared to PL. Overall, EPI results in impaired nutrient digestibility with a greater filling of the GIT with digesta. The elongation of the small intestine does not represent “stretching” of the intestine, but rather increased synthesis of intestinal tissue.

The nutritional benefits of rye (and therefore rye bran) are mainly due to its high content of fermentable dietary fiber, the non-starch polysaccharides (NSP). Microorganisms in the large intestine are able to convert these into short-chain fatty acids (SCFA), including butyrate. Butyrate strengthens the epithelial barrier function in the colon by nourishing the enterocytes and inhibiting the spread of Salmonella in the intestinal tract. Therefore, the aim of this study was to test under field conditions whether a diet with rye or rye bran as the main ingredient for gilts, sows, and weaned piglets is associated with a lower Salmonella prevalence. Depending on the age groups, between 20–30% rye or between 15–20% rye bran was used in the experimental group. A total of n = 1983 boot swabs, n = 356 fecal samples, and n = 1909 serum samples were examined. The results of this study show that rye or rye bran at the levels used had no apparent effect on the number of positive Salmonella samples. However, the Salmonella OD values in the experimental groups were significantly lower than in the control group. This suggests that the use of rye leads to a lower incidence of infection, but this effect could not be proven from swabs.

Foodborne zoonotic diseases can be transferred into the food chain at the stage of livestock farming. As an emerging public health challenge, practicable reduction measures in porcine health management for Salmonella are constantly being investigated. This in vitro study aimed to determine the influence of six different sodium butyrate (SB) concentrations (0, 5, 10, 20, 40, and 80 mM) on the growth of three different Salmonella enterica serovars at a constant pH value of 6.0, corresponding to conditions in the pig's hindgut. S. Derby and S. Typhimurium, isolated from a pig farm, and S. Typhimurium DSM 19587, which served as control, were used. Broth microdilution assay was applied to record Salmonella growth in the presence of different SB-concentrations over six different incubation periods (0, 1, 2, 4, 6, and 24 h). Results were quantified in the log colony-forming units (log10 CFU/mL). For 1 h incubation, the addition of SB showed no significant differences in the range of initial Salmonella dose of about 5.7 log10 between concentrations (0–80 mM, 5.26 ± 0.10–5.60 ± 0.07 log10, p > 0.05). After 6 h, for SB addition, the range of Salmonella counts was significantly lower compared to no addition of SB (5–80 mM, p < 0.05), 6.78 ± 0.84–7.90 ± 0.10 log10 for 5 mM, and 7.53 ± 0.04–8.71 ± 0.22 log10 for 0 mM. Moreover, for SB concentrations of 40 and 80 mM, no difference in the range of Salmonella counts over 6 h was obtained (5.23 ± 0.11–5.38 ± 0.05 log10, p > 0.05), and minor Salmonella growth was recorded at the earliest after 24 h incubation. Growth rates for varying SB concentrations and incubation times were confirmed in a similar manner for the three serovars. Obtained results suggest that increasing SB concentrations suppress Salmonella growth for concentrations of 5–20 mM over a 6 h incubation period and for 40 and 80 mM over a 24 h incubation period. When transferring these in vitro findings to the porcine organism, it may be assumed that Salmonella reduction can be achieved by increased butyrate content in the chyme of the large intestine.

Green liver discoloration (GL) in fattening turkeys is suspected to be a multifactorial disease complex with a compromised immune system as the key factor. This study aimed to identify the formal pathogenesis of GL and to investigate possible nutritional influences. A total of 360 Bronze turkey hens out of 10 flocks from 5 fattening farms were necropsied for detection of GL during 2 consecutive trials on 2 examination dates each (70th to 75th and 120th to 127th day of fattening, respectively). At each examination date, hematological and clinical chemistry analyses, as well as determination of vitamin E and selenium concentrations in the liver, were carried out in 6 hens with (if applicable) and 6 hens without GL, representing a total of 130 individuals. Raw nutrient, energy, amino acid, bulk and trace element, and vitamin E and D3 concentrations were analyzed in feed samples for each of the five feeding phases during each trial. The results of the hematological analyses, clinical chemistry analyses, and determination of vitamin E and selenium liver concentrations were statistically evaluated between: (i) individuals with and without GL, and (ii) individuals from flocks with and without turkeys with GL. At both fattening stages, the occurrence of GL was characterized by an inflammatory reaction. A subacute inflammatory reaction was detected in the early fattening stage, indicating a viral cause of the disease. In the late fattening stage, acute inflammation indicated a bacterial cause of the disease. The results of the feed sample analyses of the different flocks were generally quite homogeneous. However, the nutrient and energy content of the feed likely contribute to GL pathogenesis.

Mammals respond to amino acid (AA) deficiency by initiating an AA response pathway (AAR) that involves the activation of general control nonderepressible 2 (GCN2), phosphorylation of eukaryotic translation initiation factor 2α (eIF2α), and activation of transcription factor 4 (ATF4). In this study, the effects of protein (N) and/or phosphorus (P) restriction on the GCN2/eIF2α/ATF4 pathway in the liver and the induction of fibroblast growth factor 21 (FGF21) in young goats were investigated. An N-reduced diet resulted in a decrease in circulating essential AA (EAA) and an increase in non-essential AA (NEAA), as well as an increase in hepatic mRNA expression of GCN2 and ATF4 and protein expression of GCN2. Dietary N restriction robustly increased both hepatic FGF21 mRNA expression and circulating FGF21 levels. Accordingly, numerous significant correlations demonstrated the effects of the AA profile on the AAR pathway and confirmed an association. Furthermore, activation of the AAR pathway depended on the sufficient availability of P. When dietary P was restricted, the GCN2/eIF2α/ATF4 pathway was not initiated, and no increase in FGF21 was observed. These results illustrate how the AAR pathway responds to N- and/or P-reduced diets in ruminants, thus demonstrating the complexity of dietary component changes.

In animal feeds and pet food, meat industry rendered by-products as a source of high-quality protein are commonly used. Among all rendered protein sources, poultry carcasses and neck meal are frequently used as ingredients in commercial pet foods due to their agreeable fatty acid and amino acid profiles, and they have no impact on the palatability of the diet. Nonetheless, it is unclear how poultry by-product meal affects companion animals regarding diet digestibility and fecal quality. This study either aimed to provide information on poultry by-product meal, including coarsely, finely, or very finely ground varieties, regarding their nutrient digestibility and characteristics of feces in dogs. One type of animal by-product meal was used in the three aforementioned particle sizes. Beagle dogs (n = 6; body weight, 16.6 kg ± 2.03) participated in a crossover experiment design. Each trial consisted of a five day adaptation period to the diet, and five days of fecal samples were collected and measured for individual apparent nutritional digestibility and fecal scores. The animal by-product supplementation in the diet of dogs was well accepted, with an acceptable percentage of apparent nutrient digestibility. Different particle sizes had no significant effect on the organic matter, crude protein, and crude fat digestibility as well as the fecal fatty acid concentrations. In addition, feces remained firm and well-formed and increased fecal dry matter. This indicates that poultry by-products should be taken into account as a potential dietary protein source in dog food.

Characterisation of the variations in physico-chemical properties of grains may help to improve the feeding value of grains for animal nutrition. Thus, this study aimed to obtain more extensive quantitative ideas concerning different physico-chemical properties of wheat, hybrid rye, and barley. The samples were ground in a hammer mill using screen size of 1, 3, and 6 mm, respectively. The cumulative mean particle distribution at >1.0 mm of the ground grains showed significant differences between wheat and hybrid rye (4.63 and 9.13%, respectively). At dry sieve analysis of 6 mm screen size, hybrid rye had significantly higher mean particle size distribution of >1.0 mm (26.8%) than for ground wheat and barley. Ground wheat using a 1 mm mesh sieve had the lowest water holding capacity and swelling capacity (1.89 g H2O/g dry matter (DM); p = .001 and 1.33 mL H2O/g DM; p = .021, respectively) compared to hybrid rye and barley. Ground hybrid rye using a 1 mm mesh sieve had the significantly highest extract viscosity (6.22 mPa s). Ground wheat had the lowest (p 1 mm regardless of the grinding size. Ground wheat had the lowest water holding capacity irrespective of the grinding mesh sieve. Finally, hybrid rye in general is characterised by high extract viscosity (6.22 mPa s at 1 mm grinding size), which decreased with coarser grinding (3.75 and 3.10 mPa s at 3 and 6 mm, respectively).Highlights Particle size distribution is directly affected by the grinding process. Water holding capacity and swelling capacity are two complementary measurements. Extract viscosity seems to be affected by grinding; however, the sedimentation rate is influenced by the grain type.

Abstract Background Fibre sources as feed components with specific physical characteristics like a high swelling capacity (SC), viscosity and water-binding capacity (WBC) have been discussed to affect sow behaviour and to have long-term effects on lactational performance. The present studies aim to analyse the effects of different fibre sources in diets for sows on behaviour in gestation, reproductive performance as well as piglet development. Methods Twenty-eight feedingstuffs (four grain varieties, 16 by-products, three oilseeds and five leguminous plants) were compared concerning swelling capacity, viscosity and water binding capacity to select fibre sources with optimal physical characteristics. Following this a digestibility study was carried out with eight castrates for determining digestibilities of gross energy, crude protein, crude fibre, crude fat and crude ash. Additionally, a practical feeding experiment during gestation was performed with 96 sows of Danish genetics. Two supplements for sows with different fibre sources were composed, namely a control diet (based on wheat bran and lignocellulose) and a test diet containing sugar beet pulp, alfalfa, rapeseed meal, soybean hulls, grape pomace and lignocellulose. Six pens with eight sows each were video-monitored for 2 weeks (evaluation of interactions and fights). Furthermore, the animals were subjected to weekly scoring to count skin injuries. To check the fibre effect on reproductive performance and piglet development, the body condition development of the sows as well as the number and weight of live and stillborn piglets, litter weight- and weaning weight of the litters were recorded. Results Digestibility of crude fibre increased significantly in the experimental group (58.8% ± 3.3 vs. 49.0% ± 4.3, p = 0.01). At the sow trial, there was a tendency to observe less aggressive interactions and fewer fights in sows in the fibre group without significance. No significant differences could be measured between the two groups concerning performance parameters of sows and piglets. Conclusion Only changing the fibre source in a gestational diet does not have significant effects on the sows´ behaviour and performance of sows and piglets in lactation. It should be investigated how the amount of fibre can be increased without having any negatives effects on the performance so that the positive effects on the behaviour of the sows become more obvious.

25. Effects of poultry by-products inclusion in dry food on nutrient digestibility and fecal quality in Beagle dogs.

Animal by-product meals show large variability in diet digestibility. This study aimed to provide information on including bone protein meal (BPM) or feather meal (FM) in extruded dog diets with regard to digestibility and fecal characteristics in two trials. In the first trial, compared to the control (BPM0), 6, 12, and 24% of the basic diets were replaced by BPM (BPM6, BPM12, and BPM24, respectively). In the second trial, in comparison to the control (FM0), 5, 10, and 20% of the basic diets were replaced by FM (FM5, FM10, and FM20, respectively). In both trials, six Beagle dogs (BW 17.3±2.14 and 18.1±2.04 kg for trials 1 and 2, respectively) participated in a crossover experiment design. Five days were used as wash-out before each experimental period for each trial. The fecal consistency scores were based on a 5-point scale (1 = very hard, 2 = solid, well formed "optimum", and 5 = watery diarrhea). In the first trial, results showed that the apparent digestibility of dry matter, crude protein, and crude fat was significantly lower for dogs fed BPM6 compared to those fed BPM24. There was a lower number of dogs with a fecal consistency score value > 2 (16.7%) among those fed BPM6 (median = 2, Interquartile range (IQR) = 0) compared to those fed BPM24 (83.3%). The fecal dry matter content was significantly (p < 0.05) the highest (39.4%±2.15) for dogs fed BPM6. In the second trial, the data revealed that dogs fed FM0 had significantly (p < 0.05) the highest organic matter digestibility (87.2%±1.05), while dogs fed FM20 had significantly (p < 0.05) the lowest crude fat digestibility (95.0%±0.95). Inclusion of FM at 10% or 20% in the diet decreased fecal dry matter significantly (29.0%±2.10 and 27.9%±2.46, respectively) compared to those animals offered FM0 (31.1%±2.56). Among those dogs fed FM0 and FM5, there was a lower significant number of dogs with a fecal score value > 2 (16.7% and 16.7%, respectively; p < 0.05). While the fecal score was significantly a higher (median = 4, IQR = 0) for dogs fed FM20. Including FM at any level in the diet resulted in significantly higher levels of iso-butyric and iso-valeric acids compared to FM0. These findings in both trials suggest that apparent crude protein digestibility was not affected when diets containing BPM up to 24% and FM up to 20% were offered, but fecal quality was reduced.

In order to enhance the health and welfare of obese dogs and to facilitate the required loss of body weight, commercial diets are produced with fibrous ingredients. Cellulose is a common dietary fiber used mainly in powdered form. However, other processing forms and fibers are available as fibrous additives. This work aimed to test the effects of different fiber sources on apparent total tract digestibility and fecal quality in dogs. Four diets were fed to eight dogs (experimental design: 4 × 4 Latin square) for a 14-day period each. In addition to a basal diet (CO), three experimental diets varying in fiber sources were used: powdered cellulose (CE), granulated cellulose (GC), and lignocellulose (LC). Dogs fed the CO had lower crude fiber digestibility than those fed the other experimental diets (p < 0.0033). Dogs fed diets supplemented with fiber sources had lower gross energy digestibility (range: 76.2–77.3%) compared with those fed the CO (84.4%). In all groups, the fecal score (consistency and shape) ranged within the optimal values; solely wet fecal output was increased for the fiber groups compared with those on the CO. This study demonstrated that various sources of fiber such as GC and LC can be used as alternatives to CE without restrictions.

Chronic stress is recognized as a secret killer in poultry. It is associated with systemic inflammation due to cytokine release, dysbiosis, and the so-called leaky gut syndrome, which mainly results from oxidative stress reactions that damage the barrier function of the cells lining the gut wall. Poultry, especially the genetically selected broiler breeds, frequently suffer from these chronic stress symptoms when exposed to multiple stressors in their growing environments. Since oxidative stress reactions and inflammatory damages are multi-stage and long-term processes, overshooting immune reactions and their down-stream effects also negatively affect the animal’s microbiota, and finally impair its performance and commercial value. Means to counteract oxidative stress in poultry and other animals are, therefore, highly welcome. Many phytogenic substances, including flavonoids and phenolic compounds, are known to exert anti-inflammatory and antioxidant effects. In this review, firstly, the main stressors in poultry, such as heat stress, mycotoxins, dysbiosis and diets that contain oxidized lipids that trigger oxidative stress and inflammation, are discussed, along with the key transcription factors involved in the related signal transduction pathways. Secondly, the most promising phytogenic substances and their current applications to ameliorate oxidative stress and inflammation in poultry are highlighted.

The entire gastrointestinal tract (GIT) of 61 adult ostriches (both sexes; av. age of 15 mo), which had previously been fed a ration consisting of either haylage and pelleted compound feed (HP) or haylage, corn silage and pelleted compound feed (HCP), was the subject of the present investigations. Sorghum is the fifth most produced cereal in the world and is a source of nutrients and bioactive compounds for the human diet. This study examined the nutrient composition and in vitro fermentation characteristics of sorghum varieties grown in 2020 and 2021 (n = 15 × 3 × 2) across three locations in the north of Italy (Bologna, Padova, and Rovigo). In 2020, the crude protein content of sorghum was significantly higher in the region of Padova than in the region of Bologna (124 vs. 95.5 g/kg dry matter). However, crude fat, sugar, and gross energy levels showed no significant differences among the different regions in 2020. In 2021, the levels of crude protein, crude fat, sugar, and gross energy had no significant differences among different sorghum varieties harvested from the three regions. Significant differences in some mineral contents were found among the samples particularly in manganese and zinc in both years. After 24 h of fermentation of two different sorghum hybrids (hybrids 1 and 2 of both years harvested in Bologna, n = 4 × 2 × 2), the pH value was significantly higher in hybrid 1 of year 2021 (3.98) than in the other fermented samples (range: 3.71–3.88). The sorghum harvested from the region of Bologna had a significantly higher viscosity value (1.22 mPa·s) compared to other regions (1.8–1.10 mPa·s) in 2021 only. The results show that the nutritional value and viscosity of different sorghum varieties could differ depending on the location and year of cultivation.

Little is known regarding the interplay between microbiota and pancreas functions in humans as investigations are usually limited to distal sites, namely the analyses of fecal samples. The aim of this study was to investigate both ileal and fecal microbiota in response to pancreatic enzyme replacement therapy (PERT) in a porcine model of exocrine pancreatic insufficiency (EPI). PERT was stopped for ten days in ileo-cecal fistulated minipigs with experimentally induced EPI (n = 8) and ileal digesta as well as fecal samples were obtained before withdrawal, during withdrawal and after the reintroduction of PERT. Profound community changes occurred three days after enzyme omission and were maintained throughout the withdrawal phase. A reduction in α-diversity together with relative abundance changes in several taxa, in particular increases in Bifidobacteria (at both sites) and Lactobacilli (only feces) were observed. Overall, dysbiosis events from the ileum had accumulating effects in distal parts of the gastrointestinal tract with additional alterations occurring only in the colon. Changes were reversible after continuing PERT, and one week later, bacterial communities resembled those at baseline. Our study demonstrates the rapid and profound impacts of enzyme withdrawal in bacterial communities, contributing to our understanding of the interplay between pancreas function and microbiota.

Abstract Background The conventional farrowing crate is criticised due to the limited mobility of sows during farrowing and lactation. The present study aims to investigate the effects of three different farrowing systems on the performance of suckling neonates on the basis of immunocrit (IC; a quantification of immunoglobulins), serum amino acid (AA) concentrations and growth performance. Methods From a total of 149 sows placed in three housing systems (farrowing crate – FC, loose housing – LH, group housing – GH), 18 sows and their respective litters, formed the basis for a two-factorial study design (farrowing system and body weight (BW) of neonates). Therefore, also blood samples of two light (1.0–1.4 kg) and two heavy (≥ 1.4 kg) piglets were taken within 48 h post natum (p.n.) and on the day of weaning (day 26) to determine the immunocrit (IC; a quantification of immunoglobulins) and levels of serum AAs. Results The IC (FC: 0.148a, LH: 0.153a, GH: 0.117b) as well as serum levels of arginine, leucine, lysine, proline and threonine within 48 h p.n. were significantly lower in GH. Additionally, in general, these piglets showed (except for the first week of life) the lowest average daily weight gain. On the day of weaning, piglets in GH had the lowest levels of arginine (in mg/dL; FC: 3.68a, LH: 3.40ab, GH: 2.94b) and threonine (in mg/dL; FC: 3.59a, LH: 3.02ab, GH: 2.49b). The concentrations of leucine, lysine, proline and valine at this time were significantly lower in LH. Conclusion The observed significant lower IC indicates a lower Ig intake of piglets in the tested GH. No significant differences regarding the IC and AA levels within 48 h p.n. of the piglets in FC and LH could be seen. In principle, differences at weaning in AA levels were rather small, although the body weight of GH piglets at weaning was lower. Therefore, further research needs to clarify whether there are medium-term effects on health and performance.

Lignocellulose (LC) might be used as a substitute fiber source for dogs as a strategy to decrease energy density and enhance gastrointestinal functionality. The objective of the present study was to compare the effects of including different levels of LC on apparent nutrient digestibility and fecal parameters (dry matter (DM), fecal score, and daily fecal output), as well as fecal fatty acid concentrations. Four diets were tested: control diet (no supplementation of LC; LC0), and three control diets diluted with increasing levels of LC: 1, 2, and 4% (LC1, LC2, and LC4). Six Beagle dogs (BW 17.1 kg ± 1.22) participated in a crossover experimental design. Before each experimental period, five days were used as a wash-out period. The fecal consistency was scored based on a 5-point scale (1 = very hard; 2 = solid, well-formed “optimum”; 3 = soft, still formed; 4 = pasty, slushy; and 5 = watery diarrhea). The results demonstrated that the organic matter apparent digestibility (p = 0.01) and nitrogen-free extract (NFE) (p < 0.01) was significantly lower for dogs fed LC4 compared to those fed LC0. Dogs fed LC2 and LC4 had a lower fecal consistency score (2.39 and 2.18, respectively; p < 0.05). The fecal DM was significantly the highest (p < 0.01), and the daily fecal output on fresh matter was lower (p < 0.05) when dogs were fed the LC4 diet compared with the LC0 diet. Including LC at 1% in the diet resulted in significantly higher fecal acidic pH levels. However, no differences among treatments were noted regarding fecal fatty acid concentrations.

Evidence has already been provided that feed-borne mold spores and endotoxins can trigger chronic, non-infectious respiratory disease if inhaled. Furthermore, deficiencies in feed microbiology are suspected to trigger gastrointestinal and liver disorders in horses, but the connection needs further clarification. Most of the previous studies regarding horse feed hygiene focused on forage, whereas research regarding hygienic quality of concentrates is scarce. In the present study, results of reports on hygienic quality of compound feed and cereals for horses were evaluated secondarily. Results included sensory findings, and counts of aerobic bacteria, molds and yeasts determined by cultivation and lipopolysaccharide (LPS) contents. It was found that microbial counts of compound feed exceeded VDLUFA orientation values significantly more frequently than cereals (38.4 vs. 22.6%). However, average counts of bacteria, molds and yeasts were higher in cereals than in compound feeds (p < 0.0001, respectively). Mold counts in grains were significantly higher if dry matter contents were below 86% (p = 0.0201). No relation could be established between the anamnestically reported gastrointestinal disorders or elevated liver enzyme activities and microbiological deviations. Mold counts of concentrates which were suspected to cause coughing in horses were significantly higher than mold counts of control samples (3.29 vs. 2.40 log10 cfu g−1, p = 0.0313). These results indicate that hygienic status of concentrates is relevant for horse health in the respiratory tract.

Bacterial fermentation of undigested carbohydrates in the hindgut has considerable potential for the stimulation or inhibition of the growth of distinct bacteria within microbiota. The aim of the present study was to evaluate whether high levels of rye affect porcine gut microbiota composition with subsequent effects on the load of Salmonella Typhimurium, an intestinal pathogen with zoonotic relevance. Therefore, forty-two 25-day-old piglets were allocated to two groups and fed a diet containing either 69% wheat or 69% rye for 35 days. One week after introducing the two different diets, the piglets were experimentally infected with Salmonella Typhimurium. The microbiota composition of cecal and fecal samples of the piglets were evaluated 28 days after infection. In the cecum, promoted growth of Bifidobacterium, several lactic acid bacteria and Faecalibacterium prausnitzii were seen in pigs fed the diet containing 69% rye. Bacterial species belonging to the genera Bifidobacterium and Catenisphaera were associated with differing bacterial counts of Salmonella Typhimurium detected in the cecal contents of all piglets in both feeding groups via cultural cultivation. The high intake of rye instead of wheat seems to promote the growth of beneficial intestinal bacteria accompanied by impaired growth conditions for the foodborne pathogen Salmonella Typhimurium.

35. Influence of Using Perforated Plastic Flooring Beneath the Waterline on Growth Performance, Litter Quality, and Footpad Health of Broiler Chickens: A Field Study

The aim of this study was to evaluate the effect of slatted flooring beneath waterlines in broiler barns on litter quality and, subsequently, footpad health. The hypothesis tested was that installing slatted flooring underneath waterlines helps to improve litter quality and thus reduces footpad diseases, enhancing animal welfare as a result. Five experimental runs with two groups were conducted. Each run was defined as one fattening period of 32 days and consisted of 15,000 broiler Ross 308 of both sexes. Every barn was divided into three areas (drinkers, feeders, and comfort area) for weekly sampling. No influence on growth performance was noted. The slatted flooring influenced the litter quality by preventing the litter in the experimental group (EG = 690 ± 167 g/kg DM) from becoming moisture until day 14 of the fattening period compared to the control group (CON = 636 ± 198 g/kg DM). The footpad health was also influenced by using slatted flooring, with lower camera-based footpad scores in the EG (8.80) compared to CON (22.0) at the slaughterhouse (p = 0.0258). Installing slatted flooring beneath the waterline reduced the moisture of the litter compared to the control barn in the first two weeks of age and showed a positive effect on the footpad health of the broilers at the end of fattening, which indicates an improvement in animal welfare.

36. Impacts of Reducing Protein Content in Milk Replacer on Growth Performance and Health of Young Calves

In the present study, a drinking amount of 10 L of milk replacer (MR) was allowed to dairy calves in order to approach the natural drinking behavior. The question is: how much protein is required by calves in order to achieve an intended growth rate? For this reason, sixty-eight pre-weaned Holstein calves were divided into two groups and fed with 10 L/d of MR containing either 22% protein (MR22) or 19% protein (MR19) at an almost comparable energy intake. Effects on performance, metabolic status, and health were compared. Feed intake, growth performance, and health status were monitored during the pre-transition, transition, and postweaning phase (until 157 d of age). Total feed intake, and intake of MR, body weight (BW), and average daily gain (ADG) were not significantly different between MR22 and MR19 during the entire experimental period (p > 0.05). At d 42, calves in MR19 group showed greater serum levels of growth hormone (16.2 vs. 22.2 ng/mL; p = 0.02), insulin-like growth factor 1 (262 vs. 291 ng/mL; p = 0.03), and urea (2.86 vs. 3.04 mmol/L; p < 0.01). The results of the present study suggested that when high amounts of MR are provided, the protein content in MR can be reduced to 19% without any adverse effects on growth performance as well as on health status of dairy calves.

In piglet production, the beginning of pork production, Salmonella prevalence requires greater attention as having an impact on the subsequent production steps. The aim of this study was to investigate Salmonella prevalence in three sow herds with attached piglet rearing units. Salmonella prevalence was investigated either directly by boot swabs and feces or indirectly by serum samples taken during gilt integration, the peripartal period, and piglet rearing. Boot swabs and feces were analyzed by real-time PCR and subsequent microbiology. Results indicated that high biosecurity measures in sow husbandry do not necessarily result in a low Salmonella prevalence. Furthermore, the sow herds’ Salmonella prevalence should not be used to infer the situation in the associated piglet rearing. The proportion of positive boot swabs was 10.5, 3.6, and 21.3% for sows (gilts and peripartal) with an inverse situation in piglet rearing with 50.0, 63.3, and 5.8% positive swabs for farms A, B, and C, respectively. Boot swabs are suitable as a direct sampling method to gain an overview of Salmonella prevalence in both sows and piglets. Indirect serum antibody testing can be useful, although it should be evaluated considering age-dependent levels of antibody titres.

Feed supplements such as Saccharomyces cerevisiae fermentation products (SCFP) alter immune responses in horses. The purpose of this study was to analyze whether a prebiotic activity of the SCFP alters the gut microbiome in horses. Racehorses were fed either SCFP (Olimond BB, OLI, n = 6) or placebo pellets (PLA, n = 5) for 43 days. Fecal microbiota analysis was performed using 16S rRNA gene sequencing. The numbers and function of circulating immune cell subpopulations were analyzed by flow cytometry. SCFP supplementation resulted in non-consistent differences in fecal microbiota between the PLA and OLI during the feeding period. Rather, the individual animal had the highest impact on fecal microbiota composition. OLI and PLA horses displayed the same changes in numbers of blood leukocyte subpopulations over time. One day after a booster vaccination against equine influenza during the feeding period, the alpha diversity of fecal microbiota of PLA horses was significantly higher compared to OLI horses. This suggests that SCFP feeding altered the vaccination-induced spectrum of released mediators, potentially affecting gut microbiota. The overall non-consistent findings argue against a strong prebiotic effect of Olimond BB on the microbiota in racehorses. Fecal microbiota differences between the groups were also noticed outside the feeding period and, hence, are most likely not caused by the SCFP additive.

To investigate the influence of two dietary sources of vitamin D on the vitamin D status, bone metabolism, welfare and birth progress of gestating and lactating sows, forty-nine multiparous sows were randomly assigned to one of two diets: “CON” (n = 25; 50 μg vitamin D3/kg feed) and “HYD” (n = 24; 50 μg 25-hydroxycholecalciferol/kg feed). The basal diets were protein- and phosphorus-reduced. The trial started on day 3 ante insemination of the sows and ended with weaning of the piglets on day 28 postpartum. Dietary supplementation of 25-hydroxycholecalciferol resulted in improved maternal vitamin D status (p < 0.001), fewer gait changes (p < 0.01) and longer standing time after feeding (day 5 ante partum; p < 0.05) compared to vitamin D3. However, the bone markers CrossLaps and osteocalcin were not affected. Overall, the present results suggest that sows fed 25-hydroxycholecalciferol instead of vitamin D3 showed improved locomotion and stance strength. However, this outcome is probably not related to altered bone metabolism. The underlying mechanisms must be investigated in further studies.

Pet owners are increasingly concerned about the links between health status, animal welfare, environmental impacts, climate change and consumption of animal products. Accordingly, many owners are increasingly interested in vegetarian diets for themselves and their companion animals. However, such diets should be investigated nutritionally regards digestibility as well as on fecal quality and nitrogen output. In light of this trend, six Beagle dogs were included in a cross-over experimental design and offered a vegetarian diet containing wheat gluten (8.81%), rice protein (8.81%) and sunflower oil (6.84%) or an meat-based diet containing poultry meal (19.5%) and poultry fat (5.23%). The dogs received extruded complete diets for 12 days (adaptation and collection period, each 6 days). The dogs fed both diets showed a high and identical palatability (scoring of food intake) of the experimental diets. No significant differences occurred regarding digestibility of organic matter, crude protein and crude fat between vegetarian and meat-based diets. However, dogs fed the meat-based diet had higher (p < 0.05) nitrogen-free extract digestibility (89.5%) compared to those fed the vegetarian diet (88.6%). The amount of nitrogen excreted in feces (g)/kg BW0.75 was slightly, but not significantly, higher for dogs fed the vegetarian diet compared to those fed the meat-based diet (0.88 vs 0.79). The fecal consistency scores were considered to be within an acceptable range (well formed and firm). The mass of the feces between both groups were similar (62.9 g wet feces/100 g dry matter food) for vegetarian and meat-based diets. Additionally, the fecal dry matter content was comparable between both groups (29.0% and 29.6% for vegetarian and meat-based diets, respectively). In conclusion, the results of this study appear to indicate that virtually the only significant difference between the two diets was lower nitrogen-free extract digestibility in the vegetarian diet. However, the vegetarian diet did not result in a significant difference in amount of nitrogen excreted in feces.

The hygienic quality of forage for horses is discussed as a potential health hazard, especially regarding respiratory diseases, colic, and hepatopathies. Therefore, the purpose of this study was to investigate the possible relations between microbiological counts, as well as endotoxin levels and disease symptoms. Data from microbiological examination reports were analyzed retrospectively, including the results of sensory examination, microbiological counts, and lipopolysaccharide contents. Sensory analysis gave an indication of deficiencies in microbiological analysis, but both methods did not give consistently equivalent results regarding the hygienic status of forage. The strongest agreements between sensory and microbiological findings were demonstrated in haylage regarding mold contamination. The influences of dry matter content on microbiological quality could be shown in haylage and hay, whereas this did not apply to straw. Deviations regarding molds and the detection of Aspergillus species occurred, especially in haylage, with values above 70% DM detected (39.6%, p=0.0021 and 47.2%, p = 0.0393). Aspergillus was detected more frequently, and average counts were higher in samples that were suspected to induce coughing in horses (p = 0.0118 and p = 0.0313, respectively). The results of the present study emphasize the importance of feed hygiene for equine respiratory health and the need for the microbiological examination of feedstuffs, since sensory analysis cannot provide an error-free prediction of microbial counts.

The pH of the fermented rye at a DM content of 25% after 24 h had the lowest values (3.57) compared to that at a DM content of 75% (6.42). In fermented sorghum, pH values were lower than 4 already after incubation at 25% DM for 12 h (3.93) in comparison to that at DM content 75% (6.51). The L-lactic acid concentration in the fermented rye with 25% DM content after 24 h was significantly the highest (18.7 g/kg DM), as was that of sorghum with 25% DM content after 24 h (22.2 g/kg DM). Moreover, the acetic acid level in the fermented rye with 25% DM content after 24 h was significantly the highest (3.02 g/kg DM) compared to the other DM contents of fermented rye. Also, in fermented sorghum (25% DM), the acetic acid content was significantly the greatest (1.49 g/kg DM) in comparison to the other DM contents of fermented sorghum. Overall, fermented rye and sorghum containing 25 or 37.5% DM for 24 h and 12 h for rye and sorghum, respectively are sufficient for fermentation to be optimized based on the values of pH and lactic acid content, except for acetic acid content, which may negatively affect the palatability in animals.

Meat and bone meal (MBM) is one animal by-product used in pets. This study purposed to provide information on MBM including either coarsely (MBMc) or finely (MBMf) ground with regard to diet digestibility and fecal characteristics in dogs. Three different levels of MBM (6%, 12% and 24%) of each grinding form (MBM6, MBM12 and MBM24) were added to a basic diet. Six Beagle dogs (body weight 16.7 kg ± 0.42) participated in a Latin Square experiment. Each trial began with the animals adapting to the food for a five-day period, followed by five days of fecal collection. The feed particle size had no effect on the apparent digestibility of organic matter, crude protein and crude fat. The fecal score was significantly affected by the particle size × level interaction among treatments (p < 0.0001). It was noted that the different particle sizes or levels of MBM as main effects had no significant effect on the fecal fatty acid concentrations. These findings suggest that using coarse or fine grinding even including MBM up to 24% in dog diets does not affect the apparent digestibility of protein nor fecal quality negatively in our experimental study.

The aim of this study was to investigate the influence of ad libitum access to compound feeds rich in fibre (ante partum (a.p.) and peri partum) on the DM intake, body mass development and performance of sows as well as excretion of Clostridium (C.) perfringens via sows’ faeces. From day 109 (d-7) of gestation, 25 of 48 sows (23 considered as control) received access to one of two different high-fibre pellets from d-7 until the second day post partum (p.p.) (d2) (fibre groups (FG) 1 and 2) in additional to a lactation diet. The additional DM intake of the high-fibre pellets a.p. was 2.13 ± 1.15 kg in FG 1 and 3.14 ± 0.68 kg in FG 2. This led to higher DM intake in the first lactation week and significantly lower losses of weight and back fat thickness during lactation. The bacterial counts of C. perfringens in sows’ faeces directly p.p. were 10 times lower in FG 1 and 100 times lower in FG 2 compared to the controls. High amounts of fibre led to higher DM intake throughout lactation, which had beneficial effects on sows’ body conditions. It seems that high fibre intake influenced the excretion of C. perfringens at parturition, which could improve the health of newborns.

The gut microbiota has been designated as a hidden metabolic ‘organ’ because of its enormous impact on host metabolism, physiology, nutrition, and immune function. The connection between the intestinal microbiota and their respective host animals is dynamic and, in general, mutually beneficial. This complicated interaction is seen as a determinant of health and disease; thus, intestinal dysbiosis is linked with several metabolic diseases. Therefore, tractable strategies targeting the regulation of intestinal microbiota can control several diseases that are closely related to inflammatory and metabolic disorders. As a result, animal health and performance are improved. One of these strategies is related to dietary supplementation with prebiotics, probiotics, and phytogenic substances. These supplements exert their effects indirectly through manipulation of gut microbiota quality and improvement in intestinal epithelial barrier. Several phytogenic substances, such as berberine, resveratrol, curcumin, carvacrol, thymol, isoflavones and hydrolyzed fibers, have been identified as potential supplements that may also act as welcome means to reduce the usage of antibiotics in feedstock, including poultry farming, through manipulation of the gut microbiome. In addition, these compounds may improve the integrity of tight junctions by controlling tight junction-related proteins and inflammatory signaling pathways in the host animals. In this review, we discuss the role of probiotics, prebiotics, and phytogenic substances in optimizing gut function in poultry.

Abstract Background Lawsonia intracellularis is one of the world’s most important infectious diseases in pork production with regard to economic losses. So far, studies are missing that describe the effects of a natural infection of piglets on the digestibility of nutrients, possible effects on performance and the morphometrics of the intestine depending on whether piglets are vaccinated, clinically healthy or clinically affected with regard to Lawsonia intracellularis induced diarrhoea. Results Digestibility studies were performed on a total of 27 eight-week-old piglets with naturally occurring Lawsonia intracellularis infection in a trial with three repetitions. Nine out of 27 animals were vaccinated as suckling pigs with a commercial Lawsonia intracellularis vaccine (vac; Enterisol®Ileitis). Half of the remaining 18 animals were without clinical signs of infection (non-vac/cs-), half showed moderate clinical signs of Lawsonia intracellularis induced diarrhoea (non-vac/cs+). All three groups were fed one identical complete diet ad libitum. Faecal shedding of Lawsonia intracellularis was found in all groups (25 out of 27 animals). Numerically, the mean excretion in the group non-vac/cs + (7.69 ± 1.65 log10 copies/ g faeces) was higher in comparison to the group non-vac/cs- (5.83 ± 2.35 log10 copies/ g faeces) and vaccinated animals (vac: 6.00 ± 2.89log10 copies/ g faeces). The average daily weight gain (ADG; Ø 8.66 day period) differed significantly (vac: 894a ± 73.3, non-vac/cs-: 857ab ± 86.3, non-vac/cs+: 785b ± 137 g/day). The apparent total tract digestibility (ATTD) of nitrogen was significantly lower in clinically affected animals (vac: 83.0a ± 1.72, non-vac/cs-: 83.9a ± 2.03, non-vac/cs+: 80.7b ± 2.57).The total length of the small intestine in clinically affected animals increased significantly (vac: 15.9ab ± 1.57, non-vac/cs-: 14.6b ± 1.12, non-vac/cs+: 16.2a ± 1.37 m). The relative body weight depending on the length of the small intestine was lower for clinically affected animals (vac: 1.72a ± 0.21, non-vac/cs-: 1.83a ± 0.17, non-vac/cs+: 1.56b ± 0.12 kg/m). Conclusion These studies show that clinically moderate L. intracellularis infections lead to significantly lower ADGs in comparison to vaccinated animals. The disease is also found in altered intestinal morphometry and reduced total N digestibility if clinical signs occur.

Abstract Background Despite years of effort, the proportion of serologically Salmonella-conspicuous pig farms has not been significantly reduced. Incoming piglets are considered to be a significant source of Salmonella for feeder-to-finish-farms. Therefore it is important for farrow-to-feeder-farms to deliver Salmonella-inconspicuous piglets. The aim of the present study was to establish a possible link between an inadequate colostrum supply as a side effect of steadily increasing number of piglets born alive and weaned per sow and increasing Salmonella seroprevalence in piglet rearing on Salmonella-conspicuous farms. Methods Twenty four farms in total were selected for this study. Half of the farms (n = 12) had been detected as Salmonella-conspicuous in previous serological tests on piglets (25 kg) and remaining farms (n = 12) had appeared Salmonella-inconspicuous. Every farm was visited once 24–28 h after the main day of farrowing. For sampling, four sows were randomly selected on each farm. The parity, the litter weight and the litter size were recorded. The sow and six of her piglets were selected for blood sampling (two light-weight, two medium-weight and two heavy-weight piglets respectively). In addition, the colostrum supply of newborn piglets was estimated by using the immunocrit. Results The lightest piglets on Salmonella-inconspicuous and Salmonella-conspicuous farms showed a significant difference (p 0.05) in the factors body weight, litter weight, parity and litter size. Conclusion The study provides preliminary evidence that when comparing Salmonella-inconspicuous farms with Salmonella-conspicuous ones, the colostrum supply may be a critical factor that needs to be considered. The fact that there is no difference in body weight between the two groups of farms suggests that there may be differences in farrowing management and especially colostrum management. Further studies are now required to investigate what causes the various colostrum supply on the respective farms and what long-term effects the individual colostrum supply might have on Salmonella prevalence at abattoir.

Abstract Background Lawsonia intracellularis is one of the most economically important pathogens in swine production. This study tested the hypothesis that the composition of diets for pigs has an impact on the excretion of L. intracellularis in a natural infection model. Results Fifty boars (~ 90 kg BW) from a SPF-farm with a strict hygiene and management regime for reducing the spread of an L. intracellularis infection up to the beginning of the final fattening period were transported, regrouped and randomly allotted to groups of five animals each at the research facility. After a 1-week acclimatisation period groups were fed one of five diets 4 weeks before slaughter. These were either a finely ground pelleted diet (FP) or a coarsely ground meal diet (CM), both consisting of wheat (40.0%), barley (39.3%), soybean meal (16.0%), soybean oil (2.0%) and minor components. In the other meal diets parts of wheat, barley and soybean meal were substituted either with 22% cracked corn (CORN), 16.9% dried whey (WHEY) or 30% raw potato starch (RPS). The animals had a comparable serological status in a blocking-ELISA immediately before the start and at the end of the feeding experiment. Values increased significantly during the trial. In all subgroups (FP/CM/CORN/WHEY/RPS), shedding was detected in week 0 (genome equivalents = GE; log10 GE L. intracellularis/g faeces: 2.46 ± 2.64/3.58 ± 2.54/3.43 ± 2.37/2.30 ± 3.16/2.58 ± 2.73). The average number of L. intracellularis microbes in faeces during the trial period did not differ between the groups (log10 GE L. intracellularis/g faeces: 3.40 ± 1.53/3.01 ± 1.41/3.80 ± 1.71/3.98 ± 2.20/4.08 ± 2.13). In animals fed the WHEY-diet, significantly lower counts of L. intracellularis were found in the caecal content. The acetate content in the caecum was negatively correlated with the serological results at the end of the trial (r = − 0.36; P = 0.010). Butyrate concentrations in the caecal content were negatively correlated with the number of L. intracellularis in the caecum (r = − 0.32; P = 0.023). Conclusion Therefore, this study provides preliminary evidence that there might be specific dietary effects on the course of a L. intracellularis infection.

Monitoring of sows’ health is the key to preventing and controlling diseases in sows, and it guarantees optimal rearing conditions for piglets. The aim of this study was to investigate the relationship between the health status of sows shortly after parturition, and to analyze thermographic images of the mammary gland and the sows’ performance. Clinical examination of a total of 513 db.Viktoria hybrid sows was bundled individually using a modified score system. According to this, animals were divided into three health classes: healthy, clinically suspicious, and diseased. Simultaneously, the mammary glands were investigated by infrared thermography. Total born piglets (TBP), number of piglets born alive (NBA), and the daily weight gain of the piglets were significantly lower in the diseased group (p < 0.05). Regarding the results of the thermographic images of the mammary gland, significantly higher mean value of the warmest pixels was found in the diseased group (38.3 °C ± 0.57), while the significantly lowest value was reported in the healthy group (37.2 °C ± 0.54; p < 0.05). The results of this study show that thermography of the mammary gland at birth contains information that can help to identify diseased animals whose disease has negative effects on their piglets.

50. Effects of Early Nutrition of Hatched Chicks on Welfare and Growth Performance: A Pilot Study

This study aimed to investigate the possibility of rearing newly hatched chicks with immediate access to feed and water in the same hatching unit one week prior to transferring them to the conventional broiler house with special regards to foot pad health and growth performance. Two trials were performed with a total of 6900/6850 (trials 1/2) broiler chickens (ROSS 308). A total of 3318/3391 chicks (trials 1/2) were transported from the hatchery (duration of about 3 h) and reared in a conventional broiler house (control group: delayed nutrition on-hatchery hatched). The control group did not receive any form of nutrition until they were taken to conventional broiler housing. Additionally, a total of 3582/3459 (trials 1/2) embryonated eggs (d 18) were obtained from the same parent flock of the same commercial hatchery and taken to the farm facility. After on-farm hatch, the chicks had immediate access to water and feed (experimental group: early nutrition on-farm hatched). After d 6/7 of life, the on-farm hatched chicks (trials 1/2) were transferred to the broiler house on the same facility. The delayed nutrition on-hatchery hatched groups displayed a significantly lower dry matter content in the litter compared to the early nutrition on-farm hatched groups (two-factorial analysis) at d 6/7 and d 14 of life. However, thereafter, no significant differences were noted. Based upon two-factorial analysis, the early nutrition on-farm hatched groups revealed lower foot pad lesions from d 14 of life onwards and showed a higher body weight (BW) throughout the rearing period compared to the delayed nutrition on-hatchery hatched groups (p < 0.05). Overall, early nutrition on-farm hatched chickens is of critical importance together with using new litter at d 7 to maintain healthy foot pads as well as to enhance nutrient utilization and optimize the growth performance.
