Objective: Syzygium aromaticum and Coffea canephora are acknowledged for their outstanding antioxidant, anti-inflammatory, and nerve-stimulant properties, showcasing potential in brain protection. Therefore, this study aims to quantitatively review existing literature and assess the potential of using it to formulate a herbal tea blend for managing stress and anxiety. Methods: Data was retrieved from the Scopus database, and a bibliometric analysis was performed using VOSviewer software. Results: Following a screening process, a total of 121 articles were identified, with S. aromaticum yielding a higher number compared to C. canephora. A detailed exploration of each plant revealed active components such as eugenol, β-caryophyllene, α-humulene, caffeine, mangiferin, and chlorogenic acids, each exhibiting stimulatory effects alongside antioxidant and anti-inflammatory properties. The neuroprotective effects were attributed to the reduction of oxidative stress and inflammation, coupled with the stimulation of neurotransmitters and hormones like dopamine, serotonin, cortisol, and adrenaline. Conclusions: The review showed that these plants positively affect mood and cognition by influencing the brain's pleasure system. This suggests the need for further research to combine these plant extracts for developing 'Tenang tea', a potential herbal blend for managing stress and anxiety. [ABSTRACT FROM AUTHOR]

This study evaluated consumers' perceptions of beverages obtained from the intervarietal hybrids of Coffea canephora, Conilon and Robusta, produced in the Western Amazon, through a home-use test with 127 participants. An acceptance test and a Check-All-That-Apply procedure were applied. Two clones, BRS 2314 and BRS 2357, were studied (both in natural and fermented versions) and their roasted coffee composition was also evaluated. All beverages were described as having a mild aroma, roasted flavor, and slightly sour taste; consumers noticed both the effect of the fermentation process and genetics. Natural coffees had greater sensory acceptance than fermented ones. Natural coffees were most associated with a slightly bitter taste, and were well accepted by almost half of the participants, mainly women. Fermented coffees were both associated with fermented flavor, but also presented specific characteristics. BRS 2314F was most associated with a fruity flavor and slightly bitter taste and was well accepted by younger consumers with higher levels of education. BRS 2357F was most associated with tobacco flavor and bitter taste and was well accepted by older consumers with a lower education level and a higher coffee consumption frequency. This research aimed to select conilon coffee tree clones (Coffea canephora Pierre ex A. Froehner) for agroforestry and/or intercropping systems. The experiment was carried out at Bananal do Norte Experimental Farm (20º45' S and 41º17' W), Espírito Santo, Brazil. The clones were evaluated in an agroforestry system with "urograndis" hybrid eucalyptus trees (Eucalyptus grandis W.Hill × E. urophylla S.T.Blake) plus prata-type banana herbs (Musa spp.) and an intercropping with 'Folha Murcha' sweet orange trees (Citrus sinensis (L.) Osbeck). An augmented block experiment design, with two plants per plot, four controls, in six blocks in the agroforestry system and four in the intercropping, was adopted. Thirteen morphophysiological characteristics were evaluated. Data analysis was performed using the restricted maximum likelihood method and best linear unbiased prediction and the significance of genetic effects by the likelihood ratio test. To select superior clones the Mulamba-Rank index was performed. Twenty clones were selected for the agroforestry and 20 for the intercropped system. The clones 16, 17, 35, 43, 48, 61, 64, 68 and T3 were the most promising for both systems. Background and Aims Plant breeders are increasingly turning to crop wild relatives (CWRs) to ensure food security in a rapidly changing environment. However, CWR populations are confronted with various human-induced threats, including hybridization with their nearby cultivated crops. This might be a particular problem for wild coffee species, which often occur near coffee cultivation areas. Here, we briefly review the evidence for wild Coffea arabica (cultivated as Arabica coffee) and Coffea canephora (cultivated as Robusta coffee) and then focused on C. canephora in the Yangambi region in the Democratic Republic of the Congo. There, we examined the geographical distribution of cultivated C. canephora and the incidence of hybridization between cultivated and wild individuals within the rainforest. Methods We collected 71 C. canephora individuals from home gardens and 12 C. canephora individuals from the tropical rainforest in the Yangambi region and genotyped them using genotyping-by-sequencing (GBS). We compared the fingerprints with existing GBS data from 388 C. canephora individuals from natural tropical rainforests and the INERA Coffee Collection, a Robusta coffee field gene bank and the most probable source of cultivated genotypes in the area. We then established robust diagnostic fingerprints that genetically differentiate cultivated from wild coffee, identified cultivated–wild hybrids and mapped their geographical position in the rainforest. Key Results We identified cultivated genotypes and cultivated–wild hybrids in zones with clear anthropogenic activity, and where cultivated C. canephora in home gardens may serve as a source for crop-to-wild gene flow. We found relatively few hybrids and backcrosses in the rainforests. Conclusions The cultivation of C. canephora in close proximity to its wild gene pool has led to cultivated genotypes and cultivated–wild hybrids appearing within the natural habitats of C. canephora. Yet, given the high genetic similarity between the cultivated and wild gene pool, together with the relatively low incidence of hybridization, our results indicate that the overall impact in terms of risk of introgression remains limited so far. Abstract Agroforestry is a management strategy for mitigating the negative impacts of climate and adapting to sustainable farming systems. The successful implementation of agroforestry strategies requires that climate risks are appropriately assessed. The spatial scale, a critical determinant influencing climate impact assessments and, subsequently, agroforestry strategies, has been an overlooked dimension in the literature. In this study, climate risk impacts on robusta coffee production were investigated at different spatial scales in coffee-based agroforestry systems across India. Data from 314 coffee farms distributed across the districts of Chikmagalur and Coorg (Karnataka state) and Wayanad (Kerala state) were collected during the 2015/2016 to 2017/2018 coffee seasons and were used to quantify the key climate drivers of coffee yield. Projected climate data for two scenarios of change in global climate corresponding to (1) current baseline conditions (1985–2015) and (2) global mean temperatures 2 °C above preindustrial levels were then used to assess impacts on robusta coffee yield. Results indicated that at the district scale rainfall variability predominantly constrained coffee productivity, while at a broader regional scale, maximum temperature was the most important factor. Under a 2 °C global warming scenario relative to the baseline (1985–2015) climatic conditions, the changes in coffee yield exhibited spatial-scale dependent disparities. Whilst modest increases in yield (up to 5%) were projected from district-scale models, at the regional scale, reductions in coffee yield by 10–20% on average were found. These divergent impacts of climate risks underscore the imperative for coffee-based agroforestry systems to develop strategies that operate effectively at various scales to ensure better resilience to the changing climate.

Introduction: The genetic diversity of Robusta coffee (Coffea canephora), a cornerstone in the global coffee industry, remains not fully explored, leading to a significant gap in our understanding of its sensory intricacies. Our study evaluated the sensory quality potential of the Robusta cultivars from the INERA Coffee Collection in Yangambi (the Democratic Republic of the Congo), the local wild diversity, and their hybrids. Methods: We evaluated the sensory attributes of 70 genotypes representing the genetic structure of the coffee collection. Of those 70, 22 genotypes were evaluated for two consecutive years to assess the consistency of the sensory quality. Standard coffee cupping with the Fine Robusta Standards and Protocols was enhanced through sensory descriptors from the Coffee Taster's Flavor Wheel. Each genotype's sensory profile was constructed based on the Total cupping score and the frequency of reported sensory descriptors. The Total cupping score ranged from 75.75 to 84.75, with a substantial variation in sensory profiles, even within a genetic cluster. Results and discussion: Nutty/Cocoa was the most frequently reported descriptor class. The sensory profile ideotype exhibits a high frequency of Fruity, Sweet, and Sour/Fermented descriptors and a low frequency of Green/Vegetative, Other, and Roasted descriptors. Evidence suggests that the sensory profile of a genotype is consistent over two harvest years. Genotypes with promising and unique sensory profiles were discovered within the cultivars and the wild - cultivar hybrids. Background: The utilization of coffee cascara, a byproduct of coffee cultivation, in cancer therapy research. This research begins with the rationale of exploring medicinal plants, especially coffee, to obtain compounds that can target cancer cells with fewer side effects. Objectivity: This research aims to extract and evaluate the secondary metabolites from robusta coffee cascara, such as friedelin, lupeol, stigmasterol, ursolic acid, caffeine, chlorogenic acid, caffeic acid, and catechin, for their cytotoxic activity against Hela and MCF-7 cells. The aim of this research is also to identify and understand the cytotoxic mechanisms of compounds like stigmasterol, which showed significant cytotoxicity against cancer cells, paving the way for developing targeted cancer therapies from natural sources. Methods: Robusta coffee cascara then goes to the process of extraction using ethanol, fractionation, isolation, purification, and characterization, followed by bioactivity evaluation using in vitro method through breast cancer cell line MCF-7 and cervical cancer cell line HeLa and determination of active compound levels. Results: The cascara, a byproduct of coffee cultivation, is rich in proteins, polysaccharides, and bioactive compounds. Through extraction and purification processes, eight compounds were isolated and characterized, including (1) friedelin, (2) lupeol, (3) Stigmasterol, (4) Ursolic acid, (5) caffeine, (6) Chlorogenic acid, (7) caffeic acid, and (8) catechin. Bioactivity evaluation shows that stigmasterol (3) is the most cytotoxic compound with a value against Hela cells with an IC50 value of 25.85 µg/mL in the toxic category and against MCF-7 cells with an IC50 value of 12.83 µg/mL in the very toxic category. The results of determining the levels of active compounds in robusta coffee cascara extract showed that friedelin (1) 0.539±0.137%; lupeol (2) levels were 0.087±0.015%; (3) stigmasterol 0.126±0.046%; ursolic acid (4) 0.627±0.002%; caffeine (5) 3,203±0.069%; chlorogenic acid (6) 0.679±0.003%; caffeic acid (7) 0.153±0.003% and catechin (8) 0.359 0.012% mg/g extract. Consumers are increasingly looking for healthy foods without the addition of synthetic additives. The aim of this study was to evaluate the efficiency of coffee extracts as a natural antioxidant in fresh pork sausage. Firstly, the conditions for obtaining coffee green extracts were optimized (Central Composite Rotatable Design 23, variables: extraction time, ethanol–water ratio, and sample–solvent ratio) in an ultrasound bath (70 °C). The response variables were the bioactive compounds levels and antioxidant activity. Valid models were obtained (p ≤ 0.05, R2 > 0.751), with higher bioactive content and antioxidant activity in the central point region. Extracts of Robusta and Arabica coffee green (RG and AG) and medium roast (RR and AR) obtained, and central point (10 min, an ethanol concentration of 30%, and a sample–solvent ratio of 10 g/100 mL) and optimized (14.2 min, 34.2%, and 5.8 g/100 mL) parameters were characterized. The RG presented a significantly (p ≤ 0.05) higher content of caffeine (3114.8 ± 50.0 and 3148.1 ± 13.5 mg/100 g) and 5-CQA (6417.1 ± 22.0 and 6706.4 ± 23.5 mg/100 g) in both extraction conditions, respectively. The RG and RR coffee presented the highest antioxidant activity. Two concentrations of RG and RR coffee extracts were tested in fresh pork sausage. The Robusta coffee extract presented the highest antioxidant activity in both roasted and green states. However, when applied to a meat product, the extract prepared with RG coffee showed better results, with efficiency in replacing synthetic antioxidants (content of malonaldehyde/kg of sample below 0.696 ± 0.059 in 20 days of storage), without altering the sensory attributes of the product (average scores above 7.16 ± 1.43 for all attributes evaluated). Recent investigations into the microbiota and fermentation of Coffea canephora fruits and beans have yielded limited advancements globally, highlighting a developing field of study. Consequently, this review seeks to consolidate existing literature through an analysis focusing on the microbiota and fermentation processes inherent in the postharvest processing of Canephora coffee. To this end, a comprehensive examination of the principal microorganisms inherent to this species, the application of starter cultures in fermentation, and the repercussions of fermentation on the chemical and sensory attributes of the beverage will be expounded. These investigations underscore the influence exerted by the fermentation process and the introduction of microorganism inoculation on Canephora coffees' chemical composition and sensory characteristics. Fermentation emerges as a mechanism facilitating the modification of coffee flavor and aroma, thereby presenting avenues for innovative enhancements in producing distinct Canephora coffee beverages. Research to assess the microbiota of Canephora coffees from various origins has the potential to advance our understanding of the microbial ecology specific to this species. Coffee leaf rust (CLR) is caused by the biotrophic pathogenic fungus Hemileia vastatrix. Despite being the most researched coffee disease, mysteries still exist relating to its epidemiology and biology. The objective of this work is to highlight past and present events concerning this prominent coffee disease. We start with an historical overview of the homeland of Coffea arabica, the Afromontane forest of south‐west Ethiopia, and then follow its journey across the globe linked to colonial trade and power struggles. We report the relevance of CLR to coffee production today, with a focus on the Americas and summarize unproven hypotheses in relation to the cause of recent epidemics. We present an original hypothesis concerning the first major outbreak occurring in 1869 in Ceylon (Sri Lanka), based on geopolitical connections to global trade. We review old and new options for management of the disease. The likelihood of a yet undetected alternate host of H. vastatrix is also considered as an additional piece of the epidemiological puzzle. Finally, we reflect on the interactions between H. vastatrix and a changing climate. By better understanding past events, linked to CLR, we may be better prepared for future outbreaks. Simple Summary: Rain acts as a triggering factor for the mass emergence of females of the coffee berry borer, Hypothenemus hampei (CBB), which shelter in old and unharvested berries, mainly in regions with a dry season. As they emerge from the old berries, thousands of CBB females fly out to search for and infest the coffee berries of the new crop. Very little information is available, however, about the effect microclimate on CBB or, in particular, about the interaction between the shade trees, the microclimate, and the mass emergence of CBB females. We conducted a field experiment in two Robusta coffee plots, one grown under shade trees and the other in full sun exposure. During the coffee intercropping period, when mass emergence of the CBB occurs, we set up a meteorological station to record microclimate data and installed traps to capture the females that fly out during the day, collecting caught individuals every hour. We found that female emergence increased after midday. Although more flying females were captured in the coffee plot grown under shade trees, the relationship between microclimatic variables and CBB mass emergence was similar for both shade conditions. This behavior has important implications for monitoring and pest management, because female CBB populations are more concentrated and exposed to natural and artificial mortality factors. The rainfall regime has a significant impact on the microclimate and mass emergence of the coffee berry borer, Hypothenemus hampei (Coleoptera: Curculionidae) (CBB). Little is known, however, about the shade tree–microclimate–CBB mass emergence interaction. The objective of the present study was to compare the effect of microclimate on the mass emergence of CBB in a full sun-exposed plot with a plot shaded by trees. The experiment was conducted on a Robusta coffee farm in southern Chiapas, Mexico. In each plot, 18 traps baited with an alcohol mixture were installed to capture flying females, collecting caught individuals every hour from 8:00 to 18:00 h. A meteorological station recorded several microclimatic variables on 13 weekly sampling dates from February to May 2022. Significantly more CBB females were captured in the shaded plot. The largest number of CBB captures was recorded between 14:00 and 16:00 h for the shade plot and between 15:00 and 17:00 h for the sun-exposed plot. The mass emergence of CBB showed a positive association with precipitation, dew point, and wind speed samples and a negative association with maximum air temperature, average relative humidity, ultraviolet radiation, wind speed, and equilibrium moisture content. Our observations show that the relationship between shade trees, microclimate, and mass emergence of CBB is complex and that its study helps us to gain deeper insight into CBB bioecology and advance control techniques against this important pest. Specialty coffee has become increasingly consumed in recent years. The growth of the specialty coffee market has also led to a greater appreciation of the unique flavors and characteristics of different coffee species. Coffea arabica (Arabica coffee) and Coffea canephora (Canephora coffee, known internationally as Robusta) are the main species of most significant economic importance worldwide. This study aims to evaluate the use of Meyerozyma guilliermondii in the self-induced anaerobiosis fermentation (SIAF) of Coffea arabica and Coffea canephora. SIAF without inoculation (spontaneous fermentation) and with inoculation of yeast M. guilliermondii was conducted for five days in closed bioreactors. Volatiles and sensory analyses were performed in the roasted coffees. Twenty volatile compounds were identified in Arabica coffee and fifteen in Canephora coffee. The classes found were esters (7), pyrazines (7), ketones (4), alcohols (2), aldehydes (2), phenols (1), and pyrrole (1). Furfuryl alcohol was a significant compound for Arabica coffee, while 4-ethenyl-2-methoxyphenol was notable for Canephora coffee. M. guilliermondii increased significantly (p < 0.05) the score by over 3 points, improved the production of desirable volatile compounds (2-methylpyrazine, 2-ethylpyrazine, furfuryl alcohol, and 5-methylfurfural), enhanced the perception of sweetness and the sensory complexity of Canephora coffee. The SIAF method combined with M. guilliermondii can be a practical approach for improving the sensory quality of the Canephora coffee beverage. Water stress in plants due to the effects of climate change resulted in reduced growth and yield of coffee in the tropics. Efficient use of water through the use of SMART irrigation systems available provides a promising solution to address such a problem. Thus, a study utilizing a combined climate- based and soil moisture SMART irrigation system has been conducted to assess its effects on the growth of Robusta coffee seedlings compared to practiced manual irrigation. The experimental design utilized a Complete Block Design (CBD) approach. Additionally, water consumption was measured to determine any significant differences. Over the course of the two-month experiment, 20 replications per treatment, it was observed that coffee seedlings subjected to the automated irrigation system (T1) exhibited significant improvements in both plant height and number of leaves, with growth rates of 72.56% and 65.29%, respectively, compared to manually irrigated plants (T0). Although the water savings achieved under the automated system were not statistically significant, there was an 18.95% reduction in water consumption. These findings highlight the potential benefits of automating irrigation systems using an IoT approach, specifically in terms of enhancing coffee seedling productivity. By utilizing real-time data from sensors, the SMART irrigation system effectively optimized water usage and resulted in improved growth and yield. This research underscores the importance of implementing such technological advancements to mitigate the adverse effects of climate change on coffee cultivation in tropical regions. A new cafestol-type diterpenoid, 5β-hydroxy-2-oxocafestol named coffecanepholide C (1) along with three known diterpenoids including cafestol (2), tricalysiolide A (3) and atractyligenin (4) were identified from the Coffea canephora trunks collected at Lam Dong province, Vietnam. Their structures were elucidated by HRESIMS and NMR spectroscopic analysis (1H, 13C, COSY, HSQC, HMBC, and NOESY NMR) as well as compared with data in the literature. Upon evaluation of the α-glucosidase inhibitory activity, compound 1 (IC50 = 142.0 ± 0.2 µM) and compound 3 (IC50 = 286.2 ± 1.2 µM) exhibited activity against α-glucosidase, while structures 2 and 4 showed no activity. Furthermore, the docking simulations revealed that the carbonyl groups of compounds 1 and 3 formed hydrogen bonds with Lys506 residue at the enzyme pocket, which may induce the α-glucosidase inhibitory activity. Due to the concern with the contamination of food by pesticides, especially coffee, the aim of this study was to analyze the chemical composition and glyphosate residue in Conilon coffee beans with and without defects and harvested at different times after applying the herbicide. The experiment was in a split-plot design, with the presence or absence of defects in the beans comprising the subplots, and the periods of 15, 30, 45 and 60 days after applying the herbicide corresponding to the split plots. We analyzed the volatile compounds, bioactive compounds, and glyphosate residue in the beans. Regardless of the presence or absence of defects, there was an increase in the concentrations of bioactive compounds in the raw Conilon coffee beans as the interval between applying the herbicide and harvesting the beans was increased. The most abundant volatile compounds per percentage area belonged to the pyrazines, furans and phenols. The amount of glyphosate residue found in the beans exceeded the maximum detectable limit by the ELISA method regardless of the time between application and harvest, corresponding to values that are unacceptable to several purchasing countries, and making the samples unsuitable for export and a risk to food safety. Plant growth regulators (PGR) are essential for somatic embryogenesis (SE) in different species, and Coffea canephora is no exception. In our study model, previously, we have been able to elucidate the participation of various genes involved in SE by using different strategies; however, until now, we have not used a proteomic approach. This research seeks to contribute to understanding the primary cellular pathways involved in developing SE in C. canephora. The process of our model consists of two stages: (1) preconditioning in MS medium with auxin (NAA) and cytokinin (KIN), and (2) induction in Yasuda liquid medium added with cytokinin (BA). Therefore, in this study, we analyzed different days of the SE induction process using shotgun label-free proteomics. An amount of 1630 proteins was found among different sampling days of the process, of which the majority were accumulated during the induction stage. We found that some of the most enriched pathways during this process were the biosynthesis of amino acids and secondary metabolites. Eighteen proteins were found related to auxin homeostasis and two to cytokinin metabolism, such as ABC, BIG, ILR, LOG, and ARR. Ten proteins and transcription factors related to SE were also identified, like SERK1, SKP1, nuclear transcription factor Y, MADS-box, and calreticulin, and 19 related to other processes of plant development, among which the 14-3-3 and PP2A proteins stand out. This is the first report on the proteomic approach to elucidate the mechanisms that operate during the induction of SE in C. canephora. Worldwide coffee production is threatened by climate change, which highlights the importance of heat tolerance studies. Here we tested the hypothesis that photosynthetic heat tolerance in coffee varieties changes according to acclimation to distinct light conditions. Furthermore, we tested if heat tolerance is associated with the habitat of origin of the coffee species.We evaluated heat tolerance using chlorophyll fluorescence in varieties of Coffea arabica (Mundo Novo and Catuai Amarelo) and C. canephora (Conilon) grown in a common garden under two conditions: high (HS) and low (LS) sunlight. Leaf traits associated with leaf cooling were evaluated in plants grown in LS and HS and associations of heat tolerance with these traits were determined.The varieties tested had high photosynthetic heat tolerance, with temperatures above 54 °C leading to a 50% reduction in Fv/Fm (T50). The heat tolerance of each Coffea variety was unaffected by growth in distinct light conditions. Leaves of plants grown in LS were larger and had a lower fraction of the leaf area occupied by stomata (nast). Heat tolerance was positively associated with leaf size and negatively with nast. C. canephora exhibited higher heat tolerance than C. arabica.The limited plasticity of heat tolerance in response to acclimation under distinct light conditions contradicts the prediction that plants acclimated to HS would have higher photosynthetic heat tolerance than those acclimated to LS. We analyzed the volatile compounds, bioactive compounds, and glyphosate residue in the beans. Regardless of the presence or absence of defects, there was an increase in the concentrations of bioactive compounds in the raw Conilon coffee beans as the interval between applying the herbicide and harvesting the beans was increased. The most abundant volatile compounds per percentage area belonged to the pyrazines, furans and phenols. The amount of glyphosate residue found in the beans exceeded the maximum detectable limit by the ELISA method regardless of the time between application and harvest, corresponding to values that are unacceptable to several purchasing countries, and making the samples unsuitable for export and a risk to food safety.

IntroductionThe genetic diversity of Robusta coffee (Coffea canephora), a cornerstone in the global coffee industry, remains not fully explored, leading to a significant gap in our understanding of its sensory intricacies. Our study evaluated the sensory quality potential of the Robusta cultivars from the INERA Coffee Collection in Yangambi (the Democratic Republic of the Congo), the local wild diversity, and their hybrids.MethodsWe evaluated the sensory attributes of 70 genotypes representing the genetic structure of the coffee collection. Of those 70, 22 genotypes were evaluated for two consecutive years to assess the consistency of the sensory quality. Standard coffee cupping with the Fine Robusta Standards and Protocols was enhanced through sensory descriptors from the Coffee Taster’s Flavor Wheel. Each genotype’s sensory profile was constructed based on the Total cupping score and the frequency of reported sensory descriptors. The Total cupping score ranged from 75.75 to 84.75, with a substantial variation in sensory profiles, even within a genetic cluster.Results and discussionNutty/Cocoa was the most frequently reported descriptor class. The sensory profile ideotype exhibits a high frequency of Fruity, Sweet, and Sour/Fermented descriptors and a low frequency of Green/Vegetative, Other, and Roasted descriptors. Evidence suggests that the sensory profile of a genotype is consistent over two harvest years. Genotypes with promising and unique sensory profiles were discovered within the cultivars and the wild – cultivar hybrids. The genetic diversity of wild and cultivated Robusta in the Democratic Republic of the Congo could play an essential role in understanding and improving its sensory quality.

Recent investigations into the microbiota and fermentation of Coffea canephora fruits and beans have yielded limited advancements globally, highlighting a developing field of study. Consequently, this review seeks to consolidate existing literature through an analysis focusing on the microbiota and fermentation processes inherent in the postharvest processing of Canephora coffee. To this end, a comprehensive examination of the principal microorganisms inherent to this species, the application of starter cultures in fermentation, and the repercussions of fermentation on the chemical and sensory attributes of the beverage will be expounded. These investigations underscore the influence exerted by the fermentation process and the introduction of microorganism inoculation on Canephora coffees’ chemical composition and sensory characteristics. Fermentation emerges as a mechanism facilitating the modification of coffee flavor and aroma, thereby presenting avenues for innovative enhancements in producing distinct Canephora coffee beverages. Research to assess the microbiota of Canephora coffees from various origins has the potential to advance our understanding of the microbial ecology specific to this species. Such studies will play a crucial role in identifying pertinent starter cultures that could be used to produce high-quality coffees.

This study evaluated consumers’ perceptions of beverages obtained from the intervarietal hybrids of Coffea canephora, Conilon and Robusta, produced in the Western Amazon, through a home-use test with 127 participants. An acceptance test and a Check-All-That-Apply procedure were applied. Two clones, BRS 2314 and BRS 2357, were studied (both in natural and fermented versions) and their roasted coffee composition was also evaluated. All beverages were described as having a mild aroma, roasted flavor, and slightly sour taste; consumers noticed both the effect of the fermentation process and genetics. Natural coffees had greater sensory acceptance than fermented ones. Natural coffees were most associated with a slightly bitter taste, and were well accepted by almost half of the participants, mainly women. Fermented coffees were both associated with fermented flavor, but also presented specific characteristics. BRS 2314F was most associated with a fruity flavor and slightly bitter taste and was well accepted by younger consumers with higher levels of education. BRS 2357F was most associated with tobacco flavor and bitter taste and was well accepted by older consumers with a lower education level and a higher coffee consumption frequency. The positive acceptance of C. canephora intervarietal hybrid beverages allowed us to confirm its material market potential.

Perawatan luka bakar memerlukan penanganan khusus. Penggunaan sediaan obat topikal seperti silver sulfadiazine serta hydrocolloids yang berfungsi sebagai absorptive dressings merupakan salah satu prosedur terapi yang digunakan untuk mempercepat penyembuhan luka. Perawatan kasus luka bakar di Indonesia masih kurang optimal karena adanya pembatasan biaya perawatan oleh sistem jaminan kesehatan Indonesia, oleh karena itu, perlu adanya alternatif perawatan luka bakar dengan mengembangkan bahan-bahan herbal sebagai dressing topikal seperti ekstrak dari kopi. Tujuan dari penelitian ini ialah untuk membuktikan pengaruh pemberian gel ekstrak kopi robusta (Coffea canephora) terhadap gambaran makroskopis dan mikroskopis luka bakar derajat IIB pada tikus long evans. Metode penelitian yang digunakan adalah true experimental laboratories dengan rancangan post-test only control group design. Tikus sebanyak 24 ekor terbagi menjadi 3 kelompok perlakuan. Kelompok kontrol negatif yaitu diberi basis gel, kelompok positif diberi silver sulfadiazine, dan kelompok perlakuan diberi gel ekstrak kopi robusta 2,5%. Data luas luka bakar, perubahan intensitas warna, dan ketebalan epitel luka bakar yang telah diperoleh selanjutnya diuji secara statistik menggunakan One Way ANOVA maupun Kruskal-Wallis dan dilanjutkan dengan uji Post-Hoc. Hasil penelitian menunjukkan pada kelompok perlakuan terjadi penurunan luas makroskopis (p < 0,05), perbaikan intensitas warna (p < 0,05), dan peningkatan ketebalan epitel (p < 0,05) antara gel ekstrak kopi robusta dibandingkan dengan kelompok kelompok negatif. Kesimpulan penelitian ini adalah bahwa gel ekstrak kopi robusta (Coffea canephora) terbukti memberikan pengaruh terhadap penyembuhan luka baik secara makroskopis maupun mikroskopis sehingga gel ekstrak kopi dapat terus dikembangkan menjadi salah satu alternatif dressing dalam perawatan luka bakar.

Abstract The objective of this work was to evaluate the morphophysiological responses and cutting production of clones of Conilon coffee (Coffea canephora) cultivars in a super-dense clonal garden in the state of Espírito Santo, Brazil. The super-dense clonal garden was built in 2019 using 39 clones: 9, 9, 9, and 12 of cultivars Centenária ES8132, Diamante ES8112, ES8122 (Jequitibá), and Marilândia ES8143, respectively. The experiment was carried out in a randomized complete block design, with three replicates. Cutting production and the following morphophysiological traits were evaluated at 9 and 18 months after planting: chlorophyll index, normalized difference vegetation index, plant height, canopy height, canopy diameter, number of shoots, number of viable cuttings, number of leaves, fresh leaf mass, and plant fresh and dry matter mass. The super-dense clonal garden caused different morphophysiological responses among the studied clones. In general, clones C2, C5, C6, C8, D1, D8, D9, J8, M2, M9, M10, and M12 showed a higher mean cutting production, whereas C4, J1, J4, M4, and M5 were the most sensitive to the super-dense regime. Under these conditions, it is recommended to increase the proportion of matrix plants of the latter clones.

Coffee genotypes cultivated in the Amazonian region have been gaining increasing prominence in Brazilian plantations. This study aimed to quantify nutrient accumulation in the fruits, grains, and husks of Robusta coffee genotypes cultivated in the Brazilian Amazon and estimate genetic diversity. The experiment was conducted in Alta Floresta D'Oeste—Rondônia, Brazil. To assess nutrient accumulation, fresh fruits were collected. These were dried, processed, separated into grains and husks, and subjected to chemical analysis. Nutrient accumulation in fruits, grains, and husks, as well as the grain/husk ratio, underwent analysis of variance through the F-test (p < 0.01. For each evaluated trait, the experimental coefficient of 337 variation (CVe), genetic coefficient of variation (CVg), and genotypic determination coefficient (H2) were also estimated. Variability was observed among Robusta coffee genotypes, with VP06, AS4, and AS10 being the most dissimilar. LB080 had the lowest dry fruit weight and the lowest percentage of grains in relation to husks. ZD156 accumulated more K in the grains, while VP06 and AS10 were the genotypes that accumulated more nutrients in the husks. Nutrients N, K, Ca, and P are accumulated in larger quantities, necessitating the calibration of mineral fertilization dosages and distribution. [ABSTRACT FROM AUTHOR]

The coffee quality is affected by 40% pre-harvest, 40% post-harvest, and 20% export handling. Besides, future risks for the coffee industry are related with climate change and increased pathogens. Considering the importance of the aroma profile and unique flavor of Arabica coffee, most literature focuses on this variety because of the high market share; however, nowadays, Robusta coffee stands out for its increasing industrial value and resistance to drought. In this review, both species are emphasized, highlighting sensory aspects of possible new products mixed with a higher proportion of Robusta given market trends for bitter beverages. In the present work, a systematic search of peer-reviewed literature evaluates how the coffee cup quality and physicochemical characteristics of Robusta and Arabica are influenced by environmental, agronomic, and further processing factors. Somatic embryogenesis (SE) is an excellent example of mass plant propagation. Due to its genetic variability and low somaclonal variation, coffee SE has become a model for in vitro propagation of woody species, as well as for large-scale production of vigorous plants that are advantageous to modern agriculture. The success of the large-scale propagation of an embryogenic system is dependent on the development, optimization, and transfer of complementary system technologies. In this study, two successful SE systems were combined with a SETIS™ bioreactor immersion system to develop an efficient and cost-effective approach for the in vitro development of somatic embryos of Coffea spp. This study used an efficient protocol for obtaining somatic embryos, utilizing direct and indirect SE for both C. canephora and C. arabica. Embryos in the cotyledonary stage were deposited in a bioreactor to complete their stage of development from embryo to plant with minimal manipulation. Following ten weeks of cultivation in the bioreactor, complete and vigorous plants were obtained. Different parameters such as fresh weight, length, number of leaves, and root length, as well as stomatal index and relative water content, were recorded. In addition, the survival rate and ex vitro development of plantlets during acclimatization was assessed. The best substrate combination was garden soil (GS), peat moss (PM), and agrolite (A) in a 1:1:0.5 ratio, in which the bioreactor-regenerated plants showed an acclimatization rate greater than 90%. This is the first report on the use of SETIS™ bioreactors for the in vitro development of somatic embryos in Coffea spp., providing a technology that could be utilized for the commercial in vitro propagation of coffee plants. A link between research and innovation is necessary to establish means of communication that facilitate technology transfer. This protocol can serve as a basis for the generation and scaling of different species of agroeconomic importance. However, other bottlenecks in the production chains and the field must be addressed. In the present work, it was phenotyped 29 Coffea canephora clones for resistance to Hemileia vastatrix races II and XXXIII and investigated the presence of molecular markers associated with Coffee Leaf Rust (CLR) and Coffee Berry Disease (CBD) resistance loci in 67 clones. Phenotyping for resistance was performed by evaluating six components of quantitative resistance using the leaf disc technique. The averages of the components were submitted to the Scott-Knott test and to the multivariate analysis to obtain the generalized Mahalanobis distance, which was used to group the clones by the unweighted pair-group method using arithmetic averages. In screening resistance to CLR, molecular markers were evaluated associated with two resistance loci. Three sequence characterized amplified regions (SCAR) and a simple sequence repeats (SSR) markers associated with the locus of SH3 gene (Locus A); a SCAR and a cleaved amplified polymorphic site (CAPS) associated with a locus of resistance to races I, II and pathotype 001 of H. vastatrix (Locus B). For screening resistance to CBD, two SSRs were associated with the locus of the resistance Ck-1 gene (Locus C) were evaluated. The phenotyped clones were grouped into five resistance classes for each H. vastatrix race. Fifteen clones have locus of resistance to races I, II, and the pathotype 001 of H. vastatrix. Three clones have the Ck-1 gene. These results support growers to choose resistant clones for crop composition and indicate potential sources of CLR and CBD resistance for C. canephora breeding programs. [ABSTRACT FROM AUTHOR]

Consumers are increasingly looking for healthy foods without the addition of synthetic additives. The aim of this study was to evaluate the efficiency of coffee extracts as a natural antioxidant in fresh pork sausage. Firstly, the conditions for obtaining coffee green extracts were optimized (Central Composite Rotatable Design 23, variables: extraction time, ethanol–water ratio, and sample–solvent ratio) in an ultrasound bath (70 °C). The response variables were the bioactive compounds levels and antioxidant activity. Valid models were obtained (p ≤ 0.05, R2 > 0.751), with higher bioactive content and antioxidant activity in the central point region. Extracts of Robusta and Arabica coffee green (RG and AG) and medium roast (RR and AR) obtained, and central point (10 min, an ethanol concentration of 30%, and a sample–solvent ratio of 10 g/100 mL) and optimized (14.2 min, 34.2%, and 5.8 g/100 mL) parameters were characterized. The RG presented a significantly (p ≤ 0.05) higher content of caffeine (3114.8 ± 50.0 and 3148.1 ± 13.5 mg/100 g) and 5-CQA (6417.1 ± 22.0 and 6706.4 ± 23.5 mg/100 g) in both extraction conditions, respectively. The RG and RR coffee presented the highest antioxidant activity. Two concentrations of RG and RR coffee extracts were tested in fresh pork sausage. The Robusta coffee extract presented the highest antioxidant activity in both roasted and green states. However, when applied to a meat product, the extract prepared with RG coffee showed better results, with efficiency in replacing synthetic antioxidants (content of malonaldehyde/kg of sample below 0.696 ± 0.059 in 20 days of storage), without altering the sensory attributes of the product (average scores above 7.16 ± 1.43 for all attributes evaluated). Therefore, the RG coffee extract was a suitable alternative as a natural antioxidant applied to fresh pork sausage.

In-field data were collected in Costa Rica between 2018–2021 on newly planted grafted and non-grafted coffee plants grown under artificial shade nets and across an elevation gradient (1050, 1250 and 1450 m.a.s.l). The coffee plants consisted of Coffea arabica F1 hybrid plants (‘H3 i.e. Caturra cv. X Ethiopian 531’), which were derived from a somatic embryogenesis clonal propagation process, an American C. arabica pure line (‘Villa Sarchi’) and C. canephora ‘Nemaya’ (the latter two both being produced by seed). Data from eight different coffee types (including these three genotypes) and different grafting combinations (including reverse and auto-grafting) were collected. Data concerned plant traits such as grafting compatibility (plant collar diameters above and below graft union), agronomic characteristics (aerial and root traits), leaf ecophysiology (leaf gas-exchange and chlorophyll fluorescence), yield and quality attributes (bean size, peaberry percentage, WB100 and SCA note). Climate data were also included for comparison on the farm plots along the elevation gradient. Linear mixed models were used to test for effects of elevation (test sites), coffee types (grafted or non-grafted combinations) and interaction between coffee types and elevations. Least square mean estimates were calculated for significant fixed effects and Tukey tests applied for pairwise tests. A tangential hyperbola curve was used to analyse leaf gas-exchange data. These datasets and R scripts can be re-used as a guide for future analyses concerning coffee agronomy or eco-physiological interactions for other plant species. Other potential re-uses could be meta-analyses aimed at comparing coffee yield, quality, or other agronomic traits across different environmental conditions (such as under shade of an agroforestry system or across different elevation sites).

Coffee processing is a major contributor to the creation of food and product waste. Using coffee co-products can play an essential role in addressing environmental problems and issues with nutritionally unbalanced foods, population growth, and food-related diseases. This research aimed to determine the quality and sensory parameters (aw, pH, dry matter, TAC, TPC, fat, fatty acids profile, fiber, caffeine, chlorogenic acids, color, and sensory analysis) of different botanical origins of cascara (coffee husks) and silverskin (thin layer). The results of this study show that silverskin and cascara are a good source of TAC (1S 58.17 ± 1.28%, 2S 46.65 ± 1.20%, 1C 36.54 ± 1.84%, 2C 41.12 ± 2.11%). Cascara showed the presence of polyphenols (2C 49.135 g GAE·kg−1). Coffee co-products are good sources of fiber. Silverskin had higher values of caffeine than cascara. Palmitic, stearic, oleic, linoleic, and arachidic acids were the most represented acids in the samples. Given the obtained results, cascara can be considered "low-fat" (1C 4.240 g·kg−1 and 2C 5.4 g·kg−1). Based on the sensory evaluation, no sample reached the acceptable index value of 70%. Understanding the link between the character, identification properties, and composition of coffee co-products of different botanical origins can enable their application in the food industry. The research attempts to analyze the phytochemical compositions of both the leaves and the cherries of coffee and examine preference through the sensory evaluation of consumers consuming tea products of Robusta coffee from Songkhla, Thailand. The methods used for the phytochemical analysis were conducting an antioxidant content analysis using the Ferric ion reducing antioxidant power method, conducting total phenolic and tannin contents analysis using the Folin-Ciocalteau method, and conducting a total flavonoid content measurement using the aluminum chloride colorimetry method. For the sensory evaluation of tea products (leaves and cherries of coffee), a set of questionnaires was used as the survey observation tools to collect the data from consumers at some coffee and tea shops. The data analysis was using T-test for phytochemical composition and F-test on sensory evaluation. This study shows that Robusta coffee leaves and cherries contained phytochemical compounds of different values. Compared to the cherries, the leaves had higher values of phytochemical compositions. Based on the sensory evaluation of consumers, tea products made from leaves and cherries were highly rated. Coffee cherry tea demonstrated a higher percentage of interest compared with coffee leaf tea.

Humans have had a major influence on the dissemination of crops beyond their native range, thereby offering new hybridization opportunities. Characterizing admixed genomes with mosaic origins generates valuable insight into the adaptive history of crops and the impact on current varietal diversity. We applied the ELAI tool—an efficient local ancestry inference method based on a two-layer hidden Markov model to track segments of wild origin in cultivated accessions in the case of multiway admixtures. Source populations—which may actually be limited and partially admixed—must be generally specified when using such inference models. We thus developed a framework to identify local ancestry with admixed source populations. Using sequencing data for wild and cultivated Coffea canephora (commonly called Robusta), our approach was found to be highly efficient and accurate on simulated hybrids. Application of the method to assess elite Robusta varieties from Vietnam led to the identification of an accession derived from a likely backcross between two genetic groups from the Congo Basin and the western coastal region of Central Africa. Admixtures resulting from crop hybridization and diffusion could thus lead to the generation of elite high-yielding varieties. Evaluation of genetic uniformity is important in a perennial plant like coffee propagated through somatic embryogenesis. In this study, we compared the genetic homogeneity of somatic embryo-derived plants and the seed-derived plants obtained from a single mother plant of robusta coffee cultivar by employing Sequence Related Amplified Polymorphism (SRAP) and Start Codon Targeted (SCoT) molecular markers and sequencing four functional genes. The SRAP and SCoT molecular markers revealed 96% genetic similarity between somatic embryo-derived and seed-derived plants with the mother plant. The sequencing analysis has shown the minimum sequence variability in the chloroplast maturase K gene and the highest sequence variability in the zinc finger protein gene compared to NAC25 and UDP-glycosyltransferase genes. The presence of SNP and indels created polymorphism in gene sequences. The percent frequency of SNP varied from 0.0 to 0.45 in matK, 0.36 to 1.40 in NAC25, 0.81 to 2.63 in the UDP-glycosyltransferase gene and 3.30 to 8.70 in the zinc finger protein gene. In the case of NAC25 and UDP-glycosyltransferase genes, the seed-derived plants have shown a higher frequency of SNP compared to somatic embryo-derived plants. However, in the case of zinc finger protein gene, tissue culture-derived plants accumulated a two-fold higher frequency of SNP compared to the seed-derived plants. The sequence characterization of the four genes has revealed the presence of a higher frequency of non-synonymous SNPs compared to synonymous SNPs. This is the first report on the comparative genetic uniformity analysis of somatic embryo-derived and seed-derived plants in robusta coffee cultivar with significant practical implications. Phytochemical investigation of the trunks of Coffea canephora yielded two new ent-kaurane diterpene diastereomers, which have been named coffecanepholide A, ent-3β,16β,17-trihydroxykauran-18-al (1) and coffecanepholide B, ent-3β,16β,17-trihydroxykauran-19-al (2). Structural elucidation and configurational assignment were deduced from extensive spectroscopic NMR/HRESIMS analysis and by comparison with the spectral data of the literature relevant structures. The isolated compounds were assayed for in vitro inhibitory activities against α-glucosidase. Structure 2 showed the α-glucosidase inhibitory activity with an IC50 value of 294.7 ± 0.9 μM, while compound 1 exhibited inactivity. In addition, the docking results revealed that structure 2 can form more interactions with amino acid residues at the active site of α-glucosidase, which gave a more negative binding energy (−9.56 kcal/mol) compared with 1 (−8.60 kcal/mol). Coffea canephora plant stem cells can have bioactive compounds with tissue repairing and anti-inflammatory action. This study aimed to develop a liposomal stem cell extract formulation obtained from the leaves of C. canephora (LSCECC) and to investigate its capacity to contribute to the dynamic mechanisms of tissue repair. The liposome cream was developed and characterized through the dynamic light scattering technique, atomic force microscopy, and transmission electron microscopy. The excisional full-thickness skin wound model was used and daily topically treated with the LSCECC formulation or vehicle control. On days 2, 7, 14, and 21 after wounding, five rats from each group were euthanized and the rates of wound closure and re-epithelialization were evaluated using biochemical and histological tests. LSCECC resulted in faster re-epithelialization exhibiting a significant reduction in wound area of 36.4, 42.4, and 87.5% after 7, 10, and 14 days, respectively, when compared to vehicle control. LSCECC treated wounds exhibited an increase in granular tissue and a proper inflammatory response mediated by the reduction of pro-inflammatory cytokines like TNF-α and IL-6 and an increase of IL-10. Furthermore, wounds treated with LSCECC showed an increase in the deposition and organization of collagen fibers at the wound site and improved scar tissue quality due to the increase in transforming growth factor-beta and vascular endothelial growth factor. Our data showed that LSCECC improves wound healing, the formation of extracellular matrix, modulates inflammatory response, and promotes neovascularization being consider a promising bioactive extract to promote and support healthy skin. The graphical presents the action of LSCECC in all four phases of wound healing and tissue repair. The LSCECC can reduce the inflammatory infiltrate in the inflammatory phase by decreasing the pro-inflammatory cytokines like IL-6 and TNF-α, in addition to maintaining this modulation through lesser activation and recruitment of macrophages. The LSCECC can also increase the release of IL-10, an anti-inflammatory cytokine, decreasing local edema. The increase in VEGF provides neovascularization and the supply of nutrients to newly repaired tissue. Coffea canephora has high but inadequately exploited genetic diversity. This diversity, if well exploited, can sustain coffee productivity amidst climate change effects. Drought and heat stress are major global threats to coffee productivity, quality, and tradable volumes. It is not well understood if there is a selectable variation for drought stress tolerance in Robusta coffee half-sibs as a result of watering deficit pre-exposure at the germination stage. Half-sib seeds from selected commercial clones (KR5, KR6, KR7) and a pipeline clone X1 were primed with deficit watering at two growth stages followed by recovery and later evaluated for tolerance to watering deficit stress in three different temperature environments by estimation of plant growth and wilt parameters. Overall, the KR7 family performed the best in terms of the number of individuals excelling for tolerance to deficit watering. In order of decreasing tolerance, the 10 most promising individuals for drought and heat tolerance were identified as: 14.KR7.2, 25.X1.1, 35.KR5.5, 36.KR5.6, 41.KR7.5, 46.KR6.4, 47.KR6.5, 291.X1.3, 318.X1.3, and 15.KR7.3. Agronomic performance of Robusta coffee (Coffea canephora) is affected by genotype by environment interaction, which demands multi-environment testing of genotypes to identify superior genotypes. The objectives of this study were to (1) estimate genetic parameters and analyze the effects of genotype (G), environment (E), and genotype by environment (G × E) interaction on growth and yield traits of C. canephora genotypes, (2) identify superior genotypes that exhibit high stability by combining high growth and yield with broad or specific environmental adaptation, and (3) identify environments that best represent the target environment for high trait expression. Thirty-nine C. canephora hybrids were evaluated using a randomized-complete-block design with three replications at New Tafo-Akim and Bechem in the Eastern and Ahafo regions of Ghana, respectively. Additive main effect and multiplicative interaction (AMMI) analysis of variance revealed highly significant (p < 0.001) differences among G, E, and G × E interaction effects for all traits. AMMI2 biplot analysis revealed the presence of 3, 2, 3, 2, and 3 mega environments for trunk cross-sectional area, height, span, number of laterals, and yield, respectively. Identifying mega environments for these traits will help expedite Robusta coffee breeding through the reduction of the number of test environments needed for phenotype evaluations. The AMMI analyses indicated genotypes G17 (E138 × C180), G35 (PA193 × C180), G4 (197 × PA413), G1 (149 × C193) and G30 (E152 × 149) have high and stable yields across all test environments. ABSTRACT This work evaluated the initial performance and genetic diversity of Coffea canephora genotypes cultivated in environments at contrasting altitudes. Fourteen morphophysiological traits and seven descriptors of the genus Coffea spp. of coffee trees cultivated at altitudes of 140 m and 700 m were evaluated. The design used was Federer’s augmented block in a 2 × 112 factorial scheme with six blocks. The first factor was the two environments, and the second was the 112 genotypes, with eight common treatments, being five conilon coffee clones and three arabica coffee cultivars. The data were analyzed by the method of REML/BLUP and genetic correlation method. Genetic diversity was evaluated by estimating the distance matrix, applying the Gower methodology followed by the clustering method by Tocher and UPGMA. The phenotypic means were higher in the environment at an altitude of 700 m, except for plant height, number of leaves, and canopy height (CH). Genotypic effects were significant for most traits except for leaf width, CH, unit leaf area, and total leaf area. A wide genetic diversity was verified, with distances varying from 0.037 to 0.593 for the pairs of genotypes 26 × 93 and T7 × 76, respectively. Most of the traits studied showed high genotypic correlation with the environment and expressive genetic correlation between the evaluated traits thereby demonstrating the possibility of indirect selection. There is an adaptation of conilon coffee genotypes to high altitudes and the possibility of developing a specific cultivar for the southern state of Espírito Santo.

Cultivation of Robusta coffee is likely to gain importance because of its high disease resistance and climate envelope. Robusta coffee genetic resources conserved in field genebanks can play an important role to further improve its cupping quality and other agronomic traits, but such Coffea canephora collections are limited and still poorly characterized. In this study, we characterized the genetic composition of the historically important but until recently neglected INERA Coffee Collection in Yangambi (the Democratic Republic of Congo). We used GBS to discover genome-wide genetic diversity, created and validated a novel multiplex amplicon sequencing (HiPlex) screening assay to genetically screen 730 coffee shrubs of the Yangambi Coffee Collection, grouped clonal material and delineated 263 accessions with unique genetic fingerprints. Comparison to reference material of three genetic origins revealed that the majority of the Yangambi accessions were assigned a ‘Lula’ cultivar origin, four accessions were assigned to Congolese subgroup A and nine accessions were most closely related to local wild accessions. About one-quarter of the accessions was likely derived from hybridization between these groups, which could result from seed-based propagation of the collection, breeding efforts, or natural cross-pollination. Parental analyses discovered eight preferentially used accessions, which may correspond to historically selected founders, or direct descendants thereof, whose seed material was once widely used to establish coffee plantations. Finally, two core collections were proposed using the maximization strategy (CC-I; 100 accessions) and genetic distance method (CC-X; 10 accessions). Our study demonstrates a method for the genetic characterization of Robusta coffee collections in general and contributes to the re-evaluation and exploration of the Robusta coffee genetic resources in the Democratic Republic of the Congo in particular.

Plant cell cultures have become a promising production platform of bioactive compounds for biomedical and cosmetic uses in the last decades. However, the success so far has been limited. The study aimed to evaluate the effectiveness of this unique biotechnology process to obtain a bioactive stem cell extract of Coffea canephora (SCECC) with antioxidant, anti-inflammatory, and regenerative properties. Total phenolic and flavonoid contents were determined in the SCECC by spectrophotometry. The chemical composition of the extracts was characterized by mass spectrometry. Antioxidant activity was evaluated using the colorimetric methods of free radical scavenging 2,2'-azinobis-3-ethylbenzothiazoline-6-sulfonic acid (ABTS) and the ferric reducing ability of plasma (FRAP). The anti-inflammatory activity was determined in lipopolysaccharide-stimulated RAW 264.7 macrophages through the production of superoxide anion (O2•-), nitric oxide (NO), tumor necrosis factor-alpha (TNF-α), interleukin-6 (IL-6), and the activity of nuclear factor kappa B (NF-κB). Moreover, the ability of SCECC to stimulate the proliferation and migration of fibroblasts was assessed. Five compounds were tentatively identified, two flavonoids, two phenolic acids, and one sugar. High phenolic content and antioxidant activity were observed in the SCECC. SCECC promoted the proliferation and migration of fibroblasts and suppressed the pro-inflammatory mediators O2•-, NO, TNF-α, and IL-6 in a dose-dependent manner. Moreover, SCECC inhibited the NF-κB transcription factor. Therefore, we obtained evidence that the extract from C. canephora stem cells can be used as a natural agent against skin damage. Hence, it could be of interest in cosmetics for preventing skin aging.

The rainfall regime has a significant impact on the microclimate and mass emergence of the coffee berry borer, Hypothenemus hampei (Coleoptera: Curculionidae) (CBB). Little is known, however, about the shade tree–microclimate–CBB mass emergence interaction. The objective of the present study was to compare the effect of microclimate on the mass emergence of CBB in a full sun-exposed plot with a plot shaded by trees. The experiment was conducted on a Robusta coffee farm in southern Chiapas, Mexico. In each plot, 18 traps baited with an alcohol mixture were installed to capture flying females, collecting caught individuals every hour from 8:00 to 18:00 h. A meteorological station recorded several microclimatic variables on 13 weekly sampling dates from February to May 2022. Significantly more CBB females were captured in the shaded plot. The largest number of CBB captures was recorded between 14:00 and 16:00 h for the shade plot and between 15:00 and 17:00 h for the sun-exposed plot. The mass emergence of CBB showed a positive association with precipitation, dew point, and wind speed samples and a negative association with maximum air temperature, average relative humidity, ultraviolet radiation, wind speed, and equilibrium moisture content. Our observations show that the relationship between shade trees, microclimate, and mass emergence of CBB is complex and that its study helps us to gain deeper insight into CBB bioecology and advance control techniques against this important pest.

Plant growth regulators (PGR) are essential for somatic embryogenesis (SE) in different species, and Coffea canephora is no exception. In our study model, previously, we have been able to elucidate the participation of various genes involved in SE by using different strategies; however, until now, we have not used a proteomic approach. This research seeks to contribute to understanding the primary cellular pathways involved in developing SE in C. canephora. The process of our model consists of two stages: (1) preconditioning in MS medium with auxin (NAA) and cytokinin (KIN), and (2) induction in Yasuda liquid medium added with cytokinin (BA). Therefore, in this study, we analyzed different days of the SE induction process using shotgun label-free proteomics. An amount of 1630 proteins was found among different sampling days of the process, of which the majority were accumulated during the induction stage. We found that some of the most enriched pathways during this process were the biosynthesis of amino acids and secondary metabolites. Eighteen proteins were found related to auxin homeostasis and two to cytokinin metabolism, such as ABC, BIG, ILR, LOG, and ARR. Ten proteins and transcription factors related to SE were also identified, like SERK1, SKP1, nuclear transcription factor Y, MADS-box, and calreticulin, and 19 related to other processes of plant development, among which the 14-3-3 and PP2A proteins stand out. This is the first report on the proteomic approach to elucidate the mechanisms that operate during the induction of SE in C. canephora. This article reports the first case of Pseudopestalotiopsis theae, a fungal plant disease, causing leaf spot on robusta coffee in the Philippines. The disease was observed on robusta coffee trees in New Bantangan, Columbio, Sultan Kudarat, and affected approximately 85% of the leaves on affected plants. The fungus was identified as Pseudopestalotiopsis theae through morphological and molecular analysis. This finding is significant as robusta coffee is a major crop in the Philippines, and further research is needed to understand the impact of this pathogen and develop management strategies to protect coffee production. The black twig borer, Xylosandrus compactus (Eichhoff), was detected in Tepexilotla, town of Zoquitlán, municipality of Sierra Negra de Puebla, Mexico in Nov 2021. A subsequent visit of the infested area and the municipalities surrounding Zoquitlán, carried out in Jun and Jul 2022, showed that X. compactus infested mainly Robusta coffee but also to a lesser extent some Arabica coffee plants. In 2022, the insect pest was detected in coffee plantations in communities in the municipality of Tezonapa, Veracruz and in communities in the municipality of Santa María Chilchotla, Oaxaca, close to the infested area in Puebla. Now, with this report, there are 3 species of the genus Xylosandrus in Mexico: X. curtulus, X. morigerus, and X. compactus; of these, X. morigerus and X. compactus are economically important pests for coffee crops. El taladrador negro de las ramas, Xylosandrus compactus (Eichhoff) fue detectado en Tepexilotla, localidad de Zoquitlán, municipio de la Sierra Negra de Puebla, México en Noviembre de 2021. Una revisión posterior del área infestada y de los municipios aledaños a Zoquitlán, realizada en junio y julio de 2022, mostró que X. compactus infestaba principalmente café Robusta, pero también algunas plantas de café Arábica. En 2022, el insecto plaga se detectó en cafetales de comunidades del municipio de Tezonapa, Veracruz y en comunidades del municipio de Santa María Chilchotla, Oaxaca, cercanas a la zona infestada en Puebla.
