Epibenthic biofilms are important in regulating nitrogen (N) fluxes in stream ecosystems. The efficiency of the regulation is controlled by hydraulic and biological processes and their interactions. However, knowledge on the underlying physical and biological processes, their controlling parameters, and interactions in stream ecosystems is still limited. To analyze the relative importance of hydraulic and biological controls on biofilm N uptake, we measured turbulence, biofilm N uptake using a stable isotope tracer, and biofilm biomass in two gravel-bed streams with contrasting nutrient concentrations for two seasons. We found high within-stream vari ability in biofilm areal N uptake and uptake velocity, which exceeded variability between streams and seasons by 60% and 30%, respectively. Sixty-four percent of the within-stream variability in uptake velocity was explained by hydraulic mass transfer and biofilm characteristics, which were described in terms of the turbulent dissipation rate and the biofilm biomass, respectively. We show that surface renewal theory based on scales of the smallest turbulent eddies can be used to estimate transfer velocities at the sediment-water interface and can be extrapolated to larger scales by spatial averaging. Our results improved the mechanistic understanding of the processes regulating biofilm N uptake at small scale which contributes to the understanding of ecosystem func tioning in low-order streams and supports upscaling to larger spatiotemporal scales along stream networks.

Benthic biofilms are often assumed to control terrestrially-derived dissolved organic carbon (tDOC) uptake in streams. We tested this by comparing 13C-enriched ryegrass leachate uptake in an agricultural and a forest stream, hypothesizing that a greater abundance of autotrophic biofilms in the agricultural stream would cause its whole-stream tDOC uptake to be comparatively low. We measured whole-stream and biofilm tDOC tracer uptake, metabolism, bacterial and algal diversity, and nutrient status of benthic epilithic biofilms, and assessed whole-stream hydromorphology. Whole-stream uptake of tDOC was six times lower in the agricultural (3.0 mg m−2 day−1) than in the forest (19.0 mg m−2 day−1) stream, and tDOC uptake velocity indicated lower tDOC demand in the agricultural (1.2 mm min−1) than in the forest (1.9 mm min−1) stream. The agricultural stream differed from the forest stream by slightly lower transient storage capacity and higher benthic biofilm bacterial abundance and production, lower biofilm biomass and lower biofilm molar C:N, C:P, and N:P ratios. Changes in epilithic biofilms contributed little to the differences in whole-stream tDOC tracer uptake between streams, as biofilm tDOC uptake only amounted to 4% and 13% of whole-stream uptake in the forest and agricultural stream, respectively. This comparison of a forest and an agricultural stream suggests that agricultural stressors have the potential to diminish both whole-stream tDOC uptake and uptake efficiency. Furthermore, the weak link between biofilm and whole-stream tDOC uptake implies that benthic biofilms characteristics are poor predictors for human impacts on tDOC uptake in agricultural streams and that hot spots of tDOC uptake are likely situated in the hyporheic zone or in the stream water column.

Nitrogen (N) uptake is a key process in stream ecosystems that is mediated mainly by benthic microorganisms (biofilms on different substrata) and has implications for the biogeochemical fluxes at catchment scale and beyond. Here, we focused on the drivers of assimilatory N uptake, especially the effects of hydromorphology and other environmental constraints, across three spatial scales: micro, meso and reach. In two seasons (summer and spring), we performed whole-reach 15N-labelled ammonium injection experiments in two montane, gravel-bed stream reaches with riffle–pool sequences. N uptake was highest in epilithic biofilms, thallophytes and roots (min–max range 0.2–545.2 mg N m−2 day−1) and lowest in leaves, wood and fine benthic organic matter (0.05–209.2 mg N m−2 day−1). At the microscale, N uptake of all primary uptake compartments except wood was higher in riffles than in pools. At the mesoscale, hydromorphology determined the distribution of primary uptake compartments, with fast-flowing riffles being dominated by biologically more active compartments and pools being dominated by biologically less active compartments. Despite a lower biomass of primary uptake compartments, mesoscale N uptake was 1.7–3.0 times higher in riffles than in pools. At reach scale, N uptake ranged from 79.6 to 334.1 mg N m−2 day−1. Highest reach-scale N uptake was caused by a bloom of thallopyhtes, mainly filamentous autotrophs, during stable low discharge and high light conditions. Our results reveal the important role of hydromorphologic sorting of primary uptake compartments at mesoscale as a controlling factor for reach-scale N uptake in streams.

This study addressed the influence of common shoreline engineering structures (off-bankline revetment, rip rap and wing dike) on richness, biomass and secondary production of native and non-native macroinvertebrates in the navigation channel and near-shore habitats in the Elbe River (Germany). Within the navigation channel, only marginal differences among engineering structures were observed, and non-native species were absent from all samples. At the shoreline, secondary production of non-native species was significantly greater at the rip rap and represented 59% of total secondary production in near-shore habitats. Conversely, secondary production of native species at the shoreline was 9-fold lower at the rip rap and more than twice the rates at the wing dike. Differences in secondary production among engineering structures were attributed to differential distribution of substrate types. Boulder substrates, the dominant substrate type in the rip rap, promoted contributions of non-native species while macrophytes and silt were associated with high contributions of native species at the off-bankline revetment. Our results reveal that the morphological configuration of engineering structures in large rivers not only controls the rate of secondary production for macroinvertebrates but also the contribution of non-native species to total community functioning.

Previous laboratory and on-site experiments have highlighted the importance of hydrodynamics in shaping biofilm composition and architecture. In how far responses to hydrodynamics can be found in natural flows under the complex interplay of environmental factors is still unknown. In this study we investigated the effect of near streambed turbulence in terms of turbulent kinetic energy (TKE) on the composition and architecture of biofilms matured in two mountainous streams differing in dissolved nutrient concentrations. Over both streams, TKE significantly explained 7% and 8% of the variability in biofilm composition and architecture, respectively. However, effects were more pronounced in the nutrient richer stream, where TKE significantly explained 12% and 3% of the variability in biofilm composition and architecture, respectively. While at lower nutrient concentrations seasonally varying factors such as stoichiometry of dissolved nutrients (N/P ratio) and light were more important and explained 41% and 6% of the variability in biofilm composition and architecture, respectively. Specific biofilm features such as elongated ripples and streamers, which were observed in response to the uniform and unidirectional flow in experimental settings, were not observed. Microbial biovolume and surface area covered by the biofilm canopy increased with TKE, while biofilm thickness and porosity where not affected or decreased. These findings indicate that under natural flows where near bed flow velocities and turbulence intensities fluctuate with time and space, biofilms became more compact. They spread uniformly on the mineral surface as a film of densely packed coccoid cells appearing like cobblestone pavement. The compact growth of biofilms seemed to be advantageous for resisting hydrodynamic shear forces in order to avoid displacement. Thus, near streambed turbulence can be considered as important factor shaping the composition and architecture of biofilms grown under natural flows.

Dissolved organic matter (DOM) is an important part of the global carbon cycle and significantly influences aquatic ecosystem functions. Recent studies suggest that its amount and composition in freshwaters may be altered by agricultural land use, whereby the influence of preceding in-stream production and processing is not clear. To assess the land use effect on DOM amount and composition for the export from terrestrial to freshwater systems at the land–water interface, we sampled headwater streams draining agricultural and near-pristine catchments (forested and wetland) in the North German plains. To account for spatial and seasonal variation, we conducted a screening of DOM amount (53 sites) and composition (42 sites), and conducted bi-weekly samplings to investigate seasonal variation at eight sites over one year. Concentrations of dissolved organic carbon (DOC) were significantly higher for agricultural and wetland catchments than for forested catchments. Moreover, DOC loads exhibited higher seasonal variation for agricultural and wetland catchments than for forested catchments, which was due to higher variation in discharge. Parallel Factor Analysis revealed that the composition of DOM in agricultural catchments was significantly different from the other studied catchment types, and was characterized by low redox state and high structural complexity. Moreover, a gradient from protein- to humic-like fluorescence significantly separated forested from agricultural and wetland catchments. The contribution of humic-like DOM was strongly and positively related to DOC concentration, suggesting a mechanistic coupling of both. The effects of land use on patterns of DOC concentration and DOM composition were consistent across seasons, implying that land use strongly regulates DOM export. Overall, this study clearly shows the seasonally independent importance of agricultural land use for the amount and composition of DOM fluxes from the terrestrial zone to surface waters. These altered fluxes may affect ecosystem metabolism and health of agricultural headwaters and downstream situated aquatic ecosystems.
