In insect-pollinated plants, the foraging behavior of pollinators affects their pattern of movement. If distinct bee species vary in their foraging behaviors, different models may best describe their movement. In this study, we quantified and compared the fine scale movement of three bee species foraging on patches of Medicago sativa. Bee movement was described using distances and directions traveled between consecutive racemes. Bumble bees and honey bees traveled shorter distances after visiting many flowers on a raceme, while the distance traveled by leafcutting bees was independent of flower number. Transition matrices and vectors were calculated for bumble bees and honey bees to reflect their directionality of movement within foraging bouts; leafcutting bees were as likely to move in any direction. Bee species varied in their foraging behaviors, and for each bee species, we tested four movement models that differed in how distances and directions were selected, and identified the model that best explained the movement data. Premise: Pollinator foraging behavior can influence pollen dispersal and gene flow. In many plant species a pollinator trips a flower by applying pressure to release its sexual organs. We propose that differences in tripping rate among grooming pollinators could generate distinct pollen deposition curves, the pattern of pollen deposition over successive flowers visited. This study compares the pollen deposition curves of two grooming pollinators, a social bumble bee and a solitary leafcutting bee, with distinct tripping rates on Medicago sativa flowers. We predict a steeper deposition curve for pollen moved by leafcutting bees, the pollinator with the higher tripping rate., Methods: Medicago sativa plants carrying a gene (GUS) whose product is easily detected by staining, were used as pollen donors. After visiting the GUS plants, a bee was released on a linear array of conventional M. sativa plants. The number of GUS pollen grains deposited over successive flowers visited or over cumulative distances was examined. Distinct mixed effect Poisson regression models, illustrating different rates of decay in pollen deposition, were fitted to the pollen data for each bee species., Results: Pollen decay was steeper for leafcutting bees relative to bumble bees for both models of flowers visited and cumulative distance, as predicted by their higher tripping rate., Conclusions: This is the first report of a difference in pollen deposition curves between two bee species, both grooming pollinators. Pollen and nectar resources are unevenly distributed over space and bees must make routing decisions when navigating patchy resources. Determining the patch selection process used by bees is crucial to understanding bee foraging over discontinuous landscapes. To elucidate this process, we developed four distinct probability models of bee movement where the size and the distance to the patch determined the attractiveness of a patch. A field experiment with a center patch and four peripheral patches of two distinct sizes and distances from the center was set up in two configurations. Empirical transition probabilities from the center to each peripheral patch were obtained at two sites and two years. The best model was identified by comparing observed and predicted transition probabilities, where predicted values were obtained by incorporating the spatial dimensions of the field experiment into each model's mathematical expression. Bumble bees used both patch size and isolation distance when selecting a patch and could assess the total amount of resources available in a patch. Bumble bees prefer large, nearby patches. This information will facilitate the development of a predictive framework to the study of bee movement and of models that predict the movement of genetically engineered pollen in bee-pollinated crops.

Identifying the factors that determine habitat suitability and hence patterns of wildlife abundances over broad spatial scales is important for conservation. Ecosystem productivity is a key aspect of habitat suitability, especially for large mammals. Our goals were to a) explain patterns of moose (Alces alces) abundance across Russia based on remotely sensed measures of vegetation productivity using Dynamic Habitat Indices (DHIs), and b) examine if patterns of moose abundance and productivity differed before and after the collapse of the Soviet Union. We evaluated the utility of the DHIs using multiple regression models predicting moose abundance by administrative regions. Univariate models of the individual DHIs had lower predictive power than all three combined. The three DHIs together with environmental variables, explained 79% of variation in moose abundance. Interestingly, the predictive power of the models was highest for the 1980s, and decreased for the two subsequent decades. We speculate that the lower predictive power of our environmental variables in the later decades may be due to increasing human influence on moose densities. Overall, we were able to explain patterns in moose abundance in Russia well, which can inform wildlife managers on the long-term patterns of habitat use of the species.

Pollen viability affects the probability that a pollen grain deposited on a plant's stigma will produce a viable seed. Because a mature seed is needed before a gene flow event can occur, pollen viability will influence the risk of escape for genetically engineered (GE) crops. Pollen viability was measured at intervals for up to 2 h following removal of the pollen from the anthers. It was quantified at three temperatures and for different alfalfa varieties, including both conventional and Roundup Ready (RR) varieties. Pollen viability was assessed using in vitro germination. Time since removal from the anthers was the most prevalent factor affecting pollen viability in alfalfa. Pollen viability declined with increasing time at all three temperatures and for all varieties tested. Pollen viability was not affected by temperatures ranging between 25 and 37 °C and did not vary among plant varieties, including conventional and RR varieties. Bee foraging behaviour suggested pollen viability within the first 10 min following pollen removal from a flower to most affect seed production. Pollen longevity was predicted to have little impact on seed set and gene flow. Foraging behaviors that impact gene flow can guide the design of pollinator strategies to mitigate gene flow. Reduced gene flow is expected to minimize the impact of genetically engineered (GE) crops on feral and natural populations and to facilitate the coexistence of different agricultural markets. The goal of this study is to link foraging behavior to gene flow and identify behaviors that can help predict gene flow for different bee species. To reach this goal, we first examined and compared the foraging behaviors of three distinct bee species, the European honey bee, Apis mellifera L., the common eastern bumble bee, Bombus impatiens Cr., and the alfalfa leafcutting bee, Megachile rotundata F., foraging on Medicago sativa flowers. Each foraging behavior investigated differed among bee species. Both social bees exhibited directionality of movement and had similar residence, in contrast to the random movement and shorter residence of the solitary bee. Tripping rate and net distance traveled differed among the three bee species. We ranked each behavior among bee species and used the relative ranking as gene flow predictor before testing the predictions against empirical gene flow data. Tripping rate and net distance traveled, but not residence, predicted relative gene dispersal among bee species. Linking specific behaviors to gene flow provides mechanisms to explain differences in gene flow among bee species and guides the development of management practices to reduce gene flow. Increasing popularity of high-throughput phenotyping technologies, such as image-based phenotyping, offer novel ways for quantifying plant growth and morphology. These new methods can be more or less accurate and precise than traditional, manual measurements. Many large-scale phenotyping efforts are conducted to enable genome-wide association studies (GWAS), but it is unclear exactly how alternative methods of phenotyping will affect GWAS results. In this study we simulate phenotypes that are controlled by the same set of causal loci but have differing heritability, similar to two different measurements of the same morphological character. We then perform GWAS with the simulated traits and create receiver operating characteristic (ROC) curves from the results. The areas under the ROC curves (AUCs) provide a metric that allows direct comparisons of GWAS results from different simulated traits. We use this framework to evaluate the effects of heritability and the number of causative loci on the AUCs of simulated traits; we also test the differences between AUCs of traits with differing heritability. We find that both increasing the number of causative loci and decreasing the heritability reduce a trait's AUC. We also find that when two traits are controlled by a greater number of causative loci, they are more likely to have significantly different AUCs as the difference between their heritabilities increases. When simulation results are applied to measures of tassel morphology, we find no significant difference between AUCs from GWAS using manual and image-based measurements of typical maize tassel characters. This finding indicates that both measurement methods have similar ability to identify genetic associations. Premise of the Study: The ability to attract pollinators is crucial to plants that rely on insects for pollination. We contrasted the roles of floral display size and flower color in attracting three bee species and determined the relationships between plant attractiveness (number of pollinator visits) and seed set for each bee species., Methods: We recorded pollinator visits to plants, measured plant traits, and quantified plant reproductive success. A zero-inflated Poisson regression model indicated plant traits associated with pollinator attraction. It identified traits that increased the number of bee visits and traits that increased the probability of a plant not receiving any visits. Different components of floral display size were examined and two models of flower color contrasted. Relationships between plant attractiveness and seed set were determined using regression analyses., Key Results: Plants with more racemes received more bee visits from all three bee species. Plants with few racemes were more likely not to receive any bee visits. The role of flower color varied with bee species and was influenced by the choice of the flower color model. Increasing bee visits increased seed set for all three bee species, with the steepest slope for leafcutting bees, followed by bumble bees, and finally honey bees., Conclusions: Floral display size influenced pollinator attraction more consistently than flower color. The same plant traits affected the probability of not being visited and the number of pollinator visits received. Wildfire is globally an important ecological disturbance affecting biochemical cycles and vegetation composition, but also puts people and their homes at risk. Suppressing wildfires has detrimental ecological effects and can promote larger and more intense wildfires when fuels accumulate, which increases the threat to buildings in the wildland-urban interface (WUI). Yet, when wildfires occur, typically only a small proportion of the buildings within the fire perimeter are lost, and the question is what determines which buildings burn. Our goal was to examine which factors are related to building loss when a wildfire occurs throughout the United States. We were particularly interested in the relative roles of vegetation, topography, and the spatial arrangement of buildings, and how their respective roles vary among ecoregions. We analyzed all fires that occurred within the conterminous United States from 2000 to 2010 and digitized which buildings were lost and which survived according to Google Earth historical imagery. We modeled the occurrence as well as the percentage of buildings lost within clusters using logistic and linear regression. Overall, variables related to topography and the spatial arrangement of buildings were more frequently present in the best 20 regression models than vegetation-related variables. In other words, specific locations in the landscape have a higher fire risk, and certain development patterns can exacerbate that risk. Fire policies and prevention efforts focused on vegetation management are important, but insufficient to solve current wildfire problems. Background: To prevent cognitive impairment, phenylketonuria requires lifelong management of blood phenylalanine (Phe) concentration with a low-Phe diet. The diet restricts intake of Phe from natural proteins in combination with traditional amino acid medical foods (AA-MFs) or glycomacropeptide medical foods (GMP-MFs) that contain primarily intact protein and a small amount of Phe., Objective: We investigated the efficacy and safety of a low-Phe diet combined with GMP-MFs or AA-MFs providing the same quantity of protein equivalents in free-living subjects with phenylketonuria., Design: This 2-stage, randomized crossover trial included 30 early-treated phenylketonuria subjects (aged 15-49 y), 20 with classical and 10 with variant phenylketonuria. Subjects consumed, in random order for 3 wk each, their usual low-Phe diet combined with AA-MFs or GMP-MFs. The treatments were separated by a 3-wk washout with AA-MFs. Fasting plasma amino acid profiles, blood Phe concentrations, food records, and neuropsychological tests were obtained., Results: The frequency of medical food intake was higher with GMP-MFs than with AA-MFs. Subjects rated GMP-MFs as more acceptable than AA-MFs and noted improved gastrointestinal symptoms and less hunger with GMP-MFs. ANCOVA indicated no significant mean ± SE increase in plasma Phe (62 ± 40 μmol/L, P = 0.136), despite a significant increase in Phe intake from GMP-MFs (88 ± 6 mg Phe/d, P = 0.026). AA-MFs decreased plasma Phe (-85 ± 40 μmol/L, P = 0.044) with stable Phe intake. Blood concentrations of Phe across time were not significantly different (AA-MFs = 444 ± 34 μmol/L, GMP-MFs = 497 ± 34 μmol/L), suggesting similar Phe control. Results of the Behavior Rating Inventory of Executive Function were not significantly different., Conclusions: GMP-MFs provide a safe and acceptable option for the nutritional management of phenylketonuria. The greater acceptability and fewer side effects noted with GMP-MFs than with AA-MFs may enhance dietary adherence for individuals with phenylketonuria. Introduction: Metabolic control of phenylketonuria (PKU) and compliance with the low-phenylalanine (phe) diet are frequently assessed by measuring blood phe concentrations in dried blood spots (DBS) collected by patients instead of plasma phe concentrations., Objective: Our objective was to investigate the difference in blood phe concentrations in DBS collected by subjects and analyzed using either a validated newborn screening tandem mass spectrometry (MS/MS) protocol or ion-exchange chromatography (IEC) compared to plasma phe concentrations obtained simultaneously and analyzed using IEC., Design: Three to four fasting blood samples were obtained from 29 subjects with PKU, ages 15-49 years. Capillary blood was spotted on filter paper by each subject and the DBS analyzed using both MS/MS and IEC. Plasma was isolated from venous blood and analyzed using IEC., Results: Blood phe concentrations in DBS analyzed using MS/MS are 28% ± 1% (n = 110, p < 0.0001) lower than plasma phe concentrations analyzed using IEC resulting in a blood phe concentration of 514 ± 23 μmol/L and a plasma phe concentration of 731 ± 32 μmol/L (mean ± SEM). This discrepancy is larger when plasma phe is > 600 μmol/L. Due to the large variability across subjects of 13.2%, a calibration factor to adjust blood phe concentrations is not recommended. Analysis of DBS using IEC reduced the discrepancy to 15 ± 2% lower phe concentrations compared to plasma analyzed using IEC (n = 38, p = 0.0001). This suggests that a major contributor to the discrepancy in phe concentrations is the analytical method., Conclusion: Use of DBS analyzed using MS/MS to monitor blood phe concentrations in individuals with PKU yields significantly lower phe levels compared to plasma phe levels analyzed using IEC. Optimization of current testing methodologies for measuring phe in DBS, along with patient education regarding the appropriate technique for spotting blood on filter paper is needed to improve the accuracy of using DBS to measure phe concentrations in PKU management.

Pulses of respiration from coarse woody debris (CWD) have been observed immediately following canopy disturbances, but it is unclear how long these pulses are sustained. Several factors are known to influence carbon flux rates from CWD, but few studies have evaluated more than temperature and moisture. We experimentally manipulated forest structure in a second-growth northern hardwood forest and measured CO2 flux periodically for seven growing seasons following gap creation. We present an analysis of which factors, including the composition of the wood-decay fungal community influence CO2 flux. CO2 flux from CWD was strongly and positively related to wood temperature and varied significantly between substrate types (logs vs. stumps). For five growing seasons after treatment, the CO2 flux of stumps reached rates up to seven times higher than that of logs. CO2 flux of logs did not differ significantly between canopy-gap and closed-canopy conditions in the fourth through seventh post-treatment growing seasons. By the seventh season, the seasonal carbon flux of both logs and stumps had decreased significantly from prior years. Linear mixed models indicated the variation in the wood inhabiting fungal community composition explained a significant portion of variability in the CO2 flux along with measures of substrate conditions. CO2 flux rates were inversely related to fungal diversity, with logs hosting more species but emitting less CO2 than stumps. Overall, our results suggest that the current treatment of CWD in dynamic forest carbon models may be oversimplified, thereby hampering our ability to predict realistic carbon fluxes associated with wood decomposition.

Stable isotopes of carbon, nitrogen, and sulfur are used as ecological tracers for a variety of applications, such as studies of animal migrations, energy sources, and food web pathways. Yet uncertainty relating to the time period integrated by isotopic measurement of animal tissues can confound the interpretation of isotopic data. There have been a large number of experimental isotopic diet shift studies aimed at quantifying animal tissue isotopic turnover rate λ (%·day(-1), often expressed as isotopic half-life, ln(2)/λ, days). Yet no studies have evaluated or summarized the many individual half-life estimates in an effort to both seek broad-scale patterns and characterize the degree of variability. Here, we collect previously published half-life estimates, examine how half-life is related to body size, and test for tissue- and taxa-varying allometric relationships. Half-life generally increases with animal body mass, and is longer in muscle and blood compared to plasma and internal organs. Half-life was longest in ecotherms, followed by mammals, and finally birds. For ectotherms, different taxa-tissue combinations had similar allometric slopes that generally matched predictions of metabolic theory. Half-life for ectotherms can be approximated as: ln (half-life) = 0.22*ln (body mass) + group-specific intercept; n = 261, p<0.0001, r2 = 0.63. For endothermic groups, relationships with body mass were weak and model slopes and intercepts were heterogeneous. While isotopic half-life can be approximated using simple allometric relationships for some taxa and tissue types, there is also a high degree of unexplained variation in our models. Our study highlights several strong and general patterns, though accurate prediction of isotopic half-life from readily available variables such as animal body mass remains elusive.

Large carnivores are difficult to monitor because they tend to be sparsely distributed, sensitive to human activity, and associated with complex life histories. Consequently, understanding population trend and viability requires conservationists to cope with uncertainty and bias in population data. Joint analysis of combined data sets using multiple models (i.e., integrated population model) can improve inference about mechanisms (e.g., habitat heterogeneity and food distribution) affecting population dynamics. However, unobserved or unobservable processes can also introduce bias and can be difficult to quantify. We developed a Bayesian hierarchical modeling approach for inference on an integrated population model that reconciles annual population counts with recruitment and survival data (i.e., demographic processes). Our modeling framework is flexible and enables a realistic form of population dynamics by fitting separate density-dependent responses for each demographic process. Discrepancies estimated from shared parameters among different model components represent unobserved additions (i.e., recruitment or immigration) or removals (i.e., death or emigration) when annual population counts are reliable. In a case study of gray wolves in Wisconsin (1980-2011), concordant with policy changes, we estimated that a discrepancy of 0% (1980-1995), -2% (1996-2002), and 4% (2003-2011) in the annual mortality rate was needed to explain annual growth rate. Additional mortality in 2003-2011 may reflect density-dependent mechanisms, changes in illegal killing with shifts in wolf management, and nonindependent censoring in survival data. Integrated population models provide insights into unobserved or unobservable processes by quantifying discrepancies among data sets. Our modeling approach is generalizable to many population analysis needs and allows for identifying dynamic differences due to external drivers, such as management or policy changes.

Applications of remote sensing for biodiversity conservation typically rely on image classifications that do not capture variability within coarse land cover classes. Here, we compare two measures derived from unclassified remotely sensed data, a measure of habitat heterogeneity and a measure of habitat composition, for explaining bird species richness and the spatial distribution of 10 species in a semi-arid landscape of New Mexico. We surveyed bird abundance from 1996 to 1998 at 42 plots located in the McGregor Range of Fort Bliss Army Reserve. Normalized Difference Vegetation Index values of two May 1997 Landsat scenes were the basis for among-pixel habitat heterogeneity (image texture), and we used the raw imagery to decompose each pixel into different habitat components (spectral mixture analysis). We used model averaging to relate measures of avian biodiversity to measures of image texture and spectral mixture analysis fractions. Measures of habitat heterogeneity, particularly angular second moment and standard deviation, provide higher explanatory power for bird species richness and the abundance of most species than measures of habitat composition. Using image texture, alone or in combination with other classified imagery-based approaches, for monitoring statuses and trends in biological diversity can greatly improve conservation efforts and habitat management.

Invasive species are leading drivers of environmental change. Their impacts are often linked to their population size, but surprisingly little is known about how frequently they achieve high abundances. A nearly universal pattern in ecology is that species are rare in most locations and abundant in a few, generating right-skewed abundance distributions. Here, we use abundance data from over 24,000 populations of 17 invasive and 104 native aquatic species to test whether invasive species differ from native counterparts in statistical patterns of abundance across multiple sites. Invasive species on average reached significantly higher densities than native species and exhibited significantly higher variance. However, invasive and native species did not differ in terms of coefficient of variation, skewness, or kurtosis. Abundance distributions of all species were highly right skewed (skewness>0), meaning both invasive and native species occurred at low densities in most locations where they were present. The average abundance of invasive and native species was 6% and 2%, respectively, of the maximum abundance observed within a taxonomic group. The biological significance of the differences between invasive and native species depends on species-specific relationships between abundance and impact. Recognition of cross-site heterogeneity in population densities brings a new dimension to invasive species management, and may help to refine optimal prevention, containment, control, and eradication strategies.

National-scale analyses of fire occurrence are needed to prioritize fire policy and management activities across the United States. However, the drivers of national-scale patterns of fire occurrence are not well understood, and how the relative importance of human or biophysical factors varies across the country is unclear. Our research goal was to model the drivers of fire occurrence within ecoregions across the conterminous United States. We used generalized linear models to compare the relative influence of human, vegetation, climate, and topographic variables on fire occurrence in the United States, as measured by MODIS active fire detections collected between 2000 and 2006. We constructed models for all fires and for large fires only and generated predictive maps to quantify fire occurrence probabilities. Areas with high fire occurrence probabilities were widespread in the Southeast, and localized in the Mountain West, particularly in southern California, Arizona, and New Mexico. Probabilities for large-fire occurrence were generally lower, but hot spots existed in the western and south-central United States The probability of fire occurrence is a critical component of fire risk assessments, in addition to vegetation type, fire behavior, and the values at risk. Many of the hot spots we identified have extensive development in the wildland--urban interface and are near large metropolitan areas. Our results demonstrated that human variables were important predictors of both all fires and large fires and frequently exhibited nonlinear relationships. However, vegetation, climate, and topography were also significant variables in most ecoregions. If recent housing growth trends and fire occurrence patterns continue, these areas will continue to challenge policies and management efforts seeking to balance the risks generated by wildfires with the ecological benefits of fire.

Changes in land use and land cover have affected and will continue to affect biological diversity worldwide. Yet, understanding the spatially extensive effects of land-cover change has been challenging because data that are consistent over space and time are lacking. We used the U.S. National Land Cover Dataset Land Cover Change Retrofit Product and North American Breeding Bird Survey data to examine land-cover change and its associations with diversity of birds with principally terrestrial life cycles (landbirds) in the conterminous United States. We used mixed-effects models and model selection to rank associations by ecoregion. Land cover in 3.22% of the area considered in our analyses changed from 1992 to 2001, and changes in species richness and abundance of birds were strongly associated with land-cover changes. Changes in species richness and abundance were primarily associated with changes in nondominant types of land cover, yet in many ecoregions different types of land cover were associated with species richness than were associated with abundance. Conversion of natural land cover to anthropogenic land cover was more strongly associated with changes in bird species richness and abundance than persistence of natural land cover in nearly all ecoregions and different covariates were most strongly associated with species richness than with abundance in 11 of 17 ecoregions. Loss of grassland and shrubland affected bird species richness and abundance in forested ecoregions. Loss of wetland was associated with bird abundance in forested ecoregions. Phenylketonuria (PKU) is caused by a mutation in the phenylalanine (phe) hydroxylase gene and requires a low-phe diet plus amino acid (AA) formula to prevent cognitive impairment. Glycomacropeptide (GMP) contains minimal phe and provides a palatable alternative to AA formula. Our objective was to compare growth, body composition, and energy balance in Pah(enu2) (PKU) and wild-type mice fed low-phe GMP, low-phe AA, or high-phe casein diets from 3-23 wk of age. The 2 × 2 × 3 design included main effects of genotype, sex, and diet. Fat and lean mass were assessed by dual-energy X-ray absorptiometry, and acute energy balance was assessed by indirect calorimetry. PKU mice showed growth and lean mass similar to wild-type littermates fed the GMP or AA diets; however, they exhibited a 3-15% increase in energy expenditure, as reflected in oxygen consumption, and a 3-30% increase in food intake. The GMP diet significantly reduced energy expenditure, food intake, and plasma phe concentration in PKU mice compared with the casein diet. The high-phe casein diet or the low-phe AA diet induced metabolic stress in PKU mice, as reflected in increased energy expenditure and intake of food and water, increased renal and spleen mass, and elevated plasma cytokine concentrations consistent with systemic inflammation. The low-phe GMP diet significantly attenuated these adverse effects. Moreover, total fat mass, %body fat, and the respiratory exchange ratio (CO(2) produced/O(2) consumed) were significantly lower in PKU mice fed GMP compared with AA diets. In summary, GMP provides a physiological source of low-phe dietary protein that promotes growth and attenuates the metabolic stress induced by a high-phe casein or low-phe AA diet in PKU mice.

Model averaging is gaining popularity among ecologists for making inference and predictions. Methods for combining models include Bayesian model averaging (BMA) and Akaike's Information Criterion (AIC) model averaging. BMA can be implemented with different prior model weights, including the Kullback-Leibler prior associated with AIC model averaging, but it is unclear how the prior model weight affects model results in a predictive context. Here, we implemented BMA using the Bayesian Information Criterion (BIC) approximation to Bayes factors for building predictive models of bird abundance and occurrence in the Chihuahuan Desert of New Mexico. We examined how model predictive ability differed across four prior model weights, and how averaged coefficient estimates, standard errors and coefficients' posterior probabilities varied for 16 bird species. We also compared the predictive ability of BMA models to a best single-model approach. Overall, Occam's prior of parsimony provided the best predictive models. In general, the Kullback-Leibler prior, however, favored complex models of lower predictive ability. BMA performed better than a best single-model approach independently of the prior model weight for 6 out of 16 species. For 6 other species, the choice of the prior model weight affected whether BMA was better than the best single-model approach. Our results demonstrate that parsimonious priors may be favorable over priors that favor complexity for making predictions. The approach we present has direct applications in ecology for better predicting patterns of species' abundance and occurrence.

Fluxes of organic matter across habitat boundaries are common in food webs. These fluxes may strongly influence community dynamics, depending on the extent to which they are used by consumers. Yet understanding of basal resource use by consumers is limited, because describing trophic pathways in complex food webs is difficult. We quantified resource use for zooplankton, zoobenthos, and fishes in four low-productivity lakes, using a Bayesian mixing model and measurements of hydrogen, carbon, and nitrogen stable isotope ratios. Multiple sources of uncertainty were explicitly incorporated into the model. As a result, posterior estimates of resource use were often broad distributions; nevertheless, clear patterns were evident. Zooplankton relied on terrestrial and pelagic primary production, while zoobenthos and fishes relied on terrestrial and benthic primary production. Across all consumer groups terrestrial reliance tended to be higher, and benthic reliance lower, in lakes where light penetration was low due to inputs of terrestrial dissolved organic carbon. These results support and refine an emerging consensus that terrestrial and benthic support of lake food webs can be substantial, and they imply that changes in the relative availability of basal resources drive the strength of cross-habitat trophic connections.

The aim of this work was to compare use of an o-phthaldialdehyde (OPA) colorimetric assay (OPA-C), which responds to both free AA and peptides, with an OPA fluorimetric assay (OPA-F), which is insensitive to peptides, to quantify rates of ruminal protein degradation in the inhibitor in vitro system using Michaelis-Menten saturation kinetics. Four protein concentrates (expeller-extracted soybean meal, ESBM; 2 solvent-extracted soybean meals, SSBM1 and SSBM2; and casein) were incubated in a ruminal in vitro system treated with hydrazine and chloramphenicol to inhibit microbial uptake of protein degradation products. Proteins were weighed to give a range of N concentrations (from 0.15 to 3 mg of N/mL of inoculum) and incubated with 10 mL of ruminal inoculum and 5 mL of buffer; fermentations were stopped after 2 h by adding trichloroacetic acid (TCA). Proteins were analyzed for buffer-soluble N and buffer extracts were treated with TCA to determine N degraded at t=0 (FD0). The TCA supernatants were analyzed for ammonia (phenol-hypochlorite assay), total AA (TAA; OPA-F), and TAA plus oligopeptides (OPA-C) by flow injection analysis. Velocity of protein degradation was computed from extent of release of 1) ammonia plus free TAA or 2) ammonia plus free TAA and peptides. Rate of degradation (kd) was quantified using nonlinear regression of the integrated Michaelis-Menten equation. The parameters Km (Michaelis constant) and kd (Vmax/Km), where Vmax=maximum velocity, were estimated directly; kd values were adjusted (Akd) for the fraction FD0 using the equation Akd=kd-FD0/2. The OPA-C assay yielded faster degradation rates due to the contribution of peptides to the fraction degraded (overall mean=0.280/h by OPA-C and 0.219/h by OPA-F). Degradation rates for SSBM samples (0.231/h and 0.181/h) and ESBM (0.086/h) obtained by the OPA-C assay were more rapid than rates reported by the National Research Council (NRC). Both assays indicated that the 2 SSBM differed in rumen-undegradable protein (RUP) content; the more slowly degraded SSBM had RUP content (35% by OPA-C) similar to that reported by the NRC. The RUP content of ESBM (42% by OPA-C) was lower than the NRC value. Preliminary studies with 4 additional protein concentrates confirmed that accounting for peptide formation increased degradation rate; however, a trend for an interaction between assay and protein source suggested that peptide release made a smaller contribution to rate for more slowly degraded proteins. Global demand for agricultural products such as food, feed, and fuel is now a major driver of cropland and pasture expansion across much of the developing world. Whether these new agricultural lands replace forests, degraded forests, or grasslands greatly influences the environmental consequences of expansion. Although the general pattern is known, there still is no definitive quantification of these land-cover changes. Here we analyze the rich, pan-tropical database of classified Landsat scenes created by the Food and Agricultural Organization of the United Nations to examine pathways of agricultural expansion across the major tropical forest regions in the 1980s and 1990s and use this information to highlight the future land conversions that probably will be needed to meet mounting demand for agricultural products. Across the tropics, we find that between 1980 and 2000 more than 55% of new agricultural land came at the expense of intact forests, and another 28% came from disturbed forests. This study underscores the potential consequences of unabated agricultural expansion for forest conservation and carbon emissions.

Background: Quantifying changes in forest bird diversity is an essential task for developing effective conservation actions. When subtle changes in diversity accumulate over time, annual comparisons may offer an incomplete perspective of changes in diversity. In this case, progressive change, the comparison of changes in diversity from a baseline condition, may offer greater insight because changes in diversity are assessed over longer periods of times. Our objectives were to determine how forest bird diversity has changed over time and whether those changes were associated with forest disturbance., Methodology/principal Findings: We used North American Breeding Bird Survey data, a time series of Landsat images classified with respect to land cover change, and mixed-effects models to associate changes in forest bird community structure with forest disturbance, latitude, and longitude in the conterminous United States for the years 1985 to 2006. We document a significant divergence from the baseline structure for all birds of similar migratory habit and nest location, and all forest birds as a group from 1985 to 2006. Unexpectedly, decreases in progressive similarity resulted from small changes in richness (<1 species per route for the 22-year study period) and modest losses in abundance (-28.7 - -10.2 individuals per route) that varied by migratory habit and nest location. Forest disturbance increased progressive similarity for Neotropical migrants, permanent residents, ground nesting, and cavity nesting species. We also documented highest progressive similarity in the eastern United States., Conclusions/significance: Contemporary forest bird community structure is changing rapidly over a relatively short period of time (e.g., approximately 22 years). Forest disturbance and forest regeneration are primary factors associated with contemporary forest bird community structure, longitude and latitude are secondary factors, and forest loss is a tertiary factor. Importantly, these findings suggest some regions of the United States may already fall below the habitat amount threshold where fragmentation effects become important predictors of forest bird community structure.

Phenylketonuria (PKU) requires a lifelong low-phenylalanine (phe) diet where protein needs are met by consumption of a phe-free amino acid (AA) formula; complaints of persistent hunger are common. Foods made with glycomacropeptide (GMP), an intact protein that contains minimal phe and may promote satiety, provide an alternative to AA formula. The objective was to assess the ability of a GMP breakfast to promote satiety and affect plasma concentrations of AAs, insulin, and the appetite stimulating hormone ghrelin in those with PKU, when compared to an AA-based breakfast. Eleven PKU subjects (8 adults and 3 boys ages 11-14) served as their own controls in an inpatient metabolic study with two 4-day treatments: an AA-based diet followed by a diet replacing all AA formula with GMP foods. Plasma concentrations of AAs, insulin and ghrelin were obtained before and/or 180 min after breakfast. Satiety was assessed using a visual analog scale before, immediately after and 150 min after breakfast. Postprandial ghrelin concentration was significantly lower (p=0.03) with GMP compared to an AA-based breakfast, with no difference in fasting ghrelin. Lower postprandial ghrelin concentrations were associated with greater feelings of fullness after breakfast suggesting greater satiety with GMP compared to AAs. Postprandial concentrations of insulin and total plasma AAs were higher after a GMP breakfast compared to an AA-based breakfast consistent with slower absorption and less degradation of AAs from GMP. These results show sustained ghrelin suppression, and suggest greater satiety with ingestion of a meal containing GMP compared with AAs., (Copyright 2010 Elsevier Inc. All rights reserved.)

Chemical communication by many insect species involves complex signals of both insect and plant origin. Much attention has been focused on the behavioral activities of these components but less on their sources of variation, despite implications for evolutionary theory and pest management. We studied variation in chemical signaling at host, tree-within-host, and beetle-on-tree scales using tunneling male pine engravers [Ips pini (Say)] on jack, Pinus banksiana Lamb, red, P. resinosa Aiton, and white, P. strobus L. pines. Pine engravers are distributed transcontinentally, and stereoisomeric ratios of their principal pheromone component ipsdienol varies regionally. Linear mixed-effects models were used to examine variation in monoterpene and pheromone volatile profiles, determined by gas chromatography. Phloem from white pine had the greatest concentration of monoterpenes, although insects tunneling in white pine produced the smallest ratios of monoterpenes to pheromones (1:2) in their volatile plumes relative to jack and red pine (1:1). Beetle-to-beetle variation in plume composition was approximately 2-9 times greater than the inter-tree variation within a tree species. The stereoisomeric ratio of ipsdienol was highly consistent within the pheromone component of the plume. The little variation present existed almost entirely at the level of the insects. Within the pheromone component of the plume in a given host species, there was up to 13 times more beetle-to-beetle than tree-to-tree variation. This magnitude was almost double the magnitudes of the ratios among components within the entire plumes. Implications to the behavioral ecology of bark beetle communication, such as potential strategies of cheating and predator avoidance, are discussed.

Historical land use can influence forest species composition and structure for centuries after direct use has ceased. In Wisconsin, USA, Euro-American settlement in the mid- to late 1800s was accompanied by widespread logging, agricultural conversion, and fire suppression. To determine the maximum magnitude of change in forest ecosystems at the height of the agricultural period and the degree of recovery since that time, we assessed changes in forest species composition and structure among the (1) mid-1800s, at the onset of Euro-American settlement; (2) 1930s, at the height of the agricultural period; and (3) 2000s, following forest regrowth. Data sources included the original U.S. Public Land Survey records (mid-1800s), the Wisconsin Land Economic Inventory (1930s), and U.S. Forest Service Forest Inventory and Analysis data (2000s). We derived maps of relative species dominance and tree diameters for the three dates and assessed change using spatial error models, nonmetric multidimensional scaling ordination, and Sørenson distance measures. Our results suggest that since the mid-1800s, hemlock and white pine have declined in absolute area from 22% to 1%, and the proportion of medium (25-<50 cm) and large-diameter (> or = 50 cm) trees of all species has decreased from 71% to 27% across the entire state. Early-successional aspen-birch is three times more common than in the mid-1800s (9% vs. 3%), and maple and other shade-tolerant species are increasing in southern areas formerly dominated by oak forests and savannas. Since the peak agricultural extent in the 1930s, species composition and tree size in northern forests have shown some recovery, while southern forests appear to be on a novel trajectory of change. There is evidence of regional homogenization, but the broad north-south environmental gradient in Wisconsin constrains overall species composition. Although the nature of the future forests will be determined in part by climate change and other exogenous variables, land use is likely to remain the driving factor.

One-third of net CO(2) emissions to the atmosphere since 1850 are the result of land-use change, primarily from the clearing of forests for timber and agriculture, but quantifying these changes is complicated by the lack of historical data on both former ecosystem conditions and the extent and spatial configuration of subsequent land use. Using fine-resolution historical survey records, we reconstruct pre-EuroAmerican settlement (1850s) forest carbon in the state of Wisconsin, examine changes in carbon after logging and agricultural conversion, and assess the potential for future sequestration through forest recovery. Results suggest that total above-ground live forest carbon (AGC) fell from 434 TgC before settlement to 120 TgC at the peak of agricultural clearing in the 1930s and has since recovered to approximately 276 TgC. The spatial distribution of AGC, however, has shifted significantly. Former savanna ecosystems in the south now store more AGC because of fire suppression and forest ingrowth, despite the fact that most of the region remains in agriculture, whereas northern forests still store much less carbon than before settlement. Across the state, continued sequestration in existing forests has the potential to contribute an additional 69 TgC. Reforestation of agricultural lands, in particular, the formerly high C-density forests in the north-central region that are now agricultural lands less optimal than those in the south, could contribute 150 TgC. Restoring historical carbon stocks across the landscape will therefore require reassessing overall land-use choices, but a range of options can be ranked and considered under changing needs for ecosystem services.

Background: Phenylketonuria (PKU) requires a lifelong low-phenylalanine diet that provides the majority of protein from a phenylalanine-free amino acid (AA) formula. Glycomacropeptide (GMP), an intact protein formed during cheese production, contains minimal phenylalanine., Objective: The objective was to investigate the effects of substituting GMP food products for the AA formula on acceptability, safety, plasma AA concentrations, and measures of protein utilization in subjects with PKU., Design: Eleven subjects participated in an inpatient metabolic study with two 4-d treatments: a current AA diet (AA diet) followed by a diet that replaced the AA formula with GMP (GMP diet) supplemented with limiting AAs. Plasma concentrations of AAs, blood chemistries, and insulin were measured and compared in AA (day 4) and GMP diets (day 8)., Results: The GMP diet was preferred to the AA diet in 10 of 11 subjects with PKU, and there were no adverse reactions to GMP. There was no significant difference in phenylalanine concentration in postprandial plasma with the GMP diet compared with the AA diet. When comparing fasting with postprandial plasma, plasma phenalyalanine concentration increased significantly with the AA but not with the GMP diet. Blood urea nitrogen was significantly lower, which suggests decreased ureagenesis, and plasma insulin was higher with the GMP diet than with the AA diet., Conclusions: GMP, when supplemented with limiting AAs, is a safe and highly acceptable alternative to synthetic AAs as the primary protein source in the nutritional management of PKU. As an intact protein source, GMP improves protein retention and phenylalanine utilization compared with AAs.

We address the development of methods for analyzing crossclassified categorical data that are spatially autocorrelated. We first extend the autologistic model to accommodate two variables. Two bivariate autologistic models are constructed, namely a two-step model and a symmetric model. Importance sampling is used to approximate the complex normalizing factors that arise in these models, and Markov chain Monte Carlo techniques are used to generate simulations of posterior distributions. The resulting models then are expanded to accommodate trend surfaces and directional effects. Simulation studies and real data are used to illustrate this method.

Rural America is witnessing widespread housing development, which is to the detriment of the environment. It has been suggested to cluster houses so that their disturbance zones overlap and thus cause less habitat loss than is the case for dispersed development. Clustering houses makes intuitive sense, but few empirical studies have quantified the spatial pattern of houses in real landscapes, assessed changes in their patterns over time, and quantified the resulting habitat loss. We addressed three basic questions: (1) What are the spatial patterns of houses and how do they change over time; (2) How much habitat is lost due to houses, and how is this affected by spatial pattern of houses; and (3) What type of habitat is most affected by housing development. We mapped 27 419 houses from aerial photos for five time periods in 17 townships in northern Wisconsin and calculated the terrestrial land area remaining after buffering each house using 100- and 500-m disturbance zones. The number of houses increased by 353% between 1937 and 1999. Ripley's K test showed that houses were significantly clustered at all time periods and at all scales. Due to the clustering, the rate at which habitat was lost (176% and 55% for 100- and 500-m buffers, respectively) was substantially lower than housing growth rates, and most land area was undisturbed (95% and 61% for 100-m and 500-m buffers, respectively). Houses were strongly clustered within 100 m of lakes. Habitat loss was lowest in wetlands but reached up to 60% in deciduous forests. Our results are encouraging in that clustered development is common in northern Wisconsin, and habitat loss is thus limited. However, the concentration of development along lakeshores causes concern, because these may be critical habitats for many species. Conservation goals can only be met if policies promote clustered development and simultaneously steer development away from sensitive ecosystems.

In the United States, housing density has substantially increased in and adjacent to forests. Our goal in this study was to identify how housing density and human populations are associated with avian diversity. We compared these associations to those between landscape pattern and avian diversity, and we examined how these associations vary across the conterminous forested United States. Using data from the North American Breeding Bird Survey, the U.S. Census, and the National Land Cover Database, we focused on forest and woodland bird communities and conducted our analysis at multiple levels of model specificity, first using a coarse-thematic resolution (basic models), then using a larger number of fine-thematic resolution variables (refined models). We found that housing development was associated with forest bird species richness in all forested ecoregions of the conterminous United States. However, there were important differences among ecoregions. In the basic models, housing density accounted for < 5% of variance in avian species richness. In refined models, 85% of models included housing density and/or residential land cover as significant variables. The strongest guild response was demonstrated in the Adirondack-New England ecoregion, where 29% of variation in richness of the permanent resident guild was associated with housing density. Model improvements due to regional stratification were most pronounced for cavity nesters and short-distance migrants, suggesting that these guilds may be especially sensitive to regional processes. The varying patterns of association between avian richness and attributes associated with landscape structure suggested that landscape context was an important mediating factor affecting how biodiversity responds to landscape changes. Our analysis suggested that simple, broadly applicable, land use recommendations cannot be derived from our results. Rather, anticipating future avian response to land use intensification (or reversion to native vegetation) has to be conditioned on the current landscape context and the species group of interest. Our results show that housing density and residential land cover were significant predictors of forest bird species richness, and their prediction strengths are likely to increase as development continues.

Periodic wildfire maintains the integrity and species composition of many ecosystems, including the mediterranean-climate shrublands of California. However, human activities alter natural fire regimes, which can lead to cascading ecological effects. Increased human ignitions at the wildland-urban interface (WUI) have recently gained attention, but fire activity and risk are typically estimated using only biophysical variables. Our goal was to determine how humans influence fire in California and to examine whether this influence was linear, by relating contemporary (2000) and historic (1960-2000) fire data to both human and biophysical variables. Data for the human variables included fine-resolution maps of the WUI produced using housing density and land cover data. Interface WUI, where development abuts wildland vegetation, was differentiated from intermix WUI, where development intermingles with wildland vegetation. Additional explanatory variables included distance to WUI, population density, road density, vegetation type, and ecoregion. All data were summarized at the county level and analyzed using bivariate and multiple regression methods. We found highly significant relationships between humans and fire on the contemporary landscape, and our models explained fire frequency (R2 = 0.72) better than area burned (R2 = 0.50). Population density, intermix WUI, and distance to WUI explained the most variability in fire frequency, suggesting that the spatial pattern of development may be an important variable to consider when estimating fire risk. We found nonlinear effects such that fire frequency and area burned were highest at intermediate levels of human activity, but declined beyond certain thresholds. Human activities also explained change in fire frequency and area burned (1960-2000), but our models had greater explanatory power during the years 1960-1980, when there was more dramatic change in fire frequency. Understanding wildfire as a function of the spatial arrangement of ignitions and fuels on the landscape, in addition to nonlinear relationships, will be important to fire managers and conservation planners because fire risk may be related to specific levels of housing density that can be accounted for in land use planning. With more fires occurring in close proximity to human infrastructure, there may also be devastating ecological impacts if development continues to grow farther into wildland vegetation.

IGF binding protein-5 (IGFBP-5) modulates the availability of IGF-I to its receptor and potentiates the intestinotrophic action of IGF-I. Our aim was to test the hypothesis that stimulation of intestinal growth due to coinfusion of IGF-I with total parenteral nutrition (TPN) solution is dependent on increased expression of IGFBP-5 through conducting our studies in IGFBP-5 knockout (KO) mice. IGFBP-5 KO, heterozygote (HT) and wild type (WT) male and female mice were maintained with TPN or TPN plus coinfusion of IGF-I [recombinant human (rh)IGF-I; 2.5 mg x kg(-1) x day(-1)] for 5 days. The concentration of IGF-I in serum was 73% greater (P < 0.0001) in mice given TPN + IGF-I infusion compared with TPN alone. IGF-I attenuated the 2-3 g loss of body weight associated with TPN in WT mice, whereas KO and HT mice did not show improvement in body weight with IGF-I treatment. KO and HT mice had significantly greater levels of circulating IGF-I binding proteins (IGFBPs) compared with WT mice. Intestinal growth due to IGF-I was observed in all groups treated with IGF-I based on greater concentrations of protein and DNA in small intestine and colon and significantly greater crypt depth and muscularis thickness in jejunum. Jejunal expression of IGFBP-5 mRNA was greater in WT mice, whereas IGFBP-3 mRNA was greater in KO mice treated with IGF-I. In summary, the absence of the IGFBP-5 gene did not block the ability of IGF-I to stimulate intestinal growth, possibly because greater jejunal expression of IGFBP-3 compensates for the absence of IGFBP-5.

Reasons for Performing Study: There is little information available about conformational changes in the forelimbs of growing foals., Objectives: To describe the conformation of the carpus and fetlock of Thoroughbred foals from birth to yearling sale age., Methods: Subjective assessments of the fetlock and carpal conformation of 119 Thoroughbred foals were made within the first month of life and then at 30 day intervals until at least age 120 days. At least 70 subjects were examined further at 60 day intervals until September of their second year. Conformation grades are reported for 5 age groups: first 7 days and at a mean of 46, 176, 362 and 525 days. The conformation of all available sires and dams of subjects was also graded., Results: All subjects demonstrated carpal deviations, such as valgus, outward rotation and offset, and approximately 30% had fetlock deviations. Heavier birth weights were associated with carpal offset and fetlock inward conformation at most ages, and heavier yearlings were more likely to be carpal valgus. The carpal conformation of the sire (offset and outward rotation) was associated with similar yearling carpal conformation. During the study period, the carpal conformation of Thoroughbred foals became less valgus and more offset. Fetlock conformation became more inwardly deviated during the first 6 months of the study., Conclusions: Carpal and fetlock conformation change greatly in Thoroughbred foals up to age 18 months. The phenotype of the sire can be associated with yearling carpal conformation and bodyweight, particularly at birth and as yearlings, is associated with yearling fetlock and carpal conformation., Potential Relevance: New factors associated with forelimb conformational deviations have been identified that may help breeders better to manage young racing stock.

Roads remove habitat, alter adjacent areas, and interrupt and redirect ecological flows. They subdivide wildlife populations, foster invasive species spread, change the hydrologic network, and increase human use of adjacent areas. At broad scales, these impacts cumulate and define landscape patterns. The goal of this study was to improve our understanding of the dynamics of road networks over time, and their effects on landscape patterns, and identify significant relationships between road changes and other land-use changes. We mapped roads from aerial photographs from five dates between 1937 and 1999 in 17 townships in predominantly forested landscapes in northern Wisconsin, U.S.A. Patch-level landscape metrics were calculated on terrestrial area outside of a 15-m road-effect zone. We used generalized least-squares regression models to relate changes in road density and landscape pattern to concurrent changes in housing density. Rates of change and relationships were compared among three ecological regions. Our results showed substantial increases in both road density and landscape fragmentation during the study period. Road density more than doubled, and median, mean, and largest patch size were reduced by a factor of four, while patch shape became more regular. Increases in road density varied significantly among ecological subsections and were positively related to increases in housing density. Fragmentation was largely driven by increases in road density, but housing density had a significantly positive relationship with largest patch area and patch shape. Without protection of roadless areas, our results suggest road development is likely to continue in the future, even in areas where road construction is constrained by the physical environment. Recognizing the dynamic nature of road networks is important for understanding and predicting their ecological impacts over time and understanding where other types of development are likely to occur in the future. Historical perspectives of development can provide guidance in prioritizing management efforts to defragment landscapes and mitigate the ecological impacts of past road development.

Modelling disease clustering over space and time can be helpful in providing indications of possible exposures and planning corresponding public health practices. Though a considerable number of studies focus on modelling spatio-temporal patterns of disease, most of them do not directly model a spatio-temporal clustering structure and could be ineffective for detecting clusters. In this paper, we extend a purely spatial cluster model to accommodate space-time clustering. Inference is performed in a Bayesian framework using reversible jump Markov chain Monte Carlo. This idea is illustrated using data on female breast cancer mortality from Japan. The optimal level of energy for critically ill patients who require parenteral nutrition (PN) is unclear. Our objective was to determine whether 50% energy (50%E) restriction due to a reduction in carbohydrate or fat, with provision of adequate protein and micronutrients, ameliorates the detrimental effects of dexamethasone (Dex) on body protein catabolism, insulin resistance, and insulin-like growth factor-I (IGF-I) responses in rats administered PN. The experiment included 6 PN groups, adequate energy (AE) +/- Dex, 50% AE with high carbohydrate (50%E CHO) +/- Dex and 50% AE with high fat (50%E FAT) +/- Dex. There was a significant interaction between energy level and Dex such that the increase in body catabolism due to 50%E from CHO or FAT was reduced by approximately 50%, although the amount of body weight and nitrogen lost over 7 d was significantly greater with 50%E than with AE. AE+Dex induced a 60% increase in liver mass, whereas 50%E+Dex reduced the increase to 26%. AE+Dex induced a 5-fold increase in serum insulin level, whereas 50%E+Dex normalized the insulin to glucose ratio. Serum IGF-I levels were reduced 14-18% by Dex and 30% by 50%E. Hepatic immunoreactive IGF-I was significantly correlated with serum IGF-I and nitrogen balance. 50%E CHO and 50%E FAT had differential effects on hepatic IGF-I mRNA with a 40% decrease in IGF-I mRNA due to 50%E FAT+Dex. In summary,CHO or FAT hypoenergetic PN with adequate protein had similar effects in normalizing hyperinsulinemia, attenuating hepatomegaly, and reducing the increment, but not the total amount of body protein catabolism, induced by glucocorticoid excess.

Objective: To quantify the adhesion of Staphylococcus aureus to 4 equine bone surfaces passivated in a balanced polyionic solution (Plasmalyte) or hyperimmune equine plasma (Polymune plasma)., Study Design: In vitro comparative study., Sample Population: Third metacarpal bone (MC3) surface explants from 9 equine cadavers., Methods: Approximately 1 cm(2) sections of periosteum were removed from MC3 and stapled to sterile stainless steel screens. Three bone surface explants were cut using a surgical saw to present 1 cm(2) surfaces of subperiosteal bone, cut cortical bone, or endosteum. Duplicate explants of each surface were immersed for 1 hour in Plasmalyte or hyperimmune equine plasma. Each explant was then placed in a well of a 6-well sterile tissue culture plate with the surface of interest exposed. Each surface was inoculated with approximately 100 colony-forming units of S. aureus in 10 microL of Mueller Hinton broth and incubated for 6 hours at 37 degrees C. After gentle rinsing to remove non-adherent bacteria, samples were sonicated for 5 minutes at 60 kHz to loosen adhered bacteria. The number of adherent bacteria was determined by serial dilutions and incubation of the sonicate. Scanning electron microscopy (SEM) was performed on samples identically treated from an additional horse to confirm bacterial removal by sonication from all surfaces and support quantitative culture results., Results: Less S. aureus adhered to periosteum than to cortical bone, cut cortical bone, and endosteal surfaces, which were all similar. Exposure of all surfaces to hyperimmune plasma reduced S. aureus adherence compared with Plasmalyte exposure; SEM supported these conclusions., Conclusion: Less bacteria adhere to periosteum than other bone surfaces. Hyperimmune plasma reduces bacterial adhesion to all bone tissue surfaces., Clinical Relevance: Understanding the factors that affect bacterial adhesion to bone will facilitate development of improved intraoperative lavage solutions to reduce the morbidity and mortality associated with postoperative infection.

Multiple predator species feeding on a common prey can lead to higher or lower predation than would be expected by simply combining their individual effects. Such emergent multiple predator effects may be especially prevalent if predators share feeding habitat. Despite the prevalence of endophagous insects, no studies have examined how multiple predators sharing an endophytic habitat affect prey or predator reproduction. We investigated density-dependent predation of Thanasimus dubius (Coleoptera: Cleridae) and Platysoma cylindrica (Coleoptera: Histeridae) on a bark beetle prey, Ips pini (Coleoptera: Scolytidae), in a laboratory assay. I. pini utilize aggregation pheromones to group-colonize and reproduce within the stems of conifers. T. dubius and P. cylindrica exploit these aggregation pheromones to arrive simultaneously with the herbivore. Adult T. dubius prey exophytically, while P. cylindrica adults enter and prey within the bark beetle galleries. Larvae of both predators prey endophytically. We used a multiple regression analysis, which avoids confounding predator composition with density, to examine the effects of varying predator densities alone and in combination on herbivore establishment, herbivore reproduction, and predator reproduction. Predators reduced colonization success by both sexes, and decreased I. pini reproduction on a per male and per female basis. The combined effects of these predators did not enhance or reduce prey establishment or reproduction in unexpected manners, and these predators were entirely substitutable. The herbivore's net replacement rate was never reduced significantly below one at prey and predator densities emulating field conditions. Similar numbers of each predator species emerged from the logs, but predator reproduction suffered from high intraspecific interference. The net replacement rate of P. cylindrica was not affected by conspecifics or T. dubius. In contrast, the net replacement rate of T. dubius decreased with the presence of conspecifics or P. cylindrica. Combinations of both predators led to an emergent effect, a slightly increased net replacement rate of T. dubius. This may have been due to predation by larval T. dubius on pupal P. cylindrica, as P. cylindrica develops more rapidly than T. dubius within this shared habitat.

Serotonin (5-hydroxytryptamine, 5-HT) is an endogenous signalling molecule capable of altering small intestinal motility. Serotonin is normally present in the intestinal lumen and released by enterochromaffin cells of the mucosal epithelium. We found that intraduodenal infusion of exogenous serotonin causes a dose-dependent myoelectric response in the smooth muscle of the small intestine in the conscious rat. The response consists of repetitive bursts of action potentials (RBAP) that are characterized as short bursts of non-propagative myoelectric spiking. RBAP occur intermittently and only during the first 15 min after intralumenal serotonin infusion. After the initial 15 min period, the frequency of RBAP declines, and the myoelectric pattern shifts to prolonged and continuous spiking, eliminating the interdigestive migrating myoelectric pattern. The effects of intralumenal serotonin are not replicated by parenteral or intraperitoneal infusion nor by intralumenal infusion of 5-hydroxytryptophan or 5-hydroxyindoleacetic acid. The response to intralumenal serotonin was eliminated by several specific 5-HT receptor antagonists. On repeated intralumenal administration of serotonin, the RBAP response decreased demonstrating a decreased sensitivity of the muscle contraction on re-exposure to serotonin. We conclude that intralumenal infusion of serotonin can temporarily initiate specific small intestinal muscle events that are not generated by serotonin from other non-lumenal administration sites. We speculate that an afferent neuro-pathway is necessary for the induction of RBAP, since RBAP are not observed from in vitro muscle preparations.

Maps of regional disease rates are potentially useful tools in examining spatial patterns of disease and for identifying clusters. Bayes and empirical Bayes approaches to this problem have proven useful in smoothing crude maps of disease rates. In recent years, models including both spatially correlated random effects and spatially unstructured random effects have been very popular. The spatially correlated random effects have been proposed in an attempt to capture a general clustering in the data. As an alternative, we propose replacing the spatially structured random effect with fixed clustering effects associated with particular areas. A reversible jump Markov chain Monte Carlo (RJMCMC) algorithm for posterior inference is described. ABSTRACT Successful spread of an organism to a new habitat requires both immigration to and growth on that habitat. Field experiments were conducted to determine the relative roles of dispersal (i.e., immigration) and bacterial multiplication in spread of Pseudomonas syringae pv. syringae in the phyllosphere. To study spread, individual plots consisted of three nested concentric squares with the inner 6 m(2) planted to snap beans serving as the sink. Each sink, in turn, was surrounded by a barrier zone, usually 6 m wide, which was surrounded by a 6-m-wide source area. The source areas were planted with snap bean seeds inoculated with doubly marked strains derived from wild-type P. syringae pv. syringae B728a. The treatments were designed to test the effects of the nature and width of the barrier zone and suitability of the habitat in the sinks on spread of P. syringae pv. syringae. The marked strains introduced into the source areas at the time of planting were consistently detected in sink areas within a day or two after emergence of bean seedlings in the sources as assessed by leaf imprinting and dilution plating. The amounts of spread (population sizes of the marked strain in sinks) across barrier zones planted to snap bean (a suitable habitat for growth of P. syringae pv. syringae), soybean (not a favorable habitat for P. syringae pv. syringae), and bare ground were not significantly different. Thus, the nature of the barrier had no measurable effect on spread. Similarly, spread across bare-ground barriers 20 m wide was not significantly different from that across barriers 6 m wide, indicating that distance on this scale was not a major factor in determining the amount of spread. The suitability of the sink for colonization by P. syringae pv. syringae had a measurable effect on spread. Spread to sinks planted to clean seed was greater than that to sinks planted with bean seeds inoculated with a slurry of pulverized brown spot diseased bean leaves, sinks planted 3 weeks before sources, and sinks planted to a snap bean cultivar that does not support large numbers of P. syringae pv. syringae. Based of these results, we conclude that the small amount of dispersal that occurred on the scale studied was sufficient to support extensive spread, and suitability of the habitat for multiplication of P. syringae pv. syringae strongly influenced the amount of spread.

Two sampling strategies were compared and sources of variability in the sampling protocols analyzed to optimize sampling methods for studies of cranberry fruit rot that occurs in the field (i.e., field rot). For the first method, fruit were dry picked by hand from randomly assigned quadrats; for the second method, fruit were scooped from harvest floodwaters. Rot incidence, which ranged from 1.8 to 9.7%, did not differ significantly between upland and lowland sites or, in general, between dry-picked and wet-harvested samples. There were no consistent differences between upland and lowland sites in the frequency of isolation of any fungus from either rotten or sound fruit. The incidence of certain saprophytic and soilborne fungi was greater in wet-harvested compared with dry-picked fruit. In general, rot incidence and incidence of various fungal taxa isolated from fruit varied more among samples within sites than among sites. Site type (i.e., upland or lowland) was never a major source of variability. These findings suggest that if the goal were to assess the occurrence of cranberry fruit rot within a region, intensive within-site sampling would be necessary, but site type would not be an important consideration, at least in Wisconsin, where this study was conducted.

Objective: To evaluate whether body size and anatomic site influence the quantity of bone microdamage in dogs without osteosarcoma (OS)., Sample Population: Pairs of radii were collected from 10 small dogs (< 15 kg) and 10 large dogs (> 25 kg)., Procedure: Specimens were stained in basic fuchsin for bone microdamage. Transverse sections were cut from each proximal and distal radial metaphysis at 15 and 85% of bone length. The following variables were determined for each region: mean microcrack length (CrLe, microM), microcrack density (CrDn, microcracks/mm2), microcrack surface density (CrSDn, microm/mm2), and estimated activation frequency (Acf, microcracks/mm2/y)., Results: Metaphyseal region did not significantly influence CrDn, CrLe, and CrSDn. The CrDn and CrSDn were influenced by body size, with microdamage being increased in large dogs, compared with small dogs. However, mean CrLe was not significantly influenced by body size. Acf significantly decreased with age and was significantly decreased in large dogs and in the distal radial metaphysis, compared with small dogs and the proximal radial metaphysis, respectively., Conclusions and Clinical Relevance: Our data did not reveal an increase in microdamage or remodeling at the OS predilection site (ie, the distal metaphysis of the radius), suggesting that induction of microdamage and an associated increase in bone remodeling are unlikely to be an important risk factor for induction of OS.

Nerve growth factor (NGF) synthesized in peripheral organs plays a critical role in the development and maintenance of the nervous system and also participates in processing nociceptive stimuli. Previous studies suggest that reproductive hormones may regulate the expression of NGF. Ovariectomies were performed on female mice, and mice were killed 24 h after hormone replacement to evaluate the effects of estrogen and progesterone on NGF in peripheral organs, specifically the uterus, bladder, heart, and salivary gland. Sham-operated intact mice and untreated ovariectomized mice served as controls. Immunohistochemistry demonstrated the presence of NGF, estrogen receptor-alpha, estrogen receptor-beta, and progesterone receptors in these organs. Ovariectomy caused a significant decrease in NGF protein content in the uterus, and short term treatment of ovariectomized mice with estrogen and/or progesterone increased uterine NGF mRNA and restored NGF protein to concentrations similar to intact control mice. Ovariectomy did not affect NGF protein concentrations in the salivary gland, but treatment of ovariectomized mice with estrogen alone or in conjunction with progesterone stimulated concentrations of NGF protein that exceeded those observed in intact control or ovariectomized, untreated mice. NGF mRNA was increased in salivary glands from ovariectomized mice treated with progesterone alone or in combination with estrogen relative to other groups. NGF protein content of the hearts of ovariectomized mice treated with estrogen alone or in conjunction with progesterone was increased relative to intact controls and ovariectomized, untreated mice, but neither ovariectomy or hormone replacement affected NGF mRNA content in the heart. NGF protein content of the bladder was unaffected by ovariectomy or hormone treatment, and bladder NGF mRNA was unaffected by ovariectomy or hormone treatment. Collectively, these results indicate that reproductive hormones have the capacity to regulate NGF message and protein in a manner that varies among organs. Fluctuations in the expression of NGF, in conjunction with other factors, may help to explain gender differences in pain sensation and inflammatory response.
