In the study of rapidity distributions of charged pions produced in e e annihilation at 29 GeV, we observe a significant dip in the rapidity distributions at y≈0 for events with low sphericity. 21. Chemically Cross-Linked Polycyclooctene:  Synthesis, Characterization, and Shape Memory Behavior

Abstract

A novel polymeric shape memory system of chemically cross-linked polycyclooctene (PCO) was developed and characterized. PCO was synthesized via ring-opening metathesis polymerization of cyclooctene using the dihydroimidazolylidene-modified Grubb's catalyst. After dicumyl peroxide was added to PCO, the mixture was compression-molded into a film and further cured through chemical cross-linking upon heating. The chemically cross-linked PCO samples were fully characterized using differential scanning calorimetry (DSC), dynamic mechanical analysis (DMA), and wide-angle X-ray scattering (WAXS) in order to gain insight into the rapid shape memory behavior. We observe that the transition temperature of PCO is tunable through the change of the trans/cis ratio of vinylene groups. A fast shape memory behavior was observed, where the primary stress-free shape was recovered within 1 s on immersion in hot water above the melting point of the crystalline PCO phase. In contrast with glassy shape memory polymers, chemically cross-linked PCO behaves as an elastomer capable of arbitrary shaping above the sharp melting temperature of the PCO crystalline phase and subsequent shape fixing during crystallization.

23. Morphology of Model A/B/(C-block-D) Ternary Blends and Compatibilization of Two Immiscible Homopolymers A and B with a C-block-D Copolymer

Abstract

The morphology of A/B/(C-block-D) ternary blends was investigated by transmission electron microscopy (TEM), and its implications in the compatibilization of two immiscible homopolymers A and B with a C-block-D copolymer are discussed. Emphasis is placed on the thermodynamic requirements and the role of order−disorder transition temperature (TODT) of block copolymer for compatibilization. For the investigation, the following model ternary blends were prepared:  (i) 63/27/10 PS/PB/(PαMS-block-PI) with PS denoting polystyrene, PB denoting polybutadiene, PαMS denoting poly(α-methylstyrene), and PI denoting polyisoprene; (ii) 63/27/10 PPO/PP/(PS-block-PEB) with PPO denoting poly(phenylene oxide), PP denoting polypropylene, and PEB denoting poly(ethylene-co-1-butene); and (iii) 63/27/10 PMMA/PP/(PαMS-block-PI) with PMMA denoting poly(methyl methacrylate). Blend specimens were prepared by melt blending in a Mini-Max mixer at temperatures above and below the TODT of block copolymer. The TEM images of ternary blends showed a uniform distribution of (i) PαMS-block-PI layer on the surface of PB droplets dispersed in the PS matrix and (ii) PS-block-PEB layer on the surface of PP droplets dispersed in the PPO matrix only when melt blending was carried out at a temperature above the TODT of block copolymer. The formation of a uniform interphase between two immiscible homopolymers is attributed to the attractive thermodynamic interactions existing (i) between homopolymer PB and PI block in the 63/27/10 PS/PB/(PαMS-block-PI) ternary blend and (ii) between homopolymer PPO and PS block and between homopolymer PP and PEB block in the 63/27/10 PPO/PP/(PS-block-PEB) ternary blend. The present study indicates that the TODT of block copolymer, in relation to melt blending temperature, determines whether the block copolymer can play the role of an effective compatibilizing agent even when thermodynamic requirements (attractive interactions) for compatibilization are satisfied. Thus, we have concluded that attractive thermodynamic interactions alone are not sufficient for compatibilization of two immiscible homopolymers unless the TODT of block copolymer is lower than melt blending temperature, because the viscosity of block copolymer in an ordered state (at T < TODT) is a few orders of magnitude higher than that in the disordered state (at T > TODT). That is, the mobility of block copolymer plays an important role in the compatibilization of two immiscible homopolymers.

24. The Role of the Order−Disorder Transition Temperature of Block Copolymer in the Compatibilization of Two Immiscible Homopolymers

Abstract

The important role that the order−disorder transition temperature (TODT) of a block copolymer plays in the compatibilization of two immiscible homopolymers is demonstrated, using the model ternary blend systems consisting of a polystyrene-block-polybutadiene (SB diblock) copolymer and two immiscible homopolymers, polystyrene (hPS) and polyisoprene (hPI). For the study, SB diblock copolymers having different microstructures were employed. We investigated via transmission electron microscopy (TEM) the morphology of the blends. We found that an SB diblock copolymer was very poorly distributed at the interface between hPS and hPI in an hPS/hPI/SB ternary blend when the specimen was annealed at a temperature below the TODT of the block copolymer, while a more uniform distribution of the SB diblock copolymer was observed when a specimen was annealed at a temperature above its TODT. We have shown that the miscibility (or the interaction parameter) between the hPI and PB block in an SB diblock copolymer plays a decisive role in controlling the morphology at the interfaces between hPS and hPI. We conclude that a block copolymer must be designed, such that its TODT is below the targeted melt blending temperature, in order for the block copolymer to be able to act as an effective compatibilizing agent for two immiscible homopolymers. This conclusion is supported further by investigating the tensile properties and morphology of ternary blends consisting of polypropylene (PP), hPS, and polystyrene-block-poly(ethylene-co-1-butene)-block-polystyrene (SEBS triblock) copolymer (Kraton G1650), which were prepared by melt blending at 200 °C in a batch mixer. That is, little improvement in the tensile properties of the ternary blends was observed when Kraton G1650 was added to PP/hPS binary blends. This observation is explained by a very poor distribution, observed by TEM, of Kraton G1650 at the interface between PP and hPS in the ternary blend. This is attributed to the very high TODT, estimated to be above 350 °C from currently held mean-field theory, of Kraton G1650 compared to the melt blending temperature employed.

Abstract

The effect of the microstructure on the phase behavior of mixtures of polybutadiene (PB) and poly(ethylene-co-1-butene) (PEB) with polystyrene (PS) has been investigated. A series of PBs with 1,2-addition content ranging from 7 to 93% were synthesized by anionic polymerization, and a portion of each was subsequently hydrogenated to yield PEB. Polymer pairs with blend compositions from 10 to 90 wt % were cast from toluene for each of the 16 PS/PB pairs and 7 PS/PEB pairs. Laser light scattering was used to obtain cloud point measurements, which were then used to construct phase diagrams. It was found that, for constituent components with equivalent degrees of polymerization, PS/PEB pairs give rise to higher upper critical solution temperatures than PS/PB pairs, indicating that PS/PEB pairs are less miscible than PS/PB pairs. Experimental phase diagrams were curve-fitted to theoretical phase diagrams predicted from the Flory−Huggins theory with the expression for the interaction parameter α:  α = a + b/T + cφPS/T, where α is related to the Flory−Huggins interaction parameter χ by χ = αVr, where Vr is the molar reference volume, T is the absolute temperature, and φPS is the volume fraction of PS in the mixture. α values for PS/PB mixtures increase with increasing 1,2-addition (miscibility decreases) while α values for PS/PEB mixtures decrease (miscibility increases) with increasing 1-butene content. Using these α values, the Helfand−Wasserman theory was applied to predict the order−disorder transition temperatures of PS-block-PB and PS-block-PEB copolymers with varying 1,2-addition and 1-butene content, respectively.

27. EVALUATION OF THE LONGITUDINAL COUPLED VIBRATIONS IN ROTATING, FLEXIBLE DISKS/SPINDLE SYSTEMS

Abstract

The natural frequencies and modes of the longitudinal coupled vibrations in a flexible shaft with multiple flexible disks are determined through a substructure synthesis technique. The analysis rotor model consists of multiple flexible disks attached to a flexible shaft with varying annular cross-section, as used for steam turbines or computer storage devices. In modelling the system, disk flexibility and centrifugal stiffening effects are taken into account. Commercial computer hard disk drive spindle systems are selected as illustrative examples. In particular, the effects of disk flexibility on the longitudinal coupled vibrations between the shaft and disks are investigated with varying spindle rotational speed. The results indicate that the substructure synthesis technique allows a clear physical insight into the effect of disk flexibility with reasonable computational effort.

Abstract

IDA derivatives of three substituted benzothiazol, and two substituted chlorophenyl and one substituted pyrazoline compounds have been labeled with 99mTc and screened with four rat models with hepatocellular dysfunction manifesting varying degrees of change of liver architecture and hepatocellular damage associated with an active parenchymal destruction, fatty metamorphosis and cirrhosis. Organ distribution studies at 1 h postinjection have been compared in normal and diseased animal models for each agent labeled with 99mTc and with 99mTc-Disofenin (Disida) and Lidofenin (Hida) and 131I-Rose Bengal. From the data obtained with the six new IDA derivatives, the distribution kinetics of 99mTc-Arclophenin, (N-N'-2-benzoyl-4-chlorophenyl)carbamoylmethyl) imino diacetic acid (Phenida), are closely comparable to 99mTc-Disofenin in all animal models.

Abstract

In this study, we have investigated the mechanisms underlying organ susceptibility to candida infection. Infection of BALB/c mice with Candida albicans led to both an early (1-8 h) and late (24-48 h) activation of NF-kappaB in the organs resistant to C. albicans, including the lung and spleen. In susceptible organs such as the kidneys, early activation of NF-kappaB was not observed. The kinetics of TNF-alpha mRNA expression paralleled those of NF-kappaB activation in all organs examined. Blocking the effects of endogenous platelet-activating factor (PAF) by pretreatment with the PAF antagonist BN50739 or antioxidants significantly reduced the early activity of NF-kappaB and TNF-alpha mRNA expression, and increased the recovery of C. albicans in the lung and spleen. Importantly, administration of PAF 5 min prior to the infection resulted in the appearance of early activities of NF-kappaB and TNF-alpha mRNA expression, followed by a nearly complete clearance of the organisms in the kidneys. Pretreatment with anti-TNF-alpha Ab resulted in an enhanced susceptibility to C. albicans, and the PAF-mediated resistance was abrogated by anti-TNF-alpha in all organs examined. These data indicated that endogenously produced PAF in response to C. albicans is a key molecule involved in the early activation of NF-kappaB, which, in turn, renders the organ resistant to the fungus by promoting the production of anti-candidal proinflammatory cytokines such as TNF-alpha. Susceptible organs, including the kidneys, lack the capacity to generate a sufficient PAF-induced early NF-kappaB response.

Abstract

Epidemiological studies have demonstrated that nonsteroidal anti-inflammatory drugs (NSAIDs) decrease the incidence of and mortality from colon cancer. In addition, NSAIDs reduce the number and the size of polyps in patients with familial adenomatous polyposis. The mechanisms responsible for the antineoplastic effect of NSAIDs are not yet completely understood, but one of the possible mechanisms is an induction of apoptosis. We explored the role of caspase-3, a major apoptosis-executing enzyme, in NSAID-induced apoptosis of colon cancer cell line HT-29. Treatment of HT-29 cells with indomethacin induced a dramatic increase in caspase-3-like protease activity measured by a cleavage of the fluorogenic substrate Ac-DEVD-AMC. Western blot analysis showed that indomethacin treatment led both to decrease in procaspase-3 and to cleavage of its substrate poly(ADP-ribose) polymerase (PARP). Furthermore, the caspase-3-like protease inhibitor Ac-DEVD-CHO attenuated indomethacin-induced DNA fragmentation dose dependently. However, mRNA expression of CASP genes was not affected by the addition of indomethacin, highlighting the importance of posttranslational modification of this enzyme for the activation. These results suggest that NSAIDs, including indomethacin, induce apoptosis in colon cancer cells through a caspase-3 dependent mechanism which may contribute to the chemopreventive functions of these agents.

Abstract

Tumor necrosis factor (TNF)-alpha and platelet-activating factor (PAF) are important mediators of inflammatory reactions, and their release is controlled by a positive feedback network. However, the regulatory mechanisms underlying the interaction of these two molecules are unknown. Within 10 min of the injection of lipopolysaccharide (LPS) into C57BL/6 mice, effects inducible by PAF such as anaphylactic shock-like symptoms, disseminated intravascular coagulation, and hemorrhage in renal medullae were observed, and all these pathological changes were prevented by the PAF antagonist, BN 50739. The plasma level of PAF after LPS injection reached a peak at 5 min. TNF-alpha gene expression was evident 20 min after LPS injection and was maximal at 40 min, and the level of serum TNF-alpha reached a peak at 1 h. Pretreatment with BN 50739 inhibited LPS-induced TNF-alpha gene expression and protein synthesis in a dose-dependent manner. Injection of PAF or treatment of the macrophage cell line, J774A.1, with PAF activated the transcription factor, nuclear factor (NF)-kappa B, which is essential for inducible TNF-alpha transcription. The activation of NF-kappa B by PAF preceded the LPS-mediated TNF-alpha gene expression. Pretreatment with BN 50739 inhibited LPS-induced mobilization of NF-kappa B in a dose-dependent manner in vivo as well as in vitro. These data suggest that PAF, which is released immediately or shortly after LPS injection, induces the expression of TNF-alpha through the activation of NF-kappa B.

Abstract

The effect of platelet-activating factor (PAF) on experimental pulmonary metastasis by the B16F10 murine melanoma and the possible involvement of PAF in the activities of tumor necrosis factor alpha (TNF-alpha) and interleukin 1alpha (IL-1alpha) in tumor metastasis were investigated. i.p. injection of PAF enhanced the lung colonization in a dose- and time-dependent manner. PAF enhanced lung colonization when it was administered after, but not before, B16F10 inoculation. Multiple injections of PAF were more effective than a single injection. Neutralization of endogenous PAF with PAF antagonist BN50739 decreased lung colonization, suggesting that endogenous PAF plays an important role in pulmonary metastases. A single i.p. injection of TNF-alpha or IL-1alpha caused a marked enhancement in lung colonization. TNF-alpha- and IL-1alpha-mediated enhancement in lung colonies was significantly inhibited by BN50739. These results demonstrate that PAF has a metastasis-enhancing effect and is a mediator of the metastatic activities of TNF-alpha and IL-1alpha.

Abstract

The Schwanniomyces occidentalis (formerly castellii) ATCC 26077 (CBS 2863) alpha-amylase (AMY 26077) gene was cloned in Saccharomyces cerevisiae and sequenced. An open-reading frame encoding the AMY consists of 1536 base pairs and contains 512 amino-acid residues, which is almost the same in size as the AMY of Sch. occidentalis ATCC 26076 and CCRC 21164. The amino-acid sequence of AMY 26077 differed from that of ATCC 26076 alpha-amylase (AMY 26076) at two residues and from that of CCRC 21164 alpha-amylase (AMY 21164) at three residues. Comparison of the AMY 26077 gene with its homologues from two other strains (Sch. alluvius CBS 1153 and Sch. persoonii CBS 2169) using several restriction enzymes revealed that the AMY 26077 was very similar to AMY CBS 1153 but different from that of CBS 2169.

Abstract

A number of p-aminohippuric acid analogues were synthesized in order to develop clinically useful 99mTc-labeled radiopharmaceuticals for evaluation of renal function measurements. Stable 99mTc-labeled complexes were formed at pH 5.7 using a Sn(II) reduction method with all derivatives. The newly synthesized complexes were screened utilizing biodistribution studies in small animals. All complexes were excreted via the GU tract within 60 min post iv administration, with no significant activity in GI tract and liver. The [99mTc]methyl-PAHIDA complex showed optimal biodistribution among these analogues. Further investigation is needed to determine if these derivatives may be used to replace [131I]o-iodohippuric acid for the evaluation of renal function.
