The complex interplay between genetic and environmental factors is considered the cause of neurodegenerative diseases including Parkinson’s disease (PD) and Amyotrophic Lateral Sclerosis (ALS). Among the environmental factors, toxins produced by cyanobacteria have received much attention due to the significant increase in cyanobacteria growth worldwide. In particular, L-BMAA toxin, produced by diverse taxa of cyanobacteria, dinoflagellates and diatoms, has been extensively correlated to neurodegeneration. The molecular mechanism of L-BMAA neurotoxicity is still cryptic and far from being understood. In this research article, we have investigated the molecular pathways altered by L-BMAA exposure in cell systems, highlighting a significant increase in specific stress pathways and an impairment in autophagic processes. Interestingly, these changes lead to the accumulation of both α-synuclein and TDP43, which are correlated with PD and ALS proteinopathy, respectively. Finally, we were able to demonstrate specific alterations of TDP43 WT or pathological mutants with respect to protein accumulation, aggregation and cytoplasmic translocation, some of the typical features of both sporadic and familial ALS.

Pathological mutations in the LRRK2 gene are the major genetic cause of Parkinson’s disease (PD). Although several animal models with either LRRK2 down- or over-expression have been developed, the physiological function of LRRK2 remains elusive. LRRK2 is constitutively expressed in various tissues including neurons and glial cells, but importantly, it is expressed at low levels in dopaminergic neurons, further contributing to the cryptic function of LRRK2. Significant levels of LRRK2 protein and mRNA have been detected in peripheral blood mononuclear cells, lymph nodes, the spleen, and primary microglia, strongly suggesting the contribution of inflammatory cells to neuronal degeneration. In this research article, using Drosophila LRRK2 models, we were able to demonstrate a significant contribution of glial cells to the LRRK2 pathological phenotype. Furthermore, in Drosophila, neurodegeneration is associated with a significant and important increase in specific inflammatory peptides. Finally, levetiracetam, a compound widely used in human therapy to treat epilepsy, was able to rescue both neuronal degeneration and neuroinflammation.

Thromboembolic conditions are the most common cause of death in developed countries. Anticoagulant therapy is the treatment of choice, and heparinoids and warfarin are the most adopted drugs. Sulphated polysaccharides extracted from marine organisms have been demonstrated to be effective alternatives, blocking thrombus formation by inhibiting some factors involved in the coagulation cascade. In this study, four acidic glycan fractions from the marine sponge Sarcotragus spinosulus were purified by anion-exchange chromatography, and their anticoagulant properties were investigated through APTT and PT assays and compared with both standard glycosaminoglycans and holothurian sulphated polysaccharides. Moreover, their topographic localization was assessed through histological analysis, and their cytocompatibility was tested on a human fibroblast cell line. A positive correlation between the amount of acid glycans and the inhibitory effect towards both the intrinsic and extrinsic coagulation pathways was observed. The most effective anticoagulant activity was shown by a highly charged fraction, which accounted for almost half (about 40%) of the total hexuronate-containing polysaccharides. Its preliminary structural characterization, performed through infrared spectroscopy and nuclear magnetic resonance, suggested that it may consist of a fucosylated chondroitin sulphate, whose unique structure may be responsible for the anticoagulant activity reported herein for the first time.

The white-clawed crayfish Austropotamobius pallipes complex populations are decreasing in the Foreste Casentinesi, Monte Falterona and Campigna National Park (Central Italy), due to several factors, including illegal poaching, predatory fishes, drought, and invasive alien species. Recently, the Northern raccoon Procyon lotor has been reported to be present in the area of the National Park and has started to predate on the white-clawed crayfish. The aim of the study was to update the distribution and population status of A. pallipes in the reserves, other sites of the National Park, and surrounding areas to assess the potential effects of the raccoon. Crayfish were sampled by hand or by traps in 14 sites; sampled individuals were sexed and measured. Signs of raccoon presence (e.g., footprints and predated crayfish) were also recorded. Our study confirms the impact of raccoon on native crayfish: indeed, where the invasive mammal is present (six sites), crayfish disappeared, or their populations have been dramatically reduced in number, with a size distribution skewed towards juveniles. In two sites, close to urban settlements, fresh footprints of P. lotor and predated specimens of A. pallipes were also observed. Populations of crayfish are still abundant or even increasing as compared with samplings conducted in the past where raccoon is absent (five sites). Urgent actions (e.g., control of raccoons, and monitoring and restocking of A. pallipes populations if feasible and where possible) should be taken into account to guarantee the survival of this protected species.

Mountain wetlands are among the most vulnerable habitats in the Mediterranean basin. Their conservation requires knowledge of plant species assemblages and their environmental drivers. In this study, we investigated what the main environmental factors driving species composition in mountain wetlands are. Differences in environmental control and floristic composition between palustrine and lacustrine wetlands were explored. We used a dataset of 168 vegetation plots (relevés), sampled at 45 mountain wetlands in the northern Apennines (central Italy). Direct ordination showed that water depth, geology type and altitude were the main factors responsible for species distribution. The most important gradient was linked to soil moisture, with hygrophilous species increasing with moisture levels. Indicator Species Analysis underlined a clear distinction in the distribution of aquatic plants between wetland subsystems. Geology and rainfall affected species assemblages in lacustrine and palustrine subsystems. Indirect ordination and Generalized Additive Models revealed that plant species and their attributes significantly changed in the wetland subsystems with an increase in hydrophytes with increasing rainfall in palustrine wetlands and a decrease in thermophilous species along an altitudinal gradient in lacustrine wetlands. Management and conservation guidelines for northern Apennines wetlands are suggested.

This workshop summary on natural language processing (NLP) markers for psychosis and other psychiatric disorders presents some of the clinical and research issues that NLP markers might address and some of the activities needed to move in that direction. We propose that the optimal development of NLP markers would occur in the context of research efforts to map out the underlying mechanisms of psychosis and other disorders. In this workshop, we identified some of the challenges to be addressed in developing and implementing NLP markers-based Clinical Decision Support Systems (CDSSs) in psychiatric practice, especially with respect to psychosis. Of note, a CDSS is meant to enhance decision-making by clinicians by providing additional relevant information primarily through software (although CDSSs are not without risks). In psychiatry, a field that relies on subjective clinical ratings that condense rich temporal behavioral information, the inclusion of computational quantitative NLP markers can plausibly lead to operationalized decision models in place of idiosyncratic ones, although ethical issues must always be paramount. Six hundred and thirty individual serum samples from dairy sheep were tested for the presence of antibodies to Toxoplasma gondii using an indirect immunofluorescence antibody test (IFAT). The sampled animals came from 33 dairy herds representative of the southern area of the Tuscany region. Questionnaires exploring the management system were filled in by the veterinarian in charge of the herds. Using a cut-off of 1:64, 214 animals (33.97%) were positive with a herd prevalence of 96.97%. Using a sensitivity and specificity of 80% and 95% respectively, the true prevalence was calculated as 38.62% (CI, 34.82%, 42.43%). Abstract Background Serum samples from 630 milk sheep, in 33 dairy flocks representative of the southern area of the Tuscany region, were tested for the presence of antibodies to Toxoplasma gondii using an indirect immunofluorescence antibody test (IFAT). Questionnaires exploring the management system were completed by the veterinarian in charge of the flocks. Results At least one seropositive animal was found in 32 of the 33 flocks tested (97.0%; 95% CI: 84.2%, 99.9%). In the positive flocks, median seroprevalence was 29.4% (interquartile range: 15.9%-46.1%). Overall animal-level seroprevalence, adjusted for sampling weights and test sensitivity and specificity, was 33.3% (95% CI: 24.8%, 42.7%). In a multivariable negative binomial regression model the number of seropositive animals in a flock decreased with increasing flock size (for >400 vs. CR) = 0.62; 95% CI: 0.41, 0.95; P = 0.028) and was greater on farms where stray cats had access to animals’ water (CR = 1.54; 95% CI: 1.05, 2.26; P = 0.027). Conclusions Small flock size and access of cats to water are potential risk factors for Toxoplasma infection in sheep in the Grosseto district in Tuscany, Italy. Sheep could be an important source of T. gondii infection in humans, since we estimate that between 25% and 43% of sheep in the district were seropositive. Toxoplasmosis is also likely to be an important cause of abortion in sheep in the district. Control and prophylactic measures must be adopted to improve the rearing system and the implementation of health promoting programmes in a joint effort between sheep farmers, farmers’ associations and veterinarians to inform about the means of transmission of the infection and for a better understanding of the disease.
