Abstract Introduction Nowadays, there are no strong diabetic pig models, yet they are required for various types of diabetes research. Using cutting‐edge techniques, we attempted to develop a type 2 diabetic minipig model in this study by combining a partial pancreatectomy (Px) with an energetic overload administered either orally or parenterally. Methods Different groups of minipigs, including Göttingen‐like (GL, n = 17) and Ossabaw (O, n = 4), were developed. Prior to and following each intervention, metabolic assessments were conducted. First, the metabolic responses of the Göttingen‐like (n = 3) and Ossabaw (n = 4) strains to a 2‐month High‐Fat, High‐Sucrose diet (HFHSD) were compared. Then, other groups of GL minipigs were established: with a single Px (n = 10), a Px combined with a 2‐month HFHSD (n = 6), and long‐term intraportal glucose and lipid infusions that were either preceded by a Px (n = 4) or not (n = 4). Results After the 2‐month HFHSD, there was no discernible change between the GL and O minipigs. A direct method to study essential genes is to construct conditional knock-down mutants by replacement of their native promoter by an inducible one. In Mycobacteria, replacement of an essential gene promoter with an anhydrotetracycline inducible one was successfully used but required a multi-step approach. In this work, we describe a gene cassette for the engineering of a conditional knock-down mutant, which allows the one-step targeted replacement of mycobacterial promoters by an anhydrotetracycline-inducible promoter. The functionality of this cassette was successfully tested by engineering conditional clpP and SecA1 mutants of Mycobacterium smegmatis.

We investigated the kinetics, mechanism and secondary organic aerosols formation of the ozonolysis of trans-2-pentenal (T2P) using four different reactors with Fourier Transform InfraRed (FTIR) spectroscopy and Gas Chromatography (GC) techniques at T = 298 ± 2 K and 760 Torr in dry conditions. The rate coefficients and branching ratios were also evaluated using the canonical variational transition (CVT) state theory coupled with small curvature tunneling (CVT/SCT) in the range 278–350 K. The experimental rate coefficient at 298 K was (1.46 ± 0.17) × 10−18 cm3 molecule−1 s−1, in good agreement with the theoretical rate. The two primary carbonyls formation yields, glyoxal and propanal, were 57 ± 10% and 42 ± 12%, respectively, with OH scavenger compared to 38 ± 8% for glyoxal and 26 ± 5% for propanal without OH scavenger. Acetaldehyde and 2-hydroxypropanal were also identified and quantified with yields of 9 ± 3% and 5 ± 2%, respectively, in the presence of OH scavenger. For the OH production, an upper limit of 24% was estimated using mesitylene as OH tracer. Combining experimental and theoretical findings enabled the establishment of a chemical mechanism. Finally, the SOA formation was observed with mass yields of about 1.5%. This work provides additional information on the effect of the aldehyde functional group on the fragmentation of the primary ozonide.

Through their involvement in the integration and excision of a large number of mobile genetic elements, such as phages and integrative and conjugative elements (ICEs), site‐specific recombination systems based on heterobivalent tyrosine recombinases play a major role in genome dynamics and evolution. However, despite hundreds of these systems having been identified in genome databases, very few have been described in detail, with none from phages that infect Bacillota (formerly Firmicutes). In this study, we reanalyzed the recombination module of Lactobacillus delbrueckii subsp. bulgaricus phage mv4, previously considered atypical compared with classical systems. Our results reveal that mv4 integrase is a 369 aa protein with all the structural hallmarks of recombinases from the Tn916 family and that it cooperatively interacts with its recombination sites. Using randomized DNA libraries, NGS sequencing, and other molecular approaches, we show that the 21‐bp core‐attP and attB sites have structural similarities to classical systems only if considering the nucleotide degeneracy, with two 7‐bp inverted regions corresponding to mv4Int core‐binding sites surrounding a 7‐bp strand‐exchange region. We also examined the different compositional constraints in the core‐binding regions, which define the sequence space of permissible recombination sites. [ABSTRACT FROM AUTHOR]

Inhalation of welding fumes (WFs) containing high levels of transition metals (Cr, Cu, Fe, Mn, Ni...) is associated with numerous health effects including oxidative stress. However, the measurements of the oxidative potential (OP) and bioaccessibility of WF transition metals depend on several physicochemical parameters and may be subject to several experimental artifacts. In this work, we investigated the influence of the experimental conditions that may affect the bioaccessibility of transition metals and their OP on stainless-steel WF extracts. WFs were produced using a generation bench and sampled on filters. The soluble fraction of the metals was analysed. Two different extraction fluids mimicking physiological pulmonary conditions were studied: phosphate buffer and Hatch's solution. Three extraction times were tested to determine the optimal time for a significant OPDTT using the dithiothreitol (DTT) method. The storage conditions of WFs after filter sampling such as duration, temperature and atmospheric conditions were investigated. The results indicate that experimental conditions can significantly affect the OPDTT and metal bioaccessibility analyses. Cr, Cu and Ni show higher solubility in Hatch's solution than in the phosphate buffer. Mn is highly sensitive to DTT and shows close solubility in the two fluids. An extraction time of 0.5 h in phosphate buffer allows a better sensitivity to OPDTT, probably by limiting complexations, interactions between metals and precipitation. Storage time and temperature can influence the physical or chemical evolution of the WFs, which can affect their OPDTT and Mn solubility. However, storage under N2(g) limits these changes. On-line measurements of OPDTT could provide an alternative to filter sampling to overcome these artifacts. [ABSTRACT FROM AUTHOR]

Galactosaminogalactan (GAG) is an insoluble aminosugar polymer produced by Aspergillus fumigatus and has anti-inflammatory properties. Here, the minimum glycosidic sequences required for the induction of IL-1Ra by peripheral blood mononuclear cells (PBMCs) was investigated. Using chemical degradation of native GAG to isolate soluble oligomers, we have found that the de-N-acetylation of galactosamine residues and the size of oligomer are critical for the in vitro immune response. A minimal oligomer size of 20 galactosamine residues is required for the anti-inflammatory response but the presence of galactose residues is not necessary. In a Dextran sulfate induced colitis mouse model, a fraction of de-N-acetylated oligomers of 13 < dp < 20 rescue inflammatory damage like the native GAG polymer in an IL-1Ra dependent pathway. Our results demonstrate the therapeutic suitability of water-soluble GAG oligosaccharides in IL-1 mediated hyper-inflammatory diseases and suggest that α-1,4-galactosamine oligomers chemically synthesized could represent new anti-inflammatory glycodrugs.

Previous studies have provided some insight into the Saharan dust deposition at a few specific locations from observations over long time periods or intensive field campaigns. However, no assessment of the dust deposition temporal variability in connection with its regional spatial distribution has been achieved so far from network observations over more than 1 year. To investigate dust deposition dynamics at the regional scale, five automatic deposition collectors named CARAGA (Collecteur Automatique de Retombées Atmosphériques insolubles à Grande Autonomie in French) have been deployed in the western Mediterranean region during 1 to 3 years depending on the station. The sites include, from south to north, Lampedusa, Majorca, Corsica, Frioul and Le Casset (southern French Alps). Deposition measurements are performed on a common weekly period at the five sites. The mean dust deposition fluxes are higher close to the northern African coasts and decrease following a south–north gradient, with values from 7.4 g m−2 year−1 in Lampedusa (35°31′ N, 12°37′ E) to 1 g m−2 year−1 in Le Casset (44°59′ N, 6°28′ E). The maximum deposition flux recorded is of 3.2 g m−2 wk−1 in Majorca with only two other events showing more than 1 g m−2 wk−1 in Lampedusa, and a maximum of 0.5 g m−2 wk−1 in Corsica. The maximum value of 2.1 g m−2 year−1 observed in Corsica in 2013 is much lower than existing records in the area over the 3 previous decades (11–14 g m−2 year−1). From the 537 available samples, 98 major Saharan dust deposition events have been identified in the records between 2011 and 2013. Complementary observations provided by both satellite and air mass trajectories are used to identify the dust provenance areas and the transport pathways from the Sahara to the stations for the studied period. Despite the large size of African dust plumes detected by satellites, more than 80 % of the major dust deposition events are recorded at only one station, suggesting that the dust provenance, transport and deposition processes (i.e. wet vs. dry) of dust are different and specific for the different deposition sites in the Mediterranean studied area. The results tend to indicate that wet deposition is the main form of deposition for mineral dust in the western Mediterranean basin, but the contribution of dry deposition (in the sense that no precipitation was detected at the surface) is far from being negligible, and contributes 10 to 46 % to the major dust deposition events, depending on the sampling site.

To evaluate the chemical behavior and the health impact of welding fumes (WF), a complex and heterogeneous mixture of particulate metal oxides, two certified reference materials (CRMs) were tested: mild steel WF (MSWF-1) and stainless steel WF (SSWF-1). We determined their total chemical composition, their solubility, and their oxidative potential in a phosphate buffer (PB) solution under physiological conditions (pH 7.4 and 37 °C). The oxidative potential (OPDTT) of WF CRMs was evaluated using an acellular method by following the dithiothreitol (DTT) consumption rate (µmol DTT L−1 min−1). Pure metal salts present in the PB soluble fraction of the WF CRMs were tested individually at equivalent molarity to estimate their specific contribution to the total OPDTT. The metal composition of MSWF-1 consisted mainly of Fe, Zn, Mn, and Cu and the SSWF-1 composition consisted mainly of Fe, Mn, Cr, Ni, Cu, and Zn, in diminishing order. The metal PB solubility decreased from Cu (11%) to Fe (approximately 0.2%) for MSWF-1 and from Mn (9%) to Fe (DTT of SSWF-1 is 2.2 times the OPDTT of MSWF-1 due to the difference in oxidative capacity of soluble transition metals. Cu (II) and Mn (II) are the most sensitive towards DTT while Cr (VI), Fe (III), and Zn (II) are barely reactive, even at higher concentrations. The OPDTT measured for both WF CRMs extracts compare well with simulated extracts containing the main metals at their respective PB-soluble concentrations. The most soluble transition metals in the simulated extract, Mn (II) and Cu (II), were the main contributors to OPDTT in WF CRMs extracts. Mn (II), Cu (II), and Ni (II) might enhance the DTT oxidation by a redox catalytic reaction. However, summing the main individual soluble metal DTT response induces a large overestimation probably linked to modifications in the speciation of various metals when mixed. The complexation of metals with different ligands present in solution and the interaction between metals in the PB-soluble fraction are important phenomena that can influence OPDTT depletion and therefore the potential health effect of inhaled WF.

A novel mechanism is revealed by which clinical isolates of adherent-invasive Escherichia coli (AIEC) penetrate into the epithelial cell layer, replicate, and establish biofilms in Crohn's disease. AIEC uses the FimH fimbrial adhesin to bind to oligomannose glycans on the surface of host cells. Oligomannose glycans exposed on early apoptotic cells are the preferred binding targets of AIEC, so apoptotic cells serve as potential entry points for bacteria into the epithelial cell layer. Thereafter, the bacteria propagate laterally in the epithelial intercellular spaces. We demonstrate oligomannosylation at two distinct sites of a glycoprotein receptor for AIEC, carcinoembryonic antigen related cell adhesion molecule 6 (CEACAM6 or CD66c), on human intestinal epithelia. After bacterial binding, FimH interacts with CEACAM6, which then clusters. The presence of the highest-affinity epitope for FimH, oligomannose-5, on CEACAM6 is demonstrated using LC-MS/MS. As mannose-dependent infections are abundant, this mechanism might also be used by other adherent-invasive pathogens.

The reactions of five structurally similar unsaturated alcohols, i.e., (Z)-2-penten-1-ol, (E)-2-hexen-1-ol, (E)-3-hexen-1-ol, (Z)-3-hexen-1-ol, and 1-octen-3-ol, with Cl atoms in the gas phase, were investigated at 296 ± 2 K and 1 atm by the relative-rate kinetic technique using a 600-L Teflon reaction chamber. Selected ion flow tube mass spectrometry (SIFT-MS) was used simultaneously to monitor the decay of the alcohols of interest and selected reference compounds. Tetrahydrofuran (THF), propan-1-ol, and octane were used as reference compounds. Chlorine atoms were produced by the photolysis of molecular chlorine (Cl2) using broadband actinic lamps near 365 nm. The estimated rate constant values (in 10−10 cm3∙molecule−1∙s−1) followed the order 2.99 ± 0.53 ((Z)-2-penten-1-ol) < 3.05 ± 0.59 ((E)-3-hexen-1-ol) < 3.15 ± 0.58 ((Z)-3-hexen-1-ol) < 3.41 ± 0.65 ((E)-2-hexen-1-ol) < 4.03 ± 0.77 (1-octen-3-ol). The present work provides the first value of the rate constant for the reaction of 1-octen-3-ol with Cl atoms. The results are discussed and interpreted in relation to other studies where literature data are available. The structure−activity relationship and the atmospheric implications are discussed as well.
