2. Correlates of Using Medically-Authorized Cannabis in a Large Cohort of People Living with HIV Who Use Cannabis.

- Abstract
Many people living with HIV (PLWH) use cannabis to manage symptoms, but a large proportion do so without medical cannabis authorization and use cannabis obtained outside the medical stream. In jurisdictions where non-medical cannabis use is legal, PLWH who hold medical cannabis authorization may represent a unique subgroup; yet, research on the correlates of using medical cannabis (authorized by a healthcare provider) in the context of non-medical cannabis legalization is lacking. Thus, this study examined the cannabis- and health-related correlates of medical cannabis use among PLWH in Ontario, Canada, where non-medical cannabis is legal. PLWH (N = 868; 85.37% men, mean age 51.34 years [SD = 12.25]) who were enrolled in the Ontario HIV Treatment Network Cohort Study in 2022 and who reported past-year cannabis use completed an assessment of sociodemographic characteristics, HIV-related variables, cannabis and other substance use, and health-related quality of life. Relatively few participants (n = 122; 14.06%) reported any medical cannabis use, with most (n = 746; 85.94%) reporting exclusive use of non-medical cannabis. Logistic regression analyses showed that greater HIV symptom distress, poorer physical-health-related quality of life, more frequent cannabis use, and using smokeless forms of cannabis were each uniquely associated with increased likelihood of using medical cannabis relative to exclusively using non-medical cannabis. Results suggest that even in jurisdictions where non-medical cannabis is legal, a subset of PLWH continue to access cannabis through the medical stream, and these individuals report distinct patterns of cannabis use and poorer physical health. 3. Proximal antecedents and acute outcomes of simultaneous alcohol and cannabis use: Systematic review of daily- and event-level studies.

- Abstract
Many studies have revealed that individuals who engage in simultaneous use of alcohol and cannabis report elevated substance-related consequences relative to those who use only alcohol or cannabis; however, evidence from emerging studies examining within-person differences across simultaneous use and single substance use occasions is less consistent. This systematic review aimed to synthesize findings from existing day- and event-level studies of within-person differences in the proximal antecedents and acute outcomes associated with simultaneous use versus single substance use episodes. Our search strategy revealed 30 eligible articles. Two categories of antecedents (i.e., internal [e.g., motives] and external [e.g., social context]) and three categories of outcomes (i.e., consumption behavior, general positive and negative consequences, and specific consequences) were identified. The current literature consistently suggests that greater day- or event-level social and enhancement motives, as well as being in a social context, predict greater likelihood of engaging in simultaneous use compared with alcohol- or cannabis-only use. However, there was heterogeneity in findings regarding the role of other person-level antecedents. Further, while most evidence pointed to heavier alcohol consumption on simultaneous use occasions versus alcohol-only occasions, findings for elevations in acute negative and positive substance-related consequences on simultaneous use versus single substance use occasions were mixed. Additionally, four studies found that increased consequences on simultaneous use occasions depended on the level of alcohol consumed. This review identifies several antecedents for simultaneous use events but suggests that simultaneous use occasions are not always associated with more acute harms than single substance use occasions. Given the extent to which the current literature is mixed, this review emphasizes the importance of methodological improvements and future research examining the mechanisms linking simultaneous use with substance-related consequences to help reconcile findings across within-person and between-person studies., (© 2024 The Author(s). 4. Prospective associations of behavioral economic demand for cannabis and alcohol with simultaneous cannabis and alcohol use among young adults.

Behavioral economic demand for cannabis and alcohol is robustly associated with cannabis use and alcohol use, respectively. However, few studies have examined the contributions of cannabis and alcohol demand to simultaneous cannabis and alcohol use, which is common among young adults. We examined prospective associations of cannabis demand and alcohol demand with propensity for simultaneous use (broadly defined as using both cannabis and alcohol in the same day) and with cannabis and alcohol consumption during simultaneous use days among young adults. Young adults reporting simultaneous use ( N = 107) completed a Marijuana Purchase Task assessing cannabis demand and an Alcohol Purchase Task assessing alcohol demand. They then completed daily smartphone surveys over 21 days assessing cannabis and alcohol use. Multilevel models revealed that higher cannabis demand (i.e., higher O
max , Pmax , and intensity; lower elasticity) was uniquely associated with greater propensity for simultaneous use relative to nonuse. In addition, higher alcohol demand (lower elasticity) was uniquely associated with greater propensity for simultaneous use relative to cannabis-only use, and higher cannabis demand (higher break point and intensity, lower elasticity) was uniquely associated with greater propensity for simultaneous use relative to alcohol-only use. Furthermore, in models limited to simultaneous use days, greater cannabis demand (higher Omax , lower elasticity) and lower alcohol demand (higher elasticity) were uniquely associated with greater overall cannabis flower consumption, and higher alcohol demand (higher Omax , lower elasticity) was uniquely associated with greater overall alcohol consumption. Results suggest that individual differences in cannabis and alcohol demand may contribute to simultaneous cannabis and alcohol use behaviors in a substance-specific pattern. 5. Within-person and between-person associations of access to environmental reward with alcohol and cannabis use and consequences among young adults.

- Abstract
Background: Recent behavioural economic models of substance use suggest that low access to environmental reward may increase risk for heavy substance use and associated harms. Most prior studies of these associations have been cross-sectional and have focused on alcohol. The current study extends this research using longitudinal data to examine the within-person and between-person associations of environmental reward access with both alcohol and cannabis outcomes., Method: Young adults (N = 119, 64.71 % female) completed an online survey at three time points, spaced six months apart. The survey included measures of alcohol and cannabis use and consequences, and two facets of environmental reward access: reward probability (i.e., likelihood of experiencing environmental reward) and environmental suppression (i.e., diminished availability of environmental reward)., Results: Multilevel models revealed that at the between-person level (i.e., averaged across time points), greater environmental suppression (but not reward probability) was significantly associated with more frequent cannabis use, and greater reward probability (but not environmental suppression) was significantly associated with heavier alcohol use. Higher environmental suppression (but not reward probability) was also associated with greater alcohol and cannabis consequences at the between-person level, over and above level of use. A significant within-person association also was observed, wherein participants reported relative increases in cannabis consequences during time periods when they also reported relative decreases in the availability of environmental reward., Conclusions: Results highlight environmental suppression as a risk factor for more frequent cannabis use and for both alcohol and cannabis consequences, and provide novel support for a within-person association between environmental suppression and cannabis consequences over time. - Abstract
The Gambling Craving Scale (GACS) is a multifaceted measure of gambling craving. Initial validation work by Young and Wohl (2009) in university student samples showed that the GACS had a three-factor structure capturing dimensions of Desire, Anticipation, and Relief. Despite its potential clinical utility as a measure of craving, the GACS has yet to be validated in people seeking treatment for gambling problems. Accordingly, we examined the psychometric properties in a sample of people (N = 209; M
age = 37.66; 62.2% female) participating in a randomized controlled trial testing a novel online treatment for problem gambling. We predicted the GACS would have a three-factor structure. In addition, we also examined measurement invariance across sex and problem gambling risk status. Finally, we assessed concurrent validity of the factors with other measures of problem gambling severity and involvement. Exploratory structural equation modeling findings supported a three-factor structure that was invariant across the groups tested. Each of the Desire, Anticipation, and Relief subscales were significant positive predictors of problem gambling severity and symptoms, and some form of gambling behaviour. Findings show the GACS is a promising scale to assess multidimensional craving experiences among people in treatment for gambling problems., (© 2024. 7. Energy requirement for primiparous Holstein × Gyr crossbred dairy cows.

Our objective was to estimate the requirements of metabolizable energy (ME) and NE
M of lactating and dry cows, the efficiency of ME utilization for milk production (kl ) and tissue gain (kg ), and the use of body energy mobilization for milk production (kt ) throughout the lactation of primiparous crossbred Holstein × Gyr cows, using open-circuit respiration chambers. Twenty-nine primiparous Holstein × Gyr crossbred cows with an initial BW averaging 563 ± 40.1 kg and 2.5 ± 0.09 yr old were used throughout lactation and dry periods. The cows were kept nonpregnant throughout the study to eliminate possible confounding factors. Apparent digestibility assays, followed by calorimeter measurements, were performed 6 times throughout the lactation period. In the dry-off period, the cows were also evaluated but fed with restricted intake (DMI = 1.1% BW/d) to achieve heat production close to maintenance. After 21 d of diet adaptation, an apparent digestibility assay followed by calorimeter measurements was performed. Parameter estimates for lactation period were obtained by mixed models including lactation stage as repeated measures. For restricted feeding at dry-off and fasting period assays, the requirements were estimated by exponential regression. For whole lactation, the values of the ME requirement for maintenance (MEM ) and NEM were 0.588 and 0.395 MJ/BW0.75 , respectively. The efficiencies of kl , kg , and kt were 0.672, 0.771, and 0.814, respectively. However, MEM and NEM were higher in early and mid lactation than late, whereas kl was higher in early than other lactation stages. Dry and nonpregnant cows had MEM of 0.434 MJ/BW0.75 and NEM of 0.351 MJ/BW0.75 for maintenance level, and MEM of 0.396 MJ/BW0.75 and NEM of 0.345 MJ/BW0.75 for fasting metabolism level, and efficiency of ME utilization for maintenance was 0.80. Our findings confirmed that F1 crossbred Holstein × Gyr dairy cows have differences in energy requirement and efficiency throughout the lactation stages, suggesting the use of different values in each stage. The estimated values of energy requirement for maintenance and efficiencies for primiparous lactating crossbred Holstein × Gyr were similar to those reported in the literature in specific studies and requirements systems., (The Authors. This study aimed to assess the effect of bulk tank milk (BTM), waste milk (WM), and pasteurized waste milk (PWM) on nutrient digestibility, ruminal and cecal fermentation, gastrointestinal tract (GIT) development, and antimicrobial resistance of fecal Escherichia coli from dairy calves at 2 periods (30 and 60 d of age). Calves were grouped according to BW, serum protein levels, and breed composition. Three treatments were included: BTM (n = 21), WM from cows under antibiotic treatment (n = 21), and PWM (WM submitted to HTST pasteurization; n = 21). A total of 63 calves were used, of which 18 animals (n = 6 per treatment) were evaluated in the period of 4 to 30 d, and 45 (n = 15 per treatment) from 4 to 60 d. During the experimental period, a daily intake of 6 L of milk was divided into 2 equal meals, with ad libitum access to water and starter. Milk and feed intakes were recorded daily. Apparent total-tract digestibility and nitrogen balance were conducted from 25 to 29 d of age (n = 6) and from 53 to 57 d of age (n = 15). Animals were slaughtered at 30 ± 1 and 60 ± 1 d of age for the assessment of ruminal and cecal fermentation and GIT development. Antimicrobial susceptibility testing was conducted at 1, 30, and 60 d of age (n = 15/treatment). Statistical analysis used a linear mixed-effects model for continuous outcomes and generalized linear models for single measurements (R software). Treatments WM and PWM had lower rumen pH, higher ruminal acetate concentration, larger reticulorumen and liver, and a higher prevalence of fecal-resistant E. coli compared with BTM at both 30 and 60 d. Up to 60 d, both BTM and WM treatments exhibited higher digestibility of ether extract and gross energy compared with the PWM, whereas WM and PWM treatments showed increased nitrogen intake and retention compared with the BTM treatment. These findings suggest that pasteurization of WM negatively affects nutrient digestibility and calf performance, while also affecting rumen development. Additionally, the use of milk containing antibiotic residue leads to the selection of resistant E. coli in the GIT over time., (The Authors. 9. How Much THC Is in That Joint? A Daily Diary Study of Young Adults' Knowledge of the Cannabinoid Content of Cannabis Products.

Objective: Many young adults report frequent cannabis use and are at risk for cannabis harms. Knowledge of the tetrahydrocannabinol (THC) and cannabidiol (CBD) concentrations of cannabis products may promote harm reduction, but few studies have characterized cannabinoid concentration knowledge in this population. This study used day-level data to examine predictors of cannabinoid concentration knowledge and associations of cannabinoid concentration knowledge with substance-related consequences among young adults., Method: Participants ( n = 131; mean age = 22.11 years; 64.12% female) from a larger study of cannabis and alcohol co-use completed daily surveys over 21 days assessing knowledge of the cannabinoid concentrations of cannabis used, forms of cannabis used, motives for cannabis use (medicinal, nonmedicinal, both), and substance-related consequences., Results: On average, participants reported at least some knowledge of the THC and CBD concentrations of their cannabis on 48% and 32% of their cannabis use days, respectively. Generalized linear mixed models revealed that participants with a greater propensity to use nonflower (relative to flower) cannabis products and to report medicinal (relative to exclusively nonmedicinal) motives for cannabis use reported greater cannabinoid concentration knowledge overall across days, controlling for sociodemographic factors and level of cannabis involvement. Participants with greater overall cannabinoid concentration knowledge reported positive substance-related consequences more often. In addition, participants were more likely to report negative substance-related consequences on days during which cannabinoid concentrations were known versus unknown., Conclusions: Findings suggest that cannabinoid concentration knowledge may be higher among young adults who report primarily nonflower and medicinally motivated cannabis use, although cannabinoid concentration knowledge, alone, may not protect against negative substance-related consequences at the day level.

Behavioral economic demand for cannabis is robustly associated with cannabis consumption and cannabis use disorder (CUD). However, few studies have examined the processes underlying individual differences in the relative valuation of cannabis (i.e., demand). This study examined associations between executive functions and cannabis demand among young adults who use cannabis. We also examined indirect associations of executive functions with cannabis consumption and CUD symptoms through cannabis demand. Young adults ( N = 113; 58.4% female; mean age 22 years) completed a Marijuana Purchase Task. Participants also completed cognitive tasks assessing executive functions (set shifting, inhibitory control, working memory) and semistructured interviews assessing past 90-day cannabis consumption (number of grams used) and number of CUD symptoms. Poorer inhibitory control was significantly associated with greater O
max (peak expenditure on cannabis) and greater intensity (cannabis consumption at zero cost). Poorer working memory was significantly associated with lower elasticity (sensitivity of consumption to escalating cost). Lower inhibitory control was indirectly associated with greater cannabis consumption and CUD symptoms through greater Omax and intensity, and poorer working memory was indirectly associated with greater cannabis consumption and CUD symptoms through reduced elasticity. This study provides novel evidence that executive functions are associated with individual differences in cannabis demand. Moreover, these results suggest that cannabis demand could be a mechanism linking poorer executive functioning with heavier cannabis use and CUD, which should be confirmed in future longitudinal studies. 11. Association of passive immunity and genetic composition, health, and performance of tropical dairy calves.

This retrospective cohort study aimed to identify risk factors for the failure of transfer of passive immunity (FPI) in preweaned dairy calves, explore its associations with morbidity, mortality, genetics, and determine a standardized cut-off point for FPI. Analyzing data from 6011 calves, factors such as sire predicted transmitted ability for milk (PTA), birth season, retained placenta (RP), total serum protein concentration (TSP), morbidity (neonatal calf diarrhea - NCD, bovine respiratory disease - BRD, tick-borne disease - TBD), mortality, and average daily weight gain (ADG) were considered. The calves were categorized into predominantly Gyr (PG) and predominantly Holstein (PH) genetic composition groups. Multivariate mixed logistic regression revealed optimal TSP cut-off points for predicting morbidity (7.6 g/dL) and mortality (6.9 g/dL). PH calves exhibited 1.35 times higher odds of FPI and 1.48 times greater odds of disease. Calves from multiparous cows and those born to dams with RP had increased FPI odds. Disease prevalence was 53%, with 41% NCD, 18% BRD, and 10% TBD. Season, parity, PTA, and birth weight were associated with disease odds, though FPI was not a reliable predictor. The mortality percentage was 6%, with PH calves and those with a positive PTA having higher odds. The ADG was 0.64 kg, and FPI-affected calves gained less weight. The study compared and identified various risk factors that potentially impact calf immunity. 12. Interactive effects of alcohol and cannabis quantities in the prediction of same-day negative consequences among young adults.

Background: Simultaneous alcohol and cannabis use is common, but observational studies examining negative consequences of simultaneous use have rarely considered dose-related interactions between alcohol and cannabis. This study examined interactions between quantities of cannabis and alcohol consumed in predicting negative consequences on simultaneous use days., Methods: Young adults (N = 151; 64% female; 62% White) reporting recent simultaneous use and at least weekly alcohol and cannabis use completed 21 daily, smartphone-based surveys assessing previous day quantities of cannabis and alcohol used, types of cannabis used (flower, concentrates, edibles), and negative substance-related consequences. Multilevel models examined: (1) whether negative consequences differed within-person across simultaneous use days and single-substance use days; and (2) whether quantities of alcohol and cannabis consumed on simultaneous use days interacted, within-person, to predict negative consequences. We focused on quantities of cannabis flower (grams) in primary analyses and explored quantities of other forms of cannabis (concentrates, edibles) in supplementary analyses., Results: Participants reported fewer negative consequences on alcohol-only (243 observations) and cannabis-only (713 observations) days than they did on simultaneous use days (429 observations). On simultaneous use days involving cannabis flower use (313 observations across 81 participants), the within-person association between number of standard drinks and negative consequences was weaker on days during which larger (vs. smaller) amounts of cannabis flower were consumed. Inspection of simple slopes revealed that decreased alcohol use was associated with less of a decline in negative consequences when combined with relatively greater amounts of cannabis flower., Conclusions: Although simultaneous use was associated with more negative consequences than alcohol-only and cannabis-only use, negative consequences on simultaneous use days varied as a function of the interaction between alcohol and cannabis quantities. As findings suggest that using larger amounts of cannabis may attenuate declines in negative consequences associated with lighter drinking, interventions for higher-risk simultaneous use patterns may benefit from a focus on quantities of both alcohol and cannabis., (© 2024 The Authors. - Abstract
Diarrhea and respiratory diseases pose significant challenges in the rearing of pre-weaned calves, motivating the investigation of tools to improve gastrointestinal tract development, health, and overall performance in young calves. Consequently, the primary objective of this study was to assess the effectiveness of an additive incorporated into milk replacer to promote the development and health of the animals. Forty-six dairy calves were randomly assigned into two treatments: control (CON, n = 23; with 15 females and 8 males), and sodium butyrate (SB, n = 23; with 15 females and 8 males). The calves in the SB treatment group were supplemented with 4 g/d of unprotected sodium butyrate (Adimix, Adisseo, China), added to the milk replacer from 4 to 60 days of age. Water and starter were fed ad libitum. The study evaluated several parameters, including feed intake, nutrient digestibility, ruminal pH, ammonia and volatile fatty acids, blood metabolites (glucose, insulin-like growth factor type 1, urea, β-hydroxybutyrate), hemogram, health scores, performance, and feed efficiency. Bull calves were euthanized at 60 days of age for organ comparison, while heifer calves were assessed for carryover effects up to 90 days of age. Data were analyzed independently using linear mixed models using the nlme package in R, and the Artools package for non-parametric categorical outcomes. Although the feed intake and performance variables exhibited differences within weeks, no divergence was observed between treatment groups. Notably, a positive treatment-by-week interaction was identified for starter feed intake ( p = 0.02) and total dry matter intake ( p = 0.04) during pre-weaning for CON animals. Ruminal parameters, blood metabolites, and hemogram values such as glucose, urea, insulin-like growth factor type 1, mean corpuscular value, lymphocytes, and neutrophils displayed differences within weeks during the pre-weaning stage, but similar results within groups. No differences between supplemented and non-supplemented calves were found across nutrient digestibility, organ development, and histology. Regarding health scores, differences were noted within weeks for fecal and respiratory scores during the pre-weaning stage, and only the respiratory score during the post-weaning stage. Consequently, butyrate supplementation did not elicit improvements or negative effects in the body development or health status of dairy calves.

14. Screening for cannabis use disorder among young adults: Sensitivity, specificity, and item-level performance of the Cannabis Use Disorders Identification Test - Revised.

- Abstract
The Cannabis Use Disorders Identification Test Revised (CUDIT-R) is a widely used screening tool. However, its utility in screening for cannabis use disorder (CUD) among young adults requires further investigation. The current study evaluated the accuracy of the CUDIT-R in distinguishing between young adults with and without CUD. We also examined the individual item properties of the CUDIT-R in this sample. A community sample of young adults (N = 153, ages 19-26 years) who reported at least one lifetime period of regular cannabis use (i.e., twice or more per month for at least six months) completed the CUDIT-R and the Structured Clinical Interview for DSM-5 (SCID-5) Substance Use Module. The CUDIT-R showed good accuracy in distinguishing between young adults with and without CUD as determined by the SCID-5 (area under the receiver operating characteristic curve = 0.84). A cut-point of nine yielded 87.80 % sensitivity and 70.42 % specificity, with positive and negative predictive values of 77.42 % and 83.33 %, respectively. Item response theory analyses revealed that CUDIT-R items assessing impaired control, failure to meet expectations, time devoted to use, and memory and concentration problems displayed high discrimination and were relatively difficult, whereas items assessing frequency of use and attempts to reduce use showed moderate discrimination and relatively low difficulty. Items assessing hours high and use with potential for physical hazard displayed poor discrimination. The CUDIT-R appears to validly distinguish between young adults with and without CUD and may be useful in screening for CUD in this high-risk population. 15. Dispositional and contextual correlates of in-play sports betting and related harms: A 2-week ecological momentary assessment study.

Objective: In-play betting is a novel form of sports betting that allows players to make continuous bets during a game. The present study examined the dispositional and contextual correlates of in-play sports betting and related harms., Method: Participants were 84 individuals (73.81% men, M
age = 41.02) who engaged in in-play betting. Participants first completed an online questionnaire including measures of problem gambling severity, childhood trauma, impulsivity, and emotion dysregulation. Participants then completed a 14-day ecological momentary assessment (EMA) study involving completing brief surveys twice per day. EMA surveys assessed in-play betting behaviors, motives, contexts, and harms., Results: A total of 1,365 EMA surveys were completed, of which 32.89% involved placing at least one in-play bet. A total of 77 (91.67%) participants placed at least one in-play bet during the study period. Participants reported greater in-play betting involvement (the number of bets placed, money spent) and an increased likelihood of experiencing in-play betting-related financial and relationship harms when using substances while betting and when motivated to place in-play bets by an interest in sports or to "be in the game." Participants, who used substances during a greater proportion of in-play betting occasions, who more frequently endorsed coping motives for in-play betting, and who exhibited elevated problem gambling severity, reported greater in-play betting involvement and the likelihood of experiencing in-play betting-related harms overall., Conclusions: Responsible gambling initiatives targeting contextual risk factors, such as using alcohol and cannabis while in-play betting, may help to reduce the intensity of in-play betting and its associated harms. Objective: People who use cannabis for medicinal (vs. nonmedicinal) reasons report greater cannabis use and lower alcohol use, which may reflect a cannabis-alcohol substitution effect in this population. However, it is unclear whether cannabis is used as a substitute or complement to alcohol at the day level among people who use cannabis for both medicinal and nonmedicinal reasons. This study used ecological momentary assessment to examine this question., Method: Participants ( N = 66; 53.1% men; mean age 33 years) completed daily surveys assessing previous-day reasons for cannabis use (medicinal vs. nonmedicinal), cannabis consumption (both number of different types of cannabis used and grams of cannabis flower used), and number of standard drinks consumed., Results: Multilevel models revealed that, in general, greater cannabis consumption on a given day was associated with greater same-day alcohol use. Further, days during which cannabis was used for medicinal (vs. exclusively nonmedicinal) reasons were associated with reduced consumption of both cannabis and alcohol. The day-level association between medicinal reasons for cannabis use and lower alcohol consumption was mediated by using fewer grams of cannabis on medicinal cannabis use days., Conclusions: Day-level cannabis-alcohol associations may be complementary rather than substitutive among people who use cannabis for both medicinal and nonmedicinal reasons, and lower (rather than greater) cannabis consumption on medicinal use days may explain the link between medicinal reasons for cannabis use and reduced alcohol use. 17. Invited review: Total solids concentration in milk or milk replacer for dairy calves.

Restricting milk or milk replacer (MR) fed to dairy calves to 10% of body weight at 12.5% total solids (TS) concentration is a common feeding strategy that promotes early weaning and lowers total feed costs for raising young calves. However, this strategy has been associated with inferior performance and compromised welfare. Because a restricted liquid nutrition plan limits calf growth due to limited supply of nutrients, research on increased supply of nutrients from liquid has shown improved growth, health, and welfare of calves. Nutrient supply may be increased by feeding larger amounts of milk or MR averaging 12.5% TS, or by increasing TS above the usual 12.5% TS by increasing MR TS concentration alone (TS+), or by increasing TS concentration and volume simultaneously (TSV+). The objective of this review is to discuss liquid nutrition plans with greater TS concentration in MR or in milk diets, considering the effects of such strategies on performance and digestive tract development before weaning and the future performance of dairy calves. This review will focus on liquid nutrition plans that are based on increased TS concentration (i.e., TS+ or TSV+); therefore, enhanced liquid nutrition plans based only on increased volumes will not be the discussed. Improved growth rates have been observed in calves fed TSV+ programs. However, reduced starter intake preweaning can also have negative effects on dry matter intake, average daily gain, and digestibility postweaning. Feeding a TS+ program may reduce impairment of starter intake and therefore have positive effects on performance, nutrient digestibility, and health. When considering an accelerated nutrition plan with TS+ or TSV+, it is important to consider osmolality of the liquid diet. Further studies are necessary to confirm this hypothesis and the cost-benefit of alternative liquid nutrition plans, as well as the optimal TS concentration of milk or MR fed to dairy calves., (© 2023, The Authors. 18. Associations of self-regulation strategy use with in-play betting intensity and associated harms: An ecological momentary assessment study.

Self-regulation strategies, such as limit setting, can be effective in reducing harms associated with gambling. However, limited research has examined their applicability to in-play sports betting. The current study therefore examined the utility of self-regulation strategies in reducing in-play betting intensity and associated harms. Adults engaging in-play betting (N = 77; M
age = 41.23 years, 71.43 % men) completed 14 days of ecological momentary assessment surveys in which they reported their in-play betting behaviours (number of bets placed, money spent), use of self-regulation strategies for in-play betting (setting monetary limits, setting time limits, stopping betting when no longer fun, taking frequent breaks), and in-play-betting-related harms. Multilevel models revealed that participants with a greater propensity to set monetary limits were overall less likely to experience financial, interpersonal, and health harms. In addition, participants were less likely to experience emotional/psychological harms following in-play betting occasions during which they set monetary limits (versus when they did not). In contrast, participants with a greater propensity to set time limits tended to spend more money on in-play bets, experienced more harms on average, and were overall more likely to experience financial and work/study harms. Results provide preliminary support for the utility of monetary limit setting, but not time limit setting, in reducing in-play-betting-related harms. 19. Knowledge of Cannabinoid Content Among People Living with HIV Who Use Cannabis: a Daily Diary Study.

Background: Many people living with HIV (PLWH) use cannabis for medicinal reasons. Patients' knowledge of the tetrahydrocannabinol (THC) and cannabidiol (CBD) concentrations of the cannabis products they use may be important in helping patients achieve symptom relief while guarding against potential risks of cannabis use. However, no studies have examined cannabinoid concentration knowledge among PLWH., Method: PLWH (N = 29; 76% men, mean age 47 years) reporting cannabis use for both medicinal and nonmedicinal reasons completed daily surveys over 14 days assessing cannabis products used, knowledge of cannabinoid concentrations of cannabis products used, cannabis use motives (medicinal, nonmedicinal, both), and positive and negative cannabis-related consequences. Across the 361 cannabis use days captured on the daily surveys, at least some knowledge of cannabinoid concentrations was reported on an average of 43.1% (for THC) and 26.6% (for CBD) of the days., Results: Generalized linear mixed models revealed that participants were more likely to report knowing THC and CBD concentrations on days when they used non-flower forms of cannabis relative to days when they used cannabis flower only. Participants who used cannabis for medicinal reasons on a greater proportion of days had greater knowledge of cannabinoid concentration overall across days. Further, greater overall knowledge of cannabinoid concentrations was associated with fewer reported negative cannabis-related consequences., Conclusions: Findings suggest that among PLWH, knowledge of cannabinoid concentrations may be higher when using non-flower cannabis products and among those reporting primarily medicinal cannabis use. 20. Residual Feed Intake as an Efficiency Metric for Pre-Weaning Dairy Calves: What Do We Know?

Dairy cattle systems have targeted improvements in feed efficiency by selecting animals that can convert less feed into more products. Residual feed intake (RFI) has been the index of choice when selecting dairy cattle for feed efficiency. Nonetheless, RFI studies have focused on lactating cows, and the crucial importance of pre-weaning efficiency on farm profitability and cow productivity has been mostly neglected. This review discusses the current knowledge of how RFI divergence relates to nutrient metabolism in pre-weaning dairy calves, including the advantages and limitations of evaluating RFI in this phase. Existing literature indicates that nutrient utilization, energy metabolism, protein metabolism, vitamin metabolism, intestinal development, and hindgut bacterial populations may be implicated in RFI divergence between pre-weaning calves. Techniques developed to date to evaluate RFI in this phase are still evolving to better adapt to the unique characteristics of this phase, and more research is needed to fill in the gaps in our current understanding of early-life feed efficiency divergence in cattle. However, current results suggest great potential for selecting high-efficiency calves while in pre-weaning to accelerate the progress of genetic selection in dairy cattle.

21. Characterizing heterogeneity among people who use cannabis for medicinal reasons: A latent class analysis of a nationally representative Canadian sample.

Background: Many individuals who use cannabis report doing so for medicinal reasons. Few studies have explored heterogeneity within this population, which may be important to inform targeted interventions. This study used latent class analysis to identify subgroups of people who use cannabis for medicinal reasons and their sociodemographic and cannabis-risk-related correlates., Method: Data were drawn from the 2019 Canadian Alcohol and Drugs Survey, which is a representative survey of Canadians ages 15 years and older. Data from 814 individuals reporting past-year use of cannabis for medicinal or mixed medicinal and non-medicinal reasons were included. Latent class analysis was conducted with forms of cannabis used, cannabis use frequency, concurrent non-medicinal cannabis use, and the medical conditions and symptoms cannabis was used to manage as indicators., Results: Four distinct latent classes of medicinal cannabis use were identified: a non-daily cannabis flower for mental health and sleep class (39.56% of the sample), a non-daily cannabis flower for pain class (26.41% of the sample), a non-daily cannabis oil for physical health class (20.15% of the sample), and a daily multi-form cannabis for mental health and non-medical reasons class (13.88% of the sample). Sociodemographic factors and risk level for cannabis-related harms were associated with latent class membership., Conclusions: Results of this study reveal considerable heterogeneity among people reporting medicinal cannabis use and suggest that the distinct patterns of cannabis use behaviors and motives observed may be important for understanding risk for cannabis-related harms in this population. Background and Aims: Sports betting has increased markedly in recent years, in part due to legislative changes and the introduction of novel forms of sports betting (e.g., in-play betting). Some evidence suggests that in-play betting is more harmful than other types of sports betting (i.e., traditional and single-event). However, existing research on in-play sports betting has been limited in scope. To address this gap, the present study examined the extent to which demographic, psychological, and gambling-related constructs (e.g., harms) are endorsed by in-play sports bettors relative to single-event and traditional sports bettors., Methods: Sports bettors (N = 920) aged 18+ from Ontario, Canada completed an online survey containing self-report measures of demographic, psychological, and gambling-related variables. Participants were classified as either in-play (n = 223), single-event (n = 533), or traditional bettors (n = 164) based on their sports betting engagement., Results: In-play sports bettors reported higher problem gambling severity, endorsed greater gambling-related harms across several domains, and reported greater mental health and substance use difficulties compared to single-event and traditional sports bettors. There were generally no differences between single-event and traditional sports bettors., Discussion: Results provide empirical support for the potential harms associated with in-play sports betting and inform our understanding of who may be at risk for increased harms associated with in-play betting., Conclusions: Findings may be important for the development of public health and responsible gambling initiatives to reduce the potential harms of in-play betting, particularly as many jurisdictions globally move towards legalization of sports betting.

23. A Systematic Review of Eye-Tracking Studies of Gambling-Related Attentional Biases.

Previous research has identified attentional biases towards addiction-related stimuli, including gambling-related stimuli. Eye-tracking is considered the gold standard methodology for measuring attentional biases, yet no review to date has examined its use in measuring gambling-related attentional biases. This systematic review synthesized the literature using eye-tracking to examine attentional biases among people who gamble. We reviewed articles from Web of Science and PubMed that were published from 1990 to 2021. A total of 11 articles were included, with sample sizes ranging from 38 to 173 participants. Of these studies, seven examined attentional biases for gambling-related visual stimuli. These seven studies provided support that gambling can result in the development of an attentional bias for gambling-related stimuli. With respect to correlates of gambling-related attentional biases, there were mixed results. Some studies identified significant positive associations between gambling-related attentional biases and psychosocial variables, such as problem gambling severity, gambling expectancies, gambling cravings, gambling motives, depressive symptom severity, alcohol use severity, daily stress, affective impulsivity, and immersion. Four studies examined attentional biases for responsible gambling messaging and advertisements, finding that both people who do and do not gamble attend less to responsible gambling messaging compared to other types of information such as the betting odds. Research using eye-tracking to examine attentional biases among people who gamble is in its infancy. Yet, the preliminary results support the identification of attentional biases using the gold-standard methodology. Attentional biases for gambling-related stimuli are a robust correlate of problem gambling. Free-viewing eye-tracking paradigms are considered the gold standard for measuring attentional bias in addiction research, but their reliability in measuring biases for gambling-related stimuli remains unclear. Using secondary data from two different free-viewing eye-tracking paradigms (two-image and four-image displays), this study examined the internal consistency of fixation indices in samples with varying degrees of gambling involvement and problem gambling risk. Cronbach's alpha, McDonald's omega, and split-half reliability coefficients were used to assess internal consistency of several fixation indices (total dwell time, total dwell time percent, total fixation count, time of first fixation) for gambling-related images, neutral images, and computed attentional bias difference scores. For both two- and four-image displays, internal consistency estimates for total dwell time, total dwell time percent, and total fixation count were good to excellent for gambling-related images, neutral images, and attentional bias scores. Only time of first fixation exhibited low internal consistency. - Abstract
The objective of this study was to evaluate blood cells and metabolites, insulin-like growth factor-1 (IGF-1), digestibility, internal organs weight and histology, gene expression, and spleen cell proliferation of pre-weaned bull calves supplemented with a blend of essential oils in milk replacer (MR). Sixteen newborn Holstein × Gyr crossbred dairy bull calves, with body weight at birth of 33.3 ± 3.7 kg, were housed in individual sand bedded pens, blocked by genetic composition, and randomly assigned to 1 of 2 treatments in a randomized complete block design: Control (CON, n = 8) and blend of essential oils supplementation (BEO, n = 8, 1 g/day/calf, Apex Calf, Adisseo, China). The commercial blend was composed by plant extracts derived from anise, cinnamon, garlic, rosemary, and thyme. Animals were fed 5 L of MR/day reconstituted at 15% (dry matter basis), divided into two equal meals. Water and starter were provided ad libitum. ß-hydroxybutyrate, urea, and glucose were evaluated weekly, IGF-1 was evaluated biweekly, and total blood cell count was performed every four weeks until the end of the trial at eight weeks of age. Feed samples were collected three times a week and polled for weekly analysis. Apparent total nutrient digestibility was determined from d 56 to 60 of age. On d 60 ± 1, animals were euthanized for organ weight, histology, spleen cell proliferation, and intestinal gene expression analysis. Data were analyzed independently using linear mixed models using the REML method in the nlme package in R for continuous outcomes. A non-parametric test was used for ordered categorical outcomes using the Artools package in R. There were no differences between groups for blood evaluations, digestibility, gene expression, and a spleen cell proliferation assay. However, BEO calves presented a heavier pancreas, heavier intestines, bigger ileum villi, and higher cecum butyrate levels (P < 0.05), demonstrating that the EO supplementation helped intestinal development and symbiotic bacteria. It was also observed in CON animals' heavier respiratory tract and a higher eosinophil count (P < 0.05). Therefore, the organs where eosinophils are more active had a better response for BEO animals. No differences were found in the intestinal gene expression in the immune context. These results demonstrate that supplementing essential oils in MR could contribute to gut development and immune function. The aim of this study was to evaluate the effects of dose, start time, and application interval of recombinant bovine somatotropin (rbST) on performance, health, and reproduction parameters of crossbred Holstein × Gyr dairy cows (3/4 and 7/8). A 2 × 2 × 2 factorial study was designed to test the effects of application dose (250 mg rbST or 500 mg rbST), start of application (40 or 63 days in milk (DIM)), and application interval (12 or 14 days); 180 crossbred Holstein × Gyr cows were enrolled. Treatments with 500 mg rbST resulted in increased milk production, 3.5% fat-corrected milk (FCM), and energy-corrected milk (ECM). In the factorial design analysis, greater FCM, ECM, and milk solid production (kg/day) were observed when 500 mg rbST was started at 63 DIM, while 250 mg rbST resulted in greater milk production (kg/day), FCM, ECM, and milk solids (kg/day) when administered at 12-day intervals. Administration of 500 mg rbST resulted in an increase of days open and number of services per pregnancy (P = 0.01). Holstein × Gyr cows (3/4 and 7/8) presented a superior productive response to rbST when treated with 500 mg starting at 63 DIM and at 14-day intervals., (© 2023. Objectives: This study examined children's perspectives about returning to in-person school following lockdown due to the pandemic and about mask-wearing in class, as well as the mental health of children and parents during the pandemic., Methods: This cross-sectional study was part of a 2-day school simulation exercise that randomized students to different masking recommendations. Parent-report of mental health and post-simulation child-report of COVID-19-related anxiety and mask-wearing were analyzed using descriptive and multiple regression analyses. Semi-structured focus groups were conducted with older students to supplement questionnaire data., Results: Of 190 students in this study, 31% were in grade 4 or lower 95% looked forward to returning to in-person school. Greater child anxiety about COVID-19 was predicted by increased parent/caregiver anxiety (β=0.67; P<0.001), and lower parental educational attainment (β=1.86; P<0.002). Older students were more likely than younger students to report that mask-wearing interfered with their abilities to interact with peers (χ
2 (1)=31.16; P<0.001) and understand the teacher (χ2 (1)=13.97; P<0.001). Students in the group that did not require masks were more likely than students in the masking group to report worries about contracting COVID-19 at school (χ2 (1)=10.07; P<0.05), and anticipated difficulty wearing a mask (χ2 (1)=18.95; P<0.001)., Conclusions: For children anxious about COVID-19, parental anxiety and education about COVID-19 may be targets for intervention. Bovine anaplasmosis causes considerable economic losses in dairy cattle production systems worldwide, ranging from $300 million to $900 million annually. It is commonly detected through rectal temperature, blood smear microscopy, and packed cell volume (PCV). Such methodologies are laborious, costly, and difficult to systematically implement in large-scale operations. The objectives of this study were to evaluate (1) rumination and activity data collected by Hr-Tag sensors (SCR Engineers Ltd.) in heifer calves exposed to anaplasmosis; and (2) the predictive ability of recurrent neural networks in early identification of anaplasmosis. Additionally, we aimed to investigate the effect of time series length before disease diagnosis (5, 7, 10, or 12 consecutive days) on the predictive performance of recurrent neural networks, and how early anaplasmosis disease can be detected in dairy calves (5, 3, and 1 d in advance). Twenty-three heifer calves aged 119 ± 15 (mean ± SD) d and weighing 148 ± 20 kg of body weight were challenged with 2 × 10
7 erythrocytes infected with UFMG1 strain (GenBank no. EU676176) isolated from Anaplasma marginale. After inoculation, animals were monitored daily by assessing PCV. The lowest PCV value (14 ± 1.8%) and the finding of rickettsia on blood smears were used as a criterion to classify an animal as sick (d 0). Rumination and activity data were collected continuously and automatically at 2-h intervals, using SCR Heatime Hr-Tag collars. Two time series were built including last sequence of -5, -7, -10, or -12 d preceding d 0 or a sequence of 5, 7, 10, or 12 d randomly selected in a window from -50 to -15 d before d 0 to ensure a sequence of days in which PCV was considered normal (32 ± 2.4%). Long short-term memory was used as a predictive approach, and a leave-one-animal-out cross-validation (LOAOCV) was used to assess prediction quality. Anaplasmosis disease reduced 34 and 11% of rumination and activity, respectively. The accuracy, sensitivity, and specificity of long short-term memory in detecting anaplasmosis ranged from 87 to 98%, 83 to 100%, and 83 to 100%, respectively, using rumination data. For activity data, the accuracy, sensitivity, and specificity varied from 70 to 98%, 61 to 100%, and 74 to 100%, respectively. Predictive performance did not improve when combining rumination and activity. The use of longer time-series did not improve the performance of models to predict anaplasmosis. The accuracy and sensitivity in predicting anaplasmosis up to 3 d before clinical diagnosis (d 0) were greater than 80%, confirming the possibility for early identification of anaplasmosis disease. These findings indicate the great potential of wearable sensors in early identification of anaplasmosis diseases. Evaluating the dermal absorption of sunscreen UV filters requires the development of a bio-predictable in vitro permeation test (IVPT). This work describes the comparison of two IVPT methods and rank order correlations of in vitro absorption (skin permeation and retention) with the in vivo absorption (AUC and skin retention) of sunscreens. The IVPT was compared regarding the following elements: (1) application of a single finite dose vs. an infinite dose and (2) the use of heat-separated human epidermis vs. dermatomed skin models. The IVPT was used to evaluate dermal absorption of six UV filters (avobenzone, homosalate, octinoxate, octisalate, octocrylene, and oxybenzone) in commercial sunscreens. Both the in vivo and in vitro permeation studies demonstrated that all UV filters were absorbed following a single-dose application. Sunscreens were rank ordered by the amount of the UV filters absorbed. Data obtained from the IVPT method using a single finite dose and heat-separated human epidermis was found to correlate with the clinical data. Rank orders of the cumulative in vitro skin permeation and the in vivo AUC were found comparable for oxybenzone, homosalate, octisalate, and octinoxate. Rank orders of the in vitro and in vivo skin retention of oxybenzone and octinoxate were also comparable. Additional IVPT parameters may be optimized to enhance the discriminatory power for UV filters with low skin permeation potential (e.g., avobenzone and octocrylene)., (© 2022. 32. Evaluation of Ingestive Behavior, Ruminal and Blood Parameters, Performance, and Thermography as a Phenotypic Divergence Markers of Residual Feed Intake in Rearing Dairy Heifers.

The objectives of this study were: (1) to identify and rank phenotypically divergent animals for residual feed intake (RFI) regarding their efficiency (high: HE or low: LE); (2) to evaluate their relationships with ingestive behavior, ruminal and blood parameters, performance, and infrared thermography; and (3) to determine if such measurements can be used as feed efficiency markers in rearing dairy heifers. Thirty-eight heifers, 143 d ± 4 (Mean ± SD) of age and 108.7 kg ± 17.9 of body weight were used. The animals were fed with a total mixed ration during the 91 d of the trial. A phenotypic divergence of DMI for RFI was observed between -0.358 and 0.337 kg/d for HE and LE, respectively. Dry matter intake (DMI) was lower in the HE (2.5 kg DMI/d vs. 3.1 kg DMI/d), as was the number of visits to the feed bin with consumption (59 vs. 71). Feed intake was the best predictor of said divergence. Water intake and number of visits to the feed bin were presented moderate correlations with RFI. The ruminal fermentation variables, blood metabolites, blood hormones (such as the other ingestive behavior variables), and infrared thermography were not able to accurately predict HE or LE animals.

33. Effects of Bulk Tank Milk, Waste Milk, and Pasteurized Waste Milk on the Intake, Ruminal Parameters, Blood Parameters, Health, and Performance of Dairy Calves.

- Abstract
The aim of this study was to evaluate the effects of bulk tank milk (BTM), WM, and PWM on the intake, ruminal parameters, blood parameters, health, and performance of dairy calves. Forty-five male crossbred dairy calves (Gyr × Holstein) were used. On their fourth day of age, animals were grouped according to body weight, serum protein levels, and genetic composition. Three treatments were assessed: BTM ( n = 15), WM from cows in antibiotic treatment ( n = 15), and PWM via high-temperature, short-time pasteurization (72-74 °C for 16 s) ( n = 15). During the experimental period (from 4 to 60 d of age), animals were fed 6 L of milk/d, divided into two equal meals. Water and concentrate were provided ad libitum . Daily measurements were made for milk, concentrate, and water intakes, as well as for fecal and respiratory scores. Rumen fluid and blood were sampled weekly. The following parameters were evaluated: volatile fatty acids (VFAs), pH and ammonia-N in rumen fluid, and β-hydroxybutyrate (BHB) and glucose in blood. Animals were weighed at birth, 4 d of age, and weekly up to 60 d of age. At the end of the experimental period (60 ± 1 d), all animals were euthanized for pulmonary evaluation. The randomized complete design with an interaction between treatment and week was the experimental method of choice for testing the hypothesis of the treatment's effect on all evaluated outcomes. Animals in the BTM treatment had higher milk dry matter intake (DMI), followed by WM and PWM calves. Concentrate DMI was lower for BTM in comparison to WM and PWM calves. However, total DMI showed no significant differences between treatments. The rumen fluid from calves receiving PWM had higher concentrations of acetate and propionate than that of BTM and WM animals. No differences were observed between treatments for blood glucose and BHB concentrations. Health parameters (fecal and respiratory scores) and pneumonia occurrence showed no significant difference between treatments. No differences were observed for average daily gain (ADG) or body growth. Feeding WM and PWM did not show significant negative effects on the intake, ruminal parameters, blood parameters, health, or performance of dairy calves.

34. Adoption of Precision Technologies by Brazilian Dairy Farms: The Farmer's Perception.

The use of precision farming technologies, such as milking robots, automated calf feeders, wearable sensors, and others, has significantly increased in dairy operations over the last few years. The growing interest in farming technologies to reduce labor, maximize productivity, and increase profitability is becoming noticeable in several countries, including Brazil. Information regarding technology adoption, perception, and effectiveness in dairy farms could shed light on challenges that need to be addressed by scientific research and extension programs. The objective of this study was to characterize Brazilian dairy farms based on technology usage. Factors such as willingness to invest in precision technologies, adoption of sensor systems, farmer profile, farm characteristics, and production indexes were investigated in 378 dairy farms located in Brazil. A survey with 22 questions was developed and distributed via Google Forms from July 2018 to July 2020. The farms were then classified into seven clusters: (1) top yield farms; (2) medium-high yield, medium-tech; (3) medium yield and top high-tech; (4) medium yield and medium-tech; (5) young medium-low yield and low-tech; (6) elderly medium-low yield and low-tech; and (7) low-tech grazing. The most frequent technologies adopted by producers were milk meters systems (31.7%), milking parlor smart gate (14.5%), sensor systems to detect mastitis (8.4%), cow activity meter (7.1%), and body temperature (7.9%). Based on a scale containing numerical values (1-5), producers indicated "available technical support" (mean; σ
2 ) (4.55; 0.80) as the most important decision criterion involved in adopting technology, followed by "return on investment-ROI" (4.48; 0.80), "user-friendliness" (4.39; 0.88), "upfront investment cost" (4.36; 0.81), and "compatibility with farm management software" (4.2; 1.02). The most important factors precluding investment in precision dairy technologies were the need for investment in other sectors of the farm (36%), the uncertainty of ROI (24%), and lack of integration with other farm systems and software (11%). Farmers indicated that the most useful technologies were automatic milk meters systems (mean; σ2 ) (4.05; 1.66), sensor systems for mastitis detection (4.00; 1.57), automatic feeding systems (3.50; 2.05), cow activity meter (3.45; 1.95), and in-line milk analyzers (3.45; 1.95). Overall, the concerns related to data integration, ROI, and user-friendliness of technologies are similar to those of dairy farms located in other countries. Increasing available technical support for sensing technology can have a positive impact on technology adoption.- Published

35. How Divergence for Feed Efficiency Traits Affects Body Measurements and Metabolites in Blood and Ruminal Parameters on Pre-Weaning Dairy Heifers.

The objectives of this study were: (1) to evaluate feed efficiency indexes and their relationships with body measurements and blood and ruminal metabolites in the pre-weaning period; (2) to determine if such measurements can be used as feed-efficiency markers during the pre-weaning period. Holstein-Gyr heifer calves ( n = 36), enrolled between 4 and 12 weeks of age, were classified into two residual feed intake (RFI) and residual body weight gain (RG) groups: high efficiency (HE; RFI, n = 10; and RG, n = 9), and low efficiency (LE; RFI, n = 10; and RG, n = 8). Calves were fed whole milk (6 L/day) and solid feed ad libitum. Body developments were measured weekly and feed intake (milk and solid feed) daily during the whole period. Blood samples were collected at 12 weeks of age and analyzed for glucose, insulin and β-hydroxybutyrate (BHB). Samples of ruminal content were collected on the same day and analyzed for pH, NH
3 -N, and volatile fatty acids (VFA). Among the growth characteristics, only the initial hip width differed between the RFI groups, and withers height differed between the RG groups. Concentration of BHB was greater and glucose: insulin ratios tended to be greater in LE-RG animals. Butyric acid proportions were similar among RFI groups, but tended to be greater for HE-RG than for LE-RG. Overall, correlation coefficients between RFI or RG and blood, rumen, or morphometric markers were low. Thus, it is unlikely that measurements of metabolic indicators, per se, will be useful in the early identification of more efficient animals. Understanding the underlying physiological basis for improved feed efficiency in dairy heifers requires further investigation.- Published

36. Screening microchip sites to predict body temperature in young calves.

Thermal microchip sensors can automate body temperature measurements. The best site of implantation is still unknown, and the accuracy and precision of body temperature predictions based on microchip data need to be investigated. The aim of this study was to investigate the best site for microchip implant for monitoring body temperature in dairy calves. Seventeen calves were used (32.2 ± 5.2 kg of body weight) and the microchips were implanted four days after birth. The microchips were implanted at navel, ear and tail base (subcutaneous), neck (cleidocephalicus) and internal face of leg (gracilis) (intramuscular). Rectal temperature (RT, °C), obtained with a clinical thermometer, was considered as core temperature. Air temperature (AT), relative humidity (RH) and the temperature and humidity index (THI) were evaluated at the same time of rectal and microchip temperature measurements over 56 days. The range of AT, RH and THI was 7.6-34.4 °C, 17.5-99.0% and 50.6 to 91.5. The average for rectum, ear, neck, tail, leg, and navel were 38.7; 36.9; 38.0; 37.0, 37.8 and 37.0 °C. The intramuscular implantations had closest values to RT. The correlations between RT and ear, neck, tail, leg, and navel temperatures were 0.56, 0.60, 0.60, 0.53 e 0.48. The RT prediction based on microchip data had precision (r
c ) ranged between 0.49 and 0.60 and accuracy (Cb ) between 0.79 and 0.88. The inclusion of AT, RH and THI as predictive variables in models decrease the mean absolute error (23%) and increase the precision (21.3%) and accuracy (10.2%). The Concordance Correlation Coefficient and root-mean-square error for equations using tail or neck microchips were 0.68 and 0.67, and 0.29 and 0.28 °C, respectively. The tail base is a promising site for microchip implantation to predict rectal temperature. The aim of this study was to identify possible effects of different vaccination strategies (concomitantly or not) against brucellosis and clostridia on intake, performance, feeding behavior, blood parameters, and immune responses of dairy heifers calves. Fifty heifers calves were enrolled [38 Gyr (Zebu, Bos taurus indicus) and 12 5/8 Holstein × Gyr]. At 120 d of age, animals were randomly distributed among 3 groups: B (n = 18), vaccinated against brucellosis; C (n = 14), vaccinated against clostridia and CB (n = 18), vaccinated concomitantly for both. Rectal and thermographic temperatures were evaluated on days -1, 0, 1, 2, 3, 5, 7,10, 14, and 28 relatives to the vaccination day. Feed and water intake, body weight (BW), and feeding behavior were monitored daily by an electronic feeding system. Blood was sampled on days 0, 3, 7, 14, and 28, relative to the vaccination day for determination of glucose and β -hydroxybutyrate (BHBA) concentrations. Blood sampled on day 0 (prevaccination) and on days 28 and 42 were used to evaluate the immune response against Brucella abortus and clostridia. There was an increase in rectal temperature between the first and the third day postvaccination in the 3 groups. The thermography revealed an increase of local temperature for 7 d on groups B and CB. Group C had increased local temperature for a longer period, lasting for up to 14 d. Dry mater intake was reduced for groups B and CB, but no alteration was observed for group C. No alterations regarding initial BW, final BW, average daily weight gain, and feed efficiency were observed. No differences were observed for the 3 vaccination groups for blood parameters throughout the evaluation period. The concomitant vaccination against brucellosis and clostridia led to lower neutralizing antibody titers against epsilon toxin of Clostridium perfringens and botulinum toxin type C of C. botulinum (C > CB > B). When cellular proliferation assay and serological tests to B. abortus were evaluated, no differences were observed between groups B and CB. The present results indicate that the concomitant vaccination against brucellosis and clostridia has no relevant impact on the intake, performance, and feeding behavior of dairy calves. - Abstract
Sunscreen products contain UV filters as active ingredients for the protection of the skin against UVR. The US Food and Drug Administration (FDA) issued a new proposed rule in 2019 (84.FR.6204) for sunscreens and identified the need for additional safety data for certain UV filters including their dermal absorption data. Dermal absorption data reveal systemic exposure of UV filters in humans, which can be obtained from clinical maximal usage trials. FDA guidance recommends conducting in vitro skin permeation tests (IVPTs) to help select formulations for maximal usage clinical trials as IVPT results may be indicative of in vivo absorption. This case study reports in vitro methodologies used for the selection of sunscreen products for an FDA-sponsored proof-of-concept maximal usage clinical trial. An IVPT method was developed using human cadaver skin. Commercially available sunscreen products were tested to determine the skin absorption potential of common UV filters using the IVPT. All the studied sunscreen products demonstrated a certain degree of skin absorption of UV filters using IVPT, and a formulation rank order was obtained. These sunscreen products were also characterized for several formulation properties including the globule size in emulsions, which was found to be an indicator for the rank order., (Copyright © 2020 The Authors. The objectives of this study were: i) to classify animals into groups of high and low feed efficiency (FE) using three FE indexes (Residual feed intake (RFI), Residual weight gain (RG) and Feed conversion efficiency (FCE)), and ii) to evaluate whether crossbreed Holstein x Gyr heifers divergent for FE indexes exhibit differences in nutrient intake and digestibility, energy partitioning, heat production, methane emissions, nitrogen partitioning and blood parameters. Thirty-five heifers were housed in a tie-stall, received ad libitum TMR (75:25, corn silage: concentrate) and were ranked and classified into high (HE) or low efficiency (LE) for RFI, RG and FCE. The number of animals for each HE group were 13 (< 0.5 standard deviation (SD) for RFI, 11 for RG and 11 for FCE (> 0.5 SD) and for the LE were 10 (> 0.5 SD) for RFI, 11 for RG and 12 for FCE (< 0.5 SD). Gas exchanges (O2 consumption, CO2 and CH4 production) in open-circuit respiratory chambers and whole tract digestibility trial was performed. A completely randomized experimental design was used and the data were analyzed by ANOVA and correlation study. High efficiency animals for RFI produced less CO2, consumed less O2 and had lower heat production (HP). Methane production was positively correlated with RFI. High efficiency RG had higher O2 consumption and CO2 production in relation to LE-RG. High efficiency FCE had greater NFC digestibility, higher positive energy balance (EB) and excreted (11.4 g/d) less nitrogen in urine. High efficiency RG and FCE groups emitted less CH4 per kg of weight gain than LE animals. Animals HE for RFI and FCE had lower β-hydroxybutyrate and higher glucose concentrations, respectively. The differences in intake, digestibility, energy and nitrogen partition, CH4 emission, blood metabolic variables and heat production between the HE and LE groups varied according to the efficiency indexes adopted. The objective of this study was to evaluate whether pre-weaning heifer calves divergent for residual feed intake (RFI) or residual feed intake and body weight gain (RIG) exhibit differences in thermography, blood, and ruminal parameters. Thirty-two Gyr heifer calves were enrolled in a 63-d trial and classified into 2 feed efficiency (FE) groups based on RFI and RIG (mean ± 0.5 SD). The groups were classified as high efficiency (HE) RFI (HE RFI, n = 9), HE RIG (HE RIG, n = 10), low efficiency (LE) RFI (LE RFI, n = 10), and LE RIG (LE RIG, n = 11). The amount of whole milk provided for each calf was calculated based on their metabolic weight at birth (42% x BW0.75). The liquid diet was divided into two meals at 0700 and 1400 h. The total solid diet (TSD) was composed of 92% concentrate and 8% of Tifton 85 hay chopped in 5-cm lengths, as fed. Intake was measured daily. Blood concentrations of insulin, beta hydroxybutyrate, urea, and glucose, and ruminal pH, N-NH3, and volatile fatty acids (VFA) were evaluated at 14, 28, 42, 56, and 70 days of age. Thermal images of the calves were taken with an infrared camera (FLIR T420, FLIR Systems Inc., Wilsonville, OR) on d 56 (±3) at 0600 h, before the morning feeding. Total VFA concentration and propionate as % of total VFA were 24.2% and 22.2% lower in HE RFI compared to LE RFI calves, respectively. On the other hand, acetate as % of total VFA was 10.6% greater in HE RFI than LE RFI calves. Blood urea concentration tended to be greater in LE RFI than HE RFI calves. High efficiency HE RIG tended to have 6.8% greater acetate and 15.4% lower propionate as % of total VFA than LE RIG. Blood insulin concentration was greater and blood glucose tended to be greater for LE RIG than HE RIG group. Low efficiency RIG group had greater left rib, left flank, and anus surface temperature measured by infrared thermography than the HE RIG group. Differences in ruminal fermentation do not seem to be associated with pre-weaning calves efficiency, while differences in protein metabolism seem to affect RFI during this phase. - Abstract
The authors report a rare clinical case of a patient with neurofibromatosis type 1 (NF1) complicated by pulmonary hypertension (PH), which presents with rapid progression. An exhaustive investigation was performed to identify the main aetiology of the PH. It was concluded that the PH could be associated with NF1, and so belonged in group 5 of the clinical classification of PH. In general, such patients have a poor long-term prognosis due to the inexistence of proven, effective treatment. The objective of this study was to evaluate the effect of the physical form of starter and inclusion of hay in the diet of preweaning dairy calves on performance, digestibility, ruminal development, and mRNA expression of genes involved in ruminal metabolism. Holstein × Gyr crossbred male calves (n = 38 1day old) were assigned to 3 treatments for 9 weeks: Control (n = 13; pellet starter with 4 mm diameter and 18 mm length and 4% steam-flaked corn), Ground (n = 12; same starter of the control but ground pass through a 4.0 mm sieve), or Ground plus 5% chopped Tifton hay GH (n = 13). All calves were fed 4 L/d of whole milk up to 63 d of age and were abruptly weaned at 64 d of age. Water and diets were offered ad libitum. Samples of ruminal contents were obtained from all animals at 30, 45, and 60 d of age to evaluate pH, ammonia nitrogen, and volatile fatty acids (VFA). At 55 d of age, an apparent digestibility assay was performed using 18 animals (n = 6/ treatment). At 65 d of age, the 18 animals were euthanized to evaluate the development of the digestive tract. The physical form of starter and the dietary inclusion of hay did not influence starter intake (Control 326 g/d, Ground 314 g/d and GH 365 g/d), daily weight gain (Control 541g/d, Ground 531g/d and GH 606g/d), feed efficiency, apparent nutrient digestibility, energy partitioning, nitrogen balance, ruminal pH, ammonia nitrogen concentration, VFA, the development of the digestive tract and the mRNA expression of genes involved in AGV metabolism., Competing Interests: Embrapa Dairy Cattle (Minas Gerais, Brazil) provided the premises and animals for use in the survey. - Abstract
Marfan syndrome is an autosomal dominant connective tissue disease with an estimated incidence of 1 in 5000 individuals. In 90% of cases it is caused by mutations in the gene for fibrillin-1, the main constituent of extracellular microfibrils. Studies on animal models of Marfan syndrome have revealed that fibrillin-1 mutations interfere with local TGF-β signaling, in addition to impairing tissue integrity. The cardinal features involve the cardiovascular, ocular and skeletal systems. The diagnosis of Marfan syndrome is made according to the revised Ghent nosology. Early identification and appropriate management are critical for patients with Marfan syndrome, who are prone to the life-threatening cardiovascular complications of aortic aneurysms and aortic dissection. The standard treatment includes prophylactic beta-blockers in order to slow down dilation of the ascending aorta, and prophylactic aortic surgery. The success of current medical and surgical treatment of aortic disease in Marfan syndrome has substantially improved mean life expectancy, extending it above 72 years. - Abstract
Human ingestion of antimicrobial residues in dairy products is a problem of public health. In this study we evaluated antibacterial effectiveness of aqueous and ethanolic leaf extracts of Mangifera indica L. as a natural alternative for reduction of Staphylococcus spp. biofilms. Eight Staphylococcus spp. strains from cows with mastitis and the Staphylococcus aureus strain (ATCC 25923) were evaluated. High performance liquid chromatography indicated the presence of phenolic compounds in the aqueous extract. Agilent Technologies gas chromatography revealed the presence of phenolic compounds, carbohydrates, and gallic acid in ethanolic extract. The tannin contents were 2.27 ± 0.01 and 2.67 ± 0.01% for ethanolic extract and aqueous extract, respectively. Extracts were evaluated in agar diffusion test and the size of inhibition zones were compared. The ethanolic extract was the most effective, showing minimum inhibitory concentrations and minimum bactericidal concentrations between 1.8-7.5 and 15.1-45.3 mg/ml, respectively. After tannin removal, the extracts were less effective, indicating these secondary metabolites as one of the main antibacterial compounds. The ethanolic extract at 45.3 mg/ml reduced mature biofilms in teat rubber (average: 3.91 log reduction) and stainless steel (average: 3.87 log reduction) after 5 min of contact. The extracts of M. indica leaves represent natural alternatives against Staphylococcus spp. strains and the ethanolic extract shows potential as a natural sanitizer.

- Abstract
Background: Broad spectrum sunscreens with a sun protection factor (SPF) of 15 or greater are indicated to decrease the risk of skin cancer and early skin aging caused by the sun if used as directed with other sun protection measures. To determine whether sunscreen product performance is compromised under solar exposure and to test spectral uniformity of protection across the UVA spectrum, we tested broad spectrum sunscreens with a variety of active pharmaceutical ingredients (APIs) and in a variety of dosage forms., Methods: A cross-sectional market survey of 32 sunscreen drug products containing either organic or inorganic APIs with SPFs of 15, 30, 50, and 70 was tested. UV doses were delivered via natural sun in Silver Spring, Maryland between June and September of 2017., Results: Of the 32 sunscreen drug products, 6 products failed to meet their broad spectrum claim under solar exposure. Using FDA's new proposal to strengthen sunscreen broad spectrum requirements, spectral uniformity based on the mean sunscreen absorbance of UVA1(340-400 nm)/UV (290-400 nm) indicated that ~40% of sunscreen drug products tested had suboptimal UVA protection., Conclusion: US consumers may unknowingly be receiving up to 36% more transmitted UVA when selecting between similarly labeled broad spectrum sunscreen drug products with equivalent SPF values. FDA's new proposal may help decrease consumers' overall lifetime UVA burden. Spectral absorbance data on sunscreen performance can be used to further improve the coupling of broad spectrum protection to a product's SPF value so that consumers have improved proportional increases in UV protection., (Published 2019. The objective was to evaluate reproductive characteristics of crossbred Girolando (Gyr x Holstein) bulls from two breed compositions and correlate these results with infrared thermography data. Evaluations were performed considering sperm motility, vigor and morphology; scrotal circumference; body morphology and temperament. Infrared thermography was performed to determine surface temperatures of ocular and scrotal areas. Thermoregulation capacity was assessed by differences between air and rectal temperatures, air and maximum temperatures in ocular and scrotal areas, and dorsal and ventral lines of the scrotum. Data analysis was performed using a linear mixed model (breed composition as fixed effect and year of evaluation as random effect). Spearman correlation coefficient was used to associate thermography and reproductive data. Girolando 3/4 Holstein bulls had higher (P ≤ 0.001) scrotal circumference and higher average body morphology and temperament, whereas 5/8 Holstein bulls had a higher (P < 0.001) percentage of major-type and total sperm defects. Girolando 3/4 Holstein bulls had scrotal temperatures 0.8 °C higher (P < 0.001) and 5/8 Holstein bulls had 9.8% and 10.6% higher differences on "rectal - scrotal area" and "ocular area - scrotal area" temperatures, respectively. Ocular area temperatures had negative correlations (P ≤ 0.1) with sperm motility and vigor, and positive correlations (P < 0.001) with minor-type and total sperm defects. Ventral line scrotal temperatures had positive correlations (P < 0.001) with minor-type and total sperm defects. Girolando 3/4 Holstein bulls were found to be superior to 5/8 Holstein bulls for reproductive characteristics. Under non-stressing climatic conditions, semen characteristics of Girolando bulls were more influenced by breed composition than by the capacity for scrotal thermoregulation. Correlations between semen quality and scrotal temperatures can aid in the identification of bulls for breeding, particularly when a large number of animals are in the tests., (Copyright © 2019 Elsevier Ltd. Tick-borne disease is a hemolytic disease complex that causes extensive losses to the dairy industry in tropical regions. Its timely diagnosis is challenging and labor intensive. Our objective was to investigate whether tick-borne disease was associated with changes in ingestive behavior of weaned dairy calves. Weaned calves (n = 35) enrolled at 108 ± 8 d of age were group housed (n = 12, 12, and 11/group) for 4 wk in the experimental pen, which had 12 electronic feed bins and 2 electronic water bins. Data were automatically collected at each feed or water bin visit via radio frequency identification ear tags. Water and total mixed ration were provided ad libitum. Calves were examined weekly for tick-borne disease detection. A disease bout was confirmed if packed cell volume was ≤25% at any exam. Diseased calves received antimicrobial and antipyretic treatment. Data were summarized by day within bin type (feed or water) as intake (kg/d; as-fed basis), frequency of visits (visits/d), and total duration of visits (min/d). Day of detection was set as d 0; a 9-d behavioral screening period for evaluation of behavioral changes was set according to d 0 (d -4 to +4). A within-calf mean was calculated for the healthy period (HP; mean of all days between 2 consecutive negative exams) for each response variable. Data were analyzed as within-calf differences between HP and each day of the behavioral screening period. Tick-borne disease was detected in 12 calves; thus, only data referent to these animals were included in the analyses. Compared with HP, daily feed intake was reduced on d -1, 0, and +1, and daily frequency and total duration of feed bin visits were reduced from d -3 to d +4. Daily feed intake was reduced by 35% on d -1 (3.5 ± 0.4 vs. 5.4 ± 0.5 kg/d for HP), and daily frequency and duration of visits were reduced by 27% (56.3 ± 7.8 vs. 76.7 ± 8.1 visits/d for HP) and 24% (27.0 ± 3.8 vs. 35.4 ± 3.9 min/d for HP) on d -3, respectively. Daily water intake on d 0 (9.1 ± 1.4 kg/d) was lower than at HP (12.9 ± 1.6 kg/d), but other drinking behaviors were not different from healthy means during the screening period. Feeding behavior, but not drinking behavior, was different from HP means before detection at weekly exams. The aims of this study were (1) to assess if there is phenotypical divergence for feed efficiency (FE) during the preweaning phase; (2) if FE is correlated with heat production (HP) measured by the face mask method or (3) by surface skin temperature via thermography, and (4) whether these methods are applicable to preweaned calves. Holstein × Gyr heifer calves (n = 36, birth body weight = 32.4 ± 6.6 kg) were enrolled and on trial between 4 and 12 wk of age and were classified into 2 residual feed intake (RFI) and residual body weight gain (RG) groups: high efficiency (HE; RFI, n = 10; and RG, n = 9) and low efficiency (LE; RFI, n = 10; and RG, n = 8). Calves were fed milk (6 L/d) and solid feed (95% starter and 5% chopped Tifton 85 hay, as fed). Growth was monitored weekly and feed intake (milk and solid feed) daily, during the whole period. Gas exchanges (O
2 consumption and production of CO2 and CH4 ) were obtained using a face mask at 45 ± 5 d of age and HP was estimated. Maximum temperatures were measured at 7 sites with an infrared camera at 62 ± 7 d of age. There was divergence in RFI and RG. Respectively, HE and LE calves had RFI of -0.14 and 0.13 kg/d, and RG of 0.05 and -0.07 kg/d. Dry matter intake was 15% lower in HE-RFI compared with LE-RFI, but no differences were observed in average daily weight gain. Within the RG test, no differences were observed in dry matter intake or average daily gain. The HE-RFI calves consumed less O2 (L/d) and produced less CO2 (L/d). Heart rate and HP were lower for HE-RFI calves compared with LE-RFI. Residual feed intake was correlated with HP (r = 0.48), O2 consumption (r = 0.48), CO2 production (r = 0.48), and heart rate (r = 0.40). No differences were observed in HP and gas exchanges between RG groups. Methane production was null in both groups. Eye temperature measured by thermography was 0.5°C greater in HE-RG than LE-RG calves. Differences in skin temperature between HE and LE calves were not observed at the other sites. These results support the hypothesis that calves are divergent for RFI, RG, and FE during preweaning and divergence tests are applicable during this phase. The face mask method described here is a useful tool for estimating differences in HP among phenotypically divergent RFI calves. 50. Validation of a system for monitoring individual feeding and drinking behaviour and intake in young cattle.

- Abstract
The objective of this study was to validate an electronic system for monitoring individual feeding and drinking behaviour and intake developed for young cattle housed in group. A total of 35 Holstein-Gyr crossbred heifers (BW: 180±52 kg; age: 121.5±32.5 days), fitted with an ear tag containing a unique passive transponder, were distributed in three groups of 12, 12 and 11 animals per period and had free access to 12 electronic feed bins and two electronic water bins (Intergado® Ltd). The dimensions of feed and water bins, as well as the sensors position were appropriate for young cattle. The system documented the visit frequency and duration, as well as the feed and water intakes, by recording the animal's identification tag, bin number, initial and final times of visits and the difference of feed/water weight at the start and end of each bin visit. Feed bins were monitored using time-lapse video recording over 4 days and the water bins were monitored over 6 days. For each feed bin, two feeding events were monitored using manual weighings with an external scale immediately before and after the animal's visit and the difference between them was assumed as feed intake (n=24 observations). For the water bins, 60 manual weighings were made. Video and manual weighing data were regressed on the electronic feeding and drinking behaviour and intake data to evaluate the system's precision and accuracy. The system showed high specificity (98.98% and 98.56% for the feed and water bins, respectively) and sensitivity (99.25% and 98.74%, respectively) for identifying an animal's presence or absence. Duration of feed and water bin visits as well as feed and water consumption per visit estimated by the system were highly correlated and precise compared with the observed video and manual weighing data (r 2=0.917, 0.963, 0.973 and 0.986, respectively). It was concluded that Intergado® system is a useful tool for monitoring feeding and drinking behaviour as well as water and feed intakes in young cattle housed in groups.
