Abstract Brontispa longissima is a highly destructive pest that affects coconut and ornamental palm plants. It is widely distributed across Southeast and East Asia and the Pacific region, causing production losses of up to 50–70%. While control methods and ecological phenomena have been the primary focus of research, there is a significant lack of studies on the molecular mechanisms underlying these ecological phenomena. The absence of a reference genome has also hindered the development of new molecular-targeted control technologies. In this study, we conducted a karyotype analysis of B. longissima and assembled the first high-quality chromosome-level genome. The assembled genome is 582.24 Mb in size, with a scaffold N50 size of 63.81 Mb, consisting of 10 chromosomes and a GC content of 33.71%. The BUSCO assessment indicated a completeness estimate of 98.1%. A total of 23,051 protein-coding genes were predicted. Our study provides a valuable genomic resource for understanding the mechanisms of adaptive evolution and facilitates the development of new molecular-targeted control methods for B. longissima.

Abstract Sintering of active metal species often happens during catalytic reactions, which requires redispersion in a reactive atmosphere at elevated temperatures to recover the activity. Herein, we report a simple method to redisperse sintered Cu catalysts via O2-H2O treatment at room temperature. In-situ spectroscopic characterizations reveal that H2O induces the formation of hydroxylated Cu species in humid O2, pushing surface diffusion of Cu atoms at room temperature. Further, surface OH groups formed on most hydroxylable support surfaces such as γ-Al2O3, SiO2, and CeO2 in the humid atmosphere help to pull the mobile Cu species and enhance Cu redispersion. Both pushing and pulling effects of gaseous H2O promote the structural transformation of Cu aggregates into highly dispersed Cu species at room temperature, which exhibit enhanced activity in reverse water gas shift and preferential oxidation of carbon monoxide reactions. These findings highlight the important role of H2O in the dynamic structure evolution of supported metal nanocatalysts and lay the foundation for the regeneration of sintered catalysts under mild conditions.

Understanding the invasion of moso bamboo (Phyllostachys edulis) into adjacent evergreen broadleaf forest based on functional traits is crucial due to its significant influence on ecosystem processes. However, existing research has primarily focused on above- or below-ground traits in isolation, lacking a comprehensive integration of both. In this study, we conducted a trait-based analysis including 23 leaf traits and 11 root traits in three forest types - bamboo forest, mixed bamboo and broadleaf forest, and evergreen broadleaf forest - to investigate trait differences, phenotypic integration, and above- and below-ground resource strategies in bamboo and broadleaf species. Our findings demonstrated significant differences in leaf and root key traits between bamboo and broadleaf species, strongly supporting the “phenotypic divergence hypothesis”. Bamboo exhibited stronger trait correlations compared to broadleaf species, indicating higher phenotypic integration. Above- and below-ground strategies were characterized by trade-offs rather than coordination, resulting in a multi-dimensional trait syndrome. Specifically, a unidimensional leaf economics spectrum revealed that bamboo with higher leaf N concentrations (LNC), P concentrations (LPC), and specific leaf area (SLA) adopted a “fast acquisitive” above-ground strategy, while broadleaf species with thicker leaves employed a “slow conservative” above-ground strategy. A two-dimensional root trait syndrome indicated a “conservation” gradient with bamboo adopting a “slow conservative” below-ground strategy associated with higher root tissue density (RTD), and broadleaf species exhibiting a “fast acquisitive” below-ground strategy linked to higher root N concentrations (RNC) and P concentrations (RPC), and a “collaboration” gradient probably ranging from broadleaf species with a “do-it-yourself” strategy characterized by high specific root length (SRL), to bamboo adopting an “outsourcing” strategy with thicker roots. In conclusion, key trait divergence from coexisting broadleaf species, higher phenotypic integration, and multi-dimensional opposite above- and below-ground resource strategies confer competitive advantages to moso bamboo, shedding light on the mechanistic understanding of its invasion into subtropical evergreen broadleaf forest and providing theoretical guidance for maintaining the stability of subtropical forest ecosystem.

Slow and deep breathing (SDB) is a relaxation technique that can increase vagal activity. Respiratory sinus arrhythmia (RSA) serves as an index of vagal function usually quantified by the high-frequency power of heart rate variability (HRV). However, the low breathing rate during SDB results in deviations when estimating RSA by HRV. Besides, the impact of the inspiration-expiration (I: E) ratio and guidelines ways (fixed breathing rate or intelligent guidance) on SDB is not yet clear. In our study, 30 healthy people (mean age = 26.5 years, 17 females) participated in three SDB modes, including 6 breaths per minute (bpm) with an I:E ratio of 1:1/ 1:2, and intelligent guidance mode (I:E ratio of 1:2 with guiding to gradually lower breathing rate to 6 bpm). Parameters derived from HRV, multimodal coupling analysis (MMCA), Poincaré plot, and detrended fluctuation analysis were introduced to examine the effects of SDB exercises. Besides, multiple machine learning methods were applied to classify breathing patterns (spontaneous breathing vs. SDB) after feature selection by max-relevance and min-redundancy. All vagal-activity markers, especially MMCA-derived RSA, statistically increased during SDB. Among all SDB modes, breathing at 6 bpm with a 1:1 I:E ratio activated the vagal function the most statistically, while the intelligent guidance mode had more indicators that still significantly increased after training, including SDRR and MMCA-derived RSA, etc. About the classification of breathing patterns, the Naive Bayes classifier has the highest accuracy (92.2%) with input features including LFn, CPercent, pNN50, $\alpha 2$ , SDRatio, $\alpha 1$ , and LF. Our study proposed a system that can be applied to medical devices for automatic SDB identification and real-time feedback on the training effect. We demonstrated that breathing at 6 bpm with an I:E ratio of 1:1 performed best during the training phase, while intelligent guidance mode had a more long-lasting effect.

Abstract Alpha-synuclein (α-syn) aggregation and immune activation represent hallmark pathological events in Parkinson’s disease (PD). The PD-associated immune response encompasses both brain and peripheral immune cells, although little is known about the immune proteins relevant for such a response. We propose that the upregulation of CD163 observed in blood monocytes and in the responsive microglia in PD patients is a protective mechanism in the disease. To investigate this, we used the PD model based on intrastriatal injections of murine α-syn pre-formed fibrils in CD163 knockout (KO) mice and wild-type littermates. CD163KO females revealed an impaired and differential early immune response to α-syn pathology as revealed by immunohistochemical and transcriptomic analysis. After 6 months, CD163KO females showed an exacerbated immune response and α-syn pathology, which ultimately led to dopaminergic neurodegeneration of greater magnitude. These findings support a sex-dimorphic neuroprotective role for CD163 during α-syn-induced neurodegeneration.

Background: Gastric cancer is a highly prevalent malignant neoplasm. Metabolic reprogramming is intricately linked to both tumorigenesis and cancer immune evasion. The advent of single-cell RNA sequencing technology provides a novel perspective for evaluating cellular metabolism. This study aims to comprehensively investigate the metabolic pathways of various cell types in tumor and normal samples at high resolution and delve into the intricate regulatory mechanisms governing the metabolic activity of malignant cells in gastric cancer.Methods: Utilizing single-cell RNA sequencing data from gastric cancer, we constructed metabolic landscape maps for different cell types in tumor and normal samples. Employing unsupervised clustering, we categorized malignant cells in tumor samples into high and low metabolic subclusters and further explored the characteristics of these subclusters.Results: Our research findings indicate that epithelial cells in tumor samples exhibit significantly higher activity in most KEGG metabolic pathways compared to other cell types. Unsupervised clustering, based on the scores of metabolic pathways, classified malignant cells into high and low metabolic subclusters. In the high metabolic subcluster, it demonstrated the potential to induce a stronger immune response, correlating with a relatively favorable prognosis. In the low metabolic subcluster, a subset of cells resembling cancer stem cells (CSCs) was identified, and its prognosis was less favorable. Furthermore, a set of risk genes associated with this subcluster was discovered.Conclusion: This study reveals the intricate regulatory mechanisms governing the metabolic activity of malignant cells in gastric cancer, offering new perspectives for improving prognosis and treatment strategies.

Background: In recent years, research on exosomal miRNAs has provided new insights into exploring the mechanism of viral infection and disease prevention. This study aimed to investigate the serum exosomal miRNA expression profile of dengue-infected individuals through a community survey of dengue virus (DENV) infection. Methods: A seroprevalence study of 1253 healthy persons was first conducted to ascertain the DENV infection status in Baiyun District, Guangzhou. A total of 18 serum samples, including 6 healthy controls (HC), 6 asymptomatic DENV infections (AsymptDI), and 6 confirmed dengue fever patients (AcuteDI), were collected for exosome isolation and then sRNA sequencing. Through bioinformatics analysis, we discovered distinct serum exosomal miRNA profiles among the different groups and identified differentially expressed miRNAs (DEMs). These findings were further validated by qRT-PCR. Results: The community survey of DENV infection indicated that the DENV IgG antibody positivity rate among the population was 11.97 % in the study area, with asymptomatic infected individuals accounting for 93.06 % of the anti-DENV IgG positives. The age and Guangzhou household registration were associated with DENV IgG antibody positivity by logistic regression analysis. Distinct miRNA profiles were observed between healthy individuals and DENV infections. A total of 1854 miRNAs were identified in 18 serum exosome samples from the initial analysis of the sequencing data. Comparative analysis revealed 23 DEMs comprising 5 upregulated and 18 downregulated miRNAs in the DENV-infected group (mergedDI). In comparison to AcuteDI, 18 upregulated miRNAs were identified in AsymptDI. Moreover, functional enrichment of the predicted target genes of DEMs indicated that these miRNAs were involved in biological processes and pathways related to cell adhesion, focal adhesion, endocytosis, and ECM-receptor interaction. Eight DEMs were validated by qRT-PCR. Conclusion: The Baiyun District of Guangzhou exhibits a notable proportion of asymptomatic DENV infections as suggested in other research, highlighting the need for enhanced monitoring and screening of asymptomatic persons and the elderly. Differential miRNA expression among healthy, symptomatic and asymptomatic DENV-infected individuals suggests their potential as biomarkers for distinguishing DENV infection and offers new avenues of investigating the mechanisms underlying DENV asymptomatic infections.

Additive manufacturing (AM) has gained significant attention in automotive applications, while the low weldability of AM material poses challenges when assembling AM materials with conventionally manufactured materials. This study investigated the laser welding of AM AlSi10Mg and AA1050 sheet metal in an overlap configuration, specifically focusing on the impact of placement configuration of two materials on weld properties. This aims to provide valuable insights for optimizing the localized geometry of AM components to enhance the joint performance. Results showed that positioning AM AlSi10Mg on top leads to a higher weld penetration and higher Si content within the molten pool, especially in the upper sheet. Additionally, weld zone grain structure was greatly influenced by alternating the placement configuration, where welds with AM AlSi10Mg on top exhibiting more equiaxed grains in the weld centre and refined columnar grains near the fusion boundary of upper sheet. Furthermore, significantly reduced weld porosity was determined by positioning AM AlSi10Mg at the bottom (3.2 ± 0.4 %∼7.1 ± 0.9 %) compared to placing it on top (16.9 ± 0.9 %∼20.1 ± 0.9 %) in all scenarios ranging from full penetration to partial penetration. This can be related to the fact that the hydrogen bubbles originating from AM AlSi10Mg experienced a shorter exposure in the molten pool, limiting the merging and growth of small bubbles. However, a slightly lower joint strength was determined in welds with AM AlSi10Mg at the bottom which may be due to the plastic deformation incompatibility between large columnar grains in the fusion zone and the fine grains in the as-printed material.

Investigations and research on the giant African snail (GAS) mainly focus on Achatina fulica. However, in recent years, a more harmful GAS, Achatina immaculata, has been reported. In order to understand the invasive status of A. immaculata in China, we take Shenzhen, a coastal city in Southern China, as an example to carry out an ecological survey on the field populations of the two species of GAS in various districts. We explore the differences in the invasive characteristics of the two species of snails in terms of their dietary intake, cold adaptation and drought resistance. The results indicate that, based on the phylogenetic tree analysis, more than half of the sampled snails exhibit higher similarity to A. immaculata. The number of wild A. immaculata is significantly greater than that of A. fulica, and 70.64% of the 746 GAS are A. immaculata. At the same time, it is also found that the maximum shell length of A. immaculata is 135.83 mm, with an average shell length of 76.00 mm, which is significantly different from the average shell length of A. fulica (56.57 mm, p < 0.01). The food intake assay shows that there is no difference in the food preferences of the two species, but the food demand of A. immaculata is significantly greater than that of A. fulica (2.32 fold, p < 0.01). In the cold adaptation assay, A. immaculata recovers from the cold dormancy state significantly faster than A. fulica (1.92 fold, p < 0.05), and the speed with which A. immaculata enters the dormancy state in the drought environment is significantly slower than that of A. fulica (0.706 fold, p < 0.05). With the characteristics of a large body size, large food intake and strong resistance to cold and drought resistance, A. immaculata has the potential to be dominant in competition with A. fulica in the same ecological niche, and it has become the main invasive species of GAS in Shenzhen.

Drilling is one of the most commonly used techniques in hydrogeological exploration and is employed to obtain rock samples and create boreholes. During conventional drilling, it is necessary to raise all the drilling tools in the borehole when switching between coring drilling and non-coring drilling, which causes large auxiliary operation time consumption and poor drilling efficiency. Based on the structure of wireline coring tools, a large diameter modular drill bit was designed to switch between coring drilling and non-coring drilling without lifting the whole set of drilling tools. In the COMSOL simulation environment, a simulation model of the modular bit was constructed. Drilling fluid velocity and pressure characteristics flowing through the modular bit were studied. According to the analysis results, with the same input flow rate, similar velocities and lower pressure loss can be obtained in non-coring drilling as with the coring bit, and thus drilling cuttings can be removed effectively even if there are more cuttings produced in non-coring drilling than in coring drilling for a borehole drilled at the same diameter. When the outside diameter of the modular bit is 216 mm, the recommended clearance value is 9 mm or 10 mm in order to obtain lower pressure loss and larger diameter core. To generate low pressure loss and ensure bit strength, a layout with four nozzles on the internal non-coring bit is recommended. The modular bit enables fast switching between coring drilling and non-coring drilling without raising the drilling tools. The simulation model can be used for drilling parameter selection and drill bit optimization.

Abstract Background Low back pain (LBP) is one of the leading causes of disability worldwide. Differences in social backgrounds and lifestyles in various regions and countries may contribute to the discrepancies in the disease burden of LBP. Methods Based on the GBD 2019, we collected and analyzed numbers and age-standardized rates (ASR) of LBP disability-adjusted life years (DALYs). Temporal trends in ASR were also analyzed using estimated annual percentage change (EAPC). The Age-period-cohort (APC) model was used to estimate age, period and cohort trends in DALYs of LBP. An autoregressive integrated moving average (ARIMA) model was used to forecast DALYs of LBP trends from 2020 to 2035. Results The DALYs due to LBP increased from 1990 to 2019. The APC model showed that the risk of DALYs for global LBP increased with age and year and that the risk of DALYs was lower in the later-born cohort than in the earlier-born cohort. The main risk factors which GBD estimates were available for DALYs of LBP include smoking, occupational ergonomic factors and high BMI. It is expected that DALYs of LBP will continue to rise until 2035. Conclusion From 1990 to 2019, the global disease burden of LBP remained high. It is necessary to pay attention to the influence of social factors and lifestyle on LBP. Focusing on the impact of social factors as well as lifestyle on the prognosis of LBP and targeting interventions may further reduce the disease burden of LBP.

Abstract Background Relationships between adulthood modifiable risk factors and chronic kidney disease (CKD) are well-established, but associations with childhood risk factors are unclear. This study systematically assesses the published evidence about childhood modifiable risk factors and adulthood CKD. Methods We searched MEDLINE, EMBASE, and Web of Science to 6th May 2022. Articles were included if (1) they were population-based longitudinal studies, (2) exposures were potentially modifiable, for example through pharmacological or lifestyle modifications, including clinical conditions/measures (diabetes, blood pressure, adiposity, and dyslipidaemia); health behaviours (smoking, alcohol consumption, physical activity, fitness, and poor nutrition); and socio-economic factors (socio-economic position), and occurred during childhood (ages 2–19 years), and (3) outcome was CKD or surrogate markers of CKD in adulthood (ages 20 years or older). Three reviewers independently extracted the data. Results 15,232 articles were identified after deduplication; 17 articles met the inclusion criteria, reporting childhood blood pressure (n = 8), adiposity (n = 4), type 2 diabetes (n = 1), socio-economic position (n = 1), famine (n = 1), cardiorespiratory fitness (n = 1), and a healthy lifestyle score (n = 1). The results suggested positive associations of childhood adiposity, type 2 diabetes, and low socio-economic position and cardiorespiratory fitness in females with CKD in adulthood. Findings were inconsistent on associations between childhood BP and CKD in adulthood. Childhood healthy lifestyle score and exposure to famine were not associated with risk of CKD in adulthood. Conclusions The limited evidence suggests childhood factors may contribute to the CKD risk in adulthood, particularly adiposity, type 2 diabetes, and low socio-economic position and cardiorespiratory fitness in females. Further high-quality community-based studies are needed with long-term follow-up and investigation of a broader range of modifiable risk factors.

Objectives: To investigate the association between diabetes and cognitive dysfunction in the elderly population, and examine the impact of cognitive dysfunction on level of activities of daily living (ADL) in patients with diabetes. Methods: Data analysis was conducted on 2,951 individuals aged over 65 years from the Chinese Longitudinal Healthy Longevity Survey cohort. Propensity score matching was utilized to mitigate selection bias. Multivariate binary logistic regression was performed to analyse the association between diabetes and cognitive dysfunction in the study subjects. In addition, the relationship between ADL and cognitive function in patients with diabetes was analysed using the Wilcoxon rank-sum test. Results: A significant association (p = 0.017) was found between diabetes and the occurrence of cognitive dysfunction in older adults. Background and purposesTreating intracranial stenosis with distal thrombosis (IS&DT) using traditional mechanical thrombectomy (MT) techniques has proven challenging. This study aimed to summarize the experience of utilizing the balloon-assisted tracking (BAT) technique for IS&DT.MethodsDemographic and morphologic characteristics of patients with IS&DT were collected for this study. The BAT technique, involving a half-deflated balloon outside the intermediate catheter tip, was used in all patients to navigate through the proximal stenosis. Various parameters were recorded, including the sequence of vascular reperfusion, the puncture-to-reperfusion time (PRT), the residual stenosis rate, and the occurrence of re-occlusion. The thrombolysis in cerebral infarction (TICI) scale was used to assess the reperfusion of intracranial vessels, with a TICI score of ≥2b considered as successful perfusion. The clinical status of patients was evaluated at three time points: pre-procedure, post-procedure, and at discharge using the modified Rankin score (mRS).ResultsIn this study, a total of 10 patients were diagnosed with IS&DT, consisting of 9 male patients (90.0%) and 1 female patient (10.0%). The patients’ mean age was 63.10 years (ranging from 29 to 79 years). The mean National Institute of Health Stroke Scale (NIHSS) score before treatment was 24.3 (ranging from 12 to 40), indicating the severity of their condition. Following the procedure, all patients achieved successful reperfusion with a thrombolysis in cerebral infarction (TICI) score of ≥2b. The average puncture-to-reperfusion time (PRT) was 51.8 min (ranging from 25 to 100 min), indicating the time taken for the procedure. During the perioperative period, three patients (30.0%) experienced complications. One patient had hemorrhage, while two patients had contrast extravasation. Among these cases, only the patient with hemorrhage (10%) suffered from a permanent neurological function deficit. At discharge, the patient’s condition showed improvement. The mean NIHSS score decreased to 13.2 (ranging from 1 to 34), indicating a positive response to treatment. The mean mRS score at discharge was 3.2 (ranging from 1 to 5), showing some level of functional improvement.ConclusionIn conclusion, the use of the balloon-assisted tracking (BAT) technique for treating intracranial stenosis with distal thrombosis (IS&DT) showed promising results. However, a moderate rate of perioperative complications was observed, warranting further investigation and refinement of the procedure.

Reverse circulation impact drilling has the advantages of high drilling efficiency and less dust, which can effectively form holes in hard rock and gravel layer. As integral reverse circulation drill bits used in the conventional down-the-hole (DTH) hammers are only suitable for specific formations, the whole set of DTH hammer needs to be replaced when drilling different formations. In this paper, several types of split drill bits for different drilling technologies are designed. The flow field characteristics of one of the split drill bits is analyzed based on the computational fluid dynamics (CFD) method, with four technic parameters considered, which are input flow rate, number of inlet holes, angle of injection exhaust holes, and diameter of injection exhaust holes, respectively. Three parameters are selected as indicators to evaluate the rationality and performance of the split drill bit, which are injection exhaust hole outlet mass flow rate, ratio of the mass flow rate out of injection exhaust holes to the whole inlet mass flow rate, and maximum pressure at the upper end of the split drill bit. According to the CFD analysis results, the above four technic parameters influence the flow rate and pressure in different rules. Considering the injection capacity, pressure loss, and bit strength, inlet holes of 10, injection exhaust holes with an angle of 50°, and injection exhaust holes with a diameter of 12 mm are recommended to obtain ideal reverse circulation. Different types of split drill bits were manufactured, and drilling experiments were carried out in unconsolidated formations. The maximum drilling rate can reach 1.5 m/min in the drilling experiments. The split drill bit proposed in this paper exhibits excellent adaptability for reverse circulation drilling in loose formations.

Postmenopausal osteoporosis is the most common type of osteoporosis in women. To date, little is known about their transcriptome signatures, although biomarkers from peripheral blood mononuclear cells are attractive for postmenopausal osteoporosis diagnoses. Here, we performed bulk RNA sequencing of 206 samples (124 postmenopausal osteoporosis and 82 normal samples) and described the clinical phenotypic characteristics of postmenopausal women. We then highlighted the gene set enrichment analyses between the extreme T-score group and the heathy control group, revealing that some immune-inflammatory responses were enhanced in postmenopausal osteoporosis, with representative pathways including the mitogen-activated protein kinase (NES = 1.6, FDR

Chikungunya virus (CHIKV) and Dengue virus (DENV) are vector-borne diseases transmitted by Aedes aegypti and Aedes albopictus that pose a significant threat to global public health. Cases of acute Chikungunya fever often present similar clinical symptoms to other vector-borne diseases, such as Dengue fever. In regions where multiple vector-borne diseases coexist, CHIKV is often overlooked or misdiagnosed as Dengue virus, West Nile virus, Zika virus or other viral infections, which delays its prevention and control. However, IgM antibodies directed against the E2 protein of CHIKV have not yet been generalized to clinical settings due to the low sensitivity and high cost in commercial kits. Indirect ELISA with peptides provides an effective supplementary tool for detecting CHIKV IgM antibodies. Our study aims at examining the potential of linear epitopes on the E2 glycoprotein that specifically bind to IgM antibodies as serodiagnostic tool for CHIKV. The sensitivity of the established peptide indirect ELISA method for detecting clinical samples is significantly better than that of commercial kits, realizing a beneficial supplement to the existing IgM antibody assay. It also established the groundwork for comprehending the biological mechanisms of the CHIKV E2 protein and the advancement of innovative epitope peptide vaccines.

Creep is a long-term property of concrete that poses a significant challenge to structural engineering. The research content of this paper concerns how to accurately predict the creep behavior of concrete. The overall purpose is to develop a robust predictive model based on the Northwestern University (NU) database that overcomes the challenges encountered when handling tabular creep data, including the lack of spatial correlations between features and the presence of noise. To achieve this goal, a novel deep learning methodology that leverages convolutional neural networks (CNNs) is introduced. First, the data preprocessing phase involves feature selection and K-nearest neighbor-based data filling to enhance the quality of the given data. A soft-sorted 1D CNN is introduced to transform tabular data into multichannel images, thereby taking advantage of spatial locality and feature sorting. Additionally, this model incorporates noise reduction mechanisms through residual network encoders, achieving enhanced data clarity and model accuracy. The mean absolute error (MAE) and root mean square deviation (RMSE) of the model increase from 7.48 and 10.76 to 5.83 and 8.88, respectively. The major findings of this research indicate a significant predictive accuracy improvement over the existing methods. The model effectively captures the complex relationships between concrete creep and a range of influencing factors, enabling precise and quantitative predictions to be obtained. These findings offer deep insights for the field of civil engineering, enabling enhanced structural design, safety, and performance prediction in concrete applications. This paper contributes to the ongoing exploration of advanced deep learning techniques for addressing complex challenges in the domains of concrete mechanics and structural engineering.

Its distinct phenotypic traits, especially its polyphagy and ability to migrate long distances, contribute to its widening distribution and increasing difficulty of control. However, knowledge about these traits is still limited. Results We generated a high-quality chromosome-level assembly of A. ipsilon using PacBio and Hi-C technology with a contig N50 length of ~ 6.7 Mb. Comparative genomic and transcriptomic analyses showed that detoxification-associated gene families were highly expanded and induced after insects fed on specific host plants. Knockout of genes that encoded two induced ABC transporters using CRISPR/Cas9 significantly reduced larval growth rate, consistent with their contribution to host adaptation. A comparative transcriptomic analysis between tethered-flight moths and migrating moths showed expression changes in the circadian rhythm gene AiCry2 involved in sensing photoperiod variations and may receipt magnetic fields accompanied by MagR and in genes that regulate the juvenile hormone pathway and energy metabolism, all involved in migration processes. Conclusions This study provides valuable genomic resources for elucidating the mechanisms involved in moth migration and developing innovative control strategies.

Abstract Background G-quadruplexes (G4s) are unique noncanonical nucleic acid secondary structures, which have been proposed to physically interact with transcription factors and chromatin remodelers to regulate cell type-specific transcriptome and shape chromatin landscapes. Results Based on the direct interaction between G4 and natural porphyrins, we establish genome-wide approaches to profile where the iron-liganded porphyrin hemin can bind in the chromatin. Hemin promotes genome-wide G4 formation, impairs transcription initiation, and alters chromatin landscapes, including decreased H3K27ac and H3K4me3 modifications at promoters. Interestingly, G4 status is not involved in the canonical hemin-BACH1-NRF2-mediated enhancer activation process, highlighting an unprecedented G4-dependent mechanism for metabolic regulation of transcription. Furthermore, hemin treatment induces specific gene expression profiles in hepatocytes, underscoring the in vivo potential for metabolic control of gene transcription by porphyrins. Conclusions These studies demonstrate that G4 functions as a sensor for natural porphyrin metabolites in cells, revealing a G4-dependent mechanism for metabolic regulation of gene transcription and chromatin landscapes, which will deepen our knowledge of G4 biology and the contribution of cellular metabolites to gene regulation.

This paper introduced the use of an adjustable-ring-mode (ARM) laser beam to optimise the microstructure and mechanical performance of laser welded 6xxx high-strength aluminium alloy joints. A remote laser welding system equipped with transverse beam oscillation was employed and welding parameters, including the beam oscillation width and core/ring power ratio of the ARM laser beam, were investigated sequentially. Results showed that the core/ring power ratio at a constant total power has a limited control on the weld geometry when integrated with beam oscillation. Increasing the beam oscillation width, up to 2.5 mm, resulted in a wider weld interface and a threshold value of 3.1 mm was determined to activate the transition from interface failure to the fusion boundary failure subjected to tensile lap shear loading, leading to a significant improvement in the joint strength from 170 N/mm to 277 N/mm. Furthermore, results revealed that proper selection of core/ring power ratio at a constant total power can restrict the formation of columnar grains near the interface between two plates, resulting in grain refinement within the weld zone. Both extremely high power ratio, for example core alone mode (infinite power ratio) and extremely low power ratio, e.g. ring alone mode (zero power ratio), lead to a low thermal gradient and cooling rate at the solidification front. A compromised solution which balances the wider formation of equiaxed dendrites and better grain refinement was determined at the power ratio of 0.33, translating to the improvement in joint strength up to 400 N/mm.

Abstract Background Membrane nephropathy (MN) often presents as nephrotic syndrome with characteristic lipid metabolism that could not be explained by lipid indicators commonly used in clinical practice. Studies have shown that invigorating spleen and qi, activating blood and detoxication in the treatment of MN is an effective method proved by randomized controlled clinical trial. However, the alterations of lipid profile before and after traditional Chinese medicine (TCM) treatment and the related lipid markers that affect the therapeutic effect have not been fully clarified. Methods We analyzed plasma lipid profiles of 92 patients with MN before and after TCM treatment by high-coverage targeted lipidomics. Results 675 lipids were identified, of which 368 stably expressed lipids (coefficient of variation less than 30% and deletion value less than 10%) were eventually included for statistical analysis. 105 lipids were altered mainly including spingolipids, glycerides, glycerophosholipid, fatty acyl and steroids, among which, the abundance of ceramides (Cers), sphingomyelins (SMs), diacylglycerols (DGs), phosphatidylcholines (PCs) were lower than those before treatment with statistically significant difference. The WGCNA network to analyze the correlation between the collective effect and the therapeutic effect showed that the triglyceride (TG) molecules were most relevant to the therapeutic effect. Analysis of 162 triglyceride molecules showed that 11 TGs were significantly down-regulated in the effective group which were concentrated in carbon atom number of 52–56 and double bond number of 0–4. TGs molecules including TG56:2-FA20:0, TG56:2-FA20:1, TG56:3-FA20:0 and TG56:5-FA20:2 were most closely related to the therapeutic effect of TCM after adjusting the influence of clinical factors. ROC curve analysis showed that these four lipids could further improve the predictive efficacy of treatment based on clinical indicators. Conclusion Our work demonstrated that the therapeutic effect of invigorating spleen and qi, activating blood and detoxication in the treatment of MN may be exerted by regulating lipid metabolism. High-coverage targeted lipidomics provided a non-invasive tool for discovery of lipid markers to improve the predictive efficacy of TCM therapy.

BackgroundThe global cancer burden is substantial and spiraling. Although rehabilitation specialists could offer assistance, oncologic rehabilitation is still underutilized and not a routine part of clinical oncology guidelines worldwide. Global investigations of disease prevalence and years lived with disability (YLDs) for tracheal, bronchus, and lung (TBL) cancer are valuable for facilitating clinical practice improvement and health resource management. The objective of this study is to report the global estimates of rehabilitation needs and disease burden of TBL cancers from 1990 to 2019 and provide predictions for 2045.MethodsTo estimate the need for rehabilitation, the data used from the Global Burden of Disease Study 2019 to calculate the prevalence, YLDs, and the attributable risk factors of TBL cancer. The Bayesian age-period-cohort model and Auto-Regressive Integrated Moving Average model were established to forecast the future health burden. All analyses were done at the global level and then some in the aggregation with the seven World Bank regions. All the data were analyzed by R software (x64 version 4.2.1) and Microsoft Excel (version 2019).ResultsGlobally in 2019, 3,212,307 cases of TBL cancer (95% UI 2,937,037-3,488,346) could have benefitted from rehabilitation, contributing to 544,215 (95% UI 396,134-700,099) YLDs. Over the past 30 years, the age-standardized rate (ASR) of prevalence (EAPC = 0.51) and YLDs (EAPC = 0.03) increased. Throughout this period, the global prevalence and YLDs counts were greater in males than females. The ASR of prevalence and YLDs are projected to show a slight downward trend by 2045 on the global scale, the overall prevalence and YLDs due to TBL cancer are likely to increase further, but all indicators show a growing trend in females.ConclusionTBL cancer remains one of the major public health issues globally. According to the forecasted results, the burden of YLDs due to TBL cancer will continue to rise, and the increment is higher in females than males. A rising number of patients worldwide will benefit from rehabilitation services in the future to achieve precise control and management throughout the TBL cancer patient lifecycle.

Invasive plant species pose significant biodiversity and ecosystem threats. Real-time identification of invasive plants is a crucial prerequisite for early and timely prevention. While deep learning has shown promising results in plant recognition, the use of deep learning models often involve a large number of parameters and high data requirements for training. Unfortunately, the available data for various invasive plant species are often limited. To address this challenge, this study proposes a lightweight deep learning model called IPMCNet for the identification of multiple invasive plant species. IPMCNet attains high recognition accuracy even with limited data and exhibits strong generalizability. Simultaneously, by employing depth-wise separable convolutional kernels, splitting channels, and eliminating fully connected layer, the model’s parameter count is lower than that of some existing lightweight models. Additionally, the study explores the impact of different loss functions, and the insertion of various attention modules on the model’s accuracy. The experimental results reveal that, compared with eight other existing neural network models, IPMCNet achieves the highest classification accuracy of 94.52%. Furthermore, the findings suggest that focal loss is the most effective loss function. The performance of the six attention modules is suboptimal, and their insertion leads to a decrease in model accuracy.

The J-TEXT capability is enhanced compared to two years ago with several upgrades of its diagnostics and the increase of electron cyclotron resonance heating (ECRH) power to 1 MW. With the application of electron cyclotron wave (ECW), the ECW assisted plasma startup is achieved; the tearing mode is suppressed; the toroidal injection of 300 kW ECW drives around 24 kA current; fast electrons are generated with toroidal injected ECW and the runaway current conversion efficiency increases with ECRH power. The mode coupling between 2/1 and 3/1 modes are extensively studied. The coupled 2/1 and 3/1 modes usually lead to major disruption. Their coupling can be either suppressed or avoided by external resonant magnetic perturbation fields and hence avoids the major disruption. It is also found that the 2/1 threshold of external field is significantly reduced by a pre-excited 3/1 mode, which can be either a locked island or an external kink mode. The disruption control is studied by developing prediction methods capable of cross tokamak application and by new mitigation methods, such as the biased electrode or electromagnetic pellet injector. The high-density operation and related disruptions are studied from various aspects. Approaching the density limit, the collapse of the edge shear layer is observed and such collapse can be prevented by applying edge biasing, leading to an increased density limit. The density limit is also observed to increase, if the plasma is operated in the poloidal divertor configuration or the plasma purity is increased by increasing the pre-filled gas pressure or ECRH power during the start-up phase.

A fast frequency hopping scheme based on SDR was proposed to solve the problem that the existing fast frequency hopping communication system relied on satellite timing.Firstly, a true random sequence was used to perform frequency hopping operation on the modulated signal in this scheme.And the frequency hopping table was obtained by channel down-conversion, low-pass filtering and amplitude peak detection operations.Secondly, the receiver searched for the precise starting point by the peak-to-peak distribution characteristics of the quadrature demodulation amplitude of the baseband signal after de-hopping.Finally, down-conversion was performed and the peak-to-peak amplitude of the quadrature demodulation was checked for exceeding the threshold value to achieve de-hopping for frequency hopping signals.The effectiveness and adaptability of the proposed scheme is verified by using two general-purpose SDR devices with a hopping rate of four hops per symbol period in a real channel with other continuous signals.

Abstract Aiming at solving the effective data delivery and energy hole problem in multi-hop cognitive radio sensor networks (CRSNs), a weighted energy consumption minimization-based uneven clustering (ECMUC) routing protocol is proposed in this paper. For the first time, the impact of control overhead on the network performance is taken into consideration, to be specific, the energy consumption of control overhead is integrated with that of data communication to model the network energy consumption. Through effective transformation and theoretical analysis, cluster radius of each ring is derived by minimizing the network energy consumption and balancing the residual energy among nodes in different rings. Distributed cluster heads (CHs) selection and cluster formation are carried out within this range to control the cluster size and the corresponding energy cost. Expected times for being CHs metric is defined to measure nodes’ energy and spectral potential and help select powerful CHs. Simulation results show that ECMUC protocol is superior to most clustering protocols designed for CRSNs in terms of network surveillance capability and network lifetime, and it is also demonstrated that taking control overhead into consideration is beneficial for improving the network performance.

PurposeElongation denotes the regularity of an aneurysm and parent artery. This retrospective research study was conducted to identify the morphological factors that could predict postoperative in-stent stenosis (ISS) after Pipeline Embolization Device (PED) implantation for unruptured intracranial aneurysms (UIAs).MethodsPatients with UIA and treated with PED at our institute between 2015 and 2020 were selected. Preoperative morphological features including both manually measured shape features and radiomics shape features were extracted and compared between patients with and without ISS. Logistic regression analysis was performed for factors associated with postoperative ISS.ResultsA total of 52 patients (18 men and 34 women) were involved in this study. The mean angiographic follow-up time was 11.87 ± 8.26 months. Of the patients, 20 of them (38.46%) were identified with ISS. Multivariate logistic analysis showed that elongation (odds ratio = 0.008; 95% confidence interval, 0.001–0.255; p = 0.006) was an independent risk factor for ISS. The area under the curve (AUC) of the receiver operating characteristic curve(ROC) was 0.734 and the optimal cut-off value of elongation for ISS classification was 0.595. The sensitivity and specificity of prediction were 0.6 and 0.781, respectively. The ISS degree of elongation of less than 0.595 was larger than the ISS degree of elongation of more than 0.595.ConclusionElongation is a potential risk factor associated with ISS after PED implantation for UIAs. The more regular an aneurysm and parent artery, the less likelihood of an ISS occurrence.

Abstract Background The conversion of astrocytes activated by nerve injuries to oligodendrocytes is not only beneficial to axonal remyelination, but also helpful for reversal of glial scar. Recent studies have shown that pathological niche promoted the Sox10-mediated astrocytic transdifferentiation to oligodendrocytes. The extracellular factors underlying the cell fate switching are not known. Methods Astrocytes were obtained from mouse spinal cord dissociation culture and purified by differential adherent properties. The lineage conversion of astrocytes into oligodendrocyte lineage cells was carried out by Sox10-expressing virus infection both in vitro and in vivo, meanwhile, epidermal growth factor (EGF) and epidermal growth factor receptor (EGFR) inhibitor Gefitinib were adopted to investigate the function of EGF signaling in this fate transition process. Pharmacological inhibition analyses were performed to examine the pathway connecting the EGF with the expression of oligodendrogenic genes and cell fate transdifferentiation. Results EGF treatment facilitated the Sox10-induced transformation of astrocytes to O4+ induced oligodendrocyte precursor cells (iOPCs) in vitro. The transdifferentiation of astrocytes to iOPCs went through two distinct but interconnected processes: (1) dedifferentiation of astrocytes to astrocyte precursor cells (APCs); (2) transformation of APCs to iOPCs, EGF signaling was involved in both processes. And EGF triggered astrocytes to express oligodendrogenic genes Olig1 and Olig2 by activating extracellular signal-regulated kinase 1 and 2 (Erk1/2) pathway. In addition, we discovered that EGF can enhance astrocyte transdifferentiation in injured spinal cord tissues. Conclusions These findings provide strong evidence that EGF facilitates the transdifferentiation of astrocytes to oligodendrocytes, and suggest that targeting the EGF-EGFR-Erk1/2 signaling axis may represent a novel therapeutic strategy for myelin repair in injured central nervous system (CNS) tissues.

Abstract Background Chikungunya virus (CHIKV) is a re-emerged mosquito-borne alphavirus that can cause musculoskeletal diseases, imposing a substantial threat to public health globally. High-affinity antibodies are need for diagnosis and treatment of CHIKV infections. As a potential diagnostic and therapeutic agent, the multivalent VHH antibodies is a promising tookit in nanomedicine. Here, we developed potent multivalent VHH antibodies from an alpaca naïve phage display library targeting the E2 glycoprotein of the CHIKV virus. Results In the present study, we generated 20 VHH antibodies using a naïve phage display library for binders to the CHIKV E2 glycoprotein. Of these, multivalent VHH antibodies Nb-2E8 and Nb-3C5 had specific high-affinity binding to E2 protein within the nanomolar range. The equilibrium dissociation constant (KD) was between 2.59–20.7 nM, which was 100-fold stronger than the monovalent antibodies’ affinity. Moreover, epitope mapping showed that Nb-2E8 and Nb-3C5 recognized different linear epitopes located on the E2 glycoprotein domain C and A, respectively. A facile protocol of sandwich ELISA was established using BiNb-2E8 as a capture antibody and HRP-conjugated BiNb-3C5 as a detection antibody. With advantages of high efficiency and low cost, DTH hammer drilling has been highly applied in various drilling projects. When drilling in unconsolidated formations, it is prone to drilling accidents such as drilling tools sticking or burying. Thus, a bidirectional pneumatic DTH hammer is designed to drill boreholes using forward impact and release sticking drilling tools using backward impact. With a floating gas distribution mechanism, impact strokes of the DTH hammer piston can be changed when flat keys are in a different position of the key grooves on the gas distribution shaft. In drilling mode, the piston has a larger impact stroke and can impact the anvil at high speeds to drive the bit breaking rocks. When drilling tools become stuck, by changing to a smaller impact stroke, the piston can impact backward on the gas distribution valve to break rocks above the DTH hammer so sticking drilling tools can be released. According to the structure and working principle of the bidirectional pneumatic DTH hammer, a physical model based on the pneumatic transmission circuit is established; then, a simulation model is built with pneumatic transmission module components in software of SimulationX 4.1 student version. Piston velocities, displacements, and impact energy are analyzed, with main factors including piston mass, total weight of the DTH hammer, compressed air pressure, and backward impact stroke being considered. Analysis results show that working characteristics of the DTH hammer are fairly affected by piston mass and compressed air pressure. Based on the changing laws of the impact frequency, peak of impact velocity, and impact energy, a piston mass of 18 kg, total weight of 125 kg, gas source pressure of 2.2 MPa, and lifting distance of 60 mm for backward impact were recommended. To verify the performance of the bidirectional pneumatic DTH hammer, field experiments were carried out in the gravel stratums. The bidirectional DTH hammer was in good working condition and the maximum drilling rate can reach up to 1.5 m/min. By lifting the DTH hammer away from the bottom of the borehole and pumping compressed air, the DTH hammer piston could achieve a high frequency backward impact. There are no drilling tools’ sticking or burying accidents in the drilling experiments. The bidirectional pneumatic DTH hammer can effectively drill boreholes in loose formations and deal with drilling tools’ sticking or burying accidents.

Abstract Background The relationships of healthy lifestyle scores (HLS) of various kinds in adulthood with the risk of chronic kidney disease (CKD) have been reported, but little is known about the association of childhood lifestyle with later life CKD. This study examined the relationship of HLS from childhood to adulthood with subclinical kidney damage (SKD) in midlife, a surrogate measure for CKD. Methods Data were collected in an Australian population-based cohort study with 33 years follow-up. 750 participants with lifestyle information collected in childhood (ages 10–15 years) and midlife (ages 40–50 years), and measures of kidney function in midlife were included. The HLS was generated from the sum scores of five lifestyle factors (body mass index, smoking, alcohol consumption, physical activity, and diet). Each factor was scored as poor (0 point), intermediate (1 point), or ideal (2 points). Log-binomial regression was used to investigate the relationship of HLS in childhood and from childhood to adulthood with SKD defined as either 1) estimated glomerular filtration rate (eGFR) 30–60 mL/min/1.73m2 or 2) eGFR> 60 mL/min/1.73m2 with urine albumin-creatinine ratio ≥ 2.5 mg/mmol (males) or 3.5 mg/mmol (females), adjusting for socio-demographic factors and the duration of follow-up. Results The average HLS was 6.6 in childhood and 6.5 in midlife, and the prevalence of SKD was 4.9% (n = 36). Neither HLS in childhood nor HLS from childhood to adulthood were significantly associated with the risk of SKD in midlife. Conclusions A HLS from childhood to adulthood did not predict SKD in this middle-aged, population-based Australian cohort.

ObjectiveThis study aimed to investigate the effects of explicit and implicit online intervention methods for self-compassion on improving sleep quality among Chinese adults.MethodsA total of 328 adult participants were recruited to complete the Pittsburgh Sleep Quality Questionnaire and Self-compassion Scale, and 168 participants were randomly assigned to one of three different conditions: two self-compassion intervention groups (self-compassion writing intervention asked participants to write several sentences with self-compassion, and self-compassion combination intervention asked participants to combine words into complete sentences with self-compassion) and one control group. After 1 week of online self-compassion intervention with daily sleep quality measured simultaneously, 150 participants completed the posttest of the Pittsburgh Sleep Quality Questionnaire and Self-Compassion Scale.ResultsThe pretest and posttests results showed that the self-compassion level and sleep quality of the self-compassion writing intervention group were significantly better than those of the control group. However, there was no significant difference between the self-compassion combination intervention group and the control group. For the diary tracking results, sleep quality was significantly better for both the self-compassion writing intervention group and self-compassion combination intervention group than the control group, however, the self-compassion writing intervention group showed great improvements.ConclusionBoth self-compassion writing and combination interventions were effective in improving sleep quality, and the effect of self-compassion writing was more stable.

Objective. This evidence mapping is aimed at identifying, summarizing, and analyzing the available evidence on cognitive behavioral therapy (CBT) for neuropathic pain (NP). Methods. This study was conducted following the methodology of Global Evidence Mapping (GEM). Searches were conducted in PubMed, Embase, the Cochrane Library, and PsycINFO to identify systematic reviews (SRs) with or without meta-analysis published before February 15, 2022. The authors independently assessed eligibility, extracted data, and evaluated the methodological quality of the included SRs using AMSTAR-2. The results were presented in the tables and a bubble plot based on the identified population-intervention-comparison-outcome (PICO) questions. Results. A total of 34 SRs met the eligibility criteria. According to the AMSTAR-2, 2 SRs were rated “high,” 2 SRs were rated “moderate,” 6 SRs were rated “low,” and 24 SRs were rated “critically low.” The most common study design utilized to evaluate the efficacy of CBT for NP was the randomized controlled trial. In total, 24 PICOs were identified. Migraine was the most studied population. CBT for NP usually reaches the “potentially better” result at follow-up. Conclusions. Evidence mapping is a useful way to present existing evidence. Currently, the existing evidence on CBT for NP is limited. Overall, the methodological quality of the included SRs was low. Further improvements in the methodological quality of SRs and more research on the most efficient CBT formats for NP are recommended in the future.

Background and Object. There is a growing body of evidence highlighting the significant role of gut microbiota in various neurological and psychiatric disorders. We performed an evidence mapping to review the association between different microbiota and these disorders and assessed the strength of evidence for these associations. Methods. We searched PubMed, Cochrane Library, and Epistemonikos to identify systematic reviews and meta-analysis (SRs). We searched for neurological diseases and psychiatric disorders, including Alzheimer’s disease (AD), attention deficit hyperactivity disorder (ADHD), amyotrophic lateral sclerosis (ALS), autism spectrum disorder (ASD), anorexia nervosa (AN), bipolar disorder (BD), eating disorder (ED), generalized anxiety disorder (GAD), major depressive disorder (MDD), multiple sclerosis (MS), obsessive compulsive disorder (OCD), Parkinson’s disease (PD), posttraumatic stress disorder (PTSD), spinal cord injury (SCI), schizophrenia, and stroke. We used A Measurement Tool to Assess Systematic Reviews (AMSTAR-2) to evaluate the quality of included SRs. We also created an evidence map showing the role of gut microbiota in neurological diseases and the certainty of the evidence. Results. In total, 42 studies were included in this evidence mapping. Most findings were obtained from observational studies. According to the AMSTAR-2 assessment, 21 SRs scored “critically low” in terms of methodological quality, 16 SR scored “low,” and 5 SR scored “moderate.” A total of 15 diseases have been investigated for the potential association between gut microbiome alpha diversity and disease, with the Shannon index and Simpson index being the most widely studied. A total of 12 diseases were investigated for potential link between beta diversity and disease. At the phylum level, Firmicutes, Bacteroidetes, Actinobacteria, Proteobacteria, and Verrucomicrobia were more researched. At the genus level, Prevotella, Coprococcus, Parabacteroides, Phascolarctobacterium, Escherichia Shigella, Alistipes, Sutteralla, Veillonella, Odoribacter, Faecalibacterium, Bacteroides, Bifidobacterium, Dialister, and Blautia were more researched. Some diseases have been found to have specific flora changes, and some diseases have been found to have common intestinal microbiological changes. Conclusion. We found varied levels of evidence for the associations between gut microbiota and neurological diseases; some gut microbiota increased the risk of neurological diseases, whereas others showed evidence of benefit that gut microbiota might be promising therapeutic targets for such diseases.

Abstract As a unique group of advanced artificial materials, metamaterials have found many interesting applications in various devices due to the ability to control electromagnetic fields. The metamaterial provides the prospect of new opportunities for the wireless power transfer (WPT) system to improve efficiency and reduce magnetic flux density. This review paper evaluates the performance of the WPT system with metamaterial from both principles and functions aspects. The objectives of this review include: (1) to present the research process and bottlenecks of metamaterial in the WPT system; (2) to clarify the design solution and checking method of the metamaterial and (3) to identify the challenges and opportunities based on the functional of the WPT system integration with metamaterial. Therefore, the process on metamaterial design, working principle of metamaterial, optimal method, equivalent model, and experimental method are reviewed. The recent efforts of the WPT system with metamaterial in efficiency enhancement, misalignments, magnetic field reduction are illustrated. Finally, the challenges and prospects of metamaterial based on the WPT system are concluded.

Transposable elements (TEs) and transcription factors (TFs) are involved in the precise regulation of gene expression during the preimplantation stage. Activation of TEs is a key event for mammalian embryonic genome activation and preimplantation early embryonic development. TFs are involved in the regulation of drastic changes in gene expression patterns, but an inventory of the interplay between TEs and TFs during normal/abnormal human embryonic development is still lacking. Here we used single-cell RNA sequencing data generated from biparental and uniparental embryos to perform an integrative analysis of TE and TF expression. Our results showed that endogenous retroviruses (ERVs) are mainly expressed during the minor embryonic genome activation (EGA) process of early embryos, while Alu is gradually expressed in the middle and later stages. Some important ERVs (e.g., LTR5_Hs, MLT2A1) and Alu TEs are expressed at significantly lower levels in androgenic embryos. Integrative analysis revealed that the expression of the transcription factors CTCF and POU5F1 is correlated with the differential expression of ERV TEs. Comparative coexpression network analysis further showed distinct expression levels of important TFs (e.g., LEUTX and ZSCAN5A) in dizygotic embryos vs. parthenogenetic and androgenic embryos. This systematic investigation of TE and TF expression in human early embryonic development by single-cell RNA sequencing provides valuable insights into mammalian embryonic development.

The invasion of Spartina alterniflora (S. alterniflora) has resulted in significant losses in the diversity of coastal ecosystems. However, the impact of seasonal changes on the accurate identification of S. alterniflora remains to be explored, which is of great significance due to its early monitoring and warning. In this study, S. alterniflora in Beihai, Guangxi, was selected as the research object. Unmanned aerial vehicles (UAVs) and deep convolutional neural networks (CNNs) were used to explore the identification of S. alterniflora in different seasons. Through comparative analysis, the ResNet50 model performed well in identifying S. alterniflora, with an F1-score of 96.40%. The phenological characteristics of S. alterniflora differ in different seasons. It is difficult to accurately monitor the annual S. alterniflora using only single-season data. For the monitoring of S. alterniflora throughout the year, the autumn-winter two-season model was selected from the perspective of time cost, the four-season model was selected from the perspective of identification performance, and the three-season model of summer, autumn and winter was selected from the perspective of time cost and identification performance. In addition, a method was developed to predict and evaluate the diffusion trend of S. alterniflora based on time series UAV images. Using the spring data to predict the diffusion trend of S. alterniflora in summer and autumn, the results showed that the highest recall reached 84.28%, the F1-score was higher than 70%, and most of the diffusion of S. alterniflora was correctly predicted. This study demonstrates the feasibility of distinguishing S. alterniflora from native plants in different seasons based on UAV and CNN recognition algorithms. The proposed diffusion early warning method reflects the actual diffusion of S. alterniflora to a certain extent, which is of great significance for the early management of invasive plants in coastal wetlands.

In this study, we introduce a novel denoising transformer-based neural network (DTNN) model for predicting the remaining useful life (RUL) of lithium-ion batteries. The proposed DTNN model significantly outperforms traditional machine learning models and other deep learning architectures in terms of accuracy and reliability. Specifically, the DTNN achieved an R2 value of 0.991, a mean absolute percentage error (MAPE) of 0.632%, and an absolute RUL error of 3.2, which are superior to other models such as Random Forest (RF), Decision Trees (DT), Multilayer Perceptron (MLP), Recurrent Neural Network (RNN), Long Short-Term Memory (LSTM), Gated Recurrent Unit (GRU), Dual-LSTM, and DeTransformer. These results highlight the efficacy of the DTNN model in providing precise and reliable predictions for battery RUL, making it a promising tool for battery management systems in various applications.

As one of the 100 most-threatening invasive alien species, the giant African snail (Achatina immaculata) has successfully invaded and established itself in most areas of southern China. Protection against recurrent pathogen infections is vital to biological invasion. Enhanced immune protection has been previously found in other invertebrates, but not in the unique immune system of the giant African snail. In the present study, the survival rate of the giant African snail was recorded following a second infection with lethal doses of Escherichia coli after a previous first injection using lipopolysaccharide (LPS), and the mechanism of immune enhancement was investigated by examining the cellular and transcriptomic response of the giant African snail after two successive stimuli using LPS. Snails injected first with LPS, sterilized physiologic (0.9%) saline (SPS), phosphate-buffered saline (PBS) or untreated (Blank) were rechallenged at 7d with E. coli (Ec), and were named as LPS + Ec, SPS + Ec, PBS + Ec, Ec, and Blank. The log-rank test shows the survival rate of the LPS + Ec group as significantly higher than that of other control groups after the second injection (p < 0.05). By performing cell counting and BrdU labeling on newly generated circulating hemocytes, we found that the total hemocyte count (THC) and the ratio of BrdU-positive cells to total cells increased significantly after primary stimulation with LPS and that they further increased after the second challenge. Then, caspase-3 of apoptosis protease and two antioxidant enzyme activities (CAT and SOD) increased significantly after infection, and were significantly higher in the second response than they had been in the first round. Moreover, transcriptome analysis results showed that 84 differentially expressed genes (DEGs) were expressed at higher levels in both the resting and activating states after the second immune response compared to the levels observed after the first challenge. Among them, some DEGs, including Toll-like receptor 4 (TLR4) and its downstream signaling molecules, were verified using qRT-PCR and were consistent with the transcriptome assay results. Based on gene expression levels, we proposed that these genes related to the TLR signaling cascade participate in enhanced immune protection. All results provide evidence that enhanced immune protection exists in the giant African snail.

Abstract Wireless power transfer (WPT) is a promising charging method without physical contact for the electronic products energy supply. However, low transfer efficiency, short transmission distance, as well as the electromagnetic (EM) leakage are commonly main bottlenecks of the WPT technology in the practical applications. The electromagnetic metamaterials (EMMMs) used in WPT system have opened up a brand‐new approach for better transfer properties and EM shielding. This paper is intended to provide an overview of the current studies and breakthroughs of the WPT system based on EMMMs. First, we demonstrate the fundamentals of the WPT based on the magnetic resonant coupling. Then, various key considerations of the EMMMs, such as working mechanisms, fabrications have been taken into accounted in detail. Moreover, the applications of EMMM in WPT systems are elaborated. Finally, the technical challenges in connection with working frequency and design miniaturization are investigated in this paper. This novel interdisciplinary technology shows important meanings on the extensive application of energy transmission in the future.

Abstract In an interweave cognitive radio system, cooperative spectrum sensing has been recognised as a key technology to enable secondary users to opportunistically access licensed spectrum band without harmful interference to primary users. At the same time, the unmanned aerial vehicle equipped with spectrum sensing and data transmission facilities is gaining more popularity in different applications. An unmanned aerial vehicle‐based interweave cognitive radio is investigated in which the unmanned aerial vehicle is used as a secondary user, but unlike the participation of multiple secondary users in traditional cooperative spectrum sensing, a virtual cooperative spectrum sensing model is introduced into the periodic spectrum sensing frame structure. Afterwards, the authors further propose an energy‐efficient virtual cooperative spectrum sensing with the sequential 0/1 fusion rule to reduce the average number of decisions without any loss in the detection performance. Sequentially, the authors formulate the optimisation of virtual cooperative spectrum sensing for unmanned aerial vehicle‐based interweave cognitive ratio system as the optimal sequential 0/1 fusion problem on the basis of the K‐out‐of‐N fusion rule and prove the formulated problem indeed has one optimal K, which yields the highest throughput. Finally, numerical simulations are presented to demonstrate the correctness of theoretical analyses and the effectiveness of the virtual cooperative spectrum sensing with the sequential 0/1 fusion rule.

Breast cancer (BC) accounts for the highest proportion of the all cancers among women, and necroptosis is recognized as a form of caspase-independent programmed cell death. We created prognostic signatures using univariate survival analysis, and lasso regression, to assess immune microenvironments between subgroups. We then used network pharmacology to bind our drugs to target differentially expressed genes (DEGs). A signature comprising a set of necroptosis-related genes was established to predict patient outcomes based on median risk scores. Those above and below the median were classified as high-risk group (HRG) and low-risk group (LRG), respectively. Patients at high risk had lower overall survival, and poorer predicted tumor, nodes, and metastases stages (TNM). The novel prognostic signature can effectively predict the prognosis of breast cancer patients docking of β,β-dimethyl acryloyl shikonin (DMAS) to possible targets to cure breast cancer. We found that all current prognostic models do not offer suitable treatment options. In additional, by docking drugs DMAS that have been initially validated in our laboratory to treat breast cancer. We hope that this novel approach could contribute to cancer research.

Objective To compare the clinical outcomes of fresh embryo transfers (ETs) and frozen-thawed embryo transfers (FETs) after a freeze-all cycle in women ≥40 years old with poor ovarian response (POR). Methods We performed a single-center, retrospective, case-control study of patients who underwent in vitro fertilization between January 2014 and June 2019. We included a total of 192 patients aged 40 years or older from whom 3 or fewer oocytes had been retrieved and who were receiving cleavage-stage ET in this study. Of these patients, 101 and 91 patients underwent fresh ET and FET, respectively. The primary outcome was the live birth rate (LBR) after the first ET. Logistic regression analysis was used to compare the IVF outcomes and neonatal characteristics between the fresh ET and FET groups, adjusting for maternal age, body mass index, luteinizing hormone, and the number of good quality embryos transferred. Results The mean maternal ages and number of oocytes retrieved (43.2 years and 2.3 in both groups, P=0.902 and P=0.927, respectively) were similar in the fresh ET and FET groups. No significant difference was observed between the LBRs of the fresh ET and FET groups (adjusted odds ratio, 1.28; 95% confidence interval, 0.29–5.70). The clinical pregnancy and miscarriage rates, and neonatal characteristics (birth weights and premature infant rates) were similar between the 2 groups. Conclusions FET after the freeze-all strategy had no beneficial impact on the clinical outcomes of women ≥40 years with POR.

Wireless power transfer (WPT) technology offers a potential solution for the energy-supply problem. In this paper, we propose a novel magnetic coupling mechanism for omnidirectional and multiple-pickup energy transfer based on the magnetic dipole coils. Firstly, a cubic transmitter structure capable of generating three-dimension (3D) homogeneous magnetic field is fabricated to weaken directional sensitivity of receiving coil. Furthermore, the direction of current flowing through each dipole coil is also studied in detail. Numerical and simulated analysis is implemented to verify the omnidirectionality of the transmitter. Secondly, the equivalent circuit model for WPT systems with ferrite is analyzed. In addition, optimal loads, power distribution and transfer efficiency for multiple receivers are discussed and used to achieve the proper system design. Finally, experimental prototype is set up to validate the transmission performance of the proposed WPT system. The results have showed that genuine 3D high degree of freedom (DoF) can be achieved. Meantime, above 60% efficiency at least 30 W of total output power can be obtained for WPT systems with eight pick-ups. We believe the proposed system will give a new guideline for future low power electronic applications, such as monitoring sensors, miniature robots and household devices.
