1. Obesity as risk factor for subtypes of breast cancer: results from a prospective cohort study

Abstract

Abstract Background Earlier epidemiological studies indicate that associations between obesity and breast cancer risk may not only depend on menopausal status and use of exogenous hormones, but might also differ by tumor subtype. Here, we evaluated whether obesity is differentially associated with the risk of breast tumor subtypes, as defined by 6 immunohistochemical markers (ER, PR, HER2, Ki67, Bcl-2 and p53, separately and combined), in the prospective EPIC-Germany Study (n = 27,012). Methods Formalin-fixed and paraffin-embedded (FFPE) tumor tissues of 657 incident breast cancer cases were used for histopathological analyses. Associations between BMI and breast cancer risk across subtypes were evaluated by multivariable Cox regression models stratified by menopausal status and hormone therapy (HT) use. Results Among postmenopausal non-users of HT, higher BMI was significantly associated with an increased risk of less aggressive, i.e. ER+, PR+, HER2-, Ki67low, Bcl-2+ and p53- tumors (HR per 5 kg/m2: 1.44 [1.10, 1.90], p = 0.009), but not with risk of more aggressive tumor subtypes. Among postmenopausal users of HT, BMI was significantly inversely associated with less aggressive tumors (HR per 5 kg/m2: 0.68 [0.50, 0.94], p = 0.018). Finally, among pre- and perimenopausal women, Cox regression models did not reveal significant linear associations between BMI and risk of any tumor subtype, although analyses by BMI tertiles showed a significantly lower risk of less aggressive tumors for women in the highest tertile (HR: 0.55 [0.33, 0.93]). Conclusion Overall, our results suggest that obesity is related to risk of breast tumors with lower aggressiveness, a finding that requires replication in larger-scale analyses of pooled prospective data.

9. Toward preresonant impulsive Raman preparation of large amplitude vibrational motion.

Abstract

This article investigates a new approach to the optical generation of large-amplitude coherent molecular vibrations in condensed media. On the basis of analytical results using pulse propagators in the classical Franck approximation, we are led to investigate the efficacy of driving vibrational motion in the electronic ground state by impulsive stimulated Raman scattering with a timed sequence of electronically preresonant femtosecond laser pulses. Numerically exact computations are performed on a model system of dilute molecular Iodine in a low-temperature host crystal. Vibrational relaxation is incorporated via Redfield theory. The results indicate that under a variety of conditions, chemically significant (greater than 0.1 Å) displacements can be produced in a Raman active mode with a fair measure of control over wave packet spreading, and without substantial population loss due to electronic absorption. © 1996 American Institute of Physics. Abstract

The nonlinear effects of ultrashort phase-locked electronically resonant pulse pairs on the ground state nuclear motion are investigated theoretically. The pulse-pair propagator, momentum impulse, and displacement are determined in the weak field limit for pulse pairs separated by a time delay short on a nuclear time scale. Possible application to large amplitude vibrational excitation of the 104 cm-1 mode of α-perylene is considered and comparisons are made to other Raman excitation methods. [ABSTRACT FROM AUTHOR]

Abstract

The recently developed technique of time-resolved spectroscopy with phase-locked optical pulse pairs is further explored with additional experimental data and more detailed comparison to theory. This spectroscopic method is sensitive to the overall phase evolution of an optically prepared nuclear wave packet. The phase locking scheme, demonstrated for the B←X transition of gas phase molecular iodine, is extended through the use of in-quadrature locked pulses and by examination of the dispersed fluorescence signal. The excited state population following the interaction with both pulses is detected as the resultant two-field-dependent fluorescence emission from the B state. The observed signals have periodically recurring features that result from rovibrational wave packet dynamics of the molecule on the excited state electronic potential energy curve. Quantum interference effects cause the magnitude and sign of the periodic features to be strongly modulated. The two-pulse phase-locked interferograms are interpreted with first order time-dependent perturbation theory. Excellent agreement is found between the experimental interferograms and those calculated from literature values of the parameters governing the electronic, vibrational and rotational structure of I2. A relationship between the phase-locked interferograms and the time-dependent linear susceptibility is obtained. The in-phase and in-quadrature phase-locked interferograms together provide a complete record of the optical free induction decay. Thus by combining the in-phase and in-quadrature data, we obtain the contributions to both the absorptive and dispersive linear susceptibilities arising from transitions within the pulse spectrum. 20. The Relaxation Dynamics and Short-Time Optical Response of a Multimode Open System

Abstract

When dividing a large system into a subsystem and a bath, one sometimes needs to include multiple nuclear coordinates in the subsystem in order to treat the remaining bath modes by Markovian relaxation theory (which assumes fast bath relaxation). This paper examines the effects of Redfield relaxation on the time-resolved fluorescence signal and on the loss of energy from a three-mode system. Simulations are compared for system coordinates coupled to the same bath mode and to independent bath modes, for coupling linear and quadratic in the system coordinates, and for different temperatures. To make these comparisons meaningful, a criterion is proposed for normalization of the coupling strengths in the different cases. The fluorescence Stokes shift and the system energy are shown to be sensitive to different relaxation processes. The coupling of multiple system coordinates to the same bath mode results in a wide range of relaxation rates. Simple descriptions in terms of individual population relaxation and coherence dephasing rates are inadequate due to sequential processes and to coupling between populations and coherences. These results have implications not only for Redfield treatments but also for other relaxation theories such as the Brownian oscillator model.

Abstract

Background & Aims: Despite the thorough mapping of brain pathways involved in eating behavior, no treatment aimed at modulating eating dysregulation from its neurocognitive root has been established yet. We aimed to evaluate the effect of N.I.R. H.E.G. (Near Infra-Red Hemoencephalography) neurofeedback training on appetite control, weight and food-related brain activity., Methods: Six healthy male participants with overweight or mild obesity went through 10 N.I.R. H.E.G. neurofeedback sessions designed to practice voluntary activation of the prefrontal cortex. Weight, eating behavior, appetite control and brain activity related to food and self-inhibition based on fMRI were evaluated before and after neurofeedback training., Results: Our study group demonstrated a positive trend of increased self-control and inhibition related to food behavior, reduced weight and increased activation during an fMRI response-inhibition task (Go-No-Go - GNG) in the predefined region of interest (ROI): superior orbitofrontal cortex (sOFC)., Conclusions: N.I.R. H.E.G. holds a promising potential as a feasible neurofeedback platform for modulation of cortical brain circuits involved in self-control and eating behavior and should be further evaluated and developed as a brain modifying device for the treatment and prevention of obesity., (Copyright © 2018 Elsevier Ltd and European Society for Clinical Nutrition and Metabolism. Abstract

The difference boundary sedimentation velocity technique of Schachman and co-workers is demonstrated to be applicalbe to the measurement of binding constants (Kobsd) in the range 10(2)-10(5) M(-1) for the nonspecific interactions of proteins with DNA. The difference technique can reproducibly detect a 2% change in the sedimentation coefficient of the DNA upon binding ligands, corresponding to average extents of association as low as 10 molecules of protein (in the cases of Escherichia coli lac repressor and E. coli RNA polymerase) per molecule of bacteriophage T7 DNA. At these low binding densities, it is plausible to assume that the primary effect of ligand binding is on the buoyant mass of the complex and not on the frictional coefficient of the flexible DNA coil. Binding constants calculated by using this assumption agree well with literature values for the nonspecific interactions of RNase and lac repressor proteins with double-stranded DNA. Advantages of the method are that it is relatively rapid, requires the optical detection of the DNA only, and can be performed on small amounts of sample. The method appears useful for surveying (to an accuracy of +/-50% in Kobsd or +/-10% in log Kobsd) the effects of solution variables on Kobsd of protein-DNA interactions. Applications of the method to the nonspecific interactions of RNA polymerase core and holoenzymes with T7 DNA are discussed.
