Abstract

Kuphus polythalamius (Teredinidae) is one of the world's largest, most rarely observed, and least understood bivalves. Kuphus polythalamius is also among the few shallow-water marine species and the only teredinid species determined to harbor sulfur-oxidizing chemoautotrophic (thioautotrophic) symbionts. Until the recent discovery of living specimens in the Philippines, this species was known only from calcareous hard parts, fossils, and the preserved soft tissues of a single large specimen. As a result, the anatomy, biology, life history, and geographic range of K. polythalamius remain obscure. Here we report the collection and description of the smallest living specimens of K. polythalamius yet discovered and confirm the species identity of these individuals by using sequences of three genetic markers. Unlike previously collected specimens, all of which have been reported to occur in marine sediments, these specimens were observed burrowing in wood, the same substrate utilized by all other members of the family. These observations suggest that K. polythalamius initially settles on wood and subsequently transitions into sediment, where this species may grow to enormous sizes. This discovery led us to search for and find previously unidentified and misidentified wood-boring specimens of this species within museum collections, and it allowed us to show that the recent geographic range (since 1933) of this species extends across a 3000-mile span from the Philippines to Papua New Guinea and the Solomon Islands., IntroductionSince the initial misidentification of Kuphus polythalamius as a serpulid polychaete by Linnaeus in 1758, the biology of this species has remained enshrouded in mystery. Abstract

Somatostatin (SS) is a peptide hormone with diverse physiological roles. By investigating a deep-water clade of fish-hunting cone snails, we show that predator-prey evolution has generated a diverse set of SS analogs, each optimized to elicit specific systemic physiological effects in prey. The increased metabolic stability, distinct SS receptor activation profiles, and chemical diversity of the venom analogs make them suitable leads for therapeutic application, including pain, cancer, and endocrine disorders. Our findings not only establish the existence of SS-like peptides in animal venoms but also serve as a model for the synergy gained from combining molecular phylogenetics and behavioral observations to optimize the discovery of natural products with biomedical potential.

Predators sometimes attract prey using the prey’s own pheromones, in a strategy known as aggressive mimicry. Abstract

The cone snails (family Conidae) are the best known and most intensively studied venomous marine gastropods. However, of the total biodiversity of venomous marine mollusks (superfamily Conoidea, >20,000 species), cone snails comprise a minor fraction. The venoms of the family Drilliidae, a highly diversified family in Conoidea, have not previously been investigated. In this report, we provide the first biochemical characterization of a component in a Drilliidae venom and define a gene superfamily of venom peptides. A bioactive peptide, cdg14a, was purified from the venom ofClavus davidgilmouriFedosov and Puillandre, 2020. The peptide is small (23 amino acids), disulfide-rich (4 cysteine residues) and belongs to the J-like drillipeptide gene superfamily. Other members of this superfamily share a conserved signal sequence and the same arrangement of cysteine residues in their predicted mature peptide sequences. The cdg14a peptide was chemically synthesized in its bioactive form. It elicited scratching and hyperactivity, followed by a paw-thumping phenotype in mice. Using the Constellation Pharmacology platform, the cdg14a drillipeptide was shown to cause increased excitability in a majority of non-peptidergic nociceptors, but did not affect other subclasses of dorsal root ganglion (DRG) neurons. This suggests that the cdg14a drillipeptide may be blocking a specific molecular isoform of potassium channels. The potency and selectivity of this biochemically characterized drillipeptide suggest that the venoms of the Drilliidae are a rich source of novel and selective ligands for ion channels and other important signaling molecules in the nervous system.

Abstract

Predatory gastropods of the superfamily Conoidea number over 12,000 living species. The evolutionary success of this lineage can be explained by the ability of conoideans to produce complex venoms for hunting, defense, and competitive interactions. Whereas venoms of cone snails (family Conidae) have become increasingly well studied, the venoms of most other conoidean lineages remain largely uncharacterized. In the present study, we present the venom gland transcriptomes of two species of the genus Clavus that belong to the family Drilliidae. Venom gland transcriptomes of two specimens of Clavus canalicularis and two specimens of Clavus davidgilmouri were analyzed, leading to the identification of a total of 1,176 putative venom peptide toxins (drillipeptides). Based on the combined evidence of secretion signal sequence identity, entire precursor similarity search (BLAST), and the orthology inference, putative Clavus toxins were assigned to 158 different gene families. The majority of identified transcripts comprise signal, pro-, mature peptide, and post-regions, with a typically short (<50 amino acids) and cysteine-rich mature peptide region. Thus, drillipeptides are structurally similar to conotoxins. However, convincing homology with known groups of Conus toxins was only detected for very few toxin families. Among these are Clavus counterparts of Conus venom insulins (drillinsulins), porins (drilliporins), and highly diversified lectins (drillilectins). The short size of most drillipeptides and structural similarity to conotoxins were unexpected, given that most related conoidean gastropod families (Terebridae and Turridae) possess longer mature peptide regions. Our findings indicate that, similar to conotoxins, drillipeptides may represent a valuable resource for future pharmacological exploration.

Abstract

Shipworms, assisted by intracellular γ-proteobacteria in their gills, are the principal degraders of wood in the sea. Shipworm symbionts have been cultivated in the laboratory. The genomes of these symbionts, in addition to being replete with lytic enzymes capable of degrading wood and/or enzymes of thioautotrophic metabolism, are among the bacterial genomes richest in secondary metabolite genes. These cultivated symbionts represent the dominant species in the gills in diverse shipworm species. We investigated how the isolate secondary metabolites might impact the host animals: which bacterial pathways are present, how widely distributed they are, and how they vary. Focusing on 14 wood-eating shipworm specimens, we found between one to three major bacterial species in each gill, with each species comprising a complex mixture of closely related strains. The mixture allows the shipworm host to access a much more complex metabolism than can be afforded by a single symbiont strain. We analyzed sequences from 22 shipworm gill metagenomes from seven shipworm species and compared them with the genomes of 23 cultivated bacterial isolates from shipworm gills. Limiting our analyses to well-characterized biosynthetic gene families, we found more than 400 polyketide, nonribosomal peptide, and related biosynthetic gene clusters, only a handful of which resemble known ones, comprising over 100 gene cluster families (GCFs). Only four GCFs are found in all specimens investigated. Several GCFs exhibited a host species-specific distribution, but most occurred stochastically. Shipworms and their symbionts thus provide a model system for understanding the role of secondary metabolism in symbioses. Significance Shipworms play critical roles in recycling wood in the sea and in shaping mangrove habitats. Symbiotic bacteria supply the enzymes that they need for nutrition and wood degradation. Here, we show that the same nutritional symbionts also have an immense capacity to produce a multitude of diverse and likely novel bioactive secondary metabolites. The compounds likely support the ability of shipworms to degrade wood in marine environments and include a compound under investigation for its therapeutic potential. Because many of the symbionts can be cultivated, they provide a model for understanding how secondary metabolism impacts microbial symbiosis in animals.

Abstract

Shipworms are a group of wood-boring and wood-feeding bivalves of extraordinary economic, ecological and historical importance. Known in the literature since the fourth century BC, shipworms are both destructive pests and critical providers of ecosystem services. All previously described shipworms are obligate wood-borers, completing all or part of their life cycle in wood and most are thought to use wood as a primary source of nutrition. Here, we report and describe a new anatomically and morphologically divergent species of shipworm that bores in carbonate limestone rather than in woody substrates and lacks adaptations associated with wood-boring and wood digestion. The species is highly unusual in that it bores by ingesting rock and is among the very few known freshwater rock-boring macrobioeroders. The calcareous burrow linings of this species resemble fossil borings normally associated with bivalve bioerosion of wood substrates (ichnospecies Teredolites longissimus) in marginal and fully marine settings. The occurrence of this newly recognized shipworm in a lithic substrate has implications for teredinid phylogeny and evolution, and interpreting palaeoenvironmental conditions based on fossil bioerosion features.

Abstract

Two novel cyclic hexapeptides were isolated from a Philippine ascidian, Lissoclinum bistatum. The oxazole-containing hexapeptides are given the name bistratamide A and B. Structural characterization of the peptides is presented. Intracerebral injection of bistratamide D in mice produced depressant effects on the central nervous system. A 65 microgram dose produced reduction of motor activity, sluggishness and sedation in the subjects.
