Abstract

The Guinea fowl (Numidameleagris) presents a highly resistant eggshell compared to the other birds. We will described in this study the particular ultrastructure of the Guinea fowl shell that confers exceptional mechanical properties and how changes in organic matrix components controls the development of this structure. The inner part of the shell is similar to other birds, but an additional change in the size and orientation of crystals (switch) is observed at about 1/3 of the calcified layer. Large columnar calcite units break into smaller crystal units with varying crystallographic orientations forming a microstructure with an intricate interlacing of calcite crystals.We recently reported the Guinea fowl shell structural organization from the micro to the Angstrom, which underlined that this particular shell is a bilayer structure. Organic matrix is suspected to firstly induce the initial microstructure shift and then the secondary nucleation events resulting in smaller crystals with increasing misorientations. Consequently, the change of the intra-crystalline organic matter level during the crystal switch was also investigated. A proteomic survey allowed us to identify and characterize 149 proteins in Guinea fowl shell. These proteins were quantified at five calcification stages corresponding to the first events of mineral deposition, the growth of calcite units just prior the shift of crystal orientation, then to the period of the deposition of newly formed crystalline shape and to later stage when the growth of the newly formed crystals is stabilized after the microstructure shift and the secondary nucleation events. We have observed 61 matrix proteins only present in the shift period and potentially responsible of the change of mineral in the shell. Amongst them are calcium binding proteins (NPNT-X1, CALBP1, Protein S100-A6, ANXA1 and 2, CDH2…), core proteins of proteoglycans (TSKU, GPC4…), and other proteins regulating the activity of proteins driving the mineralization (SSP1, OC-116GDF6…). These proteins interact with mineral to produce changes in crystal size and orientation and consequently the new shell structure and its resulting mechanical properties. Data obtained will allow the determination of biological markers that will be used for the genomic selection of chicken layers with improved mechanical shell mechanical properties. Additionally, they provided a list of organic products that will be tested as additives for material and ceramics.

Abstract

International audience; Water stress and sulfur-deficiency are two constraints increasingly faced by crops due to climate change and low-input practices. To investigate their interplay in the grain legume pea (Pisum sativum L.), sulfate was depleted at mid-vegetative stage and a moderate 9-day water stress period was imposed during the early reproductive phase. The combined stress accelerated seed production, lowering yield, one-seed weight and seed number per plant, but rebalanced seed globulin composition. In fact, the moderate water stress mitigated the negative effect of sulfur-deficiency on the accumulation of sulfur-rich globulins in seeds, probably due to a lower seed sink strength for nitrogen, enabling a readjustment of the sulfur-poor/sulfur-rich globulin ratio. These results uncovered the importance of sulfur for stabilizing seed yield in pea facing short and moderate water stress episodes, and showed that the adaptive response of sulfur-deprived plants to water stress is much more complicated than a simple additive response. A transcriptome analysis of developing seeds at the end of the combined stress period clearly advanced our knowledge of the seed response to the single or double stresses. Among the results that will be presented is a specific up-regulation of a set of genes in response to the combined stress, indicating the establishment of unique regulatory processes when sulfur-deficiency is combined with water stress and highlighting candidate genes to fine-tune the regulation of globulin synthesis under stressed conditions.

Abstract

Water stress and sulfur (S) deficiency are two constraints increasingly faced by crops due to climate change and low-input agricultural practices. To investigate their interaction in the grain legume pea (Pisum sativum), sulfate was depleted at the mid-vegetative stage and a moderate 9-d water stress period was imposed during the early reproductive phase. The combination of the stresses impeded reproductive processes in a synergistic manner, reducing seed weight and seed number, and inducing seed abortion, which highlighted the paramount importance of sulfur for maintaining seed yield components under water stress. On the other hand, the moderate water stress mitigated the negative effect of sulfur deficiency on the accumulation of S-rich globulins (11S) in seeds, probably due to a lower seed sink strength for nitrogen, enabling a readjustment of the ratio of S-poor (7S) to 11S globulins. Transcriptome analysis of developing seeds at the end of the combined stress period indicated that similar biological processes were regulated in response to sulfur deficiency and to the combined stress, but that the extent of the transcriptional regulation was greater under sulfur deficiency. Seeds from plants subjected to the combined stresses showed a specific up-regulation of a set of transcription factor and SUMO ligase genes, indicating the establishment of unique regulatory processes when sulfur deficiency is combined with water stress., Moderate water stress combined with sulfur deficiency during seed development in pea synergistically affects reproductive processes, but mitigates the negative impact of the deficiency on the seed transcriptome and rebalances seed globulin composition.

Abstract

Polymorphonuclear neutrophils (PMNs) can contribute to the regulation of the host immune response by crosstalk with innate and adaptive leukocytes, including NK cells. Mechanisms by which this immunoregulation process occurs remain incompletely understood. Here, we focused on the effect of human neutrophil-derived serine proteases on NKp46, a crucial activating receptor expressed on NK cells. We used flow cytometry, Western blotting, and mass spectrometry (MS) analysis to reveal that cathepsin G [CG; and not elastase or proteinase 3 (PR3)] induces a time- and concentration-dependent, down-regulatory effect on NKp46 expression through a restricted proteolytic mechanism. We also used a functional assay to demonstrate that NKp46 cleavage by CG severely impairs NKp46-mediated responses of NK cells, including IFN-? production and cell degranulation. Importantly, sputa of cystic fibrosis (CF) patients, which have high concentrations of CG, also alter NKp46 on NK cells. Hence, we have identified a new immunoregulatory mechanism of neutrophils that proteolytically disarms NK cell responses.
