International audience; Drought is the major constraint causing considerable yieldreduction in chickpea. Roots play a critical role in enhancingdrought tolerance. The main objective of the study was to introgressdrought tolerant root traits into Kenyan chickpea varietiesthrough marker assisted breeding (MABC). Eight simplesequence repeat (SSR) markers, linked to quantitative trait loci(QTL) for root and yield traits, were used to screen the parentsat ICRISAT, India. In addition, 1144 single nucleotide polymorphicmarkers (SNPs) were also used in genotyping theseparents at Legume Genomics Center, United Kingdom. Crosseswere made between two selected varieties, ICCV 92944 (ChaniaDesi II) and ICCV 00108 (LDT 068) and ‘QTL-hotspot’ donorparent ICC 4958 that has extensive rooting system. Polymorphic phic SSR and SNP markers were used to select progenies withroot QTL at F1, BC1F1 and BC2F1 that were later advanced toBC2F3. The BC2F3 populations were evaluated for root traitsat Egerton University in randomized complete block designwith two replications in pot experiment. The BC2F3 familieswere significantly different for root dry weight (RDW), shootdry weight (SDW), total plant dry weight (PDW) and root toshoot dry weight (R/S) ratio (R/S) for Chania Desi II x ICC 4958and R/S for LDT 068 x ICC 4958. These lineshad significantlyimproved root traits compared the recurrent parents. MABCis aneffective and efficient method of introgressing complexdrought tolerant traits which leads to improvement in yield especiallyunder drought conditions.

Abstract

International audience; Water scarcity associated with climate change particularly threatens the sustainability of viticulture in most cultivated, drought prone areas. Breeding grapevine for reduced water use and maintained production (that is high water-use efficiency) is therefore of major interest. This requires a comprehensive knowledge of the physiological impacts of drought which are the most influential on yield and quality. Special attention should be paid to those mechanisms involved in the regulation of water status in plant tissues as the primary parameter affected by drought. Transpiration rate, which has major influence on plant water status, together with water-use efficiency, therefore require special attention in breeding programs. To progress on the determinism of transpiration rate and water-use efficiency in grapevine, we used a F1 progeny made of 188 genotypes from a cross between two widespread cultivars, Syrah and Grenache, well-known for their contrasted water use. We showed the benefits of combining quantitative genetics (for QTL detection) and physiological experiments to study this population both in the vineyard and on potted plants. On the one hand, we developed an original experimental design in the field coupled to geostatistical modelling to take into account the spatial variability of soil water status inherent to vineyard conditions. This helped to identify significant genetic variability for the traits of interest. On the other hand, we combined powerful phenotyping tools on potted plants (high-throughput platform and controlled chambers) to control water deficit conditions and improve QTL detection. First, we found evidence that a dual physiological mechanism controls the decline of leaf water status under drought with a key role for plant hydraulic conductance beside that of stomatal control of transpiration. Contrasted combination of these two controls may lead to more or less efficient maintenance of leaf water status in response to soil drying (i.e. iso- or an isohydric behaviour). An indirect role of abscisic acid on stomatal conductance was also identified, mediated by the downregulation of leaf hydraulic conductance, with a genetic variability which correlated with genetic variation in iso- or aniso-hydric behaviour. We then revealed wide genetic variations in nocturnal transpiration, which correlated with variations in whole plant water-use efficiency (WUE), and identified corresponding genetic and physiological determinants. Lastly, we showed some consistency between QTLs detected for daytime WUE in pots and in the vineyard. Beyond the potential interest of the QTLs detected in this study for breeding prospects, this work demonstrated the interest of quantitative genetics to shed light on ecophysiological and physiological processes.

Leeway to improve WUE depends on how water losses are coupled with biomass gain. As water loss at night is not associated with any carbon gain, its reduction could be a good strategy to limit waste of water without altering photosynthesis rate in the daytime. However, genetic and physiological bases of variations in night transpiration are still unclear. The present study aimed at elucidating whether and how variations in night transpiration are genetically controlled in grapevine. A 2 year experiment was conducted on the F1 progeny from a cross between Syrah and Grenache using a phenotyping platform coupled to a controlled-environment chamber. Potted plants of the whole progeny were analysed for night transpiration, both in well-watered and water deficit conditions. A high genetic variability was found and 7 QTLs were detected, of which 3 were stable between years. These results evidenced for the first time the genetic control of water losses at night by identifying underlying locus. We then explored the physiological bases of this genetic variability, deciphering the respective roles of stomata and cuticle. 20 genotypes, chosen within the progeny for their contrasting transpirations at night, were studied in experiments with detached leaves. By feeding leaves with abscisic acid, we showed that stomata remain open at night, and that this residual opening differs among genotypes. We took advantage of the hypostomatic feature in grapevine to study cuticular conductance on the adaxial face. We found a significant genetic control of cuticular conductance, which accounted for up to 30% of the total leaf transpiration at night. These results open new avenues to breed grapevine for lower water losses at night, whose genetic variability appears to be driven both by stomata and cuticle.; Face à la raréfaction des ressources hydriques qui accompagne les changements climatiques, il devient crucial d'augmenter l'efficience d'utilisation de l'eau, notamment dans les systèmes viticoles en zone méditerranéenne. Abstract

Increasing water scarcity challenges crop sustainability in many regions. As a consequence, the enhancement of transpiration efficiency (TE)—that is, the biomass produced per unit of water transpired—has become crucial in breeding programs. This could be achieved by reducing plant transpiration through a better closure of the stomatal pores at the leaf surface. However, this strategy generally also lowers growth, as stomatal opening is necessary for the capture of atmospheric CO2 that feeds daytime photosynthesis. Here, we considered the reduction in transpiration rate at night (En) as a possible strategy to limit water use without altering growth. For this purpose, we carried out a genetic analysis for En and TE in grapevine, a major crop in drought-prone areas. Using recently developed phenotyping facilities, potted plants of a cross between Syrah and Grenache cultivars were screened for 2 y under well-watered and moderate soil water deficit scenarios. High genetic variability was found for En under both scenarios and was primarily associated with residual diffusion through the stomata. Five quantitative trait loci (QTLs) were detected that underlay genetic variability in En. Interestingly, four of them colocalized with QTLs for TE. Moreover, genotypes with favorable alleles on these common QTLs exhibited reduced En without altered growth. These results demonstrate the interest of breeding grapevine for lower water loss at night and pave the way to breeding other crops with this underexploited trait for higher TE.

Abstract

Modeling plant growth and development underwent considerable development with strong incentives from various consortia. It emerged as an efficient tool in ecology and genetics to face new challenges raised by competition for resources and to benefit from breakthroughs in biotechnology. In this presentation, we propose a classification of approaches used in modeling plants based on our experience at the Laboratoire d'Ecophysiologie des Plantes sous Stress Environnementaux (Montpellier, France). We present 5 types of models and discuss how they participate in a general workflow aimed at designing plant ideotypes for future climates.Type 1 are frameworks for spatiotemporal analysis of growth and development. They consist in quantitative description of when and where growth occurs in a specific organ, or how growth is synchronized between organs at whole plant level or, more generally, how development occurs. They are developed almost systematically when a novel species or organ are to be studied in the lab, as they are critical to set up experimental designs (what to measure, when, ...) or to reason the sampling of tissues for omics studies (which part of the organ should be taken, when).Type 2 are data-driven (statistical) representations of the responses to the environment and of determinant of growth. Often based on specialized statistical methods (multivariate analysis, structural equation models, mixed models,...), they allow us to formalize responses, reveal linkages and hierarchies between processes or multiple factors affecting growth, and demonstrate causalities between them. They are used as such to produce synthetic representations of growth processes or decipher between genetic and environmental effects. They are also used as bases for building other types of models.Type 3 are empirical models of response to environmental factors specially designed for parameterizations in high throughput phenotyping. This entails parsimony in the parameterization (hence in input variables), high heritability of parameters (traits) and compatibility with high throughput measurements. Such models have proved to be very efficient in genetic studies, for characterizing allelic diversity or finding QTLs. They also are ready to use and genetically well parameterized components for more integrative models.Type 4 are computer-assisted simulation models, used to make predictions a priori. They are based on hypothetical frames of integration of biological processes within the plant, and on physical models for the interactions with soil and atmospheric compartments. They allow simulation of complex system (cropmodels). They are used in the lab to simulate the behavior of plants in different climatic scenarios and thus to assess the relevance of a given set of traits for a given location. They are also produced to help reasoning agricultural practices, and especially irrigation.Type 5 are computer simulation models, used to reconstruct or interpret experiments a posteriori. Such models are designed to be fitted to experimental data, either to compute hidden variables (like sink strengths, radiation interception and efficiency), interpolate variables of interest from fragmental inputs in time or space (e.g. LAI time course) or interpret a macroscopic response by extracting meaningful parameters (e.g. hydraulic characteristics from difference in growth kinetics).During this presentation, new challenges are proposed to modelers in each type of models. They consist in bottlenecks which currently hamper plant scientists from achieving multiple goals in biology, ecology, agronomy or breeding.

Non disponible / Not available, Ce travail a permis de mettre au point i) dispositif de traitements lumineux en conditions contrôlées permettant de faire varier de manière indépendante le photoéquilibre phytochromique [phi]c, le rayonnement actif sur le cryptochrome Bleu, et le rayonnement actif, pour la photosynthèse PAR, et ii) une méthode de quantification des niveaux trophiques effectifs. Par ailleurs, nous avons développé une méthodologie originale de quantification et d'analyse de la colonisation de l'espace par la plante, basée sur la digitalisation 3D et le calcul de rayons de dispersion des noeuds et des extrémités des pétioles. Cette méthode permet de faire la part des contributions de la vitesse de développement, de la croissance et du port des espaceurs (entre-n?uds et pétioles) dans le déploiement 3D de la plante. Les résultats obtenus montrent que les traitements lumineux influencent tous les colonisations spatiales horizontale et verticale du trèfle blanc. Il est donc difficile d'assigner des rôles tranchés aux trois composantes du rayonnement. Mais on observe des nuances, ainsi l'effet de la lumière Bleu sur la colonisation semble s'expliquer essentiellement par son effet sur la croissance des pétioles. L'effet du niveau de PAR porte surtout sur la vitesse de développement global et sur la croissance des pétioles, alors que le phytochrome contrôle en plus le port des pétioles et la géométrie du réseau de stolons. Dans tous les cas, on assiste à la modulation plastique d'évolutions ontogénétiques qui sont conservées quel que soit le traitement. L'évolution du rayon de dispersion horizontale des extrémités des pétioles présente toujours une allure logarithmique en fonction du nombre total de phytomères, alors que le rayon de dispersion verticale présente une allure non monotone, avec une longue phase de décroissance...

Abstract

Phenotypic plasticity is considered as an important mechanism for plants to cope with environmental challenges. Leaf growth is one of the first macroscopic processes to be impacted by modification of soil water availability. In this study, we intended to analyze and compare plasticity at different scales. We examined the differential effect of water regime (optimal, moderate water deprivation and recovery) on growth and on the expression of candidate genes in leaves of different growth stages. Candidates were selected to assess components of growth response: abscisic acid signaling, water transport, cell wall modification and stomatal development signaling network. At the tree scale, the four studied poplar hybrids responded similarly to water regime. Meanwhile, leaf growth response was under genotype × environment interaction. Patterns of candidate gene expression enriched our knowledge about their functionality in poplars. For most candidates, transcript levels were strongly structured according to leaf growth performance while response to water regime was clearly dependent on genotype. The use of an index of plasticity revealed that the magnitude of the response was higher for gene expression than for macroscopic traits. In addition, the ranking of poplar genotypes for macroscopic traits well paralleled the one for gene expression. [ABSTRACT FROM AUTHOR]

Abstract

Grapevine production and wine quality are affected by climate change, and identifying varieties adapted to climate change becomes an urgent necessity. Searching for genotypes that are more water-efficient and able to maintain high photosynthesis in drought condition seems to be the key to maintain production while preserving vineyards on the long term. In order to evaluate the genetic variability for these traits, it is a key requirement to phenotype hundreds of genotypes in the vineyard. However, this is often hampered because conventional methods for measuring water and carbon related traits are typically expensive, destructive, and not usable at high-throughput. In this project, we aimed at developing and testing new high-throughput phenotyping methods, based on the use of NIRS (Near InfraRed Spectroscopy) and leaf chlorophyll fluorescence, for studying the genetic variability and determinism of functional traits in grapevine. We used two populations, one resulting from a semi-diallel cross (600 genotypes), the other corresponding to a diversity panel of 279 Vitis vinifera varieties. In this "calibration" phase, we combined, on a subset of genotypes, conventional low-throughput measurements of photosynthesis, stomatal conductance, leaf water potential, nitrogen and carbohydrates content, with fast leaf measurements using porometry/fluorimetry and NIRS. We used these datasets to calibrate models to predict the traits of interest from NIRS and porometry/fluorimetry, based on PLS (Partial Least Squares) regressions. In the next phase of the project, we are deploying the high-throughput methods alone on the whole populations, in order to predict the traits of interest using models established in the calibration phase, and to further analyse the genetic determinants of these traits with Genome Wide Association Study (GWAS). Abstract

Maintaining grapevine productivity with limited inputs is crucial in Mediterranean areas. Apart from water, nitrogen (N) is also an important limiting factor in grape growing. The effects of N deficiency on grapevine growth were investigated in this study. Two-year-old Vitis vinifera L.cv. Shiraz plants grafted on 110 R were grown in pots placed outside and exposed to various N supplies (0, 0.6, 1.2, 2.4 and 12g plant-1) under well-watered conditions. At veraison, plants were harvested and organs separately dried, weighed and analysed for N. During plant growth, the length of the primary and secondary axes and the number of leaves on them were recorded. The N content of leaves was also analysed at three phenological stages (flowering, bunch closure and veraison). All growth processes were inhibited by N deficiency in an intensity-dependent manner. Quantitative relationships with N supply were established. Vegetative growth responded negatively to N stress when comparing control N supply with no N supply: primary axis elongation (-61%), leaf emergence on the primary axis (-47%), leaf emergence on the secondary axis (-94%) and lamina area expansion (-45%). Significant differences on the plant N status were observed from flowering onwards which might be useful for managing fertilisation.
