11. In vivo imaging of TSPO in mouse model of neuroinflammation using small-animal PET with [18F]DPA-714

Abstract

Introduction: Neuroinflammation (NI) may play an important role in stroke, neuroinflammatory and neurodegenerative diseases, involving microglia, astrocytes and peripheral immune cells. The translocator protein 18 kDa (TSPO) is an interesting marker of in vivo neuroinflammation because it is highly expressed by activated microglia under pathological conditions. In vivo PET imaging may provide new insight on the dynamic, topography and extent of NI since the earliest stage. Aim: To validate biodistribution and specific binding of [18F]DPA-714, a promising fluorinated radiotracer that targets the TSPO, in healthy mice and in mouse models of neurodegeneration and neuroinflammation using small-animals PET and ex-vivo methods. Methods: PET/CT was used to measure kinetics of [18F]DPA-714 in brain and peripheral tissues of normal mice, while specific binding to TSPO was evaluated using competitive studies with 1mg/kg of DPA-714 and 3mg/kg of PK11195. Metabolites assay was performed in plasma and peripheral organs by radio-HPLC. For mouse models of amyotrophic lateral sclerosis (SOD1 G93A) and of multiple sclerosis (PLP-EAE) and their control animals, in vivo [18F]DPA-714 PET/CT and ex-vivo immunofluorescence studies were performed. Results: In healthy mice [18F]DPA-714 reached high concentration in lung, heart, kidney and spleen, tissues well known to be rich in TSPO sites. Abstract

Introduction: The translocator protein 18 kDa (TSPO), a biochemical marker of neuroinflammation, is highly expressed in the brain activated microglia and it is also expressed by peripheral inflammatory cells and normal peripheral tissues. Thus, development of radioligands for the TSPO may contribute to further understanding the in vivo TSPO function in central and peripheral inflammatory processes and other pathologies. Here, we report the biodistribution, the specific binding and the radiometabolites of [(18)F]DPA-714, a promising fluorinated PET radiotracer, in normal mice using a microPET/CT scanner. Methods: The in vivo biodistribution and kinetics of [(18)F]DPA-714 were measured in mice brain and peripheral tissues. Abstract

Insulin-like growth factor binding protein 6 (IGFBP6) is a secreted protein with a controversial role in human malignancies, being downregulated in most types of human cancer, but upregulated in selected tumors. Ovarian cancer (OC) is a human malignancy characterized by IGFBP6 downregulation; however, the significance of its low expression during ovarian carcinogenesis is still poorly understood. In the present study, IGFBP6 expression and activation of its associated signaling pathway were evaluated in two matched OC cell lines derived from a high-grade serous OC before and after platinum resistance (PEA1 and PEA2 cells, respectively). A whole genome gene expression analysis was comparatively performed in both cell lines upon IGFBP6 stimulation using Illumina technology. IGFBP6 gene expression data from human OC cases were obtained from public datasets. Gene expression data from public datasets confirmed the downregulation of IGFBP6 in primary and metastatic OC tissues compared with in normal ovarian tissues. The comparative analysis of platinum-sensitive (PEA1) and platinum-resistant (PEA2) cell lines showed quantitative and qualitative differences in the activation of IGFBP6 signaling. Notably, IGFBP6 enhanced ERK1/2 phosphorylation only in PEA1 cells, and induced more evident and significant gene expression reprogramming in PEA1 cells compared with in PEA2 cells. Furthermore, the analysis of selected genes modulated by IGFBP6 (i.e., FOS, JUN, TNF, IL6, IL8 and EGR1) exhibited an inverse regulation in PEA1 versus PEA2 cells. In addition, selected hallmarks (TNFA_signaling_via_NFKB, TGF_beta_signaling, P53_pathway) and IL-6 signaling were positively regulated in PEA1 cells, whereas they were inhibited in PEA2 cells in response to IGFBP6. These data suggested that dysregulation of IGFBP6 signaling may serve a role in the progression of OC, and is likely associated with the development of platinum resistance. [ABSTRACT FROM AUTHOR]

Abstract

Background: Preclinical perfusion studies are useful for the improvement of diagnosis and therapy in dermatologic, cardiovascular and rheumatic human diseases. The Laser Doppler Perfusion Imaging (LDPI) technique has been used to evaluate superficial alterations of the skin microcirculation in surgically induced murine hindlimb ischemia. We assessed the reproducibility and the accuracy of LDPI acquisitions and identified several critical factors that could affect LDPI measurements in mice., Methods: Twenty mice were analysed. Statistical standardisation and a repeatability and reproducibility analysis were performed on mouse perfusion signals with respect to differences in body temperature, the presence or absence of hair, the type of anaesthesia used for LDPI measurements and the position of the mouse body., Results: We found excellent correlations among measurements made by the same operator (i.e., repeatability) under the same experimental conditions and by two different operators (i.e., reproducibility). A Bland-Altman analysis showed the absence of bias in repeatability (p = 0.29) or reproducibility (p = 0.89). The limits of agreement for repeatability were -0.357 and -0.033, and for reproducibility, they were -0.270 and 0.238. Significant differences in perfusion values were observed in different experimental groups., Conclusions: Different experimental conditions must be considered as a starting point for the evaluation of new drugs and strategic therapies.
