Many desert fishes, which evolved in isolated aquatic “islands” with limited predation pressure, have been severely impacted by non-native predators. These impacts have been attributed to the evolutionary loss of antipredator competence, known as the predator naiveté hypothesis. Recent work provided support for this hypothesis for one desert fish species. We sought to examine the generality of the predator naiveté hypothesis by evaluating antipredator competence in five populations of Red River pupfish (Cyprinodon rubrofluviatilis), a species that occupies habitats that vary in the degree of isolation and levels of fish species richness. Fish were exposed to a conspecific chemical alarm cue released from damaged epidermal tissue as a general assay of antipredator response. We found that pupfish from all five populations exhibited antipredator behavior in response to the alarm cue, regardless of the exposure to predation risk. These data provide evidence that antipredator responses to alarm cues are conserved in Red River pupfish, even in populations isolated from piscivorous species.

Water quality and quantity within the upper San Antonio River (Bexar County, Texas) were supported historically from spring flows of the Edwards Aquifer. Since the 1700s, water quality, quantity, physical aquatic habitats, and presumably fish communities have been altered in the upper San Antonio River, including loss of spring flow and replacement of base flow by treated wastewater. The upper San Antonio river fish community provides a unique opportunity to fill a gap in knowledge on spring-associated fish responses to spring flow loss. The purpose of our study was to describe temporal changes in the upper San Antonio River fish community in relation to increased anthropogenic alterations from reported fish collections and infer from reference conditions. Comparisons with reference conditions were necessary since anthropogenic alterations occurred before the first fish collections within the basin. We found the upper San Antonio River fish community changed through time with increases in native and introduced fishes and decreases in spring-associated fish richness and relative abundances. Decreases in spring-associated fish richness and relative abundances were attributed to decreases in spring flow and to changes in water quality; however, specific mechanisms between loss of spring flow and spring-associated fishes are unknown at this time. Quantifying historical fish community changes through time in systems with a long history of urbanization is difficult, but our assessment provides a greater understanding of how spring flow loss due to increasing water demand in highly urbanized areas can alter communities with spring-associated species.

Pelagic broadcast spawning cyprinids are a mobile group of fishes commonly found within semi-arid areas of the Great Plains of North America. In several species, upstream movements of 50 to 200 km have been reported and associated with reproductive migration. Long distance migrations also are associated with finding food and refuge. Goals of this study were to detect synchronous movement of Prairie Chub at specific life stages and relate movement to functions associated with migration (i.e., reproduction, feeding, and refuge). Movement was inferred by quantifying monthly occurrences of age groups using length frequency distributions among five sites in two rivers and supported with range-wide assessment of genetic structuring. Monthly occurrences of age groups among sites were related to reproductive season (i.e., gonadosomatic indices, ovarian condition) for assessing reproductive migration, to food consumption (i.e., gut content assessments) for assessing feeding migration, and to habitat associations (e.g., specific conductance) for assessing refuge migration. Spatial and temporal patterns in age-0, age-1 and age-2 fish occurrences and abundances suggested movement within only one river. Non-synchronized movements were more consistently associated with specific conductance following a high flow event, which was more consistent with a refuge function than reproductive season or food consumption. Our findings suggest that Prairie Chubs are capable of moving long distances, but movement was inconsistently associated with reproductive efforts as previously described. Without synchronized movements, we question whether movement is migration or dispersal and provide a contrasting narrative to the drift compensation theory used to describe broadcast spawning cyprinid migrations.

Purposes of this study were to assess population status and life history attributes of the Texas Shiner Notropis amabilis, a minnow endemic to the Edwards Plateau region in Texas and the Rio Grande drainage of U.S.A. and Mexico. Despite possible extirpation from three stream reaches, populations of Texas Shiners persist with abundances categorized as occasional and frequent among multiple and independent stream reaches, streams, and drainages. Therefore, Texas Shiner population is considered stable. Quantification of life history traits suggest Texas Shiner has a protracted reproductive season (i.e., 9 mo), a life span of 2 y, and feeds on primarily aquatic invertebrates. Protracted spawning seasons are consistent with Texas Shiner's reported associations with spring systems of the Edwards Plateau. Current and future conservation of N. amabilis and other spring-associated fishes are explicitly linked to spring complexes, although exact mechanisms of association are unknown.

Species checklists and dichotomous keys are valuable tools that provide many services for ecological studies and management through tracking native and non-native species through time. We developed nine drainage basin checklists and dichotomous keys for 196 inland fishes of Texas, consisting of 171 native fishes and 25 non-native fishes. Our checklists were updated from previous checklists and revised using reports of new established native and non-native fishes in Texas, reports of new fish occurrences among drainages, and changes in species taxonomic nomenclature. We provided the first dichotomous keys for major drainage basins in Texas. Among the 171 native inland fishes, 6 species are considered extinct or extirpated, 13 species are listed as threatened or endangered by U.S. Fish and Wildlife Service, and 59 species are listed as Species of Greatest Conservation Need (SGCN) by the state of Texas. Red River drainage basin was the most speciose with 120 fishes. Rio Grande & Pecos drainage basin had the greatest number of threatened or endangered fishes (N = 7) and the greatest number of SGCN fishes (N = 28). We revised drainage basin occurrences for 77 species. Drainage basin checklists and dichotomous keys provide finer resolution of species distributions within the geopolitical boundaries of Texas and can reduce probability of errors in fish identification errors by removing species not occurring within a natural boundary.

It has long been recognized that the process of preserving biological specimens results in alterations of body shape, though detailed studies examining the degree to which morphological changes occur throughout the preservation process are lacking. We utilize geometric morphometric analyses, an increasingly common tool for examining shape variation in a wide variety of biological disciplines, to examine the effects of formalin and ethanol preservation on the body shape of 10 freshwater fish species over time: from fresh specimens to eight weeks after preservation. We found significant changes in body shape among fresh and formalin fixed specimens. Furthermore, changes in body shape continue to occur after subsequent ethanol preservation. Two fish species collected at multiple localities show significant morphological differences for a limited number of morphometric characters. However, the significance, or lack thereof, often changed inconsistently from one stage of preservation to another. We conclude that morphometric analyses would ideally be performed on fresh specimens. However, recognizing that this is not always feasible, it is important to be aware of the morphometric changes that can occur during preservation.

We report the occurrence of a single population of Etheostoma parvipinne (Goldstripe Darter) in a small, acidic, spring-fed stream within the Colorado River drainage following an extended period of below-average precipitation (January–September 2011), a wildland fire (September 2011), and subsequent debris and sediment flows (November 2011–May 2012). Goldstripe Darters were taken from Alum Creek (Bastrop County, TX) along with a cyprinid, a poeciliid, and several species of centrarchids in June 2012. Occurrence of Goldstripe Darters in Alum Creek provides a regional example of fish-community responses to wildland fires. In additio n, understanding the mechanism of long-term persistence in a single tributary on the periphery of a species range might offer insight into the origin of endemic fishes and persistence of disjunct fish populations within aquatic evolutionary refugia of central Texas.

Within its southwestern native range, Ameiurus nebulosus (Brown Bullhead) had been found in the Red River drainage within Oklahoma, Arkansas, and Louisiana but not Texas. We report for the first time the occurrence of Brown Bullhead within the Cypress River system of Texas, a tributary of the Red River. We tentatively conclude that the Cypress River system of Texas is within the natural range of Brown Bullhead, but we cannot exclude human introduction as a possible mechanism for its occurrence, given that Brown Bullhead are introduced worldwide.

Aquifers and spring complexes within the arid and semi-arid karst Edwards Plateau region of Texas are evolutionary refugia supporting a large number of endemic flora and fauna. Spring fishes, many of which are endemic, are associated with the spring complexes, whereas river fishes generally avoid spring complexes. Purposes of this study were to experimentally test affinities of spring fishes and river fishes for spring complexes and rivers among six independent spring-river systems of the Edwards Plateau and to quantitate relationship among spring fishes, river fishes and spring flow magnitude within spring complexes. We found that spring fishes did not exclusively use spring complexes but were more abundant in spring complexes than rivers and that species richness, relative abundance, and density of spring fishes were directly related to spring flow magnitude. Patterns in affinities for river fishes were less distinct. Quantitation of fish–habitat relationships will assist efforts in conserving aquatic resources within the Edwards Plateau and provides predictive models to assess biological integrity of other spring complexes. In addition, consistency in distributional patterns of spring and river fishes provides a framework for testing underlying processes of species segregation and diversification, establishes boundaries of aquatic evolutionary refugia, and expands the value of evolutionary refugia from historical refugia to contemporary and future refugia for Edwards Plateau aquatic biota.

beta-peptides possess several features that are desirable in peptidomimetics: they are easily synthesized, fold into stable secondary structures in physiologic buffers, and resist proteolysis. They can also bind to a diverse array of proteins to inhibit their Interactions with alpha-helical ligands. beta-peptides are usually not cell-permeable, however, and this feature limits their utility as research tools and potential therapeutics. Appending an Arg(8) sequence to a beta-peptide improves Uptake but adds considerable mass. We previously reported that embedding, a small cationic patch within a PPII, alpha-, or beta-peptide helix improves uptake without the addition of significant mass. In another mass-neutral strategy, Verdine, Walensky, and others have reported that insertion of a hydrocarbon bridge between the i and i + 4 positions of an alpha-helix also increases cell Uptake. Here we describe a series of beta-peptides containing diether and hydrocarbon bridges and compare them on the basis of cell uptake and localization, affinities for hDM2, and 14-helix structure. Our results highlight the relative merits of the cationic-patch and hydrophobic-bridge strategies for improving beta-peptide Uptake and identify a surprising correlation between uptake efficiency and hDM2 affinity.

Previous work has shown that certain β(3)-peptides can effectively mimic the side chain display of an α-helix and inhibit interactions between proteins, both in vitro and in cultured cells. Here we describe a β(3)-peptide analog of GLP-1, CC-3(Act), that interacts with the GLP-1R extracellular domain (nGLP-1R) in vitro in a manner that competes with exendin-4 and induces GLP-1R-dependent cAMP signaling in cultured CHO-K1 cells expressing GLP-1R.

Advances in the foldamer field in recent years are as diverse as the backbones of which they are composed. Applications have ranged from cellular penetration and membrane disruption to discrete molecular recognition, while efforts to control the complex geometric shape of foldamers has entered the realm of tertiary and quaternary structure. This review will provide recent examples of progress in the foldamer field, highlighting the significance of this class of compounds and the advances that have been made towards exploiting their full potential.

We recently described the high-resolution X-ray structure of a helical bundle composed of eight copies of the β-peptide Zwit-1F. Like many proteins in Nature, the Zwit-1F octamer contains parallel and antiparallel helices, extensive inter-helical electrostatic interactions, and a solvent-excluded hydrophobic core. Here we explore the stability of the Zwit-1F octamer using circular dichroism (CD) spectroscopy, analytical ultracentrifugation (AU), differential scanning calorimetry (DSC), and NMR. These studies demonstrate that the thermodynamic and kinetic properties of Zwit-1F closely resemble those of α-helical bundle proteins. Together these studies should provide a model for the design of β-peptide proteins with biological functions.

We reported recently that certain beta-peptides self-assemble spontaneously into cooperatively folded bundles whose kinetic and thermodynamic metrics mirror those of natural helix bundle proteins. The structures of four such beta-peptide bundles are known in atomic detail. These structures reveal a solvent-sequestered, hydrophobic core stabilized by a unique arrangement of leucine side chains and backbone methylene groups. Here we report that this hydrophobic core can be re-engineered to contain a fluorous subdomain while maintaining the characteristic beta-peptide bundle fold. Like alpha-helical bundles possessing fluorous cores, fluorous beta-peptide bundles are stabilized relative to hydrocarbon analogues and undergo cold denaturation. Beta-peptide bundles with fluorous cores represent the essential first step in the synthesis of orthogonal protein assemblies that can sequester selectively in an interstitial membrane environment.
